Acceleration a yeast-based biodegradation process of polyethylene terephthalate microplastics by Tween 20: Efficiency, by-product analysis, and metabolic pathway Prediction

Polyethylene terephthalate is a widely produced plastic polymer that exhibits considerable biodegradation resistance, making its derived microplastics ubiquitous environmental pollutants. In this study, a new yeast strain (Vanrija sp. SlgEBL5) was isolated and found to have lipase and esterase-positive capabilities for degrading polyethylene terephthalate microplastics. This isolate changed the microplastic surface charge from −19.3 to +31.0 mV and reduced more than 150 μm of its size in addition to reducing the intensity of the terephthalate, methylene, and ester bond functional groups of the polymer in 30 days. Tween 20 as a chemical auxiliary treatment combined with biodegradation increased the microplastic degradation rate from 10 to 16.6% and the thermal degradation rate from 85 to 89%. Releasing less potentially hazardous by-products like 1,2 diethyl-benzene despite the higher abundance of long-chain n-alkanes, including octadecane and tetracosane was also the result of the bio + chemical treatment. Altogether, the findings showed that Vanrija sp. SlgEBL5 has the potential as a biological treating agent for polyethylene terephthalate microplastics, and the simultaneous bio + chemical treatment enhanced the biodegradation rate and efficiency.

Entry pathways determined the effects of MPs on sludge anaerobic digestion system: The views of methane production and antibiotic resistance genes fates

Sludge is one of the primary reservoirs of microplastics (MPs), and the effects of MPs on subsequent sludge treatment raised attention. Given the entry pathways, MPs would exhibit different properties, but the entry pathway-dependent effect of MPs on sludge treatment performance and the fates of antibiotic resistance genes (ARGs), another high-risk emerging contaminant, were seldom documented. Herein, MPs with two predominant entry pathways, including wastewater-derived (WW-derived) and anaerobic digestion-introduced (AD-introduced), were used to investigate the effects on AD performance and ARGs abundances. The results indicated that WW-derived MPs, namely the MPs accumulated in sludge during the wastewater treatment process, exhibited significant inhibition on methane production by 22.8%–71.6%, while the AD-introduced MPs, being introduced in the sludge AD process, slightly increased the methane yield by 4.7%–17.1%. Meanwhile, MPs were responsible for promoting transmission of target ARGs, and polyethylene terephthalate MPs (PET-MPs) showed a greater promotion effect (0.0154–0.0936) than polyamide MPs (PA-MPs) (0.0013–0.0724). Compared to size, entry pathways and types played more vital roles on MPs influences. Investigation on mechanisms based on microbial community structure revealed characteristics (aging degree and types) of MPs determined the differences of AD performance and ARGs fates. WW-derived MPs with longer aging period and higher aging degree would release toxics and decrease the activities of microorganisms, resulting in the negative impact on AD performance. However, AD-introduced MPs with short aging period exhibited marginal impacts on AD performance. Furthermore, the co-occurrent network analysis suggested that the variations of potential host bacteria induced by MPs with different types and aging degree attributed to the dissemination of ARGs. Distinctively from most previous studies, the MPs with different sizes did not show remarkable effects on AD performance and ARGs fates. Our findings benefited the understanding of realistic environmental behavior and effect of MPs with different sources.

Adsorption of Macrolide Antibiotics and a Metabolite onto Polyethylene Terephthalate and Polyethylene Microplastics in Aquatic Environments.

Microplastics (MPs) and antibiotics are emerging pollutants widely found in aquatic environments, potentially causing environmental harm. MPs may act as carriers for antibiotics, affecting their environmental distribution. This study investigates the adsorption of four macrolide antibiotics and a metabolite onto two types of MPs: polyethylene terephthalate (PET) and polyethylene (PE). Results revealed a linear isotherm adsorption model, with higher adsorption to PET than to PE (R2 > 0.936 for PE and R2 > 0.910 for PET). Hydrophobic interactions and hydrogen bonding could be the main adsorption mechanisms, with pore filling potentially involved. Reduced particle size enhances adsorption due to the increase of active adsorption sites. This increasement is more pronounced in PE than in PET, leading to an 11.6% increase in the average adsorption of all macrolides to PE, compared to only 5.1% to PET. Dissolved organic matter inhibits adsorption (azithromycin adsorption to PE was reduced from 12% to 5.1%), while salinity enhances it just until 1% salinity. pH slightly influences adsorption, with maximal adsorption at neutral pH. Results in real samples showed that complexity of the matrix decreased adsorption. Overall, these findings indicate that PE and PET MPs can be a vector of macrolides in aquatic environments.

Development and characterization of a bacterial enzyme cascade reaction system for efficient and stable PET degradation

The widespread use of polyethylene terephthalate (PET) in various industries has led to a surge in microplastics (MPs) pollution, posing a significant threat to ecosystems and human health. To address this, we have developed a bacterial enzyme cascade reaction system (BECRS) that focuses on the efficient degradation of PET. This system harnesses the Escherichia coli (E. coli) surface to display CsgA protein, which forms curli fibers, along with the carbohydrate-binding module 3 (CBM3) and PETases, to enhance the adsorption and degradation of PET. The study demonstrated that the BECRS achieved a notable PET film degradation rate of 3437 ± 148 μg/(d*cm²), with a degradation efficiency of 21.40% for crystalline PET MPs, and the degradation products were all converted to TPA. The stability of the system was evidenced by retaining over 80% of its original activity after multiple uses and during one month of storage. Molecular dynamics simulations confirmed that the presence of CsgA did not interfere with the enzymatic activity of PETases. This BECRS represents a significant step forward in the biodegradation of PET, particularly microplastics, offering a practical and sustainable solution for environmental pollution control.

Toxic effects of fragmented polyethylene terephthalate particles on the marine rotifer Brachionus koreanus: Based on ingestion and egestion assay, in vivo toxicity test, and multi-omics analysis

Microplastics (MPs) are a major concern in marine ecosystem because MPs are persistent and ubiquitous in oceans and are easily consumed by marine biota. Although many studies have reported the toxicity of MPs to marine biota, the toxicity of environmentally relevant types of MPs is little understood. We investigated the toxic effects of fragmented polyethylene terephthalate (PET) MP, one of the most abundant MPs in the ocean, on the marine rotifer Brachionus koreanus at the individual and molecular level. No significant rotifer mortality was observed after exposure to PET MPs for 24 and 48 h. The ingestion and egestion assays showed that rotifers readily ingested PET MPs in the absence of food but not when food was supplied; thus, there were also no chronic effects of PET MPs. In contrast, intracellular reactive oxygen species levels and glutathione S-transferase activity in rotifers were significantly increased by PET MPs. Transcriptomic and metabolomic analyses revealed that genes and metabolites related to energy metabolism and immune processes were significantly affected by PET MPs in a concentration-dependent manner. Although acute toxicity of PET MPs was not observed, PET MPs are potentially toxic to the antioxidant system, immune system, and energy metabolism in rotifers.

Mechanism of microplastics effects on the purification of heavy metals in piggery effluents by microalgae

Microalgae is an effective bioremediation technique employed for treating piggery effluent. However, there is insufficient study on how the presence of microplastics (MPs) in wastewater affects the ability of microalgae to remove heavy metals from piggery effluent. This study aims to investigate the influence of two prevalent heavy metals found in piggery wastewater, Cu2+ (2 mg/L) and Zn2+ (2 mg/L), on their removal by microalgae (Desmodesmus sp. CHX1) in the presence of four types of MPs: polyethylene (PE), polyvinyl chloride (PVC), polypropylene (PP), and polyethylene terephthalate (PET). The results revealed that smaller particle size MPs promoted chlorophyll accumulation, while larger particles inhibits it. Additionally, higher concentrations of MPs promoted chlorophyll accumulation, while lower concentrations inhibited it. As for heavy metals, the presence of microplastics reduced the removal efficiency of Cu2+ and Zn2+ by Desmodesmus sp. CHX1. The highest inhibition of Cu2+ was 30%, 10%, 19%, and 16% of the control (CK), and the inhibition of Zn2+ was 7%, 4%, 4%, and 13%, respectively, under the treatments of PE, PVC, PP and PET MPs. Furthermore, Desmodesmus sp. CHX1 can secrete more extracellular polymeric substances (EPS) and form heterogeneous aggregates with MPs to counteract their pressure. These findings elucidate the impact of MPs on microalgae in bioremediation settings and offer useful insights into the complex relationships between microalgae, MPs, and heavy metals in the environment.

Impact of Polyethylene Terephthalate Microplastics on Drosophila melanogaster Biological Profiles and Heat Shock Protein Levels.

Microplastics and nanoplastics are abundant in the environment. Further research is necessary to examine the consequences of microplastic contamination on living species, given its widespread presence. In our research, we determined the toxic effects of PET microplastics on Drosophila melanogaster at the cellular and genetic levels. Our study revealed severe cytotoxicity in the midgut of larvae and the induction of oxidative stress after 24 and 48 h of treatment, as indicated by the total protein, Cu-Zn SOD, CAT, and MDA contents. For the first time, cell damage in the reproductive parts of the ovaries of female flies, as well as in the accessory glands and testes of male flies, has been observed. Furthermore, a decline in reproductive health was noted, resulting in decreased fertility among the flies. By analyzing stress-related genes such as hsp83, hsp70, hsp60, and hsp26, we detected elevated expression of hsp83 and hsp70. Our study identified hsp83 as a specific biomarker for detecting early redox changes in cells caused by PET microplastics in all the treated groups, helping to elucidate the primary defense mechanism against PET microplastic toxicity. This study offers foundational insights into the emerging environmental threats posed by microplastics, revealing discernible alterations at the genetic level.

Pristine and UV-Weathered PET Microplastics as Water Contaminants: Appraising the Potential of the Fenton Process for Effective Remediation

Polyethylene terephthalate (PET) microplastics constitute a significant portion of plastic pollution in the environment and pose substantial environmental challenges. In this study, the effectiveness of the Fenton process and post-oxidation coagulation for the removal of non-weathered and UV-weathered PET microplastics (PET MPs) were investigated. A response surface methodology was used to investigate the interplay between PET concentration and ferrous ion (Fe2+) concentration. The models revealed an intricate interplay between these variables, highlighting the need for a balanced system for optimal PET MP removal. For non-weathered PET, the simultaneous increase in the concentrations of both PET microplastics and Fe2+ was found to enhance the removal efficiency. However, this synergistic effect was not observed in UV-weathered PET, which also demonstrated a more pronounced effect from the Fe2+ concentration. The statistical analysis provided a strong basis for the validity of the models. X-ray photoemission spectroscopy (XPS) further elucidated the mechanisms behind these findings, revealing that UV weathering results in surface changes, which facilitate hydroxyl radical oxidation. These findings underline the complexity of the Fenton process in PET microplastic removal and the different behavior of non-weathered and UV-weathered microplastics. This has significant implications for tailoring remediation strategies and underscores the importance of considering environmental weathering in these strategies.

Baking releases microplastics from polyethylene terephthalate bakeware as detected by optical photothermal infrared and quantum cascade laser infrared

The use of plastic bakeware is a potential source of human exposure to microplastics (MPs). However, characterizing MPs remains a challenge. This study aims to employ optical photothermal infrared (O-PTIR) and quantum cascade laser infrared (QCL-IR) technology to characterise polyethylene terephthalate (PET) MPs shed from PET bakeware during the baking process. The bakeware, filled with ultrapure water, underwent baking cycles at 220 °C for 20 min, 60 min, and three consecutive cycles of 60 min each. Subsequently, particles present in the ultrapure water were collected using an Al2O3 filter. O-PTIR and QCL-IR were used to characterise PET MPs collected from the filtration. Analysis revealed that QCL-IR spectra exhibited broader absorption peaks, compared to O-PTIR. Notably, MP spectra obtained from both techniques displayed common absorption peaks around 1119, 1623, 1341 and 1725 cm−1. The dominant size of PET MPs detected by O-PTIR and QCL-IR was 1–3 μm and 5–20 μm, respectively. The quantity of identified PET MPs using O-PTIR was 18 times greater than that with QCL-IR, which was attributed to variations in spatial resolution, sampling methods for spectra collection, and data analysis employed by the two methods. Importantly, findings from both techniques highlighted a notably large quantity of MPs released from PET bakeware, particularly evident after 3 cycles of 60 min of baking, suggesting a substantial increase in the potential ingestion of MPs, especially in scenarios involving extended baking durations. The research outcomes will guide consumers on minimizing the intake of microplastics by using PET bakeware for shorter baking time. Additionally, the study will yield valuable insights into the application of O-PTIR and QCL-IR for MPs detection, potentially inspiring advancements in MPs detection methodologies through cutting-edge technologies.

Potential Effects of Orally Ingesting Polyethylene Terephthalate Microplastics on the Mouse Heart.

Polyethylene terephthalate microplastics (PET MPs) are widespread in natural environment, and can enter organisms and accumulate in the body, but its toxicity has not been well studied. Therefore, in order to investigate the toxic effects of PET microplastics on mammals, this study investigated the toxic effects of PET MPs on ICR mice and H9C2 cells by different treatment groups. The results indicated the cardiac tissue of mice in the PET-H (50µg/mL) group showed significant capillary congestion, myocardial fiber breakage, and even significant fibrosis compared to the PET-C (control) group (P < 0.01). Results of the TUNEL assay demonstrated significant apoptosis in myocardial tissue in the PET-H and PET-M (5µg/mL) groups (P < 0.01). Meanwhile, Western blotting showed increased expression of the apoptosis-related protein Bax and decreased expression of PARP, caspase-3, and Bcl-2 proteins in both myocardial tissues and H9C2 cells. In addition, flow cytometry confirmed that PET MPs decreased the mitochondrial membrane potential and apoptosis in H9C2 cells; however, this trend was reversed by N-acetylcysteamine application. Moreover, PET MP treatment induced the accumulation of reactive oxygen species (ROS) in H9C2 cells, while the MDA level in the myocardial tissue was elevated, and the activities of catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) were decreased (P < 0.01), indicating a change in the redox environment. In conclusion, PET MPs promoted cardiomyocyte apoptosis by inducing oxidative stress and activating mitochondria-mediated apoptotic processes, ultimately leading to myocardial fibrosis. This study provides ideas for the prevention of PET MP toxicity and promotes thinking about enhancing plastic pollution control.

Elucidating adsorption mechanisms of benzalkonium chlorides (BACs) on polypropylene and polyethylene terephthalate microplastics (MPs): Effects of BACs alkyl chain length and MPs characteristics

Since the onset of the COVID-19 pandemic, there has been an increase in the use of disposable plastics and disinfectants. This study systematically investigated the adsorption behavior and mechanisms of benzalkonium chlorides (BACs), commonly used disinfectants, on polypropylene (PP) and polyethylene terephthalate (PET) microplastics (MPs), considering various factors, such as characteristics of MPs, alkyl chain length of BACs, and environmental conditions. Our results demonstrated a higher adsorption capacity for PP-MPs with relatively hydrophobic properties compared to PET-MPs, where longer alkyl chains in BACs (i.e., higher octanol-water partition coefficients, Kow) significantly enhanced adsorption through hydrophobic interactions. The inverse relationship between particle size of MPs and adsorption was evident. While changes in pH minimally affected adsorption on PP-MPs, adsorption on PET-MPs increased with rising pH, highlighting the influence of pH on electrostatic interactions. Moreover, MP aging with UV/H2O2 amplified BAC adsorption on PP-MPs due to surface oxidation and fragmentation, whereas the properties of PET-MPs remained unaltered, resulting in unchanged adsorption capacities. Spectroscopy studies and density functional theory (DFT) calculations confirmed hydrophobic and electrostatic interactions as the primary adsorption mechanisms. These findings improve our understanding of MPs and BACs behavior in the environment, providing insights for environmental risk assessments related to combined pollution.

When polyethylene terephthalate microplastics meet Perfluorooctane sulfonate in thermophilic biogas upgrading system: Their effect on methanogenesis

Microplastics (MPs) and Perfluorooctane sulfonate (PFOS) are two hard-biodegradable pollutants widely existing in the waste streams treated by anaerobic digestion. However, their synergistic effect on methanogenic metabolism is still unknown. This study investigated the impact of polyethylene terephthalate (PET) MPs alone and co-existing with PFOS on CO2 conversion to CH4 in a thermophilic biogas upgrading system. The results showed that either PET MPs addition alone or coexisting with PFOS improved the ultimate CH4 percentage and increased CO2 utilization rate. When Fe0 was added into the reactors with PET to enhance the interspecies electron transfer, a potential defluorination was observed with a defluorination rate of 15.88 ± 1.53%. Exposure of the reactor to PFOS of 300 μg/L could change the methanogenic pathway, resulting in a newly emerged Methanomassiliicoccus with dominance of 16%. Furthermore, under the exposure of PFOS, the number of predicted genes regulating enzymes in methanogenic steps from CO2 increased. These results suggest that the co-existence of PET MPs and PFOS will not inhibit the activity of hydrotrophic methanogenes, and a portion of PFOS may be biodegraded during the methanogenesis under Fe0 regulation.

Thermal degradation evaluation of polyethylene terephthalate microplastics: Insights from kinetics and machine learning algorithms using non-isoconversional TGA data

The thermal stability of polyethylene terephthalate (PET) microplastics (MPs) is crucial in diverse applications, necessitating a comprehensive understanding of their behavior under high temperatures. This study explores the impact of chemical modifications and aging on the thermal degradation of different PET MPs, including pristine (Pr-PET), acid-modified (Mod-PET), and oxidatively aged (Ag-PET), using kinetics, thermodynamics, and machine learning algorithms using non-isoconversional thermogravimetric analysis (TGA) data. The calculated activation energy (E) and pre-exponential factor (A) for Coats-Redfern model showed Pr-PET with highest E value of 21.09 and 15.24 kJ/mol for zero-order and first order kinetic model, indicating its increased energy requirement for chemical reactions, while Mod-PET and Ag-PET demonstrated improved thermal resistance with lower E values. Thermodynamic data from kinetic parameters showed positive values of Gibbs free energy (ΔG°), enthalpy (ΔH°) and negative values of entropy (ΔS°) (−0.17. kJ/mol·K to −0.20 kJ/mol·K) indicated non-spontaneous degradation process, necessitated energy input, and reduced disorder with decreased reactivity. For accurate predictions of thermal behavior, various machine learning algorithms, including Support Vector Machine (SVM), Decision Tree Regressor (DTM), Random Forest Regressor (RFM), and Artificial Neural Network (ANN), were utilized. The RFM consistently outperformed other models, exhibiting the lowest error metrics and providing reliable predictions of weight loss and derivative thermogravimetric (DTG) for different PET MPs with a predicted-R2 of 0.999. This study sheds light on the thermal stability of PET MPs, considering chemical modifications and aging. The integration of these methods offers a powerful approach to understanding and predicting PET MPs under thermal conditions.

Polyethylene Terephthalate Hydrolases in Human Gut Microbiota and Their Implications for Human Health.

Polyethylene terephthalate (PET), primarily utilized for food and beverage packaging, consistently finds its way into the human gut, thereby exerting adverse effects on human health. PET hydrolases, critical for the degradation of PET, have been predominantly sourced from environmental microbial communities. Given the fact that the human gut harbors a vast and intricate consortium of microorganisms, inquiry into the presence of potential PET hydrolases within the human gut microbiota becomes imperative. In this investigation, we meticulously screened 22,156 homologous sequences that could potentially encode PET hydrolases using the hidden Markov model (HMM) paradigm, drawing from 4984 cultivated genomes of healthy human gut bacteria. Subsequently, we methodically validated the hydrolytic efficacy of five selected candidate PET hydrolases on both PET films and powders composed of micro-plastics (MPs). Notably, our study also unveiled the influence of both diverse PET MP powders and their resultant hydrolysates on the modulation of cytokine expression in macrophages. In summary, our research underscores the ubiquitous prevalence and considerable potential of the human gut microbiota in PET hydrolysis. Furthermore, our study significantly contributes to the holistic evaluation of the potential health hazards posed by PET MPs to human well-being.

Succession of bacterial community during electroactive methanogenic biofilm development under microplastic manipulation

Electrochemical methanogenesis is a promising and reliable process to convert waste streams into CH4, where the electroactive methanogenic biofilms play a key role. However, given that microplastics (MP) have now been spread ubiquitously in the environment, their regulation on the performance and succession of methanogenesis biofilm remains an enigma. Herein, we developed the single-chamber electrochemical methanogenic systems to investigate how poly (ethylene terephthalate) MP (PET-MP) regulates methanogenic biofilm formation and microbial metabolisms. The microbial volume of biofilm formed under MP exposure was similar to that without exposure. However, the live/dead cell ratio of the microbes in the biofilm under the PET-MP exposure decreased significantly (p < 0.05). Correspondingly, the richness and diversity of the microbial community in the presence of MP were also lower. Network analysis implied the interspecific cooperation among the microbial communities to cope with the MP stress. Meanwhile, the biofilm produced more extracellular polymer substrates during the biofilm thickening, possibly as a defense against MP invasion. At the gene level, the content of methanogenic gene mcrA was found to positively linearly correlate with the cultivation cycles, both in presence (r = 0.945, p < 0.05) or absence of PET-MP (r = 0.913, p < 0.05). The outcomes of this study could provide insights into the practical application of electrochemical methanogenesis technology to upcycle the MP-polluted biowastes and to implement the Power&Waste-to-X concept better.

The Effect of Polyethylene Terephthalate Microplastics on the Growth of Mice

Over the course of the previous decade, there has been a growing apprehension regarding the presence of MPs. This concern has arisen because of the detrimental effects of MPs pollution on the well-being of both animals and people. This study explores the potential implications of MPs contamination on mice. This study examined the impact of microplastics on mice by feeding four food experiment; P0 (pellet BR1), P1 (potato from Pujon Farm), P2 (potato mixed 300 µg PET) and P3 (potato mixed 600 µg PET). Body weight, feces weight, and left feed were observed in this study. Food contaminated with microplastics (MPs) has a negative effect on appetite, feces weight and body weight of mice. This process has an impact on reducing the weight of feces, appetite, body weight of mice and weight of mice. It is suspected that MPs in rat digestion increase concentrations of mice corticosterone metabolites and carcass fat, suggesting an adaptation of metabolism to this situation. Another impact is expected the decrease in microbial richness in the intestines of mice, causing a decrease in mice weight and body weight of mice. The high concentration of MPs entering the digestive tract can also reduce the rats’ appetite.

Bioengineering Comamonas testosteroni CNB-1: a robust whole-cell biocatalyst for efficient PET microplastic degradation

The escalating crisis of polyethylene terephthalate (PET) microplastic contamination in biological wastewater treatment systems is a pressing environmental concern. These microplastics inevitably accumulate in sewage sludge due to the absence of effective removal technologies. Addressing this urgent issue, this study introduces a novel approach using DuraPETase, a potent enzyme with enhanced PET hydrolytic activity at ambient temperatures. Remarkably, this enzyme was successfully secreted from Comamonas testosteroni CNB-1, a dominant species in the active sludge. The secreted DuraPETase showed significant hydrolytic activity toward p-NPB and PET nanoplastics. Furthermore, the CNB-1 derived whole-cell biocatalyst was able to depolymerize PET microplastics under ambient temperature, achieving a degradation efficiency of 9% within 7 days. The CNB-1-based whole biocatalysts were also capable of utilizing PET degradation intermediates, such as terephthalic acid (TPA) and ethylene glycol (EG), and bis(2-hydroxyethyl)-TPA (BHET), for growth. This indicates that it can completely mineralize PET, as opposed to merely breaking it down into smaller molecules. Hydrolase and plastic-degrading microbiota explain degradation of polyethylene terephthalate microplastics during high-temperature composting

This research aims to explore the degradation properties of polyethylene terephthalate (PET) by PET hydrolase (WCCG) in high-temperature composting and its impact on microbial communities. PET degradation, composting parameters and microbial communities were assessed in 220 L sludge composters with PET and WCCG using high-throughput sequencing. Results showed that WCCG addition led to a deceleration of the humification process and a reduction in the relative abundance of thermophilic genera. Potential PET degrading microbiota, e.g. Acinetobacter, Bacillus, were enriched in the plastisphere in the composters where PET reduced by 26 % without WCCG addition. The external introduction of the WCCG enzyme to compost predominantly instigates a chemical reaction with PET, concurently curtailing the proliferation of plastic-degrading bacteria, leading to a 35 % degradation of PET. Both the WCCG enzyme and the microbiota associated with plastic-degradation showed the potential for reducing PET, offering a novel method for mitigating pollution caused by environmental microplastics.

Dynamic tissue model in vitro and its application for assessment of microplastics-induced toxicity to air-blood barrier (ABB)

The replication of the hominine physiological environment was identified as an effectual strategy to develop the physiological model in vitro to perform the intuitionistic assessment of toxicity of contaminations. Herein, we proposed a dynamic interface strategy that accurately mimicked the blood flow and shear stress in human capillaries to subtly evaluate the physiological damages. To proof the concept, the dynamic air-blood barrier (ABB) model in vitro was developed by the dynamic interface strategy and was utilized to assess the toxicity of polyethylene terephthalate microplastics (PET-MPs). The developed dynamic ABB model was compared with the static ABB model developed by the conventional Transwell® system and the animal model, then the performance of the dynamic ABB model in evaluation of the PET-MPs induced pulmonary damage via replicating the hominine ABB. The experimental data revealed that the developed dynamic ABB model in vitro effectively mimicked the physiological structure and barrier functions of human ABB, in which more sophisticated physiological microenvironment enabled the distinguishment of the toxicities of PET-MPs in different sizes and different concentrations comparing with the static ABB model constructed on Transwell® systems. Furthermore, the consistent physiological and biochemical characters adopted dynamic ABB model could be achieved in a quick manner referring with that of the mouse model in the evaluation of the microplastics-induced pulmonary damage. The proposed dynamic interface strategy supplied a general approach to develop the hominine physiological environment in vitro and exhibited a potential to develop the ABB model in vitro to evaluate the hazards of inhaled airborne pollutants.
