Exploring the dynamics of ISR signaling in maize upon seed priming with plant growth promoting actinobacteria isolated from tea rhizosphere of Darjeeling.

Plant growth-promoting rhizobacteria (PGPR) offer an eco-friendly alternative to agrochemicals for better plant growth and development. Here, we evaluated the plant growth promotion abilities of actinobacteria isolated from the tea (Camellia sinensis) rhizosphere of Darjeeling, India. 16S rRNA gene ribotyping of 28 isolates demonstrated the presence of nine different culturable actinobacterial genera. Assessment of the in vitro PGP traits revealed that Micrococcus sp. AB420 exhibited the highest level of phosphate solubilization (i.e., 445 ± 2.1µg/ml), whereas Kocuria sp. AB429 and Brachybacterium sp. AB440 showed the highest level of siderophore (25.8 ± 0.1%) and IAA production (101.4 ± 0.5µg/ml), respectively. Biopriming of maize seeds with the individual actinobacterial isolate revealed statistically significant growth in the treated plants compared to controls. Among them, treatment with Paenarthrobacter sp. AB416 and Brachybacterium sp. AB439 exhibited the highest shoot and root length. Biopriming has also triggered significant enzymatic and non-enzymatic antioxidative defense reactions in maize seedlings both locally and systematically, providing a critical insight into their possible role in the reduction of reactive oxygen species (ROS) burden. To better understand the role of actinobacterial isolates in the modulation of plant defense, three selected actinobacterial isolates, AB426 (Brevibacterium sp.), AB427 (Streptomyces sp.), and AB440 (Brachybacterium sp.) were employed to evaluate the dynamics of induced systemic resistance (ISR) in maize. The expression profile of five key genes involved in SA and JA pathways revealed that bio-priming with actinobacteria (Brevibacterium sp. AB426 and Brachybacterium sp. AB440) preferably modulates the JA pathway rather than the SA pathway. The infection studies in bio-primed maize plants resulted in a delay in disease progression by the biotrophic pathogen Ustilago maydis in infected maize plants, suggesting the positive efficacy of bio-priming in aiding plants to cope with biotic stress. Conclusively, this study unravels the intrinsic mechanisms of PGPR-mediated ISR dynamics in bio-primed plants, offering a futuristic application of these microorganisms in the agricultural fields as an eco-friendly alternative.

Bacillus Strains as Effective Biocontrol Agents Against Phytopathogenic Bacteria and Promoters of Plant Growth.

Modern crop production relies on the application of chemical pesticides and fertilizers causing environmental and economic challenges. In response, less environmentally impactful alternatives have emerged such as the use of beneficial microorganisms. These microorganisms, particularly plant growth-promoting bacteria (PGPB), have demonstrated their ability to enhance plant growth, protect against various stresses, and reduce the need for chemical inputs. Among the PGPB, Bacillus species have garnered attention due to their adaptability and commercial potential. Recent reports have highlighted Bacillus strains as biocontrol agents against phytopathogenic bacteria while concurrently promoting plant growth. We also examined Bacillus plant growth-promoting abilities in Arabidopsis thaliana seedlings. In this study, we assessed the potential of various Bacillus strains to control diverse phytopathogenic bacteria and inhibit quorum sensing using Chromobacterium violaceum as a model system. In conclusion, our results suggest that bacteria of the genus Bacillus hold significant potential for biotechnological applications. This includes developments aimed at reducing agrochemical use, promoting sustainable agriculture, and enhancing crop yield and protection.

Bio-Priming with Bacillus Isolates Suppresses Seed Infection and Improves the Germination of Garden Peas in the Presence of Fusarium Strains

Seed infection caused by Fusarium spp. is one of the major threats to the seed quality and yield of agricultural crops, including garden peas. The use of Bacillus spp. with multiple antagonistic and plant growth-promoting (PGP) abilities represents a potential disease control strategy. This study was performed to evaluate the biocontrol potential of new Bacillus spp. rhizosphere isolates against two Fusarium strains affecting garden peas. Six Bacillus isolates identified by 16S rDNA sequencing as B. velezensis (B42), B. subtilis (B43), B. mojavensis (B44, B46), B. amyloliquefaciens (B50), and B. halotolerans (B66) showed the highest in vitro inhibition of F. proliferatum PS1 and F. equiseti PS18 growth (over 40%). The selected Bacillus isolates possessed biosynthetic genes for endoglucanase (B42, B43, B50), surfactin (B43, B44, B46), fengycin (B44, B46), bacillomycin D (B42, B50), and iturin (B42), and were able to produce indole-3-acetic acid (IAA), siderophores, and cellulase. Two isolates, B. subtilis B43 and B. amyloliquefaciens B50, had the highest effect on final germination, shoot length, root length, shoot dry weight, root dry weight, and seedling vigor index of garden peas as compared to the control. Their individual or combined application reduced seed infection and increased seed germination in the presence of F. proliferatum PS1 and F. equiseti PS18, both after seed inoculation and seed bio-priming. The most promising results were obtained in the cases of the bacterial consortium, seed bio-priming, and the more pathogenic strain PS18. The novel Bacillus isolates may be potential biocontrol agents intended for the management of Fusarium seed-borne diseases.

Genomic insights into Streptomyces hygroscopicus subsp. hygroscopicus SRF1: a potential biocontrol agent against fusarium wilt with plant growth-promoting abilities in tomatoes

ABSTRACT Streptomyces sp. isolate SRF1 has previously demonstrated promising results in controlling various plant fungi under in vitro conditions. However, further exploration of its genome and its efficacy in disease control and plant growth promotion under in vivo conditions is warranted. This study aimed to utilise whole genome sequencing (WGS) to classify Streptomyces sp. isolate SRF1 at the species level and predict genes associated with antagonistic ability and plant growth promotion, thereby assessing its potential as a biocontrol agent against fusarium wilt and a promoter of plant growth. WGS was employed to elucidate the taxonomic classification of isolate SRF1 and predict genes involved in disease suppression and plant growth promotion. In vitro assays were conducted to evaluate its antagonistic activity against Fusarium oxysporum f. sp. lycopersici (Fol) and its production of cellulase, amylase, indole-3-acetic acid (IAA), and siderophores. Pot experiments were carried out using SRF1 cell suspension and culture filtrate to assess its efficacy against Fol and its impact on tomato growth. Real-time PCR analysis was performed to correlate gene expression levels with observed activities. The results showed that isolate SRF1 was classified as S. hygroscopicus subsp. hygroscopicus and demonstrated antagonism against Fol. Furthermore, WGS revealed genes or gene clusters associated with disease control, including those encoding antifungal agents such as butyrolactol A, nigericin, and hygromycin A. Additionally, genes responsible for auxin biosynthesis, siderophore production, and carbohydrate-digesting enzymes such as chitinase, cellulase, and amylase were identified, suggesting potential roles in disease suppression and plant growth promotion.

Phosphate-Solubilizing Microorganisms Stimulate Physiological Responses of Perennial Ryegrass to Phosphorus Deficiency with Assistance of Straw Compost

Biofertilizers with phosphate-solubilizing microorganism (PSM) inoculations have been suggested to diminish the limitation of phosphorus (P) deficiency in plants. However, their applications in agriculture are restricted due to the inconstant effects of various PSMs. Proper carriers for the inoculations may overcome this shortcoming and improve PSMs’ effectiveness. The objective of this study was to investigate whether straw compost, a type of organic material, can act as a carrier for improving the efficiencies of phosphate-solubilizing bacteria and fungi named Acinetobacter sp. and Aspergillus niger, respectively, in soils. We monitored the growth and cellular physiological responses of one type of model plants, named perennial ryegrass (Lolium perenne L.), under four soil treatments, including non-fertilization, PSM inoculation alone, straw compost addition alone, and the combined applications of both PSMs and straw compost. We found the combined treatments significantly improved the growth by 14.7% for shoot height and 79.7% for shoot weight, respectively, on average. P and potassium (K) uptakes of ryegrass were also increased by 102.5% and 65.3%, respectively, after the application of both PSMs and straw compost. Furthermore, physiological properties, such as photosynthetic efficiency and P-transportation capacity, of ryegrass were also significantly improved under combined treatments when compared to other treatments, regardless of the types of PSM included. The piecewise structural equation model further indicated that PSM inoculation and straw compost input are synergistically contributing to the nutrient uptake of ryegrass through many direct and indirect ways. We propose that straw compost is a good carrier material for PSMs’ survival and would improve their plant growth promotion ability in soil. Our results provide valuable insights into the exploitation and utilization of P-biofertilizers in agriculture.

Evaluation of Trichoderma spp. as a plant growth promoter and antagonist of major pulse pathogens

Trichoderma spp. is mostly used for the management of soil-borne diseases and some foliage and fruit diseases in a variety of crop plants. It can help the environment by reducing agrochemical pollution, promoting plant growth, and enhancing plant resistance in addition to preventing plant diseases. Trichoderma spp. also functions as a secure, affordable, efficient, and environmentally friendly biocontrol agent for several crop species. In the present study, we obtained different Trichoderma isolates from rhizospheric soil samples of different locations and tested them for their antagonistic activity against major pulse pathogens. Among seven isolates, three isolates, viz., Pipal TH-2, ATH-Kashipur, and Mz/AP-2 were found to be highly effective by inhibiting the growth of Fusarium udum (64.04 to 78.65%), Fusarium ciceris (77.77 to 82.12%), Sclerotium rolfsii (59.09 to 69.30%), Macrophomina phaseolina (52.42 to 62.72%) and Alternaria alternata (80.12 to 83.22%). These isolates were also tested for growth-promoting traits (PGPR) in the present study and isolates having both plant growth-promoting ability and biocontrol potentiality were selected and preserved for further studies. These isolates of Trichoderma spp. would be a crucial partner for achieving the Green Earth goal due to their contribution to the sustainable growth of agriculture.

Isolation of endophytes from Dioscorea nipponica Makino for stimulating diosgenin production and plant growth.

Both bacterial and fungal endophytes exhibited one or more plant growth-promoting (PGP) traits. Among these strains, the Paenibacillus peoriae SYbr421 strain demonstrated the greatest activity in the direct biotransformation of tuber powderfrom D. nipponica into diosgenin. Endophytes play crucial roles in shaping active metabolites within plants, significantly influencing both the quality and yield of host plants. Dioscorea nipponica Makino accumulates abundant steroidal saponins, which can be hydrolyzed to produce diosgenin.However, our understanding of the associated endophytes and their contributions to plant growth and diosgenin production is limited. The present study aimed to assess the PGP ability and potential of diosgenin biotransformation by endophytesisolatesassociated withD. nipponica for the efficient improvement of plant growth and development of a clean and effective approach for producing the valuable drug diosgenin. Eighteen bacterial endophytes were classified into six genera through sequencing and phylogenetic analysis of the 16S rDNA gene. Similarly, 12 fungal endophytes were categorized into 5 genera based on sequencing and phylogenetic analysis of the ITS rDNA gene. Pure culture experiments revealed that 30 isolated endophytic strains exhibited one or more PGP traits, such as nitrogen fixation, phosphate solubilization, siderophore synthesis, and IAA production. One strain of endophytic bacteria, P. peoriae SYbr421, effectively directly biotransformed the saponin components in D. nipponica. Moreover, a high yield of diosgenin (3.50%) was obtained at an inoculum size of 4% after 6days of fermentation. Thus, SYbr421 could be used for a cleaner and more eco-friendly diosgenin production process. In addition, based on the assessment of growth-promoting isolates and seed germination results, the strains SYbr421, SYfr1321, and SYfl221 were selected for greenhouse experiments. The results revealed that the inoculation of these promising isolates significantly increased the plant height and fresh weight of the leaves and roots compared to the control plants. These findings underscore the importance of preparing PGP bioinoculants from selected isolates as an additional option for sustainable diosgenin production.

Screening and evaluation of bacterial endophytes of cowpea [Vigna unguiculata (L.) Walp.] for plant growth promotion and biocontrol potential

Cowpea [Vigna unguiculata (L.) Walp], is a popular leguminous vegetable crop grown in the wetland fallows of Kerala, India. Web blight caused by Rhizoctonia solani has turned out to be a problem in cowpeas, leading to substantial yield loss. We isolated 22 endophytic bacteria from fodder cowpea var. Aiswarya and 16 from bush cowpea var. Bhagyalakshmi, and evaluated their plant growth promotion ability and potential for web blight pathogen suppression. Initially, the isolates were screened in vitro for their plant growth promotional traits and antagonism against R. solani. The potential isolates were identified as Bacillus subtilis, B. amyloliquefaciens and B. velezensis through 16S rDNA cataloguing. Plant growth-promoting traits like the production of IAA (Indole acetic acid), ammonia, phosphorus, and siderophore by the promising isolates were also investigated. B. amyloliquefaciens CBSE5 recorded the highest IAA (3.54 microgram/mL) and siderophore production. B. velezensis CBRE5 showed the highest ammonia (177.29 micro moles/mL) production and phosphate solubilization. In the in vivo assay, seed biopriming along with foliar application on 20 and 40 days after sowing, of a consortium of B. amyloliquefaciens CBSE5 and B. velezensis CBRE5 recorded maximum values for all growth parameters viz. shoot length, root length, shoot fresh weight, root fresh weight, shoot dry weight, root dry weight, pod yield and number of pods per plant. The present study reports for the first time the use of endophytic Bacillus spp. isolated from cowpeas as plant growth-promoting bacteria, with biocontrol potential against the web blight pathogen R. solani.

Draft genome sequence of halotolerant plant growth-promoting Bacillus paralicheniformis MHN12.

Bacillus paralicheniformis MHN12 possesses a 4,245,453-base pair genome with 45.9% G + C content, including 1 CRISPR, 80 tRNA, 8 rRNA genes, and 4,418 predicted coding sequences . MHN12 exhibits high salinity tolerance and plant growth-promoting abilities, making it a promising bioinoculant for enhancing plant growth in saline soils.

Mitigation of Salt Stress in Rice by the Halotolerant Plant Growth-Promoting Bacterium Enterobacter asburiae D2.

Salinity is a major abiotic stress that seriously affects crop growth worldwide. In this work, we aimed to isolate potential halotolerant plant growth-promoting rhizobacteria (PGPR) to mitigate the adverse impacts of salt stress in rice. An isolate, D2, with multiple plant growth-promoting (PGP) characteristics was identified as Enterobacter asburiae D2. Strain D2 could produce indole-3-acetic acid and siderophore. It also exhibited phosphate solubilization and 1-aminocyclopropane-1-carboxylic deaminase activity. Genome analysis further provided insights into the molecular mechanism of its PGP abilities. Strain D2 inoculation efficiently stimulated rice growth under both normal and saline conditions. Compared with the non-inoculated plants, a significant increase in plant height (18.1-34.7%), root length (25.9-57.1%), root dry weight (57.1-150%), and shoot dry weight (17.3-50.4%) was recorded in inoculated rice seedlings. Meanwhile, rice seedlings inoculated with strain D2 showed improvement in chlorophyll and proline content, while the oxidant damage was reduced in these plants in comparison with the control group. Moreover, the K+/Na+ ratio of the inoculated rice seedlings exposed to NaCl and Na2CO3 was higher than that of the uninoculated groups. These results imply that Enterobacter asburiae D2 is a potential PGPR that can be used for alleviation of salt stress in rice.

Panchagavya-Derived Brevibacillus brevis S1-3: Insights from the Draft Genome on its Antimicrobial and Plant Growth-Promoting Ability

Panchagavya has traditionally been used in Indian Ayurvedic practices because of its pro-agricultural and medicinal properties. This study presents the draft genome of a new Brevibacillus brevis S1-3 strain isolated from the fermented product Panchagavya. Through whole-genome sequencing, we determined that the genome of B. brevis S1-3 was 6,348,716 base pairs with a GC content of 54.3%. Genome assembly revealed the presence of 6107 protein-coding genes, 186 tRNA genes, and 13 rRNA genes. Genome annotation and analysis identified the genes involved in metabolism and other cellular processes. We also predicted the presence of several gene clusters associated with plant growth promotion, including indole acetic acid (IAA), gibberellic acid, ammonia, and nitrogen. Our study also revealed the genes responsible for the production of secondary metabolites that displayed a significant correlation with antimicrobial activity. Our results provide new insights into the genomic basis of the plant growth-promoting abilities of B. brevis and pave the way for further research in this field.

Biochemical Analysis and Characterization of Silver Nanoparticles Synthesized by the Endophytic Fungus Plectosphaerella oligotrophica S8A26 Associated with Anaphalis contorta

Endophytic fungi associated with medicinal plants produce compounds that have vast biological activities. Anaphalis contorta is a highly therapeutic plant that has been used in traditional medicine by various communities. In the present study, the endophytic fungus Plectosphaerella oligotrophica S8A26 was isolated from the stem part of A. contorta and assessed for its plant growth promotion abilities, and showed positive results for phosphate solubilization and ammonia production. The secondary metabolites were analyzed for the presence of seven biochemicals, which indicate the presence of flavonoids, phenols, saponins, and steroids. The total flavonoid content and total phenolic content were recorded to be 25.67 µg of QE/mg of EE and 5.06 µg of GAE/mg of EE, respectively. The isolate was allowed to synthesize silver nanoparticles and characterized using X-ray diffraction, Energy Dispersive X-ray, Particle size distribution, and Zeta potential. XRD analysis has shown the AgNPs are crystalline with an average size of 24.81 nm. EDX study reveals silver as the main component with 48.63%. Particle size distribution indicates that the majority of the particles occur in the size range of 65–70 nm. Zeta potential value of −31.8 mV shows that the AgNPs are stable. Studies like the biological activities of the endophyte as well as AgNPs, secondary metabolite profiling, and the isolation of bioactive compounds would constitute important future research.

Whole genome sequencing and characterization of Pantoea agglomerans DBM 3797, endophyte, isolated from fresh hop (Humulus lupulus L.).

This paper brings new information about the genome and phenotypic characteristics of Pantoea agglomerans strain DBM 3797, isolated from fresh Czech hop (Humulus lupulus) in the Saaz hop-growing region. Although P. agglomerans strains are frequently isolated from different materials, there are not usually thoroughly characterized even if they have versatile metabolism and those isolated from plants may have a considerable potential for application in agriculture as a support culture for plant growth. P. agglomerans DBM 3797 was cultured under aerobic and anaerobic conditions, its metabolites were analyzed by HPLC and it was tested for plant growth promotion abilities, such as phosphate solubilization, siderophore and indol-3-acetic acid productions. In addition, genomic DNA was extracted, sequenced and de novo assembly was performed. Further, genome annotation, pan-genome analysis and selected genome analyses, such as CRISPR arrays detection, antibiotic resistance and secondary metabolite genes identification were carried out. The typical appearance characteristics of the strain include the formation of symplasmata in submerged liquid culture and the formation of pale yellow colonies on agar. The genetic information of the strain (in total 4.8 Mb) is divided between a chromosome and two plasmids. The strain lacks any CRISPR-Cas system but is equipped with four restriction-modification systems. The phenotypic analysis focused on growth under both aerobic and anaerobic conditions, as well as traits associated with plant growth promotion. At both levels (genomic and phenotypic), the production of siderophores, indoleacetic acid-derived growth promoters, gluconic acid, and enzyme activities related to the degradation of complex organic compounds were found. Extracellular gluconic acid production under aerobic conditions (up to 8 g/l) is probably the result of glucose oxidation by the membrane-bound pyrroloquinoline quinone-dependent enzyme glucose dehydrogenase. The strain has a number of properties potentially beneficial to the hop plant and its closest relatives include the strains also isolated from the aerial parts of plants, yet its safety profile needs to be addressed in follow-up research.

Screening of rhizobacteria for multi-trait plant growth-promoting ability and antagonism against B. fabae, the causative agent of chocolate spot disease of faba bean

This study aimed to isolate and characterize plant growth-promoting rhizobacteria from the faba bean rhizosphere for future inoculum production. For this purpose,127 dissimilar rhizobacterial colonies were isolated. All isolated colonies were tested for plant growth-promoting traits. Based on their multiple plant growth-promoting traits, eight potential isolates were selected and identified GY01, GY03, and GY08 are affiliated with Acinetobacter sp. GY04 and GY05 are affiliated with Chryseobacterium sp. GY06 and GY07 are affiliated with Pseudomonas costantinii and Pseudomonas chlororaphis, respectively; and GY02 is affiliated with the Bacterium strain. All eight isolates were evaluated for their effects on seed germination and vigor index and potential antagonism against Botrytis fabae. Selected isolates showed positive effects on seed germination and vigor index with different potentials. Isolate GY04 resulted in the highest vigor index (501), while isolate GY08 achieved the lowest (218). B. fabae radial growth inhibition was found in all eight isolates. The isolates inhibited the radial growth of the test pathogen with an inhibition efficacy of 72.38 % in GY04 to 25.57 % in GY-03. Generally, the results of this study indicated the potential of these isolates as a microbial inoculant with multiple functions for faba beans.

Isolation of Azospirillum spp and Evaluation of its Plant Growth Promoting Abilities

Background: This study aimed to isolate Azospirillum species from the soil environment and assess their plant growth-promoting abilities. Soil samples were collected from 10 different agricultural sites. The bacteria were isolated by serially diluting the soil samples and each aliquot was transferred into a Nitrogen-free bromothymol blue medium and then incubated for 7 days at 37°C. The isolates were further screened based on their morphological, biochemical and plant growth phytohormone production. The Fourier Transform Infrared Spectrophotometer will be used to elucidate the functional groups present in the isolates. Furthermore, the isolates were subjected to molecular identification. The results showed that Seventeen (17) bacteria were isolated from the soil samples of different crop plants using nitrogen-free bromothymol blue medium with all forming a white subsurface pellicle and they were further screened to seven (7) isolates, based on their morphological, biochemical, and plant growth phytohormones production characteristics, they include A1, A2, A3, A4, A5, A6 and A7. Three of the isolates (A1, A3, and A6) were randomly selected for molecular identification. The Fourier Transform Infrared Spectrophotometer identified the functional groups that are present in the Azospirillum strains to be aromatic compounds. All isolates showed ammonium production potential with A7 having the highest production of 781.3 µg/ml. Only four (4) isolates showed the ability to solubilize Calcium triphosphate with A7 having the highest solubilization (776 µg/ml). Furthermore, all isolates exhibited the ability to produce Indole acetic acids in the growth medium with A6 having the highest production (798.5 µg/ml). The molecular analysis and the sequence analysis of the three isolates 16SrRNA showed two of the isolates to be Azospirillum brasilense (A1, A3) and the other Azospirillum lipoferum (A6). Therefore, this study shows that this strain of Azospirillum has a high ability to produce Indole Acetic Acids, which aids in the production of phytohormones such as Auxins, Solubilize Calcium triphosphate and hence proffer a sustainable solution to improving plant growth.

Unlocking the plant growth-promoting potential of yeast spp.: exploring species from the Moroccan extremophilic environment for enhanced plant growth and sustainable farming.

In this study, we successfully isolated two distinct yeasts from Moroccan extreme environments. These yeasts were subjected to molecular characterization by analyzing their Internal Transcribed spacer (ITS) regions. Our research thoroughly characterizes plant growth-promoting abilities and their drought and salt stress tolerance. In a greenhouse assay, we examined the impact of selected yeasts on Medicago sativa's growth. Four treatments were employed: (i) control without inoculation (NI), (ii) inoculation with L1, (iii) inoculation with L2, and (iv) inoculation with the mixture L1+L2. L1 isolated from Toubkal Mountain shared 99.83% sequence similarity to Rhodotorula mucilaginosa. Meanwhile, L2, thriving in the arid Merzouga desert, displayed a similar identity to Naganishia albida (99.84%). Yeast strains were tolerant to NaCl (2M) and 60% PEG (polyethylene glycol P6000) in case of drought. Both strains could solubilize phsphorus, with L2 additionally demonstrating potassium solubilization. In addition, both strains produce indole acetic acid (up to 135µl ml-1), have siderophore ability, and produce aminocyclopropane-1-carboxylic acid deaminase. Isolates L1 and L2, and their consortium showed that the single or combined strain inoculation of M. sativa improved plant growth, development, and nutrient assimilation. These findings pave the way for harnessing yeast-based solutions in agricultural practices, contributing to enhanced crop productivity and environmental sustainability.

Transcriptomic analysis and carbohydrate metabolism-related enzyme expression across different pH values in Rhizopus delemar.

pH is one of the important factors affecting the growth and performance of microorganisms. We studied the pH response and plant growth-promoting (PGP) ability of Rhizopus delemar using cultivation experiments and transcriptomics, and verified the expression profiles using quantitative real-time PCR. pH affected the growth and PGP properties of R. delemar. At pH 7, the growth rate of R. delemar was rapid, whereas pH 4 and 8 inhibited mycelial growth and PGP ability, respectively. In the pot experiment, the plant height was the highest at pH 7, 56 cm, and the lowest at pH 4 and pH 5, 46.6 cm and 47 cm, respectively. Enzyme activities were highest at pH 6 to pH 7. Enzyme activities were highest at pH 6 to pH 7. Among the 1,629 differentially expressed genes (DEGs), 1,033 genes were up-regulated and 596 were down-regulated. A total of 1,623 DEGs were annotated to carbohydrate-active enzyme coding genes. The PGP characteristics, e.g., Phosphorus solubilization ability, of R. delemar were strongest at pH 7. The results provide useful information regarding the molecular mechanism of R. delemar pH response.

Enhancing rice growth and yield with weed endophytic bacteria Alcaligenes faecalis and Metabacillus indicus under reduced chemical fertilization.

Endophytic bacteria, recognized as eco-friendly biofertilizers, have demonstrated the potential to enhance crop growth and yield. While the plant growth-promoting effects of endophytic bacteria have been extensively studied, the impact of weed endophytes remains less explored. In this study, we aimed to isolate endophytic bacteria from native weeds and assess their plant growth-promoting abilities in rice under varying chemical fertilization. The evaluation encompassed measurements of mineral phosphate and potash solubilization, as well as indole-3-acetic acid (IAA) production activity by the selected isolates. Two promising strains, tentatively identified as Alcaligenes faecalis (BTCP01) from Eleusine indica (Goose grass) and Metabacillus indicus (BTDR03) from Cynodon dactylon (Bermuda grass) based on 16S rRNA gene phylogeny, exhibited noteworthy phosphate and potassium solubilization activity, respectively. BTCP01 demonstrated superior phosphate solubilizing activity, while BTDR03 exhibited the highest potassium (K) solubilizing activity. Both isolates synthesized IAA in the presence of L-tryptophan, with the detection of nifH and ipdC genes in their genomes. Application of isolates BTCP01 and BTDR03 through root dipping and spraying at the flowering stage significantly enhanced the agronomic performance of rice variety CV. BRRI dhan29. Notably, combining both strains with 50% of recommended N, P, and K fertilizer doses led to a substantial increase in rice grain yields compared to control plants receiving 100% of recommended doses. Taken together, our results indicate that weed endophytic bacterial strains BTCP01 and BTDR03 hold promise as biofertilizers, potentially reducing the dependency on chemical fertilizers by up to 50%, thereby fostering sustainable rice production.

Duckweed-associated bacteria as plant growth-promotor to enhance growth of Spirodela polyrhiza in wastewater effluent from a poultry farm.

Duckweed has been highlighted as an invaluable resource because of its abilities to remove nitrogen and phosphorus from wastewater coupling with the production of high starch/protein-containing plant biomass. Duckweed recruits microbes and particularly forms a stable "core" bacterial microbiota, which greatly reduces the colonization efficiency of plant growth-promoting bacteria (PGPB). In this study, natural duckweeds were enriched in a sterilized-partially treated wastewater effluent from a poultry farm. After 24days of cultivation, the duckweed-associated bacteria (DAB) were isolated and evaluated for their plant growth-promoting (PGP) potentials by co-cultivation with axenic Spirodela polyrhiza. Ten species were found in more than one location and could be considered candidates for the stable "core" DAB. Among them, all isolates of Acinetobacter soli, Acidovorax kalamii, Brevundimonas vesicularis, Pseudomonas toyotomiensis, and Shinella curvata increased duckweed growth in Hoagland medium. The highest PGP ability was observed in Sh. curvata W12-8 (with EPG value of 208.72%), followed by Paracoccus marcusii W7-16 (171.31%), Novosphingobium subterraneum W5-13 (156.96%), and Ac. kalamii W7-18 (156.96%). However, the highest growth promotion in the wastewater was observed when co-cultured with W7-16, which was able to increase biomass dry weight and root length of duckweed by 3.17 and 2.26 folds, respectively.

Endophytic Plant Growth-Promoting Rhizobacteria Promote Dendrobium Orchid Growth

Dendrobium orchid is one of the major export products of Thailand. Endophytic bacteria inhabiting the roots have the ability to promote plant growth. They could be used to replace the chemical fertiliser. In this study, 42 bacterial isolates were isolated from the orchid roots and were screened for plant growth- promoting rhizobacteria (PGPR). We selected six isolates that exhibited plant growth promotion (PGP) abilities, including siderophores, IAA, and ACC deaminase production, phosphate-solubilisation, and nitrogen-fixation. The 16S rRNA gene sequencing identified these isolates as Enterobacter sp. and Bacillus sp. with a prominence of Bacillus halotolerans. All six isolates were confirmed to inhabit the orchid roots and were inoculated into the orchid hybrid (D. anosmum × D. parishii) seedlings. We found that B. halotolerans O-SWU-17 enhanced orchid seedling growth in vivo. Thus, B. halotolerans O-SWU- 17 has great potential as a biofertiliser in enhancing orchid production.
