Effects of rumen-protected lysine and methionine supplementation in low-crude protein diets on lactation performance, nitrogen metabolism, rumen fermentation, and blood metabolites in Holstein cows

The objective of the study was to investigate the effects of supplementing a low-protein diet with rumen-protected lysine (RP-Lys) and RP-methionine (RP-Met) on milk production, nitrogen metabolism, and rumen fermentation in dairy cows. Forty-eight Holstein dairy cows were blocked by parity (2.1 ± 0.17), milk production (32.1 ± 3.41 kg/d), and days in milk (124 ± 28 d), and randomly assigned to 1 of 4 dietary treatments: control diet (CON, 16 % CP), low-protein diet (LP, 14 % CP, negative control), a moderate diet supplemented with RP-Met (31 g/d) and RP-Lys (MPML, 15 % CP, 110 g/d), and LP diet with RP-Met (50 g/d) and RP-Lys (LPML, 14 % CP, 150 g/d). Dry matter intake (DMI), feed efficiency, and the apparent total tract digestibility of nutrients was not affected by treatments. Compared with the CON, the milk yield was increased in the MPML treatment (P = 0.03) while decreased in the LP treatment (P = 0.02). Milk protein yield in the MPML treatment was higher than that in the other treatments (P < 0.05). The LP, LPML, and MPML treatments had lower concentrations of milk urea-N and plasma urea-N than CON treatments (P < 0.01). The LP, LPML, and MPML treatments decreased the plasma histidine (His) concentration (P < 0.01), while the LP and LPML treatments decreased plasma arginine (Arg) concentration (P < 0.05) compared to the CON treatment. Plasma Met concentration in the MPML treatment was increased (P < 0.01), whereas was decreased in the LP treatment, compared with the CON (P < 0.05). The N intake (g/d) in the LP, LPML, and MPML treatments was lower than that in the CON treatment (P < 0.01), while fecal N (g/d) was not affected by treatments. However, CON treatment increased the urine N (g/d), total excreta N (g/d), N secretion and excretion (g/d), and the proportions of urine N to N intake, whereas decreased the proportion of milk N to N intake and milk N efficiency, compared to the other three treatments (P < 0.01). The rumen concentration of ammonia nitrogen in the LP, LPML, and MPML treatments was lower than that in the CON treatment (P < 0.05). Collectively, the low-protein diets supplemented with RP-Met and RP-Lys could increase milk N efficiency and dramatically decrease urinary N losses. From the perspective of N emission reduction, a 2% reduction in dietary CP was rational.

Plasma histidine concentrations as a specific biomarker for intestinal mucosal damage in calves with cryptosporidiosis

Specific alterations in plasma histidine concentrations and diamine oxidase (DAO) activity were recently reported as a potential biomarker for intestinal mucosal damage in diarrheic calves. However, there are no data on the comparison of precision between histidine concentration and DAO activity in bovine plasma. The aim of the present study was to compare precision of histidine concentrations and DAO activities in plasma as a biomarker for the Cryptosporidium parvum (C. parvum)-associated intestinal mucosal damage in diarrheic calves. Thirty-two Holstein calves aged 12.2 ± 4.1 days old were enrolled in the present study; they were divided into C. parvum (n = 9), diarrhea (n = 11), and control (n = 12) groups based on the presence or absence of diarrhea and with or without C. parvum infection. Receiver operating characteristic (ROC) curves were used to characterize the sensitivity and specificity of each parameter for the C. parvum-associated intestinal mucosal damage. The proposed cut-off points for plasma histidine concentrations and plasma DAO activities for cryptosporidiosis in calves based on ROC analyses were < 55.8 nM and < 246.0 IU/ml, respectively. The sensitivities and specificities of the proposed diagnostic cut-offs were 88.9% and 82.6% for plasma histidine concentrations and 100.0% and 34.8% for plasma DAO activities, respectively. It was concluded that plasma histidine concentrations may be superior to plasma DAO activities as a specific biomarker for the C. parvum-associated intestinal mucosal damage in diarrheic calves.

Abstract This experiment was conducted to clarify the effects of feeding rice on growth performance and protein (amino acids) metabolism in weaning piglets. Sixteen weaning piglets with an average initial weight of 7.5 kg were divided into two groups. One group was fed a corn-soybean meal based diet, and the other was fed a rice-soybean meal diet. Each diet contained about 46% of corn or rice. A 2-week growth trial was conducted and growth performance, dry matter and crude protein digestibility, liver lysine-ketoglutarate reductase activity, plasma urea nitrogen, glucose, insulin, and free amino acids concentrations were measured. The average daily gain (g/d) and feed efficiency (gain/feed) in rice-fed piglets were significantly higher than those in corn-fed piglets. Average feed intake, however, was not significantly different between corn- and rice-fed piglets. Dry matter and and crude protein digestibility were not significantly different between two groups. Liver lysine-ketoglutarate reductase activity tended to be lower (P = 0.073) in rice-fed piglets than that in corn-fed piglets. Plasma urea nitrogen concentration in rice-fed piglets was significantly lower than that in corn-fed piglets. Plasma glucose and insulin concentrations were significantly higher in rice-fed piglets than those in corn-fed piglets. Plasma free valine, isoleucine, tryptophan, and glysine concentrations were significantly higher in rice-fed piglets than those in corn-fed piglets. On the other hand, plasma histidine concentration in rice-fed piglets was significantly lower than that in corn-fed piglets. In conclusion, these results clearly show that rice feeding affects the protein and amino acids metabolism and improves the growth performance in weaning piglets. Rice could be considered to be a good feedstuff for weaning piglets.

In all, 16 weanling piglets with an average initial weight of 7.5kg were divided into two groups. The average daily gain (p=.025) and feed efficiency (p=.011) in rice-fed piglets were significantly higher than those in corn-fed piglets. Liver lysine-ketoglutarate reductase activity tended to be lower (p=.073) in rice-fed piglets than in corn-fed piglets. Plasma urea nitrogen concentration in rice-fed piglets was significantly lower than that in corn-fed piglets. Mammary Protein Synthesis upon Long-Term Nutritional Restriction Was Attenuated by Oxidative-Stress-Induced Inhibition of Vacuolar H+-Adenosine Triphosphatase/Mechanistic Target of Rapamycin Complex 1 Signaling.

To determine how nutritional restriction compromised milk synthesis, sows were fed 100% (control) or76% (restricted) of the recommended feed allowance from postpartum day (PD)-1 to PD-28. In comparison to the control, more body reserves loss, increased plasma triglyceride and high-density lipoprotein cholesterol levels, and decreased plasma methionine concentrations were observed in the restricted group at PD-21. The increased plasma malondialdehyde level, decreased plasma histidine and taurine concentrations, and decreased glutathione peroxidase activity were observed at PD-28 when backfat loss further increased in the restricted group. In mammary glands, vacuolar H+-adenosine triphosphatase (v-ATPase), as the upstream of the mechanistic target of rapamycin (mTOR) signaling, showed decreased activity, while phosphorylation of mTOR, S6 kinase, and eukaryotic translation initiation factor 4E-binding protein 1 and β-casein abundance all decreased following feed restriction. Altogether, long-term nutrition restriction could induce progressively aggravated oxidative stress and compromise mammary protein synthesis through repression of v-ATPase/mTORC1 signaling.

Amino Acid-Based Metabolic Profile Provides Functional Assessment and Prognostic Value for Heart Failure Outpatients.

Functional capacity is a crucial parameter correlated with outcomes. The currently used New York Heart Association functional classification (NYHA Fc) system has substantial limitations, leading to inaccurate classification. This study investigated whether amino acid-based assessment on metabolic status provides an objective way to assess functional capacity and prognosis in heart failure (HF) outpatients. Plasma concentrations of histidine, ornithine, and phenylalanine (HOP) were measured on 890 HF outpatients to assess metabolic status by calculating the HOP score. Cardiopulmonary exercise testing (CPET) was performed in 387 patients to measure metabolic equivalents (MET) in order to define the functional class based on MET (MET Fc). Patients were followed for composite events (death/HF-related rehospitalization) up to one year. We found only 47% concordance between the MET Fc and NYHA Fc. HOP scores worked better than NYHA Fc for discriminating patients with MET Fc II and III from those with MET Fc I, with the optimal cutoff value set at 8.8. HOP scores ≥ 8.8 were associated with risk factors for composite events in different kinds of HF populations and were a powerful predictor of composite events in univariate analysis. In multivariable analysis, HOP scores ≥ 8.8 remained a powerful event predictor, independent of other risk factors. Kaplan-Meier curves revealed that HOP scores of ≥8.8 stratified patients at higher risk of composite events in a variety of HF populations. In conclusion, amino acid-based assessment of metabolic status correlates with functional capacity in HF outpatients and provides prognostic value for a variety of HF populations.

Blood glucose, amino acid profiles and nutrient transporter gene expressions in the small intestine of low and normal birthweight piglets during the early suckling period

Birthweight and age can play a role in adaptational changes in plasma levels of glucose and amino acids by regulating the intestinal expression of specific nutrients transporters in piglets. The aim of the present study was to compare hematological features, circulating levels of glucose and amino acids, and small intestinal gene expression for transporters for glucose (SGLT1, GLUT2), amino acids (LAT2, b0,+AT) and peptides (PepT1) between low birthweight (LBW) and normal birthweight (NBW) piglets at birth prior to suckling and day 7 after birth. A secondary aim was to evaluate correlations between circulating levels of glucose or amino acids and their transporters.There were significant age-dependent changes in the immune cell repertoire in the circulating blood (P<0.004). Blood glucose levels were lower in LBW piglets than in NBW piglets (P<0.05); in both birthweight groups, blood glucose increased with age. Plasma concentrations of histidine, isoleucine, proline and tryptophan showed significant changes with age (P<0.003). With regard to mRNA levels for the nutrient transporters, there were no differences between the two birthweight groups or between the two age groups. At day 7 postpartum, intestinal expression of SGLT1 or GLUT2 mRNA did not correlate with blood glucose level. Expression of LAT2 mRNA in the ileum correlated positively with plasma level of methionine (r=0.92, P<0.001). This is the first detailed analysis of hematological features, plasma glucose and amino acid levels, and nutrient transporter expression in LBW and NBW piglets during the early suckling period.

Effects of spray-dried plasma protein product on early-lactation dairy cows

Spray-dried plasma protein (SDP) compared with blood meal (BM) may contain various functional and active components that may benefit animal health. The objective of this experiment was to investigate the effects of feeding SDP or BM on production and blood profile in dairy cows during the transition and early-lactation periods. Seventy-two Holstein cows at 14 d before calving were used in a randomized block design. During the prepartum period, cows were fed a typical late-gestation diet containing BM (100 g/cow per day; 100BM, n = 24) or SDP (100 g/cow per day; 100SDP; n = 48). After calving, cows that were fed BM prepartum were fed a typical lactation diet formulated to provide 100 g/d of BM (100BM). Half the cows that were fed 100SDP prepartum were fed a lactation diet formulated to provide 100 g/d of SDP (100SDP; n = 24), and half were fed a diet formulated to provide 400 g/d of SDP (400SDP; n = 24) on a dry matter basis where SDP replaced BM (100SDP) or BM and soybean products (400SDP). All diets were balanced for crude protein concentration and metabolizable protein supply assuming BM and SDP were equal in rumen-degradable protein and rumen-undegradable protein. All data were analyzed using the MIXED procedure of SAS (SAS Institute Inc., Cary, NC) as a randomized block design where contrasts were made for 100BM versus 100SDP for prepartum variables and 100BM versus 100SDP and 100SDP versus 400SDP for postpartum variables. Prepartum supplementation of SDP had no effect on plasma fatty acids and β-hydroxybutyrate (2 d before calving). Plasma fatty acids (255 ± 29 µEq/mL) and β-hydroxybutyrate (675 ± 70 µmol/L) at 8 and 14 d of lactation were not affected by SDP in the diet. Feeding SDP at 100 g/d compared with 100BM increased or tended to increase milk fat, protein, and lactose contents for 16 wk after parturition. Providing SDP at 400 g/d in the diet increased milk yield (42 vs. 39 kg/d), energy-corrected milk (44 vs. 41 kg/d), energy-corrected milk per kilogram of dry matter intake, and yields of milk fat (1.60 vs. 1.48 kg/d), protein (1.21 vs. 1.16 kg/d), and lactose compared with 100SDP. Body weight losses tended to be lower for 100SDP compared with 100BM without a difference between 100SDP and 400SDP. Plasma histidine concentration (d 14 of lactation) was lower for SDP compared with 100BM. In addition, plasma 1-methyl-l-histidine tended to be lower as inclusion rate of SDP increased. In conclusion, SDP at 400 g/d increased milk and milk component yields without an increase in feed intake. Studies evaluating effects of functional and active compounds in SDP on gut microbiome, gut health, and immune functions may be needed to determine mode of action.

Chronic lung inflammation affects plasma amino acid concentrations in pigs1

Metabolic changes associated with inflammatory processes and immune response can modify protein and AA requirements. Improved knowledge of these processes will provide opportunities for nutritional intervention to sustain growth and animal defense at the same time. The objective of the study was to identify AA whose metabolism is affected by chronic lung inflammation. Six pairs of littermate piglets were selected at 28 d of age on the basis of their BW. After catheterization of the jugular vein, one littermate received complete Freund's adjuvant (CFA) intravenously, whereas its littermate was injected with a sterile saline solution (CON). Piglets within a litter were pair-fed in order to avoid confounding effects of feed intake and inflammation on plasma AA concentrations. Blood samples were taken after an overnight fast and 2 h after the morning meal for 9 d. Rectal temperature, food consumption, weight gain, plasma haptoglobin, and AA concentrations were measured. The CFA injection decreased food intake, and increased body temperature and plasma haptoglobin concentration. Plasma tryptophan, glutamine, proline, glycine, tyrosine, ornithine, total AA concentrations, and the ratio of tryptophan to large neutral AA were less in CFA than in CON (P < 0.05), independent of time and meal. In contrast, plasma histidine concentration was higher (P < 0.05) in CFA than in CON pigs. Plasma serine, arginine, alanine, asparagine, and total AA concentrations were lower in CFA than in CON pigs only in the fed state (P < 0.05). Among essential AA, only plasma tryptophan concentration was lower (P < 0.01) in CFA than in CON pigs in both fasted and fed state. These results show that chronic lung inflammation affects individual AA differently and suggest that the utilization of some AA increased during chronic lung inflammation in pigs. Activation of tryptophan catabolism enzyme indoleamine 2,3-dioxygenase seems a relevant hypothesis to explain the increased tryptophan utilization, although its incorporation in acute-phase proteins and the existence of other catabolic pathways cannot be excluded.

Comparison of heat-treated rapeseed expeller and solvent-extracted soya-bean meal as protein supplements for dairy cows given grass silage-based diets

AbstractSixteen early to mid lactation Finnish Ayrshire dairy cows were used in a cyclic change-over experiment with four 21-day experimental periods and a 4 5 2 factorial arrangement of treatments to evaluate the effects of heat-treated rapeseed expeller and solvent-extracted soya-bean meal protein supplements on animal performance. Dietary treatments consisted of grass silage offered ad libitum supplemented with a fixed amount of a cereal based concentrate (10 kg/day on a fresh weight basis) containing 120, 150, 180 or 210 g crude protein (CP) per kg dry matter (DM). Concentrate CP content was manipulated by replacement of basal ingredients (g/kg) with either rapeseed expeller (R; 120, 240 and 360) or soya-bean meal (S; 80, 160 and 240). Increases in concentrate CP stimulated linear increases (P &lt; 0·05) in silage intake (mean 22·5 and 23·8 g DM per g/kg increase in dietary CP content, for R and S, respectively) and milk production. Concentrate inclusion of rapeseed expeller elicited higher (P &lt; 0·01) milk yield and milk protein output responses (mean 108 and 3·71 g/day per g/kg DM increase in dietary CP content) than soya-bean meal (corresponding values 62 and 2·57). Improvements in the apparent utilization of dietary nitrogen for milk protein synthesis (mean 0·282 and 0·274, for R and S, respectively) were associated with higher (P &lt; 0·05) plasma concentrations of histidine, branched-chain, essential and total amino acids (35, 482, 902 and 2240 and 26, 410, 800 and 2119 µmol/l, respectively) and lower (P &lt; 0·01) concentrations of urea (corresponding values 4·11 and 4·52 mmol/l). Heat-treated rapeseed expeller proved to be a more effective protein supplement than solvent-extracted soya-bean meal for cows offered grass silage-based diets.

Effects of Dietary Long-Chain Polyunsaturated Fatty Acids on Plasma Amino Acids and Indices of Protein Metabolism in Infants: Results from a Randomized Clinical Trial

Background/Aim: Previous studies in vitro and in animals in vivo found that α-linolenic acid (C18:3ω3) may enhance oxidative damage of essential amino acids. We investigated whether the addition of the long-chain polyunsaturated fatty acids (LCPUFA) arachidonate (C20:4ω-6; AA) and docosahexaenoate (C22:6-ω3; DHA) in the form of egg phospholipids to infant formula affects plasma amino acid concentrations and indices of protein metabolism in term infants. Methods: In a double-blind, randomized clinical trial, healthy infants were fed from day 5 of life formula with or without preformed LCPUFA (n = 10 and 12, respectively). At the age of 5 days and 1, 2, 3 and 4 months, blood samples were obtained and analyzed for plasma amino acids by high-performance liquid chromatography and for plasma phospholipid fatty acid composition by gas chromatography. Results: At the age of 3 months, plasma threonine concentrations were significantly lower in infants receiving dietary LCPUFA than in controls (124 ± 16 vs. 216 ± 28 µmol/l, p < 0.05). Values of other plasma essential amino acids, total protein, albumin, creatinine and urea nitrogen did not differ between the two feeding groups throughout the study. At the age of 5 days, plasma phospholipid AA and DHA concentrations were inversely correlated with histidine concentrations (AA: r = –0.60, p = 0.01; DHA: r = –0.53, p < 0.05). At the age of 3 months, DHA concentrations were inversely related to plasma histidine, methionine and threonine concentrations (r = –0.66, –0.62, and –0.64, respectively, p < 0.05). Conclusions: The dietary LCPUFA supplementation of infant formula used in this study has no adverse effects on infant plasma amino acid concentrations and indicators of protein metabolism. Nonetheless, the apparent interaction of LCPUFA with some amino acids in formula-fed infants warrants further investigation.

Quick responses of plasma amino acid concentration to changes in dietary amino acid levels in rats.

In order to evaluate the availability of plasma amino acid (AA) concentration as a parameter to determine AA requirements of rats within a short period, the response of the plasma AA concentration to changes in dietary AA levels was studied. In Experiment 1, to clarify whether the plasma AA concentration responded to changes in dietary AA levels and how long the response of the plasma AA concentration was maintained, 10 male rats were fed a casein diet for a 7 day period, and then the food was changed to AA diets. The serine (Ser), glycine (Gly) and threonine (Thr) levels were decreased from 120, 90 to 60% of the casein diet at 7-day intervals successively. Blood samples were taken at 2, 4 and 7 days after changing the diet, from the vena caudalis by vein puncture. In all cases, the plasma AA concentration responded to changes in dietary AA within 2 days, and the response was maintained until the 7th day. Experiment 2 was conducted to confirm that the plasma concentration of each AA responded to the changes in dietary AA levels at the first day and the responses of the plasma concentration of AA were maintained until the 4th day after changing the dietary AA levels, when the combination of metabolically unrelated AA was added. Five rats in A groups were fed diets with added arginine + Thr, histidine + methionine, lysine or isoleucine + Ser, and five rats in B groups were fed diets with added Gly + valine, leucine + phenylalanine or Thr + tyrosine + valine for a 4-day period successively. Blood samples were taken at 1 and 4 days after changing diets. Although the plasma concentrations of histidine, leucine, methionine, phenylalanine and Ser did not respond to changes in dietary AA levels, those of other AA responded at 1 day and the responses were maintained until the 4th day.

L-Canavanine Influences Feed Intake, Plasma Basic Amino Acid Concentrations and Kidney Arginase Activity in Chicks

L-Canavanine [2-amino-4-(guanidinooxy) butyric acid], a non-protein amino acid that is structurally analogous to arginine, has been proposed as a major antinutritional factor responsible for the toxic effects induced by raw Canavalia ensiformis (L.) seeds in chicks. We investigated the effects of L-canavanine on performance and select metabolic responses of growing chicks. Canavanine was added to a control diet, in an amount equivalent to that provided by 300 g raw canavalia seeds/kg diet (10 g free base canavanine/kg diet). Growth, plasma basic amino acids and kidney arginase, activity were measured. The incorporation of canavanine into a nutritionally balanced diet for growing chicks depressed feed intake and growth by approximately 25% (P < 0.01) compared with the control diet. Performance was unaffected by equimolar amounts of arginine. Canavanine exerted its growth-depressing effect exclusively by reducing feed intake, because this effect was not observed in a pair-feeding experiment. Chicks fed a diet containing 473 mmol canavanine sulfate/kg for 11 d were given an intracrop dose of 946 mmol of canavanine sulfate or arginine hydrochloride. In both cases, plasma histidine and lysine concentrations were significantly decreased compared with a placebo group dosed with water. Plasma arginine concentration was unaffected by the canavanine sulfate dose but, as expected, was significantly increased by the arginine hydrochloride dose. Free base canavanine significantly (P < 0.05) reduced kidney arginase activity. No overt toxic effects were observed at any point during the study. These data indicate that, although canavanine is not the principal antinutritional factor in Canavalia ensiformis seeds, its presence in the diet precludes optimum performance of chicks.

Apportioning histidine requirements for maintenance versus growth

Purified diets containing six levels of histidine were fed to growing rats to evaluate histidine requirements for growth and for maintenance. A model was developed to subdivide the requirement for maintenance from the requirement for growth. Based on weight gain response, the histidine need for growth was 4.02 mg/g weight gain and the daily histidine need for maintenance was .130 times metabolic body size (Wg .75). Based on nitrogen gain response, the histidine need for growth was .168 mg/mg N gain and the daily histidine need for maintenance (mg) was .137 times metabolic body size (Wg .75). The plateau of plasma histidine concentration was reached at 22 mg intake/day. Previous histidine requirements for growth (for less than 5 g daily weight gain or 120 mg daily N retention) and for maintenance were in very close agreement with these estimates. As a percentage of retained protein, according to theses equations, histidine was calculated to be 2.7%; the reported histidine content of rat muscle CP is 2.85%.

Malnutrition in Hemodialysis Patients

Twenty-nine hemodialysis patients were studied to evaluate different laboratory measures in assessing muscle protein stores and detecting protein malnutrition. Arm muscle circumference (AMC) was used as a reference for somatic protein stores. AMC correlated with serum complement C3, plasma histidine, isoleucine, leucine, methionine and threonine concentrations as well as with body mass index. The lowest quartile of AMC was found most reliably by measuring plasma methionine, histidine, leucine and isoleucine concentrations. Protein malnutrition was detected in five patients (17%). They had significantly lower serum prealbumin and plasma leucine concentrations than the others. In the follow-up of 3 years every malnourished patient died, three from septic infection. Of the others only seven died, none from infection. The analysis of plasma essential amino acid and serum prealbumin concentrations had an important role in assessing muscle protein stores as well as the protein nutrition status in hemodialysis patients. Patients with malnutrition should be detected in view of their unfavourable prognosis.

Nutritional status affects renal 3-methylhistidine handling in humans

The plasma concentration of 3-methylhistidine (3MH) has been used in the assessment of nutritional status. Because urinary excretion is the sole route of elimination, and hence a potential determinant of the plasma concentration, 3MH renal handling was examined under different nutritional conditions. Ten obese women consumed a meat-free maintenance energy diet for 4 days before undergoing a 21-day total fast, after which they were assigned into two groups (n = 5) that received 7 days of hypocaloric feeding with 80 g of either milk protein or carbohydrate. 3MH excretion and plasma levels decreased in parallel during fasting and protein refeeding, suggesting that the plasma concentration accurately reflected a decrease of myofibrillar protein turnover, but carbohydrate refeeding was associated with a 31% increase in plasma 3MH ( P<.05) despite a concurrent 28% decrease in its excretion ( P<.05). This was due to a 41% decrease in 3MH renal clearance ( P<.05). Creatinine and urea clearances changed insignificantly in all diet periods. Plasma histidine (HIS) concentrations decreased with fasting and carbohydrate refeeding ( P<.05). 3MH clearance was found to be related to the plasma HIS 3MH concentration ratio ( r=.60, P<.001). Under some circumstances, an altered plasma HIS concentration appears to influence renal 3MH clearance and hence its plasma concentration.

Metabolic Effects of Histidine-Deficient Diets Fed to Growing Rats by Gastric Tube

Effects of histidine deficiency on muscle carnosine and anserine levels and on activities of enzymes associated with histidine catabolism and protoporphyrin synthesis were investigated. Male Sprague-Dawley (150 g) rats were tubefed isonitrogenous, isocaloric, defined diets containing 0%, low (0.013%) or adequate (0.45%) histidine for 8–13 days. While histidine-deficient animals maintained body weight, muscle and plasma histidine and carnosine concentrations decreased rapidly and remained low following a 3-day histidine repletion period. Hepatic histidine ammonia-lyase and histidine-pyruvate transaminase activities were decreased in histidine-deficient animals, whereas formiminotransferase activity was unchanged. Hematocrit levels and hemoglobin concentrations declined progressively during histidine depletion and the activity of erythrocyte and hepatic δ-aminolevulinic acid dehydratase also decreased relative to controls. Evidence is presented indicating that decreased histidine catabolism combined with carnosine and hemoglobin degradation can provide sufficient histidine to explain the slow onset of negative nitrogen balance associated with histidine deficiency and that impaired protoporphyrin synthesis may partially explain the anemia observed in the absence of dietary histidine.

UREA AS A SUPPLEMENTAL NITROGEN SOURCE FOR LACTATING COWS

Fifteen Holstein cows in early lactation were allocated to three groups for a 17-wk period. During a 4-wk preliminary period, all cows received a soybean meal concentrate to give a total average ration crude protein of 15%. In the next 4 wk, cows were gradually adapted to zero (ZU), low (LU), and high (HU) urea-containing rations. Based on total intake, cows consumed rations with 9, 11 and 12% crude protein. Depressed concentrate intake in the HU group due to decreased palatability limited total crude protein intake of that group to 12%. Milk yield was not stimulated by increasing dietary crude protein from 9 to 11 or 12% with urea. The percent milk persistency was not maintained by urea feeding when compared to persistency in the previous lactation at which time all cows received a 13% crude protein diet. Nitrogen balance for cows in ZU, LU and HU groups was −11.9, 13.3 and −3.6 g/day, respectively. The inclusion of urea in the ration increased ruminal NH3-N (mg/100 ml) from 1.32 for the ZU group to 3.94 and 5.78 for the LU and HU groups. Rumen microbial numbers (measured by DNA concentration) were decreased in all three groups when intact protein was removed from the ration. The sum of the plasma concentrations of histidine, isoleucine, leucine, lysine, threonine and valine were significantly decreased in cows fed the high urea-containing ration.

Effects of dietary histidine and arginine on plasma amino acid and urea concentrations of men fed a low nitrogen diet

The effects of dietary histidine and arginine on fasting and 1 and 2 hour postprandial plasma free amino acid and urea concentrations were studied in six young men. For 1 week each, they were fed six different diets containing 6.3 g of nitrogen daily. Each diet contained eight indispensable amino acids, cystine and tyrosine proportioned as in casein and a different mixture of dispensable nitrogen: A) six dispensable amino acids plus argine (diet 1) or plus histidine and arginine (diet 2) in the casein pattern, or B) an isonitrogenous amount of glycine and diammonium citrate alone (diet 3), with histidine (diet 4), with arginine (diet 5) or with histidine and arginine (diet 6). The fasting plasma concentrations of the seven indispensable amino acids assayed and their similar postprandial patterns were unaffected by the dietary treatments. Both fasting and postprandial plasma histidine concentrations were significantly lower when the histidine-low diets were fed than when the histidine-supplemented diets were fed. Histidine supplementation promoted a reduction in fasting plasma urea nitrogens. Proline concentrations were lowered significantly when proline was removed from the dietary amino acid mixtures, but plasma arginine concentrations were unaffected by arginine removal. Plasma histidine was maintained at lower concentrations in dietary histidine deficiency than when histidine was added to the low nitrogen diets.
