Suppression effects of betaine-enriched spinach on hyperhomocysteinemia induced by guanidinoacetic acid and choline deficiency in rats.

Betaine is an important natural component of rich food sources, especially spinach. Rats were fed diets with betaine or spinach powder at the same level of betaine for 10 days to investigate the dose-dependent effects of spinach powder supplementation on hyperhomocysteinemia induced by guanidinoacetic acid (GAA) addition and choline deprivation. The GAA-induced hyperhomocysteinemia in rats fed 25% casein diet (25C) was significantly suppressed by supplementation with betaine or spinach, and it was completely suppressed by taking 11.0% spinach supplementation. The choline deprivation-induced enhancement of plasma homocysteine concentration in rats fed 25% soybean protein diet (25S) was markedly suppressed by 3.82% spinach. Supplementation with betaine or spinach partially prevented the effects of GAA on hepatic concentrations of methionine metabolites. The decrease in activity of betaine-homocysteine S-methyltransferase (BHMT) and cystathionine β-synthase (CBS) in GAA-induced hyperhomocysteinemia was recovered by supplementation with betaine or spinach. Supplementation with betaine or spinach did not affect BHMT activity, whereas it partially restored CBS activity in choline-deprived 25S. The results indicated that betaine or spinach could completely suppress the hyperhomocysteinemia induced by choline deficiency resulting from stimulating the homocysteine removal by both remethylation and cystathionine formation.

Factors Contributing to the Resistivity of a Higher Casein Diet against Choline Deficiency-Induced Hyperhomocysteinemia in Rats

The mechanism by which feeding a higher casein diet results in resistance to choline deprivation-induced hyperhomocysteinemia was investigated in rats. Plasma homocysteine concentration was significantly lower in rats fed a 30% casein diet (30C) than in rats fed a 10% casein diet (10C). Choline deprivation did not enhance plasma homocysteine concentration in rats fed 30C, while it significantly enhanced plasma homocysteine concentration in rats fed 10C. The choline deprivation-induced enhancement of plasma homocysteine concentration in rats fed 10C was significantly suppressed by methionine supplementation in a dose-dependent manner in the range of 0.1 to 0.3%, but the suppressive effect of methionine became smaller with an increase in supplementation level in the range of 0.3 to 0.5%. At a 0.5% supplementation level, methionine did not exhibit any suppressive effect on choline deprivation-induced hyperhomocysteinemia. The higher plasma homocysteine concentration in rats fed choline-deprived 10C+0.5% methionine was significantly decreased by concurrent supplementation with 0.32% glycine+0.94% serine to the level of rats fed 10C. Raising dietary total amino acid level by adding 3.61% branched-chain amino acids (BCAA)+4.5% acidic amino acids (AAA) to choline-deprived 10C+0.5% methionine+0.32% glycine+0.94% serine resulted in a further decrease in plasma homocysteine concentration to a level lower than the level in rats fed 10C. Choline deprivation-induced increases in hepatic S-adenosylhomocysteine and homocysteine concentrations were significantly suppressed by supplementation with glycine+serine and further suppressed by BCAA+AAA. Hepatic cystathionine β-synthase activity and its gene expression were significantly increased by BCAA+AAA. Hepatic triglyceride concentration changed in a manner similar to that of plasma homocysteine concentration. The results indicate that there are at least three factors contributing to the resistivity of rats fed a higher casein diet (30C) to choline deprivation-induced hyperhomocysteinemia, i.e., higher intake of methionine, higher intake of glycine and serine, and higher intake of other amino acids such as BCAA and AAA.

Effects of Betaine Supplementation and Choline Deficiency on Folate Deficiency-Induced Hyperhomocysteinemia in Rats

The effect of betaine status on folate deficiency-induced hyperhomocysteinemia was investigated to determine whether folate deficiency impairs homocysteine removal not only by the methionine synthase (MS) pathway but also by the betaine-homocysteine S-methyltransferase (BHMT) pathway. For this purpose, we investigated the effect of dietary supplementation with betaine at a high level (1%) in rats fed a folate-deprived 10% casein diet (10C) and 20% casein diet (20C). We also investigated the effect of choline deprivation on folate deficiency-induced hyperhomocysteinemia in rats fed 20C. Supplementation of folate-deprived 10C and 20C with 1% betaine significantly suppressed folate deprivation-induced hyperhomocysteinemia, but the extent of suppression was partial or limited, especially in rats fed 10C, the suppression of plasma homocysteine increment being 48.5% in rats fed 10C and 69.7% in rats fed 20C. Although betaine supplementation greatly increased hepatic betaine concentration and BHMT activity, these increases did not fully explain why the effect of betaine supplementation was partial or limited. Folate deprivation markedly increased the hepatic concentration of N,N-dimethylglycine (DMG), a known inhibitor of BHMT, and there was a significant positive correlation between hepatic DMG concentration and plasma homocysteine concentration, suggesting that folate deficiency increases hepatic DMG concentration and thereby depresses BHMT reaction, leading to interference with the effect of betaine supplementation. Choline deprivation did not increase plasma homocysteine concentration in rats fed 20C, but it markedly enhanced plasma homocysteine concentration when rats were fed folate-deprived 20C. This indicates that choline deprivation reinforced folate deprivation-induced hyperhomocysteinemia. Increased hepatic DMG concentration was also associated with such an effect. These results support the concept that folate deficiency impairs homocysteine metabolism not only by the MS pathway but also by the BHMT pathway.

High Casein Diet Decreases Plasma Homocysteine Concentration in Rats

Experiments were conducted to clarify the relationship between dietary protein level and plasma homocysteine concentration in rats. Male Wistar rats were fed diets differing in casein level from 5 to 50% for 14 d (Expt. 1). Plasma total homocysteine concentration was positively correlated with dietary casein level in the range of 5 to 10% but inversely correlated with dietary casein level in the range of 10 to 50%. Hepatic cystathionine beta-synthase (CBS) and betaine-homocysteine S-methyltransferase (BHMT) activities and renal CBS activity increased in response to dietary casein level in the range of 10 to 50%, whereas hepatic serine and betaine concentrations decreased with increasing dietary casein levels. When rats were fed the 10% casein diet or 10% casein+17.2% amino acid mixture diet for 14 d, plasma homocysteine concentration was significantly lower in rats fed the amino acid mixture-added diet than in rats fed the 10% casein diet (Expt. 2), indicating that the hypohomocysteinemic effect of high casein diets was elicited by amino acids, not by casein contaminants. The degree of increase in plasma homocysteine concentration caused by dietary supplementation with 0.75% L-methionine was significantly lower in rats fed the 40% casein diet than in rats fed the 10% casein diet (Expt. 3). These results indicate that high casein diets do not increase but rather decrease plasma homocysteine concentration and cause resistance to hyperhomocysteinemic treatment, and suggest that such effects of high casein diets are mediated at least by increased activities of CBS and BHMT.

High-Casein Diet Suppresses Guanidinoacetic Acid-Induced Hyperhomocysteinemia and Potentiates the Hypohomocysteinemic Effect of Serine in Rats

To determine the effect of dietary protein level on experimental hyperhomocysteinemia, rats were fed 10% casein (10C) and 40% casein (40C) diets with or without 0.5% guanidinoacetic acid (GAA) for 14 d. In addition, rats were fed 10C + 0.75% methionine (10CM) and 40C + 0.75% methionine (40CM) diets with or without 2.5% serine for 14 d to determine the relationship between the dietary protein level and intensity of the hypohomocysteinemic effect of serine. GAA supplementation markedly increased the plasma homocysteine concentration in rats fed with the 10C diet, whereas it did not increase the plasma homocysteine concentration in rats fed with the 40C diet. Although serine supplementation significantly suppressed the methionine-induced enhancement of plasma homocysteine concentration, the decreased plasma homocysteine concentration was significantly lower in rats fed with the 40CM diet than in rats fed with the 10CM diet. The hepatic cystathionine beta-synthase and betaine-homocysteine S-methyltransferase activities were significantly higher in rats fed with the 40C or 40CM diet than in rats fed with the 10C or 10CM diet, irrespective of supplementation with GAA and serine. These results indicate that the high-casein diet was effective for both suppressing GAA-induced hyperhomocysteinemia and potentiating the hypohomocysteinemic effect of serine, probably through the enhanced activity of homocysteine-metabolizing enzymes.

Effect of Dietary Soybean Protein Level on the Plasma Homocysteine Concentration in Rats

There was an inverse correlation between the plasma homocysteine concentration and dietary protein level or protein intake when a soybean protein isolate (SPI) was used as a protein source for rats. The hepatic cystathionine beta-synthase activity increased in response to the dietary SPI level. The results suggest that a high-protein diet might be an effective means to lower the plasma homocysteine concentration, probably through enhancement of the homocysteine-metabolizing activity.

Phosphocreatine and genistein supplementation attenuate plasma homocysteine concentrations in rats

Hyperhomocysteinemia is an independent risk factor for cardiovascular disease. S‐adenosylmethionine (SAM) is the methyl group donor for most biological methylations. The product of this reaction, S‐adenosylhomocysteine, is converted to homocysteine (Hcy), which is remethylated back to methionine or irreversibly catabolized. Thus, Hcy pools reflect a balance between production and metabolism. Biosynthesis of phosphocreatine (PCr) from guanidinoacetate (GAA) is a transmethylation reaction that requires SAM‐derived methyl groups, contributing significantly to Hcy production. Therefore, dietary supplementation with PCr should decrease the demand on SAM for GAA synthesis. The isolflavone, genistein (G), may promote homeostatic methyl group metabolism. However, mechanisms underlying the effects of G are not well understood. We examined the effects of PCr and G supplementation on elevated Hcy during moderate folate deficiency. Half of the rats received a FD diet for 5 and ½ wk, followed by supplementation with either PCr (4g/kg/diet) or G (300mg/kg/diet) for 10 d. As expected, FD increased total plasma Hcy concentrations by 95%. PCr significantly reduced Hcy during FD (p = 0.049). However, Hcy was attenuated in all control and FD groups receiving PCr and G. Taken together, PCr and G may represent a viable dietary strategy to reduce plasma Hcy.

Acute Exposure to Cyclosporine Does Not Increase Plasma Homocysteine in Rats

There is interest in the postulate that cyclosporine a (CsA) contributes to the elevated homocysteine levels seen in organ transplant recipients, as hyperhomocysteinemia is now considered an independent risk factor for cardiovascular disease (CVD) and may partially explain the increased prevalence of CVD in this population. The main purpose of this investigation was to determine the effect of CsA administration on plasma homocysteine. Eighteen female Sprague Dawley rats (4 months old) were randomly assigned to either a treatment or a control group. For 18 days the treatment group received of CsA (25 mg/kg/d) while the control group received the same volume of the vehicle. Blood samples obtained following sacrifice to measure CsA, total homocysteine, and plasma creatinine. There were no significant differences in plasma homocysteine (mean values ± SD: treatment = 4.79 ± 0.63 μmol/L, control = 4.46 ± 0.75 μmol/L; P = .37). Homocysteine was not significantly correlated with final CsA concentrations ( r = .17; P = .69). There was a significant difference in plasma creatinine values between the two groups (treatment = 60.44 ± 7.68 μmol/L, control = 46.33 ± 1.66 μmol/L; P < .001). Furthermore, plasma homocysteine and creatinine were positively correlated with the treatment group ( r = .73; P < .05) but not the controls ( r = −.10; P = .81). In conclusion, CsA does not influence plasma homocysteine concentrations in rats.

Triiodothyronine Treatment Attenuates the Induction of Hepatic Glycine N-Methyltransferase by Retinoic Acid and Elevates Plasma Homocysteine Concentrations in Rats

Recent studies indicated that hormonal imbalances have a role in modulating the metabolism of methyl groups and homocysteine, interrelated pathways that when disrupted, are associated with a number of pathologies. Retinoic acid (RA) was shown to induce hepatic glycine N-methyltransferase (GNMT), a key regulatory protein in methyl group metabolism, and to reduce circulating homocysteine levels. Because thyroid status influences the hepatic folate-dependent one-carbon pool and retinoids can alter thyroid hormone levels, the aim of this study was to examine the interaction between retinoids and thyroid function. For hypothyroid studies, rats were administered 0.5 g/L propylthiouracil in the drinking water for 15 d, and RA [30 micromol/(kg . d)] for the final 5 d. For hyperthyroid studies, rats were treated with RA [30 micromol/(kg . d)] for 8 d and triiodothyronine [T(3); 50 microg/(100 g . d)] the last 4 d. T(3) treatment prevented the RA-mediated increase in GNMT activity. However, GNMT abundance remained elevated, indicating that GNMT regulation by T(3) in RA-treated rats may be, at least in part, at the post-translational level. In addition, T(3) treatment elevated plasma levels of homocysteine 177%, an elevation that was prevented by RA. T(3)-mediated hyperhomocysteinemia may be due to a 70% decrease in hepatic betaine-homocysteine S-methyltransferase, the enzyme that catalyzes folate-independent remethylation of homocysteine, whereas the RA-mediated stimulation of hepatic homocysteine remethylation by folate-dependent methionine synthase may contribute to lowering plasma homocysteine levels. These findings indicate that thyroid hormones, alone and in conjunction with RA, play an important role in the regulation of methyl group and homocysteine metabolism.

Acute stressor-selective effect on total plasma homocysteine concentration in rats

Stress produces several physiological and behavioral alterations that increase cardiovascular morbidity and mortality. Nonetheless, there is a dearth of studies that have evaluated the effects of stress on total plasma homocysteine, an important amino acid associated with cardiovascular disease. We used four distinct acute stressors in rats, i.e., swimming, restrain, novelty and cold exposure, in order to examine whether any acute effect on total plasma homocysteine concentrations would occur. Plasma corticosterone and adrenocorticotropic hormone concentrations were also measured to demonstrate the ability of the chosen manipulations to activate the hypothalamic–pituitary–adrenal (HPA) axis. Three of the four stressors activated the HPA axis and only restrain affected total plasma homocysteine concentrations (+37%, P=.006) compared with the control group. The complexity of the physiological responses to stress, the peculiarities of stress responses and the intricate regulatory systems involved in homocysteine metabolism must be taken into account in order to clarify the increasing effect of restrain (mainly a psychological stressor) on total plasma homocysteine concentrations in rats and to evaluate its meaning in human pathology.

Retinoic Acid and Glucocorticoid Treatment Induce Hepatic Glycine N-Methyltransferase and Lower Plasma Homocysteine Concentrations in Rats and Rat Hepatoma Cells

Perturbation of folate and methyl group metabolism is associated with a number of pathological conditions, including cardiovascular disease and neoplastic development. Glycine N-methyltransferase (GNMT) is a key protein that functions to regulate the supply and utilization of methyl groups for S-adenosylmethionine (SAM)-dependent transmethylation reactions. Factors or conditions that have the ability to regulate GNMT and the generation of homocysteine, a product of transmethylation, have important implications in the potential perturbation of methyl group metabolism. We showed that retinoid compounds induce active hepatic GNMT, resulting in compromised transmethylation processes. Because retinoids can stimulate gluconeogenesis, a condition known to alter methyl group and homocysteine metabolism, the current study was undertaken to determine the relationship between all-trans-retinoic acid (RA) and gluconeogenic hormones on these metabolic pathways. Intact adrenal function was not required for RA to induce and activate hepatic GNMT; however, treatment of rats with dexamethasone (DEX) was as effective as RA in inducing GNMT in rat liver. The marked increase in plasma total homocysteine levels observed in adrenalectomized rats was reduced to normal levels by treatment with either RA or DEX, indicating that the transsulfuration and/or remethylation pathways may be enhanced. Moreover, coadministration of RA and DEX had an additive effect on GNMT induction. Similar findings were also observed in a rat hepatoma cell culture model using H4IIE cells. Taken together, these results demonstrate that both RA and DEX independently induce GNMT, thereby having substantial implications for the potential interaction of retinoid administration with diabetes.

Influence of folic acid-fortified foods on folate status in a folate depletion-repletion rat model.

An increasing number of foods fortified with varying levels of folic acid are appearing in the market place, targeted either at the general population or at specific consumer groups. Although it is assumed that the folate in these products should be highly bioavailable, there is a need to carry out studies to ascertain that this is, in fact, the case. The present study investigated the ability of selected folic acid-fortified foods (targeted at different types of consumer) to increase the folate status of folate-deficient rats. Forty-two weanling male rats (Wistar strain) were fed a folate-deficient diet containing 1 % succinyl sulfathiazole (w/w) for 28 d. Following depletion, seven rats were randomly assigned to each of five repletion diets containing folic acid, Complan, Slim Fast, Opti-Fuel2 or Cola Coa calculated to provide 200 microg folate/kg of each diet. Calculations were based on folate information from the product labels. After a further 28 d, plasma, liver and kidney folate concentrations were determined by microbiological assay. Plasma homocysteine was measured by HPLC as a functional indicator of folate status. The folate content of the foods was measured by tri-enzyme extraction followed by microbiological assay. Our analyses suggest that there may be considerable inaccuracies on the part of the manufacturers in relation to the folate declarations on the product labels. Despite this, the four foods evaluated were highly effective in elevating plasma, liver and kidney folate and lowering plasma homocysteine concentrations in rats. These results lend support to the policy of food fortification with folic acid as a means of raising the folate status of the population, and in particular to the fortification of specific foods which may target areas of the population where increased folate status is most needed.

The effect of l-dopa administration and folate deficiency on plasma homocysteine concentrations in rats

O-Methylation of the anti-Parkinson's disease drug l-dopa leads to significant decreases of S-adenosylmethionine and significant increases of S-adenosylhomocysteine concentrations in tissues. Based on these observations, we hypothesized that l-dopa administration would also lead to increased production of homocysteine and hyperhomocysteinemia. This hypothesis was tested in two separate experiments. In experiment 1, control and folate-deficient male rats were injected intraperitoneally with 100 mg of l-dopa per kilogram body weight. After 1 hr, blood was collected and analyzed for homocysteine. Plasma homocysteine concentration was significantly higher in the rats treated with l-dopa than in the rats treated with vehicle alone. Furthermore, the apparent increase of plasma homocysteine due to l-dopa was greater in the folate-deficient rats than in the replete controls, suggesting a significant interaction between l-dopa administration and folate deficiency on plasma homocysteine concentration. In experiment 2, nondeficient female rats were injected intraperitoneally with 100 mg of l-dopa per kologram of body weight for 1, 1, or 17 consecutive days (one injection per day). Blood was collected 1 hr after the last dose and analyzed for homocysteine. Plasma homocysteine concentration was significantly higher in the rats treated for 17 days than in the nontreated controls, indicating that the effect of l-dopa persisted with chronic administration. However, plasma homocysteine concentration was significantly higher in the rats treated with l-dopa for only 1 day than in those treated for 17 days, suggesting that there is some attenuation of the effect of l-dopa with chronic administration. Measurements of S-adenosylmethionine and S-adenosylhomocysteine in brain and liver were consistent with the hypothesis that the hyperhomocysteinemia was a consequence of significant O-methylation.
