The association of exposure to polychlorinated biphenyls with lipid profile and liver enzymes in umbilical cord blood samples

Evidence on prenatal exposure to polychlorinated biphenyls (PCBs) and its effects on newborns and potential biological mechanisms is not well defined yet. Therefore, this study aimed to examine whether PCBs are associated with lipid profile and non-invasive markers of hepatocyte injuries in samples of blood obtained from the umbilical cord. This study included 450 mothers-newborn pairs. Umbilical levels of PCBs were measured using Gas Chromatography/Mass Spectrophotometry (GC/MS). Lipid profile including low-density lipoprotein (LDL-C), total cholesterol (TC), triglycerides (TG), and high-density lipoprotein (HDL-C), as well as liver enzymes i.e., alanine amino transferase (ALT), aspartate amino transferase (AST), γ-glutamyl-transferase (GGT) and alkaline phosphatase (ALP) were determined from umbilical cord blood samples. Quantile g-computation analysis was applied to evaluate the collective influence of PCBs on both lipid profiles and liver enzymes, along with the impact of lipid profiles on liver enzymes. Exposure to the mixture of PCBs was significantly associated with increases in ALP, AST, ALT, and GGT levels in cord blood samples, with increments of 90.38 U/L (95%CI: 65.08, 115.70, p < 0.01), 11.88 U/L (95%CI: 9.03, 14.74, p < 0.01), 2.19 U/L (95%CI:1.43, 2.94, p < 0.01), and 50.67 U/L (95%CI: 36.32, 65.03, p < 0.01), respectively. Additionally, combined PCBs exposure was correlated with significant increases in umbilical TG, TC, and LDL-C levels, with values of 3.97 mg/dL (95%CI: 0.86, 7.09, p = 0.01), 6.30 mg/dL (95%CI: 2.98, 9.61, p < 0.01), and 4.63 mg/dL (95%CI: 2.04, 7.23, p < 0.01) respectively. Exposure to the mixture of lipids was linked to elevated levels of AST and GGT in umbilical cord blood samples. Furthermore, a noteworthy mediating role of TC and LDL-C was observed in the association between total PCBs exposure and umbilical cord blood liver enzyme levels. Overall our findings suggested that higher levels of umbilical cord blood PCBs and lipid profile could affect liver function in newborns.

Using Plasma Vitellogenin in Loggerhead Sea Turtles to Assess Reproductive Maturation and Estrogen-Like Contaminant Exposure.

Vitellogenin (VTG), an egg yolk precursor, is abnormally produced by male and juvenile oviparous species after exposure to estrogens. Plasma VTG in loggerhead sea turtles (Caretta caretta) helped us understand their reproductive maturation and investigate it as a biomarker of contaminant exposure. The presence of VTG was screened in plasma from 404 loggerheads from the northwestern Atlantic Ocean using a freshwater turtle antibody in western blots. The concentrations of VTG were semiquantified using band intensities calibrated to results from a loggerhead antibody enzyme-linked immunoassay. The detection and concentrations of VTG were in (from highest to lowest): nesting females, in-water adult females, subadult females, smaller females, unknown sex, and males. Loggerheads from this region begin vitellogenesis at ≅77 cm straight carapace length. We classified VTG expression as abnormal in nine male or juvenile turtles. Organochlorine contaminant (OC) concentrations were measured in blood and/or fat biopsies of some turtles. One abnormal VTG female had the second highest fat polychlorinated biphenyl (PCB) and 4,4'-dichlorodiphenyldichloroethylene concentrations compared among 43 VTG-negative juveniles. The nine VTG-abnormal turtles had average blood PCB concentrations 8.5% higher, but not significantly different, than 46 VTG-negative juveniles (p = 0.453). In turtles less than 77 cm, blood PCB concentrations were significantly, but weakly, correlated with semiquantified VTG concentrations (tau = 0.1, p = 0.004). Greater blood OC concentrations were found in adult females than in males, which motivated the creation of a conceptual model of OC, VTG, and hormone concentrations across a reproductive cycle. A decision tree is also provided incorporating VTG as a sexing tool. Abnormal VTG expression cannot conclusively be linked to endocrine disruption caused by these OC concentrations. Exposure levels of PCDD/Fs and PCBs in human blood and the transplacental transfer characteristics in cord blood of newborns near the industrialized area.

Mono-to octa-chlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) and polychlorinated biphenyls (PCBs) in pooled blood from the general population living near a typical industrialized area were investigated. Less chlorinated PCDD/Fs (mean: 2602pgL-1) were 7.5 times those of highly chlorinated ones (mean: 349pgL-1). The average ΣPCBs and Σdl-PCBs concentrations in human (cord) blood were 2741 (117) and 18 (0.31) ng L-1, respectively. Higher concentrations of highly chlorinated PCDD/Fs were found in females than in males across different ages. The mean concentrations (and toxic equivalents (TEQs)) of PCDD/Fs were 282 (27) pg L-1 in males and 312 (32) pg L-1 in females. The concentrations of the PCDD/Fs and PCBs increased with age for both males and females, which might be caused by the long half-lives of these compounds and decreases in metabolic rates with age as the metabolic of nutrients, food, and also PCDD/Fs and PCBs would trend to slow. The TEQ of total PCDD/Fs and PCBs was higher in blood from orthopedics patients (107pgL-1) than other patients. This result may be associated with the bone density and pollutant bioaccumulation. In addition, total concentration of PCDD/Fs and PCBs in blood of women at reproductive age were 6.6 and 37 times the cord blood of newborns, respectively. Positive correlation of PCDD/Fs and PCBs especially for the higher chlorinated compounds between female and cord blood were discovered, which might be caused by the transplacental transfer characteristics and blood barrier for macromolecules and reduce the chemical exposure risks for newborns.

Polychlorinated Biphenyls (PCBs) in the Environment: Occupational and Exposure Events, Effects on Human Health and Fertility

In the last decade or so, polychlorinated biphenyls (PCBs) garnered renewed attention in the scientific community due to new evidence pointing at their continued presence in the environment and workplaces and the potential human risks related to their presence. PCBs move from the environment to humans through different routes; the dominant pathway is the ingestion of contaminated foods (fish, seafood and dairy products), followed by inhalation (both indoor and outdoor air), and, to a lesser extent, dust ingestion and dermal contact. Numerous studies reported the environmental and occupational exposure to these pollutants, deriving from building materials (flame-retardants, plasticizers, paints, caulking compounds, sealants, fluorescent light ballasts, etc.) and electrical equipment. The highest PCBs contaminations were detected in e-waste recycling sites, suggesting the need for the implementation of remediation strategies of such polluted areas to safeguard the health of workers and local populations. Furthermore, a significant correlation between PCB exposure and increased blood PCB concentrations was observed in people working in PCB-contaminated workplaces. Several epidemiological studies suggest that environmental and occupational exposure to high concentrations of PCBs is associated with different health outcomes, such as neuropsychological and neurobehavioral deficits, dementia, immune system dysfunctions, cardiovascular diseases and cancer. In addition, recent studies indicate that PCBs bioaccumulation can reduce fertility, with harmful effects on the reproductive system that can be passed to offspring. In the near future, further studies are needed to assess the real effects of PCBs exposure at low concentrations for prolonged exposure in workplaces and specific indoor environments.

Burden levels of polychlorinated dibenzo-p-dioxins, dibenzofurans, and polychlorinated biphenyls in typical edible meat animals

The concentrations and distributions of polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), and polychlorinated biphenyls (PCBs) in the whole blood and meat of eight typical edible animals (chicken, donkey, horse, cattle, rabbit, sheep, duck, and pig) were illustrated. Total concentrations of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) and PCBs (on a basis of liquid volume) in animal bloods were 142-484 pg/L and 46-62 ng/L, respectively. Total concentrations of PCDD/Fs and PCBs (on a basis of dry weight (dw) and lipid weight (lw)) in animal meat samples were 0.47-1090 pg/g dw (0.47-4513 pg/g lw) and 7.2-23 ng/g dw (10-776 ng/g lw), respectively. TEQs for both PCDD/Fs and PCBs in animal blood and meat samples were (67 ± 27) pg/L and (5.3 ± 14) pg/g dw (24 ± 56 pg/g lw), respectively. Besides, the dietary intakes of PCDD/Fs and PCBs were also estimated. Chicken and pig contributed more TEQs than other animals. Chicken contributed the most (95%) with high toxicity, followed by pig (3.4%) with high consumption. The dietary intake of chicken might pose risks to consumers who prefer to eat chicken products, who should comprehensively consider the essential nutrients and contaminants in food during dietary intake.

Accumulation and decontamination kinetics of PCBs and PCDD/Fs from grass silage and soil in a transgenerational cow-calf setting

Polychlorinated biphenyls (PCBs) and dibenzo-p-dioxins/dibenzofurans (PCDD/Fs) are bioaccumulative pollutants that endanger bovine food safety. Bioaccumulation depends, among others, on the physiological dynamics of the cow's reproductive cycle. However, recent studies have focused only on near steady-state situations. Thus, the effects of animal physiology on PCB + PCDD/F transfer from grass silage and soil to cows' blood, adipose tissue, and milk and subsequently to suckling calves during gestation and lactation were investigated. In the exposed group, nine cows ate a grass silage/contaminated soil mixture (6.6 ± 0.8 μg iPCBs and 2.6 ± 0.4 ng dlPCB + PCDD/F TEQ kgDM−1) for 109 days prepartum until 288 days in milk (DIM). Four of these cows underwent decontamination after DIM164, receiving the same clean grass silage as the four control cows during the experiment. Calves were fed the milk of their respective mothers. In the exposed group, transgenerational bioaccumulation occurred until DIM164, with calf blood and adipose tissue PCB + PCDD/F concentrations reaching levels twice as high as those in their respective mothers. Transfer rates from oral intake to milk ranged from 0.1 up to 42%, depending on pollutant congener, dietary treatment, and reproductive parity of the cow. Congener and parity also influenced the decontamination half-lives of milk. In decontaminated calves, declines in adipose tissue PCB + PCDD/F concentrations coincided with increases in body fat mass. Therefore, it is essential to know the physiological characteristics of cattle, exposure dose and duration, and physicochemical compound properties to perform reliable transfer assessments.

PCBs in indoor air and human blood in Pittsfield, Massachusetts

Concentrations of polychlorinated biphenyls (PCBs) and three chlorinated pesticides were determined in serum from 21 residents of Pittsfield, MA and in the basement, living room and outdoor air of the 10 homes in which they lived. Median serum PCB levels were 4.2 ng/g, which are at least four times the average level in the US population, and consisted primarily of more highly chlorinated, persistent congeners. This reflects contamination with PCBs coming from the local General Electric facility. Median basement air concentration was 20.3 ng/m3, while the median living room air was 11.4 ng/m3 and median outdoor air concentration was 3.0 ng/m3. The PCB congeners detected in air were primarily low chlorinated (four and fewer) congeners, reflecting the greater volatility of PCBs with fewer chlorines. The congener pattern between basement and living room air showed a 95% correlation, while correlation with outdoor air was much less. While the congener pattern in air is very different from that of the PCB products used in Pittsfield (Aroclors 1254 and 1260), low chlorinated PCBs are detected in the vapor phase after air is blown across the commercial mixtures. The human serum samples did not show detectible levels of many of the congeners seen in the basement air samples, reflecting rapid metabolism of lower chlorinated PCBs by the human body. However, with continuous inhalation of indoor air, especially in the living room, the exposure to these non-persistent congeners may still have adverse health effects. Cellular studies of some of these non-persistent, low chlorinated congeners indicate that they are neurotoxic, mutagenic and cytotoxic. These results demonstrate the importance of consideration of inhalation of PCBs as a route of exposure, especially in indoor sites, and suggest that monitoring serum PCB concentration may not always provide a good measurement of exposure, especially to congeners that are relatively rapidly metabolized but have significant toxicity.

Inhalation Exposure to PCB from Contaminated Indoor Air—How Much Is Absorbed into the Blood?

Polychlorinated biphenyls (PCBs) were used in many construction products until their banning in the 1970s and 1980s. Nonetheless, exposure to PCBs from contaminated indoor air is still an important public health issue. The aim of our study was to estimate the contribution of PCB congeners in indoor air to the levels of PCBs in human blood. We analyzed all 209 PCB congeners in the blood of 35 individuals exposed to PCBs from contaminated indoor air. For each individual, we measured the six marker indicators PCB28, PCB52, PCB101, PCB138, PCB153 and PCB180 in indoor air at the workplace. Statistically significant correlations between PCB-contaminated indoor air and the existence of the sum of mono-, di-, tri-, tetra- and pentachlorinated biphenyls (∑PCB1–127) in the blood of the exposed individuals were found. We quantified the proportions of PCBs that are absorbed into the blood via inhalation of contaminated indoor air. Inhalation of PCBs from contaminated indoor air, especially in children, adolescents and younger adults, may lead to PCB blood burdens that are higher than general PCB background levels or in approximately the same range.

Altered functional activations of prefrontal brain areas during emotional processing of fear in Inuit adolescents exposed to environmental contaminants

Exposure to mercury, lead and polychlorinated biphenyls (PCBs) have been associated with emotional dysregulation, but their neuronal correlates have yet to be examined. Inuit from Nunavik (Northern Quebec, Canada) face internalizing problems and are among the most exposed individuals to these environmental contaminants in the world. The aim of this study was to examine the link between pre- and postnatal exposure to these contaminants and brain fear-circuitry in Inuit adolescents. Facial expression stimuli were presented to participants (mean age = 18.3 years) in a magnetic resonance imaging (MRI) scanner. Fear conditioning and extinction tasks included neutral faces as the conditioned threat and safety cues and a fearful face paired with a shrieking scream as the unconditioned stimulus. Functional MRI data were gathered at the conditioning phase (n = 71) and at the extinction phase (n = 62). Mercury, lead and PCB 153 concentrations were measured in blood samples at birth (cord blood) and at the time of the adolescent testing to estimate pre- and postnatal exposure, respectively. For each time point, exposures were categorized in tertiles (low, moderate and high exposed groups). Mixed analyses of variance were conducted for each contaminant of interest controlling for sex, age, socioeconomic status, drug/alcohol use, food insecurity and contaminant co-exposure. Results revealed greater differential activation during the conditioning phase in the right orbitofrontal cortex in participants with moderate and high concentrations of cord blood PCB 153 compared to those in the low exposure group. During the extinction phase, the high prenatal mercury exposed group showed a lower differential activation in the right and left anterior cingulate cortex compared to those in the low-exposed group; whereas there was a higher differential activation in right dorsolateral prefrontal cortex in the high postnatal lead exposed group compared to the moderate- and low-exposed groups. Our study is the first to show alterations in the prefrontal brain areas in fear conditioning and extinction tasks in relation to environmental contaminant exposures. The observed brain correlates may advance our understanding of the emotional problems associated with environmental chemical toxicity.

PCB in serum and hand wipes from exposed residents living in contaminated high-rise apartment buildings and a reference group.

Exposure to polychlorinated biphenyls (PCB) in buildings constructed with PCB-containing materials can lead to increased body burden of PCB for building users. Exposure to PCB from building related sources was assessed by measuring PCB in serum and hand wipes in two groups of tenants living in the same building estate in apartments constructed with and without PCB. The median serum levels of the sum of 19 PCB congeners was 777ng/g lw (5-95th percentile: 219-2576ng/g lw) for the exposed group and 282ng/g lw (5-95th percentile: 49.8-797ng/g lw) for the reference group. The congener pattern in serum of the exposed was shifted towards lower chlorinated congeners and was dominated by tri- and tetrachlorinated congeners. The largest difference in serum levels between the groups was observed for PCB-28, with median levels of the exposed being 70 times higher than in the reference group. For very persistent lower chlorinated congeners, like PCB-28, 66 and 74, an increase with residence times was observed as a result of accumulation over time. Less persistent congeners such as PCB-44 and 70 were also elevated in the exposed group -but independent of residence time. The less persistent congeners can therefore be used as markers of recent exposure to original PCB sources. The hand wipes also showed a large exposure contrast, e.g. PCB-28 being more than 60 times higher in the exposed group compared to the reference group (medians 14.4 and 0.23 ng/wipe, respectively) and no overlap between the groups. All measured di- to pentaPCB congeners were significantly higher in hand wipes from the exposed group compared with the reference group. Thus hand wipes seem to be a good, non-invasive screening tool for ongoing indoor exposure to PCB. Furthermore, the measured level of PCB on hand wipes was significantly correlated to PCB in blood for almost all congeners. In conclusion, PCB exposure in contaminated buildings can contribute significantly to the exposure and total body burden of PCB and the lower chlorinated congeners can make up the majority of the total PCB body burden.

Biomonitoring of mercury and persistent organic pollutants in Michigan urban anglers and association with fish consumption.

The 32-mile Detroit River and surrounding tributaries have been designated as a Great Lakes Area of Concern due to pollution from decades of municipal and industrial discharges, sewer overflows and urban development. Key pollutants in fish samples from the Detroit River include mercury, polychlorinated biphenyls (PCBs), dichlorodiphenyldichloroethylene (DDE), dioxins and furans. A biomonitoring study was conducted to assess exposures to these persistent toxic substances in Detroit urban shoreline anglers who may be at high exposure risk due to consumption of locally caught fish. Using a modified venue-based sampling approach, 287 adult shoreline anglers along the Detroit River were recruited and participated in the program. Study participants provided blood and urine specimens and completed a questionnaire following informed consent. We examined percentile estimates for total blood mercury, PCBs, DDE, and dioxin-like total toxic equivalency (TEQ) concentrations among study participants. Multiple linear regression was used to identify important predictors of contaminant concentrations. Participants consumed a median of 64 Detroit River caught fish meals in the past year. The Detroit urban anglers' median total blood mercury concentrations was 3.2 times higher than that for the general adult U.S. population. PCB concentrations among the Detroit anglers aged 18-39 years were higher than the U.S. population of the same race/ethnicity. Elevated levels of DDE and total TEQ concentrations were not observed in the cohort. Eating more locally caught fish was associated with higher total blood mercury and serum PCB concentrations. The biomonitoring data served to inform public health officials and help guide environmental public health actions to reduce harmful exposures.

Effect of dioxin-related compounds on oral pigmentation in patients affected by the Yusho incident

Background/aimToxins such as polychlorinated biphenyls (PCBs) and dioxins dramatically affect patients even decades after exposure. Patients with Yusho, a condition caused by exposure to PCBs and dioxins, have diverse mental and physical complaints, even though it is almost 50 years since the Yusho incident. Oral pigmentation is one of the major symptoms in Yusho patients. Patients and methodsA total of 183 participants in the Yusho health study were examined. Oral examinations, including recording the prevalence of oral pigmentation, were performed by two oral surgeons. Demographic and clinical characteristics, including blood concentration of PCB and 2,3,4,7,8-pentachlorodibezofuran (2,3,4,7,8-PeCDF), which are the major causes of Yusho, were obtained from the results of recent surveys conducted by the Yusho Study Group. ResultsThe mean serum PCB and 2,3,4,7,8-PeCDF levels of the 183 Yusho patients were 1.59 ± 1.25 ppb and 29.0 ± 42.9 pg/g lipid, respectively. There was a significant correlation between the levels of PCB and 2,3,4,7,8-PeCDF (r = 0.64, p < 0.01). The rate of oral pigmentation in Yusho patients (25.7%) was significant higher than among potential victims of Yusho (13 of 183, 7.1%) (p < 0.05). ConclusionThe prevalence of oral pigmentation was still significantly higher in Yusho patients, even 50 years after exposure, although blood PCB levels have decreased in that time.

Quantile regression was used to investigate whether eating locally caught fish had a differential effect at various quantiles of each contaminant. Participants reported eating an average of 20 Detroit River caught fish meals in the past year. The Detroit urban anglers&amp;#8217; total blood mercury and PCB concentrations were 2 to 3 times higher than the general U.S. population. Elevated levels of DDE and total TEQ concentrations were not observed in the cohort. Eating more locally caught fish was associated with higher total blood mercury and serum PCB concentrations, and this effect had a larger impact at higher quantiles of these biomarkers. The biomonitoring data served to inform public health officials and guide state and local public health actions to reduce harmful exposures.

Dietary patterns related to exposure to persistent organic pollutants based on the Ewha Birth and Growth Cohort

Persistent organic pollutants (POPs) are endocrine disruptors that are typically ingested in food. Using data from the Ewha Birth and Growth Cohort study, we identified dietary patterns related to blood persistent organic pollutant (POP) levels in children. In total, 188 Korean children (95 boys and 93 girls) aged 7–9 years were included in this study. Daily food intake data derived from a semi-quantitative food-frequency questionnaire were grouped into 33 food groups. Dietary patterns were analyzed using the reduced rank regression method, with blood POP levels as dependent variables.The marker polychlorinated biphenyl (PCB) pattern showed more weighted loading factors in the intake of shellfish/salted seafood, cheese, nuts and seeds, and total PCBs showed similar results. The dioxin-like PCB pattern was characterized by a higher intake of yogurt, beverages, and fruit and a lower intake of grain, seaweeds, and processed meat. Additionally, the total organochlorine pesticide (OCP) pattern showed positive factor loading values in shrimp and beverages, and negative factor loading values in seaweeds and processed meat. Each dietary pattern accounted for 25.0% of total OCPs, 22.9% of dioxin-like PCBs, 21.6% of marker PCBs, and 21.3% of total PCBs in blood. Dietary pattern scores were linearly related to blood POP levels, and this association remained after adjusting for sex, age, follow-up survey year, maternal educational level, body mass index, and total energy. Dietary patterns differed slightly among the individual POPs. Further studies are needed to develop an intervention strategy for children.

The role of heavy metals and polychlorinated biphenyls (PCBs) in the oncogenesis of head and neck tumors and thyroid diseases: a pilot study

Previous literature has highlighted the mechanisms of molecular toxicity induced by substances such as arsenic, cadmium, chromium, nickel, lead, barium and PCBs. The research was carried out on 20 volunteers, all the patients gave their consent to the research: the aim of the study was to evaluate the presence of metals and PCBs in these different matrices (blood and hair), correlating the biochemical data to pathological conditions present, and also to the area in which patients resided. Various quantitative determinations were carried out on samples of blood and hair for 14 heavy metals and on blood samples for 12 PCBs. For the 11 patients the results indicated that blood levels for half of the 14 displayed heavy metals measured considerably higher compared to the reference values, whilst the levels measured in hair evidenced some positive values significantly higher than the maximum reference. Of the 12 PCBs assayed in blood some showed higher positive values compared to the maximum tabular reference (although there is no clear reference quantified in the WHO-2005 report). In the 9 healthy patients heavy metals in the blood were within the expected target range, with those showing positive results (≤ 3 out of 14 heavy metals for each patient) having values only slightly higher than the reference maximum. The levels of 14 heavy metals measured in hair were below thresholds, and levels for the 12 PCBs measured in blood showed negativity or positivity with values close to the minimum benchmarks. The analyses carried out on biological matrices have uncovered important and significant differences between healthy and unhealthy subjects, both qualitative and quantitative differences with respect to heavy metals and PCBs. All patients with head and neck cancer enlisted for the study had heavy metal and PCB blood levels at least twice the maximum reference level. The levels of heavy metals in hair were at least double the maximum reference. In contrast, all healthy volunteers enrolled showed no significant levels for either metals or PCBs.

Fetal exposure markers of dioxins and dioxin-like PCBs

Fetal exposure to polychlorinated biphenyls (PCBs), polychlorinated-p-dibenzodioxins (PCDDs), and polychlorinated dibenzofurans (PCDFs) have been associated with a number of adverse health outcomes. Although the placenta acts as a barrier between the mother and the fetus, these contaminants transfer through the placenta exposing the fetus. Several studies have investigated placental transfer, but few have assessed the co-variation among these contaminants. Maternal blood, cord blood, and cord tissue were collected from 41 Japanese mother-infant pairs and analyzed for dioxin-like PCBs and PCDD/Fs. Hierarchical cluster analysis followed by principal component analysis were used to assess the co-variation. Two stable clusters of dioxin-like PCBs were found in maternal and cord blood. One cluster of low/medium chlorinated dioxin-like PCBs was present in all three matrices with 2,3’,4,4’,5-PeCB(#118) and 3,3’,4,4’,5-PeCB(#126) explaining the majority of the clusters’ variances. Medium/high chlorinated dioxin-like PCBs clustered in maternal blood and cord blood but not in cord tissue. 2,3,4,4’,5-PeCB(#114) and 2,3,3’,4,4’,5,5’-HpCB(#189) explained the majority of the clusters’ variances. There was a substantial correlation between the sum of dioxin-like PCBs and total PCDD/F in all three matrices. The sum of the four suggested PCBs plus 3,3’,4,4’-TeCB(#77) correlated well with total PCDD/F in all three matrices. Apart from the dioxin-like PCBs, little co-variation existed among the studied contaminants. The five PCBs can be used as fetal exposure markers for dioxin and dioxin-like PCBs in maternal and cord blood respectively. In cord tissue, more higher chlorinated dioxin-like PCBs need to be measured as well.

Gender-specific association of exposure to non-dioxin-like polychlorinated biphenyls during pregnancy with methylation levels of H19 and long interspersed nuclear element-1 in cord blood in the Hokkaido study.

Associations between prenatal exposure to polychlorinated biphenyls (PCBs) and reduced birth-size, and between DNA methylation of insulin-like growth factor-2 (IGF-2), H19 locus, and long interspersed nuclear element-1 (LINE-1) and reduced birth-size are well established. To date, however, studies on the associations between prenatal exposure to PCBs and alterations in methylation of IGF-2, H19, and LINE-1 are lacking. Thus, in this study, we examined these associations with infant-gender stratification. We performed a prospective birth cohort study using the Sapporo cohort from the previously described Hokkaido Birth Cohort Study on Environment and Children's Health conducted between 2002 and 2005 in Japan. In the final 169 study participants included in this study, we measured the concentrations of various non-dioxin-like PCBs in maternal blood during pregnancy using high-resolution gas chromatography/high-resolution mass spectrometry. IGF-2, H19 and LINE-1 methylation levels in cord blood were measured using the bisulfite pyrosequencing methods Finally, we assessed the associations between prenatal exposure to various PCBs and the gene methylation levels using multiple regression models stratified by infant gender. We observed a 0.017 (95% confidence interval [CI]: 0.003-0.031) increase in the log10-transformed H19 methylation levels (%) in cord blood for each ten-fold increase in the levels of decachlorinated biphenyls (decaCBs) in maternal blood among all infants. Similarly, a 0.005 (95% CI: 0.000-0.010) increase in the log10-transformed LINE-1 methylation levels (%) in cord blood was associated with each ten-fold increase in heptachlorinated biphenyls (heptaCBs) in maternal blood among all infants. In particular, we observed a dose-dependent association of the decaCB levels in maternal blood with the H19 methylation levels among female infants (P value for trend=0.040); likewise a dose-dependent association of heptaCB levels was observed with LINE-1 methylation levels among female infants (P value for trend=0.015). Moreover, these associations were only observed among infants of primiparous women. Our results suggest that the dose-dependent association between prenatal exposure to specific non-dioxin-like PCBs and increases in the H19 and LINE-1 methylation levels in cord blood might be more predominant in females than in males.

Pollutants and diet influence carotenoid levels and integument coloration in nestlings of an endangered raptor

Carotenoid-based traits or ornaments, such as yellow-red integuments (feathers, beaks, legs or eye-rings) displayed by birds, play key roles in social communication by reliably advertising an individual's quality or health. In some species, these traits are displayed not only by adults but also by nestlings, and function in parent-offspring communication or sibling competition by advertising an individual's physical or physiological condition. Pollutants such as organochlorine compounds (OCs) could have disruptive effects on the coloration of these traits, thereby interfering with communication processes. Such effects have been reported in adult birds, but are still largely unknown for nestlings. Here we investigated associations between polychlorinated biphenyl (PCB) and dichlorodiphenyltrichloroethane (DDT) blood-levels, circulating carotenoid levels and the yellow-orange coloration of the cere and tarsi of wild Black Harrier Circus maurus nestlings, a scarce raptor endemic to southern Africa. As carotenoid pigments must be acquired through the diet, we also tested for an effect of dietary composition. The orangeness-purity of cere and tarsi coloration positively correlated with circulating carotenoid levels, and increased with both nestling age and the proportion of birds consumed in the diet. Circulating carotenoid levels and the orangeness-purity of colored integuments were unrelated to blood PCB levels, although the brightness of integuments (i.e. lack of pigmentation) increased with PCB levels. Nestlings with more DDT had lower levels of circulating carotenoids and reduced carotenoid-based coloration (i.e. higher hue and lower saturation, reflecting a yellow rather than orange and less intense color, respectively). Together, our results are consistent with the hypothesis that OC contaminants, in particular DDT, may disrupt carotenoid-based signaling in exposed nestlings.

Environmental exposure to human carcinogens in teenagers and the association with DNA damage

BackgroundWe investigated whether human environmental exposure to chemicals that are labeled as (potential) carcinogens leads to increased (oxidative) damage to DNA in adolescents. Material and methodsSix hundred 14–15-year-old youngsters were recruited all over Flanders (Belgium) and in two areas with important industrial activities. DNA damage was assessed by alkaline and formamidopyrimidine DNA glycosylase (Fpg) modified comet assays in peripheral blood cells and analysis of urinary 8-hydroxydeoxyguanosine (8-OHdG) levels. Personal exposure to potentially carcinogenic compounds was measured in urine, namely: chromium, cadmium, nickel, 1-hydroxypyrene as a proxy for exposure to other carcinogenic polycyclic aromatic hydrocarbons (PAHs), t,t-muconic acid as a metabolite of benzene, 2,5-dichlorophenol (2,5-DCP), organophosphate pesticide metabolites, and di(2-ethylhexyl) phthalate (DEHP) metabolites. In blood, arsenic, polychlorinated biphenyl (PCB) congeners 118 and 156, hexachlorobenzene (HCB), dichlorodiphenyltrichloroethane (DDT) and perfluorooctanoic acid (PFOA) were analyzed. Levels of methylmercury (MeHg) were measured in hair. Multiple linear regression models were used to establish exposure-response relationships. ResultsBiomarkers of exposure to PAHs and urinary chromium were associated with higher levels of both 8-OHdG in urine and DNA damage detected by the alkaline comet assay. Concentrations of 8-OHdG in urine increased in relation with increasing concentrations of urinary t,t-muconic acid, cadmium, nickel, 2,5-DCP, and DEHP metabolites. Increased concentrations of PFOA in blood were associated with higher levels of DNA damage measured by the alkaline comet assay, whereas DDT was associated in the same direction with the Fpg-modified comet assay. Inverse associations were observed between blood arsenic, hair MeHg, PCB 156 and HCB, and urinary 8-OHdG. The latter exposure biomarkers were also associated with higher fish intake. Urinary nickel and t,t-muconic acid were inversely associated with the alkaline comet assay. ConclusionThis cross-sectional study found associations between current environmental exposure to (potential) human carcinogens in 14–15-year-old Flemish adolescents and short-term (oxidative) damage to DNA. Prospective follow-up will be required to investigate whether long-term effects may occur due to complex environmental exposures.

Quantification of all 209 PCB congeners in blood-Can indicators be used to calculate the total PCB blood load?

Polychlorinated biphenyls (PCBs) are a substance group of 209 theoretically possible compounds. The human body burden of PCBs is commonly calculated based on so-called indicator congeners such as PCB 138, PCB 153 and PCB 180, which are analyzed in human blood. The German "Human Biomonitoring (HBM) Commission" assumes that the sum of these indicator congeners multiplied by a factor of 2 represents the total PCB burden. This norm is based on data obtained from exposure studies after dietary intake. Data from indoor air shows a different congener pattern, which might lead to a relatively higher intake of lower chlorinated PCBs by inhalation. In two independent studies with adult participants from two regions in Germany, we measured all 209 PCB congeners in 44 whole blood and 42 plasma samples. Participants from the whole blood study group had additional exposure to PCBs via indoor air. With our analytical method, 141 individual PCB congeners, 27 coeluted pairs of PCB congeners and 2 records of 3 and 4 coeluted PCBs could be determined. Thus, 172 analysis results were reported per sample. In the whole blood samples, 50 congeners showed values below the limit of quantification (LOQ), whereas 94 congeners could not be detected in any of plasma samples. Total PCB concentrations (Σ 209 PCB congeners, incl. ½ LOQ) in the whole blood samples ranged from 99 to 2152ng PCB/g lipid (Median: 454ng/g lipid; 95th Percentile: 1404ng/g lipid). The sum of all 209 measured PCB (incl. ½ LOQ) in plasma samples showed levels between 52 and 933ng PCB/g lipid (Median: 226ng/g lipid; 95th Percentile: 642ng/g lipid). Our results show that the burden of PCBs on the human body is caused mainly by the three highly chlorinated indicator congeners PCB 138, PCB 153 and PCB 180. In median approximately 50% of the total PCB content in human whole blood or plasma samples can be attributed to these congeners. Total PCB, calculated by multiplying the sum of the three indicator congeners by 2, showed a strong and highly significant correlation to the sum of all 209 measured congeners for each sample. A slightly stronger correlation in the whole blood samples could be achieved by choosing six indicator congeners, including the lower chlorinated congeners (PCB 28, 52 and 101) into the calculation. Although this difference is very small, it must be considered that higher PCB levels in indoor air than those measured in the present study might be associated with a higher burden of indoor-air-related congeners in exposed individuals. For precautionary reasons, it could therefore be recommended that the assessment of individuals exposed to PCB via indoor air should be carried out based on the sum of the 6 indicator congeners PCB 28, PCB 52, PCB 101, PCB 138, PCB 153 and PCB 180 multiplied by a factor of 2.
