SpecLipIDA: a pseudotargeted lipidomics approach for polyunsaturated fatty acids in milk.

Polyunsaturated fatty acids (PUFAs), such as arachidonic acid (ARA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), play an important role in the nutritional value of milk lipids. However, a comprehensive analysis of PUFAs and their esters in milk is still scarce. In this study, we developed a novel pseudotargeted lipidomics approach, named SpecLipIDA, for determining PUFA lipids in milk. Triglycerides (TGs) and phospholipids (PLs) were separated using NH2 cartridges, and mass spectrometry data in the information-dependent acquisition (IDA) mode were preprocessed by MS-DIAL, leading to improved identification in subsequent targeted analysis. The target matching algorithm, based on specific lipid cleavage patterns, demonstrated enhanced identification of PUFA lipids compared to the lipid annotations provided by MS-DIAL and GNPS. The approach was applied to identify PUFA lipids in various milk samples, resulting in the detection of a total of 115 PUFA lipids. The results revealed distinct differences in PUFA lipids among different samples, with 44 PUFA lipids significantly contributing to these differences. Our study indicated that SpecLipIDA is an efficient method for rapidly and specifically screening PUFA lipids.

The association between human milk fatty acid composition in mothers with an elevated body mass index and infant growth changes

Few studies have investigated alternations in human milk polyunsaturated fatty acid (PUFA) composition in the context of maternal obesity and its effects on infant growth trajectories. This study explored whether maternal weight status and breastfeeding type influence human milk FA composition and infant anthropometry during the first six months of life. Mother-infant dyads were enrolled from the Prediction of Allergies in Taiwanese Children birth cohort study. Data concerning maternal pre-pregnancy weight, infants' breastfeeding practices, and anthropometric data were obtained regularly. We identified and compared between the composition of 30 FAs in the colostrum and 2-month milk, respectively, in obese/overweight (OB/OW) and normal-weight (NW) mothers. Multiple linear regression analyses were performed to determine the association between PUFA composition at different lactation stages and infant anthropometric parameter changes and to identify the independent variables for body mass index (BMI) z-scores by six months of age. We included 338 mother-infant dyads (OB/OW mothers, 16.9%). OB/OW mothers exhibited lower total n-3 PUFAs (P=0.035), higher ratios of arachidonic acid (C20:4n-6)/eicosapentaenoic acid (C20:5n-3)+docosahexaenoic acid (C22:6n-3), and n-6/n-3 PUFA in colostrum (P=0.037 and 0.011, respectively), and their offspring had higher body weight and BMI z-scores. Nevertheless, no PUFA composition or n-6/n-3 PUFA ratios in colostrum and 2-month milk were associated with anthropometric parameter changes by age 6 months. Infant birth weight z-scores were independently associated with BMI outcomes at age 6 months (adjusted β=0.16, 95% confidence interval (0.05-0.35), P=0.010) CONCLUSION: Neither n-3 nor n-6 PUFA profiles nor n-6/n-3 PUFA ratios at different lactation stages were found to be associated with anthropometric changes by age 6 months, suggesting that human milk PUFA composition may not be an important determinant of early infant growth trajectories.

Impact of corn silage substitution for dry alfalfa on milk fatty acid profile, nitrogen utilization, plasma biochemical markers, rumen fermentation, and antioxidant capacity in Mahabadi lactating goats

This study aimed to investigate the effects of substituting conventional dry forage with corn silage, a more cost-effective supplementary feed, on a variety of parameters in lactating Mahabadi goats, native to arid and semi-arid regions. A total of fifty lactating Mahabadi goats, aged between 2 and 5 years and weighing an average of 45.3 ± 7.20 kg, were randomly allocated into two experimental groups. The experimental treatments consisted of a control diet and a diet incorporating corn silage at 20 % of dry matter (DM). Various measurements were conducted to assess milk compositions, fatty acid profiles, rumen fermentation parameters, selected plasma biochemical parameters, nitrogen balance, and antioxidant capacity. The results revealed that the substitution of dry forage with corn silage had no statistically significant impact on milk yield, dry matter intake (DMI), body weight, as well as the proportions of monounsaturated and unsaturated fatty acids, and specifically C18:1C9 in milk, non-esterified fatty acid content in milk, rumen concentration of volatile fatty acids (VFA), microbial nitrogen, and nitrogen balance (p > 0.05). Nonetheless, this substitution led to a reduction in the percentage of milk polyunsaturated fatty acids and plasma urea nitrogen content (p < 0.05). It is noteworthy that goats fed with corn silage exhibited a notably higher antioxidant capacity in rumen liquor (p < 0.05), plasma, and milk (p < 0.01) compared to those receiving the control diet. The substitution of conventional dry forage with corn silage exhibited no adverse effects on the performance of lactating Mahabadi goats. On the contrary, it contributed to an enhancement in the antioxidant status. These findings offer valuable insights for optimizing dietary composition by incorporating corn silage at 20 % of DM, thereby improving milk quality, reducing costs, and promoting overall health in lactating goats, particularly within arid and semi-arid regions.

Effects of dietary PUFA patterns and FADS genotype on breast milk PUFAs in Chinese lactating mothers

BackgroundBreastfeeding affects the growth and development of infants, and polyunsaturated fatty acids (PUFAs) play a crucial role in this process. To explore the factors influencing the PUFA concentration in breast milk, we conducted research on two aspects: dietary fatty acid patterns and single nucleotide polymorphisms (SNPs) in maternal fatty acid desaturase genes.MethodsThree hundred seventy Chinese Han lactating mothers were recruited. A dietary semi-quantitative food frequency questionnaire (FFQ) was used to investigate the dietary intake of lactating mothers from 22 to 25 days postpartum for 1 year. Meanwhile, breast milk samples were collected from the participants and tested for the concentrations of 8 PUFAs and 10 SNP genotypes. We sought to determine the effect of dietary PUFA patterns and SNPs on breast milk PUFAs. We used SPSS 24.0 statistical software for data analysis. Statistical tests were all bilateral tests, with P < 0.05 as statistically significant.ResultsUnder the same dietary background, PUFA contents in breast milk expressed by most major allele homozygote mothers tended to be higher than that expressed by their counterparts who carried minor allele genes. Moreover, under the same gene background, PUFA contents in breast milk expressed by the mother’s intake of essential PUFA pattern tended to be higher than that expressed by their counterparts who took the other two kinds of dietary.ConclusionsOur study suggests that different genotypes and dietary PUFA patterns affect PUFA levels in breast milk. We recommend that lactating mothers consume enough essential fatty acids to ensure that their infants ingest sufficient PUFAs.

Potential sex differences in human milk fatty acids and their association with atopic dermatitis: Results of the Ulm SPATZ health study.

Polyunsaturated fatty acids (PUFAs) in human milk are essential in immune system maturation and might play a role in the development of allergic conditions, such as atopic dermatitis (AD) in infants. Immune system responses are modulated by sex, but data on the sex-specific associations with PUFAs are limited. We therefore explored sex-specific differences in human milk PUFAs and their association with AD up to 2 years. PUFAs were measured in human milk samples from the Ulm SPATZ Health Study at 6 weeks (n = 512) and 6 months (n = 367). Associations with AD up to 2 years were evaluated using crude and multivariable logistic regression. Interactions between infant sex and PUFAs were explored by including the product term. No significant associations were observed with 6-week data. At 6 months, the median relative proportion of docosahexaenoic acid (DHA) was significantly higher in milk for female than male infants (p = .001). Female infants whose milk was lower in quintile proportions of alpha-linolenic acid (ALA) at 6 months had lower odds of AD compared to males [first vs. fifth quintile OR (95% confidence interval): 0.13 (0.02, 0.66), p = .02]. This interaction was not significant when correcting for multiple testing (α threshold: p = .004). No other statistically significant associations were observed. Individual quintile PUFA proportions in human milk were not associated with AD, overall and in a sex-specific manner. More comprehensive and statistically powered longitudinal studies are needed to determine whether potential sex differences in human milk, if any, could be of clinical relevance for infants including the investigation of mediating factors.

Investigation of the possibility of obtaining butter with a high content of unsaturated fatty acids

Polyunsaturated fatty acids (PUFA) consumed with dietary fats are physiologically active substances involved in metabolic processes in the organism, in particular, carbohydrate-fat and cholesterol metabolism, regulation of redox processes. Butter is one of the main food items of the daily human diet. The basis of butter is milk fat, which is characterized by a rich fatty acid composition and unique properties. The possibility of obtaining butter with the desired ratio of saturated and unsaturated fatty acids without making adjustments to the technological process using milk with high PUFA content has been investigated. The purpose of the study was to obtain butter enriched with PUFA from the milk of Holstein cows with a modified fatty acid composition towards an increased content of unsaturated fatty acids. Material and methods. For the research, dairy raw materials from lactating Holstein cows were used, the milking herd included 881 cows. Dairy raw materials were studied, selected individually from each cow. According to the results of an in-depth assessment of the composition of milk lipid fractions, a group of cows was isolated after the first calving - 15 heads, whose milk was characterized by an increased content of unsaturated fatty acids. The content of milk fat, protein, lactose and fatty acid composition of milk were determined by infrared spectrometry with Fourier transform. Butter was produced from the milk of the experimental and control (combined) groups, which was evaluated by organoleptic (on a 20-point scale), physico-chemical parameters, including fatty acid composition determined by gas chromatography. Results. Milk obtained from cows of the experimental group differed downward from the combined milk produced in the whole herd in terms of protein and casein content by 12.8%, dry matter - by 4.4%, SOMO - by 3.1%. It varied upwards in milk sugar level (by 3.2%). In the milk fat of the milk of the experimental group of cows, the total amount of saturated fatty acids was reduced by 14.9%, and unsaturated fatty acid level was increased by 12.6%; as a result, the ratio of unsaturated to saturated fatty acids was higher by 31.7%. The butter produced from the milk of cows of the experimental group was slightly inferior to the control sample in terms of the characteristics of «taste and smell», «consistency» and total score. At the same time, the prototype fat differed from the control one by a more pronounced, saturated yellow color. Evaluation of the fatty acid composition of the experimental and control fat samples showed the advantage of the prototype in terms of PUFA content compared to the control. The total amount of unsaturated fatty acids exceeded the same indicator for the control sample by 8.3%, while the superiority of polyunsaturated fatty acids was 14.4%. At the same time, saturated fatty acid level in the experimental sample was 4.2% less, but the amount of volatile low-molecular- weight fatty acids was significantly reduced compared to the control sample - by 19.1%. Conclusion. The results obtained show the possibility and open the prospect of obtaining butter with a more favorable ratio of fatty acids in favor of PUFAs by selecting cows for milk with a higher ratio of unsaturated and saturated fatty acids and further selection aimed at an increased content of PUFAs in milk.

Oxidative stability of milk and expression of oxidative stress response, lipogenic-related, and IGF-1 genes in early-lactation cows supplemented with peanut and sunflower oils

Context Feeding dairy cows with plant oils rich in unsaturated fatty acids is a recommended strategy for increasing the proportion of polyunsaturated fatty acids in milk for human consumption. Aims We evaluated the effects of supplementation with peanut oil (high oleic acid content) and sunflower oil (high linoleic acid content) on the fatty acid profile and oxidative stability of milk and the expression of oxidative stress response, lipogenic-related and insulin-like growth factor 1 (IGF-1) genes in pasture-fed Jersey cows in early lactation. Methods Twenty-four cows were distributed in a randomised block design. Treatments were four dietary supplements of concentrate alone, or concentrate plus oil (59.6 g/kg dry matter): peanut oil, sunflower oil, or a 1:1 (v/v) mixture of peanut and sunflower oils. Analyses were performed at 30 and 60 days in milk. Key results Both peanut oil and mixed oil diets increased the hypocholesterolemic/hypercholesterolemic ratio (P &lt; 0.001) and health-promoting index (P &lt; 0.001) of milk. The sunflower oil diet increased the thrombogenicity index (P = 0.005) and conjugated diene production (P &lt; 0.001). mRNA abundance of fatty acid synthase (P = 0.003), acetyl-CoA carboxylase alpha (P = 0.001), and stearoyl-CoA desaturase (P &lt; 0.001) decreased with the sunflower oil diet. mRNA abundance of superoxide dismutase decreased (P = 0.01) with sunflower oil and mixed oil diets. The control diet tended (P = 0.08) to reduce IGF-1 mRNA abundance. Conclusions and implications Supplementation with peanut oil increases the health-promoting index of milk, whereas supplementation with sunflower oil increases susceptibility to lipid peroxidation. Supplementation with sunflower oil reduces the expression of lipogenic and antioxidant enzyme genes, and supplementation with unsaturated fats may reduce IGF-1 gene expression in early-lactation cows. The relatively small sample size means that more studies are needed.

Metagenomic insights into the modulatory effects of kelp powder (Thallus laminariae)-Treated dairy milk on growth performances and physiological lipometabolic processes of kunming mice.

Kelp powder, supplemented with a dairy cow diet, effectively improved the milk polyunsaturated fatty acids (PUFAs) content. However, little information exists on the downstream effects of the kelp-treated milk on body health, gut microbiota, and nutrient metabolism. For this purpose, 48 3-week old Kunming (KM) male mice with an average body weight of 16.1 g ± 0.2 g were randomly divided into the control treatment (CON, fed with standard chow), the common milk supplement treatment (Milk), and the kelp powder-treated milk supplement treatment (KPM). The experiment lasted for 35 days, with a 7-day long adaptive period and a 28-day long main trial. Phenotypic parameters including growth performances and serum lipids-related parameters were first measured, and results indicated that Milk and KPM supplement significantly promoted the total body weight gain (P < 0.05), while significantly decreasing the feed conversion ratio compared with CON (P < 0.05). No significant differences were observed in the blood lipids content among all three treatments, however, the triglyceride content showed a decreasing trend after KPM supplement treatment. Further, activities of liver lipometabolic-related enzymes were investigated to determine the underlying factors that impacted physiological lipid metabolism. KPM treatment showed a significant reductive effect on the activity of lipogenesis-related enzymes, such as FAS and ACC, while a significant stimulative effect on the activity of lipolysis-related enzymes included the ATGL and CPT1 compared with CON (P < 0.05). Finally, gastrointestinal tract development and cecal microbiota community that correlated with body lipid degradation and absorption were measured to determine the underlying mechanism of KPM supplementation on physiological lipid metabolism. Results indicated that supplementation with KPM significantly enhanced cecal bacteria diversity which was reflected in the significant increase of Chao1 and ACE indexes. Besides, starch-degraded bacteria such as Faecalibacterium, Ruminococcaceae, and Streptococcus are significant decreased (P < 0.05), while cellulose-degraded bacteria including Parabacteroides, Prevotella, Lactobacillus, Clostridium, and Bifidobacterium are significantly increased (P < 0.05) after KPM supplement, which may further restrict the energy generation and therefore reduce the lipid deposition. In summary, kelp supplement helped increase the milk PUFAs content, enhance the bacterial diversity and relative abundances of probiotics, which finally modulated physiological lipid metabolism, and promote growth performances.

Polyunsaturated Fatty Acid Composition in Breast Milk Plasma of HIV-infected and Uninfected Mothers in Relation to Infant Clinical Outcomes.

Background:The increased coverage of prevention of mother to child transmission (PMTCT) services has significantly reduced paediatric HIV infection incidence. The aim of the study was to compare breast milk omega-6 and omega-3 polyunsaturated fatty acid profiles of HIV infected and uninfected mothers and determine the association between fatty acid profiles and postnatal transmission of HIV, morbidity/mortality of HIV exposed and unexposed infants.Methods:A prospective cohort study of 57 HIV infected and 57 HIV uninfected lactating mothers was conducted in Gweru, Zimbabwe from July 2019 to March 2020. The women’s 114 babies (term and preterm) were also enrolled and stratified by HIV exposure and infection status. The mother-infant pairs were followed up at 6 weeks, 16 weeks and 6 months postpartum to determine, HIV transmission rate, breast milk polyunsaturated fatty acid profiles as well as infant clinical outcomes.Results:The mean breast milk docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) levels in HIV uninfected mothers (0.82 ± 0.92; 0.47 ± 0.75 μg/ml respectively) at 6 weeks postpartum were significantly higher compared to those of HIV infected mothers (0.33 ± 0.32; 0.08 ± 0.14 μg/ml) respectively. The same pattern was observed at 16 weeks postpartum in terms of DHA and EPA mean concentration. However, the arachidonic acid (AA) levels and AA/DHA ratio measured at 6 weeks postpartum were significantly higher in HIV infected mothers (2.31 ± 2.01; 17.18 ± 52.47 respectively) compared to HIV uninfected mothers (0.82 ± 0.54; 9.71 ± 21.80; P < .001). A higher morbidity rate was observed amongst HIV exposed infants than HIV unexposed infants (3.26 ± 0.13; 2.49 ± 0.09; P < .001) respectively. A significant positive correlation was observed between AA and infant morbidity (r = .388; P < .001).Conclusion:Deficiencies in breast milk omega-3 fatty acids were observed in HIV infected women. Maintaining a healthy balance between omega-6 and omega-3 fatty acid diets is critical for breast feeding mothers regardless of their HIV status. The adverse clinical outcomes observed amongst HIV exposed infants emphasise their vulnerability under conditions of maternal universal antiretroviral therapy.

Fatty Acids Profile , and ∆9 -Desaturase Index of Milk from Barki Ewes Fed Diets Supplemented with Spirulina Platensis or Fish Oil

AbstractThe current study aimed to evaluate milk fat and fatty acids profile, nutrients digestibility and lactating ewes’ performance as a response to supplementing diets with spirulina algae or fish oil in lactating barki ewes. Three experimental diets were used as follow: Control diet based on 60:10:30 concentrate feed mixture (CFM), clover hay, and bean straw (T1); control diet supplemented with 10 ml of fish oil/ kg DM (T2), control diet supplemented with 5 g Spirulina Platensis / kg DM (T3). The results showed that T2 significantly improved nutrients digestibility (DM, OM, NDF, ADF, EE) as compared with control (T1). No differences (p>0.05) were recorded between experimental groups in different blood serum parameters, milk yield, energy corrected milk (ECM), milk fat (%) and milk total solids yield. Supplementing diet with Spirulina platensis (T3) increased CLA, C18:3 n6, C18:3 n3. T2 and T3 increased milk poly-unsaturated fatty acid and declined ∆9 desaturase index as compared with T1. The results of the current study suggested that supplementing diets with fish oil or Spirulina platensis had a potential positive impact in milk fatty acid profile which have a healthy benefit.

The effect of feeding whole-crop barley or black oat silage in the fatty acid content of milk from cows in small-scale dairy systems in the highlands of Mexico.

Diets have an important effect on the fatty acid (FA) content in milk from dairy cows so that feeding strategies may modify the FA profile. There is little information on the fatty acid contents of whole-crop barley (BLY) and black oat (BKO) silages and on milk from cows fed these silages that are an alternative forage for small-scale dairy systems (SSDS). Given the growing interest in foods with favourable health attributes, the objective was to identify the FA content of milk from dairy cows fed whole-crop BLY and BKO silages in small-scale systems. Three inclusion levels of BLY and BKO silage were assessed in nine lactating cows at pasture for 8h/day and supplemented with 4.6kg/DM/cow/day commercial concentrate. Treatments were BLY: 10kg DM/cow/day BLY silage; BKO: 10kg DM/cow/d BKO silage; and BLY/BKO: 5kg BLY and 5kg DM/cow/d BKO silages (50:50). A 3 × 3 Latin square design repeated three times with nine cows and 14-day periods was utilised. Variables evaluated were the content of fatty acids present in feeds and milk. Contents of linoleic (C18:2n-6) and linolenic (C18:3n-3) acids, as precursors of polyunsaturated fatty acids, were 46 in BLY and 50 BKO g/100g, lower than for multispecies pasture (61g/100g). There were statistical differences in the content of short-chain fatty acids in milk (P < 0.05), being lower for BLY (18.9g/100g) compared with BKO (20.4g/100g) and BLY/BKO (20.6g/100g), the saturated fatty acids and linoleic acid (C18:2n-6c) were higher in BLY/BKO in relation to BLY and BKO. Content of health-promoting polyunsaturated fatty acids in milk was higher in BLY/BKO (P < 0.05). There were no differences (P < 0.05) in the atherogenic index, with values within reports. Small-grain cereal forages may produce milk with a favourable fatty acid content for human health.

Effects of partially replacing grass silage by lucerne silage cultivars in a high-forage diet on ruminal fermentation, methane production, and fatty acid composition in the rumen and milk of dairy cows

Lucerne silages of the Kometa and Verko cultivars were investigated in four consecutive experiments to evaluate their effects on ruminal fermentation and fatty acid (FA) proportions in the rumen fluid and milk of dairy cows. Initially, two successive in vitro experiments were performed using a semicontinuous culture system (RUSITEC). In Experiment 1, the treatments were: (1) MGS: a high-forage diet (65:35 forage:concentrate ratio; DM basis) containing maize silages (MS) and grass silages (GS) as the main forage sources, (2) MKLS: a high-forage diet containing MS and Kometa lucerne silage (KLS) as the main forage sources; and (3) sMKLS: a high-forage diet containing MS and KLS as the main forage sources and supplemented with sufficient saponin extract powder from KLS to give a saponin content equal to the in the KLS. The treatments in Experiment 2 (MGS, MVLS, or sMVLS) were similar to those in Experiment 1, except that Verko Lucerne silage (VLS) and its saponin extract were used. Methane concentration statistically insignificant decreased (P = 0.97) as a result of the MKLS treatment, while digestible dry matter (DDM) was increased (P = 0.002) for MKLS and sMKLS. However, neither methane concentration nor DDM were affected by MVLS or sMVLS. We also performed two in vivo experiments: in Experiment 3, we used six cannulated Polish Holstein-Friesian cows fitted with rumen cannulas, and in Experiment 4, we used 54 productive Polish Holstein-Friesian cows. The treatments for these two experiments were based on partial replacement of GS with KLS or VLS in the high-forage diet containing MS, as described earlier: MGS, MKLS, and MVLS. In Experiment 3, the methane concentration decreased (P < 0.001) for both MKLS and MVLS, with the diets lowering the total bacterial and protozoal counts and the total methanogens (P ≤ 0.002). Interestingly in Experiment 4, only MVLS led to a decrease in fat content and energy (P ≤ 0.02). MKLS and MVLS were also associated with lower levels of milk polyunsaturated fatty acid (P < 0.001) and higher levels of milk saturated fatty acid (P < 0.001). MKLS had more promising fatty acid composition than MVLS, with clearer methane mitigation. In conclusion, the Kometa lucerne silages inhibited methane emission and microbial population, but at the cost of increasing the saturated fatty acids in the milk fat.

The effects of dietary ricinoleic acid from castor oil on the zootechnical traits and haemato-biochemical profile of lactating Kankrej cows

The aim of the present study was to evaluate the effects of dietary ricinoleic acid from castor oil on the milk yield, composition, fatty acid profile and haemato-biochemical profile in lactating Kankrej cows. Twenty lactating Kankrej cows were assigned to the following groups: control (CON), basal diet without any additive and treatment (RAS), basal diet with 2 g/animal/day of ricinoleic acid from castor oil. Dietary ricinoleic acid had no (P&gt;0.05) effect on dry matter intake and feed efficiency. Milk yield (milk, 4% fat corrected milk and energy corrected milk) were not affected (P&gt;0.05) by the feeding of ricinoleic acid. The milk composition showed no significant differences between the groups. A significantly (P&lt;0.05) higher percentage of C4:0, C6:0 and C8:0 was found in the milk from the RAS group than the CON group. A lower (P&lt;0.05) milk C16:0 percentage was observed in the RAS group as compared to the CON. The percentages of C18:2 and C18:3 were found to be increased in the RAS group. There was a decrease (P = 0.055) in milk saturated fatty acids but an increase in milk unsaturated fatty acids percentage in the RAS as compared to the CON group. The milk polyunsaturated fatty acids were significantly (P&lt;0.05) increased in the RAS group. There was no difference (P&gt;0.05) in haemato-biochemical profile between the CON and RAS groups. It was concluded that supplementation of 2 g/animal/day ricinoleic acid from castor oil had no effect on milk yield, milk composition and haemato-biochemical profile. However, it increased the milk fat percentages of unsaturated fatty acids and polyunsaturated fatty acids, and decreased milk saturated fatty acids, without any adverse effect on the health status of the cows.

Sex-specific associations of human milk long-chain polyunsaturated fatty acids and infant allergic conditions.

Polyunsaturated fatty acids (PUFAs) may influence immune development. We examined the association of PUFAs in human milk with food sensitization and atopic dermatitis among breastfed infants. In a selected subgroup of 1109 mother-infant dyads from the CHILD Cohort Study, human milk was analyzed by gas-liquid chromatography to quantify PUFAs including arachidonic acid (ARA) and docosahexaenoic acid (DHA). At 1year of age, food sensitization was determined by skin-prick testing for egg, peanut, cow's milk, and soybean, and atopic dermatitis was diagnosed by pediatricians. Logistic regression analyses controlled for breastfeeding exclusivity, family history of atopy, and other potential confounders. Overall, 184 infants (17%) were sensitized to one or more food allergens and 160 (14%) had atopic dermatitis. Sex-specific associations were observed between these conditions and milk PUFAs. Girls receiving human milk with lower proportions of DHA had lower odds of food sensitization (aOR 0.35; 95% CI 0.12, 0.99 for lowest vs highest quintile), and a clear dose-dependent association was observed for the ARA/DHA ratio (aOR 2.98; 95% CI 1.10, 8.06 for lowest vs highest quintile). These associations were not seen in boys. Similar sex-specific tendencies were observed for atopic dermatitis. Human milk PUFA proportions and their ratios are associated with infant atopic conditions in a sex-specific manner. In female infants, a higher ratio of ARA/DHA may reduce the risk of food sensitization and atopic dermatitis. Further research is needed to determine the underlying mechanisms and clinical relevance of this sex-specific association.

The impact of the dietary supplementation level with Schizochytrium sp. on milk chemical composition and fatty acid profile, of both blood plasma and milk of goats

• 20 g Schizochytrium sp./goat/day enhances milk PUFA. • 20 g Schizochytrium sp./goat/day didn’t affect negatively the milk fat. • Both 40 and 60 g Schizochytrium sp./goat/day induce milk fat depression. • Both trans-10, cis-12 C 18:2 and c is-9 C 18:1 have pivotal role in goats’ milk fat. Microalgae, have gained significant attention as a source of beneficial biomolecules such as long chain n-3 polyunsaturated fatty acid (PUFA), and are considered to be an effective strategy to enhance ruminant milk with PUFA. However, milk fatty acids (FA) profile is affected by ruminant biohydrogenation process and consequently by possible adaptations of rumen microbiome related with the duration of diet supplementation. Thus, the aim of the present study was to investigate a long-term response of dietary inclusion of microalgae Schizochytrium sp., at three different levels with moderate forage to concentrate ratio (50/50) on milk chemical composition and fatty acid profile of both goats’ blood plasma and milk. For this purpose, twenty-four dairy goats were divided into four homogeneous sub-groups. All the goats were fed individually with alfalfa hay and concentrates. The concentrates of the control group (CON) had no microalgae, while those of the treated groups were supplemented daily with 20 (ALG20), 40 (ALG40) and 60 (ALG60) g Schizochytrium sp./kg concentrates. The mean milk yield was not affected, while the fat content was decreased in ALG40 and ALG60 groups, compared with the CON and ALG20 ones. In blood plasma the proportions of C 16:0 , C 18:0 , cis-9 C 18:1 and C 18:2n6 c decreased (P < 0.05), while those of trans-11 C 18:1 , C 20:3n3 , C 22:2n6 , C 22:5n6 and C 22:6n3 were increased in the microalgae treated groups compared with the CON. The proportions of C 14:0 , C 16:0 , trans-10 C 18:1 , trans-11 C 18:1 , cis-9, trans-11 C 18:2 , trans-10, cis-12 C 18:2 , C 22:6n-3 (DHA), C 22:5n-6 (DPA) and the total n-3 fatty acids were increased significantly in the milk of treated goats, while those of C 18:0 , cis-9 C 18:1 and C 18:2n6c were decreased. In conclusion, the dietary inclusion of 20 g Schizochytrium sp./goat/day, combined with a forage to concentrate ratio of 50/50, enhances the PUFA of goats’ milk without negative effects on its chemical composition.

The potential role of organic and conventional yoghurt consumption in the treatment of non-alcoholic fatty liver disease

Health is an important motivation for the consumption of both organic and functional foods. Organic food contains fewer pesticide residues and statistically more selected health-related compounds such as polyphenols in plant products and polyunsaturated fatty acids in milk and meat products. Recent studies suggest that the gut-liver axis plays an important role in the pathogenesis of non-alcoholic fatty liver disease, so probiotics could be a therapeutic tool. Comparing the health effects of yoghurt from organic origin with so-called conventional yoghurt is difficult, because there is no biomarker that would signal the difference with good specificity and sensitivity. The aim of this study was to investigate numerous biomarkers to evaluate the difference between yoghurt from conventional and organic origin and their health effects in NAFLD. We performed a prospective, cohort study consisting of 37 (age = 51.73 ± 11.82, male = 21, female = 16) patients with NAFLD at the 2nd Department of Internal Medicine of the Semmelweis University, Budapest. Diagnosis of NAFLD was based on ultrasonography and the exclusion of other etiololgy. The patients were examined also with shear wave elastography to evaluate the hepatic fibrosis stage. We divided the patients randomly into two groups. The patients consumed individually daily 300 grams of yoghurt from organic (n = 21) or conventional (n = 16) origin for 8 weeks. We collected 37 routine laboratory data, measured 4 cytokines, 3 markers of the redox-homeostasis and 14 body composition values before, after, and 12 weeks after the yoghurt consumption. We found a mild elevation of vitamin D and a minimal reduction of LDL after the yoghurt consumption, but in the other 35 routine laboratory data there was no statistical difference. Adiponectin and leptin levels were elevated after the yoghurt consumption in the "conventional group". In contrast, we found significant decrease of adiponectin levels in the "organic group" after the treatment. Only the adiponectin tendency was different in the two groups. The induced free radical content was also statistically lower after the yoghurt consumption. In the body composition measurements, there were no significant differences. These data suggest that adiponectin could be a possible biomarker to evaluate the effectiveness of probiotic treatment in non-alcoholic fatty liver disease. Impact of inbreeding on milk fatty acids of a Brazilian Holstein cattle

Context Reproductive technologies may either contribute to the increase of genetic gains or inbreeding levels in dairy cattle breeding programs and herds. In general, studies have evaluated the impacts of inbreeding on traditional traits, such as production and reproduction. However, studies on novel traits, such as milk fatty acids profile, are scarce in the literature. Aim We aimed to evaluate the impact of inbreeding levels on milk fatty acids profile measured in a Brazilian Holstein cattle population. Methods Monthly records of the percentages (%) of palmitic, stearic, oleic, total saturated, unsaturated, monounsaturated and polyunsaturated fatty acids in milk were collected from 2047 Holstein cows between May and December 2012. The pedigree file contained 7963 animals over seven generations. The inbreeding trend was evaluated by linear regression of inbreeding coefficients on generations. The inbreeding impacts on breeding and phenotypic values were evaluated by linear regression of breeding values and phenotypic values on generations respectively. Breeding values were obtained by a Bayesian approach based on an animal model that included the systematic effect of contemporary group, cubic effect of days in milk as a covariate, and the random direct additive genetic and animal permanent environmental effects. Key results The average inbreeding coefficient was 0.28% for all animals, and 1.39% for inbred animals. Inbreeding tended to increase 0.24% per generation (P-value &amp;lt;0.0001). The regression coefficients of phenotypic values on inbreeding were not significant for saturated, unsaturated, monounsaturated, palmitic, stearic and oleic fatty acids (P-values &amp;gt;0.18, 0.10, 0.16, 0.26, 0.51 and 0.09 respectively); whereas for polyunsaturated fatty acids, they were significant (P-value &amp;lt;0.04), with an increase of –0.000978% on phenotypic values for each 1% increase in inbreeding. The regression coefficients of breeding values on inbreeding were not significant for unsaturated, monounsaturated, polyunsaturated, stearic and oleic fatty acids (P-values &amp;gt;0.33, 0.78, 0.48, 0.32 and 0.07 respectively). However, they were significant for saturated and palmitic fatty acids (P-values &amp;lt;0.03 and 0.0001 respectively), presenting an increase of 0.000689% and 0.000283% respectively on breeding values for each 1% increase in inbreeding. Conclusions There was a significant increase in inbreeding coefficients over the studied generations; however, the impacts on both breeding and phenotypic values were small, when significant. Implications Our study may contribute to the understanding of how inbreeding levels may affect the fatty acids profile in a Brazilian Holstein cattle population, as well as encourage future research on this subject.

Effects of flaxseed oil supplementation to lactating goats diet on milk production, composition and fatty acids profiles

Milk goats is of particular nutritional and economic importance in many parts of the world. This study aimed to define the effect of quaffing dairy goats with linseed oil )10, 20 and 30 mL/head/day) on milk productioncomposition and fatty acids profile. Twenty-four lactating Zaraibi goats at the last weeks of gestation were used. Experimental animals were randomly divided into four equal groups. Result obtained revealed that flaxseed oil supplementation increased milk yield by a mean value of 19.79% as compared with control treatment. Milk fat percentage was higher for goats received 30 mL. flaxseed oil than other treatments, while no remarkable effect of flaxseed oil supplementation on milk protein, lactose and ash contents was detected. Addition of flaxseed oil to goats' diet resulted in reducing saturated fatty acids by a mean value of 13.72% as compared with control sample, however, these decreases may be influenced partially by lactation progress. In contrast, the concentration of unsaturated fatty acids increased with a mean value of 28.32%, with pronounced increases in oleic, linoleic and linolenic acids concentration as compared with control. Moreover, results obtained revealed that ά-linolenic acid (ALA) concentrations achieved greatly affect by flaxseed oil supplementation, where their concentrations varied from 0.2-3.04%. In conclusion, feeding flaxseed to lactating goats can be used as a nutritional strategy to reduce concentrations of short-chain and saturated fatty acids and to increasing long-chain and polyunsaturated fatty acids in milk, as well as to improve milk yield. Consequently, such strategy has a significant effect for human's nutrition and health.

The effect of dietary forage to concentrate ratio and forage type on milk fatty acid composition and milk fat globule size of lactating cows

We examined the effects of 2 grass silage-based diets differing in forage:concentrate (FC) ratio and those of a red clover silage-based diet on intake, milk production, ruminal fatty acid (FA) biohydrogenation, milk FA composition, and milk fat globule (MFG) size distribution. Ten multiparous Nordic Red cows received the following treatments: grass silage-based diets containing high (70:30, HG) or low (30:70, LG) FC ratio or a red clover silage-based diet with an FC ratio of 50:50 (RC) on a dry matter basis. Determinations of MFG were performed from fresh milk samples without addition of EDTA so the results of fat globules >1 µm in diameter are emphasized instead of the entire globule population. Lower FC ratio in grass silage-based diets increased milk production with no effect on daily fat yield, leading to 13% lower milk fat concentration. The effect of FC ratio on MFG size was moderate. It did not affect the volume-weighted diameter in grass silage-based diets, although LG lowered the volume-surface diameter of MFG in the size class >1 µm compared with HG. Compared with HG, feeding LG moderately decreased the biohydrogenation of 18:2n-6, leading to a higher level of polyunsaturated fatty acids in milk fat. Feeding RC lowered milk fat concentration and daily milk fat yield compared with grass silage-based diets. The volume-weighted diameter of MFG in the size class >1 µm was smaller in RC milk compared with grass silage-based diets. Feeding RC increased the flow of 18:3n-3 at the omasum by 2.4-fold and decreased the apparent ruminal 18:3n-3 biohydrogenation compared with grass silage-based diets despite similar intake of 18:3n-3. It also resulted in the lowest amount of saturated FA and the highest amounts of cis-9 18:1, 18:3n-3, and polyunsaturated FA in milk. In conclusion, LG decreased milk fat content and induced minor changes in MFG size distribution compared with HG, whereas RC lowered milk fat production, altered milk FA composition to nutritionally more beneficial direction, and led to smaller MFG compared with grass silage-based diets.

Effects of Azgp1−/−on mammary gland, adipose tissue and liver gene expression and milk lipid composition in lactating mice

The expression of Azgp1 gene, an adipokine involved in the mobilization of body reserves, was observed in mammary gland of ruminants. Its regulation by different dietary conditions suggests a potential role in the mechanisms controlling the composition of milk fat. The aim of this study was to evaluate the role of Azgp1 during lactation. Azgp1−/− mice were compared to wild-type to determine its effects on milk fatty acid composition and offspring growth. To determine its effects on mammary gland, adipose tissue and liver gene expression, gene expression was analyzed using RT-qPCR via TLDA analyses. The body weight of Azgp1−/− mothers was slightly higher after parturition and at 10 days of lactation compared to the wild type. The milk polyunsaturated fatty acid content was increased in Azgp1−/− mice. Among the 40 genes studied, Azgp1−/− modified the expression of 9, 10 and 3 genes in mammary gland, adipose tissue and liver, respectively. These genes, involved in fatty acid synthesis, transport and triglyceride synthesis, were downregulated in Azgp1−/− mice showing a particularity during lactation. Changes in mammary gland gene expression may explain the modifications observed in milk fatty acid composition. This study supports a role of Azgp1 on lipid metabolism, in particular in mammary gland, during lactation function.
