Asymptomatic Malaria Cases and Plasmodium Species among BaAka Pygmies in Central Africa.

Malaria is a significant health problem in Africa, primarily due to the Plasmodium falciparum species, but this is not the only etiological factor responsible for malaria on the continent. The goal of the present research was to describe asymptomatic malaria cases and to identify Plasmodium species responsible for malaria in the BaAka Pygmies, inhabitants of the Central African Republic (CAR). Screening was realised in the period of August-September 2021 among 308 people, including 74 children and 234 adults reporting to a healthcare facility in Monasao (southwest CAR), an area inhabited by a semi-nomadic tribe of BaAka Pygmies. The study consisted of two phases. Phase I, which was conducted in Africa, consisted of performing malaria rapid diagnostic tests (mRDTs), taking haemoglobin measurements and collecting blood samples onto Whatman FTA cards for molecular diagnostics. Phase II, which was conducted in Poland, involved molecular tests (RT-PCR) to confirm or rule out malaria infections and to identify Plasmodium species responsible for the infections. mRDTs detected Plasmodium infections in 50.3% of children and 17.1% of adults participating in the study, whereas RT-PCR assays yielded positive results for 59.5% children and 28.6% adults. Molecular tests detected multiple Plasmodium falciparum infections but also three infections with P. malariae, three with P. ovale and one with P. vivax. The obtained results have confirmed numerous asymptomatic Plasmodium infections among the BaAka Pygmies. The rates of asymptomatic malaria cases in adults were twice as high as those in children, which may be indicative of the gradual acquisition of protective immunity with age. The study findings have also demonstrated that although most cases of malaria in Africa are caused by P. falciparum, three other species are also present in the region.

Malaria infection and predictor factors among Chadian nomads’ children

BackgroundIn Chad, malaria remains a significant public health concern, particularly among nomadic populations. Geographical factors and the mobility of human populations have shown to be associated with the diversity of Plasmodium species. The study aims to describe the malaria prevalence among nomadic children and to investigate its associated factors.MethodsA cross-sectional study was conducted in February and October 2021 among nomadic communities in Chad. Blood sample were collected and tested from 187 Arab, Fulani and Dazagada nomadic children aged 3–59 months using malaria rapid diagnostic test (RDT). A structured electronic questionnaire was administered to their parents to collect information about the socio‑economic data. Malaria testing results were categorized according to the SD BIOLINE Malaria Ag Pf/Pan RDT procedures. Logistic regression analysis was used to determine key risk factors explaining the prevalence of malaria. STATA version IC 13 was used for statistical analysis.ResultsThe overall malaria prevalence in nomadic children was 24.60%, with 65.20% being Plasmodium falciparum species and 34.8% mixed species. Boys were twice as likely (COR = 1.83; 95% CI, 0.92–3.62; p = 0.083) to have malaria than girls. Children whose parents used to seek traditional drugs were five times more likely (AOR = 5.59; 95% CI, 1.40–22.30, p = 0.015) to have malaria than children whose parents used to seek health facilities. Children whose parents reported spending the last night under a mosquito net were one-fifth as likely (AOR = 0.17; 95% CI, 0.03–0.90, p = 0.037) to have malaria compared to children whose parents did not used a mosquito net. Furthermore, Daza children were seventeen times (1/0.06) less likely (AOR = 0.06; 95% CI, 0.01–0.70, p = 0.024) to have malaria than Fulani children and children from households piped water as the main source were seven times more likely (AOR = 7.05; 95% CI, 1.69–29.45; p = 0.007) to have malaria than those using surface water.ConclusionsMalaria remains a significant public health issue in the nomadic communities of Chad. Community education and sensitization programs within nomad communities are recommended to raise awareness about malaria transmission and control methods, particularly among those living in remote rural areas. The National Malaria Control Program (NMCP) should increase both the coverage and use of long-lasting insecticidal nets (LLINs) and seasonal malaria chemoprevention (SMC) in addition to promoting treatment-seeking behaviors in nomadic communities.

Is Brazil reaching malaria elimination? A time series analysis of malaria cases from 2011 to 2023.

In Brazil, 99% of malaria cases occur in the Amazon region, mainly caused by Plasmodium vivax (~83%) and Plasmodium falciparum (Pf) species. Aligned with the Sustainable Development Goals, Brazil aims to eliminate autochthonous malaria by 2035. This study aims to analyse epidemiological patterns of malaria in Brazil to discuss if Brazil is on track to meet malaria control targets. A time-series study was conducted analysing autochthonous malaria new infections notifications in the Brazilian Amazon region from 2011 until June 2023. Descriptive analyses were conducted, along with joinpoint regression and forecast models to verify trend and future behaviour. A total of 2,067,030 malaria cases were reported in the period. Trend analysis indicated a decreasing trend in all malaria infections since late 2017 (monthly reduction = 0.81%, p-value <0.05), while Pf infections have increased progressively since 2015 (monthly increase = 0.46%, p-value <0.05). Forecast models predict over 124,000 malaria cases in 2023 and over 96,000 cases in 2024. Predictions for Pf infections are around 23,900 cases in 2023 and 22,300 in 2024. Cases in indigenous population villages are predicted to reach 48,000 cases in 2023 and over 51,000 in 2024. In gold mining areas it is expected over 21,000 cases in 2023 and over 20.000 in 2024. Malaria elimination in Brazil has advanced over the last decade, but its speed has slowed. The country exhibits noteworthy advancements in the reduction of overall malaria cases. It is imperative, however, to proactively target specific issues such as the incidence raise among indigenous populations and in gold mining areas. Pf infections remain a persistent challenge to control in the country and may require novel measures for containment. Current government supporting actions towards combating illegal goldmining activities and protecting indigenous populations may help malaria control indicators for the following years.

Malaria today

Malaria, a parasitic disease the pathogen of which was discovered by Alphonse Laveran in 1880 in the blood of febrile patients, remains in 2022 the most frequent endemic disease in tropical and subtropical countries. In its latest "World Malaria Report" available in November 2021, the WHO deals in great detail with the data collected in the field in 2019-2020, their progression over the last 20 years, and the measures to be taken to try to better control this life-threatening endemic. The number of malaria cases is estimated at 232 million in 2019 in 87 endemic countries, down from 245 million in 2000. The WHO African Region alone accounts for 94% of cases and the most frequent and severe infections due to Plasmodium falciparum species. If children under the age of 5 are not treated promptly, they can die. Globally, the number of malaria deaths declined steadily over the period 2000-2019, from 897,000 in 2000 to 568,000 in 2019, with nearly 95% of deaths occurring in 31 countries, primarily in sub-Saharan Africa. In other WHO regions, including Southeast Asia, malaria deaths decreased by 74%, with 35,000 deaths in 2000 compared to 9,000 in 2019. Malaria can be controlled worldwide, and possibly eradicated, if public information campaigns are strengthened and sufficient funds are made available.

Long-read genome assembly and gene model annotations for the rodent malaria parasite Plasmodium yoelii 17XNL

Malaria causes >600 thousand fatalities each year, with most cases attributed to the human-infectious Plasmodium falciparum species. Many rodent-infectious Plasmodium species, like Plasmodium berghei and Plasmodium yoelii, have been used as model species that can expedite studies of this pathogen. P.yoelii is an especially good model for investigating the mosquito and liver stages of development because key attributes closely resemble those of P.falciparum. Because of its importance, in 2002 the 17XNL strain of P.yoelii was the first rodent malaria parasite to be sequenced. Although this was a breakthrough effort, the assembly consisted of >5000 contiguous sequences that adversely impacted the annotated gene models. While other rodent malaria parasite genomes have been sequenced and annotated since then, including the related P.yoelii 17X strain, the 17XNL strain has not. As a result, genomic data for 17X has become the de facto reference genome for the 17XNL strain while leaving open questions surrounding possible differences between the 17XNL and 17X genomes. In this work, we present a high-quality genome assembly for P.yoelii 17XNL using PacBio DNA sequencing. In addition, we use Nanopore and Illumina RNA sequencing of mixed blood stages to create complete gene models that include coding sequences, alternate isoforms, and UTR designations. A comparison of the 17X and this new 17XNL assembly revealed biologically meaningful differences between the strains due to the presence of coding sequence variants. Taken together, our work provides a new genomic framework for studies with this commonly used rodent malaria model species.

PCR-confirmed malaria among children presenting with a decreased level of consciousness in Angola: a prospective, observational study

BackgroundIn malaria-endemic areas, children presenting to hospitals with a decreased level of consciousness remain a diagnostic dilemma. The definition of cerebral malaria in a comatose child demands exclusion of other possible reasons, which requires in-depth investigations that are not easily available. The aim of this study was to investigate the frequency and clinical characteristics of PCR-confirmed malaria in a cohort of children with a decreased level of consciousness, look for potential features that would aid in differentiating children with malaria from those without, and assess the performance of traditional thick film microscopy against the cytb-qPCR-method.MethodsA total of 345 children aged 30 days–15 years old, presenting to Hospital Pediátrico David Bernardino in Luanda, Angola, with a decreased level of consciousness (Glasgow coma scale score < 15) were prospectively enrolled during 2014–2017. Malaria was defined as a positive cytb-qPCR result on any occasion in hospital. The clinical course and laboratory parameters were compared between children with malaria and those without. The performance of thick film microscopy was analysed against the PCR method.Results161 of 345 children (46.7%) had a positive malaria PCR test result. All cases were Plasmodium falciparum species, and 82.6% (133/161) fulfilled the WHO criteria for severe malaria. Overall, children with malaria presented to hospital with a shorter duration of symptoms and less convulsions pre-admission compared to those without malaria. The median GCS score on admission was 8, which did not differ between children with or without malaria. Clinical findings on admission were mostly similar across the whole cohort, but an infection focus outside the central nervous system was more common in malaria-negative children. Moreover, severe anaemia, thrombocytopenia, and high CRP levels occurred more frequently in children with malaria. The case fatality ratio was 28.5% (91/319) and did not differ between parasitaemic children and those without malaria, although parasitaemic children died sooner after hospital admission. When neurological sequelae were also considered, a positive malaria test was associated with a better outcome. The performance of thick film microscopy against PCR yielded a sensitivity of 96.8% and a specificity of 82.7%.ConclusionsIn this cohort of children with a decreased consciousness, the frequent presence of a malarial infection could not be judged from the clinical findings on admission, but the combination of profound aneamia, thrombocytopenia, and a high CRP level increased the odds of a positive malaria test result. Mortality remained high regardless of etiology, but malaria infection associated with fewer neurological deficits at discharge. Thick film microscopy performed well compared to the cytb-qPCR method.

A systematic self-study and analysis in prevalence of malaria across the world

Malaria is a febrile mosquito borne infectious disease which is caused by plasmodium. Malaria is one of the most deadliest and serious tropical disease ever witnessed by humanity. There are five species of plasmodium which are reported to infect humans. Out of the five species plasmodium falciparum has been reported to be the most deadliest form of plasmodium.A systemic self-study was planned to analyze and study the current prevalence of malaria globally. With the application of electronic databases we searched PubMed, Google Scholar, Web of Science, WHO website, Medline Plus, Health line &amp; Cleveland Clinic web which were published in English language. This systemic self-study have reviewed the facts which were published earlier to determine the current scenario by vast study of statistics and derivation of facts. Vast analysis and along with proper examination of data were made to evaluate with final conclusion.We observed and analyzed various surveys and studies conducted by World Health Organization which has stated that malaria have affected 90 countries and also territories in tropical and subtropical regions. In 2019 World Malaria Report documented over 96% of malarial death has occurred in African region. According to World Malarial Report, India also accounts for 3% of global burden of malaria. We also have observed in over 50% of estimated cases of malaria in India Plasmodium vivax have been the most infectious type of plasmodium. Irrespective of any country in the world India has by far the greatest plasmodium vivax incidences.We hereby conclude our systematic self-study and vast analysis by stating these following facts that malaria is a mosquito borne infectious disease which is highly infectious and is caused by a unicellular microorganism known as plasmodium. Malaria is one of the most serious and fatal tropical disease ever witnessed by humanity. We analysed several studies and found that World Health Organization have estimated globally about 241 million clinical cases of malaria followed by 6,27,000 deaths of people. We also derive the fact that most of this death related malaria occurred in the African region. In 2019 World Malarial Report also confirmed that 96% of global death has occurred from the African continent. We further studied that India have also accounted 3% of total global burden of malaria. We further inferred based on our observational analysis that over 50% of urban malaria in India is caused by plasmodium vivax. We studied that there are five types of malaria along with the prevalence and inferred that plasmodium falciparum is the most fatal form of malaria and have accounted over 95% of death caused due to malaria in African region at 100% in sub-Saharan African region. We hereby positively conclude our self-study on prevalence of malaria across the world by drawing a report given by World Health Organization stating that global mortality rate has significantly reduced by 90% in the end of 20th century.

Malaria among under-five children in rural communities of Al-Mahweet governorate, Yemen

BackgroundMalaria burden among under-five children living in endemic areas of Yemen is largely unknown due to the lack of community-based studies. Therefore, this study determined the prevalence and risk factors associated with falciparum malaria among under-five children in rural communities of Al-Mahweet governorate, Yemen.MethodsThis community-based, cross-sectional study recruited 400 under-five children from two rural districts of Al-Mahweet governorate in December 2019. Demographic characteristics (gender, age, education and occupation of the child’s parents, and household size) and risk factors associated with malaria were collected through interviews with children’s caregivers using a structured questionnaire. Finger-prick blood was screened for Plasmodium falciparum and non-falciparum species using rapid diagnostic tests (RDTs), and duplicate Giemsa-stained thick and thin blood films were examined for malaria parasites. The density of asexual P. falciparum stages was also estimated. Data were then analysed, and the agreement between the results of thick-film microscopy and RDTs for diagnosing falciparum malaria was assessed using the kappa index. Statistical significance was set at a P-value of < 0.05.ResultsPlasmodium falciparum was prevalent among 9.8% (95% CI 7.0–13.1) of under-five children in the rural communities of Al-Mahweet, with a median asexual parasite density of 763 ± 2606 parasites/μl of blood (range: 132–4280) and low-to-moderate parasitaemia levels. Approximately one-third of microscopy-confirmed cases were gametocyte carriers. Multivariable logistic regression analysis confirmed that age of three years or older (AOR = 5.6, 95% CI 1.6–19.8; P = 0.007), not sleeping under a mosquito net the previous night of the survey (AOR = 8.0, 95% CI 2.4–27.4; P = 0.001), sleeping outdoors at night (AOR = 4.4, 95% CI 2.0–10.0; P < 0.001), and absence of indoor residual spraying (IRS) during the last year (AOR = 4.2, 95% CI 1.9–9.4; P < 0.001) were the independent predictors of falciparum malaria among under-five children in the rural communities of Al-Mahweet. The observed percentage agreement between thick-film microscopy and RDTs was 98.5%, with a very good agreement (k-index = 0.9) between the two methods for falciparum malaria diagnosis that was statistically significant.ConclusionApproximately one in ten under-five children in rural communities of Al-Mahweet is infected with P. falciparum based on microscopy and RDTs. Age of three years or older, not sleeping under mosquito nets, sleeping outdoors at night and absence of IRS can independently predict falciparum malaria among them. The very good agreement between thick-film microscopy and RDTs for diagnosing falciparum malaria in children supports the usefulness of using RDTs in such resource-limited rural communities.

A Case of Latent Plasmodium Falciparum Malaria in a Patient With Coexisting Systemic Lupus Erythematosus (SLE) and Neuromyelitis Optica Spectrum Disorder (NMOSD)

Malaria is a global health concern with high morbidity and mortality. It is often attributed to the Plasmodium (P.) falciparum species, particularly in sub-Saharan Africa, and it normally has an incubation period of seven to 14 days. Dormant disease secondary to P. vivax and P. ovale is well-reported, yet only a handful of cases report dormant malaria secondary to P. falciparum. Even though malaria is significantly less common in the United States in comparison to other parts of the world, it is still a growing concern given international travel from endemic regions and a growing immunocompromised population. Here, we present a case of Plasmodium falciparum malaria in a patient with systemic lupus erythematosus (SLE) with neuromyelitis optica spectrum disorder (NMOSD)and renal transplant without travel to sub-Saharan Africa in 10 years.

Co-infection of the four major Plasmodium species: Effects on densities and gametocyte carriage.

Co-infection of the four major species of human malaria parasite Plasmodium falciparum (Pf), P. vivax (Pv), P. malariae (Pm), and P. ovale sp. (Po) is regularly observed, but there is limited understanding of between-species interactions. In particular, little is known about the effects of multiple Plasmodium species co-infections on gametocyte production. We developed molecular assays for detecting asexual and gametocyte stages of Pf, Pv, Pm, and Po. This is the first description of molecular diagnostics for Pm and Po gametocytes. These assays were implemented in a unique epidemiological setting in Papua New Guinea with sympatric transmission of all four Plasmodium species permitting a comprehensive investigation of species interactions. The observed frequency of Pf-Pv co-infection for asexual parasites (14.7%) was higher than expected from individual prevalence rates (23.8%Pf x 47.4%Pv = 11.3%). The observed frequency of co-infection with Pf and Pv gametocytes (4.6%) was higher than expected from individual prevalence rates (13.1%Pf x 28.2%Pv = 3.7%). The excess risk of co-infection was 1.38 (95% confidence interval (CI): 1.09, 1.67) for all parasites and 1.37 (95% CI: 0.95, 1.79) for gametocytes. This excess co-infection risk was partially attributable to malaria infections clustering in some villages. Pf-Pv-Pm triple infections were four times more frequent than expected by chance alone, which could not be fully explained by infections clustering in highly exposed individuals. The effect of co-infection on parasite density was analyzed by systematic comparison of all pairwise interactions. This revealed a significant 6.57-fold increase of Pm density when co-infected with Pf. Pm gametocytemia also increased with Pf co-infection. Heterogeneity in exposure to mosquitoes is a key epidemiological driver of Plasmodium co-infection. Among the four co-circulating parasites, Pm benefitted most from co-infection with other species. Beyond this, no general prevailing pattern of suppression or facilitation was identified in pairwise analysis of gametocytemia and parasitemia of the four species. Repurposing of MitoTam: Novel Anti-Cancer Drug Candidate Exhibits Potent Activity against Major Protozoan and Fungal Pathogens.

ABSTRACTMany of the currently available anti-parasitic and anti-fungal frontline drugs have severe limitations, including adverse side effects, complex administration, and increasing occurrence of resistance. The discovery and development of new therapeutic agents is a costly and lengthy process. Therefore, repurposing drugs with already established clinical application offers an attractive, fast-track approach for novel treatment options. In this study, we show that the anti-cancer drug candidate MitoTam, a mitochondria-targeted analog of tamoxifen, efficiently eliminates a wide range of evolutionarily distinct pathogens in vitro, including pathogenic fungi, Plasmodium falciparum, and several species of trypanosomatid parasites, causative agents of debilitating neglected tropical diseases. MitoTam treatment was also effective in vivo and significantly reduced parasitemia of two medically important parasites, Leishmania mexicana and Trypanosoma brucei, in their respective animal infection models. Functional analysis in the bloodstream form of T. brucei showed that MitoTam rapidly altered mitochondrial functions, particularly affecting cellular respiration, lowering ATP levels, and dissipating mitochondrial membrane potential. Our data suggest that the mode of action of MitoTam involves disruption of the inner mitochondrial membrane, leading to rapid organelle depolarization and cell death. Altogether, MitoTam is an excellent candidate drug against several important pathogens, for which there are no efficient therapies and for which drug development is not a priority.

SEVERE PLASMODIUM VIVAX MALARIA IN AN ADOLESCENT

Vivax malaria is in general described and considered as benign as it less likely causes severe illness, compared to malaria caused by Plasmodium falciparum (P.falciparum)species. Of late, there have been increasing evidences of Plasmodium Vivax (P.vivax) too causing severe disease and leading to poor outcomes.. In Vitro Study: Antimalarial Activity of Rivet Sea Cucumber Extract (Holothuria atra) With Ethyl Acetate Solvent Against Plasmodium falciparum

&#x0D; Abstract &#x0D; Indonesia is experiencing a decreased cure rate of malaria caused by the species Plasmodium falciparum. Alternative medicine, in this case, uses marine biota, namely lollyfish (Holothuria atra), which has a lot of active compound content; the purpose uses the biota because many reports that there is a failure of malaria treatment therapy due to Artemisinin-based Combination Therapy (ACT) resistance. The purpose of this study was to find out the antimalarial activity of Holothuria atra extract with ethyl acetate solvent at dose 0.01; 0.1; 1; 10; and 100 Ug/ml against Plasmodium falciparum in vitro. Extract lollyfish using the maceration method. The lollyfish powder will be dissolved with ethyl acetate solvent for 24 hours. Antimalarial activity testing is measured through 4 parameters, namely the measurement of parasitemia rates, % parasite growth, % inhibitory rate, and IC50. Antimalarial activity with ethyl acetate solvent belongs to a very active category with an IC50 value of 1.52 µg/mL. These results can kill the P. falciparum parasite with a percentage that reaches 0%. A decrease in the percentage of growth occurs when the dose given is too low so that the parasite can grow and survive. The result of higher concentrations of extracts will have a higher percentage of resistance to the growth of parasites. The potential for antimalarial activity in lollyfish extract is influenced by the presence of active content such as alkaloids, flavonoids, catechins, and pyrogallol owned by marine biota animals. Malaria time series in the extra-Amazon region of Brazil: epidemiological scenario and a two-year prediction model

BackgroundIn Brazil, malaria is caused mainly by the Plasmodium vivax and Plasmodium falciparum species. Its transmission occurs in endemic and non-endemic areas. Malaria geography in Brazil has retracted and is now concentrated in the North region. The Brazilian Amazon region accounts for 99% of Brazil's cases. Brazil’s extra-Amazon region has a high frequency of imported cases and in 2019 presented a mortality rate 123 times higher than the Amazon region. Extra-Amazon cases present risks of reintroduction. This study aims to characterize the epidemiological scenario for malaria in the extra-Amazon region of Brazil from 2011 to 2020 with a two-year forecast.MethodsTime-series study with description of malaria cases and deaths registered in Brazilian extra-Amazon region from 2011 to 2020. Public data from the Notifiable Diseases Information System (Sinan) and the Mortality Information System (SIM) were used. Descriptive analysis, incidence, and notification rates were calculated. Flow charts analysed the flux between Places of Probable Infection (PI) and places of notification. The prediction model utilized a multiplicative Holt-winters model for trend and seasonality components.ResultsA total of 6849 cases were registered. Cases were predominantly white males with 9 to 11 years of education, mostly between 30 and 39 years old. Imported cases accounted for 78.9% of cases. Most frequent occupations for imported cases are related to travelling and tourism activities. Among autochthonous cases, there is a higher frequency of agriculture and domestic economic activities. In the period there were 118 deaths due to malaria, of which 34.7% were caused by P. falciparum infections and 48.3% were not specified. The most intense flows of imported cases are from Amazonas and Rondônia to São Paulo, Rio de Janeiro, and Paraná. The prediction estimates around 611 cases for each of the following two years.ConclusionThe time series allows a vast epidemiological visualization with a short-term prediction analysis that supports public health planning. Government actions need to be better directed in the extra-Amazon region so the objective of eliminating malaria in Brazil is achieved. Carrying out quality assessments for information systems and qualifying personnel is advisable. Malaria outside the Amazon region is mainly due to imported cases and delay in diagnosis is associated with a higher fatality rate. Better strategies to diagnose and treat suspected cases can lead to lower risk of deaths and local outbreaks that will be important for achieving malaria elimination in Brazil.

Characterization of Human-malarial Parasite Species based on DHFR and GST Targets Resulting in Changes in Anti-malarial Drug Binding Conformations.

In this study, we focused primarily on three anti-malarial drugs, namely chloroquine, mefloquine, and proguanil, and these were tested against two malarial targets DHFR and GST. The species Plasmodium falciparum, Plasmodium malariae, Plasmodium ovale, Plasmodium vivax were used for the study. The purpose of this study was to determine the sequence and structural similarity of the proteins DHFR and GST among four Plasmodium species as well as to discover the in silico interactions with the aforementioned drug candidates. Bioinformatics databases, such as PDB, UniProt, DrugBank, PubChem, and tools, and software like Phyre 2.0, Clustal O (1.2.4), AutoDock 4, AutoDock Vina, and Discovery Studio Visualizer were used to determine the evolutionary significance of the Plasmodium species. The variations showed a difference in the binding patterns of drugs with our target proteins. Our finding reveals the Plasmodium spp divergence or convergence as well as the structural and sequential similarity or dissimilarity features. Our result suggests that due to the deviation in the sequences and structures, variations in protein-drug binding patterns have emerged.

Species Composition and Plasmodium falciparum Infection Rates of Anopheles gambiae s.l. Mosquitoes in Six Localities of Kwara State, North Central, Nigeria

Entomological data gathering is essential for monitoring malaria vector disease risks and selection of appropriate interventions for the protection of exposed human populations. This study assessed the relative abundance, species composition, and sporozoite infection rates of indoor resting An. gambiae s.l. malaria vectors in six communities across 3 Local Government Areas in Kwara State, Nigeria. Total number of mosquitoes collected by Pyrethrum Spray Catch method over a period of eighth months were correlated with rainfall values in the area. Plasmodium falciparum sporozoite infection rates and sibling species identification of collected An. gambiae s.l. mosquito samples were determined by ELISA and PCR respectively. Results showed a positive correlation (r = 0.639, p = 0.08) between rainfall and numbers of Anopheles mosquitoes in the study areas. The overall composition of the An. gambiae s.l sibling species in the collected samples from all the six communities showed the predominance of An. gambiae s.s 298 (75.3%) compared to An. coluzzii 94(23.7%) and An. arabiensis 4(1.0%). However, the sporozoite infection rate of An. coluzzii (22.3%) was higher compared to An. gambiae s.s (12.8%) and An. arabiensis (0%). Mean numbers of An. gambiae s.l mosquitoes were significantly higher in Ilorin west LGA compared to Asa (F = 17.81, P &lt; 0.001) and Ilorin East LGAs (F = 22.81, P &lt; 0.001). Sporozoite rates of both An. gambiae s.s and An. coluzzii sibling species were higher in Ilorin West communities (Aiyede 21%, Ogundele 32%) compared to Asa (Idi Emi 11.1%, Lasoju 5.1%) and Ilorin East (Oke Oyi 2.4%, Ote-efan 0%) communities. Prevalence of sporozoite-infected An. gambiae s.s and An. coluzzii indoors highlight the need for effective insecticide treated bed-nets interventions to protect the residents from malaria risks. Higher numbers of An. coluzzii in the swampy rice marshed Ilorin West LGA communities require larval source management as an additional strategy for effective malaria vector control.

The role of hematological parameters in differentiating Plasmodium falciparum and others – a study from Somalia

Introduction and aim. Accurate identification of Plasmodium species is important because of the differences in their treatment. We aimed to investigate the role of hematological and biochemical parameters in the differentiation of Plasmodium falciparum and other plasmodium species. Material and methods. This is a retrospective study. Patients admitted to the emergency department with signs and symptoms of malaria were included into the study. Patients with malaria were grouped as P. falciparum and others. Hematological parameters of two groups were compared by univariate and multivariate analysis. Statistical analysis was performed using the Jamovi. Results. A total of 107 patients were included in the study. According to univariant and multivariant analysis there was no difference in between two groups in the terms of blood urea nitrogen, aspartate aminotransferase, total bilirubin, hemoglobin, hematocrit, white blood cell count, platelet count, and mean platelet volume (in univariate analysis p values were 0.029, 0.011, 0.019, 0.171, 0.870, 0.307, 0.042, and 0.276, respectively and in multivariate analysis p values for blood urea nitrogen, aspartate aminotransferase, total bilirubin, hemoglobin, and platelet count were 0.100, 0.535, 0.328, and 0.213, respectively). Conclusion. The investigated hematological and biochemical parameters were found to be not valuable in predicting type of malaria. On the other hand, we recommend confirming the results of our study with larger samples and multicenter studies.

Pre and post flooding malaria parasitemia in gravid women, South East, Nigeria

The effect of global change on the incidence of vector borne diseases including malaria is of great importance. Malaria has been regarded as one of the most sensitive disease that responds fast to climate change. Pregnant women tend to have reduced immunity are more vulnerable to vector borne diseases such as malaria especially with climate change like flooding where these vectors borne diseases are endemic. To measure malaria parasitaemia in gravid women pre and post flooding and also to determine the relationship between malaria and seasonal flooding in South east Nigeria. This was a hospital-based cross-sectional study involving pregnant women aged 15- 45 years attending the antenatal clinics of two maternity centres in a rural community South east Nigeria. Malaria was determined using the thick and thick blood films. Plasmodium falciparum species was examined in this study. Chi-square was used to assess the relationship between malaria and seasonal flooding. One Hundred and fifty pregnant women were recruited for the study. The prevalence of malaria in the gravid women pre and post flooding were 60.00% and 65.30% respectively. Malaria parasite was highest in the gravid women aged 28-31 years and the primigravids. There was no statistical difference between malaria and parity. The mean parasite density in the gravid women was significantly higher post flooding than pre flooding with p-value of 0.001. There was no significant relationship between malaria parasite in gravid women and the periods of investigation. The prevalence of malaria parasite among gravid women in the study area is high regardless of the seasonal flooding. Hence, the need for adherence to malaria prophylaxis protocol by the health care professionals and increase on community health education on malaria preventive strategies.

Messenger RNA expressing PfCSP induces functional, protective immune responses against malaria in mice

Human malaria affects the vast majority of the world’s population with the Plasmodium falciparum species causing the highest rates of morbidity and mortality. With no licensed vaccine and leading candidates achieving suboptimal protection in the field, the need for an effective immunoprophylactic option continues to motivate the malaria research community to explore alternative technologies. Recent advances in the mRNA discipline have elevated the long-neglected platform to the forefront of infectious disease research. As the immunodominant coat protein of the invasive stage of the malaria parasite, circumsporozoite protein (PfCSP) was selected as the antigen of choice to assess the immunogenic and protective potential of an mRNA malaria vaccine. In mammalian cell transfection experiments, PfCSP mRNA was well expressed and cell associated. In the transition to an in vivo murine model, lipid nanoparticle (LNP) encapsulation was applied to protect and deliver the mRNA to the cell translation machinery and supply adjuvant activity. The immunogenic effect of an array of factors was explored, such as formulation, dose, number, and interval of immunizations. PfCSP mRNA-LNP achieved sterile protection against infection with two P. berghei PfCSP transgenic parasite strains, with mRNA dose and vaccination interval having a greater effect on outcome. This investigation serves as the assessment of pre-erythrocytic malaria, PfCSP mRNA vaccine candidate resulting in sterile protection, with numerous factors affecting protective efficacy, making it a compelling candidate for further investigation.

Fibrinogen and C-Reactive Protein Significance in Children Infected by Plasmodium falciparum Species in Enugu, Enugu State, Nigeria

Malaria accounts for a considerable mortality and morbidity rate with children bearing the greatest burden. The study investigated fibrinogen and C-reactive protein (CRP) value alterations in children infected by Plasmodium falciparum (P. falciparum) species. A case control study with a total of ninety-five microscopically confirmed P. falciparum malaria infected children and fifty apparently healthy age and gender matched controls from Enugu State University Teaching Hospital, Parklane, Wesley Specialist Hospital and Akpugo Community Health Centre, Enugu were recruited for the study. Fibrinogen level was determined by clauss clotting time method using sodium citrated plasma. Giemsa stained thick and thin blood film was used for parasite identification and calculation of parasite density. Serum CRP values was determined by immunoturbidimetric method. Fibrinogen levels were significantly increased (p &lt; 0.05) in P. falciparum infected children (324.03 + 59.87) mg/dl as compared to the control (224.74 + 34.88) mg/dl. Parasite density showed a weak positive correlation between fibrinogen (p &lt; 0.01, r = 0.461) and CRP (p &lt; 0.01, r = 0.232). CRP was significantly increased (p &lt; 0.05) in P. falciparum malaria infected children (21.52 + 35.59) mg/l as compared to the control (2.43 + 0.97) mg/l. In conclusion, P. falciparum malaria infection demonstrated a significant impact on fibrinogen and CRP.
