Strategies for biofilm optimization of plastic-degrading microorganisms and isolating biofilm formers from plastic-contaminated environments

Abstract The perpetual disposal of plastic waste, combined with ineffective waste management strategies, has resulted in widespread environmental plastic pollution. Microbial plastic biodegradation represents an emerging solution to this problem. However, biodegradation studies tend to overlook the fundamental prerequisite of initial surface colonization via biofilm formation. This study had two independent but connected aims relating to plastic surface colonization by microorganisms: to enhance biofilm formation by known plastic degraders, with translational potential for improved plastic degradation, and to isolate microorganisms from microplastic contaminated environments with the ability to colonize plastic surfaces. Planktonic and biofilm responses to diverse carbon and energy sources were investigated over seven days, using Bacillus subtilis 168, Fusarium solani (Martius) Saccardo, Ideonella sakaiensis 201-F6, Pseudomonas putida KT2440 and Rhodococcus ruber C208. This enabled optimal conditions for biofilm formation by each strain to be determined. In parallel, environmental samples containing synthetic or natural polymeric substances (anaerobic digestate, landfill leachate, and microplastic contaminated compost) were incubated with polyethylene and polyethylene terephthalate films, to isolate microorganisms capable of colonizing their surfaces. This yielded eight bacterial isolates from three genera: Bacillus, Lysinibacillus, and Proteus. These genera contain species that have been shown to degrade plastics and other recalcitrant synthetic polymers, demonstrating the success of our approach. This study also suggests that discrete plastic types may create different ecological niches which can be exploited by unique bacterial colonizers. Our findings underscore the importance of considering plastic colonization by microbial biofilms in the context of their biodegradation.

A Novel Approach towards the Preparation of Silk-Fibroin-Modified Polyethylene Terephthalate with High Hydrophilicity and Stability

Silk fibroin (SF) has been widely used in biomedical applications for the hydrophilicity modification of high molecular polymer materials. However, the challenge remains to immobilize SF with high structure stability and strong adhesion strength between SF and the substrate. Here, we propose an effective two-step process for modifying polyethylene terephthalate (PET) with SF: dipping PET film in SF solution and subsequently carrying out plasma-assisted deposition in SF aerosol. The structure and property analysis revealed that the SF-modified PET (PET-SF) prepared using the two-step method exhibited superior structural stability and stronger adhesion strength compared to the dip-coating method and the plasma-assisted deposition method. In addition, PET-SF prepared using the two-step method resulted in a higher concentration of SF and an increased content of active groups on its surface, enhancing its hydrophilicity compared to the other two methods. Additionally, the influence of dipping time and deposition time in the two-step method was investigated. The results demonstrated that the dipping time for 6 h and the deposition time for 3 min resulted in maximum SF grafting amount with a highly stable structure. Furthermore, the PET-SF exhibited satisfactory hydrophilicity when the deposition time was more than 3 min and showed the most hydrophilicity surface at 8 min.

Flexible wafer-scale bifunctional metasurface based on nanoimprinting

Metasurfaces have demonstrated remarkable capabilities in manipulating light fields across diverse applications. However, current research tends to examine these functionalities in isolation, prompting a growing interest in integrating different functionalities within a singular metasurface device. In this paper, we propose and experimentally demonstrate a bifunctional metasurface capable of providing concealment and sensing functions simultaneously. Specifically, the proposed nanostructure effectively operates as a one-way mirror, exhibiting an average reflection rate of approximately 90% under external illumination, alongside an absorption rate of 87.9% from the opposite direction of incidence. This functionality renders it suitable for privacy-enhancing building windows. Meanwhile, this nanostructure also integrates liquid sensing capabilities boasting a sensitivity of 464 nm/RIU, which is particularly valuable for monitoring liquid-based corrosion. The experimental performance of the prepared 6-inch nanohole-patterned metasurface closely aligns with the simulations, and the utilization of flexible polyethylene terephthalate (PET) film, coupled with nanoimprint lithography technology, enables a direct and cost-effective manufacturing process that can be scaled up for widespread applications.

Ternary nano-composite wireless ammonia sensor based on Polyaniline/CuTsPc/AgNPs for food intelligent packaging

The integration of gas sensing unit and low-cost wireless communication equipment enables real-time detection of various environmental gases, which is of great significance for food safety, medical diagnosis and environmental monitoring. In this study, copper phthalocyanine powder (CuTsPc) was prepared by high temperature solid phase melting technology. Ternary ammonia sensors with exceptional sensitivity to ammonia and conductivity were successfully fabricated by growing polyaniline/copper phthalocyanine/silver nanoparticles (PANI/CuTsPc/AgNPs) composites on polyethylene terephthalate films by in-situ polymerization. The morphology and composition of the films were characterized by FTIR, XRD and SEM, respectively. The ammonia sensing performance of the films at room temperature was systematically investigated. It is found that the flexible PANI/CuTsPc/AgNPs gas sensor exhibited rapid response time (61 s), fast recovery time (19 s), low theoretical detection limit (0.234 ppm), high response value (3.6 towards 500 ppm NH3), and excellent stability at room temperature. These remarkable response characteristics can be attributed to the synergistic effect between protonic acid-doped PANI, CuTsPc derivatives with an electron-withdrawing group, and AgNPs possessing both chemical sensitisation and electron sensitisation. Finally, the sensor material was transformed into a label antenna pattern as well as sensor transducer through series and parallel antenna loop access methods to create a wireless sensor system based on near-field communication (NFC). This system can offer real-time sensing and monitoring of changes in food packaging environment by non-contact recognition way, making it highly promising for future applications in intelligent packaging.

Surface modification of polymers by 50 Hz dielectric barrier discharge (DBD) plasma produced in air at 40 Torr

This study deals with the surface modification of polymer films utilizing a custom designed cost- effective dielectric barrier discharge (DBD) plasma produced in air at reduced pressure. We comprehensively examine diverse aspects of surface modification, encompassing electrical discharge characterization, optical signal analysis, contact angle measurements, and surface morphology assessment. Our observations unveiled the presence of distinctive filamentary streamer-based micro-discharges during the DBD process, with a power consumption of approximately 5.64 watts and an electron density of 3.4 × 1011 cm−3. Optical emission spectroscopy identifies multiple emission peaks attributed to nitrogen emissions. Notably, plasma treatment substantially reduced the water contact angle and augmented surface energy on polypropylene (PP) and polyethylene terephthalate (PET) films. Surface morphology analysis illustrated an increase in surface roughness following plasma treatment. Intriguingly, the initial rapid alterations in wettability and surface morphology attained equilibrium after approximately 30 seconds of treatment. This study highlights atmospheric DBD plasma's effectiveness in customizing polymer surfaces, improving wettability and roughness, offering promising applications for enhanced adhesion and wetting.

Discovery and characterization of two novel polyethylene terephthalate hydrolases: One from a bacterium identified in human feces and one from the Streptomyces genus

Polyethylene terephthalate (PET) is widely used for various industrial applications. However, owing to its extremely slow breakdown rate, PET accumulates as plastic trash, which negatively affects the environment and human health. Here, we report two novel PET hydrolases: PpPETase from Pseudomonas paralcaligenes MRCP1333, identified in human feces, and ScPETase from Streptomyces calvus DSM 41452. These two enzymes can decompose various PET materials, including semicrystalline PET powders (Cry-PET) and low-crystallinity PET films (gf-PET). By structure-guided engineering, two variants, PpPETaseY239R/F244G/Y250G and ScPETaseA212C/T249C/N195H/N243K were obtained that decompose Cry-PET 3.1- and 1.9-fold faster than their wild-type enzymes, respectively. The co-expression of ScPETase and mono-(2-hydroxyethyl) terephthalate hydrolase from Ideonella sakaiensis (IsMHETase) resulted in 1.4-fold more degradation than the single enzyme system. This engineered strain degraded Cry-PET and gf-PET by more than 40% and 6%, respectively, after 30 d. The concentrations of terephthalic acid (TPA) in the Cry-PET and gf-PET degradation products were 37.7% and 25.6%, respectively. The discovery of these two novel PET hydrolases provides opportunities to create more powerful biocatalysts for PET biodegradation.

Development and characterization of a bacterial enzyme cascade reaction system for efficient and stable PET degradation

The widespread use of polyethylene terephthalate (PET) in various industries has led to a surge in microplastics (MPs) pollution, posing a significant threat to ecosystems and human health. To address this, we have developed a bacterial enzyme cascade reaction system (BECRS) that focuses on the efficient degradation of PET. This system harnesses the Escherichia coli (E. coli) surface to display CsgA protein, which forms curli fibers, along with the carbohydrate-binding module 3 (CBM3) and PETases, to enhance the adsorption and degradation of PET. The study demonstrated that the BECRS achieved a notable PET film degradation rate of 3437 ± 148 μg/(d*cm²), with a degradation efficiency of 21.40% for crystalline PET MPs, and the degradation products were all converted to TPA. The stability of the system was evidenced by retaining over 80% of its original activity after multiple uses and during one month of storage. Molecular dynamics simulations confirmed that the presence of CsgA did not interfere with the enzymatic activity of PETases. This BECRS represents a significant step forward in the biodegradation of PET, particularly microplastics, offering a practical and sustainable solution for environmental pollution control.

Mechanisms of permeation of helium, hydrogen, oxygen, and water vapor through silicate-based composite barrier coating layers

When considering coating of flexible films for the packaging of sensitive products, a common goal is to meet all gas barrier requirements in a single process step. One way to achieve this is to improve the barrier performance of polymeric coating layers by incorporating silicate particles. In order to tailor the gas barrier performance of the coatings, understanding the permeation mechanisms through these composite coating layers is required. In this study, polyethylene terephthalate films were coated with composite lacquers comprising montmorillonite particles and a polymer matrix. The compatibility of montmorillonite with polymer matrices of polypropylene, polyacrylate, polycarboxylic acid, and polyvinyl alcohol was tested. The permeation behavior of helium, hydrogen, oxygen, and water vapor in these coatings was investigated. For a composite coating layer comprising montmorillonite and polyvinyl alcohol at a mixing ratio of 1:1 by weight, barrier improvement factors of ▪ compared with the pure polymer coating were found for helium, hydrogen, and oxygen, respectively. It was shown that the permeability coefficients of composite coating layers decrease with increasing permeant kinetic diameter. A comparison of calculated and measured permeability values indicated that the integration of montmorillonite leads to a tortuous permeation path and changes in the free volume and crystallinity of the polymer matrix. The permeation mechanism for water vapor turned out to be completely different from that for non-polar helium, hydrogen, and oxygen and is determined by the so-called polar path effect.

Determination of Optical Properties of Commercial Thin Film Polymers Used for Solar Protection Applications

A procedure is proposed to determine the optical properties of commercial polymeric films used as solar control for building skins. The spectral variation (190-1100 nm) of the refractive index and extinction coefficient of the samples was obtained through the simulation of the experimental transmittance spectra of the samples. Polyvinyl chloride (PVC) and polyethylene terephthalate (PET) films were analyzed. Refractive index values between 1.1 and 2.5 at λ = 500 nm and extinction coefficient values of 104 cm-1 in the ultraviolet (UV) region were found. All these results show a good fit with those given in the literature for similar thin films. A new concept for the illumination and temperature control of buildings by the use of polymeric thin films as a transparent solar protection material is presented.

Photoluminescent polypropylene nanofiber-supported polyethylene terephthalate integrated with strontium aluminate phosphor

In order to develop photochromic materials with persistent afterglow emission, such as concrete and smart windows, electrospun polypropylene nanofibers (PPNF) were incorporated into polyethylene terephthalate (PET) for reinforcement. Nanoparticles of lanthanide-activated strontium aluminum oxide (NLSA) were physically embedded into polyethylene terephthalate films to produce transparent PPNF@PET smart membranes. Electrospinning was utilized to create PPNF, which were subsequently included as a roughening agent into PET membranes. The green coloration detected upon exposing the PPNF@PET film to ultraviolet light was verified by CIE Lab coordinates and photoluminescence analysis. The PPNF@PET hybrids with low quantities of NLSA were found to immediately reverse this emission activity after removing the ultraviolet source, which suggest fluorescence emission. Afterglow photoluminescence was detected at higher phosphor concentrations in PPNF@PET by slow reversibility of the light emission, which suggests glow in the dark emission. After 365 nm of excitation, a 517 nm emission band could be monitored from the PPNF@PET hybrids. The chemical structure and morphology of LSA nanoparticles and PPNF were studied, showing diameters of 80–120 nm and 75–180 nm, respectively. Different analysis methods were employed to examine the morphologies of PPNF@PET membranes. As compared to a NLSA-free polyethylene terephthalate control sample, the PPNF@PET smart membranes demonstrated improved scratch resistance. The hydrophobicity and UV resistance of the PPNF@PET smart membranes were improved with an increase in the concentration of NLSA.

Thermal Stability, Crystallization, and Melting Behaviours of Iodine Doped Semiconducting Polyethylene Terephthalate (PET) Thin Films

The influence of iodine doping on the thermal stability, degradation kinetics, and the melting and crystallization behaviors of polyethylene terephthalate (PET) films were investigated. Iodine doping was performed by immersing PET films in 0.4 M I2/l toluene. The thermal stability and degradation kinetics of PET and the semi-conducting PET/iodine complex films were studied using the thermogravimetric analysis-derivative thermogravimetric analysis (TGA-DTGA) studies and the differential scanning calorimetry (DSC). It was found that iodine incorporation accelerated the thermal degradation and weight loss through increased chain mobility owing to the imperfections formed by doping. Both pure and doped films exhibited appreciable thermal stability with the peak degradation temperature above 400 °C. DSC thermograms of the pristine PET and the PET/iodine films in the crystallization regions were recorded during the cooling scan and the significant parameters were studied. The peak crystallization temperature of PET increased from 192 to 209 °C after iodination. Doping initiated crystallization at an early stage of cooling with an increase in the rate of crystallization and smaller crystallization half time. Iodine catalyzed crystallization and acted as a nucleating agent. DSC scans were also used to study the melting characteristics of the polymer samples. The peak melting temperature was lower and the absolute crystallinity reduced for the iodine doped PET films.

Iridescent transparent passive radiative cooling enabled by cellulose nanocrystal assemblies

Recently, passive daytime radiative cooling (PDRC) materials that reflect sunlight and transmit heat through atmospheric transparent windows to outer space without any energy consumption have attracted broad interest. However, most PDRC materials have the complicated structures, white or mirror-like appearance with poor transparency. Here, we reported a transparent iridescent PDRC coating material constructed by the evaporation-induced self-assembly (EISA) of cellulose nanocrystals (CNCs). The iridescent PDRC coating not only shows high light transparency (75 %) because the right-handed circularly polarized light can transmit the unique left-handed chiral nematic liquid crystal structure, but also shows high solar reflection efficiency (95.2 %) and infrared emission efficiency (96.5 %) that endows the coating with high radiative cooling efficiency (119 W/m2). More importantly, the PDRC coating can apply on polymethyl methacrylate (PMMA), polyethylene terephthalate (PET) film and textiles, and the substrate color has no negative impact for cooling. With the excellent radiative cooling efficiency, high transparency, facile preparation process and general applicability, the CNCs-based PDRC coating opens a new pathway toward the potential application of PDRC for environmental sustainability purpose.

Preparation of robust superhydrophobic surface on PET substrate using Box-Behnken design and facile sanding method with PTFE powder

Superhydrophobic surfaces have attracted increasing interests due to their excellent features, while achieving facile preparation of superhydrophobic surface with good mechanical stability is still a challenging work. In this paper, we prepared a superhydrophobic surface by sanding polytetrafluoroethylene powder directly onto the surface of a polyethylene terephthalate (PET) film by means of a simple sanding method with sandpaper. The fabrication parameters were firstly optimized using response surface methodology. Surface morphology and chemical composition of the fabricated surface were characterized by field emission scanning electron microscopy, Fourier transform infrared and x-ray photoelectron spectroscopy. The mechanical performance of the superhydrophobic PET surfaces was evaluated by tape peeling test, and potential applications of this surface in self-cleaning and anti-icing were finally carried out. The results showed that the water contact angle up to 153.5° and sliding angle less than ∼3° on PET surface could be prepared under the optimum conditions, and its superhydrophobicity of surfaces was attributed to the synergistically effect of low surface energy and surface roughness. The fabricated superhydrophobic surfaces also exhibited good resistance to abrasion, and they have great potential for application in the fields of self-cleaning and anti-icing.

Structure motif guided mining of MHET hydrolase and development of a two-enzyme cascade for plastics depolymerization at mild temperature

The utilization of polyethylene terephthalate (PET) has caused significant and prolonged ecological repercussions. Enzymatic degradation is an environmentally friendly approach to addressing PET contamination. Hydrolysis of mono(2-hydroxyethyl) terephthalate (MHET), a competitively inhibited intermediate in PET degradation, is catalyzed by MHET degrading enzymes. Herein, we employed bioinformatic methods that combined with sequence and structural information to discover an MHET hydrolase, BurkMHETase. Enzymatic characterization showed that the enzyme was relatively stable at pH 7.5-10.0 and 30-45 ℃. The kinetic parameters kcat and Km on MHET were (24.2±0.5)/s and (1.8±0.2) μmol/L, respectively, which were similar to that of the well-known IsMHETase with higher substrate affinity. BurkMHETase coupled with PET degradation enzymes improved the degradation of PET films. Structural analysis and mutation experiments indicated that BurkMHETase may have evolved specific structural features to hydrolyze MHET. For MHET degrading enzymes, aromatic amino acids at position 495 and the synergistic interactions between active sites or distal amino acids appear to be required for MHET hydrolytic activity. Therefore, BurkMHETase may have substantial potential in a dual-enzyme PET degradation system while the bioinformatic methods can be used to broaden the scope of applicable MHETase enzymes.

Optical Anisotropy of Polyethylene Terephthalate Films Characterized by Spectral Means.

Polyethylene terephthalate (PET) films are the subject of intensive research because of great interest in using them in applications, especially in medicine. From an optical point of view, PET films with a low degree of stretching can be considered uniaxial materials, for which the determination of the linear birefringence and its dispersion is very important. Two methods were applied here for the estimation of these parameters: the ellipsometric method and the channeled spectra method. The ellipsometric method uses monochromatic radiation; therefore, the linear birefringence of the PET films is determined for a given value of the radiation wavelength. The channeled spectra method allows for the estimation of the linear birefringence and its dispersion for a large range of wavelengths in the visible spectrum. A decrease in both parameters with the increase in the wavelength was recorded. To evidence the microstructure of PET films and the conformational changes induced by elongation and to evaluate the degree of orientation, a polarized infrared spectral study in attenuated total reflection (ATR-FTIR) mode was performed. The dichroic ratio (between the absorbance measured with linearly polarized radiation parallel and orthogonal relative to the stretching direction, respectively) and the ATR absorbance ratio for the machine direction (MD) and transversal direction (TD) configurations both for the stretched and unstretched PET samples were measured.

Study of the properties of combined multilayer polymer materials for aeronautical equipment

The results of studies of changes in the physical and mechanical properties of heat-resistant polyethylene terephthalate (PET), polymide (PI) and fluoroplastic (PP) films, as well as synthetic polyethylene terephthalate (PET) and aramid fabrics in a wide range of specified exposure temperatures and temperature exposure times are presented. The most suitable heat-resistant polymer films and fabrics for use in inflatable structures of aeronautical equipment have been identified. An assessment of the operational parameters of the investigated materials showed linear character of change in properties in a given temperature and time interval of researches.

Designing Sustainable Hydrophilic Interfaces via Feature Selection from Molecular Descriptors and Time-Domain Nuclear Magnetic Resonance Relaxation Curves.

Surface modification using hydrophilic polymer coatings is a sustainable approach for preventing membrane clogging due to foulant adhesion to water treatment membranes and reducing membrane-replacement frequency. Typically, both molecular descriptors and time-domain nuclear magnetic resonance (TD-NMR) data, which reveal physicochemical properties and polymer-chain dynamics, respectively, are required to predict the properties and understand the mechanisms of hydrophilic polymer coatings. However, studies on the selection of essential components from high-dimensional data and their application to the prediction of surface properties are scarce. Therefore, we developed a method for selecting features from combined high-dimensional molecular descriptors and TD-NMR data. The molecular descriptors of the monomers present in polyethylene terephthalate films were calculated using RDKit, an open-source chemoinformatics toolkit, and TD-NMR spectroscopy was performed over a wide time range using five-pulse sequences to investigate the mobility of the polymer chains. The model that analyzed the data using the random forest algorithm, after reducing the features using gradient boosting machine-based recursive feature elimination, achieved the highest prediction accuracy. The proposed method enables the extraction of important elements from both descriptors of surface properties and can contribute to the development of new sustainable materials and material-specific informatics methodologies encompassing multiple information modalities.

Highly reliable and stretchable OLEDs based on facile patterning method: toward stretchable organic optoelectronic devices

Stretchable displays attract significant attention because of their potential applications in wearable electronics, smart textiles, and human-conformable devices. This paper introduces an electrically stable, mechanically ultra-robust, and water-resistant stretchable OLED display (SOLED) mounted on a stress-relief pillar platform. The SOLED is fabricated on a thin, transparent polyethylene terephthalate (PET) film using conventional vacuum evaporation, organic-inorganic hybrid thin film encapsulation (TFE), and a nonselective laser patterning process. This simple and efficient process yields an OLED display with exceptional stretchability, reaching up to 95% strain and outstanding durability, enduring 100,000 stretch-release cycles at 50% strain. Operational lifetime and water-resistant storage lifetime measurements confirm that the TFE provides effective protection even after the nonselective laser patterning process. A 3 × 3 array SOLED display module mounted on a stress-relief pillar platform is successfully implemented, marking the first case of water-resistant display array operation in the field of SOLEDs. This work aims to develop practical stretchable displays by offering a reliable fabrication method and device design for creating mechanically robust and adaptable displays, potentially paving the way for future advances in human-conformable electronics and other innovative applications.

Detecting ultraviolet C radiation under polyethylene terephthalate (PET) packaging by thermoluminescence analysis using commercial dosimeters

Food irradiation is a proven technology that enhances food quality and safety by removing microorganisms and extending shelf life. Ultraviolet C radiation (UVC) has recently attracted interest due to its potential to inactivate foodborne pathogens. It relies on several advantages; however, there is limited research on its efficacy and safety particularly concerning food packaging materials such as polyethylene terephthalate (PET).This study reports on the effect of UVC radiation on commercial thermoluminescence dosimeters (namely, TLD-100, TLD-200, TLD-400 and GR-200) placed under PET films with different thicknesses (0.10, 0.42, and 0.60 mm). The results indicate the potential use of these materials for the detection of UVC radiation passing through the randomly selected PET samples. Fourier transform infrared spectroscopy assesses potential structural and chemical alterations in the PET induced by UVC exposure.

Polyethylene terephthalate bottles with excellent oxygen, water vapor barrier and mechanical performances prepared by injection and blow molding

AbstractIn this work, the oxygen barrier property of polyethylene terephthalate (PET) films was enhanced by optimizing the “active” and “passive” synergistic barrier approaches, that is, integration of high barrier material polyethylene naphthalate (PEN) and oxygen scavengers along with catalyst with PET before biaxially stretching the PET blended films. When the PEN content in the PET blended films reached 50 wt%, the oxygen permeability coefficient was significantly reduced to 0.9687 cc·mil·m−2·day−1·0.1 MPa−1, showing an enhancement of 117.4 times compared to the pure PET film. Furthermore, the water vapor permeability coefficient was reduced to 8.0208 g·mil·m−2·day−1, representing a 45.5% reduction in comparison to the pristine PET film. It also maintained a higher transverse and longitudinal tensile strength (110.5 and 127.2 MPa) than PET film only with oxygen scavengers and catalyst (53.5 and 78.0 MPa). This work presents a viable and practical approach to endow PET materials with simultaneously enhanced oxygen, water barrier performance, and mechanical property.Highlights The OPC of PET film was as low as 0.9687 cc·mil·m−2·day−1·0.1 MPa−1. The “active” and “passive” barrier techniques together reduced the OPC of PET film.
