Photoinhibition and photosynthetic regulation in fluctuating light under compound stresses of drought and heat

AbstractUnder the background of global climate change, crop leaves usually experience fluctuating light (FL) under compound stresses of drought and heat in the field. However, photoinhibition and photosynthetic regulation in FL under these compound stresses are little known. To elucidate this, we measured gas exchange, chlorophyll fluorescence, P700 redox state and electrochromic shift signals for tomato (Solanum lycopersicum) leaves exposed to FL under individual and compound stresses of moderate drought and heat stresses. After treatment with short‐term FL, photosystem I (PSI) activity decreased slightly in all treatments. By comparison, photosystem II (PSII) activity was reduced slightly under individual stress but largely decreased by 46% under compound stresses. Therefore, PSI is more tolerant than PSII when exposed to FL under compound stresses of drought and heat. Under compound stresses, PSII photoinhibition largely downregulated linear electron flow and photorespiration became a major electron sink downstream. Concomitantly, cyclic electron flow (CEF) was highly enhanced to help the building up of trans‐thylakoid proton gradient. This study highlights the importance of PSII activity and CEF in crop tolerance under future compound climate extremes such as drought and heat.

Plastid-expressed AdDjSKI enhances photosystem II stability, delays leaf senescence, and increases fruit yield in tomato plants under heat stress.

Heat stress substantially reduces tomato (Solanum lycopersicum) growth and yield globally, thereby jeopardizing food security. DnaJ proteins, constituents of the heat shock protein system, protect cells from diverse environmental stresses as HSP-70 molecular co-chaperones. In this study, we demonstrated that AdDjSKI, a serine-rich DnaJ III protein induced by pathogens, plays an important role in stabilizing photosystem II (PSII) in response to heat stress. Our results revealed that transplastomic tomato plants expressing the AdDjSKI gene exhibited increased levels of total soluble proteins, improved growth and chlorophyll content, reduced malondialdehyde (MDA) accumulation, and diminished PSII photoinhibition under elevated temperatures when compared with wild-type (WT) plants. Intriguingly, these transplastomic plants maintained higher levels of D1 protein under elevated temperatures compared with the WT plants, suggesting that overexpression of AdDjSKI in plastids is crucial for PSII protection, likely due to its chaperone activity. Furthermore, the transplastomic plants displayed lower accumulation of superoxide radical (O2 •─) and H2O2, in comparison with the WT plants, plausibly attributed to higher superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities. This also coincides with an enhanced expression of corresponding genes, including SlCuZnSOD, SlFeSOD, SlAPX2, and SltAPX, under heat stress. Taken together, our findings reveal that chloroplastic expression of AdDjSKI in tomatoes plays a critical role in fruit yield, primarily through a combination of delayed senescence and stabilizing PSII under heat stress.

Photosynthetic control at the cytochrome b6f complex.

Photosynthetic control (PCON) is a protective mechanism that prevents light-induced damage to photosystem I (PSI) by ensuring the rate of NADPH and ATP production via linear electron transfer (LET) is balanced by their consumption in the CO2 fixation reactions. Protection of PSI is a priority for plants since they lack a dedicated rapid-repair cycle for this complex, meaning that any damage leads to prolonged photoinhibition and decreased growth. The imbalance between LET and the CO2 fixation reactions is sensed at the level of the transthylakoid ΔpH, which increases when light is in excess. The canonical mechanism of PCON involves feedback control by ΔpH on the plastoquinol oxidation step of LET at cytochrome b6f. PCON thereby maintains the PSI special pair chlorophylls (P700) in an oxidized state, that allows excess electrons unused in the CO2 fixation reactions to be safely quenched via charge recombination. In this review we focus on angiosperms, considering how photo-oxidative damage to PSI comes about, explore the consequences of PSI photoinhibition on photosynthesis and growth, discuss recent progress in understanding PCON regulation, and finally consider the prospects for its future manipulation in crop plants to improve photosynthetic efficiency.

Accounting for photosystem I photoinhibition sheds new light on seasonal acclimation strategies of boreal conifers.

The photosynthetic acclimation of boreal evergreen conifers is controlled by regulatory and photoprotective mechanisms that allow conifers to cope with extreme environmental changes. However, the underlying dynamics of photosystem II (PSII) and photosystem I (PSI) remain unresolved. Here, we investigated the dynamics of PSII and PSI during the spring recovery of photosynthesis in Pinus sylvestris and Picea abies using a combination of chlorophyll-a fluorescence, P700 difference absorbance measurements, and quantification of key thylakoid protein abundances. In particular, we derived a new set of PSI quantum yield equations, correcting for the effects of PSI photoinhibition. Using the corrected equations, we found that the seasonal dynamics of PSII and PSI photochemical yields remained largely in balance, despite substantial seasonal changes in the stoichiometry of PSII and PSI core complexes driven by PSI photoinhibition. Similarly, the previously reported seasonal upregulation of cyclic electron flow was no longer evident, after accounting for PSI photoinhibition. Overall, our results emphasize the importance of considering the dynamics of PSII and PSI to elucidate the seasonal acclimation of photosynthesis in overwintering evergreens. Beyond the scope of conifers, our corrected PSI quantum yields expand the toolkit for future studies aimed at elucidating the dynamic regulation of PSI.

The toxigenic red-tide-forming dinoflagellates Alexandrium leei and Alexandrium catenella differ in terms of the sensitivity to strong light and low temperature of their photosynthetic machinery

Alexandrium leei and Alexandrium catenella are noxious dinoflagellates that form red tides but the two species differ substantially in optimal growth conditions. For formation of blooms in natural environments, A. leei favors moderate temperatures (around 20 °C) while A. catenella favors much lower temperatures (around 10 °C). We examined the effects of temperature and light on cell proliferation and photosynthesis in cell cultures of the two species. Under light at 100 μmol photons m−2 s−1, the growth rate of both species was highest at 20 °C. At 25 °C, A. leei continued to proliferate but A. catenella did not survive. Conversely, at lower temperatures, namely, 12 °C and 15 °C, the growth rate of A. leei was very low while that of A. catenella remained high. The net photosynthetic activity, as determined in terms of oxygen evolution, was highest at 20 °C in A. leei. By contrast, it was highest at 15 °C in A. catenella. Illumination of cells with strong light at 1000 μmol photons m−2 s−1 decreased the activity of photosystem II (PSII), as determined in terms of Fv/Fm, in both species. In A. leei, the activity of PSII declined more rapidly at 12 °C and 15 °C than at 20 °C and 25 °C under strong light. In A. catenella, the activity of PSII declined more rapidly at 20 °C than at 12 °C and 15 °C. Thus, A. catenella appeared to be more resistant to photoinhibition of PSII at low temperatures than A. leei. In addition, the ability to repair photodamaged PSII at 12 °C and 15 °C was greater in A. catenella than that in A. leei. Thus, the tolerance of PSII to strong light and low temperatures might determine the range of temperatures that supports the formation of blooms of the two Alexandrium species.

Dysfunction of Chloroplast Protease Activity Mitigates pgr5 Phenotype in the Green Algae Chlamydomonas reinhardtii.

Researchers have described protection mechanisms against the photoinhibition of photosystems under strong-light stress. Cyclic Electron Flow (CEF) mitigates electron acceptor-side limitation, and thus contributes to Photosystem I (PSI) protection. Chloroplast protease removes damaged protein to assist with protein turn over, which contributes to the quality control of Photosystem II (PSII). The PGR5 protein is involved in PGR5-dependent CEF. The FTSH protein is a chloroplast protease which effectively degrades the damaged PSII reaction center subunit, D1 protein. To investigate how the PSI photoinhibition phenotype in pgr5 would be affected by adding the ftsh mutation, we generated double-mutant pgr5ftsh via crossing, and its phenotype was characterized in the green algae Chlamydomonas reinhardtii. The cells underwent high-light incubation as well as low-light incubation after high-light incubation. The time course of Fv/Fm values in pgr5ftsh showed the same phenotype with ftsh1-1. The amplitude of light-induced P700 photo-oxidation absorbance change was measured. The amplitude was maintained at a low value in the control and pgr5ftsh during high-light incubation, but was continuously decreased in pgr5. During the low-light incubation after high-light incubation, amplitude was more rapidly recovered in pgr5ftsh than pgr5. We concluded that the PSI photoinhibition by the pgr5 mutation is mitigated by an additional ftsh1-1 mutation, in which plastoquinone pool would be less reduced due to damaged PSII accumulation.

Nitrate nitrogen enhances the efficiency of photoprotection in Leymus chinensis under drought stress.

Global climate change exerts a significant impact on the nitrogen supply and photosynthesis ability in land-based plants. The photosynthetic capacity of dominant grassland species is important if we are to understand carbon cycling under climate change. Drought stress is one of the major factors limiting plant photosynthesis, and nitrogen (N) is an essential nutrient involved in the photosynthetic activity of leaves. The regulatory mechanisms responsible for the effects of ammonium (NH4 +) and nitrate (NO3 -) on the drought-induced photoinhibition of photosystem II (PSII) in plants have yet to be fully elucidated. Therefore, there is a significant need to gain a better understanding of the role of electron transport in the photoinhibition of PSII. In the present study, we conducted experiments with normal watering (LD), severe drought (MD), and extreme drought (HD) treatments, along with no nitrogen (N0), ammonium (NH4), nitrate (NO3), and mixed nitrogen (NH4NO3) treatments. We analyzed pigment accumulation, reactive oxygen species (ROS) accumulation, photosynthetic enzyme activity, photosystem activity, electron transport, and O-J-I-P kinetics. Analysis showed that increased nitrate application significantly increased the leaf chlorophyll content per unit area (Chlarea) and nitrogen content per unit area (Narea) (p< 0.05). Under HD treatment, ROS levels were lower in NO3-treated plants than in N0 plants, and there was no significant difference in photosynthetic enzyme activity between plants treated with NO3 and NH4NO3. Under drought stress, the maximum photochemical efficiency of PSII (Fv/Fm), PSII electron transport rate (ETR), and effective quantum yield of PSII (φPSII) were significant higher in NO3-treated plants (p< 0.05). Importantly, the K-band and G-band were higher in NO3-treated plants. These results suggest that drought stress hindered the formation of NADPH and ATP in N0 and NH4-treated L. chinensis plants, thus damaging the donor side of the PSII oxygen-evolving complex (OEC). After applying nitrate, higher photosynthetic enzyme and antioxidant enzyme activity not only protected PSII from photodamage under drought stress but also reduced the rate of damage in PSII during the growth of L. chinensis growth under drought stress.

Improved capacity for the repair of photosystem II via reinforcement of the translational and antioxidation systems in Synechocystis sp. PCC 6803.

In the cyanobacterium Synechocystis sp. PCC 6803, translation factor EF-Tu is inactivated by reactive oxygen species (ROS) via oxidation of Cys82 and the oxidation of EF-Tu enhances the inhibition of the repair of photosystem II (PSII) by suppressing protein synthesis. In our present study, we generated transformants of Synechocystis that overexpressed a mutated form of EF-Tu, designated EF-Tu (C82S), in which Cys82 had been replaced by a Ser residue, and ROS-scavenging enzymes individually or together. Expression of EF-Tu (C82S) alone in Synechocystis enhanced the repair of PSII under strong light, with the resultant mitigation of PSII photoinhibition, but it stimulated the production of ROS. However, overexpression of superoxide dismutase and catalase, together with the expression of EF-Tu (C82S), lowered intracellular levels of ROS and enhanced the repair of PSII more significantly under strong light, via facilitation of the synthesis de novo of the D1 protein. By contrast, the activity of photosystem I was hardly affected in wild-type cells and in all the lines of transformed cells under the same strong-light conditions. Furthermore, transformed cells that overexpressed EF-Tu (C82S), superoxide dismutase, and catalase were able to survive longer under stronger light than wild-type cells. Thus, the reinforced capacity for both protein synthesis and ROS scavenging allowed both photosynthesis and cell proliferation to tolerate strong light.

PSI Photoinhibition and Changing CO2 Levels Initiate Retrograde Signals to Modify Nuclear Gene Expression.

Photosystem I (PSI) is a critical component of the photosynthetic machinery in plants. Under conditions of environmental stress, PSI becomes photoinhibited, leading to a redox imbalance in the chloroplast. PSI photoinhibition is caused by an increase in electron pressure within PSI, which damages the iron-sulfur clusters. In this study, we investigated the susceptibility of PSI to photoinhibition in plants at different concentrations of CO2, followed by global gene expression analyses of the differentially treated plants. PSI photoinhibition was induced using a specific illumination protocol that inhibited PSI with minimal effects on PSII. Unexpectedly, the varying CO2 levels combined with the PSI-PI treatment neither increased nor decreased the likelihood of PSI photodamage. All PSI photoinhibition treatments, independent of CO2 levels, upregulated genes generally involved in plant responses to excess iron and downregulated genes involved in iron deficiency. PSI photoinhibition also induced genes encoding photosynthetic proteins that act as electron acceptors from PSI. We propose that PSI photoinhibition causes a release of iron from damaged iron-sulfur clusters, which initiates a retrograde signal from the chloroplast to the nucleus to modify gene expression. In addition, the deprivation of CO2 from the air initiated a signal that induced flavonoid biosynthesis genes, probably via jasmonate production.

X- and y-type thioredoxins maintain redox homeostasis on photosystem I acceptor side under fluctuating light.

Plants cope with sudden increases in light intensity through various photoprotective mechanisms. Redox regulation by thioredoxin (Trx) systems also contributes to this process. Whereas the functions of f- and m-type Trxs in response to such fluctuating light conditions have been extensively investigated, those of x- and y-type Trxs are largely unknown. Here, we analyzed the trx x single, trx y1 trx y2 double, and trx x trx y1 trx y2 triple mutants in Arabidopsis (Arabidopsis thaliana). A detailed analysis of photosynthesis revealed changes in photosystem I (PSI) parameters under low light in trx x and trx x trx y1 trx y2. The electron acceptor side of PSI was more reduced in these mutants than in the wild type. This mutant phenotype was more pronounced under fluctuating light conditions. During both low- and high-light phases, the PSI acceptor side was largely limited in trx x and trx x trx y1 trx y2. After fluctuating light treatment, we observed more severe PSI photoinhibition in trx x and trx x trx y1 trx y2 than in the wild type. Furthermore, when grown under fluctuating light conditions, trx x and trx x trx y1 trx y2 plants showed impaired growth and decreased level of PSI subunits. These results suggest that Trx x and Trx y prevent redox imbalance on the PSI acceptor side, which is required to protect PSI from photoinhibition, especially under fluctuating light. We also propose that Trx x and Trx y contribute to maintaining the redox balance even under constant low-light conditions to prepare for sudden increases in light intensity.

The impact of long-term acclimation to different growth light intensities on the regulation of zeaxanthin epoxidase in different plant species.

Proper short- and long-term acclimation to different growth light intensities is essential for the survival and competitiveness of plants in the field. High light exposure is known to induce the down-regulation and photoinhibition of photosystem II (PSII) activity to reduce photo-oxidative stress. The xanthophyll zeaxanthin (Zx) serves central photoprotective functions in these processes. We have shown in recent work with different plant species (Arabidopsis, tobacco, spinach and pea) that photoinhibition of PSII and degradation of the PSII reaction center protein D1 is accompanied by the inactivation and degradation of zeaxanthin epoxidase (ZEP), which catalyzes the reconversion of Zx to violaxanthin. Different high light sensitivity of the above-mentioned species correlated with differential down-regulation of both PSII and ZEP activity. Applying light and electron microscopy, chlorophyll fluorescence, and protein and pigment analyses, we investigated the acclimation properties of these species to different growth light intensities with respect to the ability to adjust their photoprotective strategies. We show that the species differ in phenotypic plasticity in response to short- and long-term high light conditions at different morphological and physiological levels. However, the close co-regulation of PSII and ZEP activity remains a common feature in all species and under all conditions. This work supports species-specific acclimation strategies and properties in response to high light stress and underlines the central role of the xanthophyll Zx in photoprotection.

Biophysical and molecular characteristics of senescing leaves of two Norway maple varieties differing in anthocyanin content.

Disassembly and degradation of the photosynthetic protein complexes during autumn senescence, a vital step to ensure efficient nutrient relocalization for winter storage, is poorly understood. Concomitantly with the degradation, anthocyanins are often synthesized. However, as to why leaves accumulate red pigments, no consensus exists. One possibility is that anthocyanins protect senescing leaves from excess light. In this study, we investigated the pigment composition, photosynthetic performance, radical production, and degradation of the photosynthetic protein complexes in Norway maple (Acer platanoides) and in its highly pigmented, purple-colored variety (Faassen's black) during autumn senescence, to dissect the possible roles of anthocyanins in photoprotection. Our findings show that senescing Faassen's black was indeed more resistant to Photosystem II (PSII) photoinhibition, presumably due to its high anthocyanin content, than the green maple. However, senescing Faassen's black exhibited low photosynthetic performance, probably due to a poor capacity to repair PSII. Furthermore, an analysis of photosynthetic protein complexes demonstrated that in both maple varieties, the supercomplexes consisting of PSII and its antenna were disassembled first, followed by the degradation of the PSII core, Photosystem I, Cytochrome b6 f, and ATP synthase. Strikingly, the degradation process appeared to proceed faster in Faassen's black, possibly explaining its poor PSII repair capacity. The results suggest that tolerance against PSII photoinhibition may not necessarily translate to a better fitness. Finally, thylakoids isolated from senescing and non-senescing leaves of both maple varieties accumulated very little carbon-centered radicals, suggesting that thylakoids may not be a major source of reactive oxygen species in senescing leaves.

Long-Term Alleviation of the Functional Phenotype in Chlorophyll-Deficient Wheat and Impact on Productivity: A Semi-Field Phenotyping Experiment.

Wheat mutants with a reduced chlorophyll synthesis are affected by a defective control of the photosynthetic electron flow, but tend to recover a wild-type phenotype. The sensitivity of some mutants to light fluctuations suggested that cultivation outdoors could significantly impact productivity. Six mutant lines of Triticum durum or Triticum aestivum with their respective wild-type cultivars were cultivated with a regular seasonal cycle (October-May) in a semi-field experiment. Leaf chlorophyll content and fluorescence parameters were analysed at the early (November) and late (May) developmental stages, and checked for correlation with morphometric and grain-production parameters. The alleviation of the phenotype severity concerned primarily the recovery of the photosynthetic-membrane functionality, but not the leaf chlorophyll content. Photosystem II (PSII) was less photoprotected in the mutants, but a moderate PSII photoinhibition could help control the electron flow into the chain. The accumulation of interchain electron carriers was a primary acclimative response towards the naturally fluctuating environment, maximally exploited by the mature durum-wheat mutants. The mutation itself and/or the energy-consuming compensatory mechanisms markedly influenced the plant morphogenesis, leading especially to reduced tillering, which in turn resulted in lower grain production per plant. Consistently with the interrelation between early photosynthetic phenotype and grain-yield per plant, chlorophyll-fluorescence indexes related to the level of photoprotective thermal dissipation (pNPQ), photosystem II antenna size (ABS/RC), and pool of electron carriers (Sm) are proposed as good candidates for the in-field phenotyping of chlorophyll-deficient wheat.

Physiological Role of pH-Dependent Structural Transition in Oxygen-Evolving Complex of PSII

Photosystem II (PSII) of the photosynthetic apparatus in oxygenic organisms contains a catalytic center that performs one of the most important reactions in bioenergetics: light-dependent water oxidation to molecular oxygen. The catalytic center is a Mn4CaO5 cluster consisting of four cations of manganese and one calcium cation linked by oxygen bridges. The authors reported earlier that a structural transition occurs at pH 5.7 in the cluster resulting in changes in manganese cation(s) redox potential and elevation of the Mn‑clus-ter resistance to reducing agents. The discovered effect was examined in a series of investigations that are reviewed in this work. It was found that, at pH 5.7, Fe(II) cations replace not two manganese cations as it happens at pH 6.5 but only one cation; as a result, a chimeric Mn3Fe1 cluster is produced. In the presence of exogenous calcium ions, membrane preparations of PSII with such a chimeric cluster are capable of evolving oxygen in the light (at a rate of approximately 25% of the rate in native PSII). It was found that photoinhibition that greatly depends on the processes of oxidation or reduction at pH 5.7 slows down as compared with pH 6.5. PSII preparations were also more resistant to thermal inactivation at pH 5.7 than at pH 6.5. However, in PSII preparations lacking manganese cations in the oxygen-evolving complex, the rates of photoinhibition at pH 6.5 and 5.7 did not differ. In thylakoid membranes, protonophores that abolish the proton gradient and increase pH in the lumen (where the manganese cluster is located) from 5.7 to 7.0 considerably elevated the rate of PSII photoinhibition. It is assumed that the structural transition in the Mn-cluster at pH 5.7 is involved in the mechanisms of PSII defense against photoinhibition.

Red pigments in autumn leaves of Norway maple do not offer significant photoprotection but coincide with stress symptoms.

The reasons behind autumn colors, a striking manifestation of anthocyanin synthesis in plants, are poorly understood. Usually, not all leaves of an anthocyanic plant turn red or only a part of the leaf blade turns red. In the present study, we compared green, red and yellow sections of senescing Norway maple leaves, asking if red pigments offer photoprotection, and if so, whether the protection benefits the senescing tree. Green and senescing maple leaves were illuminated with strong white, green or red light in the absence or presence of lincomycin which blocks photosystem II (PSII) repair. Irrespective of the presence of anthocyanins, senescing leaves showed weaker capacity to repair PSII than green leaves. Furthermore, the rate of photoinhibition of PSII did not significantly differ between red and yellow sections of senescing maple leaves. We also followed pigment contents and photosynthetic reactions in individual leaves, from the end of summer until abscission of the leaf. In maple, red pigments accumulated only during late senescence, but light reactions stayed active until most of the chlorophyll had been degraded. PSII activity was found to be lower and non-photochemical quenching higher in red leaf sections, compared with yellow sections of senescing leaves. Red leaf sections were also thicker. We suggest that the primary function of anthocyanin synthesis is not to protect senescing leaves from excess light but to dispose of carbohydrates. This would relieve photosynthetic control, allowing the light reactions to produce energy for nutrient translocation at the last phase of autumn senescence when carbon skeletons are no longer needed.

Drought stress delays photosynthetic induction and accelerates photoinhibition under short-term fluctuating light in tomato

Fluctuating light (FL) and drought stress usually occur concomitantly. However, whether drought stress affects photosynthetic performance under FL remains unknown. Here, we measured gas exchange, chlorophyll fluorescence, and P700 redox state under FL in drought-stressed tomato (Solanum lycopersicum) seedlings. Drought stress significantly delayed the induction kinetics of stomatal and mesophyll conductances after transition from low to high light and thus delayed photosynthetic induction under FL. Therefore, drought stress exacerbated the loss of carbon gain under FL. Furthermore, restriction of CO2 fixation under drought stress aggravated the over-reduction of photosystem I (PSI) upon transition from low to high light. The resulting stronger FL-induced PSI photoinhibition significantly suppressed linear electron flow and PSI photoprotection. These results indicated that drought stress not only caused a larger loss of carbon gain under FL but also accelerated FL-induced photoinhibition of PSI. Furthermore, drought stress enhanced relative cyclic electron flow in FL, which partially compensated for restricted CO2 fixation and thus favored PSI photoprotection under FL. To our knowledge, we here show new insight into how drought stress affects photosynthetic performance under FL.

In thylakoid membranes, protonophores that abolish the proton gradient and increase pH in the lumen (where the manganese cluster is located) from 5.7 to 7.0 considerably elevated the rate of PSII photoinhibition. It is assumed that the structural transition in the Mn-cluster at pH 5.7 is involved in the mechanisms of PSII defense against photoinhibition.

Photoinhibition of Photosystem I Induced by Different Intensities of Fluctuating Light Is Determined by the Kinetics of ∆pH Formation Rather Than Linear Electron Flow.

Fluctuating light (FL) can cause the selective photoinhibition of photosystem I (PSI) in angiosperms. In nature, leaves usually experience FL conditions with the same low light and different high light intensities, but the effects of different FL conditions on PSI redox state and PSI photoinhibition are not well known. In this study, we found that PSI was highly reduced within the first 10 s after transition from 59 to 1809 μmol photons m-2 s-1 in tomato (Solanum lycopersicum). However, such transient PSI over-reduction was not observed by transitioning from 59 to 501 or 923 μmol photons m-2 s-1. Consequently, FL (59-1809) induced a significantly stronger PSI photoinhibition than FL (59-501) and FL (59-923). Compared with the proton gradient (∆pH) level after transition to high light for 60 s, tomato leaves almost formed a sufficient ∆pH after light transition for 10 s in FL (59-501) but did not in FL (59-923) or FL (59-1809). The difference in ∆pH between 10 s and 60 s was tightly correlated to the extent of PSI over-reduction and PSI photoinhibition induced by FL. Furthermore, the difference in PSI photoinhibition between (59-923) and FL (59-1809) was accompanied by the same level of linear electron flow. Therefore, PSI photoinhibition induced by different intensities of FL is more related to the kinetics of ∆pH formation rather than linear electron flow.

RNA interference (RNAi) of 2-Cys Prx gene enhances PSII photoinhibition but does not affect PSI activity in tobacco leaves under high-temperature stress

ABSTRACT This study aimed to evaluate the effects of 2-Cys Prx gene inhibition on photochemical reaction and reactive oxygen species (ROS) metabolism under high temperature (35°C) with low light (HT + LL) or high temperature with high light (HT + HL) in tobacco. The results showed that HT significantly increased the production of and H2O2 compared with CK (25°C). Particularly, the oxidative damage of RNAi plants was significantly greater than that of wild type (WT) under HT + HL treatment, possibly due to the inhibition of superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities. HT treatment inhibited the photosystem II (PSII) activity, and the oxygen evolution complex (OEC) was the main injury site. Notably, the photosystem I (PSI) activity of WT and RNAi plants did not change significantly under HT + LL treatment compared with CK. Although the PSI activity of WT and RNAi plants decreased significantly under HT + HL treatment, there was no significant difference between WT and RNAi plants. Collectively, these findings indicate that high light increases the photoinhibition of PSII and PSI and oxidative damage under high-temperature stress. The results also revealed that 2-Cys Prx plays a crucial role in alleviating oxidative damage and PSII photoinhibition under high-temperature stress in tobacco.

Dissecting photosynthetic electron transport and photosystems performance in Jerusalem artichoke (Helianthus tuberosus L.) under salt stress.

Jerusalem artichoke (Helianthus tuberosus L.), a vegetable with medical applications, has a strong adaptability to marginal barren land, but the suitability as planting material in saline land remains to be evaluated. This study was envisaged to examine salt tolerance in Jerusalem artichoke from the angle of photosynthetic apparatus stability by dissecting the photosynthetic electron transport process. Potted plants were exposed to salt stress by watering with a nutrient solution supplemented with NaCl. Photosystem I (PSI) and photosystem II (PSII) photoinhibition appeared under salt stress, according to the significant decrease in the maximal photochemical efficiency of PSI (△MR/MR0) and PSII. Consistently, leaf hydrogen peroxide (H2O2) concentration and lipid peroxidation were remarkably elevated after 8 days of salt stress, confirming salt-induced oxidative stress. Besides photoinhibition of the PSII reaction center, the PSII donor side was also impaired under salt stress, as a K step emerged in the prompt chlorophyll transient, but the PSII acceptor side was more vulnerable, considering the decreased probability of an electron movement beyond the primary quinone (ETo/TRo) upon depressed upstream electron donation. The declined performance of entire PSII components inhibited electron inflow to PSI, but severe PSI photoinhibition was not averted. Notably, PSI photoinhibition elevated the excitation pressure of PSII (1-qP) by inhibiting the PSII acceptor side due to the negative and positive correlation of △MR/MR0 with 1-qP and ETo/TRo, respectively. Furthermore, excessive reduction of PSII acceptors side due to PSI photoinhibition was simulated by applying a specific inhibitor blocking electron transport beyond primary quinone, demonstrating that PSII photoinhibition was actually accelerated by PSI photoinhibition under salt stress. In conclusion, PSII and PSI vulnerabilities were proven in Jerusalem artichoke under salt stress, and PSII inactivation, which was a passive consequence of PSI photoinhibition, hardly helped protect PSI. As a salt-sensitive species, Jerusalem artichoke was recommended to be planted in non-saline marginal land or mild saline land with soil desalination measures.
