A Screen of Plant-Based Natural Products Revealed That Quercetin Prevents Pyroglutamylated Amyloid-β (Aβ3(pE)-42) Uptake in Astrocytes As Well As Resulting Astrogliosis and Synaptic Dysfunction.

Two connected histopathological hallmarks of Alzheimer's disease (AD) are chronic neuroinflammation and synaptic dysfunction. The accumulation of the most prevalent posttranslationally modified form of Aβ1-42, pyroglutamylated amyloid-β (Aβ3(pE)-42) in astrocytes is directly linked to glial activation and the release of proinflammatory cytokines that in turn contribute to early synaptic dysfunction in AD. At present, the mechanisms of Aβ3(pE)-42 uptake to astrocytes are unknown and pharmacological interventions that interfere with this process are not available. Here we developed a simple screening assay to identify substances from a plant extract library that prevent astroglial Aβ3(pE)-42 uptake. We first show that this approach yields valid and reproducible results. Second, we show endocytosis of Aβ3(pE)-42 oligomers by astrocytes and that quercetin, a plant flavonol, is effective to specifically block astrocytic buildup of oligomeric Aβ3(pE)-42. Importantly, quercetin does not induce a general impairment of endocytosis. However, it efficiently protects against early synaptic dysfunction following exogenous Aβ3(pE)-42 application.

Eucalyptus Essential Oil Based Nanoemulsions: Preparation and Biological Activities.

Eucalyptus essential oil has remarkable industrial importance and biological properties due to its effectiveness against various diseases, reported throughout human history. Despite the extraordinary bioactivities of essential oil, its applications are limited due to volatility, insolubility in water, and less stability. Formulation of nanoemulsion is the best way to enhance the bio-efficacy of this essential oil and eliminate the factors responsible for limited application. This review article compiles the information regarding formulation of Eucalyptus essential oil-based nanoemulsion and their several biological activities and medicinal properties including antibacterial, antifungal, larvicidal, insecticidal, and cytotoxic activities etc. The bio-efficacy of essential oil-based nanoemulsion has also been found to be enhanced as compared utilization of essential oil alone. This review can be beneficial for researchers working on medicinal plant-based natural products, specifically containing Eucalyptus essential oil, to explore new research horizons in this emerging field.

Evaluation of methyl salicylate, limonene, and thymol as potential biocidal against Trogoderma granarium everts larvae

The investigation of plant-based natural products, especially essential oils, as insecticide alternatives, responds to challenges as induced resistance and environmental drawbacks linked to prolonged synthetic insecticide use. Although essential oils demonstrate promising activity, their utilization faces hurdles related to standardization. In this study, contact/fumigation properties of three molecules (methyl salicylate, limonene and thymol) known for their activity against stored product beetles, were tested in laboratory bioassays toward young and old larval stages of Trogoderma granarium. The experiment revealed significant differences in their effects based on larval age and chemical concentration. Methyl salicylate exhibited concentration-dependent effects on both young and old larvae, with higher concentrations leading to accelerated mortality rates. For instance, young larvae exposed to 10 mg displayed an LT50 of 33.75 h, while old larvae showed an LT50 of 39.07 h. Similarly, at a concentration of 5 mg, young and old larvae exhibited LT50 values of 72.79 and 79.68 h, respectively, after treatment. Limonene also exhibited concentration-dependent effects, although with lower potency compared to methyl salicylate. Thymol showed efficacy only against young larvae of T. granarium. Statistical analyses confirmed the significant impact of each chemical on larval survival time, with tested chemicals displaying significant concentration-dependent variations in mortality rates. These findings suggest the potential of methyl salicylate, and to a lesser extent limonene and thymol, as effective biocidal agents against T. granarium larvae.

Natural product cell division cycle 25 (CDC25) inhibitors: A promising source for cancer drug discovery

Cell division cycle 25 (CDC25) is a family of proteins that play a vital role in regulating cell cycle progression. They activate cyclin-dependent kinases (CDKs), which are key enzymes controlling pivotal stages throughout the cell cycle to ensure proper transition between phases. Due to their overexpression or dysregulation in many human cancers, CDC25 phosphatases have become a prime target for cancer therapy. A vast number of structurally diverse molecules have been identified as potent and selective CDC25 phosphatase inhibitors, exhibiting promising therapeutic potential against various cancer types. However, only a small subset of CDC25 inhibitors such as pexiganan, PM-20, BN82002, and JUN1111 have progressed to clinical trials for evaluation. Natural products are being explored as potential sources of novel anticancer compounds due to their functional diversity. Plant-based and marine-based natural products are being investigated for CDC25 inhibition and for the development of phosphatase inhibitors. This review article comprehensively covers the role of CDC25 in the cell cycle, plant-based and marine-based natural products for CDC25 inhibition, and recent advances in the development of CDC25 phosphatase inhibitors as novel anticancer agents.

Immunomodulatory Plant Natural Products as Therapeutics against Inflammatory Skin Diseases.

Frequently occurring inflammatory skin conditions such as psoriasis, dermatitis, acne, including skin cancer, wounds and other disorders arising out of premature skin aging, deteriorate skin health and adversely impact human life. Even though several synthetic compounds have evolved for treating these skin conditions, natural-product-based therapeutics are gaining popularity with growing evidence of their efficacy and safety for treating skin disorders. Many of these inflammatory skin diseases have underlying disturbances in our immune system and immunomodulatory natural products provide solutions for their effective treatment and aid in understanding the underlying mechanism of such inflammatory skin conditions. Based on this premise, the present review summarizes the possible application of plant-derived immunomodulatory compositions and single molecules for treating inflammatory skin conditions. In vitro, in vivo and mechanistic studies reported the application of selected plant-derived natural products for the treatment of inflammatory skin disorders including, cancer and infections. Several online databases including PubMed, Google Scholar, and Science Direct have been searched for gathering the information covered in this review. Empirical studies demonstrated that most of these natural compounds exhibited therapeutic properties through their immunomodulatory and anti-inflammatory potential supplemented often with anti-microbial, anti-neoplastic, and anti- oxidant activities. Overall, plant-based natural products discussed here are capable of modulating the immune system to minimize or completely suppress the pro-inflammatory markers, scavenge free radicals (ROS), prevent bacteria, fungal, and virus-derived skin infections and often regress skin cancer through the induction of apoptosis. The challenges and opportunities associated with the application of plant-based immunomodulators for skin applications and their safety considerations are also discussed here. The present study indicated that immunomodulatory plant natural products being biologically validated ligands against various biological targets manifested in inflammatory skin diseases, offer an effective, safe and affordable treatment for such disorders affecting skin health. However, further clinical evaluations are needed to substantiate these findings.

Research Progress on Sesquiterpenoids of Curcumae Rhizoma and Their Pharmacological Effects.

Curcumae Rhizoma, a traditional Chinese medicine with a wide range of pharmacological activities, is obtained from the dried rhizomes of Curcuma phaeocaulis VaL., Curcuma kwangsiensis S. G. Lee et C. F. Liang, and Curcuma wenyujin Y. H. Chen et C. Ling. Sesquiterpenoids and curcuminoids are found to be the main constituents of Curcumae Rhizoma. Sesquiterpenoids are composed of three isoprene units and are susceptible to complex transformations, such as cyclization, rearrangement, and oxidation. They are the most structurally diverse class of plant-based natural products with a wide range of biological activities and are widely found in nature. In recent years, scholars have conducted abundant studies on the structures and pharmacological properties of components of Curcumae Rhizoma. This article elucidates the chemical structures, medicinal properties, and biological properties of the sesquiterpenoids (a total of 274 compounds) isolated from Curcumae Rhizoma. We summarized extraction and isolation methods for sesquiterpenoids, established a chemical component library of sesquiterpenoids in Curcumae Rhizoma, and analyzed structural variances among sesquiterpenoids sourced from Curcumae Rhizoma of diverse botanical origins. Furthermore, our investigation reveals a diverse array of sesquiterpenoid types, encompassing guaiane-type, germacrane-type, eudesmane-type, elemane-type, cadinane-type, carane-type, bisabolane-type, humulane-type, and other types, emphasizing the relationship between structural diversity and activity. We hope to provide a valuable reference for further research and exploitation and pave the way for the development of new drugs derived from medicinal plants.

Dosage Evaluation and Antioxidant Activity Test of Lotion of Ethanol Extract of Belimbing Wuluh Leaf (Averrhoa Bilimbi L.)

<div><table cellspacing="0" cellpadding="0" align="left"><tbody><tr><td align="left" valign="top"><p><em>This study aims to make and evaluate the lotion formula from ethanol extract of star fruit leaves (Averrhoa bilimbi L.) and test the antioxidant activity of the lotion. The research method used is experimental, making lotions and testing antioxidant activity from July to October 2023 at the Semi-Solid Preparation Technology Laboratory, Department of Pharmacy, Poltekkes Kemenkes Medan, and Research Laboratory, University of North Sumatra. In the preparation stage, star fruit leaf simplisia was extracted by maceration using a 95% ethanol solvent. The lotion formula was made with ingredients such as PEG 4000, stearyl alcohol, glycerin, sodium lauryl sulfate, coconut oil, nipagin, and distillate water, with varying concentrations of ethanol extract of star fruit leaves. The formulation was evaluated through organoleptic, homogeneity, pH, spreadability, and viscosity tests. The results showed that the lotion produced had a homogeneous shape and aroma to the concentration of ethanol extract. In addition, the pH of the lotion is also within the safe range for human skin. The spreadability test showed the lotion's ability to spread on the skin, which met the requirements for lotions with spreadability values between 5 and 7 cm. The viscosity of the lotion was also by the set standards. Stability tests were conducted to monitor the physical and chemical changes of the lotion over four weeks. The results showed that lotions with 1% and 3% ethanol extract concentrations were stable during the study period. However, the 5% concentration experienced inhomogeneity changes in the third week. In conclusion, lotion with ethanol extract of star fruit leaves at 1% and 3% concentration can produce a good and stable formula. However, the antioxidant activity of the lotion is still in the weak to moderate category. Reviewing the Therapeutic Potential of Plant-based Natural Products Against Multidrug-resistant Bacteria (MDR)

Abstract: The global health of individuals is severely threatened by drug-resistant microorganisms. Existing antibiotics available in the market, such as penicillin, cephalosporins, new β- lactams, aminoglycosides, macrolides, lincomycin, and quinolones, are losing their effectiveness due to overuse and inappropriate prescribing, among other factors. This situation underscores the urgent need to develop new antimicrobial agents. In response to this challenge, attention has been directed towards natural products derived from plants, which have been utilized for their medicinal benefits over an extended period. Various studies have demonstrated that plant-based natural products exhibit antibacterial action against multidrug-resistant (MDR) bacteria, making them potential candidates for the development of novel antimicrobial medicines. Plants belonging to classes, such as alkaloids, organosulfur compounds, terpenoids, coumarin, and polyphenols have been found to possess antimicrobial properties. This paper reviews the antimicrobial activity of plant-based natural products, whether used alone or in combination with conventional antibiotics. Additionally, the paper explores their mechanisms against drugresistant bacteria, the potential development of resistance against them, their current role in medicine, and their future perspectives.

Natural Compounds and Breast Cancer: Chemo-Preventive and Therapeutic Capabilities of Chlorogenic Acid and Cinnamaldehyde.

Globally, breast cancer is not only the most frequently diagnosed cancer but also the leading cause of cancer death in women. Depending on breast cancer histotype, conventional breast cancer treatment options vary greatly in efficacy and accompanying side effects. Thus, there is a need for more effective and safer strategies that impact breast cancer at all stages. Plant-based natural products are easily available, with them proving effective and inexpensive. Two such phytochemicals are chlorogenic acid and cinnamaldehyde. Studies have shown their efficacy against different molecular subtypes of breast cancers in vitro and in vivo. In this review, we discuss their current status in anticancer research with specific emphasis on chlorogenic acid and cinnamaldehyde. We describe their multiple mechanisms of action in destroying breast cancer cells, their potential uses, and the need for translational applications. We also include future directions for investigations to progress chlorogenic acid and cinnamaldehyde research from bench to bedside.

Reduced Production of Pro-Inflammatory and Pro-Catabolic Factors by Human Serum Metabolites Derived from a Patented Saffron Extract Intake.

Safe and anti-inflammatory plant-based natural products present an increasing focus in the treatment of chronic inflammatory diseases such as osteoarthritis or inflammatory bowel diseases. Among them, saffron, a spice derived from the stigma of Crocus sativus, could have anti-inflammatory properties and would be therefore a promising therapeutic agent for the treatment of such conditions. However, the anti-inflammatory molecular mechanisms of saffron in humans are still understudied and unclear. In this study, combining human serum metabolites and cell cultures, we evaluated the effect of circulating metabolites from the consumption of a patented saffron extract (Safr'InsideTM) on the chondrocytes and colon epithelial cell responses to inflammatory stress. Parametric or non-parametric Analysis of Variance with post hoc tests was performed. We demonstrated that human serum containing metabolites from saffron intake attenuated IL-1β-stimulated production of PGE2 and MMP-13 in chondrocyte cells and limited the increase in ICAM-1, MCP-1, iNOS, and MMP-3 in human epithelial cells following combined IL-1β and TNF-α inflammatory stimulation. Altogether, these data provide new findings into the mechanisms underlying the beneficial effects of saffron on chondrocytes and enterocyte cells at the cellular level and in the context of chronic inflammatory disorders.

Exploring biocidal effects of methyl salicylate and limonene toward <em>Trogoderma granarium</em> Everts

The khapra beetle, Trogoderma granarium, poses a significant threat as a post-harvest pest in stored products and stands out as a crucial quarantine concern globally. Unlike many other stored product pests, this species proved challenging to manage using conventional insecticides and alternative non-chemical methods. The exploration of plant-based natural products, particularly essential oils, as alternatives arises in response to challenges associated with the prolonged use of synthetic insecticides. While essential oils show promising activity, their use encounters challenges associated with standardization. This study investigates the feasibility of employing some single chemical compounds widespread in several essential oils with insecticidal activity as candidate insecticides, specifically limonene and methyl salicylate in contact/fumigation bioassays toward T. granarium adults. The results showed that methyl salicylate caused a lethal time of 50% (LT50) at the dose of 1 mg after 7.40 hours of exposure. The LT50 calculated for limonene was 86.83 hours while positive control using deltamethrin, used at the recommended dose of the manufacturer, was 5.20 hours after exposure. These data suggest that methyl salicylate can be exploited as a candidate for further tests in field conditions toward T. granarium also in consideration of its relative low toxicity for humans.

Guidelines for the Diagnosis and Treatment of Rotator Cuff Tear with Integrated Traditional Chinese and Western Medicine.

Rotator cuff tear is a common injury among middle-aged and elderly people, and it has a great impact on patients' physical and mental health and quality of life. Integrative medicine based on Traditional Chinese Medicine (TCM) has certain advantages in the diagnosis and treatment of rotator cuff tears. TCM, which mainly involves the use of plant-based products, has relatively stable and reliable curative effects. It is of great significance to formulate a combined diagnosis and treatment plan for rotator cuff tear based on evidence-based medicine, which can help to standardize the clinical diagnosis and treatment techniques of TCM and Western medicine and achieve better therapeutic effects. This guideline standardizes the diagnosis and treatment process of rotator cuff tear from the aspects of range, terminology and definition, diagnosis, TCM syndrome differentiation, treatment, functional exercise, and prevention and care. It makes recommendations that cover the adoption of manual therapy, acupuncture, and other integrative medicine based on TCM. Users of these guidelines are most likely to include clinicians and health managers in healthcare settings.

Phytochemistry, Biological Activities, and Chemical Profiling of Berberis asiatica

This study focused on chemical profiling and assessed the total phenolic and flavonoid content as well as the antioxidant and antibacterial effect of the medicinal plant Berberis asiatica. The results revealed that this plant has high concentrations of TPC (Total phenol content) and TFC (Total flavonoid content) of 37.686 ± 2.728 mg GAE/g and 115.568 ± 8.012 mg QE/g, respectively. The DPPH free radical scavenging assay demonstrated strong inhibition, with an IC50 of 205.7 ± 5.353 μg/mL, and also showed robust antibacterial properties against Staphylococcus aureus and Klebsiella pneumoniae with a zone of inhibition (ZOI) of 14 mm and 19 mm, respectively. The extract exhibited an excellent inhibitory potential against S. aureus, and K. pneumoniae with an MIC (Minimum inhibitory concentration) of 0.39 mg/mL, and 3.125 mg/mL respectively, indicating significant inhibitory action. Furthermore, the MBC (Minimum bactericidal concentration) for both S. aureus and K. pneumoniae was found to be 6.25 mg/mL, emphasizing the extract's consistent bactericidal effectiveness against these bacteria. These findings underscore the potential utility of the methanolic extract of Berberis asiatica as a natural antibacterial agent. GC-MS analysis of hexane fraction indicates the plant is rich in secondary metabolites, specifically 2,2-dimethyl-3-pentanol, 2-methyl-2-pentanol, 2,5-dimethyl-4-hydroxy-3-hexanone, 3-hexanol, 4-methyl-2-pentanol are identified. Overall, this study highlights the importance of plant-based natural products as potential sources of antioxidants and antibacterial agents that contributes to the future drug development process.

Unlocking the Therapeutic Potential of Medicinal Plants for Alzheimer’s Disease: Preclinical to Clinical Trial Insights

Alzheimer’s disease (AD) is a progressive, multifactorial, and unremitting neurodegenerative disease characterized by memory loss, personality changes, and cognitive impairment. It has become more prevalent in recent years. Therefore, understanding the pathophysiology of AD and developing efficient therapeutic strategies are essential. Moreover, the progression of the disease is unaffected by the pharmaceutical approaches discovered to date. Additionally, the failure of over 200 potential drug candidates in clinical trials over the past decade suggests the complexity and difficulty of both the disease and its underlying causes. Therefore, research focused on medicinal plant-based natural products in the search for novel neuroprotective therapeutic candidates for AD is essential. Indeed, several scientific investigations have demonstrated the efficacy of many medicinal plants and their principal phytochemicals in the treatment of AD. This review article covered the pathophysiological mechanisms of AD, the necessity for natural products as anti-AD treatments, and the most recent preclinical studies revealing the function of neuroprotective medicinal plants and their bioactive compounds in the effective management of AD. In addition, the review also presents clinical trial data of promising anti-AD formulations/agents of plant origin. Revealing recent findings and highlighting the clinical trial data related to the development of new treatments for AD would promote further research in this field and pave the way for the development of more effective and safe treatments for this debilitating disease.

Guidelines for the diagnosis and treatment of knee osteoarthritis with integrative medicine based on traditional Chinese medicine.

Knee osteoarthritis (KOA) is a common geriatric disease in middle-aged and elderly people. Its main pathological characteristics are articular cartilage degeneration, changes in subchondral bone reactivity, osteophyte formation at joint edges, synovial disease, ligament relaxation or contracture, and joint capsular contracture. The prevalence rate of symptomatic KOA in middle-aged and elderly people in China is 8.1%, and this is increasing. The main clinical manifestations of this disease are pain and limited activity of the knee joint, which seriously affect the quality of life of patients and may cause disability, posing a huge burden on society and the economy. Although the pathogenesis of KOA is not clear, the treatment of KOA is diverse, and Chinese medicine, which mainly relies on plant-based natural products, has a relatively stable and reliable curative effect. This guideline aims to emphasize the evidence-based staging and stepped treatment of KOA and the therapeutic effect of integrative medicine based on traditional Chinese medicine on KOA. We make recommendations that include the adoption of manual therapy, acupuncture, external application of herbs, herbal plasters, exercise therapy, and other integrative medicine based on traditional Chinese medicine. Users of the above guidelines are most likely to include clinicians and health managers in healthcare settings.

Research progress on the treatment of knee osteoarthritis combined with osteoporosis by single-herb Chinese medicine and compound.

Knee osteoarthritis (KOA) is a degenerative disease with synovial inflammation, articular surface cartilage degeneration, meniscus degeneration, ligament and muscle changes, subchondral bone changes, and osteophyte formation around the joint as the main pathological changes. Osteoporosis (OP) is a disease characterized by low bone mass and deterioration of the microstructure of bone tissue. KOA and OP are both geriatric diseases, and the incidence of KOA combined with OP is high, but there is a lack of specific drugs, and the major treatments are limited to drug therapy. Most traditional Chinese medicine (TCM) treatments use plant-based natural products, and they help patients obtain good clinical benefits and at the same time provide researchers with ideas to study the mechanism of disease occurrence and the relationship between the two diseases. This article summarizes the research progress of TCM monomers and TCM compounds that are frequently used to treat KOA combined with OP to provide ideas for future clinical treatments and related basic research.

Network pharmacology-based findings of the immunomodulatory activity of phytocompounds from Withania somnifera and Aloe barbadensis

Immunomodulation constitutes a crucial part of individual organisms’ defense systems. Moreover, the utilization of plant-based natural products as herbal medicine for immunomodulation has garnered significant interest. Herein, we examined the immunomodulatory potentials of active phytocompounds extracted from Withania somnifera and Aloe barbadensis by employing ADMET screening, network pharmacology, and molecular docking techniques. This study follows the paradigm in drug discovery, which has shifted from a “one-target, one-drug” mode to a “network-target, multiple-component-therapeutics” mode. Phyto compounds sourced from W. somnifera and A. barbadensis were mined from online databases, including Dr. Duke’s Phytochemical Ethnobotanical Database. After screening these active compounds, their potential targets were predicted through in silico ADMET property prediction models. Network pharmacology was utilized to establish a “compound-protein/gene-disease” network and reveal the regulatory mechanism of small molecules in a high-throughput manner through STRING, Cytohubba plugin in Cytoscape, and the g: Profiler software. A molecular docking simulation was performed to examine the binding affinity between the selected hub targets and bioactives. The findings showed that phytocompounds derived from the W. somnifera and A. barbadensis exhibit immunomodulatory effects by inhibiting specific protein targets, notably AKT1, HCK, JAK2, PDPK1, KIT, and IL2. Molecular docking analysis further revealed the potential of withanolide G, somniferine, and somniferanolide as promising immunomodulatory compounds against HCK, JAK2, and PDPK1 proteins, which are involved in multiple myeloma pathways, encompassing the PI3K-Akt signaling pathway, NOD-like receptor signaling pathway, and Toll-like receptor signaling pathway. In conclusion, these compounds are recommended for further in vivo and in vitro investigations to ascertain their potential as treatments for multiple myeloma.

In vitro antioxidant, antiglycation, α-glucosidase and α-amylase inhibitory activities of extracts and solvent fractions of Elaeocarpus serratus L.

Plants are an essential source of unique pharmacologically active natural products beneficial to human and animal health. In this research, crude extracts and solvent fractions from the leaves, bark, stem, seeds and fruit pulp of Elaeocarpus serratus (Cylon olive) were assessed for their antioxidant potential, antiglycation and enzymatic inhibitory activity. Phytochemical contents were analyzed by the colorimetric method. Antioxidant activities were estimated by DPPH radical scavenging, superoxide anion radical and ferrous ion chelating assays. Antiglycation properties were determined through the bovine serum albumin-methylglyoxal (BSA-MG) test and enzyme inhibitory activities were tested in vitro on α-glucosidase and α-amylase. The ethyl acetate fraction of the leaves exhibited high phenolic content (71.63 ± 0.01 mg GAE/g DW). The n-butanol fraction of the bark had high flavonoid content (37.37 ± 0.07 mg QE/g DW). The n-butanol fraction of the bark also significantly inhibited both α-glucosidase and α-amylase with IC50 values of 2.32 ± 0.62, and 2.81 ± 0.37 μg/mL, respectively, compared with quercetin (43.27 ± 3.24 μg/mL) and acarbose (17.72 ± 2.35 μg/mL). These findings indicate that E. serratus can be a source of plant-based pharmaceuticals and natural products, which may be considered for the treatment of diabetes and the formulation of healthy food supplements. However, further studies are required on the isolation, identification and toxicity of components from E. serratus.

Essential Oils of Garcinia spp. and Their Biological Activities

Garcinia is the largest genus in the Clusiaceae family. Plants of Garcinia are widely distributed in Africa, Asia, Australia, Central America, Madagascar, and Polynesia. There are over 400 species of Garcinia found in the lowland tropical forests. Garcinia are known for their medicinal values due to the presence of biologically active constituents. Plants with medicinal values play a key role in human health and disease prevention. Plant-based natural products such as essential oils (EOs) have been employed for centuries owing to their beneficial effects. Generally, EOs are a mixture of volatile chemical constituents comprising mainly terpenoids and phenylpropanoids. They play diverse roles as biologically active agents. To date, EOs have found wide applications in pharmaceutical, food, flavour and fragrance. This review discusses the volatile chemical constituents of EOs extracted from different plant parts of Garcinia. The biological activities of Garcinia EOs are highlighted to explore their therapeutic potential.
