Brown bullhead at the St. Lawrence River (Cornwall) Area of Concern: health and endocrine status in the context of tissue concentrations of PCBs and mercury.

The St. Lawrence River, at Cornwall Ontario, has accumulated sediment contaminants, mainly mercury (Hg) and polychlorinated biphenyls (PCBs), from industrial point sources over many years. Although those sources are past, the river at Cornwall remains an Area of Concern (AOC). Because of remediation and other changes in the AOC, improved knowledge of contaminants in wild-fish and their putative links to health effects could help decision makers to better assess the AOC's state. Thus, we compared tissue concentrations of Hg, PCBs, morphometric measures of health, and biomarkers of exposure, metabolic-, and reproductive health in native brown bullhead (Ameiurus nebulosus) from the AOC to those of upstream reference fish. Linear discriminant analysis separated the adult fish of both sexes among upstream and downstream sites without misclassification. Burdens of total-Hg (all sites) and PCB toxic equivalents (downstream sites) exceeded the guidance for the protection of wildlife consumers. There were subtle effects of site on physiological variables, particularly in female fish. Total-Hg in tissue correlated negatively to plasma testosterone and 17β-estradiol in female fish at Cornwall: moreover, concentrations of both hormones were lower within the AOC compared to reference site fish. A similar effect on vitellogenin, which was uncorrelated to E2/T at the downstream sites, indicated the potential for reproductive effects. Downstream fish also had altered thyroidal status (T3, TSH, and ratio of thyroid epithelial cell area to colloid area). Despite spatial and temporal variability of the endocrine-related responses, these subtle effects on fish health within the AOC warrant further study.

Examination of contaminant exposure and reproduction of ospreys (Pandion haliaetus) nesting in Delaware Bay and River in 2015

A study of ospreys (Pandion haliaetus) nesting in the coastal Inland Bays of Delaware, and the Delaware Bay and Delaware River in 2015 examined spatial and temporal trends in contaminant exposure, food web transfer and reproduction. Concentrations of organochlorine pesticides and metabolites, polychlorinated biphenyls (PCBs), coplanar PCB toxic equivalents, polybrominated diphenyl ethers (PBDEs) and other flame retardants in sample eggs were generally greatest in the Delaware River. Concentrations of legacy contaminants in 2015 Delaware Bay eggs were lower than values observed in the 1970s through early 2000s. Several alternative brominated flame retardants were rarely detected, with only TBPH [bis(2-ethylhexyl)-tetrabromophthalate)] present in 5 of 27 samples at <5 ng/g wet weight. No relation was found between p,p′-DDE, total PCBs or total PBDEs in eggs with egg hatching, eggs lost from nests, nestling loss, fledging and nest success. Osprey eggshell thickness recovered to pre-DDT era values, and productivity was adequate to sustain a stable population. Prey fish contaminant concentrations were generally less than those in osprey eggs, with detection frequencies and concentrations greatest in white perch (Morone americana) from Delaware River compared to the Bay. Biomagnification factors from fish to eggs for p,p′-DDE and total PCBs were generally similar to findings from several Chesapeake Bay tributaries. Overall, findings suggest that there have been improvements in Delaware Estuary waterbird habitat compared to the second half of the 20th century. This trend is in part associated with mitigation of some anthropogenic contaminant threats.

Reproductive success and contaminant associations in tree swallows (Tachycineta bicolor) used to assess a Beneficial Use Impairment in U.S. and Binational Great Lakes’ Areas of Concern

During 2010-2014, tree swallow (Tachycineta bicolor) reproductive success was monitored at 68 sites across all 5 Great Lakes, including 58 sites located within Great Lakes Areas of Concern (AOCs) and 10 non-AOCs. Sample eggs were collected from tree swallow clutches and analyzed for contaminants including polychlorinated biphenyls (PCBs), dioxins and furans, polybrominated diphenyl ethers, and 34 other organic compounds. Contaminant data were available for 360 of the clutches monitored. Markov chain multistate modeling was used to assess the importance of 5 ecological variables and 11 of the dominant contaminants in explaining the pattern of egg and nestling failure rates. Four of 5 ecological variables (Female Age, Date within season, Year, and Site) were important explanatory variables. Of the 11 contaminants, only total dioxin and furan toxic equivalents (TEQs) explained a significant amount of the egg failure probabilities. Neither total PCBs nor PCB TEQs explained the variation in egg failure rates. In a separate analysis, polycyclic aromatic hydrocarbon exposure in nestling diet, used as a proxy for female diet during egg laying, was significantly correlated with the daily probability of egg failure. The 8 sites within AOCs which had poorer reproduction when compared to 10 non-AOC sites, the measure of impaired reproduction as defined by the Great Lakes Restoration Initiative, were associated with exposure to dioxins and furan TEQs, PAHs, or depredation. Only 2 sites had poorer reproduction than the poorest performing non-AOC. Using a classic (non-modeling) approach to estimating reproductive success, 82% of nests hatched at least 1 egg, and 75% of eggs laid, excluding those collected for contaminant analyses, hatched.

Tissue Distribution of Polychlorinated Biphenyls and Organochlorine Pesticides and Potential Toxicity to Alaskan Northern Fur Seals Assessed Using PCBs Congener Specific Mode of Action Schemes

The fur seal (Callorhinus ursinus) population has decreased in their primary breeding grounds in the Bering Sea; contamination is among suspected causes. Our goal was to better understand the extent of contamination of seal tissues with certain organochlorine compounds by measuring the concentrations of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) in fur seal tissues from St. Paul Island, to gain a better perspective of tissue congener distribution and to evaluate the observed PCB levels against toxicologically significant levels for modes of action. Concentrations of 145 PCB congeners (Sigma(145)PCBs) and 12 OCPs were measured with gas chromatography-ion trap mass spectrometry in 8 different tissues of 10 male northern fur seals. The mean concentrations of SigmaOCPs [in ng/g lipid weight (lw)] were 1180 in blubber, 985 in the heart, 1007 in the liver, 817 in the kidney, 941 in muscle, 660 in reproductive tissues, 204 in the brain, and 322 in the lung. The mean concentrations of Sigma(145)PCBs (in ng/g lw) were 823 in blubber, 777 in the liver, 732 in the heart, 646 in reproductive tissues, 638 in muscle, 587 in the kidney, 128 in the lung, and 74.3 in brain tissues. Concentrations of PCBs affecting the aryl hydrocarbon receptor expressed as total PCB toxic equivalents (SigmaPCB-TEQs) ranged from 0.3 to 545 pg/g lw for the various tissues. The major contributors to SigmaPCB-TEQs are CB-118 in muscle, brain, lung, kidney, and liver, CB-126 in blubber, and CB-118 and CB-126 equally in the heart and reproductive tissues. Concentrations of PCBs affecting Ca(2+) homeostatsis expressed as the neurotoxic equivalent (NEQ) showed SigmaPCB-NEQs ranged from 17.7 to 215 ng/g lw in all tissues. Although no composite measure of perturbation of thyroid function is available, sufficient amounts of congeners with high binding to the thyroxine transport system were present to warrant consideration of this mode of action in future studies. Analyses of 145 PCBs and mode of action evaluation suggest that PCB contamination could potentially exert an effect on the Alaskan northern fur seal population although the PCB concentrations have been decreasing in the fur seals over the last decade.

Potential human health risks from metals (Hg, Cd, and Pb) and polychlorinated biphenyls (PCBs) via seafood consumption: Estimation of target hazard quotients (THQs) and toxic equivalents (TEQs)

Edible marine species (fish, cephalopod molluscs, crustaceans) from the Adriatic Sea were analyzed for content in heavy metals (Hg, Cd and Pb) and polychlorinated biphenyls (PCBs). Health risks to human via dietary intake of seafood were assessed by the target hazard quotients (THQs) and the toxic equivalent factors (TEFs). Mercury maximum concentrations corresponded to fish (0.07–1.56 μg g −1 w.w.), followed by cephalopod molluscs (0.10–0.55 μg g −1 w.w.), and crustaceans (0.27–0.33 μg g −1 w.w.). Cadmium levels in cephalopods (0.18–0.59 μg g −1 w.w.) were higher than those in fish (0.01–0.05 μg g −1 w.w.) and crustaceans (0.02–0.04 μg g −1 w.w.), while for Pb the concentrations were generally low (fish: ND–1.18 μg g −1 w.w., cephalopods: ND–0.17 μg g −1 w.w., crustaceans: ND–0.03 μg g −1 w.w.). For PCBs, concentrations in fish, cephalopods and crustaceans ranged between 141 and 3406 ng g −1 l.w., 190 and 542 ng g −1 l.w., and 202 and 429 ng g −1 l.w., respectively. Cd and Pb THQ values as well as estimates of PCB TEQ exposure indicated the absence of health risks through consumption of the various seafood. In contrast, mercury TEQs values due to consumption of certain fish species (albacore, rosefish and thornback ray) indicated that human health risk might be of concern.

Transplacental transfer of organochlorines in california sea lions (Zalophus californianus)

The transplacental transfer of organochlorines (OCs) in California sea lions (Zalophus californianus) was investigated by analyzing blubber samples from 20 female sea lions and their fetuses during the last trimester of pregnancy. A rapid, high-performance liquid chromatographic, photodiode-array method was used to measure blubber concentrations of polychlorinated biphenyls (PCBs), including dioxin-like congeners, as well as DDTs and hexachlorobenzene. Summed values of PCBs (sigmaPCBs), of DDTs (sigmaDDTs), and of PCB toxic equivalents (sigmaPCB TEQs) were calculated from these data. The ratios of mean blubber concentrations of fetal sigmaPCBs to maternal blubber concentrations of sigmaPCBs were 0.45 by wet weight and 0.97 by lipid weight, but these ratios varied widely among mother-fetus pairs. Mean ratios of fetal sigmaDDTs to maternal sigmaDDTs were 0.53 by wet weight and 1.12 by lipid weight. Fetuses were classified into two age groups, based on date of recovery, to examine differences in OC transfer because of gestational age. Fetal to maternal ratios for individual PCB congeners, DDT compounds, and sigmaPCBs, sigmaDDTs, and sigmaPCB TEQs were lower among premature compared with late-term fetuses. These ratios increased for both groups as the logarithmic n-octanol/water partition coefficient (log Kow) for each compound decreased. Linear predictions for sigmaPCB and sigmaDDT concentrations in fetal blubber could be obtained using the sigmaPCB and sigmaDDT concentrations in maternal blubber, maternal and fetal blubber lipid content, maternal mass, and maternal age. Fetal TEQ was explained by maternal TEQ and maternal age. The ability to predict contaminant concentrations in fetal blubber from maternal parameters is important for developing risk assessment models for marine mammals.

Relationship of serum levels of individual PCB, dioxin, and furan congeners and DDE with Great Lakes sport-caught fish consumption

Great Lakes (GL) sport fish consumption is a potential route of exposure for environmentally persistent organochlorine contaminants, which may have human health effects. In this report, relationships are explored among individual congeners in a large cohort of frequent and infrequent GL sport fish consumers. Blood samples were obtained in 1993–1995 and analyzed for 1,1-bis(4-chlorophenyl)-2,2-dichloroethene (DDE) and for 62 noncoplanar polychlorinated biphenyls (PCBs), 4 coplanar PCBs, 8 polychlorinated dibenzo- p-dioxin (dioxin), and10 dibenzofuran (furan) congeners. All GL fish eaters and referents had detectable levels of DDE, total noncoplanar PCBs, total coplanar PCBs, total dioxins, and total furans. Noncoplanar PCBs were higher in GL sport fish consumers than in a referent population from the same geographic area, were associated with GL sport-caught fish (GLSCF) consumption, and varied significantly by Great Lake. DDE, lower chlorinated dioxin and furan toxic equivalents (TEQs), and coplanar PCB TEQs were positively associated with noncoplanar PCBs but were not associated with GL sport fish consumption independent of PCB level. Highly chlorinated dioxin and furan congener TEQs were not significantly associated with noncoplanar PCBs or GL sport fish consumption, suggesting that participants were acquiring some of these TEQs from a source other than GLSCF. In epidemiologic studies, it may be important to include populations with higher organochlorine exposures as well as background exposures and to consider the effects of individual congeners or mixtures of congeners on health outcomes.

PCBs and DDT in the serum of juvenile California sea lions: associations with vitamins A and E and thyroid hormones

Top-trophic predators like California sea lions bioaccumulate high levels of persistent fat-soluble pollutants that may provoke physiological impairments such as endocrine or vitamins A and E disruption. We measured circulating levels of polychlorinated biphenyls (PCBs) and dichlorodiphenyltrichloroethane (DDT) in 12 healthy juvenile California sea lions captured on Año Nuevo Island, California, in 2002. We investigated the relationship between the contamination by PCBs and DDT and the circulating levels of vitamins A and E and thyroid hormones (thyroxine, T4 and triiodothyronine, T3). Serum concentrations of total PCBs (∑PCBs) and total DDT were 14 ± 9 mg/kg and 28 ± 19 mg/kg lipid weight, respectively. PCB toxic equivalents (∑PCB TEQs) were 320 ± 170 ng/kg lipid weight. Concentrations of ∑PCBs and ∑PCB TEQs in serum lipids were negatively correlated ( p < 0.05) with serum vitamin A and T3, potentially reflecting PCB-related toxicity. A slight but not significant negative correlation ( p < 0.1) was observed between serum T4 and the levels of ∑PCBs and ∑PCB TEQs. Conversely, no relationship was evident between the contaminant concentrations and vitamin E ( p > 0.1). As juvenile California sea lions are useful sentinels of coastal contamination, the high levels encountered in their serum is cause for concern about the ecosystem health of the area.

Exposure and effects of chemical contaminants on tree swallows nesting along the Housatonic River, Berkshire County, Massachusetts, USA, 1998–2000

Hatching success of tree swallows (Tachycineta bicolor) was assessed for three years in relation to chemical contamination along the Housatonic River, Berkshire County (MA, USA), in 1998, 1999, and 2000. Nest boxes were erected at five sites along the Housatonic River and its tributaries and at one reference location. Concentrations of total polychlorinated biphenyls (PCBs) were some of the highest ever reported in bird eggs. Mean concentrations at sites along the Housatonic River ranged between 32 and 101 microg/g wet weight. A significant negative relationship was observed between concentrations of total PCBs in clutches and hatching success. A significant negative relationship was also observed between hatching success and the sum of the total dioxins and furans and the associated toxic equivalents (TEQs) for dioxins and furans. In a combined model with PCB TEQs and dioxin/furan TEQs, PCB TEQs were not significantly correlated to hatching success, whereas dioxin/furan TEQs were. Contamination of tree swallows was from local food sources. Accumulation rates of total PCBs in 12-d-old nestlings averaged between 34 and 76 microg/d at the sites along the main stem of the Housatonic River compared to <1 microg/d at the reference location.

Body Burden Levels of Dioxin, Furans, and PCBs among Frequent Consumers of Great Lakes Sport Fish

Dioxins, furans, and polychlorinated biphenyls (PCBs) are toxic, persist in the environment, and bioaccumulate to concentrations that can be harmful to humans. Sport anglers may be exposed to these residues via consumption of contaminated Great Lakes (GL) fish. The Health Departments of five GL states, Wisconsin, Michigan, Ohio, Illinois, and Indiana, formed a consortium to study body burden levels of chemical residues in fish consumers of Lakes Michigan, Huron, and Erie. In Fall 1993, a telephone survey was administered to sport angler households to obtain fish consumption habits and demographics. A blood sample was obtained from a portion of the study subjects. One hundred serum samples were analyzed for 8 dioxin, 10 furan, and 4 coplanar PCB congeners. Multiple linear regression was conducted to assess the predictability of the following covariates: GL sport fish species, age, BMI, gender, years sport fish consumed, and lake. Of the 100 subjects, there were 58 men; 35 consumed sport fish from Lake Michigan, 29 from Lake Huron, and 36 from Lake Erie. The overall average number of GL sport fish meals consumed in the previous 12 months was 43. Lake Erie male and female consumers, on average, ate more GL sport fish, a mean of 57 and 42 meals, respectively, than men and women from the other two lake subgroups. Median total dioxin toxic equivalents (TEq), total furan TEq, and total coplanar PCB TEq were higher among all men than all women (P=0.0001). Lake trout, salmon, age, BMI, and gender were significant regression predictors of log(total coplanar PCBs). Lake trout, age, gender, and lake were significant regression predictors of log(total furans). Age was the only significant predictor of total dioxin levels.
