Interspecific root interaction enhances cadmium accumulation in Oryza sativa when intercropping with cadmium accumulator Artemisia argyi

The contamination of arable land with heavy metals, such as Cd, is a serious concern worldwide. Intercropping with Cd accumulators can be used for efficient safe crop production and phytoremediation of Cd-contaminated soil. However, the effect of intercropping on Cd uptake by main crops and accumulators varies among plant combinations. Rhizosphere interaction may mediate Cd uptake by intercropped plants, but the mechanism is unclear. Thus, in the present study, we aimed to examine the effect of rhizosphere interaction on Cd uptake by intercropping rice (Oryza sativa L.) with mugwort (Artemisia argyi Levl. et Vant.) in Cd-contaminated paddy soil. We grew O. sativa and A. argyi in pots designed to allow different levels of interaction: complete root interaction (no barrier), partial root interaction (nylon mesh barrier), and no root interaction (plastic film barrier). Our results indicated that both complete and partial root interaction increased the shoot and root mass of A. argyi, but did not decrease the shoot, root, and grain mass of O. sativa. Interspecific root interaction significantly increased the Cd content in the shoots, roots, and grains of O. sativa and the shoots of A. argyi. Increased content of total organic acids in the rhizosphere, which increased the content of available Cd, was a possible mechanism of increased Cd uptake in both plants under interspecific root interaction. Our findings demonstrate that an intercropping system can extract more Cd from contaminated soil than a monocropping system of either A. argyi or O. sativa. However, the intercropping system did not facilitate safe crop production because it substantially increased grain Cd content in O. sativa.

NPs-Ca promotes Cd accumulation and enhances Cd tolerance of rapeseed shoots by affecting Cd transfer and Cd fixation in pectin

The use of rapeseed (Brassica napus) as a hyperaccumulator plant has shown great promise for the remediation of cadmium (Cd) contaminated soils. Nanosized materials (NPs) have been shown to mitigate heavy metal toxicity in plants, but it is unknown how l-aspartate nano-calcium (NPs-Ca) affects Cd uptake, transport, and tolerance in rapeseed. A soil pot experiment was conducted with two treatments: a control treatment (CK) with 2.16 g CaCl2 and NPs-Ca treatment with 6.00 g NPs-Ca, to evaluate the effects and mechanisms of NPs-Ca on Cd tolerance in rapeseed. Compared to CaCl2, NPs-Ca promoted Cd transportation from roots to shoots by up-regulating the expression of Cd transport genes (ABCC12, HMA8, NRAM6, ZIP6, CAX4, PCR2, and HIP6). Therefore, NPs-Ca increased Cd accumulation in rapeseed shoots by 39.4%. Interestingly, NPs-Ca also enhanced Cd tolerance in the shoots, resulting in lower hydrogen peroxide (H2O2) accumulation and proline content, as well as higher antioxidant enzyme activities (POD, CAT). Moreover, NPs-Ca reduced the activity of pectin-degrading enzymes (polygalacturonase: PG, β-galactosidase: β-GAL), promoted the activity of pectin methyl esterase (PME), and changed transcription levels of related genes (PME, PMEI, PG, PGIP, and β-GAL). NPs-Ca treatment also significantly increased the Cd content in cell walls by 59.8%, that is, more Cd was immobilized in cell walls, and less Cd entered organelles in shoots of NPs-Ca treatment due to increased pectin content and degree of pectin demethylation. Overall, NPs-Ca increased Cd accumulation in rapeseed shoots by promoting Cd transport from roots to shoots. And meantime, NPs-Ca enhanced Cd tolerance of shoots by inhibiting pectin degradation, promoting pectin demethylation and increasing Cd fixation in pectin. These findings suggest that NPs-Ca can improve the potential of rapeseed as a hyperaccumulator for the remediation of Cd-contaminated soil and the protection of the environment. Furthermore, the study provides a theoretical basis for the application of NPs-Ca in the phytoremediation of Cd-contaminated soils with hyperaccumulating plants.

Effect of cadmium stress on the bacterial community in the rhizosphere of mulberry (Morus alba L.).

Mulberry has a good tolerance to cadmium (Cd) and is considered a candidate plant for phytoremediation. The rhizosphere microbial community plays an important role in phytoremediation. Nevertheless, little information on the rhizosphere microbial community mechanisms in mulberry during the phytoremediation of Cd-contaminated soil is available. In this study, the remediation efficiency of mulberry in pots subjected to three simulated Cd pollution levels and their rhizosphere bacterial communities during the remediation process were analyzed. "Yuesang 11" was used as the test mulberry variety, and three simulated Cd pollution levels were set by adding three concentrations of Cd (Cd5, 5 mg kg-1; Cd3, 3 mg kg-1; Cd2, 2 mg kg-1). The results showed that the elimination rates of Cd in the rhizosphere soil were 81.7%, 85.3%, and 57.9% under the stress of the Cd2, Cd3, and Cd5 conditions, respectively. Meanwhile, 3,082,583 high-quality sequence reads and 976 operational taxonomic units were successfully obtained from the mulberry rhizosphere soil by high-throughput absolute quantification sequencing and further assigned to 11 bacterial phyla and 26 families. Of these, decreased abundances of 19 bacteria at the family level and increased abundances of seven bacteria under Cd stress were revealed by comparative analysis. Based on the alpha diversity indices (Chaol, Shannon and Simpson) and principal component analysis, the rhizosphere bacterial diversity of the Cd5 condition was significantly decreased, but that of the Cd2 and Cd3 conditions was not different from that of soil without Cd (CK). Likewise, redundancy analysis showed that the abundances of Acidobacteria Gp2, Acidobacteria Gp13, and Sphingobacteria were significantly positively associated with the elimination rates of Cd. This study suggested that the mulberry rhizosphere contains a relatively stable bacterial community consisting of diverse Cd-resistant bacteria, providing a scientific basis for remediating heavy-metal polluted soils using mulberry.

Soil microbial community compositions and metabolite profiles of Achnatherum inebrians affect phytoremediation potential in Cd contaminated soil

Cadmium (Cd) contamination poses serious risks to soil ecosystems and human health. Herein, the effect of two drunken horse grasses (Achnatherum inebrians) including endophytes Epichloë gansuensis infected (E+ ) and uninfected (E-) on the phytoremediation of Cd-contaminated soils were analyzed by coupling high-throughput sequencing and soil metabolomics. The results showed that the high-risk soil Cd decreased and the medium- and low-risk Cd fraction increased to varying degrees after planting E+ and E- plants in the soil. Meanwhile, total Cd content decreased by 19.7 % and 35.1 % in E+ and E- A. inebrians-planted soils, respectively. Principal coordinate analysis revealed a significant impact of E+ and E- plants on the soil microbial community. Most stress-tolerant and gram-positive functional bacterial taxa were enriched to stabilize Cd(II) in E+ planted soil. Several beneficial fungal groups related to saprotroph and symbiotroph were enriched to absorb Cd(II) in E- soil. Soil metabolomic analysis showed that the introduction of A. inebrians could weaken the threat of CdCl2 to soil microbe metabolism and improve soil quality, which in turn promoted plant growth and improved phytoremediation efficiency in Cd-contaminated soil. In conclusion, A. inebrians plants alleviate soil Cd pollution by regulating soil microbial metabolism and microbial community structure. These results provide valuable information for an in-depth understanding of the phytoremediation mechanisms of A. inebrians.

Cadmium Accumulation in Plants: Insights from Phylogenetic Variation into the Evolution and Functions of Membrane Transporters

Rapid industrialization during recent decades has resulted in the widespread contamination by cadmium (Cd) of agricultural soils, which has become a ubiquitous environmental problem and poses great risk to human health via the food chain. Cd accumulation greatly varies among different plant species and even within different genotypes of the same species across the plant kingdom. A better understanding of the physiological and molecular mechanisms underlying Cd uptake, translocation, sequestration, and (re)distribution in plants will shed light on developing strategies to minimize Cd in crops. Moreover, analysis of molecular evolution of the key transporters reveals that Cd transporters were highly conserved throughout the evolutionary lineage of the entire plant kingdom and underwent lineage-specific expansion as the result of gene duplication. However, different Cd transporters may experience different evolutionary lineages from algae to angiosperms, suggesting the divergence of their roles in plant adaptation to metalliferous soil. In summary, all the knowledge in the present review can be used to predict the transfer of Cd from soils to plants, to further understand the origins of Cd-accumulating phenotypes, and to discover the plant genetic resources for the breeding of low-Cd crops and the phytoremediation of Cd-contaminated soils.

Field-scale differences in rhizosphere micro-characteristics of Cichorium intybus, Ixeris polycephala, sunflower, and Sedum alfredii in the phytoremediation of Cd-contaminated soil

Understanding the intricate interplay between Cd accumulation in plants and their rhizosphere micro-characteristics is important for the selection of plant species with profitable Cd phytoextraction and soil remediation efficiencies. This study investigated the differences in rhizosphere micro-ecological characteristics and Cd accumulation in chicory, Ixeris polycephala, sunflower, and Sedum alfredii in low-moderate Cd-contaminated soil. Data reveal that the dominant organic acids in rhizosphere soil that responded to Cd were oxalic and lactic acids in chicory and Ixeris polycephala, tartaric acid in sunflower, and succinic acid in Sedum alfredii. These unique organic acids could also influence the abundance of specific rhizobacterial communities in rhizosphere soil that were Sphingomonadaceae and Bradyrhizobiaceae in both Sedum alfredii (9.75 % and 2.56 %, respectively) and chicory (8.98 % and 2.82 %, respectively) rhizosphere soil, Xanthomonadaceae in both Sedum alfredii and Ixeris polycephala rhizosphere soil, and Gaiellaceae in chicory rhizosphere soil. In this case, the combined effects of the organic acids and unique rhizobacterial communities by plant species increased the bioavailable concentration of Cd in Sedum alfredii, Ixeris polycephala, and sunflower rhizosphere soil, while decreasing the Cd-DOM concentrations in chicory rhizosphere soil and the water-extractable Cd reduced by 88.02 % compared to the control. Though the capacity for Cd accumulation in the shoots of chicory was weaker than of Sedum alfredii but better than either Ixeris polycephala or sunflower, chicory presented better Cd translocation and harbored Cd mainly as the low toxic chemical form of pectates and proteins-bound Cd and Cd oxalate in its shoot. Generally, chicory, as an economic plant, is suitable for phytoremediation of low-moderate Cd-contaminated soil after Sedum alfredii.

Proteomic analysis reveals differential responsive mechanisms in Solanum nigrum exposed to low and high dose of cadmium

Cadmium (Cd) contamination is becoming a widespread environmental problem. However, the differential responsive mechanisms of Cd hyperaccumulator Solanum nigrum to low or high dose of Cd are not well documented. In this study, phenotypic and physiological analysis firstly suggested that the seedlings of S. nigrum showed slight leaf chlorosis symptoms under 25 μM Cd and severe inhibition on growth and photosynthesis under 100 μM Cd. Further proteomic analysis identified 105 differentially expressed proteins (DEPs) in the Cd-treated leaves. Under low dose of Cd stress, 47 DEPs are mainly involved in primary metabolic processes, while under high dose of Cd stress, 92 DEPs are mainly involved in photosynthesis, energy metabolism, production of phytochelatin and reactive oxygen species (ROS). Protein-protein interaction (PPI) network analysis of DEPs support above differential responses in the leaves of S. nigrum to low and high dose of Cd treatments. This work provides the differential responsive mechanisms in S. nigrum to low and high dose of Cd, and the theoretical foundation for the application of hyperaccumulating plants in the phytoremediation of Cd-contaminated soils.

Polyaspartic acid assisted-phytoremediation of cadmium-contaminated farmland: Phytoextraction efficiency, soil quality, and rhizosphere microbial community

Cadmium is highly toxic and one of the most dangerous metal pollutants in soil, and poses a serious threat to human health through soil-crop-food chain transmission. Polyaspartic acid (PASP) is a biodegradable additive that is environment-friendly compared to traditional chelating agents. Current studies have explored its effect on auxiliary phytoextraction at a laboratory scale; however, the method is still rarely reported at the field scale. Therefore, this study used two ecotypes of Pennisetum sinese in a field experiment for 3 years in Jiaoxi Township, Liuyang City, Hunan Province, China, to understand the effect of PASP on the phytoremediation of Cd-contaminated soil and soil quality through long-term field studies. Moreover, because the soil microbial community responds well to the phytoremediation effect of heavy metal (including Cd)-contaminated soil, the changes in rhizosphere soil microbial community diversity and composition were analyzed. After 2 years of PASP-enhanced phytoremediation, the PASP application increased the total Cd reduction in soil by 237 % and 255 %, and the soil DTPA-extractable Cd content decreased to 0.092 and 0.087 mg kg−1. When the application of PASP ceased in the third year, the two ecotypes of P. sinese obtained after harvest could achieve feed safety. Our study showed that the application of PASP could significantly increase the Cd extraction capacity and shoot biomass of P. sinese, and maintain soil health by optimizing the composition and structure of rhizosphere bacterial communities. The rhizosphere bacterial community structure was improved and dominated by Acidobacteriota, Proteobacteria, and Chloroflexi at the phylum level, and the increased abundance of Acetobacter, Enterobacter, Pseudomonas, and Stenotrophomonas at the genus level may promote heavy metal detoxification in soil, plant growth, and phytoremediation. Long-term field monitoring demonstrated that the low-cost and eco-friendly features of PASP made it a good candidate for enhancing phytoextraction efficiency and regulating soil microbial communities for remediation.

A 24-epibrassinolide treatment and intercropping willow with alfalfa increase the efficiency of the phytoremediation of cadmium-contaminated soil

Cadmium contamination in agricultural soils threatens food security and human health, and that has caused widespread concern worldwide. Willow and alfalfa are widely used for the phytoremediation of cadmium (Cd)-contaminated soil, and willow NJU513 is the promising plant for remediating Cd-contaminated soil. In order to discuss the effect of intercropping willow NJU513 with alfalfa on the phytoremediation of Cd-contaminated soil, a pot-culture experiment was conducted in the greenhouse. The result showed that the phytoremediation of Cd-contaminated soil was enhanced by this intercropping because of the 25.90 % increase in the available Cd content. In order to increase the phytoremediation efficiency of Cd in the intercropping treatment, a 24-epibrassinolide (Brs) treatment was designed in the current study. The results showed that the phytoremediation of Cd-contaminated soil by willow and alfalfa improved following a Brs treatment because of the 16.32–74.15 % and 16.91–44.48 % increases in the plant biomass and available Cd content, respectively. Additionally, the extracted Cd by plants in the intercropping treatments with and without Brs was 0.56 and 0.31 mg pot−1, respectively. Transcriptome analyses of willow leaves revealed that Brs up-regulated the expression of genes related to calcium channel activity, calcium and zinc transmembrane transport, photosynthesis, catalase/antioxidant activity, glutathione metabolic processes and detoxification, phagosomes, and vacuoles, and that these upregulated genes promoted plant remediation efficiency and resistance to Cd stress. Brs promoted the phosphate ion transporter activity in willow leaves, which may have enhanced the solubilization of insoluble phosphate minerals by bacterial species (e.g., Vicinamibacterales, Bacillus, and Gaiella) to release Cd, ultimately leading to increased phytoremediation efficiency. In addition, plants with and without Brs treatments induced the bacteria-mediated transformation of available Cd to stable Cd. The study findings may be useful for improving the phytoremediation of Cd-contaminated paddy soil.

Characteristics of Enzyme Activities during Phytoremediation of Cd-Contaminated Soil

In order to study the effects of exogenous Cd on the soil enzyme activities of three herbs, a pot experiment was conducted to study the changes of soil urease, protease, catalase and phosphatase activities in different growth periods of Solanum nigrum L., Phytolacca acinose Roxb., and Bidens pilosa L. under different concentrations of Cd stress. The results showed that the content and proportion of each form of Cd were different in different periods. Compared with the control, the activities of urease, protease and catalase in the soil of three herbs decreased under 5 mg/kg and 10 mg/kg Cd stress, while the phosphatase activities increased first and then decreased. The activities of urease, protease, catalase and phosphatase were 4.24–6.84, 2.17–5.83, 2.09–2.79 and 34.57–37.25 mg/g, respectively, and the recovery degrees were 50.81–66.41%, 32.10–90.54%, 46.97–69.28% and 54.78–56.69%, respectively. After 60 days of remediation, the activities of urease, protease, catalase and phosphatase were 6.05–8.55, 2.83–9.89, 3.32–4.48 and 37.62–41.15 mg/g, respectively, and the recovery degrees were 70.19–84.57%, 41.86–161.34%, 72.35–140.44% and 58.38–63.20%, respectively. Soil enzyme activities were affected by Cd solution stress, which could be improved to a certain extent by plant self–healing, and different grass species recovered to varying degrees under various Cd solution stresses. Different soil enzymes displayed different responses to Cd stress, the inhibition of urease and phosphatase activities was temporary, and the effect of Cd concentration on soil phosphatase activity was close, and it could stimulate the activities of soil protease and catalase, and the higher the concentration of Cd solution, the greater the degree of stimulation. Principal component analysis shows that, after 60 days of repair, the best repair effect plants were Solanum nigrum L. under 5 mg/kg Cd stress and Phytolacca acinosa Roxb. under 10 mg/kg Cd stress.

Promotion effect of graphene on phytoremediation of Cd-contaminated soil.

Echinacea purpurea (L.) Moench was selected as a remediation plant in this study, and different concentrations of graphene oxide (GO) were added to Cd-contaminated soil. Through pot experiments, the effect of E. purpurea on Cd-contaminated soil was determined at 60 days, 120 days, and 150 days. A preliminary study on the remediation mechanism of GO was explored through changes in the forms of Cd in the rhizosphere soil, soil pH, and soil functional groups. Results showed that the optimal concentration of GO was 0.4 g/kg, and under the condition, the accumulation of Cd in the roots of E. purpurea was as high as 113.69 ± 23.86 mg/kg, and the maximum EF reached 5.87 ± 1.34. Compared with those of the control group, accumulated Cd concentration and EF in the roots increased by 60.34% and 2.32, respectively. Correlation analysis showed that the absorption and accumulation of Cd was negatively correlated with the exchangeable Cd content at 120 days, and the exchangeable Cd was negatively correlated with the relative content of functional groups in the soil with 0.4 g/kg GO (E2). The artificial application of GO to the soil can be used as an effective way to improve the effect of E. purpurea in the remediation of Cd soil pollution, and it has great application potential in the stabilization of plants and vegetations and restoration of high-concentration Cd-contaminated soil.

Effect of dissolved organic matter on the phytoremediation of Cd-contaminated soil by cotton

Dissolved organic matter (DOM) assists in the phytoremediation of heavy-metal-contaminated soils, but the effect of synergistic remediation of DOM on plants is unclear. This study investigated the effect of two DOM sources (cotton straw (CM) DOM and farmyard manure (FM) DOM) on cadmium (Cd) accumulation in Cd-contaminated soil by cotton and evaluated the phytoremediation effect of DOM. The results showed that adding DOM reduced the available nitrogen and increased organic matter, available phosphorus and available potassium. Applying DOM increased the proportions of Cd acid soluble fractions and reduced the proportions of Cd residual fractions by 1–7%. DOM application increased root length, root surface area and root volume compared to the control and had a promoting or inhibiting effect on cotton biomass, depending on the soil Cd concentration. Furthermore, applying DOM improved the Cd content and bioconcentration factor of cotton. The lower the molecular weight, hydrophilic components and aromaticity of DOM, the more conducive to Cd accumulation is in cotton. The correlation and random forest analyses also showed that CM showed high remediation potential. According to our study, DOM can improve the phytoremediation efficiency of cotton, especially in low-concentration contaminated soils. This study provides a basis for applying DOM in the phytoremediation of Cd-contaminated soils.

Comparative analysis of sRNAs, degradome and transcriptomics in sweet sorghum reveals the regulatory roles of miRNAs in Cd accumulation and tolerance.

Key miRNAs including sbi-miR169p/q, sbi-miR171g/j, sbi-miR172a/c/d, sbi-miR172e, sbi-miR319a/b, sbi-miR396a/b, miR408, sbi-miR5384, sbi-miR5565e and nov_23 were identified to function in the regulation of Cd accumulation and tolerance. As an energy plant, sweet sorghum shows great potential in the phytoremediation of Cd-contaminated soils. However, few studies have focused on the regulatory roles of miRNAs and their targets under Cd stress. In this study, comparative analysis of sRNAs, degradome and transcriptomics was conducted in high-Cd accumulation (H18) and low-Cd accumulation (L69) genotypes of sweet sorghum. A total of 38 conserved and 23 novel miRNAs with differential expressions were identified under Cd stress or between H18 and L69, and 114 target genes of 41 miRNAs were validated. Furthermore, 25 miRNA-mRNA pairs exhibited negatively correlated expression profiles and sbi-miR172e together with its target might participate in the distinct Cd tolerance between H18 and L69 as well as sbi-miR172a/c/d. Additionally, two groups of them: miR169p/q-nov_23 and miR408 were focused through the co-expression analysis, which might be involved in Cd uptake and tolerance by regulating their targets associated with transmembrane transportation, cytoskeleton activity, cell wall construction and ROS (reactive oxygen species) homeostasis. Further experiments exhibited that cell wall components of H18 and L69 were different when exposed to cadmium, which might be regulated by miR169p/q, miR171g/j, miR319a/b, miR396a/b, miR5384 and miR5565e through their targets. Through this study, we aim to reveal the potential miRNAs involved in sweet sorghum in response to Cd stress and provide references for developing high-Cd accumulation or high Cd-resistant germplasm of sweet sorghum that can be used in phytoremediation.

Effect of cornstalk biochar on phytoremediation of Cd-contaminated soil by Beta vulgaris var. cicla L

Cadmium (Cd) contamination is the most common and extensive heavy metal pollution in the farmland of China. Phytoremediation is considered as a promising measure for Cd-contaminated soil remediation, but the remediation efficiency still needs to be enhanced. Biochar as an effective amendment medium is widely manufactured and studied for the soil remediation of heavy metals. In this study, a greenhouse pot trial was conducted to investigate the effects of cornstalk biochar on Cd accumulation of Beta vulgaris var. cicla L. (Beta vulgaris) in Cd contaminated soil. The Cd availability, speciation and nutrients in soil, biomass and Cd chemical forms in the Beta vulgaris root were studied to explore the mechanism that how the cornstalk biochar promoted Cd accumulation in Beta vulgaris. Biochar amendment reduced the DTPA-extractable Cd concentration and stimulated the growth of root. Compared to the Beta vulgaris without biochar treatment, the results of 5% biochar amendment showed that the root dry weight of Beta vulgaris increased to 267%, Cd accumulation in Beta vulgaris increased to 206% and the Cd concentration in leaves and roots increased by 36% and 52%, respectively. Additionally, after 5% biochar was applied to soil, the total content of organic matter-bound Cd and residual Cd increased by 38%, while the content of Fe–Mn oxides-bound Cd decreased by 40%. Meanwhile, Cd may mainly bind to the root cell wall and the ratio of NaCl-extracted Cd to HAc-extracted Cd increased to 166% with 5% biochar amendment. According to our study, Cd in soil can be removed by Beta vulgaris and phytoremediation efficiency can be improved with biochar amendment. The combination of phytoremediation and biochar amendment is a promising strategy for the Cd-contaminated soil remediation.

Responses of microbial communities and metabolic activities in the rhizosphere during phytoremediation of Cd-contaminated soil

Phytoremediation is an effective way to repair heavy metal contaminated soil and rhizosphere microorganisms play an important role in plant regulation. Nevertheless, little information is known about the variation of microbial metabolic activities and community structure in rhizosphere during phytoremediation. In this study, the rhizosphere soil microbial metabolic activities and community structure of Trifolium repensL. during Cd-contaminated soil phytoremediation, were analyzed by Biolog EcoPlate™ and high-throughput sequencing. The uptake in the roots of Trifolium repensL. grown in 5.68 and 24.23 mg/kg Cd contaminated soil was 33.51 and 84.69 mg/kg respectively, causing the acid-soluble Cd fractions decreased 7.3% and 5.4%. Phytoremediation significantly influenced microbial community and Trifolium repensL. planting significantly increased the rhizosphere microbial population, diversity, the relative abundance of plant growth promoting bacteria (Kaistobacter and Flavisolibacter), and the utilization of difficultly metabolized compounds. The correlation analysis among substrate utilization and microbial communities revealed that the relative abundance increased microorganisms possessed stronger carbon utilization capacity, which was beneficial to regulate the stability of plant-microbial system. Collectively, the results of this study provide fundamental insights into the microbial metabolic activities and community structure during heavy metal contaminated soil phytoremediation, which may aid in the bioregulation of phytoremediation.

Evaluation of hyperaccumulation potentials to cadmium (Cd) in six ornamental species (compositae)

Phytoremediation is considered as a promising soil remediation technique. In the present study, the growth responses, cadmium (Cd) accumulation and uptake capability of six popular compositae species, namely, Taraxacum mongolicum Hand.-Mazz., Tagetes erecta L., Tagetes patula L., Zinnia elegans Jacq., Centaurea cyanus L. and Gerbera jamesonii Bolus under Cd stress were investigated. Among the six compositae species, the growth of T. erecta L. and T. patula L. improved under 10 mg kg−1 Cd exposure in term of the total biomass and height increased along with the increased Cd concentration in soil, and the growth of the two plants had no significant differences at the high Cd concentration (100 mg kg−1), which indicated that they have good tolerance to Cd toxicity. At the same time, the two plants have higher biomass than four other plants. Furthermore, they can accumulate Cd above 100 μg g−1 dry tissue, which is the threshold value of a Cd-hyperaccumulator, and have higher Cd uptake ability, translocation factor (TF) and bioconcentration factor (BCF) values. According to these traits, it was shown that T. erecta L. and T. patula L. had strong tolerance and accumulation capability to Cd, therefore they can become potential hyperaccumulators in phytoremediation of Cd-contaminated soils.

Phytoremediation of Cd-contaminated Soils by Datura stramonium L.

The contamination of soils with heavy metals is a grave environmental problem. Among the heavy metals, Cadmium (Cd) is regarded as one of the most toxic trace elements and environmental pollutants. The aim of this research is investigation of phytoremediation ability of Datura stramonium L. for Cadmium (Cd) and also, the enhancing effect of EDTA (ethylenediamine tetraacetic acid), indole-3-acetic acid (IAA) and kinetin (KN) on the uptake of Cd. Four levels of cadmium (0, 75, 150 and 225 mg kg-1) and different enhancers including EDTA (0, 3, 6, 12 mmol kg-1), IAA, and kinetin (0, 1, 10, 100 mg L-1) were used as the experimental groups. Analysis of Cd in soil and plant materials was carried out using an atomic absorption spectrophotometer. The bioconcentration factor, translocation factor, protein content and enzymatic activity were also measured. The results showed that D. stramonium was a Cd accumulator plant with the TF of 1.12. Accumulation ability was increased with the application of the chelating agent and plant hormones; EDTA was found to be more effective in the enhancement of phytoremediation than others. Application of EDTA and hormones reduced the negative effects of Cd by means of increasing the total protein content and peroxidase activity. The total protein content in the leaves of D. stramonium, which had been treated with Cd/EDTA, Cd/KN and Cd/IAA, was increased 2.24, 3.59 and 3.41 times more than that in the Cd treated group, respectively. The highest peroxidase (POX) activity was determined in the group treated by 150-Cd +100-IAA (0.060 unit/mg protein). It could be concluded that D. stramonium is a Cd-accumultor plant with phytoremediation potency. This study also demonstrated that EDTA and plant hormones (IAA and Kinetin) could be regarded as good strategies for increasing the phytoremediation ability of D. stramonium.

Effect of hemoglobin on the growth and Cd accumulation of pea plants (Pisum sativum L.)

This study was set to investigate the effect of the presence of hemoglobin (Hb) in cadmium (Cd)-contaminated soil on phytotoxicity and Cd accumulation. The effect of Hb on the Cd accumulation by Pisum sativum L. (pea) and seed germination and growth was studied using pot tests with the artificially Cd-contaminated soil. The results show that the externally applied Hb to Cd-contaminated soil samples did not promote Cd accumulation by P. sativum. However, the Fe accumulation was greater in the presence of Hb. The seed germination was not affected, but the adverse effects on the plant growth increased with increasing Hb/Cd molar ratio from 0 to 0.015. This can be attributed to toxic effects of the Fe added with the Hb application. The results suggest that the presence of Hb may have harmful effects on pea plants used in phytoremediation of Cd-contaminated soil due to toxic effects imposed by Fe.

Effects of cadmium stress on seedlings of various rangeland plant species (Avena fatua L., Lathyrus sativus L., and Lolium temulentum L.): Growth, physiological traits, and cadmium accumulation

ABSTRACTA pot culture experiment was performed to study the effect of cadmium stress (Cd stress) on seedling growth, physiological traits, and remediation potency of Avena fatua, Lathyrus sativus, and Lolium temulentum. The seedlings of these native rangeland plant species were treated with 0, 2, 4, and 6 mM cadmium nitrate concentrations. Based on the results of analysis of variance (p < 0.05), the shoot height, shoot dry weight, root length, root dry weight, root: shoot ratio, total chlorophyll content, soluble sugars, and protein contentof A. fatua, L. sativus, and L. temulentum significantly decreased with increased cadmium concentrations. Generally, translocation factor (TF) and tolerance index (TI) decreased significantly as the concentration of cadmium increased. The maximum TF and TI of studied plants in various concentrations of Cd were observed in L. temulentum followed by L. sativus and A. fatua. The root concentration factor (RCF) values of all studied plants were higher than 1 under different cadmium concentrations. Our results indicate that Lolium temulentum could be labeled as an accumulator of Cd asthe values of TF and RCF are greater than 1. A. fatua and L. sativus showed a potential to be used in the phytoremediation of Cd-contaminated soils.

An indoleacetic acid-producing Ochrobactrum sp. MGJ11 counteracts cadmium effect on soybean by promoting plant growth.

To analyse whether some indoleacetic acid (IAA)-secreting plant growth-promoting bacteria can alleviate cadmium (Cd) stress, the role of an Ochrobactrum sp. MGJ11 from rhizosphere of soybean in promoting plant growth, and to evaluate the counteracting Cd effects on soybean. Ochrobactrum sp. MGJ11 produced 121·2mgl-1 of IAA. MGJ11 increased soybean root length, shoot length and biomass by 30·1, 30·8 and 13·4% respectively. In liquid medium, no IAA production was detected in Cd concentration of 100mgl-1 . In soil with 20-80mgkg-1 Cd, MGJ11 promoted soybean root elongation (29·4-161·4%) and increased the shoot length (up to 52·7%) and biomass (up to 87·2%). After growing for 38days, Cd concentrations in the roots of inoculated soybean were lower than in those of noninoculated plants. Only a little Cd (2·6-16·9μgg-1 ) was translocated from the root to shoot. Ochrobactrum sp. MGJ11 secretes IAA and shows tolerance against Cd. MGJ11 inoculation improves the root length, shoot length and biomass of soybean in both vermiculite and Cd vermiculite, and decreases Cd concentration of soybean root. The characteristics of MGJ11 suggest that it could be used for promoting soybean growth and lowering bioavailability of soil Cd for soybean root. In this study, we isolated a plant growth-promoting Ochrobactrum with the activity of mitigating Cd toxicity to plant roots. The Ochrobactrum can be considered as a potential bioaugmentation agent that promotes plant growth, especially in some agricultural systems, or that helps in the phytoremediation of Cd-contaminated soil.
