Plant Growth-Promoting Bacterial Consoprtia as a Strategy Towards Crop Growth and Fitness of Little Millet (Panicum sumatranse L.)

Little millet, a minor cereal cultivated in semi-arid regions of India, is renowned for its nutraceutical properties. The endophytic plant growth-promoting bacterial (PGPB) communities help crop plants increase growth and health, endure stress tolerance, and enhance nutrient availability. Seed biotiztion with these endophytic PGPBs can facilitate the host plant to develop a defense against pathogens and challenging environments. Thus the present study aimed to biotize the little millet seeds (var. ATL1) with PGPBs, viz., Bacillus albus LRS2, Alcaligenes faecalis LSB6, Bacillus amyloliquefaciens LAS10 and Bacillus velezensis LLB10 and to evaluate their efficiency by developing as a consortia of these isolates for improved plant growth and development. The seeds were biotized with all the strains individually and as consortia to evaluate their plant growth attributes in vitro and in vivo. The results showed significant differences in vegetative growth parameters and yield attributes compared with the control over consortia biotized seeds. During the panicle initiation stage (27 DAS), the shoot length (59.2, cm), root length (13.9 cm), total plant biomass (1.46 g), number of productive tillers plant-1 (5), panicle length (13.2 cm) and 1000 grain weight (5.12 g), grain yield plant-1 (28.45 g), straw yield (36.21 g) registered more in consortia biotized plants compared to non-biotized plant maintained as control. Thus, the present study confirmed that the inoculation of little millet seeds with the Consortia of indigenous strains could promote plant growth and productivity. Further, the Vol 110|10-12 inoculant can be recommended as a novel bio-inoculant for enhancing crop health and sustainable production in little millet.

Delineation of molecular interactions of plant growth promoting bacteria induced \u03b2-1,3-glucanases and guanosine triphosphate ligand for antifungal response in rice: a molecular dynamics approach

BackgroundThe plant growth is influenced by multiple interactions with biotic (microbial) and abiotic components in their surroundings. These microbial interactions have both positive and negative effects on plant. Plant growth promoting bacterial (PGPR) interaction could result in positive growth under normal as well as in stress conditions.MethodsHere, we have screened two PGPR’s and determined their potential in induction of specific gene in host plant to overcome the adverse effect of biotic stress caused by Magnaporthe grisea, a fungal pathogen that cause blast in rice. We demonstrated the glucanase protein mode of action by performing comparative modeling and molecular docking of guanosine triphosphate (GTP) ligand with the protein. Besides, molecular dynamic simulations have been performed to understand the behavior of the glucanase-GTP complex.ResultsThe results clearly showed that selected PGPR was better able to induce modification in host plant at morphological, biochemical, physiological and molecular level by activating the expression of β-1,3-glucanases gene in infected host plant. The docking results indicated that Tyr75, Arg256, Gly258, and Ser223 of glucanase formed four crucial hydrogen bonds with the GTP, while, only Val220 found to form hydrophobic contact with ligand.ConclusionsThe PGPR able to induce β-1,3-glucanases gene in host plant upon pathogenic interaction and β-1,3-glucanases form complex with GTP by hydrophilic interaction for induction of defense cascade for acquiring resistance against Magnaporthe grisea.Graphical abstract

Predatory activity of Acanthamoeba sp genotype T4 on different plant growth-promoting bacteria and their combined effect on rice seedling growth

Heterotrophic protists play a crucial role in plant growth promotion via nutrient cycling and shift in microbial community composition in the soil ecosystem. Selective predation pressure by protists contributes to the evaluation of plant beneficial traits in rhizospheric bacteria. However, not always all plant growth-promoting bacterial (PGPB) strains are benefitted by predation. This study aimed to examine the predatory effect of Acanthamoeba sp genotype T4 on a range of PGPB strains and their combined impact on early rice seedling growth. Acanthamoeba sp isolated from rice rhizosphere soils were used to assess predation against several PGPB such as Pseudomonas, Bacillus, Enterobacter, Morganella, Stenotrophomonas, Providencia, and Lysinibacillus on Nutrient Yeast Extract agar (NYE) plate. The controlled experiment on the germinated rice seeds (Oryza sativa L.) grown in Petri dishes containing each PGPB strain and Acanthamoeba sp was performed to evaluate the combined impact on plant performance. The PGPB-Acanthamoeba combined treatments in Petri dishes showed significant rice seedling growth compared to PGPB alone, non-PGPB and control. Our results indicated the positive but different impact of Acanthamoeba sp with different PGPB species on early rice plant growth. Further in-depth research should be carried out with diverse protists and PGPB species to assess which protist species can be linked to enhancement of indigenous soil PGPB for improved plant growth.

Shifts in the Rhizosphere and Endosphere Colonizing Bacterial Communities Under Drought and Salinity Stress as Affected by a Biofertilizer Consortium

The present research asks how plant growth-promoting bacterial (PGPB) inoculants and chemical fertilizers change rhizosphere and root endophytic bacterial communities in durum wheat, and its dependence on environmental stress. A greenhouse experiment was carried out under drought (at 40% field capacity), or salinity (150mM NaCl) conditions to investigate the effects of a chemical fertilizer (containing nitrogen, phosphorus, potassium and zinc) or a biofertilizer (a bacterial consortium of four PGPBs). High-throughput amplicon sequencing of the 16S rRNA of the rhizosphere, non-sterilized, or surface-sterilized roots, showed shifts in bacterial communities in response to stress treatments, which were greater for salinity than for drought and tended to show increased oligotrophs relative abundances compared to non-stress controls. The results also showed that Proteobacteria, Acidobacteria, Bacteroidetes, Gemmatimonadetes, Thaumarchaeota, Firmicutes, and Verrucomicrobia had a higher relative abundance in the rhizosphere, while Actinobacteria were more abundant on roots, while Candidatus_Saccharibacteria and Planctomycetes inside roots. The results indicated that the root endophytic bacterial communities were more affected by (bio-) fertilization treatments than those in the rhizosphere, particularly as affected by PGPB inoculation. This greater susceptibility of endophytes to (bio-) fertilizers was associated with increased abundance of the 16S rRNA and acdS genes in plant roots, especially under stress conditions. These changes in root endophytes, which coincided with an improvement in grain yield and photosynthetic capacity of plants, may be considered as one of the mechanisms by which PGPB affect plants.

Experimental Investigation of Chlorella vulgaris and Enterobacter sp. MN17 for Decolorization and Removal of Heavy Metals from Textile Wastewater

The present study evaluated the performance of microalgae Chlorella vulgaris in an Enterobacter sp. MN17-assisted textile industry wastewater treatment system for decolorization, removal of heavy metals (Cu, Cr, Pb, and Cd), and chemical oxygen demand (COD). Different dilutions (5, 10, and 20%) of wastewater were prepared to decrease the pollutant toxicity for culturing microalgae and bacteria. Reduction of color, COD, and metal contents by microalgal treatment of wastewater varied greatly, while removal efficiency (RE) was significantly enhanced when endophytic bacterial strain MN17 inoculum was applied. Most notable, results were found at a 5% dilution level by Enterobacter sp. MN17-inoculated C. vulgaris medium, as chromium (Cr), cadmium (Cd), copper (Cu), and lead (Pb) concentrations were decreased from 1.32 to 0.27 mg L−1 (79% decrease), 0.79–0.14 mg L−1 (93% decrease), 1.33–0.36 mg L−1 (72% decrease), and 1.2–0.25 mg L−1 (79% decrease), respectively. The values of COD and color were also significantly decreased by 74% and 70%, respectively, by a C. vulgaris–Enterobacter sp. MN17 consortium. The present investigation revealed that bacterial inoculation of microalgae significantly enhanced the removal of coloring agents and heavy metals from textile wastewater by stimulating the growth of algal biomass. This study manifested the usefulness of microalgae–bacterial mutualism for the remediation of heavy metals, COD, and color in industrial effluents. Microalgae consortia with growth promoting bacteria could be a breakthrough for better bioremediation and bioprocess economy. Thus, further studies are needed for successful integration of microalgae–plant growth promoting bacterial (PGPB) consortium for wastewater treatments.

Inoculation With a Microbe Isolated From the Negev Desert Enhances Corn Growth.

Corn (Zea mays L.) is not only an important food source, but also has numerous uses, including for biofuels, fillers for cosmetics, glues, and so on. The amount of corn grown in the U.S. has significantly increased since the 1960’s and with it, the demand for synthetic fertilizers and pesticides/fungicides to enhance its production. However, the downside of the continuous use of these products, especially N and P fertilizers, has been an increase in N2O emissions and other greenhouse gases into the atmosphere as well as run-off into waterways that fuel pollution and algal blooms. These approaches to agriculture, especially if exacerbated by climate change, will result in decreased soil health as well as human health. We searched for microbes from arid, native environments that are not being used for agriculture because we reasoned that indigenous microbes from such soils could promote plant growth and help restore degraded soils. Employing cultivation-dependent methods to isolate bacteria from the Negev Desert in Israel, we tested the effects of several microbial isolates on corn in both greenhouse and small field studies. One strain, Dietzia cinnamea 55, originally identified as Planomicrobium chinense, significantly enhanced corn growth over the uninoculated control in both greenhouse and outside garden experiments. We sequenced and analyzed the genome of this bacterial species to elucidate some of the mechanisms whereby D. cinnamea 55 promoted plant growth. In addition, to ensure the biosafety of this previously unknown plant growth promoting bacterial (PGPB) strain as a potential bioinoculant, we tested the survival and growth of Caenorhabditis elegans and Galleria mellonella (two animal virulence tests) as well as plants in response to D. cinnamea 55 inoculation. We also looked for genes for potential virulence determinants as well as for growth promotion.

Sorghum Growth Promotion by Paraburkholderia tropica and Herbaspirillum frisingense: Putative Mechanisms Revealed by Genomics and Metagenomics.

Bacteria from the genera Paraburkholderia and Herbaspirillum can promote the growth of Sorghum bicolor, but the underlying mechanisms are not yet known. In a pot experiment, sorghum plants grown on sterilized substrate were inoculated with Paraburkholderia tropica strain IAC/BECa 135 and Herbaspirillum frisingense strain IAC/BECa 152 under phosphate-deficient conditions. These strains significantly increased Sorghum bicolor cultivar SRN-39 root and shoot biomass. Shotgun metagenomic analysis of the rhizosphere revealed successful colonization by both strains; however, the incidence of colonization was higher in plants inoculated with P. tropica strain IAC/BECa 135 than in those inoculated with H. frisingense strain IAC/BECa 152. Conversely, plants inoculated with H. frisingense strain IAC/BECa 152 showed the highest increase in biomass. Genomic analysis of the two inoculants implied a high degree of rhizosphere fitness of P. tropica strain IAC/BECa 135 through environmental signal processing, biofilm formation, and nutrient acquisition. Both genomes contained genes related to plant growth-promoting bacterial (PGPB) traits, including genes related to indole-3-acetate (IAA) synthesis, nitrogen fixation, nodulation, siderophore production, and phosphate solubilization, although the P. tropica strain IAC/BECa 135 genome contained a slightly more extensive repertoire. This study provides evidence that complementary mechanisms of growth promotion in Sorghum might occur, i.e., that P. tropica strain IAC/BECa 135 acts in the rhizosphere and increases the availability of nutrients, while H. frisingense strain IAC/BECa 152 influences plant hormone signaling. While the functional and taxonomic profiles of the rhizobiomes were similar in all treatments, significant differences in plant biomass were observed, indicating that the rhizobiome and the endophytic microbial community may play equally important roles in the complicated plant-microbial interplay underlying increased host plant growth.

Isolation and Identification of Plant Growth-Promoting Bacteria Highly Effective in Suppressing Root Rot in Fava Beans.

In this study, 18 plant growth-promoting bacterial (PGPB) strains were isolated from the rhizosphere of both Red silk-cotton tree (Bombax malabaricum) and Chinese banyan (Ficus retusa). Culture morphology was observed and genotypic characterization was accomplished by sequencing partial 16S rRNA gene. Plant growth promotion traits and antagonistic activities of the strains against phytopathogenic fungi were evaluated. Among all strains, Bacillus thuringiensis (MN419208) exert the highest indole acetic acid (38µg/ml), produced exopolysaccharides (587.2µg/ml), and fixed nitrogen which in turn increased both fresh and dry weights of bean plants by 41.5% and 18.8%, respectively. In another greenhouse experiment studying the antifungal activities of seven strains and their co-culture against Rhizoctonia solani (LN735538), B. sonorensis MN419205, B. wiedmannii MN419207, B. subtilis MN419218, and the mixture of (MN419207) and (MN419208) reduced total damping off from 81.7% in control to 30%, 35%, 35%, and 38.3%, respectively, and reduced disease severity index from 33.3% to 20.5%, 22.5%, 14.2%, and 19.3% as well. Our data indicate that these strains are effective in promoting plant growth and in inhibiting R. solani infection nonetheless field experiments are needed to examine their effectiveness as a viable alternative to chemical fertilizers and conventional pesticides.

Molecular characterization of monocrotophos and chlorpyrifos tolerant bacterial strain for enhancing seed germination of vegetable crops

The main aim of this study is to investigate the toxicity of organophosphate (OPs) insecticides monocrotophos (MCP) and chlorpyrifos (CLS) on plant growth promoting (PGP) properties and seed germination of brinjal, tomato and okra vegetables inoculated by Microbacterium hydrocarbonoxydans (BHUJP-P1), Stenotrophomonas rhizophila (BHUJP-P2), Bacillus licheniformis (BHUJP-P3) and Bacillus cereus (BHUJP-P4). Maximum increase in microbial growth (52.6% & 47.9%) with enhanced EPS production (447.67 mg/ml & 75.00 mg/ml) was showed by BHUJP-P4 and BHUJP-P3 at 10× dose of MCP and CLS over control, BHUJP-2 and BHUJP-P1 respectively. Simultaneously, both strains recorded minimum reduction in PGP activities and seed germination at 3× dose of both insecticides as compared to BHUJP-2 and BHUJP-P1, respectively. Strains BHUJP-P3 and BHUJP-P4 showed 83 and 81% of monocrotophos degradation at 50 mg/kg concentration; 81 and 80% at 150 mg/kg concentration within 5days respectively. Concurrently, these strains BHUJP-P3 and BHUJP-P4 were recorded 53 and 90% of chlorpyrifos degradation at 50 mg/kg concentration; 49% and 87% at 100 mg/kg concentration within 72 h, respectively. The OPs insecticide degrading gene opdA and opd was found in strain BHUJP-P3 and BHUJP-P4, respectively. The multifarious biological activities of strain BHUJP-P3 and BHUJP-P4 showed maximum tolerance against insecticide, and minimum reduction in P-solubilisation, IAA, siderophore and HCN production for plant growth promotion and biological control under insecticide stress. Thus, these novel isolates may be used as biodegradation of organophosphate insecticide and plant growth promoting bacterial (PGPB) inoculum for enhancing seed germination of vegetables under stress insecticide. These novel strains will be environment friendly, socially acceptable and economically viable.

Mining the phytomicrobiome to understand how bacterial coinoculations enhance plant growth.

In previous work, we showed that coinoculating Rhizobium leguminosarum bv. viciae 128C53 and Bacillus simplex 30N-5 onto Pisum sativum L. roots resulted in better nodulation and increased plant growth. We now expand this research to include another alpha-rhizobial species as well as a beta-rhizobium, Burkholderia tuberum STM678. We first determined whether the rhizobia were compatible with B. simplex 30N-5 by cross-streaking experiments, and then Medicago truncatula and Melilotus alba were coinoculated with B. simplex 30N-5 and Sinorhizobium (Ensifer) meliloti to determine the effects on plant growth. Similarly, B. simplex 30N-5 and Bu. tuberum STM678 were coinoculated onto Macroptilium atropurpureum. The exact mechanisms whereby coinoculation results in increased plant growth are incompletely understood, but the synthesis of phytohormones and siderophores, the improved solubilization of inorganic nutrients, and the production of antimicrobial compounds are likely possibilities. Because B. simplex 30N-5 is not widely recognized as a Plant Growth Promoting Bacterial (PGPB) species, after sequencing its genome, we searched for genes proposed to promote plant growth, and then compared these sequences with those from several well studied PGPB species. In addition to genes involved in phytohormone synthesis, we detected genes important for the production of volatiles, polyamines, and antimicrobial peptides as well as genes for such plant growth-promoting traits as phosphate solubilization and siderophore production. Experimental evidence is presented to show that some of these traits, such as polyamine synthesis, are functional in B. simplex 30N-5, whereas others, e.g., auxin production, are not.

Inoculation with Metal-Mobilizing Plant-Growth-Promoting Rhizobacterium Bacillus sp. SC2b and Its Role in Rhizoremediation

A plant growth-promoting bacterial (PGPB) strain SC2b was isolated from the rhizosphere of Sedum plumbizincicola grown in lead (Pb)/zinc (Zn) mine soils and characterized as Bacillus sp. based on (1) morphological and biochemical characteristics and (2) partial 16S ribosomal DNA sequencing analysis. Strain SC2b exhibited high levels of resistance to cadmium (Cd) (300 mg/L), Zn (730 mg/L), and Pb (1400 mg/L). This strain also showed various plant growth-promoting (PGP) features such as utilization of 1-aminocyclopropane-1-carboxylate, solubilization of phosphate, and production of indole-3-acetic acid and siderophore. The strain mobilized high concentration of heavy metals from soils and exhibited different biosorption capacity toward the tested metal ions. Strain SC2b was further assessed for PGP activity by phytagar assay with a model plant Brassica napus. Inoculation of SC2b increased the biomass and vigor index of B. napus. Considering such potential, a pot experiment was conducted to assess the effects of inoculating the metal-resistant PGPB SC2b on growth and uptake of Cd, Zn and Pb by S. plumbizincicola in metal-contaminated agricultural soils. Inoculation with SC2b elevated the shoot and root biomass and leaf chlorophyll content of S. plumbizincicola. Similarly, plants inoculated with SC2b demonstrated markedly higher Cd and Zn accumulation in the root and shoot system, indicating that SC2b enhanced Cd and Zn uptake by S. plumbizincicola through metal mobilization or plant-microbial mediated changes in chemical or biological soil properties. Data demonstrated that the PGPB Bacillus sp. SC2b might serve as a future biofertilizer and an effective metal mobilizing bioinoculant for rhizoremediation of metal polluted soils.

Amelioration of high salinity stress damage by plant growth-promoting bacterial endophytes that contain ACC deaminase

Plant growth and productivity is negatively affected by soil salinity. However, it is predicted that plant growth-promoting bacterial (PGPB) endophytes that contain 1-aminocyclopropane-1-carboxylate (ACC) deaminase (E.C. 4.1.99.4) can facilitate plant growth and development in the presence of a number of different stresses. In present study, the ability of ACC deaminase containing PGPB endophytes Pseudomonas fluorescens YsS6, Pseudomonas migulae 8R6, and their ACC deaminase deficient mutants to promote tomato plant growth in the absence of salt and under two different levels of salt stress (165 mM and 185 mM) was assessed. It was evidence that wild-type bacterial endophytes (P. fluorescens YsS6 and P. migulae 8R6) promoted tomato plant growth significantly even in the absence of stress (salinity). Plants pretreated with wild-type ACC deaminase containing endophytic strains were healthier and grew to a much larger size under high salinity stress compared to plants pretreated with the ACC deaminase deficient mutants or no bacterial treatment (control). The plants pretreated with ACC deaminase containing bacterial endophytes exhibit higher fresh and dry biomass, higher chlorophyll contents, and a greater number of flowers and buds than the other treatments. Since the only difference between wild-type and mutant bacterial endophytes was ACC deaminase activity, it is concluded that this enzyme is directly responsible for the different behavior of tomato plants in response to salt stress. The use of PGPB endophytes with ACC deaminase activity has the potential to facilitate plant growth on land that is not normally suitable for the majority of crops due to their high salt contents.

Effect of <i>Pseudomonas aeruginosa</i> ZGKD2 on the Growth of <i>Amorpha fruticosa </i>L in Coal Gangue

The cadmium-resistant Pseudomonas aeruginosa strain ZGKD2 was isolated from gangue pile of coal area. Production of siderophores, indole-3-acetic acid (IAA) and the solubilization of phosphate were observed in the strain. Two types of siderophores were identified by UV spectrophotometer. The highest production of IAA and phosphate solubilization were 2.0 ug/mL and 7.2 ug/mL. The root length, plant height and fresh weight of Amorpha fruticosa L in the substrates of Coal gannue and losses were promoted after inoculation with ZGKD2. These data indicated that Pseudomonas aeruginosa strain ZGKD2 was a plant growth-promoting bacterial (PGPB).

Delay of flower senescence by bacterial endophytes expressing 1-aminocyclopropane-1-carboxylate deaminase

The ability of 1-aminocyclopropane-1-carboxylate (ACC) deaminase-containing plant growth-promoting bacterial (PGPB) endophytes Pseudomonas fluorescens YsS6 and Pseudomonas migulae 8R6, their ACC deaminase minus mutants and the rhizospheric plant growth-promoting bacterium Pseudomonas putida UW4 to delay the senescence of mini carnation cut flowers was assessed. Fresh cut flowers were incubated with either a bacterial cell suspension, the ethylene precursor ACC, the ethylene inhibitor l-α-(aminoethoxyvinyl)-glycine or 0·85% NaCl at room temperature for 11 days. Levels of flower senescence were recorded every other day. To verify the presence of endophytes inside the plant tissues, scanning electron microscopy was performed. Among all treatments, flowers treated with wild-type ACC deaminase-containing endophytic strains exhibited the most significant delay in flower senescence, while flowers treated with the ACC deaminase minus mutants senesced at a rate similar to the control. Flowers treated with Ps. putida UW4 senesced more rapidly than untreated control flowers. The only difference between wild-type and mutant bacterial endophytes was ACC deaminase activity so that it may be concluded that this enzyme is directly responsible for the significant delay in flower senescence. Despite containing ACC deaminase activity, Ps. putida UW4 is not taken up by the cut flowers and therefore has no effect on prolonging their shelf life. The world-wide cut flower industry currently uses expensive and potentially environmentally dangerous chemical inhibitors of ethylene to prolong the shelf life of cut flowers. The use of PGPB endophytes with ACC deaminase activity has the potential to replace the chemicals that are currently used by the cut flower industry.

Plant growth promoting bacteria enhance water stress resistance in green gram plants

Plant growth promoting bacterial (PGPB) strains Pseudomonas fluorescens Pf1 and endophytic Bacillus subtilis EPB5, EPB22, EPB 31 were tested for their capacity to induce water stress related proteins and enzymes in green gram (Vigna radiata) plants. Among the different bacteria used, P. fluorescens Pf1 increased the vigour index, fresh weight and dry weight of green gram seedlings in vitro. Quantitative and qualitative analyses of stress-related enzymes indicated the greater activity of catalase and peroxidase in green gram plants bacterized with P. fluorescens Pf1 against water stress when compared to untreated plants. The greater accumulation of proline was recorded in Pf1 treated plants compared to untreated plants. The pot culture study revealed the greater resistance to water stress by green gram plants treated with P. fluorescens Pf1 compared to untreated plants. The greater activity of stress-related enzymes in green gram plants mediated by PGPB could pave the way for developing drought management strategies.

Inoculation of plant growth promoting bacterium Achromobacter xylosoxidans strain Ax10 for the improvement of copper phytoextraction by Brassica juncea

In this study, a copper-resistant plant growth promoting bacterial (PGPB) strain Ax10 was isolated from a Cu mine soil to assess its plant growth promotion and copper uptake in Brassica juncea. The strain Ax10 tolerated concentrations up to 600 mg Cu L −1 on a Luria–Bertani (LB) agar medium and utilized 1-aminocyclopropane-1-carboxylic acid (ACC) as a sole N source in DF salts minimal medium. The strain Ax10 was characterized as Achromobacter xylosoxidans based on its 16S rDNA sequence homology (99%). The bacterium A. xylosoxidans Ax10 has also exhibited the capability of producing indole acetic acid (IAA) (6.4 μg mL −1), and solubilizing inorganic phosphate (89.6 μg mL −1) in specific culture media. In pot experiments, inoculation of A. xylosoxidans Ax10 significantly increased the root length, shoot length, fresh weight and dry weight of B. juncea plants compared to the control. This effect can be attributed to the utilization of ACC, production of IAA and solubilization of phosphate. Furthermore, A. xylosoxidans Ax10 inoculation significantly improved Cu uptake by B. juncea. Owing to its wide action spectrum, the Cu-resistant A. xylosoxidans Ax10 could serve as an effective metal sequestering and growth promoting bioinoculant for plants in Cu-stressed soil. The present study has provided a new insight into the phytoremediation of Cu-contaminated soil.
