Feed restriction of lactating cows triggers acute downregulation of mammary mTOR signaling and chronic reduction of mammary epithelial mass

While there is generally no consensus about how nutrients determine milk synthesis in the mammary gland, it is likely that the mechanistic target of rapamycin complex 1 (mTORC1) plays a role as a key integrator of nutritional and mitogenic signals that can influence a multitude of catabolic and anabolic pathways. The objectives of this study were to evaluate acute changes (<24 h) in translational signaling, in addition to chronic changes (14 d) in mammary gland structure and composition, in response to a severe feed restriction. Fourteen lactating Holstein dairy cows were assigned to either ad libitum feeding (n = 7), or a restricted feeding program (n = 7). Feed-restricted cows had feed removed after the evening milking on d 0. Mammary biopsies and blood samples were collected 16 h after feed removal, after which cows in the restricted group were fed 60% of their previously observed ad lib intake for the remainder of the study. On d 14, animals were sacrificed and mammary glands dissected. In response to feed removal, an acute increase in plasma nonesterified fatty acid concentration was observed, concurrent to a decrease in milk yield. In mammary tissue, we observed downregulation of the mTORC1-S6K1 signaling cascade, in addition to reductions in mRNA expression of markers of protein synthesis, endoplasmic reticulum biogenesis, and cell turnover (i.e., transcripts associated with apoptosis or cell proliferation). During the 14 d of restricted feeding, animals underwent homeorhetic adaptation to 40% lower nutrient intake, achieving a new setpoint of 14% reduced milk yield with 18% and 29% smaller mammary secretory tissue dry matter and crude protein masses, respectively. On d 14, no treatment differences were observed in markers of protein synthesis or mammary cell turnover evaluated using gene transcripts and immunohistochemical staining. These findings implicate mTORC1-S6K1 in the early phase of the adaptation of the mammary gland's capacity for milk synthesis in response to changes in nutrient supply. Additionally, changes in rates of mammary cell turnover may be transient in nature, returning to basal levels following brief alterations that have sustained effects.

Fat supplement for dairy cows during early lactation – potentials, challenges, and risks – a meta-analysis

ABSTRACT The most critical challenge in early lactation is to meet the increasing energy demand for milk production, which results in a negative energy balance (EB). The objective of this study was to evaluate the productive performance and plasma metabolite responses of early lactating dairy cows fed diets supplemented with increasing levels of different fat sources. Diets were categorized as diets supplemented with saturated fatty acid rich fat sources (SFAR), with Ca-soaps of fatty acids (CaFA), and with polyunsaturated fatty acid rich fat sources (PUFAR). There was an interaction between crude fat level and source for dry matter intake (DMI); each percentage-unit increase reduced DMI by 0.38 and 0.77 kg/d in CaFA and PUFAR, respectively, and increased DMI by 0.55 kg/d in SFAR. Corrected milk yield increased by 0.44 kg/d for each percentage-unit increase in crude fat level. Plasma non-esterified fatty acid (NEFA) concentration increased with 0.04 mmol/L per percentage-unit increase. In conclusion, the increased crude fat level increased corrected milk yield. Fat supplementation in early lactation does not seem to cause negative effects on productive performance or metabolism, and despite a small increase, both plasma NEFA and β-hydroxybutyrate (BHB) concentrations stayed below the threshold for incidence of subclinical ketosis.

85 Evaluation of a Saccharomyces cerevisiae fermentation product on performance, plasma metabolites, rumen parameters, and fecal pathogen shedding in feedlot steers

Abstract The objective of the experiment was to evaluate a commercial Saccharomyces cerevisiae fermentation product (SCFP; NaturSafe, Dimond V, Cedar Rapids, IA) on performance, plasma metabolites, rumen parameters, and fecal pathogen shedding in feedlot steers. Crossbred steers (n = 61; 271 ± 48.1 kg) were sourced from 3 different research units at Oklahoma State University and assigned to 1 of 3 experimental treatments in a randomized complete block design. Steers were group housed in pens equipped with an automated individual feed intake system. Treatments consisted of a negative control (CON), CON with tylosin phosphate (TYL; 9.9 mg · kg-1 of diet DM), or CON with SCFP (1.63 g · kg-1 of diet DM). Body weight and feed efficiency did not differ among treatments (P ≥ 0.12). However, steers fed SCFP tended to have greater ADG from d 35 to 56 (P = 0.10). In addition, steers fed SCFP exhibited increased DMI from d 56 to 112 (P = 0.03), a tendency for increased DMI from d 112 to final (P = 0.07), and increased DMI overall (P = 0.05). There was no treatment × time interaction for pH, pH change, or lactate concentrations in rumen fluid (P ≥ 0.11), but all were impacted by time (P &amp;lt; 0.01). A treatment × time interaction (P &amp;lt; 0.01) was detected for fecal pH and fecal pH change, with the pH being greater in TYL fed steers on d 56 and in CON fed steers on the final day. Steers that consumed CON had a reduced fecal pH (P &amp;lt; 0.01) and steers consuming SCFP tended to have a reduced fecal pH (P = 0.08) from d 35 to 56 compared with TYL fed steers, while both CON and SCFP fed steers experienced an increase in fecal pH from d 56 to 112 (P &amp;lt; 0.01). A time effect was detected (P ≤ 0.04) for plasma glucose, lactate, urea nitrogen, and non-esterified fatty acid concentrations among treatment groups. A treatment × time interaction was observed (P &amp;lt; 0.02) for stx2 genes of Shiga toxin producing Escherichia coli (STEC) in individual fecal samples where SCFP and TYL fed steers tended to exhibit an increased concentration compared with CON fed steers on d 14, while on d 190 CON and SCFP fed steers exhibited an increased concentration compared with TYL fed steers. Overall, STEC, E. coli O157:H7, and Salmonella concentrations were all impacted by time (P &amp;lt; 0.001). This experiment suggests that SCFP could be utilized as a potential alternative to TYL in feedlot cattle without adversely affecting cattle performance. However, SCFP did not impact pathogen shedding. In addition, CON fed steers performed similarly to both SCFP and TYL fed steers.

The Effect of Supplementing White Sweet Lupine Grain Meal (Lupinus albus L.) on Blood and Milk Metabolites, Milk Yield, and Quality of Dairy Cows in Ethiopia

Feed is the cornerstone of dairy production, serving as a crucial input for sustainable milk production. This trial aimed to investigate the effect of white sweet lupine grain meal inclusion on feed intake, blood and milk metabolites, milk yield, and quality. The experimental cows had a parity of 2±0.71 (mean ± SD), a daily milk yield of 12.72 ± 0.9, and an average body weight of 412.6 ± 21.5 kg. The varying proportions of treatment diets were: 75, 50, 25 and 0% (T1, T2, T3 and T4) treatment groups respectively. Data collected during the study were subjected to analysis of variance to determine significant differences. Cows fed diet T1 showed a better dry matter (10.39 kg/day), organic matter (7.31 kg/day), nitrogen (2.42 kg/day), and NDF intakes (3.75 kg/day) that had significant difference on (p&lt;0.05). When lupine inclusions dropped from 75 to 50 and even to 25%, the N digestibility decreased from 76.63, 70.66, and 65.6%, respectively, and the digestibility of N in all treatment diets significantly decreased (p &lt; 0.01). Once more, cows fed T1 diet had the highest (p&lt;0.01) plasma glucose concentration (116.81 mg/dl) among the four experimental groups. Cows fed T2 and T4 diets showed significant variation at (p&lt;0.05) in plasma glucose levels, with values of 108.7 and 110.16 mg/dl, respectively. Cows fed T1 had relatively lower plasma beta-Hydroxybutyric acid and non-esterified fatty acid concentrations (p&lt;0.05) than others, with mean values of 3.23 and 1.28 mg/dl respectively. The partial inclusion of white sweet lupine grain meal up to the level of 75% is highly suggested as a better alternative feed source.

Chromium propionate in turkeys: effects on insulin sensitivity

The objective of this study was to evaluate the effects of dietary chromium (Cr), as Cr propionate (Cr Prop), on measures of insulin sensitivity in turkeys. Plasma glucose and nonesterified fatty acid (NEFA), and liver glycogen concentrations were used as indicators of insulin sensitivity. One-day-old Nicholas Large White female poults (n=336) were randomly assigned to dietary treatments consisting of 0 (control), 0.2, 0.4, or 0.6 mg supplemental Cr/kg diet. Each treatment consisted of 12 replicate cages with 7 turkeys per cage. Final BW were taken on d 34, and on d 35 two birds from each cage were sampled for plasma glucose and NEFA, and liver glycogen determination at the initiation (fed state) and termination (fasted state) of a 24-h fast. Following a 24-h fast, 2 turkeys per cage were refed (refed state) their treatment diet for 4 h, and then harvested. Feed/gain and ADG did not differ between control and Cr-supplemented turkeys over the 34-d study, but feed intake tended (P=0.071) to be greater for controls than turkeys receiving 0.4 mg Cr/kg diet. Fed turkeys had greater plasma glucose (P=0.002) and liver glycogen (P=0.001) concentrations, and lower (P=0.001) NEFA concentrations than fasted birds. Turkeys refed after fasting had greater (P=0.001) plasma glucose and liver glycogen concentrations, and lower (P=0.001) plasma NEFA levels than fed turkeys. Liver glycogen and plasma NEFA concentrations did not differ among control and Cr-supplemented birds in the fed, fasted, or refed state. Plasma glucose concentrations were not affected by treatment in fed or fasted turkeys. Turkeys supplemented with 0.2 or 0.4 mg Cr/kg and refed after fasting had lower (quadratic, P=0.038) plasma glucose concentrations than controls. Plasma glucose concentrations in refed birds did not differ among Cr-supplemented turkeys. The lower plasma glucose concentration in Cr-supplemented turkeys following refeeding is consistent with Cr enhancing insulin sensitivity.

Dynamic fecal microenvironment properties enable predictions and understanding of peripartum blood oxidative status and nonesterified fatty acids in dairy cows

The transition period in dairy cows is a critical stage and peripartum oxidative status, negative energy balance (NEB), and inflammation are highly prevalent. Fecal microbial metabolism is closely associated with blood oxidative status and nonesterified fatty acids (NEFA) levels. Here, we investigated dynamic changes in total oxidative status markers and NEFA in blood, fecal microbiome, and metabolome of 30 dairy cows during transition (-21, -7, +7, +21 d relative to calving). Then the Bayesian network and 9 machine-learning algorithms were applied to dismantle their relationship. Our results show that the oxidative status indicator (OSI) of -21, -7, +7 d was higher than +21 d. The plasma concentration of NEFA peaked on +7 d. For fecal microenvironment, a decline in bacterial α diversity was observed at postpartum and in bacterial interactions at +7 d. Conversely, microbial metabolites involved in carbohydrate, lipid, and energy metabolism increased on +7 d. A correlation analysis revealed that 11 and 10 microbial metabolites contributed to OSI and NEFA variations, respectively (arc strength >0.5). The support vector machine (SVM) radial model showed the highest average predictive accuracy (100% and 88.9% in the test and external data sets) for OSI using 1 metabolite and 3 microbiota. The SVM radial model also showed the highest average diagnostic accuracy (100% and 91% in the test and external data sets) for NEFA with 2 metabolites and 3 microbiota. Our results reveal a relationship between variation in the fecal microenvironment and indicators of oxidative status, NEB, and inflammation, which provide a theoretical basis for the prevention and precise regulation of peripartum oxidative status and NEB.

Short communication: Predicting blood plasma non-esterified fatty acid and beta-hydroxybutyrate concentrations from cow milk-addressing systematic issues in modelling.

Negative energy status in early lactation is linked to a variety of metabolic disorders, reduced fertility, and decreased milk production. To improve the energy status of cows by breeding and management, the identification of negative energy status is crucial. While biomarkers such as non-esterified fatty acid (NEFA) concentration and beta-hydroxybutyrate (BHB) in blood plasma could be used to identify a negative energy state, measuring them directly from blood is both invasive and expensive. In this work, we developed prediction equations for blood plasma NEFA and BHB levels based on mid-IR spectral measurements of milk. The models were fitted using partial least squares regression and evaluated using both cross-validation and independent-herd validation. A total of 3183 spectral records from 606 lactations originating from three different herds were utilised. R2 values of 0.53 (RMSE=0.206mmol/l, RMSE of cross-validation (RMSECV) 0.217mmol/l) for NEFA and 0.63 (RMSE=0.326mmol/l, RMSECV=0.353mmol/l) for BHB were obtained. Furthermore, relatively similar prediction accuracies were found for BHB (RMSE of prediction (RMSEP) 0.411mmol/l and 0.422mmol/l) and NEFA (RMSEP=0.186mmol/l and 0.221mmol/l) when model training was done using two herds and validated on the third herd. The results from the model fits confirm that it is possible to build blood plasma BHB and NEFA models based on mid-IR spectra that are sufficiently accurate for practical use.

Effects of the timing of protein infusion on the daily rhythms of milk synthesis and plasma hormones and metabolites in dairy cows

Milk synthesis exhibits a daily rhythm that is modified by the timing of feed intake. However, it is unknown how specific nutrients entrain this daily rhythm. Amino acids have an important role in milk synthesis, and may have a role in entrainment of mammary circadian rhythms. The objective of this study was to determine the effects of intestinally absorbed protein on daily rhythms of milk and milk component synthesis and key plasma hormones and metabolites. Nine lactating Holstein cows were assigned to 1 of 3 treatment sequences in a 3 × 3 Latin square. Treatments included abomasal infusions of 500 g/d of sodium caseinate either continuously throughout the day (CON), for 8 h/d from 0900 to 1700 h (DAY), or for 8 h/d from 2100 to 0500 h (NGT). Cows were milked every 6 h during the final 8 d of each period. A 24-h rhythm was fit to data using cosine analysis and the amplitude and acrophase were determined. Night infusion of protein decreased the daily milk yield and milk protein yield by 8.2% and 9.2%, respectively. Milk fat yield was increased 5.5% by DAY and milk fat concentration was increased 8.8% by NGT. Milk yield exhibited a daily rhythm in all treatments, with NGT increasing the amplitude of the daily rhythm 33% compared with CON. Milk fat concentration fit a daily rhythm in CON and NGT, but not DAY, whereas milk protein concentration fit a daily rhythm in CON and DAY, but not NGT. Moreover, DAY abolished the daily rhythm of plasma glucose concentration, but induced rhythms of plasma insulin and nonesterified fatty acid concentrations. Results suggest that feeding increased protein levels during the early part of the day may increase milk fat yield and modify energy metabolism through increased daily variation in insulin-stimulated lipid release, but additional research focused on feeding multiple diets across the day is required.

Male fetuses negatively affect the vitality of the litter and the dam's metabolic and physiological state in multifetal pregnant ewe.

In sheep, ~30% of fetuses do not survive till parturition, and 17.7% of the multifetal pregnancies experience partial litter loss (PLL). In humans, multifetal pregnancies are associated with a higher risk of perinatal mortality. Therefore, the objectives were to examine the association between partial litter loss, fetal sex, dam's metabolic and physiological state, and pregnancy outcome in multifetal pregnant ewes. The study includes two parts. The first was a retrospective study, in which we analyzed data of 675 lambings and examined the PLL incidence according to male ratio (MR) for all litter sizes (range 2-6). Lambings were categorized as having a low male ratio (LMR; <50% males) or a high male ratio (LMR; >50% males). In the second part, we monitored 24 ewes from 80 to 138 days in pregnancy every 10 days, and then daily until lambing, by ultrasound scanning for maternal heart rate (HR), and Doppler ultrasound for litter vitality. Blood samples were taken from dams on the days of scanning. Male ratio strongly affected PLL, where the general survival rate (for all lambings) was reduced from 90% in LMR lambings to 85% in HMR lambings. The odds ratio for PLL in HMR vs. LMR litters was 1.82. Birth body weight and the survival rate of female was higher in LMR than HMR lambings, with no differences for male lambs in both parameters. In the second part, dams' HR during the last trimester was 9.4% higher in LMR than in HMR pregnancies, with no differences in fetuses' HR. The plasma glucose and insulin concentrations were not significantly different between groups, but plasma β-hydroxybutyrate and nonesterified fatty acid concentrations were, respectively, 31% and 20% lower in HMR vs. LMR ewes. In conclusion, male fetuses negatively affect pregnancy outcomes and influence dams' metabolic and physiological state in sheep.

Longitudinal Data to Assess Relationships among Plasma Folate, Vitamin B12, Non-esterified Fatty Acid, and β-Hydroxybutyrate Concentrations of Holstein Cows during the Transition Period.

It is well established that the plasma metabolite profile changes during metabolic dysfunction, such as elevated non-esterified fatty acid (NEFA) release when body reserve mobilization is excessive in early lactation cows. Relationships between changes in plasma concentrations of metabolites caused by a metabolic impairment and the status of vitamins, such as folates and vitamin B12, have barely been studied in cattle. This study was undertaken to assess relationships between peripartum plasma concentrations of folates, vitamin B12, NEFA, and beta-hydroxybutyrate (BHB). Longitudinal data of 48 multiparous Holstein cows from 5 studies were taken from days -14 to 21 relative to calving. Blood samples were taken weekly before calving and either twice or thrice per week postpartum, and plasma was analyzed for folate, vitamin B12, NEFA, and BHB concentrations. Postpartum plasma NEFA and BHB concentrations were negatively related to plasma folate concentration at days -14 and -7 relative to parturition, whereas the opposite relationship was noted for the plasma vitamin B12:folate ratio. The plasma folate and NEFA areas under the curve from the whole studied period were negatively associated, and the opposite was observed with the association between the plasma vitamin B12:folate ratio and NEFA as well as the BHB areas under the curve. The results suggest that there is an increased use of folate for metabolic functions during elevated concentrations of plasma NEFA and BHB. Future research should focus on finding an optimal plasma vitamin B12:folate ratio to favor cow health during the challenging period of parturition.

Beef cows' performance and metabolic response to short nutritional challenges in different months of lactation

Lactating cows can react to changes in nutrient availability with a range of behavioural and physiological mechanisms, which may differ among lactation stages. We investigated the effects of short feed restriction and refeeding periods on beef cows' performance and metabolic status in different months of lactation. For this, Parda de Montaña beef cows [n = 31; 626 ± 47.7 kg body weight (BW)] were subjected to short nutritional restriction and refeeding cycles, which were repeated in months 2, 3 and 4 of lactation. Each month, cows were consecutively fed a diet to meet 100% of their energy and protein requirements during a 4-day basal period, 55% during a 4-day restriction period, and again 100% during a 4-day refeeding period. The performance (energy balance, BW, milk yield and composition) and plasma metabolite concentrations (glucose, non-esterified fatty acids (NEFA), β-hydroxybutyrate (BHB), urea and malondialdehyde) were measured daily. Most of the traits were significantly affected by the interaction between feeding period and lactation month. Feed restriction induced milk yield loss, decreased milk protein and increased milk urea contents to different extents. The plasma NEFA concentrations rose with restriction in months 2, 3 and 4 but BHB and urea concentrations increased only in month 4. Most of these metabolites lowered to basal values during refeeding. These results suggest that beef cows use different adaptation strategies to cope with nutritional challenges as lactation advances, body fat mobilisation predominates in early lactation and protein catabolism prevails at later stages.

Influence of lactation stage on heat production and macronutrient oxidation in dairy cows during a 24-hour fasting period

Understanding nutrient utilization and partitioning is essential for advancing the efficiency of dairy cattle. Our objective was to determine if dairy cows exposed to a 24-h fasting period differ in heat production (HP) and macronutrient oxidation at different stages of lactation. Twelve primiparous, lactating German Holstein dairy cows were used in a longitudinal study design spanning from 2013 to 2014. Dairy cows were housed in respiration chambers during 3 stages of the lactation cycle: early (mean ± SD; 28.8 ± 6.42 d), mid- (89.4 ± 4.52 d), and late (293 ± 7.76 d) lactation. Individual CO2, O2, and CH4 gas exchanges were measured every 6 min for two 24-h periods, an ad libitum period and fasting period (RES). Blood was sampled at the start and end of the RES period. Gas measurements were used to calculate HP, net carbohydrate oxidation (COX), and net fat oxidation (FOX). Measurements were corrected with metabolic BW (kg of BW0.75; cBW). The RES period for each stage of lactation was further subdivided into the start (RESstart) and end (RESend) by averaging the first and last 2 h of the RES period. The net change was calculated as RESend - RESstart. All energy variables differed among lactation stage within the RES period except for HP/cBW. As expected, COX, COX/cBW, COX/HP, HP, and HP/cBW, were greater at the RESstart compared with RESend, whereas FOX, FOX/cBW, and FOX/HP were greater at the RESend except for FOX and FOX/cBW during mid lactation, which was only a tendency for a difference. The net change for COX, COX/cBW, HP, HP/cBW, and FOX/cBW did not differ among stages of lactation. Despite detecting a tendency for a difference among stage of lactation for FOX, pairwise analysis revealed no differences. Plasma triglyceride, urea, and nonesterified fatty acid concentrations were greater at RESend than RESstart. The net change for plasma glucose, urea, β-hydroxybutyrate, and nonesterified fatty acid concentrations were greater in early than late lactation. Our results demonstrate that despite differences in absolute measurements of energy variables and plasma metabolites, the change in whole-body macronutrient oxidation and HP as cows' transition from a fed-like state to a starvation-like state during a 24-h fasting period is consistent throughout lactation.

Pregnancy but Not Moderate Undernutrition Attenuates Insulin Suppression of Fat Mobilization in Sheep , ,

Nonpregnant and late-pregnant ditocous ewes were fed either to maintain zero energy balance in maternal tissues (fed) or at 50% of this level (underfed) for several weeks. Plasma concentrations of nonesterified fatty acids (NEFA) and glycerol were measured under basal conditions and during infusion of various doses of insulin while maintaining euglycemia (hyperinsulinemic, euglycemic clamp technique). Pregnancy and undernutrition separately increased basal plasma NEFA concentration in an additive manner; plasma glycerol was increased by pregnancy but unaffected by undernutrition. The molar ratio of NEFA to glycerol was significantly greater in underfed ewes. Analysis of dose-response relations between plasma insulin and metabolites during insulin infusions showed that maximally insulin-suppressed concentrations of NEFA and glycerol were significantly greater in pregnant than in nonpregnant ewes but were unaffected by undernutrition. Neither pregnancy nor undernutrition affected the maximally insulin-suppressed NEFA to glycerol ratio, or the plasma insulin concentration for 50% maximal responses to insulin of plasma NEFA, plasma glycerol, or the plasma NEFA to glycerol ratio. Thus, even in ewes at or close to zero energy balance, pregnancy seems to reduce adipose responsiveness but not sensitivity to the antilipolytic effect of insulin. This is another manifestation of the normal development of insulin resistance in maternal tissues during late pregnancy.

Roasted Soybeans and an Estrogenic Growth Promoter Affect Growth Hormone Status and Performance of Beef Steers

This study investigated the effects of a roasted soybean (RSB)-supplemented diet and an estrogen implant (SYN; Synovex-S® ear implant, 20 mg estradiol benzoate plus 200 mg progesterone) in young crossbred beef steers on their performance and plasma growth hormone (GH) response to challenge injections of thyrotropin-releasing hormone (TRH) + GH-releasing hormone (GHRH). Twenty individually fed steers (body weight 255 ± 5 kg) were assigned to the following treatments: 1) no SYN and fed a soybean meal-supplemented diet, 2) no SYN and fed the RSB-supplemented diet, 3) plus SYN and soybean meal, and 4) plus SYN and RSB. Steers were fed 1.13 MJ metabolizable energy/kg metabolic body weight daily of an 18% protein diet. After a 5-wk growth period, all steers were challenged (intravenous injection) over a 3-wk period with three levels of a combination of TRH + GHRH (0.1 + 0.01, 1.0 + 0.1, 2.5 + 0.25 µg/kg body weight, respectively). After an additional 3 wk, steers were reimplanted and a second 5-wk growth period was followed by a single challenge of the 1.0 + 0.1 TRH + GHRH dose level. Plasma nonesterified fatty acid concentration was greater when steers were fed the RSB compared with soybean meal (265 vs. 205 µmol/L; P < 0.01; SEM = 9.5). Body weight gains for treatments 1, 2, 3 and 4 were 1.35, 1.21, 1.47 and 1.38 kg/d, respectively (RSB, P < 0.10; SYN, P < 0.07; SEM = 0.06). Gain/dry matter intake (g/kg) means were 184, 167, 197 and 184 (RSB, P < 0.04; SYN, P < 0.07; SEM = 7.5). Feeding roasted soybeans resulted in depressed area under the GH response curve [907, 555, 827 and 989 µg/(L*min) (SYN × RSB, P < 0.03; SEM = 117)] and depressed peak response (37.2, 26.6, 33.5 and 41.1 µg/L [SYN × RSB, P < 0.05; SEM = 4.5]), an effect alleviated by estrogen for young growing steers (Period 1) but not for heavier steers (Period 2).

Effects of injectable vitamin E before or after transit on receiving phase growth performance, health, and blood parameters of beef steers.

The objective was to determine the effects of injectable vitamin E (VE) before or after transit on feedlot cattle receiving performance, health, and blood parameters. Angus × Simmental steers (n = 196; body weight [BW] = 163 ± 29 kg) were utilized in a randomized complete block design. Steers were blocked by BW and randomly assigned to 1 of 3 treatments: intramuscular injections of saline pre- and post-transit (CON), intramuscular injections of VE (2,000 mg d-α-tocopherol) pre-transit and saline post-transit (PRE), or intramuscular injections of saline pre-transit and VE (2,000 mg d-α-tocopherol) post-transit (POST). Pre-transit injections were administered on day 0, and steers were transported on day 7 for approximately 4 h (348 km). After arrival, steers were fed a common corn silage-based diet in GrowSafe bunks. Final BW tended to be greater (P = 0.08) for CON steers compared with POST steers while PRE steers were intermediate. From days 7 to 63, treatment affected average daily gain (ADG) with PRE and CON steers exhibiting (P = 0.04) greater ADG compared with POST steers. Dry matter intake (DMI), water intake, and gain to feed from days 7 to 63 were not affected (P ≥ 0.17) by treatment. Day 0 serum α-tocopherol concentrations were considered marginal (2.3 ± 0.2 mg/l). A treatment × day interaction (P < 0.01) was observed for serum α-tocopherol concentrations. Serum α-tocopherol concentrations were greatest for PRE steers on day 7 (prior to and post-transit), but greater for POST steers on dys 10 and 14. Plasma ferric-reducing antioxidant potential concentrations increased (P = 0.04) for POST steers compared with CON steers and PRE steers being intermediate. Plasma non-esterified fatty acids (NEFA) concentrations exhibited a treatment × day interaction (P = 0.04) with CON and POST steers being 16% and 14% greater than PRE steers on day 14, respectively. On day 21, NEFA concentrations were greatest for POST steers compared with PRE steers and CON steers being intermediate. There was no main effect (P ≥ 0.14) of treatment on the number of bovine respiratory disease morbidity treatments. Hair cortisol concentrations were decreased (P < 0.01) 14 days after transit for PRE and POST steers compared with CON steers. Overall, injectable VE administered before or after transit increased serum tocopherol concentrations while reducing stress, but did not improve the growth performance of beef steers during the receiving phase.

Effect of voluntary waiting period on metabolism of dairy cows during different phases of the lactation.

An extended calving interval (CInt) by extending the voluntary waiting period (VWP) could be associated with altered metabolism in dairy cows. The aim of this study was first to evaluate the effects of VWP on metabolism and body condition during the first 305 d after the first calving in the experiment (calving 1), around the end of the VWP, and during pregnancy (280 d before calving 2). Second, the effects of the VWP on metabolism were determined from 2 wk before until 6 wk after calving 2. Third, individual cow characteristics were used to predict milk production and body condition of cows after different VWP. Holstein-Friesian cows (N = 154, 41 primiparous [PP], 113 multiparous [MP]) were blocked for parity, milk production, and lactation persistency, randomly assigned to a VWP of 50, 125, or 200 d (VWP50, VWP125, or VWP200) and followed from calving 1 until 6 wk after calving 2. In the first 6 wk after calving 1 and from 2 wk before until 6 wk after calving 2, weekly plasma samples were analyzed for nonesterified fatty acids (NEFA), β-hydroxybutyrate, glucose, insulin, and insulin-like growth factor 1 (IGF-1). From wk 7 after calving 1 until 2 wk before calving 2, insulin and IGF-1 were analyzed every 2 wk. Fat- and protein-corrected milk (FPCM) and body weight (BW) gain were measured weekly. Cows were divided in two parity classes based on calving 1 (PP and MP) and remained in these classes after calving 2. During pregnancy, MP cows in VWP200 had greater plasma insulin and IGF-1 concentration and lower FPCM compared with MP cows in VWP125 (insulin: 18.5 vs. 13.9 µU/mL, CI 13.0-19.7, P < 0.01; IGF-1: 198.5 vs. 175.3 ng/mL ± 5.3, P = 0.04; FPCM: 22.6 vs. 30.0 kg/d ± 0.8, P < 0.01) or VWP50 (insulin: 15.8 µU/mL, P < 0.01; IGF-1: 178.2 ng/mL, P < 0.01; FPCM: 26.6 kg/d, P < 0.01) and had a greater daily BW gain compared with cows in VWP50 (3.6 vs. 2.5 kg/d ± 0.2; P < 0.01). After calving 2, MP cows in VWP200 had greater plasma NEFA concentration (0.41 mmol/liter) compared with MP cows in VWP125 (0.30 mmol/liter, P = 0.04) or VWP50 (0.26 mmol/liter, P < 0.01). For PP cows, the VWP did not affect FPCM or body condition during the first lactation in the experiment, or metabolism after calving 2. Independent of the VWP, higher milk production and lower body condition before insemination were associated with higher milk production and lower body condition at the end of the lactation. Variation in these characteristics among cows could call for an individual approach for an extended VWP.

Metabolomic exploration of the effects of habituation to livestock trailer and extended transportation in goats.

Goats raised for meat production are often transported long distances. Twelve-month-old male Spanish goats were used to determine the effects of habituation to trailers on plasma metabolomic profiles when transported for extended periods. In a split-plot design, 168 goats were separated into two treatment (TRT; whole plot) groups and maintained on two different paddocks. Concentrate supplement was fed to one group inside two livestock trailers (habituated group, H), while the other group received the same quantity of concentrate, but not inside the trailers (non-habituated, NH). Goats were subjected to a 10-h transportation stress in 4 replicates (n = 21 goats/replicate/TRT) after 4 weeks of habituation period. Blood samples were collected prior to loading, 20min after loading (0h), and at 2, 4, 6, 8, and 10h of transportation (Time; subplot). A targeted quantitative metabolomics approach was employed to analyze the samples. The data were analyzed using R software and MIXED procedures in SAS. Several amino acids (alanine, serine, glycine, histidine, glutamate, trans-hydroxyproline, asparagine, threonine, methylhistidine, ornithine, proline, leucine, tryptophan) were higher (p < 0.05) in the H group compared to the NH group. Six long-chain acylcarnitines were higher (p < 0.05), while free (C0) and short-chain (C3, C5) carnitines were lower (p < 0.05) in the NH goats compared to the H goats. In general, amino acid concentrations decreased and long-chain acylcarnitine (>C10) levels increased with transportation time (p < 0.05). Butyric acid, α-ketoglutaric acid, and α-aminoadipic acid concentrations were lower (p < 0.05) and β-hydroxybutyric acid concentrations were higher in the NH goats compared to the H goats. Plasma glucose, non-esterified fatty acid (NEFA) and urea nitrogen concentrations were significantly influenced by Time (p < 0.01). Plasma NEFA concentrations were significantly lower (p < 0.01) in the H group than the NH group. Habituation to trailers can be beneficial in enhancing stress coping abilities in goats due to higher concentrations of metabolites such as butyrate and certain amino acids that support antioxidant activities and immune function. Plasma long-chain acylcarnitines may be good indicators of stress during long-distance transportation in goats.

Using whole cottonseed to replace dried distillers grains plus solubles and prairie hay in finishing beef cattle rations balanced for physically effective neutral detergent fiber

The objective of this experiment was to determine the effects of replacing prairie hay and dried distillers grains plus solubles with whole cottonseed (WCS) in diets balanced for physically effective NDF on the growth, intake, feed efficiency, carcass characteristics, and plasma metabolites of finishing beef cattle. Crossbred heifers (n = 103) and steers (n = 104) were blocked by BW within sex and randomly allocated to pens within block (6 pens per treatment) with 17 (n = 10 pens) or 18 (n = 2 pens) animals per pen. Pens were randomly allocated to experimental treatment: either a control diet (CON; prairie hay, dried distillers grains plus solubles, dry-rolled corn, and liquid supplement) or a WCS diet (CTN; WCS, dry-rolled corn, and molasses). A common vitamin and mineral supplement and urea were included in both diets at the same rate. Animals were slaughtered in 3 groups based on BW block. Cattle fed the CTN treatment tended to have a greater final BW (P = 0.10) and had greater overall ADG and G:F (P ≤ 0.05). There was no difference in overall DMI (P = 0.23). Fecal consistency scores were greater for cattle fed the CON treatment on d 42, at the beginning of the β-agonist feeding period, and at the final collection before slaughter (P ≤ 0.03). Cattle fed the CTN treatment had a more neutral fecal pH on d 140 and at the final collection (P < 0.01). No treatment × day interactions (P ≥ 0.70) were detected for plasma glucose, lactate, urea nitrogen, or nonesterified fatty acid concentrations. Cattle fed the CON treatment had greater plasma urea nitrogen concentrations (P < 0.001) and a tendency for a greater plasma lactate concentration (P = 0.06). A day effect was also observed for all plasma metabolites (P < 0.001). This experiment suggests that feeding WCS improves the growth and feed efficiency of cattle when replacing the roughage and byproduct protein and fat sources within a finishing diet.

PSIII-A-7 Conditioning of Goats to Livestock Trailer: Effects on Physiological Stress Responses During Long-Distance Transportation

Abstract Meat goats are often transported long distances under commercial situations in the US. In a split-plot design, 168 twelve-month-old male Spanish goats were used to determine the effects of habituation to trailers on behavior and stress responses when transported for long periods. Animals were separated into two treatment (TRT) groups and maintained on two different paddocks. Concentrate supplement was fed to one group inside two livestock trailers (5.0 × 2.3 m each; habituated group, H), while the other group received the same quantity of concentrate, but not inside the trailers (non-habituated, NH). After 4 weeks of habituation period, goats were subjected to a 10-hour transportation stress in 4 replicates (n = 21 goats/replicate/TRT). Blood samples were collected prior to loading (Preload), 20 min after loading (0 h), and at 2, 4, 6, 8, and 10 h of transportation (Time). Behaviors of goats after loading were not affected by treatment. Plasma cortisol, glucose, and urea nitrogen concentrations and creatine kinase (CK) activities were significantly influenced by Time (P &amp;lt; 0.01). Plasma cortisol was not affected by TRT; however, glucose and CK in the H group tended to be lower than the NH group. Plasma non-esterified fatty acid (NEFA) concentrations were significantly lower (P &amp;lt; 0.01) in the H group than the NH group, likely in response to epinephrine release. There was a TRT × Time effect (P &amp;lt; 0.05) on CK activities, with the values increasing in the NH group and decreasing in the H group during the first two hours of transportation. Lymphocyte to neutrophil ratio also increased (P &amp;lt; 0.01) over transportation time. Habituation to trailer may be beneficial during long distance transportation of goats as indicated by certain physiological stress responses. Habituation may also facilitate the loading process, based on behavioral observations.

PSIII-A-1 Acute Stress Effects on Physiological Responses and Meat Quality Changes in Goats

Abstract Duration of stress prior to processing can significantly affect physiological status and meat quality in goats. This experiment was conducted to determine the effects of duration of preslaughter stress on physiological responses and meat quality in goats. In a Randomized Complete Block Design, fifty-four uncastrated male Spanish goats (8-mo old; BW 29.7 ± 2.03 kg) were randomly subjected to one of three treatments (n = 18 goats/treatment): (i) transported for 3 h, (ii) transported for 30 min, or (iii) or held in pens (control) on two different days. Blood samples were collected before and after transport for assessment of stress responses. Goats were slaughtered, using humane procedures, for meat quality analysis. The data were analyzed using MIXED procedures in SAS with treatment and day as fixed variables. Cortisol concentrations were higher (P &amp;lt; 0.01) in both 3 h (117.7 ± 7.96 ng/mL) and 30 min (130.1 ± 8.05 ng/mL) transported groups compared to the control group (27.5 ± 7.59 ng/mL). A similar treatment effect was noticed in plasma non-esterified fatty acid (P &amp;lt; 0.05) concentrations. Glucose concentrations were highest in 180-min, lowest in control, and intermediate in 30-min group (P &amp;lt; 0.01). There were significant positive correlations among muscle desmin concentration after 6 days of aging and 24 h pH (P &amp;lt; 0.05) and calpastatin activity (P &amp;lt; 0.05), suggesting that desmin degradation plays a role in determining the tenderness of aged meat from goats subjected to preslaughter stress. Warner-Bratzler shear force values of loin chops were highest in 180-min, lowest in control, and intermediate in 30-min groups; however, treatment did not influence the color values or cooking loss of loin chops. The results indicate that acute stress prior to processing can affect both physiological responses and meat quality in a duration-dependent manner in goats.
