Mangrove microbiota along the urban-to-rural gradient of the Cayenne estuary (French Guiana, South America): Drivers and potential bioindicators

The microbial communities inhabiting the Atlantic-East Pacific (AEP) mangroves have been poorly studied, and mostly comprise chronically polluted mangroves. In this study, we characterized changes in the structure and diversity of microbial communities of mangroves along the urban-to-rural gradient of the Cayenne estuary (French Guiana, South America) that experience low human impact. The microbial communities were assigned into 50 phyla. Proteobacteria, Chloroflexi, Acidobacteria, Bacteroidetes, and Planctomycetes were the most abundant taxa. The environmental determinants found to significantly correlated to the microbial communities at these mangroves were granulometry, dieldrin concentration, pH, and total carbon (TC) content. Furthermore, a precise analysis of the sediment highlights the existence of three types of anthropogenic pressure among the stations: (i) organic matter (OM) enrichment due to the proximity to the city and its wastewater treatment plant, (ii) dieldrin contamination, and (iii) naphthalene contamination. These forms of weak anthropogenic pressure seemed to impact the bacterial population size and microbial assemblages. A decrease in Bathyarchaeota, “Candidatus Nitrosopumilus”, and Nitrospira genera was observed in mangroves subjected to OM enrichment. Mangroves polluted with organic contaminants were enriched in Desulfobacteraceae, Desulfarculaceae, and Acanthopleuribacteraceae (with dieldrin or polychlorobiphenyl contamination), and Chitinophagaceae and Geobacteraceae (with naphthalene contamination). These findings provide insights into the main environmental factors shaping microbial communities of mangroves in the AEP that experience low human impact and allow for the identification of several potential microbial bioindicators of weak anthropogenic pressure.

Heavy metal tolerance and polychlorinated biphenyl oxidation in bacterial communities inhabiting the Pasvik River and the Varanger Fjord area (Arctic Norway)

Heavy metals (HMs) and polychlorobiphenyls (PCBs) enter the Arctic environment through a variety of anthropogenic sources with deleterious effects towards biota and public health. Bacteria first transfer toxic compounds to higher trophic levels and, due to the tight link existing between prokaryotic community functions and the type and concentration of contaminants, they may be useful indicator of pollution events and potential toxicity to other forms of life. The occurrence and abundance of HM-tolerant and PCB-oxidizing bacteria in the sub-Arctic Pasvik river area, heavily impacted by anthropogenic modifications, was related to HM and PCB contamination. This latter more likely derived from local inputs rather than a global contamination with higher PCB and HM amounts (and higher bacterial viable counts) that were determined in inner and middle sections of the River. Finally, a panel of bacteria with potential applications in the bioremediation of cold environments were selected and phylogenetically identified.

Polychlorobiphenyl levels in the serum of cats from residential flats in Italy: Role of the indoor environment

ABSTRACTThe domestic cat (Felis catus) was used as a sentinel of exposure to polychlorobiphenyls (PCBs) in indoor urban environments y. Sera from 120 cats were pooled to form 30 different groups selected by age (<2 years; > 2 ≤ 8 years; > 8 years), sex, municipality (Bologna and Turin) and environment (indoor vs. outdoor). Test portions of 1 mL were analyzed by means of gas chromatography coupled to high-resolution mass spectrometry (GC-HRMS) for six selected indicators non-dioxin-like PCBs (∑6 PCBs: congeners #28, #52, #101, #132, #153 and #180) and the results were computed in the upper-bound mode. The internal dose of PCBs attributable to the cats' alimentary lipid intake ranged from 32.4 to 1,446 ng/g (P50 165; mean 258). The Wilcoxon test revealed significantly lower PCB burden in “outdoor” groups than in “indoor” groups. Age correlated well with the heptachlorinated and most bio-accumulative congener, PCB #180, and slightly with hexachlorinated PCBs #138 and #153. Contamination attributable to house dust collected in 15 living-rooms ranged from 10.0 to 279 ng/g dry weight (P50 97.4; mean 94.4). Exposure estimates indicated a 0.6–16 ng/kg bw range of daily ∑6 PCB intake from a default value of 200 mg/cat of dust ingestion. The intake of PCBs due to dust ingestion fell within the same order of magnitude as that computed from a 60 g daily intake of commercial dry pet foods, while inhalation accounted for 0.21–8.2 ng/kg bw/day, on setting the nominal ∑6 PCB contamination in outdoor and indoor air at 0.37 and 15 ng/m3, respectively. Italian indoor cats could be exposed to higher levels of ∑6 PCBs than the Reference Dose (RfD) of 10 ng/kg/bw/day; this supports the World Health Organization's statement that the quality of the indoor environment is a major determinant of health.

Combined approaches to determine the impact of wood fire on PCDD/F and PCB contamination of the environment: A case study

Fires might be the source of persistent organic pollutants (POPs) such as dioxins, furans (PCDD/Fs) and/or polychlorobiphenyls (PCBs) in the environment. In the perspective of defining legal responsibilities a thorough characterization of the impact of such an event should be carried out. However, such characterization is not easy as the environment integrates both local and diffuse sources of such molecules. Thus, a combined approach, which includes gathering field surveys, modeling and laboratory experiments, should be conducted. The objective of this work is to illustrate different approaches to give sufficient insight to determine the actual impact of wood fire on the environment. The work was carried out at the vicinity of a burnt down parcel. The fired material was a mixture of wood and PCB-contaminated soils as the site was a former pyralene-disposal site. Modeling, soil and lichen sampling and experimental combustion were carried out to delineate the contamination for each chemical and to define the area within the fire that was responsible for the environmental contamination. Concentrations of PCDD/F and PCBs were very high on the burnt plot. The combined approach determined that the furans were the predominant compounds in the smoke emitted by the fire. Based on this tracer, it was possible to demonstrate that in terms of environmental contamination of PCDD/F, the impact of the fire was restricted to a 2km radius from the burnt down plot. For PCBs, no specific tracer was identified. In this case, the delineation of the impact could only be empirical, based on the total concentration of the chemicals.

Interlaboratory Study of a Multiresidue Gas Chromatographic Method for Determination of Organochlorine and Pyrethroid Pesticides and Polychlorobiphenyls in Milk, Fish, Eggs, and Beef Fat

An interlaboratory study was conducted to validate a gas chromatographic (GC) method for determination of 21 organochlorine pesticides, 6 pyrethroid pesticides, and 7 polychlorobiphenyl (PCB) congeners in milk, beef fat, fish, and eggs. The method was performed at low contamination levels, which represent relevant contents in food, and is an extension of the European standard (method NF-EN-1528, Parts 1-4). It enlarges the applicable scope of the reference EN method to pyrethroid pesticides and proposes the use of solid-phase extraction (SPE) as a cleanup procedure. Cryogenic extraction was made, and SPE cleanup was performed with 2 successive SPE cartridges: C18 and Florisil. After injection of the purified extract onto a GC column, residues were measured by electron capture detection. Food samples (liquid milk, beef fat, mixed fish, and mixed eggs) were prepared, tested for homogeneity, and sent to 17 laboratories in France. Test portions were spiked with 27 pesticides and 7 PCBs at levels from 26 to 45, 4 to 27, 31 to 67, and 19 to 127 ng/g into milk, eggs, fish, and fat, respectively. Based on results for spiked samples, the relative standard deviation for repeatability ranged from 1.5 to 6.8% in milk, 3 to 39% in eggs, 4.5 to 12.2% in fish, and 7 to 13% in fat. The relative standard deviation for reproducibility ranged from 33 to 50% in milk, 29 to 59% in eggs, 31 to 57% in fish, and 30 to 62% in fat. This method showed acceptable intra- and interlaboratory precision data, as corroborated by HORRAT values at low levels of pesticide and PCB contamination. The statistical evaluation of the results was performed according to the International Organization for Standardization (ISO; ISO 3534 standard) and 5725-2 Guideline.

Polychlorobiphenyls in Antarctica

The presence and the distribution of polychlorobiphenyls (PCBs) in Antarctic environmental components and the effect of the seasonal formation/melting process of pack ice on the pollution level of seawater were investigated. Seawater, marine, and lake sediment and soil samples were collected in a large area of the Ross Sea and Victoria Land during the 1988–1989, 1989–1990, 1990–1991, and 1991–1992 Italian expeditions. The results obtained highlighted a low and quite homogeneous PCB contamination of the studied area. Surface seawater samples from Gerlache Inlet and Wood Bay showed a typical PCB concentration of 130 pg/liter, and an increase after pack ice melting of about 30–40%. Marine sediment, lake sediment, and soil samples showed normalized mean PCB contents of 150, 240, and 130 (pg/g)/(m2/cm3), respectively.

Influence of relative response factor in the determination of organic microcontaminants with isotopically labeled standards

The use of isotopically labeled internal standards in the analysis of organic microcontaminants entails the determination of relative response factors (RRFs) of the unlabeled with respect to the labeled compounds. RRFs were measured daily during 28 replicate polychlorobiphenyl (PCB) analyses on two different sets of samples. Daily RRFs allowed more precise results to be attained than those provided by mean RRFs, the latter being the mean of all the daily RRFs obtained during the same series of analyses. Analytical applications to food matrices are presented, including the PCB contamination profile of an olive oil and that found in a matrix of Italian national mean diet. Based on the latter sample, also the polycyclic aromatic hydrocarbon (PAH) daily intake of Italians is assessed. Seven PAHs are determined.

PCB Contamination in Samples from the Italian Diet, Dairy Products, and Agricultural Soil

Polychlorobiphenyls (PCBs) have been determined in samples of milk and butter from areas of Italy reflecting different environmental pollution, in one sample of a local Italian diet, and in samples of agricultural soil. PCBs were determined as single congeners and their contamination was represented as a pattern including a maximum of 48 different congeners, containing from three to seven chlorine atoms. The determination was carried out by HRGC-MS with the isotope dilution technique.

Differences in polychlorobiphenyl (PCB) contamination patterns in various environmental matrices

Isomer-specific determination of polychlorobiphenyls (PCB) was performed on various matrices, including the following: sea bottom sediment and molluscs ( Mytilusgalloprovincialis and Scapharca inaequivalvis) from the Northern Adriatic sea; cheese and the milk used in its production; human milk; Italian total diet; sludge from a wastewater treatment plant. Forty-five congeners were quantitated and the results were illustrated in the form of bar graphs, for visual comparison as PCB contamination profiles. Different matrices show different PCB contamination profiles, concerning both multiplicity and congener distribution. The importance of determining mono-ortho substituted congeners is also demonstrated.

Effects of chemical pollution on the activities of hepatic xenobiotic metabolizing enzymes in fish from the river rhône

Polychlorobiphenyl (PCBs) levels and hepatic xenobiotic metabolizing enzyme activities were measured in fish from three locations of the River Rhône to study the consequences of a constant loading of PCBs from a PCB incineration plant. Our results show that levels of PCBs and enzyme activities were higher in fish living downstream from the plant than in fish from two locations upstream, suggesting enzyme induction by PCBs (known to be potent inducers in laboratory conditions). Enzyme activities were studied in spring and autumn in three species: nase ( Chondrostoma nasus), roach ( Rutilus rutilus) and grayling ( Thymallus thymallus). Induction was observed for three cytochrome P-450-dependent monooxygenase activities (MO), i.e. 7-ethoxyresorufin O-deethylase (EROD), aryl hydrocarbon hydroxylase (AHH) and 7-ethoxycoumarin O-deethylase (ECOD). There was a close correlation between EROD and AHH activities (for all species). Glutathione S-transferase activities were also shown to be related to the PCB levels. Conversely, cytochrome P-450 content and benzphetamine N-demethylase activity were not “PCB level-dependent”. This study clearly demonstrates a close relationship between PCB contamination and MO activities in fish from the field and thus clearly emphasizes the interest in MO as a monitoring tool for estimating water quality.

Contaminants in fish from Great Lakes Harbors and Tributary mouths

Composite fish samples collected from Great Lakes Harbors and Tributaries between 1980 and 1981 were analyzed by gas chromatography and gas chromatography-mass spectrometry for a wide range of pesticides and priority pollutants. Severe polychlorobiphenyl (PCBs) contamination was observed in Sheboygan River fish with moderate to severe PCB contamination in samples from the Milwaukee, Kinnickinnic, Fox, and Ashtabula Rivers. The Ashtabula River samples also contained several chlorinated aromatic and aliphatic hydrocarbons. Polynuclear aromatic hydrocarbons were detected in fish from the Black, Kinnickinnic and Menominnee Rivers, while a number of benzene derivatives were observed in samples from the Fox and Wolf Rivers.
