Influence of Cropping Sequence and Tillage System on Plant-Parasitic Nematodes and Peanut (Arachis hypogaea) Response to Fluopyram Applied at Planting

Crop sequence and tillage can affect the yield of peanut (Arachis hypogaea L.) and other crops. Research was conducted from 2006 through 2022 to determine the response of peanut to previous crop sequences including corn (Zea mays L.) and cotton (Gossypium hirsutum L.) planted in continuous conventional tillage (e.g., disking, field cultivating, and bedding with in-row sub-soiling) or strip tillage (e.g., tilling a 45 cm section on rows spaced 91 cm apart using fluted coulters, rolling baskets, and in-row sub-soiling). In 2013, 2019, and 2022, the entire test area was planted with peanut. In 2019 and 2022, peanut was planted without or with fluopyram applied in the seed furrow at planting. Decreasing the number of years between peanut planting resulted in lower peanut yields compared with fewer years of peanut planting in the rotation sequence. Continuous conventional tillage and strip tillage resulted in similar peanut yields at one location, while the yield was lower at a second location when peanut was planted in continuous strip tillage. Fluopyram did not affect peanut yield regardless of previous crop rotation sequence, the number of years separating peanut plantings, or the tillage system. However, minor differences in the populations of plant-parasitic nematodes in soil were noted when comparing fluopyram treatment. The results from these experiments indicate that while fluopyram can reduce the populations of some plant-parasitic nematodes in soil, the magnitude of reduction does not translate into increases in peanut yield.

Differences in parasitism of root-knot nematodes (Meloidogyne spp.) on oilseed radish and oat.

Oilseed radish and oat are cool season annual crops that are potentially used as “trap” or “biofumigant” crops for the suppression of plant-parasitic nematodes in soil. Cultivars of oilseed radish (Carwoodi, Cardinal, Final, Image, Concorde, Control, Eco-Till, Karakter and Cannavaro), white (Tachiibuki) and black (Pratex) oats were evaluated for their ability to reduce reproduction of three root-knot nematode species: Meloidogyne javanica, M. incognita race 3, and M. arenaria race 1. Nematode penetration and development were also evaluated using selected resistant and susceptible cultivars under greenhouse conditions. Root galling severity, number of eggs per gram of fresh root, and rate of reproduction varied among the cultivars in response to nematode infection. Oilseed radish cv. Carwoodi was resistant to M. javanica, whereas Karakter and Concorde were maintenance hosts allowing the nematode to maintain or increase its population on the plants. For M. incognita, Control and Carwoodi oilseed radish and Tachiibuki oat were resistant hosts. The cultivars that supported little reproduction of M. arenaria were Karakter and Carwoodi radish, and Tachiibuki oat. Comparable numbers of nematodes entered the roots of susceptible and resistant cultivars of oilseed radish and oat during early stages of infection. However, the development of the nematodes as evident from counting young and egg-laying females in roots were significantly decreased or inhibited in the resistant cultivars compared to the susceptible cultivars indicating that resistance occurs at post-infection stages. Histopathological examinations of galled-root tissues also revealed the susceptibility and resistance responses of selected cultivars of oilseed radish and oat to these nematode species.

Deletion of the Nonribosomal Peptide Synthetase Gene nps1 in the Fungus Clonostachys rosea Attenuates Antagonism and Biocontrol of Plant Pathogenic Fusarium and Nematodes.

Secondary metabolites produced by biological control agents may influence the outcome of their interactions with plant pathogenic microorganisms and plants. In the present study, we investigated the role of the nonribosomal peptide synthetase gene nps1 expressed by the biocontrol fungus Clonostachys rosea. A gene expression analysis showed that nps1 was induced during confrontations with the plant pathogenic fungus Botrytis cinerea. Gene deletion strains of nps1 displayed increased growth rates and conidiation. However, the nematicidal activity of culture filtrates from C. rosea Δnps1 strains was significantly weaker than that from wild-type filtrates (P ≤ 0.001); after 24 h of incubation with culture filtrates from nps1 deletion strains, only 13 to 33% of a mixed community of nematodes were dead compared with 42% of nematodes incubated with wild-type culture filtrates. The Δnps1 strains also showed reduced biocontrol efficacy during pot experiments, thus failing to protect wheat seedlings from foot rot disease caused by the plant pathogenic fungus Fusarium graminearum. Furthermore, C. rosea Δnps1 strains were not able to reduce populations of plant-parasitic nematodes in soil or in roots of wheat as efficiently as the wild-type strain. Both C. rosea wild-type and Δnps1 strains increased the dry shoot weight and shoot length of wheat by 20 and 13%, respectively. We showed that NPS1, a putative nonribosomal peptide synthetase encoded by nps1, is a biocontrol factor, presumably by producing a hitherto unknown nonribosomal peptide compound with antifungal and nematicidal properties that contributes to the biocontrol properties of C. rosea.

Performance of volatiles emitted from different plant species against juveniles and eggs of Meloidogyne incognita

Several plant species are known to control plant-parasitic nematodes in soil by producing nematicidal volatile organic compounds (VOCs). However, the VOCs activity of many plant species against root-knot nematodes is still unknown. Here, we investigated VOCs emitted from leaves of citronella (Cymbopogon nardus L.) or black pepper (Piper nigrum L.), broccoli shoots (Brassica oleracea L.) and Brazil nuts (Bertholletia excelsa Bonpl.) against second-stage juveniles (J2) and eggs of Meloidogyne incognita. In vitro assays revealed that VOCs emitted from dry plant macerates decreased J2 motility to almost 0% and reduced egg hatching by 47% in comparison with the control. The water exposed to VOCs from citronella, black paper and Brazil nuts decreased the motility of the J2 by 42%, whereas broccoli reduced it to almost 0%. Furthermore, VOCs from Brazil nuts and broccoli shoots killed J2 and reduced the number of galls and eggs in tomato roots under greenhouse conditions. Purified dimethyl sulfate (DMS), dimethyl disulfide (DMDS) and 3-pentanol, previously identified in the broccoli volatilome, were tested on eggs of M. incognita. DMDS and 3-pentanol were shown to reduce egg hatching by 96.8% at 176 mg/L and by 88.4% at 918 mg/L, respectively, whereas DMS increased egg hatching by 13% at 500 and 1000 mg/L. All plant species produced VOCs toxic to eggs and J2, which are the most important developmental stages in terms of nematode management and should be more exploited to reduce root-knot nematode populations in soils prior to host infection or used for prospection of novel nematicidal products.

Evaluation of Clonostachys rosea for Control of Plant-Parasitic Nematodes in Soil and in Roots of Carrot and Wheat.

Biological control is a promising approach to reduce plant diseases caused by nematodes. We tested the effect of the fungus Clonostachys rosea strain IK726 inoculation on nematode community composition in a naturally nematode infested soil in a pot experiment, and the effect of C. rosea on plant health. The numbers of plant-parasitic nematode genera extracted from soil and plant roots decreased by 40 to 73% when C. rosea was applied, while genera of nonparasitic nematodes were not affected. Soil inoculation of C. rosea increased fresh shoot weight and shoot length of wheat plants by 20 and 24%, respectively, while only shoot dry weight increased by 48% in carrots. Light microscopy of in vitro C. rosea-nematode interactions did not reveal evidence of direct parasitism. However, culture filtrates of C. rosea growing in potato dextrose broth, malt extract broth and synthetic nutrient broth exhibited toxicity toward nematodes and immobilized 57, 62, and 100% of the nematodes, respectively, within 48 h. This study demonstrates that C. rosea can control plant-parasitic nematodes and thereby improve plant growth. The most likely mechanism responsible for the antagonism is antibiosis through production of nematicidal compounds, rather than direct parasitism.

A novel nematode diagnostic method using the direct quantification of major plant-parasitic nematodes in soil by real-time PCR

We have developed a direct quantification method using real-time PCR for various plant-parasitic nematodes. Firstly, specific primers were designed for the root-knot nematode Meloidogyne incognita, the root-lesion nematode Pratylenchus penetrans, the potato cyst nematode Globodera rostochiensis and the soybean cyst nematode Heterodera glycines. A DNA extraction method was then developed beginning with 20 g of soil, a relatively large amount of soil but a necessary amount in the consideration of heterogeneous distribution of nematodes in soil. To estimate the density of the target nematode in soil, calibration curves for each plant-parasitic nematode were obtained by inoculating different numbers of the target nematode and then extracting DNA from the soils. The detection limit was 4-5 nematodes (20 g soil)−1. This method was applied to nematode diagnostics. Soil sampling was done when transplanting of radish and sweet potato in fields was taking place, and the density of plant-parasitic nematodes was measured using both the Baermann funnel extraction method and real-time PCR methods. In some soils, P. penetrans and M. incognita were not found with the Baermann method but were detected with the real-time PCR method. At harvest, damage to crops was evaluated and its relationship with initial densities was investigated. The real-time PCR method more precisely predicted damage to radish and sweet potato by nematodes and was considered to be a powerful tool in the diagnosis of nematode diseases.

Management of crotalaria and pigeon pea for control of yam nematode diseases

Management of plant-parasitic nematodes with the use of nematicides has not been recommended for small farmers that grow yam in the Northeastern region of Brazil, due to its high cost and residue toxicity. The use of plants with antagonistic effect to nematodes and green manure which improves soil chemical, physical and biological characteristics can be a viable and low cost alternative to control parasitic nematodes. This work aimed to evaluate the effect of crotalaria (Crotalaria juncea) and pigeon pea (Cajanus cajan) plants on the control of yam nematodes. Three experiments were carried out. The first was conducted under in vitro conditions to evaluate the nematostatic and nematicide effect of extracts from fresh and dry matter of the above ground parts of crotalaria, pigeon pea, and the combination of both. The second experiment was carried out under greenhouse conditions to evaluate the effect of soil amendment with crotalaria, pigeon pea, and the combination of both in the infectivity of Scutellonema bradys, using tomato plants as the host plant. The third experiment was conducted under field conditions to evaluate the effect of crotalaria, pigeon pea, and the combination of both, cultivated between yam planting rows and incorporated to soil surface, on yam nematodes. The aqueous extract obtained form fresh matter of crotalaria had a nematicide effect of 100% for S. bradys. Extracts from dry matter of both crotalaria and pigeon pea did not have any nematicide effect, but had a nematostatic effect. Incorporation of crotalaria to soil inhibited infectivity of S. bradys in tomato seedlings. These results showed that planting crotalaria alone or in combination with pigeon pea, between the yam planting rows, is an efficient method for controlling S. bradys and Rotylenchulus reniformis associated with yams. Crotalaria can be used for controlling these plant-parasitic nematodes in soil.

Potential of Microwaves to Control Plant-Parasitic Nematodes in Soil

Microwave radiation of2450 MHz frequency was used to irradiate sandy loam soil placed in 12 cm high and 10 cm dia columns as a function of exposure times of30, 45, 60, and 120 s. This was done to evaluate the effect of radiation on the highest soil temperature attained and subsequent temperature patterns in relation to time. Soil columns were packed to a field bulk density of approximately 1.4 g/cm3, and treatments consisted of moist soil, dry soil, and layers of moist and dry soil of varying thicknesses. Moisture contents of moist and dry soil were 10% and 2%, respectively, on a dry mass basis. An exposure time of 45 seconds was the most efficient in yielding soil temperatures high enough to kill plant-parasitic nematodes. Irradiation of soil infested with Rotylenchulus reniform nematodes for 45 seconds resulted in a 99% extermination of the organisms in all treatments. However, radiation proved to be most effective in nematode control with 6.0 cm dry soil placed over 6.0 cm moist soil.

Occurrence and importance of plant-parasitic nematodes in organic farming in Germany

Abstract In an attempt to evaluate the occurrence and economic importance of plant-parasitic nematodes in organic farming in Germany, a survey was conducted with the main emphasis on vegetable and cereal production systems. For vegetables, the survey included quantification and identification of plant-parasitic nematodes in soil samples and a questionnaire for growers querying production factors and damage levels. For cereals, the survey focused on quantification and identification of plant-parasitic nematodes in soil and plant samples. Overall, Pratylenchus and Tylenchorhynchus were the most prominent nematode genera under both production systems with an incidence of over 90% of the samples. Meloidogyne was detected in 51% of the samples in both systems. Other nematode genera showed differences between the two production systems. In production systems with a high frequency of vegetables, Paratylenchus was detected in 56% of the samples and Heterodera in 15%, whereas in rotations with a high cropping frequency of cereals, incidences of plant-parasitic nematodes were 56% for Heterodera, 47% for Trichodorus and 45% for Paratylenchus. Yield losses could exceed 50% on carrots, onions and cereals and were most pronounced on sandy soils. In many cases, nematode problems started 5 to 10 years after conversion to organic farming. The survey indicated that plant-parasitic nematodes are widely spread in organic farming in Germany and can cause severe damage which may result in complete loss of the crop.

Sensitivity of Meloidogyne javanica and Tylenchulus semipenetrans to Isothiocyanates in Laboratory Assays

ABSTRACT Isothiocyanates are released through enzymatic degradation of glucosinolates produced by plants in the family Brassicaceae. Glucosinolate profiles differ among plant species and the isothiocyanate derivatives differ in their toxicity to nematodes. Control of plant-parasitic nematodes in soil by isothiocyanates released from incorporated brassicaceous plant material has been inconsistent. Success might be improved with knowledge of the relative toxicities of various isothiocyanates against nematodes. Laboratory assays were conducted to determine lethal concentration (LC) values in sand of seven commercially available isothiocyanates against Tylenchulus semipenetrans and Meloidogyne javanica. The LC(90) values were 0.01 and 0.03 mumol/ml for 2-phenylethyl isothiocyanate and 0.48 and 0.35 mumol/ml for phenyl isothiocyanate for T. semipenetrans and M. javanica, respectively. Brassicaceous sources of benzyl or 2-phenylethyl isothiocyanate and, to a lesser extent allyl isothiocyanate, are the most promising candidates for plant-parasitic nematode management. The broader context of this research is the development of approaches for consistent and reliable use of plant-derived chemicals for nematode management. The strategy is to select plants in the family Brassicaceae based on their glucosinolate profiles and the sensitivity of the target nematode species to the associated isothiocyanates.

Nematocide residues in plants.

Although one nematocidal chemical was in use before 1900, widespread use of chemicals for the control of plant-parasitic nematodes in soil is a development of the past 16 years. The most popular and useful of these are volatile materials popularly known as “soil fumigants”. These are usually applied by placement 15 or 20 cm. beneath the soil surface in lines about 30 cm. apart. This application method is known as “area treatment”. A popular alternative method is placement of the nematocide beneath the planting rows only, leaving the intervals between rows untreated. This is known as “row treatment”. The fumigants diffuse from the application points through the upper 45 to 60 cm. of the soil. Other soil nematocides are applied in water solution or emulsion or are in granular form to be mixed with the soil. All are more or less toxic to plants and are usually applied to the soil several days or weeks before planting; a few are sufficiently non-toxic to certain kinds of plants so that they can be applied at planting time or even around the roots of growing plants.
