Comparative study on plant growth-promoting bacterial inoculation by irrigation and spraying for promoting Bidens pilosa L. phytoremediation of cadmium-contaminated soil

A field study was conducted to compare FM-1 inoculation by irrigation and spraying for promoting Bidens pilosa L. phytoremediation of cadmium (Cd)-contaminated soil. Cascading relationships between bacterial inoculation by irrigation and spraying and soil properties, plant growth-promoting traits, plant biomass and Cd concentrations in Bidens pilosa L. were explored based on the partial least squares path model (PLS-PM). The results indicated that inoculation with FM-1 not only improved the rhizosphere soil environment of B. pilosa L. but also increased the Cd extracted from the soil. Moreover, Fe and P in leaves play vital roles in promoting plant growth when FM-1 is inoculated by irrigation, while Fe in leaves and stems plays a vital role in promoting plant growth when FM-1 is inoculated by spraying. In addition, FM-1 inoculation decreased the soil pH by affecting soil dehydrogenase and oxalic acid in cases with irrigation and Fe in roots in cases with spraying. Thus, the soil bioavailable Cd content increased and promoted Cd uptake by Bidens pilosa L. To address Cd-induced oxidative stress, Fe in leaves helped to convert GSH into PCs, which played a vital role in ROS scavenging when FM-1 was inoculated by irrigation. The soil urease content effectively increased the POD and APX activities in the leaves of Bidens pilosa L., which helped alleviate Cd-induced oxidative stress when FM-1 was inoculated by spraying. This study compares and illustrates the potential mechanism by which FM-1 inoculation can improve the phytoremediation of Cd-contaminated soil by Bidens pilosa L., suggesting that FM-1 inoculation by irrigation and spraying is useful in the phytoremediation of Cd-contaminated sites.

Phytoremediation of Cadmium-Contaminated Soils: A Review of New Cadmium Hyperaccumulators and Factors Affecting their Efficiency

A promising solution for the remediation of cadmium (Cd) contaminated soil involves the use of Cd hyperaccumulators to reduce levels of soil Cd. The suitability of various plant species to act as cadmium (Cd) hyperaccumulators was reviewed by considering bioconcentration factor (BCF), translocation factor (TF) and Cd concentration in shoots relative to soil parameters such as pH and organic matter content, and planting and growing parameters. High BCF and TF values (max 81 and 13.7 respectively) were observed in soils with low soil Cd concentration, soil organic matter (SOM) content (< 2%) and low soil pH, with biannual harvesting possible for some species. Certain species such as Youngia erythrocarpa and Gnaphalium affine were efficient as hyperaccumulators in soil with > 40% SOM content.

Positive Effects and Optimal Ranges of Tea Saponins on Phytoremediation of Cadmium-Contaminated Soil

Confirming positive effects and offering optimal ranges of tea saponins on improving the efficiency of phytoremediation on cadmium is a prerequisite for applying tea saponins in field remediation. Existing studies qualitatively tested the feasibility of tea saponins on promoting the absorption of cadmium by hyperaccumulators in pots experiments, while this study investigated the effects of tea saponins on increasing the proportion of cadmium available fraction in contaminated soil quantitatively and confirmed tea saponins promoted the absorption by Portulaca oleracea in cadmium-contaminated water by independent soil experiments and hydroponic experiments. The results showed that for acquiring a higher proportion of cadmium available fraction, the concentration of tea saponins was negatively correlated with the concentration of cadmium contained in the soil, and the optimal treatment time of tea saponins was between 3–9 days depending on the cadmium concentration in contaminated soil. Using tea saponins could enhance the absorption of cadmium by Portulaca oleracea in a relatively short time to decrease the concentration of cadmium left in the contaminated water. The above findings help to deepen the understanding of tea saponins’ effects and use ranges on phytoremediation of cadmium both in soil and water and conduce studies on phytoremediation of other heavy-metal-contaminated soil and water with the help of tea saponins.

Phytoremediation of cadmium-contaminated soils by Solanum nigrum L. enhanced with biodegradable chelating agents.

The application of biodegradable chelating agents to enhance phytoremediation is a low-cost and promising method to improve the remediation efficiency of heavy metal-contaminated soil. The effects of N, N-bis glutamic acid (GLDA) on the growth and heavy metal absorption of Solanum nigrum were studied by pot experiment. The addition of chelate on the 20th day after sowing can improve the bioavailability of cadmium (Cd) in the soil. The results showed that the addition of chelating agents effectively improved the migration rate of the target heavy metal Cd in the soil, and significantly increased the accumulation of heavy metal in the roots, stems, and leaves of plants. The results showed that compared with the control group, the chelating agent could increase the extraction rate of total Cd by 28.65-68.74%. The application of GLDA significantly increased the accumulation of Cd (20mgkg-1 and 40mgkg-1), reaching 24.28-40.30 and 25.71-33.16μg of pot-1 DW, respectively. At the same time, GLDA increased Cd stress by decreasing plant biomass, inhibiting photosynthetic pigment synthesis and increasing MDA levels. These results indicated that GLDA could improve the absorption of Cd by S. nigrum, which provided a new idea for its practical application in the remediation of Cd-contaminated soil.

Phytoremediation of cadmium-contaminated soil by Bidens pilosa L.: impact of pine needle biochar amendment.

The objective of the present study was to evaluate the feasibility of pine needle biochar as a soil amendment to promote the growth of Bidens pilosa L. and enhance its ability to phytoextract the cadmium from soil. Pot experiments (50 d) were designed as control experiment (C); metal treatment (MT), 20 mg Cd kg-1; biochar treatment (BT100 or BT200), 100 or 200 mg kg-1; and metal-biochar treatment (MBT100 or MBT200), 20 mg Cd kg-1 and 100 or 200 mg biochar kg-1. The Cd (20 mg kg-1) or biochar treatment (100 mg kg-1) increased the dry weight and root length of B. pilosa. The biochar amendment enhanced the metal concentration in root and shoot of the plant. The plant could accumulate 39.47±5.44 mg Cd kg-1 in shoots (MT), which increased to 45.96±17.3 mg Cd kg-1 and 55.01±5.65 mg Cd kg-1 under biochar treatment sets MBT100 and MBT200, respectively. The Cd uptake by B. pilosa in MT, MBT100, and MBT200 treatments were 67.81 μg/plant, 78.58 μg/plant, and 76.13 μg/plant, respectively. The biochar amendments increased the proline concentrations while decreased the chlorophyll content in leaves indicating the stress on the plant. Overall, the result indicates that soil amended with pine needle biochar at 100 mg kg-1 increased the phytoremediation ability of B. pilosa.

Integration of earthworms and arbuscular mycorrhizal fungi into phytoremediation of cadmium-contaminated soil by Solanum nigrum L

Arbuscular mycorrhizal fungi (AMF) and earthworms independently enhance plant growth, heavy metal (HM) tolerance, and HM uptake, thus they are potential key factors in phytoremediation. However, few studies have investigated their interactions in HM phytoextraction by hyperaccumulators. This study highlights the independent and interactive effects of earthworms and AMF on Solanum nigrum. Plants inoculated with either AMF or earthworms exhibited ameliorated growth via enhancement of productivity, metal tolerance, and phosphorus (P) acquisition. Co-inoculation with both had more pronounced effects on plant biomass and P acquisition in shoots, but not in roots, and in Cd-polluted soils it significantly promoted (P < 0.05) shoot biomass (20.7–134.6 %) and P content (20.4–112.0 %). AMF and earthworms increased Cd accumulation in plant tissues, but only AMF affected Cd partitioning between shoots and roots. Although AMF decreased root-to-shoot translocation of Cd at high Cd levels, this was counterbalanced by earthworms. Both AMF and its combination with earthworms enhanced Cd phytoavailability by altering Cd chemical fractions and decreasing pH. Co-inoculation increased Cd removal amounts up to 149.3 % in 120 mg kg−1 Cd-spiked soils. Interactions between the two organisms were synergistic in Cd phytoextraction. Thus, earthworm-AMF-plant symbiosis potentially plays an essential role in phytoremediation of HM-polluted soils.

Phytoremediation of cadmium-contaminated soil by Sorghum bicolor and the variation of microbial community

In this paper, the growth of S. bicolor in Cd-polluted sandy clay loam soil in north China, Cd accumulation in plant and the corresponding soil microbial community were characterized when the plant matured (140 d of growth). Cadmium promoted the growth of mature S. bicolor with higher height and heavier dry mass, especially at the spiked level of 1 mg kg−1 soil (P < 0.05). The higher microbial diversity was found under Cd stress at the spiked level of 15 mg kg−1, which basically corresponded with its influence on the plant growth. High-throughput sequencing data demonstrated that the predominant bacterial phyla include Proteobacteria (35.99% for Cd-polluted soil and 35.22% for the control soil), Chloroflexi (21.33% and 20.58%), Actinobacteria (12.00% and 12.89%), Acidobacteria (7.47% and 11.14%), Bacteroidetes (7.37% and 6.96%), Gemmatimonadetes (5.60% and 6.65%), Firmicutes (2.82% and 1.86%), Planctomycetes (2.47% and 0.95%), Saccharibacteria (1.26% and 1.11%). The predominant fungal phyla was Ascomycota, with the relative abundance of 89.96% for the control soil and 86.2% for the Cd-polluted soil. S. bicolor could grow well in sandy clay loam soil in northern China at low Cd lvel, but it could not accumulate cadmium at higher cadmium level. S. bicolor could be used for phytoextraction of cadmium from the lightly Cd-polluted soil.

Phytoremediation of cadmium-contaminated soil through multipurpose tree species

Phytoremediation of heavy metal-contaminated sites using trees may be preferred over crops due to capacity of trees to accumulate heavy metals in the wood which is biologically non-hazardous. An investigation was undertaken to assess the cadmium (Cd) phytoextraction potential of four multipurpose tree species viz. eucalyptus (Eucalyptus tereticornis Sm.), subabul [Leucaena leucocephala (Lam.) de Wit], dhrek (Melia azedarach L.) and shisham (Dalbergia sissoo Roxb.) at six rates of Cd applied in solution form through CdCl2·H2O (0, 10, 20, 40, 80 and 120 mg Cd kg−1 soil) and harvested after 18 months of growth in an alkaline loamy sand soil in earthen pots. Dry matter (DM) of leaves, stems and roots, Cd content of these constituents and that of soil before planting and after harvesting the tree species were determined. Subabul had the highest and eucalyptus the lowest total DM production at all levels of Cd. Total DM did not decrease significantly up to application of 40 mg Cd kg−1 soil (P > 0.05). The highest Cd level resulted in 49, 41, 26 and 43% reduction in total DM of eucalyptus, subabul, dhrek and shisham, respectively over control. Cadmium concentration was higher in leaves whereas uptake was higher in stems than the other components. Subabul had significantly highest uptake (0.05–71.4 mg pot−1) of Cd at various levels due its higher biomass whereas shisham had the lowest (0.05–28.2 mg pot−1) (P ≤ 0.05). Available Cd in the soil after harvesting of plants was the lowest in subabul and the highest in shisham pots. The upper critical level of available Cd in the soil at 20% reduction in DM was the highest for dhrek (85 mg kg−1) and the lowest for eucalyptus (46 mg kg−1). Therefore, dhrek was more tolerant to Cd but subabul had the greatest potential for phytoextraction of Cd from the soil.

Integrated metagenomics and molecular ecological network analysis of bacterial community composition during the phytoremediation of cadmium-contaminated soils by bioenergy crops

Two energy crops (maize and soybean) were used in the remediation of cadmium-contaminated soils. These crops were used because they are fast growing, have a large biomass and are good sources for bioenergy production. The total accumulation of cadmium in maize and soybean plants was 393.01 and 263.24μg pot−1, respectively. The rhizosphere bacterial community composition was studied by MiSeq sequencing. Phylogenetic analysis was performed using 16S rRNA gene sequences. The rhizosphere bacteria were divided into 33 major phylogenetic groups according to phyla. The dominant phylogenetic groups included Proteobacteria, Acidobacteria, Actinobacteria, Gemmatimonadetes, and Bacteroidetes. Based on principal component analysis (PCA) and unweighted pair group with arithmetic mean (UPGMA) analysis, we found that the bacterial community was influenced by cadmium addition and bioenergy cropping. Three molecular ecological networks were constructed for the unplanted, soybean- and maize-planted bacterial communities grown in 50mgkg−1 cadmium-contaminated soils. The results indicated that bioenergy cropping increased the complexity of the bacterial community network as evidenced by a higher total number of nodes, the average geodesic distance (GD), the modularity and a shorter geodesic distance. Proteobacteria and Acidobacteria were the keystone bacteria connecting different co-expressed operational taxonomic units (OTUs). The results showed that bioenergy cropping altered the topological roles of individual OTUs and keystone populations. This is the first study to reveal the effects of bioenergy cropping on microbial interactions in the phytoremediation of cadmium-contaminated soils by network reconstruction. This method can greatly enhance our understanding of the mechanisms of plant-microbe-metal interactions in metal-polluted ecosystems.

Effects of plant growth-promoting bacteria on the phytoremediation of cadmium-contaminated soil by sunflower

ABSTRACTThis paper describes the effect of plant growth-promoting bacteria on sunflower growth and its phytoremediation efficiency under Cd-contaminated soils. Four levels of bacteria inoculation (non-inoculation, inoculation by Bacillus safensis, Kocuria rosea and co-inoculation by Bacillus safensis+Kocuria rosea) and four Cd concentrations (0, 50, 100 and 150 mg Cd per kg soil) were arranged as factorial experiment based on a completely randomized design (CRD). Results showed that Cd significantly decreased growth by decreasing the shoot and root length and biomass (p < 0.01). In addition, Cd dramatically decreases photosynthetic pigments, Fe transport to shoot and Zn uptake (p < 0.01). Bacterial inoculation increased Fe and Zn uptake by plants, Cd concentration in the aboveground part of plants and Cd uptake by the enhancement of Cd concentration in plant tissue and biomass production. Results showed that the highest shoot Cd uptake was ‎observed in ‎inoculated plants by Bacillus safensis at Cd100 (20.35 mg pot−1). However, in average of Cd treatments, the performance of co-inoculation in Cd uptake (13.04 mg pot−1) was better than singular inoculation (10.68 and 12.58 mg pot−1 for Bacillus safensis and Kocuria rosea, respectively). Results revealed that bacterial inoculation increased the Cd uptake performance in shoot and total biomass by 30% and 25%, respectively.

Characterization of efficient plant-growth-promoting bacteria isolated from Sulla coronaria resistant to cadmium and to other heavy metals

The inoculation of plants with plant-growth-promoting rhizobacteria has become a priority in the phytoremediation of heavy-metal-contaminated soils. A total of 82 bacteria were isolated from Sulla coronaria root nodules cultivated on four soil samples differently contaminated by heavy metals. The phenotypic characterization of these isolates demonstrated an increased tolerance to cadmium reaching 4.1mM, and to other metals, including Zn, Cu and Ni. Polymerase Chain Reaction/Restriction Fragment Length Polymorphism (PCR/RFLP) analysis showed a large diversity represented by genera related to Agrobacterium sp., R. leguminosarum, Sinorhizobium sp., Pseudomonas sp., and Rhizobium sp. Their symbiotic effectiveness was evaluated by nodulation tests. Taking into consideration efficiency and cadmium tolerance, four isolates were chosen; their 16SrRNA gene sequence showed that they belonged to Pseudomonas sp. and the Rhizobium sullae. The selected consortium of soil bacteria had the ability to produce plant-growth-promoting substances such as indole acetic acid and siderophore. The intracellular Cd accumulation was enhanced by increasing the time of incubation of the four soil bacteria cultivated in a medium supplemented with 0.1mM Cd. The existence of a cadmium-resistant gene was confirmed by PCR. These results suggested that Sulla coronaria in symbiosis with the consortium of plant-growth-promoting rhizobacteria (PGPR) could be useful in the phytoremediation of cadmium-contaminated soils.

Phytoremediation of Cadmium-Contaminated Soils by Young Douglas Fir Trees: Effects of Cadmium Exposure on Cell Wall Composition

Douglas fir trees grown on an artificially Cd-contaminated soil, can tolerate this trace element (up to 68 mg/kg in soil) during several months. Most of the absorbed Cd is retained in roots (25 mg/kg DM), but transfer to aerial part is also effective. Showing the highest content, up to 6 mg/kg DM, among all the aboveground parts, barks seem to be a preferred storage compartment. However, the transfer factor is quite low, about 0.3. Another objective of this study was to compare the cell wall components of trees exposed to increasing Cd amounts in soil. A decrease in lignin and an increase in pectin contents were observed in response to increasing soil cadmium concentration. A concurrent reduction in methyl-esterification of pectin suggests than the structure of this major binding site could therefore be modified as a reaction to cadmium contamination. Future prospects will focus on the modulation of pectin composition in response to Cd exposure.

Phytoremediation of Cadmium-Contaminated Soils by Rorippa globosa Using Two-Phase Planting (5 pp)

Phytoextraction of contaminated soils by heavy metals can provide a great promise of commercial development. Although there are more than 400 species of hyperaccumulators found in the world, phytoremediation technology is rarely applied in field practice for remedying contaminated soils, partially due to low biomass and long growth duration for most of discovered hyperaccumulating plants. In order to enhance the metal-removing efficiency in a year, the two-phase planting countermeasure of phytoextraction by harvesting anthesis biomass was investigated on the basis of the newly found Cd-hyperaccumulator Rorippa globosa (Turcz.) Thell. with 107.0 and 150.1 mg/kg of the Cd accumulation in stems and leaves, respectively, when soil Cd added was concentrated to 25.0 mg/kg. The field pot-culture experiment was used to observe the distribution property of R. globosa aboveground biomass and to examine characteristics of accumulating Cd by the plant at different growth stages. The concentration of Cd in plants and soils was determined using atomic absorption spectrophotometry (AAS). The results indicated that the total dry stem and leaf biomass of R. globosa harvested at the flowering phase was up to 92.3% of that at its full maturity and the concentration of Cd in stems and leaves harvested at the flowering phase was up to 73.8% and 87.7% of that at the mature phase, respectively. The Cd-removing ratio by shoots of R. globosa harvested at the flowering phase was up to 71.4% of that at the mature phase. It was also found, by observing the growth duration of R. globosa, that the frostless period at the experiment site was twice as long as the growth time from the seedling-transplanted phase to the flowering phase of the hyperaccumulator. R. globosa could be transplanted into contaminated soils twice in one year by harvesting the hyperaccumulator at its flowering phase based on climatic conditions of the site and traits of the plant growth. In this sense, the extraction efficiency of Cd in shoots of R. globosa increased 42.8% compared to that of at its single maturity when the plant was transplanted into contaminated soils after it had been harvested at its flowering phase and the plant accumulated Cd from soil at the same extraction ratio at its second flowering phase. Thus, the method of anthesis biomass regulation by the two-phase planting is very significant to increase the Cd-removing efficiency by phytoremediation used in practice over the course of a year. As for some hyperaccumulators that the growth duration from the seedling-transplanted phase to the flowering phase are short and the concentrations of heavy metals accumulated in their shoots at the flowering phase are high, the efficiency of phytoremediation can greatly be improved using the method of the two-phase planting.
