Full genome sequence for the African swine fever virus outbreak in the Dominican Republic in 1980

African swine fever is a lethal disease of domestic pigs, geographically expanding as a pandemic, that is affecting countries across Eurasia and severely damaging their swine production industry. After more than 40 years of being absent in the Western hemisphere, in 2020 ASF reappeared in the Dominican Republic and Haiti. The recent outbreak strain in the Dominican Republic has been identified as a genotype II ASFV a derivative of the ASF strain circulating in Asia and Europe. However, to date no full-length genome sequence from either the 1978–1980 Here we report the complete genome sequence of an African swine fever virus (ASFV) (DR-1980) that was previously isolated from blood collected in 1980 from the Dominican Republic at the end of the last outbreak, before culling of all swine on the island of Hispaniola and stored in the Plum Island Animal Disease Center ASFV repository. A contig representing the full-length genome (183,687 base pairs) was de novo assembled into a single contig using both Nanopore and Illumina sequences. DR-1980 was determined to belong to genotype I and, as determined by full genome comparison, a close relative to the sequenced Sardinia viruses that were causing outbreaks at this time.

Harley William Moon,DVM, Ph.D., an outstanding American person and researcher of comparative microbiology and pathology of intestinal diseases, the former director of the USDA, ARS, National Animal Disease Center (Iowa), of Plum Island Animal Disease Center (New York) and of Veterinary Research Institute of Iowa State University, member of the National Academy of Sciences (USA) passed away after some difficult and lonely last years of his life, on October 7, 2018 at the age of 82.

Smart Card Decontamination in a High-Containment Laboratory.

Validated procedures for decontamination of laboratory surfaces and equipment are essential to biosafety and biorisk programs at high-containment laboratories. Each high-containment laboratory contains a unique combination of surfaces, procedures, and biological agents that require decontamination methods tailored to specific facility practices. The Plum Island Animal Disease Center (PIADC) is a high-containment laboratory operating multiple biosafety level (BSL)-3, ABSL-3, and BSL-3 Ag spaces. The PIADC facility requires the use of federally issued smart cards, called personal identity verification (PIV) cards, to access information technology (IT) networks both outside and within the high-containment laboratory. Because PIV cards may require transit from the BSL-3 to office spaces, a validated procedure for disinfecting PIV card surfaces prior to removal from the laboratory is critical to ensure biosafety and biosecurity. Two high-risk select agents used in the PIADC high-containment laboratory are foot-and-mouth disease virus (FMDV) and swine vesicular disease virus (SVDV). We evaluated disinfection of PIV cards intentionally spotted with FMDV and SVDV using a modified quantitative carrier test and the liquid chemical disinfectant Virkon® S. Our experimental design modeled a worst-case scenario of PIV card contamination and disinfection by combining high concentrations of virus dried with an organic soil load and use of aged Virkon® S prepared in hard water. Results showed that FMDV and SVDV dried on PIV card surfaces were completely inactivated after immersion for 30 and 60 seconds, respectively, in a 5-day-old solution of 1% Virkon® S. Therefore, this study provided internal validation of PIADC biosafety protocols by demonstrating the efficacy of Virkon® S to inactivate viruses on contaminated smart cards at short contact times.

The Convergence of High-Consequence Livestock and Human Pathogen Research and Development: A Paradox of Zoonotic Disease.

The World Health Organization (WHO) estimates that zoonotic diseases transmitted from animals to humans account for 75 percent of new and emerging infectious diseases. Globally, high-consequence pathogens that impact livestock and have the potential for human transmission create research paradoxes and operational challenges for the high-containment laboratories that conduct work with them. These specialized facilities are required for conducting all phases of research on high-consequence pathogens (basic, applied, and translational) with an emphasis on both the generation of fundamental knowledge and product development. To achieve this research mission, a highly-trained workforce is required and flexible operational methods are needed. In addition, working with certain pathogens requires compliance with regulations such as the Centers for Disease Control (CDC) and the U.S. Department of Agriculture (USDA) Select Agent regulations, which adds to the operational burden. The vast experience from the existing studies at Plum Island Animal Disease Center, other U.S. laboratories, and those in Europe and Australia with biosafety level 4 (BSL-4) facilities designed for large animals, clearly demonstrates the valuable contribution this capability brings to the efforts to detect, prepare, prevent and respond to livestock and potential zoonotic threats. To raise awareness of these challenges, which include biosafety and biosecurity issues, we held a workshop at the 2018 American Society for Microbiology (ASM) Biothreats conference to further discuss the topic with invited experts and audience participants. The workshop covered the subjects of research funding and metrics, economic sustainment of drug and vaccine development pipelines, workforce turnover, and the challenges of maintaining operational readiness of high containment laboratories.

FOOT-AND-MOUTH DISEASE IN A SMALL SAMPLE OF EXPERIMENTALLY INFECTED PRONGHORN (ANTILOCAPRA AMERICANA).

There is limited information on the pathogenesis and epidemiology of foot-and-mouth disease (FMD) in North American wildlife and none concerning pronghorn ( Antilocapra americana ). In an experimental study of 13 pronghorn and six steers ( Bos taurus ), we compared the susceptibility of pronghorn to FMD virus (FMDV) strain O, with that of cattle ( Bos taurus ). We also determined the potential for intra- and interspecies transmission of FMDV strain O in pronghorn and cattle, assessed the application of conventional laboratory tests in their suitability to detect FMDV infection in pronghorn, and evaluated the potential role of pronghorn as efficient long-term carriers of FMDV. After acclimation to containment at Plum Island Animal Disease Center, two pronghorn and one steer were each infected by intraepithelial tongue inoculation with 10,000 bovine tongue infective doses of FMDV, strain O1 Manisa. Inoculated animals were housed with contact animals. When contact-exposed animals developed fever they were placed in rooms with previously unexposed animals. All inoculated and exposed cattle and pronghorn developed clinical disease typical of FMD. Pronghorn developed severe foot lesions and mild to moderate oral lesions, primarily on the tongue. Duration of clinical signs in both species was 2-3 wk with foot abnormalities evident to the end of the study (51 d postexposure). Other lesions included pancreatitis, myositis of the tongue, and secondary lesions including pleuritis, pneumonia, decubital ulcers, and tenosynovitis. Virus transmission occurred between pronghorn, from cattle to pronghorn, and from pronghorn to cattle. Conventional laboratory tests detected virus and antibodies against nonstructural and structural FMDV proteins in pronghorn and cattle. Virus was present in some animals for 1 wk but was not detectable by virus isolation or PCR at 3 wk postinfection or afterward.

Development, optimization, and validation of a Classical swine fever virus real-time reverse transcription polymerase chain reaction assay

Classical swine fever (CSF) is an economically devastating disease of pigs. Instrumental to the control of CSF is a well-characterized assay that can deliver a rapid, accurate diagnosis prior to the onset of clinical signs. A real-time fluorogenic-probe hydrolysis (TaqMan) reverse transcription polymerase chain reaction (RT-PCR) for CSF was developed by the United States Department of Agriculture (USDA) at the Plum Island Animal Disease Center (CSF PIADC assay) and evaluated for analytical and diagnostic sensitivity and specificity. A well-characterized panel including Classical swine fever virus (CSFV), Bovine viral diarrhea virus (BVDV), and Border disease virus (BDV) isolates was utilized in initial feasibility and optimization studies. The assay was initially designed and validated for use on the ABI 7900HT using the Qiagen QuantiTect® Probe RT-PCR chemistry. However, demonstrating equivalency with multiple one-step RT-PCR chemistries and PCR platforms increased the versatility of the assay. Limit of detection experiments indicated that the Qiagen QuantiTect® Multiplex (NoROX) and the Invitrogen SuperScript® III RT-PCR kits were consistently the most sensitive one-step chemistries for use with the CSF PIADC primer/probe set. Analytical sensitivity of the CSF PIADC assay ranged from <1-2.95 log(10) TCID(50)/ml on both the ABI 7900HT and ABI 7500 platforms. The CSF PIADC assay had 100% diagnostic sensitivity and specificity when tested on a panel of 152 clinical samples from the Dominican Republic and Colombia. The ability to perform this newly developed assay in 96-well formats provides an increased level of versatility for use in CSF surveillance programs.

How Kansas Nabbed the New Bio- and Agro-Defense Lab

If you build it, they will come. The maxim seems to have worked for Kansas State University (KSU) in Manhattan, which the U.S. Department of Homeland Security (DHS) selected last week as its preferred site for a $450 million facility to replace the aging Plum Island Animal Disease Center off Long Island, New York. What made the university stand out from the competition, KSU and federal officials say, was a recently completed $54 million biocontainment lab on campus for animal research, funded by the state, as well as the lack of any vigorous local opposition to work involving dangerous animal pathogens.

FOOT-AND-MOUTH DISEASE IN NORTH AMERICAN BISON (BISON BISON) AND ELK (CERVUS ELAPHUS NELSONI): SUSCEPTIBILITY, INTRA- AND INTERSPECIES TRANSMISSION, CLINICAL SIGNS, AND LESIONS

There is limited information about the pathogenesis and epidemiology of foot-and-mouth disease (FMD) in North American bison (Bison bison) or elk (Cervus elaphus nelsoni). In these two experimental infection studies, we compared the susceptibilities of bison and elk to FMD virus (FMDV), respectively, with that of cattle; determined whether intra- and interspecies transmission could occur in bison and cattle, and elk and cattle; determined suitability of conventional available laboratory tests to detect FMDV infection in bison and elk; and investigated whether bison or elk are efficient long-term carriers of FMDV. In both studies, after a period of acclimation to the containment at Plum Island Animal Disease Center, animals were infected by intraepithelial tongue inoculation with 10,000 bovine tongue infective doses of FMDV, strain O1 Manisa. Inoculated animals were kept with contact animals; subsequently, inoculated and/or exposed contact animals were placed in rooms with unexposed animals. All bison developed oral mucosal and foot lesions similar to those of cattle. Bison developed fever, lameness, inappetence, and ptyalism. Physical examinations on bison revealed numerous small vesicles and erosions affecting tongue, gingiva, muzzle, hard and soft palates, coronary bands, and interdigital skin. Inoculated elk developed transient fever and mild focal tongue and foot lesions. Contact elk developed neither clinical signs nor gross pathologic lesions of FMD. At necropsy, lesions in bison included numerous extensive vesicles, erosions, and/or ulcers in the oral cavities, feet, and rumen pillars depending on the stage of disease. Less extensive oral, foot, and rumen lesions were present in the inoculated elk. All bison and inoculated elk developed antibodies to FMDV and were positive for FMDV by reverse transcription-polymerase chain reaction (RT-PCR). Transmission occurred between cattle and bison, and bison and bison. It did not occur between elk and cattle. Elk-to-elk transmission studies resulted in only one contact elk developing serologic evidence of a subclinical infection. Other exposed elk developed neither clinical, pathologic, virologic, nor serologic evidence of disease. FMDV was not isolated from animals past 28 days postinfection.

Eighteen of 29 applicant sites are still in the running for a new $450 million high-security agro-biodefense lab to replace Plum Island Animal Disease Center, the aging facility off Long Island, New York ( Science , 2 September 2005, [p. 1475][1]). The Department of Homeland Security is funding the National Bio-and Agro-Defense Facility to study animal diseases and possibly human illnesses. Microscopy and Microbes at Plum Island: Protecting America's Livestock

Abstract Plum Island Animal Disease Center, located on a small island off the coast of Long Island's North Fork, has been clouded in mystery and misinformation for years. Often the topic of conspiracy theorists, this secret place has generated many myths—from aliens to anthrax and pink eels to secret submarines. But the truth of the center's mission is far less colorful yet far more crucial to the state of the nation's agriculture. In June 2003, operational responsibility for the Plum Island Animal Disease Center (PIADC) transferred from the United States Department of Agriculture (USDA) to the United States Department of Homeland Security (DHS), and PIADC became the core of the DHS effort to protect US livestock from foreign animal disease agents.

Homeland Security Ponders Future of Its Animal Disease Fortress

The Alcatraz of animal diseases may come ashore. Last week, the U.S. Department of Homeland Security (DHS) announced that the Plum Island Animal Disease Center (PIADC)—which studies the most devastating agricultural diseases on a tiny speck in the Atlantic off Long Island, New York—will be replaced by a new facility that may be located elsewhere. The state's politicians, who oppose expanding the lab's remit but don't want it to close, immediately blasted the proposal. But some scientists say they would welcome leaving the remote, impractical location.

O surto de peste suína africana ocorrido em 1978 no município de Paracambi, Rio de Janeiro

Em função das dúvidas que ainda perduram 25 anos após a ocorrência do surto de peste suína africana (PSA), em Paracambi, Estado do Rio de Janeiro, Brasil, em 1978, são apresentados os resultados, relativos a este foco, obtidos pelos estudos epidemiológico, clínico-patológico, virológico, bacteriológico e ultra-estrutural dos casos naturais, bem como os relativos à reprodução experimental da doença no Brasil e sua confirmação por isolamento e determinação de patogenicidade realizada no Plum Island Animal Disease Center, New York, EUA. Os animais se infectaram pela ingestão de restos de comida de aviões procedentes de Portugal e da Espanha, países nos quais a doença existia. De acordo com publicação do Ministério da Agricultura, após o diagnóstico do surto de PSA descrito neste trabalho, 223 novos focos foram relatados, entre 1978 e 1979, em todas Regiões do país (Norte, Nordeste, Centro-Oeste, Sudeste e Sul) e focos adicionais em 1981, sem informações exatas referentes ao seu número. O último caso foi relatado em 15 de novembro de 1981, e em 5 de dezembro 1984 o Brasil foi declarado livre da PSA. Para o diagnóstico da PSA foram processadas 54.002 amostras no Departamento de Virologia do Instituto de Microbiologia da Universidade Federal do Rio de Janeiro, no período de 1978 a 1981. No processamento das amostras foram usadas as técnicas de hemadsorção em cultura de leucócitos (HAd), imunoflorescência em cortes de tecido (FATS), imunoflorescência em cultivo celular (FATCC), imuno-eletrosmoforese (IEOP) e imunoflorescência indireta (IIF). Somente 4 amostras foram positivas pela técnica de FATCC, a única das provas que inclui o isolamento viral; não é mencionada a procedência dessas amostras, mas provavelmente trata-se das amostras oriundas de Paracambi. Com base na análise de todos os dados publicados sobre o tema, na possível ocorrência de falso-positivos, na falta de informações sobre isolamento e caracterização do virus, bem como na ausência de dados sobre epidemiologia, sinais clínicos e patologia nesses outros supostos focos, pode-se concluir que o surto de Paracambi constitui a única ocorrência de PSA no Brasil, comprovada por isolamento, identificação do vírus e determinação de sua patogenicidade, e que a doença manteve-se confinada a esse local, provavelmente em função do diagnóstico precoce e da rápida adoção de eficientes medidas de controle pelas autoridades sanitárias; o abate dos suínos desse rebanho iniciou-se 10 dias depois da primeira morte e 3 dias após o diagnóstico presuntivo.

Epidemiologic association between antibody titres against bovine virus diarrhoea virus, rinderpest disease virus and infectious bovine rhinotracheitis virus in a buffalo herd.

The association between antibody titres against bovine virus diarrhoea virus (BVDV), infectious bovine rhinotracheitis virus (IBRV) and rinderpest disease virus (RPDV) in buffaloes was investigated in a cross-sectional study. Thirty-six lactating buffaloes from a herd in Landhi Cattle Colony, Karachi were bled and serum samples subjected to a micro-neutralisation test at the Plum Island Animal Disease Center, USA, to categorise each buffalo either as positive or negative against each of 3 viral antigens. Log-linear analysis was used to evaluate the association among 3 categorical variables. The "best-fitting" log-linear model was the model of complete independence. This model includes all 3 main effects (BVDV, IBRV, RPDV), suggesting that the seropositivity of buffaloes against these viral antigens tended to occur independently. Estimates of parameters of the model showed that the proportion of buffaloes seropositive (97.2%) to RPDV antigen tended to be higher than expected, whereas the proportion of BVDV seropositive (30.6%) and IBRV seropositive (16.7%) buffaloes tended to be smaller than expected.

Survival of Viruses in “Prosciutto di Parma” (Parma Ham)

Co-operative studies were undertaken at the Plum Island Animal Disease Center, Agricultural Research Service, United States Department of Agriculture; at the Istituto Zooprofiiattico Sperimentale, Brescia, Italy and at the Istituto Zooprofiiattico Sperimentale, Sassari, Italy to determine if the causative agents for such diseases as foot-and-mouth, African swine fever and hog cholera were inactivated by the processing and curing procedures used in preparing Parma hams. Hams were prepared from pigs infected with each of these diseases when the carcass contained the greatest amount of virus. Foot-and-mouth disease virus could not be recovered from the hams at 108 days in the U.S. experiment and 170 days in the Italian experiment. African swine fever virus was not recovered at 399 days in the U.S. experiment and 300 days in the Italian experiment. The virus of hog cholera was not recovered at 313 days in the U.S. experiment and 189 days in the Italian experiment.

Survival of Swine Vesicular Disease Virus in “Prosciutto di Parma” (Parma Ham)

Experiments were conducted at the Plum Island Animal Disease Center of the Agricultural Research Service, United States Department of Agriculture and at the Istituto Zooprofilattico, Brescia, Italy, to determine if the processing and curing of the Parma hams would inactivate swine vesicular disease virus. The experimental data indicated that the virus was inactivated between 180 and 300d postslaughter in the U.S. experiment and between 90 and 182d post-slaughter in the Italian experiment. The minimal curing period for Parma hams is 365d, which would assure that the ham is free of infectious swine vesicular disease virus.

Rotavirus-like, calicivirus-like, and 23-nm virus-like particles associated with diarrhea in young pigs.

Virus particles morphologically similar to caliciviruses and rotaviruses were detected by electron microscopy (EM) in the intestinal contents of a 27-day-old diarrheic nursing pig. A third small spherical 23-nm virus-like particle was also observed. Calicivirus-like particles averaged 33 nm in diameter. Similar to rotaviruses, rotavirus-like particles were present as single-capsid 55-nm forms or double-capsid 70-nm particles. Most gnotobiotic pigs orally exposed to samples containing these three viruses developed diarrhea and villous atrophy of the small intestine, and all shed the three viruses in their intestinal contents. Attempts to propagate these viruses in cell culture were unsuccessful. The antigenic relationship of the rotavirus-like particles to known rotaviruses was explored by immune EM and immunofluorescent staining. By these techniques, the rotavirus-like particles did not cross-react with antisera to porcine, bovine, or human rotaviruses or to reovirus type 3. Antisera from gnotobiotic pigs exposed to all three viruses had enzyme-linked immunosorbent assay and virus neutralization titers of <4 against porcine rotavirus. Previous infection of gnotobiotic pigs with the mixture containing rotavirus-like particles failed to protect them against a subsequent challenge with porcine rotavirus. The antigenic relationship of the calicivirus-like particles to known caliciviruses was investigated by immune EM and virus neutralization. By these tests, the calicivirus-like particles did not react with antisera against feline calicivirus strain 255 or M-8. In a study conducted at Plum Island Animal Disease Center, antiserum against the three combined agents did not specifically neutralize any serotype of swine vesicular exanthema virus.
