Characterization of a novel root-associated diazotrophic rare PGPR taxa, Aquabacter pokkalii sp. nov., isolated from pokkali rice: new insights into the plant-associated lifestyle and brackish adaptation

Salinity impacts crop growth and productivity and lowers the activities of rhizosphere microbiota. The identification and utilization of habitat-specific salinity-adapted plant growth-promoting rhizobacteria (PGPR) are considered alternative strategies to improve the growth and yields of crops in salinity-affected coastal agricultural fields. In this study, we characterize strain L1I39T, the first Aquabacter species with PGPR traits isolated from a salt-tolerant pokkali rice cultivated in brackish environments. L1I39T is positive for 1-aminocyclopropane-1-carboxylate deaminase activity and nitrogen fixation and can promote pokkali rice growth by supplying fixed nitrogen under a nitrogen-deficient seawater condition. Importantly, enhanced plant growth and efficient root colonization were evident in L1I39T-inoculated plants grown under 20% seawater but not in zero-seawater conditions, identifying brackish conditions as a key local environmental factor critical for L1I39T-pokkali rice symbiosis. Detailed physiological studies revealed that L1I39T is well-adapted to brackish environments. In-depth genome analysis of L1I39T identified multiple gene systems contributing to its plant-associated lifestyle and brackish adaptations. The 16S rRNA-based metagenomic study identified L1I39T as an important rare PGPR taxon. Based on the polyphasic taxonomy analysis, we established strain L1I39T as a novel Aquabacter species and proposed Aquabacter pokkalii sp nov. Overall, this study provides a better understanding of a marine-adapted PGPR strain L1I39T that may perform a substantial role in host growth and health in nitrogen-poor brackish environments.

Microbacterium azadirachtae CNUC13 Enhances Salt Tolerance in Maize by Modulating Osmotic and Oxidative Stress.

Soil salinization is one of the leading threats to global ecosystems, food security, and crop production. Plant growth-promoting rhizobacteria (PGPRs) are potential bioinoculants that offer an alternative eco-friendly agricultural approach to enhance crop productivity from salt-deteriorating lands. The current work presents bacterial strain CNUC13 from maize rhizosphere soil that exerted several PGPR traits and abiotic stress tolerance. The strain tolerated up to 1000 mM NaCl and 30% polyethylene glycol (PEG) 6000 and showed plant growth-promoting (PGP) traits, including the production of indole-3-acetic acid (IAA) and siderophore as well as phosphate solubilization. Phylogenetic analysis revealed that strain CNUC13 was Microbacterium azadirachtae. Maize plants exposed to high salinity exhibited osmotic and oxidative stresses, inhibition of seed germination, plant growth, and reduction in photosynthetic pigments. However, maize seedlings inoculated with strain CNUC13 resulted in significantly improved germination rates and seedling growth under the salt-stressed condition. Specifically, compared with the untreated control group, CNUC13-treated seedlings exhibited increased biomass, including fresh weight and root system proliferation. CNUC13 treatment also enhanced photosynthetic pigments (chlorophyll and carotenoids), reduced the accumulation of osmotic (proline) and oxidative (hydrogen peroxide and malondialdehyde) stress indicators, and positively influenced the activities of antioxidant enzymes (catalase, superoxide dismutase, and peroxidase). As a result, CNUC13 treatment alleviated oxidative stress and promoted salt tolerance in maize. Overall, this study demonstrates that M. azadirachtae CNUC13 significantly enhances the growth of salt-stressed maize seedlings by improving photosynthetic efficiency, osmotic regulators, oxidative stress resilience, and antioxidant enzyme activity. These findings emphasize the potential of utilizing M. azadirachtae CNUC13 as a bioinoculant to enhance salt stress tolerance in maize, providing an environmentally friendly approach to mitigate the negative effects of salinity and promote sustainable agriculture.

Streptomyces N2A, an endophytic actinobacteria that promotes soybean growth and increases yield and seed quality under field conditions

Sustainable agriculture based on the use of soil-beneficial microbes such as plant growth-promoting rhizobacteria (PGPR) and biocontrol agents (BCA) is gaining great consideration to reduce the use of agrochemicals for crop production. With this aim, in this study, a total of 78 actinobacteria were isolated from the rhizosphere and endosphere of soybean roots. Based on in vitro compatibility with Bradyrhizobium japonicum, the ability to produce phytohormones, siderophores, exo-enzymes, antifungal compounds and phosphate solubilization (PGPR traits), two endophytic strains, named N2A and N9, were selected to evaluate their effects on plant growth and development at greenhouse and field conditions. Greenhouse trials showed significantly promoted seedling emergence compared to control and the conventional fungicide treatment. Analysis of growth and development associated parameters at reproductive stages and maturity at greenhouse, but also and most importantly, in field experiments showed significant improvements. Plant biomass, node number, pod number, and consequently yield, were higher in plants previously treated with N2A and co-inoculated with B. japonicum compared to the conventional seed treatment. Furthermore, a significant increase in health status and vigor was observed for seeds harvested from the N2A-treated plants in relation to seeds obtained from the conventional treatment. Thus, we demonstrated that Streptomyces sp. N2A can replace traditional chemical fungicides to protect the seed during germination, allowing good implantation, but also, stimulating the growth and development of soybean crop increasing yield and seed quality at field conditions. Altogether, this supports the potential use of Streptomyces N2A as a PGPR for soybean crop production more efficiently and sustainably.

Biocontrol rhizobacteria enhances growth and yield of wheat (Triticum aestivum) under field conditions against Fusarium oxysporum

ABSTRACTThe current study aimed to identify the survival of bio-control bacteria with antifungal activity against Fusarium oxysporum and assess their growth promoting activity in wheat crop field conditions. To evaluate the fungicidal activities of isolated bacteria using the dual culture method, both qualitative and quantitative bioassays were performed. Plant Growth Promoting activities such as Indole 3-Acetic Acid (IAA), phosphate solubilization, Hydrogen cyanide (HCN), and Siderophore production were assessed for three biocontrol bacterial isolates (BCB 07, BCB16, and BCB 83) out of 180 with 70% antagonistic activity against Fusarium oxysporum. Chitinase, protease, and cellulase interaction in isolates was also tested. BCB16 was selected as it had 70% antagonist activity against F. oxysporum but also had the highest PGPR (Plant Growth Promoting Rhizobacteria) traits when compared to the other two isolates. BCB16 was also tested for survival in talc powder and in wheat crop field conditions. Even after 4 months in talc powder, the survival rate remained stable. In a wheat crop field, BCB16 reduced the disease incidence of Fusarium oxysporum by 54.38%. When compared to fungus alone treatment, BCB16 increased average yield by 57% alone and 32% in challenged conditions. BCB16 was identified molecularly using the 16s rRNA gene. Bacillus amyloliquefaciens shared 97% of the deduced sequence. The sequence was submitted to genbank and assigned the accession number OM333889. Bacillus amyloliquefaciens has the potential to be used in the field as an alternative to synthetic fungicides against Fusarium oxysporum.

Evaluation of the growth-inducing efficacy of various Bacillus species on the salt-stressed tomato (Lycopersicon esculentum Mill.).

Plants are affected by salt stress in a variety of ways, including water deficiency, ion toxicity, nutrient imbalance, and oxidative stress, all of which can cause cellular damage or plant death. Halotolerant plant growth-promoting rhizobacteria (PGPR) could be a viable alternative for tomato plants growing in arid and semi-arid environments. The aim of this research was to isolate halotolerant plant growth promoting Bacillus sp. to promote tomato (Lycopersicon esculentum Mill.) growth and salt stress resistance. 107 PGPR strains were isolated from the rhizospheres of 'Kesudo' (Butea monosperma Lam.), 'Kawaria' (Cassia tora L.), and 'Arjun' (Terminalia arjuna Roxb.) plants to test their plant growth promoting abilities, including indole-3-acetic acid, phosphate solubilization, siderophore production, and ACC deaminase activity. Five bacterial strains (Bacillus pumilus (NCT4), Bacillus firmus (NCT1), Bacillus licheniformis (LCT4), Bacillus cereus (LAT3), and Bacillus safensis (LBM4)) were chosen for 16S rRNA on the basis of PGPR traits. Compared to PGPR untreated plants, tomato plants developed from PGPR-treated seeds had considerably increased germination percentage, seedling growth, plant height, dry weight, and leaf area. As comparison to PGPR non-inoculated plants, salt-stressed tomato plants treated with PGPR strains had higher levels of total soluble sugar, proline, and chlorophyll as well as higher levels of SOD, CAT, APX, and GR activity. PGPR-inoculated salt-stressed tomato plants had lower MDA, sodium, and chloride levels than non-inoculated plants. In addition, magnesium, calcium, potassium, phosphorus, and iron levels were higher in PGPR treated plants when subjected to salt stress. These results indicate that halotolerant PGPR strains can increase tomato productivity and tolerance to salt stress by removing salt stress's negative effects on plant growth.

Co-Inoculation of Non-Symbiotic Bacteria Bacillus and Paraburkholderia Can Improve the Soybean Yield, Nutrient Uptake, and Soil Parameters.

Due to its nutritional value and oil, soybean (Glycine max L.) became an economic crop in India and worldwide. The current study investigated the effect of forest-associated plant growth-promoting rhizobacteria (PGPR) on soybean yield and grain nutrient content. Five potential bacteria were used in this study based on their PGPR traits. The pot assay result with two crops (soybean and chickpea) confirmed the growth promotion activity of the two strains (Bacillus subtilis MpS15 and Paraburkholderia sabiae NvS21). The result showed significant (p < 0.05) enhancement in plant length and biomass with the seed treatment with strains (MpS15 and NvS21) compared to the control. Later both biocompatible potential strains were used in field experiments as individuals and consortia. Seed treatment of consortia significantly improves the nodulation and photosynthetic content more than individual treatments and control. Compared to the control, the co-inoculation of MpS15 and NvS21 increased soybean grain, straw yield, and grain NPK contents. Interestingly, soil parameters (organic carbon, available NPK) showed a strong correlation (p < 0.05) with plant parameters and nutrient uptake. Overall, our study provides strong relationships between soil parameters, microbial inoculum as consortia, and soybean performance, and these strains may be utilized as bioinoculant in future.

Temperature-induced modulation of stress-tolerant PGP genes bioprospected from Bacillus sp. IHBT-705 associated with saffron (Crocus sativus) rhizosphere: A natural -treasure trove of microbial biostimulants.

There is a renewed interest in sustainable agriculture wherein novel plant growth-promoting rhizobacteria (PGPR) are being explored for developing efficient biostimulants. The key requirement of a microbe to qualify as a good candidate for developing a biostimulant is its intrinsic plant growth-promoting (PGP) characteristics. Though numerous studies have been conducted to assess the beneficial effects of PGPRs on plant growth under normal and stressed conditions but not much information is available on the characterization of intrinsic traits of PGPR under stress. Here, we focused on understanding how temperature stress impacts the functionality of key stress tolerant and PGP genes of Bacillus sp. IHBT-705 isolated from the rhizosphere of saffron (Crocus sativus). To undertake the study, Bacillus sp. IHBT-705 was grown under varied temperature regimes, their PGP traits were assessed from very low to very high-temperature range and the expression trend of targeted stress tolerant and PGP genes were analyzed. The results illustrated that Bacillus sp. IHBT-705 is a stress-tolerant PGPR as it survived and multiplied in temperatures ranging from 4°C-50°C, tolerated a wide pH range (5-11), withstood high salinity (8%) and osmolarity (10% PEG). The PGP traits varied under different temperature regimes indicating that temperature influences the functionality of PGP genes. This was further ascertained through whole genome sequencing followed by gene expression analyses wherein certain genes like cspB, cspD, hslO, grpE, rimM, trpA, trpC, trpE, fhuC, fhuD, acrB5 were found to be temperature sensitive while, cold tolerant (nhaX and cspC), heat tolerant (htpX) phosphate solubilization (pstB1), siderophore production (fhuB and fhuG), and root colonization (xerC1 and xerC2) were found to be highly versatile as they could express well both under low and high temperatures. Further, the biostimulant potential was checked through a pot study on rice (Oryza sativa), wherein the application of Bacillus sp. IHBT-705 improved the length of shoots, roots, and number of roots over control. Based on the genetic makeup, stress tolerance potential, retention of PGP traits under stress, and growth-promoting potential, Bacillus sp. IHBT-705 could be considered a good candidate for developing biostimulants.

Bacillus velezensis QA2 Potentially Induced Salt Stress Tolerance and Enhanced Phosphate Uptake in Quinoa Plants.

Plant Growth-Promoting Rhizobacteria (PGPR) have attracted much attention in agriculture biotechnology as biological inputs to sustain crop production. The present study describes a halotolerant phosphate solubilizing bacterium associated with quinoa plant roots. Based on a metabolic screening, one bacterial isolate, named QA2, was selected and screened for PGPR traits. This isolate solubilized both inorganic phosphate and zinc, produced indole-3-acetic acid, ammonia, hydrogen cyanide, cellulase, and (to be deleted) protease, and induced biofilm formation. We demonstrated that QA2 exhibited both antimicrobial and ion metabolism activities and tolerated high salt concentration at up to 11% NaCl. Genotyping analyses, using 16S rRNA and chaperonin cpn60 genes, revealed that QA2 belongs to the species of Bacillus velezensis. Using the quinoa model cultivated under a saline condition, we demonstrated that QA2 promoted plant growth and mitigated the saline irrigation effects. Analysis of harvested plants revealed that QA2 induced a significant increase of both leaf chlorophyll index by 120.86% (p < 0.05) and P uptake by 41.17% (p < 0.05), while the content of Na+ was drastically decreased. Lastly, a bibliometric data analysis highlighted the panoramic view of studies carried out so far on B. velezensis strains. Our investigation presents a holistic view of the potential application of B. velezensis as a biological inoculant to promote plant growth, control pathogen attacks, and mitigate the salinity effect of quinoa plants. Further investigations are still needed to demonstrate these effects in field conditions.

Revealing plant growth-promoting mechanisms of Bacillus strains in elevating rice growth and its interaction with salt stress.

Soil salinity is a major environmental stress that has been negatively affecting the growth and productivity of rice. However, various salt-resistant plant growth-promoting rhizobacteria (PGPR) have been known to promote plant growth and alleviate the damaging effects of salt stress via mitigating physio-biochemical and molecular characteristics. This study was conducted to examine the salt stress potential of Bacillus strains identified from harsh environments of the Qinghai-Tibetan plateau region of China. The Bacillus strains NMTD17, GBSW22, and FZB42 were screened for their response under different salt stress conditions (1, 4, 7, 9, 11, 13, and 16%). The screening analysis revealed strains NMTD17, GBSW22, and FZB42 to be high-salt tolerant, moderate-salt tolerant, and salt-sensitive, respectively. The NMTD17 strain produced a strong biofilm, followed by GBSW22 and FZB42. The expression of salt stress-related genes in selected strains was also analyzed through qPCR in various salt concentrations. Further, the Bacillus strains were used in pot experiments to study their growth-promoting ability and antioxidant activities at various concentrations (0, 100, 150, and 200 mmol). The analysis of growth-promoting traits in rice exhibited that NMTD17 had a highly significant effect and GSBW22 had a moderately significant effect in comparison with FZB42. The highly resistant strain NMTD17 that stably promoted rice plant growth was further examined for its function in the composition of rhizobacterial communities. The inoculation of NMTD17 increased the relative abundance and richness of rhizobacterial species. These outcomes propose that NMTD17 possesses the potential of PGPR traits, antioxidants enzyme activities, and reshaping the rhizobacterial community that together mitigate the harmful effects of salinity in rice plants.

​Induction of Systemic Resistance against Phytophthora Blight in Pigeonpea Through the Interaction of Plant Growth Promoting Rhizobacteria: In vivo and in vitro Study

Background: Phytophthora blight (PB), caused by Phytophthora drechsleri f. sp. cajani, is soil borne disease of pigeonpea (Cajanus cajan L.). The use of plant growth promoting rhizobacteria (PGPR) in agriculture leads to alternative disease management of crop in a sustainable ecofriendly way. Methods: In vitro and in vivo experimentation was conducted at fields of Punjab Agricultural University, Ludhiana during kharif season of 2017-18 to study the percentage of blight inhibition by rhizobacteria and production potential in pigeonpea. The present study consisted of 6 treatments of dual inoculation of antagonistic bacteria with recommended rhizobium of PAU (LAR-06). Result: The selected rhizoisolates were studied against Phytophthora drechsleri caused stem blight of pigeonpea. The allelochemicals and plant growth promoting rhizobacteria traits involved in antagonistic behavior significantly inhibited the growth of the test fungus. Scanning electron microscopy between potential rhizoisolates and test fungus revealed intumescent hyphae with irregular cell surface morphology. The synergistic effects of LAR-06 + S-2 and LAR-06+ S-18 were found to be the most potent ones in inhibiting the radial growth of the test pathogen with increased yield production in the field as well as greenhouse conditions.

Coal solubilizing bacteria (CSB) are microorganisms to able to bio transformed low rank coal, releasing humified organic matter in the process. On the other hand, these bacterial genera have reported previously as plant growth promoting bacteria. The aim of this work was to assess the Plant Growth Promoting Rhizobacteria (PGPR) capacity of five CSB strains: Bacillus pumilus (CSB05), B. mycoides (CSB25), Microbacterium sp. (CSB3), Acinetobacter sp. (CSB13) and B. amyloliquefaciens (CSB02). For this, the PGPR traits of CSB were evaluated under laboratory conditions: the biological nitrogen fixation capacity, the reduction of acetylene, the synthesis of indole acetic acid (IAA) and the solubilization of phosphates. In a second experiment under plant nursery conditions, PGPR activity of strain CSB05 was evaluated in common bean plants. Under laboratory conditions, it was evidenced that all the evaluated strains produced IAA, solubilized phosphate in a liquid medium, presented atmospheric nitrogen fixation capacity, and only the CSB3 and CSB13 strains reduced acetylene. In the plant nursery experiment, PGPR activity of strain CSB05 was detected in common bean plants, reflected in increases in the height of these plants. These results show that CSB are promising in the PGPR activity, which is interesting to the design of biological products with agricultural and environmental applications, for the management of crops in disturbed soils of the Colombian dry Caribbean.

Biological Control Activity of Plant Growth Promoting Rhizobacteria Burkholderia contaminans AY001 against Tomato Fusarium Wilt and Bacterial Speck Diseases.

Simple SummaryBurkholderia contaminans belongs to B. cepacia complex (Bcc), those of which are found in various environmental conditions. In this study, a novel strain AY001 of B. contaminans (AY001) was identified from the rhizosphere soil sample. AY001 showed (i) various plant growth-promoting rhizobacteria (PGPR)-related traits, (ii) antagonistic activity against different plant pathogenic fungi, (iii) suppressive activity against tomato Fusarium wilt disease, (iv) induced systemic acquired resistance (ISR)-triggering activity, and (v) production of various antimicrobial and plant immune-inducing secondary metabolites. These results suggest that AY001 is, indeed, a successful PGPR, and it can be practically used in tomato cultivation to alleviate biotic and abiotic stresses. However, further safety studies on the use of AY001 will be needed to ensure its safe use in the Agricultural system.Plant growth promoting rhizobacteria (PGPR) is not only enhancing plant growth, but also inducing resistance against a broad range of pathogens, thus providing effective strategies to substitute chemical products. In this study, Burkholderia contaminans AY001 (AY001) is isolated based on its broad-spectrum antifungal activity. AY001 not only inhibited fungal pathogen growth in dual culture and culture filtrate assays, but also showed various PGPR traits, such as nitrogen fixation, phosphate solubilization, extracellular protease production, zinc solubilization and indole-3-acetic acid (IAA) biosynthesis activities. Indeed, AY001 treatment significantly enhanced growth of tomato plants and enhanced resistance against two distinct pathogens, F. oxysporum f.sp. lycopersici and Pseudomonas syringae pv. tomato. Real-time qPCR analyses revealed that AY001 treatment induced jasmonic acid/ethylene-dependent defense-related gene expression, suggesting its Induced Systemic Resistance (ISR)-eliciting activity. Gas chromatography–mass spectrometry (GC-MS) analysis of culture filtrate of AY001 revealed production of antimicrobial compounds, including di(2-ethylhexyl) phthalate and pyrrolo [1,2-a]pyrazine-1,4-dione, hexahydro-3-(phenylmethyl). Taken together, our newly isolated AY001 showed promising PGPR and ISR activities in tomato plants, suggesting its potential use as a biofertilizer and biocontrol agent.

Alleviation of Salt Stress via Habitat-Adapted Symbiosis

Halotolerant plant-growth-promoting rhizobacteria (PGPR) could not only promote plant growth, but also help in counteracting the detrimental effects of salt stress. In the present study, a total of 76 bacteria were isolated from the rhizosphere, non-rhizospheric soil and endophytes of the halophyte Salsola tetrandra, collected from natural saline soils in Algeria. Phylogenetic analysis based on the 16S rDNA sequence of Gram-negative bacteria (n = 51) identified, showed seventeen representative isolates grouped into four genera (Pseudomonas, Acinetobacter, Enterobacter, and Providencia). These bacterial isolates that exhibited different PGPR traits were selected and tested for their ability to tolerate different abiotic stress (NaCl, PEG8000, and pH). The majority of isolates were drought tolerant (60% of PEG8000) and had an optimal growth at high pH values (pH 9 and 11) and some strains tolerated 2 M of NaCl. Strains identified as Enterobacter xiangfangensis BE1, Providencia rettgeri BR5 and Pseudomonas stutzeri MLR6 showed high capacity of adaptation on their PGP traits. The salt-tolerant isolates were finally chosen to promote growth and enhance salt tolerance, separately or combined, of Arabidopsis thaliana (Col-0) exposed or not to 0.1 M NaCl, by following fresh and root weight, primary root elongation and lateral root number. The best bacterial effect was recorded for the MLR6 strain in increasing shoot fresh weight and for BE1 in terms of root fresh weight in the absence of salt stress. At stressed conditions, all growth parameters declined. However, inoculation of Arabidopsis thaliana with the three bacterial strains (MLR6, BE1 and BR5), single or in co-culture, conferred an increase in the shoot weight, primary root length and lateral root number. The use of these strains separately or combined as biofertilizers seems to be a powerful tool in the development of sustainable agriculture in saline soils.

The Genome of Bacillus velezensis SC60 Provides Evidence for Its Plant Probiotic Effects.

Root colonization and plant probiotic function are important traits of plant growth-promoting rhizobacteria (PGPR). Bacillus velezensis SC60, a plant endophytic strain screened from Sesbania cannabina, has a strong colonization ability on various plant roots, which indicates that SC60 has a preferable adaptability to plants. However, the probiotic function of the strain SC60 is not well-understood. Promoting plant growth and suppressing soil-borne pathogens are key to the plant probiotic functions. In this study, the genetic mechanism of plant growth-promoting and antibacterial activity of the strain SC60 was analyzed by biological and bioinformatics methods. The complete genome size of strain SC60 was 3,962,671 bp, with 4079 predicted genes and an average GC content of 46.46%. SC60 was designated as Bacillus velezensis according to the comparative analysis, including average nucleotide polymorphism (ANI), digital DNA–DNA hybridization (dDDH), and phylogenetic analysis. Genomic secondary metabolite analyses indicated two clusters encoding potential new antimicrobials. The antagonism experiments revealed that strain SC60 had the ability to inhibit the growth of a variety of plant pathogens and its closely related strains of Bacillus spp., which was crucial to the rhizospheric competitiveness and growth-promoting effect of the strain. The present results further suggest that B. velezensis SC60 could be used as a PGPR strain to develop new biocontrol agents or microbial fertilizers.

In-Depth Characterization of Plant Growth Promotion Potentials of Selected Alkanes-Degrading Plant Growth-Promoting Bacterial Isolates.

The use of plant growth-promoting rhizobacteria (PGPR) as a bioremediation enhancer in plant-assisted phytoremediation requires several steps, consisting of the screening, selection, and characterization of isolates. A subset of 50 bacterial isolates representing a wide phylogenetic range were selected from 438 morphologically different bacteria that were originally isolated from a petroleum hydrocarbon (PHC)-polluted site of a former petrochemical plant. Selected candidate bacteria were screened using six conventional plant growth-promoting (PGP) traits, complemented with the genetic characterization of genes involved in alkane degradation, as well as other pertinent functions. Finally, the bacterial isolates were subjected to plant growth promotion tests using a gnotobiotic approach under normal and stressed conditions. Our results indicated that 35 bacterial isolates (70%) possessed at least four PGP traits. Twenty-nine isolates (58%) were able to utilize n-hexadecane as a sole carbon source, whereas 43 isolates (86%) were able to utilize diesel as the sole carbon source. The presence of catabolic genes related to hydrocarbon degradation was assessed using endpoint PCR, with the alkane monooxygenase (alkB) gene found in 34 isolates, the cytochrome P450 hydroxylase (CYP153) gene found in 24 isolates, and the naphthalene dioxygenase (nah1) gene found to be present in 33 isolates. Thirty-six strains (72%) promoted canola root elongation in the growth pouch assay. After several rounds of screening, seven bacterial candidates (individually or combined in a consortium) were tested for canola root and shoot growth promotion in substrates amended by different concentrations of n-hexadecane (0%, 1%, 2%, and 3%) under gnotobiotic conditions. Our results showed that Nocardia sp. (WB46), Pseudomonas plecoglossicida (ET27), Stenotrophomonas pavanii (EB31), and Gordonia amicalis (WT12) significantly increased the root length of canola grown in 3% n-hexadecane compared with the control treatment, whereas Nocardia sp. (WB46) and Bacillus megaterium (WT10) significantly increased shoot length compared to control treatment at the same concentration of n-hexadecane. The consortium had a significant enhancement effect on root length compared to all isolates inoculated individually or to the control. This study demonstrates that the combination of PGPR traits and the PHC degradation potential of bacteria can result in an enhanced beneficial effect in phytoremediation management, which could lead to the development of innovative bacterial inoculants for plants to remediate PHC-contaminated soils.

Isolation and characterization of Klebsiella oxytoca from the rhizosphere of Lotus corniculatus and its biostimulating features.

Significant food resource shortages are occurring worldwide. Plant growth-promoting rhizobacteria (PGPR) represent an ecofriendly and efficient approach for increasing soil fertility and plant productivity. The current study explored biostimulating traits of PGPR from the rhizosphere of Lotus corniculatus growing in the Al-Ahsa region. A bacterial isolate (LCK121) was obtained, characterized for phenotypic, and identified by 16S rRNA gene sequencing. In addition, its growth-stimulating effects on barley were investigated. The strain identity was confirmed via comparative analysis of the 16S rDNA sequences with Klebsiella oxytoca (99.3% similarity level). LCK121 exhibited multiple plant growth-promoting features, including indole-3-acetic acid (IAA) production (16.34 µg mL-1), 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity (1.35±0.02 µmol α-ketobutyrate mg-1 h-1), phosphate solubilization, and nitrogen fixation. Furthermore, in vitro inoculation of barley with LCK121 significantly increased the root and shoot dry weights. The results highlight the potential of LCK121 for developing green fertilizers for sustainable agriculture.

Cork Oak Forests Soil Bacteria: Potential for Sustainable Agroforest Production.

Plant growth promoting rhizobacteria (PGPR) are in increasing demand due to their role in promoting sustainable practices, not only in agriculture but also in forestry. Keeping in mind the future application of PGPR for increasing cork oak sustainability, the aim of this study was to find cork oak PGPR isolates with increased nutrient solubilisation traits, able to promote root morphological changes and/or antagonize cork oak bark phytopathogens. Soils from three cork oak forests with distinct bioclimates (humid, semi-humid and semi-arid) were used for isolating bacteria. From the 7634 colony-forming units, 323 bacterial isolates were biochemically assayed for PGPR traits (siderophores production, phosphate solubilizing and organic acids production), and 51 were found to display all these traits. These PGPR were able to induce root morphological changes on Arabidopsis thaliana, like suppression of primary root growth, increase of lateral roots or root hairs formation. However, the most proficient PGPR displayed specific ability in changing a single root morphological trait. This ability was related not only to bacterial genotype, but also with the environment where bacteria thrived and isolation temperature. Bacteria from semi-arid environments (mainly Bacillus megaterium isolates) could hold a promising tool to enhance plant development. Other isolates (Serratia quinivorens or B. cereus) could be further explored for biocontrol purposes.

Characterization and Identification of Plant Growth Promoting Traits of a Rhizobacteria: Pantoea Agglomerans 20-19

Plant Growth Promoting Rhizobacteria (PGPR) are known to influence positively plant growth by various direct or indirect mechanisms. The objective of this study was to characterize and identified a bacterium was isolated from the wheat rhizosphere of an semi arid area Meknes (Morocco). The studied strain possess several PGPR trais such as nitrogen fixation ability, phosphate solubilization, production of auxins and Ammonia production. Based on their 16S rDNA sequences, the strain were identified as Pantoea agglomerans 20- 19. This Pantoea agglomerans 20-19 is capable of IAA, ammonia production, nitrogen fixation and solubilization phosphorus. These results showed that Pantoea agglomerans 20-19 with its PGPR traits could consitute a good biofertilizer in semi arid area.

Rhizobacteria from \u2018flowering desert\u2019 events contribute to the mitigation of water scarcity stress during tomato seedling germination and growth

Tomato (Solanum lycopersicum L.) is an important vegetable cultivated around the world. Under field conditions, tomato can be negatively affected by water scarcity in arid and semiarid regions. The application of native plant growth-promoting rhizobacteria (PGPR) isolated from arid environments has been proposed as an inoculant to mitigate abiotic stresses in plants. In this study, we evaluated rhizobacteria from Cistanthe longiscapa (syn Calandrinia litoralis and Calandrinia longiscapa), a representative native plant of flowering desert (FD) events (Atacama Desert, Chile), to determine their ability to reduce water scarcity stress on tomato seedlings. The isolated bacterial strains were characterized with respect to their PGPR traits, including P solubilization, 1-aminocyclopropane-1-carboxylate deaminase activity, and tryptophan-induced auxin and exopolysaccharide production. Three PGPR consortia were formulated with isolated Bacillus strains and then applied to tomato seeds, and then, the seedlings were exposed to different levels of water limitations. In general, tomato seeds and seedlings inoculated with the PGPR consortia presented significantly (P ≤ 0.05) greater plant growth (48 to 60 cm of height and 171 to 214 g of weight) and recovery rates (88 to 100%) compared with those without inoculation (37 to 51 cm of height; 146 to 197 g of fresh weight; 54 to 92% of recovery) after exposure to a lack of irrigation over different time intervals (24, 72 and 120 h) before transplantation. Our results revealed the effectiveness of the formulated PGPR consortia from FD to improve the performance of inoculated seeds and seedlings subjected to water scarcity; thus, the use of these consortia can represent an alternative approach for farmers facing drought events and water scarcity associated with climate change in semiarid and arid regions worldwide.

Streptomyces spp. enhance vegetative growth of maize plants under saline stress.

Saline stress is one of the abiotic stresses that most compromises the yield of crops and can be mitigated by plant growth-promoting rhizobacteria (PGPR). This work characterized rhizobacteria isolates from the genus Streptomyces as PGPR and evaluated their role on growth and alleviation of the effects caused by saline stress in maize (Zea mays L.). Production of indolic compounds (IC), siderophores, ACC deaminase, phenazines, and promotion of plant growth were determined to characterize bacterial isolates. Salinity tolerance was accessed by culturing the Streptomyces isolates under NaCl increasing concentrations (0-300 mM). Four Streptomyces isolates exhibiting PGPR traits and salinity tolerance were selected and their effect on tolerance of maize plants to saline stress was evaluated. Plants obtained from bacterized seeds and submitted to 100 and 300 mM NaCl were used. All Streptomyces spp. produced IC and siderophores, CLV178 being the best producer of these two compounds. ACC deaminase was detected in six of the 10 isolates (CLV95, CLV97, CLV127, CLV179, CLV193, and CLV205), while phenazines were found only in CLV186 and CLV194. All isolates were tolerant to salinity, growing at concentrations up to 300 mM NaCl, with exception of CLV188. Increased concentrations of IC were detected in most of the isolates exposed to salinity. CLV97 and CLV179 significantly promoted growth of roots and leaves of maize plants and attenuated the negative effects of salinity on plant growth. Root colonization by Streptomyces spp. was confirmed in plants cultivated 20 days under saline stress.
