Early auditory processing abnormalities alter individual learning trajectories and sensitivity to computerized cognitive training in schizophrenia.

Auditory system plasticity is a promising target for neuromodulation, cognitive rehabilitation and therapeutic development in schizophrenia (SZ). Auditory-based targeted cognitive training (TCT) is a 'bottom up' intervention designed to enhance the speed and accuracy of auditory information processing, which has been shown to improve neurocognition in certain SZ patients. However, the dynamics of TCT learning as a function of training exercises and their impact on neurocognitive functioning and therapeutic outcomes are unknown. Forty subjects (SZ, n = 21; healthy subjects (HS), n = 19) underwent comprehensive clinical, cognitive, and auditory assessments, including measurements of auditory processing speed (APS) at baseline and after 1-h of TCT. SZ patients additionally completed 30-hours of TCT and repeated assessments ~10-12 weeks later. SZ patients were deficient in APS at baseline (d = 0.96, p < 0.005) relative to HS. After 1-h of TCT, analyses revealed significant main effects of diagnosis (d = 1.75, p = 0.002) and time (d = 1.04, p < 0.001), and a diagnosis × time interaction (d = 0.85, p = 0.02) on APS. APS learning effects were robust after 1-h in SZ patients (d = 1.47, p < 0.001) and persisted throughout the 30-h of training. Baseline APS was associated with verbal learning gains after 30-h of TCT (r = 0.51, p = 0.02) in SZ. TCT learning metrics may have prognostic utility and aid in the prospective identification of individuals likely to benefit from TCT. Future experimental medicine studies may advance predictive algorithms that enhance TCT-related clinical, cognitive and functional outcomes.

Neuroanatomical Disposition, Natural Development, and Training-Induced Plasticity of the Human Auditory System from Childhood to Adulthood: A 12-Year Study in Musicians and Nonmusicians.

Auditory perception is fundamental to human development and communication. However, no long-term studies have been performed on the plasticity of the auditory system as a function of musical training from childhood to adulthood. The long-term interplay between developmental and training-induced neuroplasticity of auditory processing is still unknown. We present results from AMseL (Audio and Neuroplasticity of Musical Learning), the first longitudinal study on the development of the human auditory system from primary school age until late adolescence. This 12-year project combined neurologic and behavioral methods including structural magnetic resonance imaging (MRI), magnetoencephalography (MEG), and auditory tests. A cohort of 112 typically developing participants (51 male, 61 female), classified as "musicians" (n = 66) and "nonmusicians" (n = 46), was tested at five measurement timepoints. We found substantial, stable differences in the morphology of auditory cortex (AC) between musicians and nonmusicians even at the earliest ages, suggesting that musical aptitude is manifested in macroscopic neuroanatomical characteristics. Maturational plasticity led to a continuous increase in white matter myelination and systematic changes of the auditory evoked P1-N1-P2 complex (decreasing latencies, synchronization effects between hemispheres, and amplitude changes) regardless of musical expertise. Musicians showed substantial training-related changes at the neurofunctional level, in particular more synchronized P1 responses and bilaterally larger P2 amplitudes. Musical training had a positive influence on elementary auditory perception (frequency, tone duration, onset ramp) and pattern recognition (rhythm, subjective pitch). The observed interplay between "nature" (stable biological dispositions and natural maturation) and "nurture" (learning-induced plasticity) is integrated into a novel neurodevelopmental model of the human auditory system.Significance Statement We present results from AMseL (Audio and Neuroplasticity of Musical Learning), a 12-year longitudinal study on the development of the human auditory system from childhood to adulthood that combined structural magnetic resonance imaging (MRI), magnetoencephalography (MEG), and auditory discrimination and pattern recognition tests. Visuo-Acoustic Stimulation’s Role in Synaptic Plasticity: A Review of the Literature

Brain plasticity is the capacity of cerebral neurons to change, structurally and functionally, in response to experiences. This is an essential property underlying the maturation of sensory functions, learning and memory processes, and brain repair in response to the occurrence of diseases and trauma. In this field, the visual system emerges as a paradigmatic research model, both for basic research studies and for translational investigations. The auditory system remains capable of reorganizing itself in response to different auditory stimulations or sensory organ modification. Acoustic biofeedback training can be an effective way to train patients with the central scotoma, who have poor fixation stability and poor visual acuity, in order to bring fixation on an eccentrical and healthy area of the retina: a pseudofovea. This review article is focused on the cellular and molecular mechanisms underlying retinal sensitivity changes and visual and auditory system plasticity.

Sound-localisation performance in patients with congenital unilateral microtia and atresia fitted with an active middle ear implant.

This study assessed the safety and sound-localisation ability of the Vibrant Soundbridge (VSB) (Med-EL, Innsbruck, Austria) in patients with unilateral microtia and atresia (MA). This was a single-centre retrospective research study. Twelve subjects with unilateral conductive hearing loss (UCHL) caused by ipsilateral MA were recruited, each of whom underwent VSB implantation and auricular reconstruction. The bone-conduction (BC) threshold was measured postoperatively, and the accuracy of sound localisation was evaluated at least 6months after surgery. Horizontal sound-localisation performance was investigated with the VSB activated and inactivated, at varying sound stimuli levels (65, 70 and 75dB SPL). Localisation benefit was analysed via the mean absolute error (MAE). There was no statistical difference in mean BC threshold of impaired ears measured preoperatively and postoperatively. When compared with VSB-inactivated condition, the MAE increased significantly in unilateral MA patients in the VSB-activated condition. Besides, sound-localisation performance worsened remarkably when sound was presented at 70dB SPL and 75dB SPL. Regarding the side of signal location, the average MAE with the VSB device was much higher than that without the VSB when sound was from the normal-hearing ear. However, no significant difference was observed when sound was located from the impaired ear. This study demonstrates that in patients with unilateral MA, the VSB device does not affect inner-ear function. Sound-localisation ability is not improved, but deteriorated at follow-up. Our results suggest that the VSB-aided localisation abilities may be related to the thresholds between the ears, plasticity of auditory system and duration of use of VSB.

Auditory Inspired Convolutional Neural Networks for Ship Type Classification with Raw Hydrophone Data.

Detecting and classifying ships based on radiated noise provide practical guidelines for the reduction of underwater noise footprint of shipping. In this paper, the detection and classification are implemented by auditory inspired convolutional neural networks trained from raw underwater acoustic signal. The proposed model includes three parts. The first part is performed by a multi-scale 1D time convolutional layer initialized by auditory filter banks. Signals are decomposed into frequency components by convolution operation. In the second part, the decomposed signals are converted into frequency domain by permute layer and energy pooling layer to form frequency distribution in auditory cortex. Then, 2D frequency convolutional layers are applied to discover spectro-temporal patterns, as well as preserve locality and reduce spectral variations in ship noise. In the third part, the whole model is optimized with an objective function of classification to obtain appropriate auditory filters and feature representations that are correlative with ship categories. The optimization reflects the plasticity of auditory system. Experiments on five ship types and background noise show that the proposed approach achieved an overall classification accuracy of 79.2%, which improved by 6% compared to conventional approaches. Auditory filter banks were adaptive in shape to improve accuracy of classification.

Cortical auditory plasticity in children with Cochlear implants

Objectives:The purpose of the current study was to investigate plasticity of auditory system following cochlear implants (CI) in prelingually severe to profound hearing-impaired chil¬dren using cortical auditory evoked potentials (CAEPs) and correlate it with auditory perception performance.Methods:A total of 28 (15 boys, 13 girls) children with profound hearing loss, who underwent CI at Ahvaz Jundishapur University of Medical Sciences, Iran, were included in this study. Their mean age at the time of implantation was 21.3 months. All children were evaluated before implantation and 3 months after implantation using the CAEPs and categorical auditory performance (CAP). For CAEP measurement, the stimuli on the HEARLab system (/m/, /g/, and /t/) were extracted from running speech and presented at 65 dB SPL.Results:The mean CAP and P1 amplitude values were increased from pre-CI condition to 3-month post-CI condition (Paired t-test, p 0.05).Conclusion:The present study indicated that early cochlear implantation, will improve cortical auditory plasticity and auditory performance ability in pre-lingual hearing-impaired children.

N-methyl-d-aspartate-type glutamate receptor modulators and related medications for the enhancement of auditory system plasticity in schizophrenia

Deficits in N-methyl-d-aspartate-type (NMDAR) function contribute to cognitive deficits in schizophrenia, particularly dysfunction in neuroplasticity, defined as reduced learning during training on exercises that place implicit, increasing demands on early sensory (auditory and visual) information processing. Auditory mismatch negativity (MMN) can be both a target engagement biomarker for the NMDAR and a proxy measure of neurophysiological plasticity. This review covers the evidence for using NMDAR modulator and related compounds for enhancement of cognition, with a particular focus on early auditory processing/plasticity. Compounds covered include glycine site agonists, glycine and system A-type transporter inhibitors, d-amino acid oxidase inhibitors, memantine and nicotinic alpha-7 acetylcholine receptor agonists. As opposed to daily treatment studies focusing on schizophrenia in general, intermittent, non-daily treatment combining NMDAR modulators with neuroplasticity-based paradigms, using MMN as target-engagement biomarkers show promise as treatments to both remediate plasticity deficits and overall functional deficits.

Context-dependent plasticity in the subcortical encoding of linguistic pitch patterns.

Human auditory perception in dynamic listening environments requires fine-tuning of sensory signal based on behaviorally relevant regularities in listening context, i.e., online experience-dependent plasticity. Our finding suggests what partly underlie online experience-dependent plasticity are interplaying contextual processes in the subcortical auditory system that are responsive to predictability as well as novelty in listening context. These findings add to the literature that looks to establish the neurophysiological bases of auditory system plasticity, a central issue in auditory neuroscience.

Measuring the capacity for auditory system plasticity: An examination of performance gains during initial exposure to auditory-targeted cognitive training in schizophrenia

Auditory-Targeted Cognitive Training (ATCT), which aims to improve auditory information processing efficiency, has shown great promise for remediating cognitive deficits in schizophrenia (SZ). However, there is substantial heterogeneity in the degree of cognitive gains made during ATCT, and some patients show negligible benefit after completing therapeutic doses of training. Identifying individual differences that can be measured early in the course of ATCT and that predict subsequent cognitive benefits from the intervention is therefore important. The present study calculated a variety of performance metrics during the initial hour of exposure to ATCT Sound Sweeps, a frequency discrimination time-order judgment task, and investigated the relationships of these metrics to demographic, clinical, and cognitive characteristics of SZ patients.Thirty-seven SZ outpatients completed measures of auditory attention, working memory, verbal memory, and executive functioning, followed by 1h of Sound Sweeps training. Performance metrics, calculated after the first training level, the first training stage (Levels 1–4), and the entire hour of training included baseline and best auditory processing speed (APS) scores, as well as percent improvement in APS after training. The number of training levels completed by each participant was also calculated.Baseline and best APS correlated with performance in all cognitive domains, whereas APS improvements only correlated with verbal memory. Number of training levels completed was marginally associated with auditory attention only.Conclusions: Sound Sweeps performance correlates with a range of neurocognitive abilities. APS improvement may provide a particularly sensitive index of “plasticity potential” within the neural network underlying verbal learning and memory.

Single unit hyperactivity and bursting in the auditory thalamus of awake rats directly correlates with behavioural evidence of tinnitus.

Tinnitus is an auditory percept without an environmental acoustic correlate. Contemporary tinnitus models hypothesize tinnitus to be a consequence of maladaptive plasticity-induced disturbance of excitation-inhibition homeostasis, possibly convergent on medial geniculate body (MGB, auditory thalamus) and related neuronal networks. The MGB is an obligate acoustic relay in a unique position to gate auditory signals to higher-order auditory and limbic centres. Tinnitus-related maladaptive plastic changes of MGB-related neuronal networks may affect the gating function of MGB and enhance gain in central auditory and non-auditory neuronal networks, resulting in tinnitus. The present study examined the discharge properties of MGB neurons in the sound-exposure gap inhibition animal model of tinnitus. MGB single unit responses were obtained from awake unexposed controls and sound-exposed adult rats with behavioural evidence of tinnitus. MGB units in animals with tinnitus exhibited enhanced spontaneous firing, altered burst properties and increased rate-level function slope when driven by broadband noise and tones at the unit's characteristic frequency. Elevated patterns of neuronal activity and altered bursting showed a significant positive correlation with animals' tinnitus scores. Altered activity of MGB neurons revealed additional features of auditory system plasticity associated with tinnitus, which may provide a testable assay for future therapeutic and diagnostic development.

Zjawiska te określa się mianem „plastyczności słuchowej”. Rozwijające się w ostatnich latach techniki badań neuroobrazowych pozwalają w sposób nieinwazyjny mierzyć zakres oraz mechanizmy zmian plastycznych, zarówno w ich aspekcie czasowym, jak i lokalizacyjnym. Wprowadzenie do obszarów zastosowań tych metod stanowi temat niniejszego artykułu przeglądowego.

Auditory Training: Evidence for Neural Plasticity in Older Adults.

Improvements in digital amplification, cochlear implants, and other innovations have extended the potential for improving hearing function; yet, there remains a need for further hearing improvement in challenging listening situations, such as when trying to understand speech in noise or when listening to music. Here, we review evidence from animal and human models of plasticity in the brain's ability to process speech and other meaningful stimuli. We considered studies targeting populations of younger through older adults, emphasizing studies that have employed randomized controlled designs and have made connections between neural and behavioral changes. Overall results indicate that the brain remains malleable through older adulthood, provided that treatment algorithms have been modified to allow for changes in learning with age. Improvements in speech-in-noise perception and cognition function accompany neural changes in auditory processing. The training-related improvements noted across studies support the need to consider auditory training strategies in the management of individuals who express concerns about hearing in difficult listening situations. Given evidence from studies engaging the brain's reward centers, future research should consider how these centers can be naturally activated during training.

Parvalbumin immunoreactivity in the auditory cortex of a mouse model of presbycusis

Age-related hearing loss (presbycusis) affects ∼35% of humans older than sixty-five years. Symptoms of presbycusis include impaired discrimination of sounds with fast temporal features, such as those present in speech. Such symptoms likely arise because of central auditory system plasticity, but the underlying components are incompletely characterized. The rapid spiking inhibitory interneurons that co-express the calcium binding protein Parvalbumin (PV) are involved in shaping neural responses to fast spectrotemporal modulations. Here, we examined cortical PV expression in the C57bl/6 (C57) mouse, a strain commonly studied as a presbycusis model. We examined if PV expression showed auditory cortical field- and layer-specific susceptibilities with age. The percentage of PV-expressing cells relative to Nissl-stained cells was counted in the anterior auditory field (AAF) and primary auditory cortex (A1) in three age groups: young (1–2 months), middle-aged (6–8 months) and old (14–20 months). There were significant declines in the percentage of cells expressing PV at a detectable level in layers I–IV of both A1 and AAF in the old mice compared to young mice. In layers V–VI, there was an increase in the percentage of PV-expressing cells in the AAF of the old group. There were no changes in percentage of PV-expressing cells in layers V–VI of A1. These data suggest cortical layer(s)- and field-specific susceptibility of PV+ cells with presbycusis. The results are consistent with the hypothesis that a decline in inhibitory neurotransmission, particularly in the superficial cortical layers, occurs with presbycusis.

Source analysis reveals plasticity in the auditory cortex: Evidence for reduced hemispheric asymmetries following unilateral deafness

ObjectiveTo investigate the effect of acquired unilateral deafness on hemispheric asymmetries in adult humans using cortical auditory evoked potentials. MethodsN1 cortical auditory evoked potentials were measured from 30 channels in 18 unilateral profoundly-deaf participants (6 right-sided and 12 left-sided deafness) and 18 audiogram-matched controls. Stimuli were 0.5-kHz and 4-kHz tones presented monaurally, and the data were analysed using global field power and dipole source analysis. ResultsThere was a statistically significant difference in dipole source strength and orientation between the two groups. Similar changes (increased dipole strength and more medial orientation) were apparent after profound unilateral deafness of either ear and for both stimuli. ConclusionsThe results reveal evidence of central auditory system plasticity that is consistent with animal models having experimentally induced unilateral deafness. SignificanceThe trend towards reduced hemispheric asymmetries was reflected in the dipole source model by changes in dipole strength, location and orientation. These findings may explain the inconsistencies reported in previous studies involving dipole source analysis where location and orientation have not always been considered.

The Hybrid cochlear implant: a review.

The Hybrid S or 'short-electrode' cochlear implant was developed to treat patients with a severe to profound hearing loss limited to the high frequencies. The short electrode is implanted into just the base or high-frequency region of the cochlea, with the goal of preserving residual low-frequency hearing. As a result, electric stimulation can be combined with acoustic stimulation in the same ear (and the opposite ear); this is one instance of 'acoustic plus electric' (A + E) stimulation. In this paper, we will review the latest findings from the first two stages of the clinical trial for the Hybrid concept in the United States. Generally, we will review surgical techniques, clinical trial criteria, residual hearing preservation, improvements in speech perception in quiet, and predictive factors for patient benefit. We will also discuss the significant benefit of A + E stimulation for speech perception in noise and musical measures of melody and instrument recognition, as well as valuable insights into central auditory nervous system plasticity gained from the use of a very short electrode array.

Mismatch negativity (MMN) as an index of central auditory system plasticity

The mismatch negativity (MMN) is an electric brain response which is automatically elicited by any discriminable change in a repetitive sound or sound pattern. When this change is made smaller in magnitude, the MMN is attenuated in amplitude, eventually vanishing at around the discrimination threshold. Therefore the MMN provides a unique objective index for a subject or patient's discrimination accuracy. Furthermore, with the MMN, these discrimination thresholds can be separately determined for all the different auditory attributes. Moreover, the individual's ability to discriminate complex sound stimuli and patterns such as different phonemes can also be measured by using the MMN.

Effects of Agonists and Antagonists of NMDA and ACh Receptors on Plasticity of Bat Auditory System Elicited by Fear Conditioning

In big brown bats, tone-specific plastic changes [best frequency (BF) shifts] of cortical and collicular neurons can be evoked by auditory fear conditioning, repetitive acoustic stimuli or cortical electric stimulation. It has been shown that acetylcholine (ACh) plays an important role in evoking large long-term cortical BF shifts. However, the role of N-methyl-d-aspartate (NMDA) receptors in evoking BF shifts has not yet been studied. We found 1) NMDA applied to the auditory cortex (AC) or inferior colliculus (IC) augmented the auditory responses, as ACh did, whereas 2-amino-5-phosphovalerate (APV), an antagonist of NMDA receptors, reduced the auditory responses, as atropine did; 2) although any of these four drugs did not evoke BF shifts, they influenced the development of the long-term cortical and short-term collicular BF shifts elicited by conditioning; 3) like ACh, NMDA augmented the cortical and collicular BF shifts regardless of whether it was applied to the AC or IC; 4) endogenous ACh of the AC and IC is necessary to produce the long-term cortical and short-term collicular BF shifts; 5) blockade of collicular NMDA receptors by APV abolished the development of the collicular BF shift and made the cortical BF shift small and short-term; 6) blockade of cortical NMDA receptors by APV reduced the cortical and collicular BF shifts and made the cortical BF shift short-term; and 7) conditioning with NMDA + atropine applied to the AC evoked the small, short-term cortical BF shift, whereas conditioning with APV + ACh applied to the AC evoked the small, but long-term cortical BF shift.

Central auditory system plasticity and aural rehabilitation of adults

Until recently, researchers used behavioral measures of identification and discrimination of speech and nonspeech stimuli to assess the effects of auditory deprivation, enhancement, and training. Recent advances in our ability to measure electrical activity in the auditory system in response to sound have made it possible for us to study how changes in auditory input (because of hearing loss, auditory input modification, or training) affect the function of the central auditory system. This article reviews the evidence of changes in the auditory cortex in mature animals and in humans with acquired sensorineural hearing loss as well as changes associated with auditory training in persons with normal hearing. The results of studies that measure psychoacoustic and speech-recognition performance of persons with hearing loss, with and without hearing aids, are interpreted within the framework of our new knowledge about plasticity of the auditory system. Applications of electrophysiologic techniques to hearing aid research and clinical practice are highlighted.

Seasonal Plasticity of Peripheral Auditory Frequency Sensitivity

Female midshipman fish (Porichthys notatus) use the auditory sense to detect and locate vocalizing males during the breeding season. Detection of conspecific vocal signals is essential to their reproductive success and can evoke strong phonotactic responses in gravid females but not in spent females that have released all of their eggs. Here, we test the hypothesis that seasonal variation in reproductive state affects the neurophysiological response properties of the peripheral auditory system in female midshipman fish. Iso-intensity responses of eighth nerve afferents from the sacculus, the main auditory end organ of the inner ear, to individual tones were measured for spike rate and vector strength (VS) of synchronization. Most auditory saccular units in reproductive, summer females showed robust temporal encoding up to 340 Hz, whereas nonreproductive winter females showed comparable encoding only up to 100 Hz. The dramatic upward shift in temporal encoding among summer fish was paralleled by increases in best frequency (BF), maximum evoked spike rate at BF, VS values at BF, and the percentage of units that showed significant VS to iso-intensity tones >140 Hz. Reproductive summer females were best suited to encode the higher harmonic components of male advertisement calls. This first demonstration of a natural cyclicity in peripheral auditory frequency sensitivity among vertebrates may represent, in this case, an adaptive plasticity of the female midshipman's auditory system to enhance the acquisition of auditory information needed for mate identification and localization during the breeding season.

Rapid development of cortical auditory evoked potentials after early cochlear implantation.

The aim of our research was to estimate the time course of development and plasticity of the human central auditory pathways following cochlear implantation. We recorded cortical auditory-evoked potentials in 3-year-old congenitally deaf children after they were fitted with cochlear implants. Immediately after implantation cortical response latencies resembled those of normal-hearing newborns. Over the next few months, the cortical evoked responses showed rapid changes in morphology and latency that resulted in age-appropriate latencies by 8 months after implantation. Overall, the development of cortical response latencies for the implanted children was more rapid than for their normal-hearing age-matched peers. Our results demonstrate a high degree of central auditory system plasticity during early human development.
