Effects of postbiotic products from Saccharomyces cerevisiae fermentation on lactation performance, antioxidant capacities, and blood immunity in transition dairy cows

This study aimed to evaluate the effects of dietary supplementation with different types of Saccharomyces cerevisiae fermentation products (SCFP) on lactational performance, metabolism, acute phase protein response, and antioxidant capacities in dairy cows from -21 to 56 d in milk (DIM). One hundred and 80 multiparous Holstein dairy cows were blocked by parity, expected calving date, pre-trial body condition score, and previous 305-d ME yield, and then randomly assigned to 1 of 3 dietary treatments: basal diet (CON; n = 60), basal diet supplemented with 40 g/d of SCFP1 (XPC; n = 60; XPC, Diamond V, Cedar Rapids, IA), and basal diet supplemented with 19 g/d of SCFP2 (NTK; n = 60, NutriTek®, Diamond V, Cedar Rapids, IA). Blood (n = 15, 13 and 12 in the CON, XPC and NTK groups, respectively) was sampled at -7 ± 3, + 3, + 7, + 21, and + 28 d, and milk samples (n = 19, 18 and 15 in the CON, XPC and NTK groups, respectively) was sampled during 1-8 wk from a subset of cows from -21 to 56 d relative to calving. Data were analyzed using the MIXED procedure in SAS (SAS Institute Inc.). All data were subjected to repeated measures ANOVA. Dietary treatment (TRT), time, and their interaction (TRT × time) were considered as fixed effects and cow as the random effect. Cows fed XPC and NTK had greater energy-corrected milk (ECM). Supplementing NTK increased milk fat content and yield, and 3.5% fat-corrected milk (FCM) yield compared with CON. Milk urea nitrogen (MUN) was lower in XPC cows than CON. SCFP supplementation decreased plasma β-hydroxybutyrate (BHB), ceruloplasmin (CER), haptoglobin (HPT), and interleukin-1β (IL-1β) concentrations, whereas increased plasma phosphorus (P) concentrations. In addition, cows fed NTK showed lower creatinine (CR) and cortisol (COR) concentrations but increased plasma calcium (Ca) and myeloperoxidase (MPO) concentrations than those in the CON cows. In addition, cows fed NTK and XPC both had reduced plasma concentrations of serum amyloid-A (SAA) at 3 DIM of lactation compared with CON fed cows. Furthermore, SCFP cows had greater concentrations of plasma glucose (GLU) and calcium (Ca) than CON cows at 7 DIM, and greater concentrations of plasma phosphorus (P) at 21 DIM. Between different SCFP type fed groups, plasma concentrations of nonesterified fatty acids (NEFA), MDA, creatinine (CR), SAA, and HPT were lower in cows fed NTK compared with cows fed XPC at 7 DIM. Overall, our results indicate the potential benefits of supplementing SCFP in transition dairy cows by modulating immunity, liver metabolic function and supporting ECM yield. The results also suggest that NutriTek at 19 g/d appears to support the performance and health of dairy cows better compared with XPC at 40 g/d, based on improved metabolic and inflammatory status during the transition period.

Effects of prepartum concentrate feeding on reticular pH, plasma energy metabolites, acute phase proteins, and milk performance in grass silage–fed dairy cows

We investigated how concentrate feeding during the last 21 d of pregnancy affects reticular pH, inflammatory response, dry matter (DM) intake, and production performance of dairy cows. We hypothesized that adding concentrates to dairy cows' diet before calving reduces the decrease in reticular pH postpartum and thus alleviates inflammatory response. We also hypothesized that prepartum concentrate feeding increases DM intake postpartum and consequently improves milk performance. Two feeding experiments were conducted using a randomized complete block design. In each experiment, 16 multiparous Finnish Ayrshire cows were paired based on parity, expected calving date, body weight, and milk yield of the previous lactation. Within the pairs, cows were randomly allocated on one of the 2 dietary treatments 21 d before expected calving. In experiment 1 (Exp1), diets were ad libitum feeding of grass silage as a sole feed or supplemented with increasing amounts of concentrate offered separately (increased to 4 kg/d by d -7). In experiment 2 (Exp2), diets were ad libitum feeding of a total mixed ration containing either grass silage, barley straw, and rapeseed meal (64%, 28%, and 8% on DM basis, respectively) or grass silage, barley straw, and cereal-based concentrate mixture (49%, 29%, and 30% on DM basis, respectively). Following calving, all the cows were fed similarly and observed until d 56 postpartum. Feed intake and milk yield were recorded daily, and reticular pH was monitored continuously by reticular pH bolus. Blood samples were collected at the beginning of the experiments, 7 d before the expected calving date, 1 d (in Exp1) or 5 d (in Exp2), 10 d, and 21 d postpartum. In Exp1, concentrate feeding increased metabolizable energy intake and tended to increase DM and crude protein intake prepartum. Moreover, prepartum concentrate feeding increased the concentrations of plasma β-hydroxybutyrate and insulin, but differences in nonesterified fatty acids, glucose, or acute phase proteins were not observed. After calving, prepartum diet did not affect DM or nutrient intake, plasma energy metabolites, or milk production in Exp1. Although prepartum concentrate feeding increased reticular pH on the first day of lactation, it elevated plasma concentrations of serum amyloid-A and haptoglobin postpartum in the grass silage-based diet. In Exp2, adding concentrates to the diet based on a mixture of grass silage and straw did not affect prepartum DM intake or plasma concentrations of nonesterified fatty acids, glucose, or insulin. Adding concentrates to prepartum diet increased plasma concentration of β-hydroxybutyrate before calving as in Exp1. After calving, prepartum concentrate feeding increased DM and nutrient intake during the second week of lactation in Exp2, but no effects were observed thereafter. In contrast to our hypothesis, prepartum concentrate feeding decreased reticular pH after calving in Exp2, but no differences in inflammatory markers were observed. Based on this study, close-up concentrate feeding in diets based on grass silage with or without straw does not alleviate the decrease in reticular pH or mitigate inflammatory response postpartum.

Evaluation of the systemic acute phase response and endometrial gene expression of serum amyloid A and pro- and anti-inflammatory cytokines in mares with experimentally induced endometritis

Infectious infertility in the mare is clinically well described, little is however known about the systemic acute phase reaction (APR) and local immunological responses accompanying equine endometritis. The aim of this study was to monitor selected markers of the APR in the systemic circulation and to correlate them to the local innate immune response in the uterus during infectious endometritis. Six adult standard bred mares received an intrauterine infusion of 10 9 CFU Escherichia coli. Blood samples were obtained before (0 h) and 3, 6, 12, 24, 36, 48, 72, 96 and 120 h post inoculation (pi), and endometrial biopsies were sampled before, and 3, 12, 24, 48 and 72 h pi. The infectious endometritis elicited a systemic APR with significantly increased concentrations of the acute phase proteins (APPs) serum amyloid A (SAA) and fibrinogen. Relative gene expression analyses were performed on extracted RNA from endometrial biopsies using quantitative real-time PCR and specific primers for SAA and pro- and anti-inflammatory cytokines. Expression of SAA was significantly up-regulated at 3 and 12 h pi, and a significant up-regulated expression of IL-1β, TNFα, IL-8 and IL-10 was observed at 3 h pi. Plasma concentration of SAA was significantly correlated to endometrial SAA expression. The results of the present study demonstrate that endometritis gives rise to a systemic APR and an up-regulated endometrial gene expression of SAA and several pro-and anti-inflammatory cytokines. Understanding endometrial expression of acute phase proteins and selected cytokines contributing to uterine immunity in equine endometritis could improve understanding of events leading to infertility in the mare and help identify candidate genes of mediators/markers for diagnostic use.

Feeding live cultures of Enterococcus faecium and Saccharomyces cerevisiae induces an inflammatory response in feedlot steers

Two experiments were conducted to investigate the effects of oral supplementation of the lactic-acid-producing bacterium Enterococcus faecium EF212 alone or in combination with Saccharomyces cerevisiae (yeast) on mediators of the acute phase response in feedlot steers. Eight fistulated steers were used to study the effects of E. faecium alone or with yeast in a crossover design with 2 Latin squares, 4 steers within each square, and 2 periods. The length of each period was 3 wk, with a 10-d adaptation and an 11-d measurement period. The experimental diet contained 87% steam-rolled barley, 8% whole-crop barley silage, and 5% supplement (DM basis). In Exp. 1, treatments were control vs. the lactic-acid-producing bacterium E. faecium (6 x 10(10) cfu/d). In Exp. 2, treatments were control vs. E. faecium (6 x 10(10) cfu/d) and S. cerevisiae (6 x 10(10) cfu/d). The bacteria and yeast supplements were blended with calcium carbonate to supply 6 x 10(10) cfu/d when top-dressed into the diet once daily at the time of feeding (10 g/d). Steers fed the control diet received only carrier (10 g/d). Blood samples were collected from the jugular vein on d 17 and 21 of each period, and serum amyloid A (SAA), lipopolysaccharide binding protein (LBP), haptoglobin, and alpha1-acid glycoprotein (alpha1-AGP) were measured. Supplementation of feed with E. faecium had no effect on concentrations of SAA, LBP, haptoglobin, or alpha1-AGP in plasma compared with those of controls. However, feeding E. faecium and yeast increased (P = 0.02) plasma concentrations of SAA, LBP, and haptoglobin but had no effect on plasma alpha1-AGP. In conclusion, oral supplementation of E. faecium alone had no effect on the mediators of the acute phase response that were measured, whereas feeding of E. faecium and yeast induced an inflammatory response in feedlot steers fed high-grain diets. Further research is warranted to determine the mechanism(s) by which E. faecium and yeast stimulated production of acute phase proteins in feedlot steers.

Urea‐induced denaturation of apolipoprotein serum amyloid A reveals marginal stability of hexamer

Serum Amyloid A (SAA) is an acute phase reactant protein that is predominantly found bound to high-density lipoprotein in plasma. Upon inflammation, the plasma concentration of SAA can increase dramatically, occasionally leading to the development of amyloid A (AA) amyloidosis, which involves the deposition of SAA amyloid fibrils in major organs. We previously found that the murine isoform SAA2.2 exists in aqueous solution as a hexamer containing a central channel. Here we show using various biophysical and biochemical techniques that the SAA2.2 hexamer can be totally dissociated into monomer by approximately 2 M urea, with the concerted loss of its alpha-helical structure. However, limited trypsin proteolysis experiments in urea showed a conserved digestion profile, suggesting the preservation of major backbone topological features in the urea-denatured state of SAA2.2. The marginal stability of hexameric SAA2.2 and the presence of residual structure in the denatured monomeric protein suggest that both forms may interconvert in vivo to exert different functions to meet the various needs during normal physiological conditions and in response to inflammatory stimuli.

The effect of serum amyloid protein A fragment-SAA25–76 on blood platelet aggregation

The effect of the major acute phase protein serum amyloid A (SAA) on collagen induced platelet aggregation was investigated. A truncated version of SAA, SAA25-76 was tested for inhibitory capacity of collagen induced platelet aggregation in platelet-rich plasma (PRP) and whole blood (WB). The tetrapeptide RGDV, a known inhibitor of platelet aggregation, was included as a reference compound. SAA25-76 at a concentration of 390 micrograms/ml inhibited platelet aggregation induced by 1 microgram/ml of collagen in PRP. This corresponds to the plasma concentration of SAA during an acute phase response. However, the inhibitory effect of the SAA25-76 fragment was lost at higher collagen concentrations (> or = 2.0 micrograms/ml). The SAA fragment at 390 micrograms/ml had no significant effect on platelet aggregation in WB. In contrast, RGDV blocked collagen induced platelet aggregation in both PRP and WB.

Concentrations of serum Amyloid‐a (SAA) and haptoglobin (HP) as parameters of inflammatory diseases in cattle

The concentrations of the acute-phase proteins, serum amyloid-A (SAA) and haptoglobin (Hp), were determined in the plasma of healthy cows (n = 25) and cows with spontaneous acute (n = 6), subacute (n = 37), or chronic (n = 7) inflammatory diseases. The plasma concentration of SAA alone, Hp alone, and the Hp/SAA ratio, differed significantly (p < 0.001) between healthy animals and animals with inflammatory diseases. Plasma Hp concentrations in the group of cows with acute inflammatory diseases were significantly (p < 0.01) different from those in the group with chronic inflammatory diseases. Moreover, the Hp/SAA ratio in chronic inflammatory diseases was significantly different from this ratio in acute (p < 0.01) and subacute (p < 0.05) inflammatory diseases. It is therefore concluded that the plasma concentrations of SAA and Hp and the Hp/SAA ratio are useful parameters to distinguish healthy animals from animals with inflammation and can be helpful in distinguishing between acute and chronic of inflammatory diseases.

Major acute-phase reactant synthesis during chronic inflammation in amyloid-susceptible and -resistant mouse strains

Hepatic levels of mRNA specific for total serum amyloid A (SAA), the SAA1 and SAA2 isotypes, serum amyloid P (SAP), C-reactive protein (CRP), and fibronectin, as well as the plasma concentrations of SAA and SAP were examined in amyloid-resistant (A/J) and amyloid-susceptible (CBA/J) mice during azocasein-induced chronic inflammation. In both strains hepatic SAA and SAP mRNA levels and plasma SAA and SAP protein concentrations increased dramatically during the early stages of inflammation; this was followed by a decrease to concentrations that were maintained at levels considerably higher than background. The ratios of SAA1 and SAA2 mRNA and plasma protein were 1:1 throughout. This indicated that there was no preferential accumulation of mRNA specifying a particular isotype and no preferential synthesis or clearance of a particular isotype during chronic inflammation and the early stages of amyloidogenesis in either strain. Similarly, hepatic SAP mRNA levels in both strains increased dramatically during the early stages of inflammation and were subsequently maintained at elevated levels. Plasma SAP concentrations increased rapidly during the first three days of the study in both A/J and CBA/J mice; however, during the later stages of inflammation, A/J plasma SAP levels decreased to a steady-state concentration that was approximately half that observed in CBA/J mice. Our results identify differences in the hepatic mRNA and plasma protein levels of the major mouse acute-phase reactants (APR) in the amyloid-resistant A/J and amyloid-susceptible CBA/J mouse strains. These findings are consistent with circulating inflammatory APR concentrations contributing, together with other factors, to the onset and pathogenesis of secondary amyloidosis.
