Irradiance driven trophic plasticity in the coral Madracis pharensis from the Eastern Mediterranean

The distribution of symbiotic scleractinian corals is driven, in part, by light availability, as host energy demands are partially met through translocation of photosynthate. Physiological plasticity in response to environmental conditions, such as light, enables the expansion of resilient phenotypes in the face of changing environmental conditions. Here we compared the physiology, morphology, and taxonomy of the host and endosymbionts of individual Madracis pharensis corals exposed to dramatically different light conditions based on colony orientation on the surface of a shipwreck at 30 m depth in the Bay of Haifa, Israel. We found significant differences in symbiont species consortia, photophysiology, and stable isotopes, suggesting that these corals can adjust multiple aspects of host and symbiont physiology in response to light availability. These results highlight the potential of corals to switch to a predominantly heterotrophic diet when light availability and/or symbiont densities are too low to sustain sufficient photosynthesis, which may provide resilience for corals in the face of climate change.

The effects of urban thermal heterogeneity and feather coloration on oxidative stress and metabolism of pigeons (Columba livia)

Urbanization stands out as a significant anthropogenic factor, exerting selective pressures on ecosystems and biotic components. A notable outcome of urbanization is thermal heterogeneity where the emergence of Urban Heat Islands is characterized by elevated air and surface temperatures compared to adjacent rural areas. Investigating the influence of thermal heterogeneity on urban animals could offer insights into how temperature variations can lead to phenotypic shifts. Urban pigeons (Columba livia) serve as an excellent model for studying urban thermal effects, given the melanism variations, which are associated with the pleiotropy of the melanocortin system. To examine the development of physiological plasticity in response to urban thermal variations, we conducted a study on pigeons in Santiago, Chile, during the rainy season. We assessed the influence of habitat on physiological traits related to metabolism and antioxidant capacities, which are theoretically affected by feather coloration. Our findings reveal that variations in melanism significantly impact pigeon physiology, affecting both antioxidant capacities and the mitochondrial activity of red blood cells. It was found that higher urban temperatures, from both the current sampling month and the prior sampling month (from CRU TS dataset), were negatively and strongly associated with lower antioxidant and metabolic activities. This suggests that elevated urban temperatures likely benefit the energetic budgets of pigeon populations and mitigate the negative effects of oxidative metabolism, with differential effects depending on feather colorations.

Rapid Physiological Plasticity in Response to Cold Acclimation for Nonnative Italian Wall Lizards (Podarcis siculus) from New York.

AbstractThermal physiology helps us understand how ectotherms respond to novel environments and how they persist when introduced to new locations. Researchers generally measure thermal physiology traits immediately after animal collection or after a short acclimation period. Because many of these traits are plastic, the conclusions drawn from such research can vary depending on the duration of the acclimation period. In this study, we measured the rate of change and extent to which cold tolerance (critical thermal minimum [CTmin]) of nonnative Italian wall lizards (Podarcis siculus) from Hempstead, New York, changed during a cold acclimation treatment. We also examined how cold acclimation affected heat tolerance (critical thermal maximum [CTmax]), thermal preference (Tpref), evaporative water loss (EWL), resting metabolic rate (RMR), and respiratory exchange ratio (RER). We predicted that CTmin, CTmax, and Tpref would decrease with cold acclimation but that EWL and RMR would increase with cold acclimation. We found that CTmin decreased within 2 wk and that it remained low during the cold acclimation treatment; we suspect that this cold tolerance plasticity reduces risk of exposure to lethal temperatures during winter for lizards that have not yet found suitable refugia. CTmax and Tpref also decreased after cold acclimation, while EWL, RMR, and RER increased after cold acclimation, suggesting trade-offs with cold acclimation in the form of decreased heat tolerance and increased energy demands. Taken together, our findings suggest that cold tolerance plasticity aids the persistence of an established population of invasive lizards. More generally, our findings highlight the importance of accounting for the plasticity of physiological traits when investigating how invasive species respond to novel environments.

Marine primary producers in a darker future: a meta‐analysis of light effects on pelagic and benthic autotrophs

The availability of underwater light, as primary energy source for all aquatic photoautotrophs, is (and will further be) altered by changing precipitation, water turbidity, mixing depth, and terrestrial input of chromophoric dissolved organic matter (CDOM). While experimental manipulations of CDOM input and turbidity are frequent, they often involve multiple interdependent changes (light, nutrients, C‐supply). To create a baseline for the expected effects of light reduction alone, we performed a weighted meta‐analysis on 240 published experiments (from 108 studies yielding 2500 effect sizes) that directly reduced light availability and measured marine autotroph responses. Across all organisms, habitats, and response variables, reduced light led to an average 23% reduction in biomass‐related performance, whereas the effect sizes on physiological performance did not significantly differ from zero. Especially, pigment content increased with reduced light, which indicated a strong physiological plasticity in response to diminished light. This acclimation potential was also indicated by light reduction effects minimized if the experiments lasted longer. Nevertheless, the performance (especially biomass accrual) was reduced the more the less light intensity remained available. Light reduction effects were also more negative at higher temperatures if ambient light conditions were poor. Macrophytes or benthic systems were more negatively affected by light reduction than microalgae or plankton systems, especially in physiological responses were microalgae and plankton showed slightly positive responses. Otherwise, the effect magnitudes remained surprisingly consistent across habitats and aspects of experimental design. Therefore, the strong observed log–linear relationship between remaining light and autotrophic performance can be used as a baseline to predict marine primary production in future light climate.

Diurnal fluctuations in seawater pCO2 amplify the negative effects of ocean acidification on the biotic performance of the calcifying macroalga Halimeda opuntia

Although the adverse effects of increasing atmospheric CO2-induced ocean acidification (OA) on marine calcifying macroalgae have been widely reported, there are limited studies on how daily fluctuations in pCO2 (pH) within shallow ecosystems influence the growth and physiological performance of these calcifiers. Therefore, a 42-day laboratory mimetic experiment to determine how growth, biological performance and related carbon and nitrogen metabolic products of the calcifying macroalga, Halimeda opuntia are generated in response to fluctuating pCO2 under OA conditions (1200 ppmv) was performed. The results of present study showed that the adverse effects of OA were more determined by the adverse influence of elevated acidity (H+) on growth rates, calcification, photosynthesis and the related biotic performance of H. opuntia compared with the positive effects that higher CO2 provided. Moreover, diurnal fluctuations in pCO2 levels [with higher (nearly 8.10) and lower pH (nearly 7.40) values during day and night times, respectively] have amplified these negative influences on H. opuntia. To mitigate elevated pCO2-related stress, higher contents of free amino acids and proline were highly secreted and likely linked to protecting the integrity of algal cellular structures. The above results contribute to increasing our understanding of the biological consequences of pCO2 (pH) variability on calcifying Halimeda species and their physiological plasticity in response to further oceanic pCO2 changes.

Examining the Role of DNA Methylation in Transcriptomic Plasticity of Early Stage Sea Urchins: Developmental and Maternal Effects in a Kelp Forest Herbivore

Gene expression plasticity can confer physiological plasticity in response to the environment. However, whether epigenetic marks contribute to the dynamics of gene expression is still not well described in most marine invertebrates. Here, we explored the extent and molecular basis of intra- and intergenerational plasticity in the purple sea urchin, Strongylocentrotus purpuratus, by examining relationships between changes in DNA methylation, transcription, and embryo spicule length. Adult urchins were conditioned in the lab for four months to treatments that represent upwelling (~1100μatm pCO2, 13 °C) and non-upwelling conditions (~500μatm pCO2, 17 °C). Embryos spawned from conditioned adults were reared in either the same adult treatment or the reciprocal condition. Maternal conditioning resulted in significantly differentially methylated CpG sites and differential gene expression in the larvae, despite no evidence of maternal effects on embryo spicule length. In contrast, conditions experienced during development resulted in significant differences in larval spicule length. Intragenerational plasticity in spicule length was strongly correlated to transcriptomic plasticity, despite low levels of intragenerational plasticity in CpG methylation. We find plasticity in DNA methylation and gene expression in response to different maternal environments and these changes have similarities across broad functional groups of genes; yet exhibit little overlap on a gene-by-gene basis. Our results suggest that different forms of environmentally induced plasticity are observable across different time scales and that DNA methylation dynamics may be uncoupled from fast transcriptional responses to the environment and whole organism traits. Overall, this study illuminates the extent to which environmental differences can induce both intra- and intergenerational phenotypic plasticity in a common kelp forest herbivore.

Intraspecific Diversity in the Cold Stress Response of Transposable Elements in the Diatom Leptocylindrus aporus.

Transposable elements (TEs), activated as a response to unfavorable conditions, have been proposed to contribute to the generation of genetic and phenotypic diversity in diatoms. Here we explore the transcriptome of three warm water strains of the diatom Leptocylindrus aporus, and the possible involvement of TEs in their response to changing temperature conditions. At low temperature (13 °C) several stress response proteins were overexpressed, confirming low temperature to be unfavorable for L. aporus, while TE-related transcripts of the LTR retrotransposon superfamily were the most enriched transcripts. Their expression levels, as well as most of the stress-related proteins, were found to vary significantly among strains, and even within the same strains analysed at different times. The lack of overexpression after many months of culturing suggests a possible role of physiological plasticity in response to growth under controlled laboratory conditions. While further investigation on the possible central role of TEs in the diatom stress response is warranted, the strain-specific responses and possible role of in-culture evolution draw attention to the interplay between the high intraspecific variability and the physiological plasticity of diatoms, which can both contribute to the adaptation of a species to a wide range of conditions in the marine environment.

Regional heterogeneity impacts gene expression in the subarctic zooplankter Neocalanus flemingeri in the northern Gulf of Alaska

Marine pelagic species are being increasingly challenged by environmental change. Their ability to persist will depend on their capacity for physiological acclimatization. Little is known about limits of physiological plasticity in key species at the base of the food web. Here we investigate the capacity for acclimatization in the copepod Neocalanus flemingeri, which inhabits the Gulf of Alaska, a heterogeneous and highly seasonal environment. RNA-Seq analysis of field-collected pre-adults identified large regional differences in expression of genes involved in metabolic and developmental processes and response to stressors. We found that lipid synthesis genes were up-regulated in individuals from Prince William Sound and down-regulated in the Gulf of Alaska. Up-regulation of lipid catabolic genes in offshore individuals suggests they are experiencing nutritional deficits. The expression differences demonstrate physiological plasticity in response to a steep gradient in food availability. Our transcriptional analysis reveals mechanisms of acclimatization that likely contribute to the observed resilience of this population.

Bundle sheath extensions affect leaf structural and physiological plasticity in response to irradiance

Coordination between structural and physiological traits is key to plants' responses to environmental fluctuations. In heterobaric leaves, bundle sheath extensions (BSEs) increase photosynthetic performance (light-saturated rates of photosynthesis, Amax ) and water transport capacity (leaf hydraulic conductance, Kleaf ). However, it is not clear how BSEs affect these and other leaf developmental and physiological parameters in response to environmental conditions. The obscuravenosa (obv) mutation, found in many commercial tomato varieties, leads to absence of BSEs. We examined structural and physiological traits of tomato heterobaric and homobaric (obv) near-isogenic lines grown at two different irradiance levels. Kleaf , minor vein density, and stomatal pore area index decreased with shading in heterobaric but not in homobaric leaves, which show similarly lower values in both conditions. Homobaric plants, on the other hand, showed increased Amax , leaf intercellular air spaces, and mesophyll surface area exposed to intercellular airspace (Smes ) in comparison with heterobaric plants when both were grown in the shade. BSEs further affected carbon isotope discrimination, a proxy for long-term water-use efficiency. BSEs confer plasticity in traits related to leaf structure and function in response to irradiance levels and might act as a hub integrating leaf structure, photosynthetic function, and water supply and demand.

High resilience to extreme climatic changes in the CAM epiphyte Tillandsia utriculata L. (Bromeliaceae)

Climate change is expected to increase the frequency of extreme climatic events, yet few studies have addressed the capacity of plant species to deal with such events. Species that are widespread are predicted to be highly plastic and able to acclimate to highly changing conditions. To study the plasticity in physiological responses of the widely distributed epiphyte Tillandsia utriculata, we transplanted individuals from a coastal scrub and broadleaf evergreen forest to a similar coastal scrub site and forest. After a 45-day acclimation, the plants were moved to a semi-controlled greenhouse at each site, and then subjected to a 20-day drought. Physiological variables were measured during the acclimation and the drought. The individuals of scrub and forest populations had similar relative water content and carbon assimilation in the contrasting conditions of the two transplantation sites despite the high discrepancy between the environments at their original site. Electron transport rates were higher in individuals from the scrub population. Electron transport rates were also higher than estimated from carbon assimilation, suggesting that photorespiration was present. The individuals of the coastal scrub population had a higher capacity to dissipate excess energy this way. The relative distance index of plasticity was high overall, indicating that some traits are highly plastic (titratable acidity, carbon assimilation) in order to maintain the stability of others (maximum quantum yield Fv /Fm and relative water content). We conclude that T. utriculata is a highly plastic species with a high capacity to tolerate extreme environmental changes over a short time.

Prey adaptive behaviour under predation risk modify stoichiometry predictions of predator‐induced stress paradigms

Abstract Prey responses to predation risk can affect multiple phenotypic traits that have significant implications for population‐to‐ecosystem‐level properties. The general stress paradigm (hereafter GSP) proposes integrating the principles of ecological stoichiometry and prey physiological plasticity in response to predation risk to provide an explicit mechanistic framework to evaluate the direct influence of predation risk on ecosystem functioning. However, theoretical attempts evaluating the GSP have focused on only one component of phenotypic plasticity, neglecting the fact that multiple phenotypic responses can affect nutrient demand and consumption, and therefore, the overall animal effect on nutrient cycling rates and stoichiometry. In this study, we used a dynamic state variable model to examine how variation in predation and starvation risk on a landscape of patch types that vary in food quality affects the behavioural and physiological phenotypic responses of prey and the linked nutrient and carbon recycling. We found that the stress imposed by predation risk promotes prey carbon limitation, which leads to increased nutrient release and carbon uptake due to a stoichiometric mismatch between an animal's demand and food stoichiometry. Such effects can be altered by prey shifting spatially to carbon‐ or nutrient‐rich patches. However, the abilities of prey to cope with new physiological demands can be affected by absolute stress level or the predation risk distribution on a spatially implicit landscape of stoichiometric‐divergent patch type availability. Overall, starvation risk can act as a selective force favouring individuals that maximize their growth, thus reducing their nutrient release, but increasing their exposure to predation risk. Our results provide an important connection between predator‐induced stress ecology and ecosystem functioning, linking organism nutrition and ecological stoichiometry to fitness outcomes, thus improving our understanding of how non‐consumptive predator effects can scale up to ecosystems. Short-term physiological plasticity: Trade-off between drought and recovery responses in three Mediterranean Cistus species.

Short‐term physiological plasticity allows plants to thrive in highly variable environments such as the Mediterranean ecosystems. In such context, plants that maximize physiological performance under favorable conditions, such as Cistus spp., are generally reported to have a great cost in terms of plasticity (i.e., a high short‐term physiological plasticity) due to the severe reduction of physiological performance when stress factors occur. However, Cistus spp. also show a noticeable resilience ability in response to stress factors. We hypothesized that in Cistus species the short‐term physiological response to stress and that to subsequent recovery can show a positive trade‐off to offset the costs of the photosynthetic decline under drought. Gas exchange, chlorophyll fluorescence, and water relations were measured in C. salvifolius, C. monspeliensis, and C. creticus subsp. eriocephalus during an imposed experimental drought and subsequent recovery. Plants were grown outdoor in common garden conditions from seeds of different provenances. The short‐term physiological response to stress and that to recovery were quantified via phenotypic plasticity index (PIstress and PIrecovery, respectively). A linear regression analysis was used to identify the hypothesized trade‐off PIstress–PIrecovery. Accordingly, we found a positive trade‐off between PIstress and PIrecovery, which was consistent across species and provenances. This result contributes in explaining the profit, more than the cost, of a higher physiological plasticity in response to short‐term stress imposition for Cistus spp because the costs of a higher PIstress are payed back by an as much higher PIrecovery. The absence of leaf shedding during short‐term drought supports this view. The trade‐off well described the relative variations of gas exchange and water relation parameters. Moreover, the results were in accordance with the ecology of this species and provide the first evidence of a consistent trade‐off between the short‐term physiological responses to drought and recovery phases in Mediterranean species.

Environmental conditions modulate plasticity in the physiological responses of three plant species of the Neotropical savannah

Plasticity in plants could be changed due to abiotic factors, tending to increase fitness across environments. In the Neotropical savannah, a strong water deficit during the dry season is one of the main factors limiting the plasticity in physiological responses of plants. The present study aims to assess the plasticity in physiological responses and vegetative phenology of three plant species of the Neotropical savannah (Cerrado in Brazil) during the dry and the rainy seasons. The three species, Byrsonima verbascifolia, Roupala montana, and Solanum lycocarpum, occur in Serra do Cipo in the state of Minas Gerais, Brazil. The development and vegetative phenology of individuals of these three species were evaluated over the course of 1 year. In February 2012 (rainy season) and August 2012 (dry season), stomatal conductance (g s), water potential (Ψ), photosynthetic quantum yield, and concentration of leaf photosynthetic pigments were measured. The relative distance among the physiological parameters of all individuals within each season was measured using the relative distance plasticity index. B. verbascifolia has pronounced senescence in July and lost leaves completely by the early September, while R. montana and S. lycocarpum have green leaves throughout the year. The three studied species had greater control of stomatal opening during the dry season. S. lycocarpum and R. montana had negative water potential values in the dry season and in the middle of the day in both seasons. In the dry season, the three species exhibited a decrease in F v/F m, with values between 0.7 and 0.75. The relative distance plasticity index varied from 0 to 1. R. montana demonstrated the greatest plasticity and S. lycocarpum had lower plasticity. Then, a seasonal effect on physiological response was observed in all three model-species, with lower values for leaf water potential and stomatal conductance, and increased photoinhibition, in the dry season. Ecophysiological traits, such as stomatal conductance and leaf water potential, exhibited the greatest plasticity. In addition, there was a seasonal effect on the plasticity in physiological responses of the three plants species of the Neotropical savannah. The results are contradicting the idea that water restriction in the dry season would reduce the plasticity in most species of the Neotropical savannah.

Deletion of a Stay-Green Gene Associates with Adaptive Selection in Brassica napus

Chlorophyll levels provide important information about plant growth and physiological plasticity in response to changing environments. The stay-green gene NON-YELLOWING 1 (NYE1) is believed to regulate chlorophyll degradation during senescence, concomitantly affecting the disassembly of the light-harvesting complex and hence indirectly influencing photosynthesis. We identified Brassica napus accessions carrying an NYE1 deletion associated with increased chlorophyll content, and with upregulated expression of light-harvesting complex and photosynthetic reaction center (PSI and PSII) genes. Comparative analysis of the seed oil content of accessions with related genetic backgrounds revealed that the B. napus NYE1 gene deletion (bnnye1) affected oil accumulation, and linkage disequilibrium signatures suggested that the locus has been subject to artificial selection by breeding in oilseed B. napus forms. Comparative analysis of haplotype diversity groups (haplogroups) between three different ecotypes of the allopolyploid B. napus and its A-subgenome diploid progenitor, Brassica rapa, indicated that introgression of the bnnye1 deletion from Asian B. rapa into winter-type B. napus may have simultaneously improved its adaptation to cooler environments experienced by autumn-sown rapeseed.

Contrasting physiological plasticity in response to environmental stress within different cnidarians and their respective symbionts

Given concerns surrounding coral bleaching and ocean acidification, there is renewed interest in characterizing the physiological differences across the multiple host–algal symbiont combinations commonly found on coral reefs. Elevated temperature and CO2 were used to compare physiological responses within the scleractinian corals Montipora hirsuta (Symbiodinium C15) and Pocillopora damicornis (Symbiodinium D1), as well as the corallimorph (a non-calcifying anthozoan closely related to scleractinians) Discosoma nummiforme (Symbiodinium C3). Several physiological proxies were affected more by temperature than CO2, including photochemistry, algal number and cellular chlorophyll a. Marked differences in symbiont number, chlorophyll and volume contributed to distinctive patterns of chlorophyll absorption among these animals. In contrast, carbon fixation either did not change or increased under elevated temperature. Also, the rate of photosynthetically fixed carbon translocated to each host did not change, and the percent of carbon translocated to the host increased in the corallimorph. Comparing all data revealed a significant negative correlation between photosynthetic rate and symbiont density that corroborates previous hypotheses about carbon limitation in these symbioses. The ratio of symbiont-normalized photosynthetic rate relative to the rate of symbiont-normalized carbon translocation (P:T) was compared in these organisms as well as the anemone, Exaiptasia pallida hosting Symbiodinium minutum, and revealed a P:T close to unity (D. nummiforme) to a range of 2.0–4.5, with the lowest carbon translocation in the sea anemone. Major differences in the thermal responses across these organisms provide further evidence of a range of acclimation potential and physiological plasticity that highlights the need for continued study of these symbioses across a larger group of host taxa.

Physiological response to elevated temperature and pCO2 varies across four Pacific coral species: Understanding the unique host+symbiont response.

The physiological response to individual and combined stressors of elevated temperature and pCO2 were measured over a 24-day period in four Pacific corals and their respective symbionts (Acropora millepora/Symbiodinium C21a, Pocillopora damicornis/Symbiodinium C1c-d-t, Montipora monasteriata/Symbiodinium C15, and Turbinaria reniformis/Symbiodinium trenchii). Multivariate analyses indicated that elevated temperature played a greater role in altering physiological response, with the greatest degree of change occurring within M. monasteriata and T. reniformis. Algal cellular volume, protein, and lipid content all increased for M. monasteriata. Likewise, S. trenchii volume and protein content in T. reniformis also increased with temperature. Despite decreases in maximal photochemical efficiency, few changes in biochemical composition (i.e. lipids, proteins, and carbohydrates) or cellular volume occurred at high temperature in the two thermally sensitive symbionts C21a and C1c-d-t. Intracellular carbonic anhydrase transcript abundance increased with temperature in A. millepora but not in P. damicornis, possibly reflecting differences in host mitigated carbon supply during thermal stress. Importantly, our results show that the host and symbiont response to climate change differs considerably across species and that greater physiological plasticity in response to elevated temperature may be an important strategy distinguishing thermally tolerant vs. thermally sensitive species.

Neurobiological effects of neonatal maternal separation and post-weaning environmental enrichment

Throughout the lifespan, the brain has a considerable degree of plasticity and can be strongly influenced by sensory input from the outside environment. Given the importance of the environment in the regulation of the brain structure, behavior and physiology, the aim of the present work was to analyze the effects of different environmental qualities during two critical ontogenic periods (early life and peripuberty) on behavior and hippocampal physiology. Male Wistar rats were separated from their mothers for 4.5h daily during the first 3 weeks of life. They were weaned on day 21 and housed under either standard or enriched conditions. At 60 d of age, all animals were then housed in same-treatment groups, two per cage, until testing began on day 74. Emotional and cognitive responses were tested using the open field, novel object recognition test and step-down inhibitory avoidance learning. In the dorsal hippocampus, glucocorticoid receptor expression and neuronal activity were examined by immunoreactivity. Grooming behavior in the open field was found to be significantly lower in maternally separated animals, but post-weaning environmental enrichment completely reversed this tendency. Inhibitory avoidance but not object recognition memory was impaired in maternally separated animals, suggesting that early maternal separation alters learning and memory in a task-specific manner. Again, environmental enrichment reversed the effects of maternal separation on the inhibitory avoidance task. Even though maternal separation did not significantly affect Fos and glucocorticoid receptor (GR) expression, environmental enrichment increased both Fos expression in the total hippocampal area and also the overall number of GR positive cells per hippocampal area, mainly due to the changes in CA1. These findings suggest that differential rearing is a useful procedure to study behavioral and physiological plasticity in response to early experience and that, although the effects of adverse experience early in life such as maternal separation can persist until adulthood, some of them can be compensated by early favorable environments, possibly through nervous system plasticity.

Emotional and cognitive responses were tested using the open field, novel object recognition test and step-down inhibitory avoidance learning. In the dorsal hippocampus, glucocorticoid receptor expression and neuronal activity were examined by immunoreactivity.Grooming behavior in the open field was found to be significantly lower in maternally separated animals, but post-weaning environmental enrichment completely reversed this tendency. Inhibitory avoidance but not object recognition memory was impaired in maternally separated animals, suggesting that early maternal separation alters learning and memory in a task-specific manner. Again, environmental enrichment reversed the effects of maternal separation on the inhibitory avoidance task. Even though maternal separation did not significantly affect Fos and glucocorticoid receptor (GR) expression, environmental enrichment increased both Fos expression in the total hippocampal area and also the overall number of GR positive cells per hippocampal area, mainly due to the changes in CA1.These findings suggest that differential rearing is a useful procedure to study behavioral and physiological plasticity in response to early experience and that, although the effects of adverse experience early in life such as maternal separation can persist until adulthood, some of them can be compensated by early favorable environments, possibly through nervous system plasticity.

Insulin signaling as a mechanism underlying developmental plasticity: the role of FOXO in a nutritional polyphenism.

We investigated whether insulin signaling, known to mediate physiological plasticity in response to changes in nutrition, also facilitates discrete phenotypic responses such as polyphenisms. We test the hypothesis that the gene FOXO – which regulates growth arrest under nutrient stress – mediates a nutritional polyphenism in the horned beetle, Onthophagus nigriventris. Male beetles in the genus Onthophagus vary their mating strategy with body size: large males express horns and fight for access to females while small males invest heavily in genitalia and sneak copulations with females. Given that body size and larval nutrition are linked, we predicted that 1) FOXO expression would differentially scale with body size (nutritional status) between males and females, and 2) manipulation of FOXO expression would affect the nutritional polyphenism in horns and genitalia. First, we found that FOXO expression varied with body size in a tissue- and sex-specific manner, being more highly expressed in the abdominal tissue of large (horned) males, in particular in regions associated with genitalia development. Second, we found that knockdown of FOXO through RNA-interference resulted in the growth of relatively larger copulatory organs compared to control-injected individuals and significant, albeit modest, increases in relative horn length. Our results support the hypothesis that FOXO expression in the abdominal tissue limits genitalia growth, and provides limited support for the hypothesis that FOXO regulates relative horn length through direct suppression of horn growth. Both results support the idea that tissue-specific FOXO expression may play a general role in regulating scaling relationships in nutritional polyphenisms by signaling traits to be relatively smaller.

Respostas fisiológicas, morfológicas e comportamentais de Zabrotes subfasciatus (Boheman) (Coleoptera: Bruchidae) associadas ao consumo de diferentes variedades de feijão (Phaseolus vulgaris)

This study was developed aiming to verify physiological, morphological and behavioral responses of two different Zabrotes subfasciatus (Boheman) populations to different beans varieties (Phaseolus vulgaris) (Fabaceae). Female longevity, fertility and oviposition preference site, as well as size and levels of fluctuating asymmetry for males and females were described. Zabrotes subfasciatus displayed physiological plasticity in response to the diet, which was considered an important adaptive ability to maintain the insect generalist habit for food consumption and oviposition sites. The populations studied had different responses to the same treatments, indicating genetic, physiological and behavioral variation on their plastic potential. The Hopkins principle, which determines the influence of previous female experience in the choice of oviposition sites, was not confirmed. The occurrence of fluctuating asymmetry in males and females was variable, probably as a consequence of genomic factors determining this trait.
