Integrated multi-omic analysis reveals the carbon metabolism-mediated regulation of polysaccharide biosynthesis by suitable light intensity in Bletilla striata leaves

Bletilla striata, valued for its medicinal and ornamental properties, remains largely unexplored in terms of how light intensity affects its physiology, biochemistry, and polysaccharide formation. In this 5-month study, B. striata plants were exposed to three different light intensities: low light (LL) (5–20 μmol m−2·s−1), middle light (ML) (200 μmol m−2·s−1), and high light (HL) (400 μmol m−2·s−1). The comprehensive assessment included growth, photosynthetic apparatus, chlorophyll fluorescence electron transport, and analysis of differential metabolites based on the transcriptome and metabolome data. The results indicated that ML resulted in the highest plant height and total polysaccharide content, enhanced photosynthetic apparatus performance and light energy utilization, and stimulated carbon metabolism and carbohydrate accumulation. HL reduced Chl content and photosynthetic apparatus functionality, disrupted OEC activity and electron transfer, stimulated carbon metabolism and starch and glucose accumulation, and hindered energy metabolism related to carbohydrate degradation and oxidation. In contrast, LL facilitated leaf growth and increased chlorophyll content but decreased plant height and total polysaccharide content, compromised the photosynthetic apparatus, hampered light energy utilization, stimulated energy metabolism related to carbohydrate degradation and oxidation, and inhibited carbon metabolism and carbohydrate synthesis. Numerous genes in carbon metabolism were strongly related to polysaccharide metabolites. The katE and cysK genes in carbon metabolism were strongly related not only to polysaccharide metabolites, but also to genes involved in polysaccharide biosynthesis. Our results highlight that light intensity plays a crucial role in affecting polysaccharide biosynthesis in B. striata, with carbon metabolism acting as a mediator under suitable light intensity conditions.

Nitrate modulates the physiological tolerance responses of the halophytic species Sarcocornia fruticosa to copper excess

Coexistence impact of pollutants of different nature on halophytes tolerance to metal excess has not been thoroughly examined, and plant functional responses described so far do not follow a clear pattern. Using the Cu-tolerant halophyte Sarcocornia fruticosa as a model species, we conducted a greenhouse experiment to evaluate the impact of two concentration of copper (0 and 12 mM CuSO4) in combination with three nitrate levels (2, 14 and 50 mM KNO3) on plant growth, photosynthetic apparatus performance and ROS-scavenging enzymes system. The results revealed that S. fruticosa was able to grow adequately even when exposed to high concentrations of copper and nitrate. This response was linked to the plant capacity to uptake and retain a large amount of copper in its roots (up to 1500 mg kg−1 Cu), preventing its transport to aerial parts. This control of translocation was further magnified with nitrate concentration increment. Likewise, although Cu excess impaired S. fruticosa carbon assimilation capacity, the plant was able to downregulate its light-harvesting complexes function, as indicated its lowers ETR values, especially at 12 mM Cu + 50 mM NO3. This downregulation would contribute to avoid excess energy absorption and transformation. In addition, this strategy of avoiding excess energy was accompanied by the upregulation of all ROS-scavenging enzymes, a response that was further enhanced by the increase in nitrate concentration. Therefore, we conclude that the coexistence of nitrate would favor S. fruticosa tolerance to copper excess, and this effect is mediated by the combined activation of several tolerance mechanisms.

Medium-chain-length polyprenol (C45–C55) formation in chloroplasts of Arabidopsis is brassinosteroid-dependent

Brassinosteroids are important plant hormones influencing, among other processes, chloroplast development, the electron transport chain during light reactions of photosynthesis, and the Calvin-Benson cycle. Medium-chain-length polyprenols built of 9–11 isoprenoid units (C45–C55 carbons) are a class of isoprenoid compounds present in abundance in thylakoid membranes. They are synthetized in chloroplast by CPT7 gene from Calvin cycle derived precursors on MEP (methylerythritol 4-phosphate) isoprenoid biosynthesis pathway. C45–C55 polyprenols affect thylakoid membrane ultra-structure and hence influence photosynthetic apparatus performance in plants such as Arabidopsis and tomato. So far nothing is known about the hormonal or environmental regulation of CPT7 gene expression. The aim of our study was to find out if medium-chain-length polyprenol biosynthesis in plants may be regulated by hormonal cues.We found that the CPT7 gene in Arabidopsis has a BZR1 binding element (brassinosteroid dependent) in its promoter. Brassinosteroid signaling mutants in Arabidopsis accumulate a lower amount of medium-chain-length C45–C55 polyprenols than control plants. At the same time carotenoid and chlorophyll content is increased, and the amount of PsbD1A protein coming from photosystem II does not undergo a significant change. On contrary, treatment of WT plants with epi-brassinolide increases C45–C55 polyprenols content. We also report decreased transcription of MEP enzymes (besides C45–C55 polyprenols, precursors of numerous isoprenoids, e.g. phytol, carotenoids are derived from this pathway) and genes encoding biosynthesis of medium-chain-length polyprenol enzymes in brassinosteroid perception mutant bri1-116. Taken together, we document that brassinosteroids affect biosynthetic pathway of C45–C55 polyprenols.

Enhanced Cd phytoextraction by rapeseed under future climate as a consequence of higher sensitivity of HMA genes and better photosynthetic performance

This study aimed to investigate the underlying physiological, biochemical, and molecular mechanisms responsible for Brassica napu's potential to remediate Cd-contaminated soil under current (CC) vs. future (FC) climate (400 vs. 800 ppm of CO2, 21/14 °C vs. 25/18 °C). B. napus exhibited good tolerance to low Cd treatments (Cd-1, Cd-10, i.e., 1, 10 mg kg−1) under both climates without visible phytotoxicity symptoms. TI sharply decreased by 47 % and 68 % (p < 0.05), respectively, in Cd-50 and Cd-100 treated shoots under CC, but to a lesser extent (−26 % and −53 %, p < 0.05) under FC. This agreed with increased photosynthetic apparatus performance under FC, primarily due to a significant decrease in the closure of active PSII RCs ((dV/dt)o, TRo/RC) and less dissipated excitation energy (DIo/RC, φDo). Calvin Benson cycle-related enzyme activity also improved under FC with 2.2-fold and 2.4-fold (p < 0.05) increases in Rubisco and TPI under Cd-50 and Cd-100, respectively. Consequentially, a 2.2-fold and 2.3-fold (p < 0.05) boosted Pr resulted in a 2.3-fold and 2.4-fold (p < 0.05) increase in the DW of Cd-50 and Cd-100 treated shoots, respectively. This also led to a decrease (26 %, p < 0.05) in shoot Cd concentration under both high Cd treatments with a slight reduction in BCF. Translocation factor (TF) decreased (on average 42 %, p < 0.05) by high Cd treatments under both climates. However, under Cd-100, FC increased TF by 1.7-fold (p < 0.05) compared to CC, which could be explained by significant increases in the expression of HMA genes, especially BnaHMA4a and BnaHMA4c. Finally, Cd TU increased under FC by 65 % and 76 % (p < 0.05) under Cd-50 and Cd-100. This led to a shorter hypothetical remediation time for reaching the Cd pollution limit by 35 (p > 0.05) and 61 (p < 0.05) years, respectively, compared to CC.

Comparative Evaluation of the Effect of Copper Oxide and Copper Sulfate Nanoparticles on Structural and Functional Characteristics of Thalassiosira weissflogii under Conditions of Enrichment Cultivation // Russian Journal of Plant Physiology.

The effect of copper oxide nanoparticles (500, 1000, 2000 μg/L) and copper ions (37.5, 75, 150 μg/L) on the morphological, structural-functional, and fluorescent parameters of diatom microalgae Thalassiosira weissflogii under conditions of long-term cultivation was studied. It was shown that copper in ionic form is more toxic for this microalgae species than in the form of a nanooxide. The nature of the effect of copper in ionic and nanoform on cells Thalassiosira weissflogii has both similarities and significant differences. The difference in the effect of the studied toxicants on the growth of algae and the performance of photosynthetic apparatus was revealed. The addition of copper ions into the cultivation medium led to long lag phase during the initial stage of the experiment, followed by the restoration of growth on days 3–6 of exposure. An increase in the concentration of copper oxide nanoparticles in the medium caused inhibition of algae growth without a pronounced lag phase. With an increase in the concentration of Cu2+ in the culture medium, a decrease in the quantum efficiency of light energy utilization by the PS II photosynthetic complex and the maximum relative electron transport rate were observed, while copper oxide nanoparticles had an insignificant effect on the efficiency of the photosynthetic apparatus ofThalassiosira weissflogii. With an increase in the concentration of copper, both in the ionic form and in the form of nanooxide, increase in cell volume, changes of cell shape and granularity, an increase in the production of reactive oxygen species, suppression of enzymatic activity and a decrease in autofluorescence values of individual cells in the red region of the spectrum were observed.

Manipulation of light spectrum can improve the performance of photosynthetic apparatus of strawberry plants growing under salt and alkalinity stress.

Strawberry is one of the plants sensitive to salt and alkalinity stress. Light quality affects plant growth and metabolic activities. However, there is no clear answer in the literature on how light can improve the performance of the photosynthetic apparatus of this species under salt and alkalinity stress. The aim of this work was to investigate the effects of different spectra of supplemental light on strawberry (cv. Camarosa) under salt and alkalinity stress conditions. Light spectra of blue (with peak 460 nm), red (with peak 660 nm), blue/red (1:3), white/yellow (1:1) (400–700 nm) and ambient light were used as control. There were three stress treatments: control (no stress), alkalinity (40 mM NaHCO3), and salinity (80 mM NaCl). Under stress conditions, red and red/blue light had a positive effect on CO2 assimilation. In addition, blue/red light increased intrinsic water use efficiency (WUEi) under both stress conditions. Salinity and alkalinity stress decreased OJIP curves compared to the control treatment. Blue light caused an increase in its in plants under salinity stress, and red and blue/red light caused an increase in its in plants under alkalinity. Both salt and alkalinity stress caused a significant reduction in photosystem II (PSII) performance indices and quantum yield parameters. Adjustment of light spectra, especially red light, increased these parameters. It can be concluded that the adverse effects of salt and alkalinity stress on photosynthesis can be partially alleviated by changing the light spectra.

Photosynthetic apparatus performance and anatomical modulations of Alcantarea imperialis (Bromeliaceae) exposed to selenium during in vitro growth

Elements not usually included in culture medium formulations, such as selenium (Se), may have beneficial effects on micropropagated plants. We evaluated the effects of Se on the physiological and anatomical responses of Alcantarea imperialis during in vitro culture. Plants were cultured in a medium containing a gradient of Se concentrations (0, 4, 8, 16, or 32 &micro;M Se). After 56 d, the growth traits, chlorophyll a fluorescence, and root and leaf anatomy were analyzed. The fresh mass declined at the highest Se concentration. Higher Se concentrations induced bigger stomata, while the stomatal density decreased. Plants cultured with Se had improved PSII and PSI electron transport. This led to higher values of the total performance index. Thus, Se-induced plants showed a higher electron transport dynamics and energy conservation from water to PSI and developed anatomical traits that can favor tolerance to water deficit.

Photosynthetic Efficiency and Anatomical Structure of Pepper Leaf (Capsicum annuum L.) Transplants Grown under High-Pressure Sodium (HPS) and Light-Emitting Diode (LED) Supplementary Lighting Systems.

The aim of this study was to evaluate the effects of various supplemental greenhouse lighting systems, i.e., high-pressure sodium lamps and mixtures of red and blue light-emitting diodes, on the photochemical efficiency, anatomical leaf structure, and growth of the two pepper cultivars. The intensity levels of the photosynthetically active radiation were the same for both light treatments. In this study, the relative chlorophyll content was measured. Additionally, certain parameters of chlorophyll a fluorescence were measured under ambient light or after dark adaptation. The obtained results showed that the application of light-emitting diodes (LEDs) as supplemental lighting positively affected the anatomical leaf characteristics and plant growth. The leaves of both pepper cultivars were thicker and had larger palisade parenchyma cells under LED supplemental lighting compared to leaves grown under high-pressure sodium (HPS) lamps. Moreover, the mesophyll cells of seedlings grown under LEDs contained more chloroplasts than those growing under HPS lighting. The chlorophyll a fluorescence measurements of pepper seedlings grown under LEDs showed significant increases in photosynthetic apparatus performance index (PI) values compared to plants grown under HPS lamps; however, the values for this index were higher in cv. ‘Aifos’ as compared to cv. ‘Palermo’. We recommend that supplemental lighting systems are applied with caution, as their performance appears to depend not only on the light spectrum but also on the cultivar.

Blue Light Improves Photosynthetic Performance during Healing and Acclimatization of Grafted Watermelon Seedlings.

To investigate the importance of light on healing and acclimatization, in the present study, grafted watermelon seedlings were exposed to darkness (D) or light, provided by blue (B), red (R), a mixture of R (68%) and B (RB), or white (W; 35% B, 49% intermediate spectra, 16% R) LEDs for 12 days. Survival ratio, root and shoot growth, soluble carbohydrate content, photosynthetic pigments content, and photosynthetic performance were evaluated. Seedling survival was not only strongly limited in D but the survived seedlings had an inferior shoot and root development, reduced chlorophyll content, and attenuated photosynthetic efficiency. RB-exposed seedlings had a less-developed root system. R-exposed seedlings showed leaf epinasty, and had the smallest leaf area, reduced chlorophyll content, and suppressed photosynthetic apparatus performance. The R-exposed seedlings contained the highest amount of soluble carbohydrate and together with D-exposed seedlings the lowest amount of chlorophyll in their scions. B-exposed seedlings showed the highest chlorophyll content and improved overall PSII photosynthetic functioning. W-exposed seedling had the largest leaf area, and closely resembled the photosynthetic properties of RB-exposed seedlings. We assume that, during healing of grafted seedlings monochromatic R light should be avoided. Instead, W and monochromatic B light may be willingly adopted due to their promoting effect on shoot, pigments content, and photosynthetic efficiency.

Photosynthetic apparatus performance of tomato seedlings grown under various combinations of LED illumination.

It is already known that the process of photosynthesis depends on the quality and intensity of light. However, the influence of the new light sources recently used in horticulture, known as Light Emitting Diodes (LEDs), on this process is not yet fully understood. Chlorophyll a fluorescence measurement has been widely used as a rapid, reliable, and noninvasive tool to study the efficiency of the photosystem II (PSII) and to evaluate plant responses to various environmental factors, including light intensity and quality. In this work, we tested the responses of the tomato photosynthetic apparatus to different light spectral qualities. Our results showed that the best performance of the photosynthetic apparatus was observed under a mixture of red and blue light (R7:B3) or a mixture of red, green and blue light (R3:G2:B5). This was demonstrated by the increase in the effective photochemical quantum yield of PSII (Y[II]), photochemical quenching (qP) and electron transport rate (ETR). On the other hand, the mixture of red and blue light with a high proportion of blue light led to an increase in non-photochemical quenching (NPQ). Our results can be used to improve the production of tomato plants under artificial light conditions. However, since we found that the responses of the photosynthetic apparatus of tomato plants to a particular light regime were cultivar-dependent and there was a weak correlation between the growth and photosynthetic parameters tested in this work, special attention should be paid in future research.

Photosynthetic activity and chlorophyll pigment concentration in Medicago x varia T. Martyn leaves treated with the Tytanit growth regulator.

The purpose of the research was to determine the effect of the foliar use of a growth regulator with the trade name of Tytanit, containing titanium ascorbate, on photosynthetic activity and chlorophyll content in Medicago × varia T. Martyn leaves. There were two kinds of plots: C – control series; Ti – plants treated with Tytanit, containing 8.5 g of titanium in 1 dm3. The following parameters were determined: maximum photosystem II efficiency (Fv/Fm) in a dark-adapted state, actual photosystem II efficiency (ΔF/Fm’) in a light-adapted state, photochemical quenching factor (QP), non-photochemical quenching factor (QN), and chlorophyll a and b content. The Fisher-Snedecor test was used to determine whether the impact of experimental factors was significant, and the HSD 0.05 value was calculated using Tukey’s test. Compared to control, the photosynthetic apparatus performance of alfalfa was positively affected by the regulator compared to control. Tytanit applied to plant leaves increased their photosynthetic activity as a result of an increase in the content of chlorophyll pigments. It was also found that periods of rainfall deficiency did not affect the beneficial effects of the regulator.

Effects of phosphorus deficiency on the absorption of mineral nutrients, photosynthetic system performance and antioxidant metabolism in Citrus grandis.

Phosphorus (P) is an essential macronutrient for plant growth, development and production. However, little is known about the effects of P deficiency on nutrient absorption, photosynthetic apparatus performance and antioxidant metabolism in citrus. Seedlings of ‘sour pummelo’ (Citrus grandis) were irrigated with a nutrient solution containing 0.2 mM (Control) or 0 mM (P deficiency) KH2PO4 until saturated every other day for 16 weeks. P deficiency significantly decreased the dry weight (DW) of leaves and stems, and increased the root/shoot ratio in C. grandis but did not affect the DW of roots. The decreased DW of leaves and stems might be induced by the decreased chlorophyll (Chl) contents and CO2 assimilation in P deficient seedlings. P deficiency heterogeneously affected the nutrient contents of leaves, stems and roots. The analysis of Chl a fluorescence transients showed that P deficiency impaired electron transport from the donor side of photosystem II (PSII) to the end acceptor side of PSI, which showed a greater impact on the performance of the donor side of PSII than that of the acceptor side of PSII and photosystem I (PSI). P deficiency increased the contents of ascorbate (ASC), H2O2 and malondialdehyde (MDA) as well as the activities of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) in leaves. In contrast, P deficiency increased the ASC content, reduced the glutathione (GSH) content and the activities of SOD, CAT, APX and monodehydroascorbate reductase (MDHAR), but did not increase H2O2 production, anthocyanins and MDA content in roots. Taking these results together, we conclude that P deficiency affects nutrient absorption and lowers photosynthetic performance, leading to ROS production, which might be a crucial cause of the inhibited growth of C. grandis.

Special issue in honour of Prof. Reto J. Strasser - Can we predict winter survival in plants using chlorophyll a fluorescence?

In the last years, JIP-test became to be a tool widely applied to assess the performance of photosynthetic apparatus of plants growing under environmental stresses. The objective of our work was to check whether JIP-test can help to predict winter survival in plants. An experiment with outdoor vertical garden was conducted in June 2015 on a south-oriented wall in Lublin city, Poland. Plants were cultivated in pockets made of polyester felt and irrigated by automatic controlled system. After winter period (2015/2016) only 16 species and cultivars from 23 initially planted taxa substantiated successful survival. Chlorophyll fluorescence measurements were performed twice in 2015 and six times in 2016. Survival rate of examined species did not show any significant correlation with performance indices of PSII (PIABS and PItotal) and parameters related to quantum yields. On the other hand, changes of some parameters related to specific energy fluxes per active reaction centres were found to be connected, to some extent, to plants winter survival. These changes could play a key role in plants ability to survive winter conditions.

Differences in the functioning of photosynthetic electron transport between metallicolous and non-metallicolous populations of the pseudometallophyte Viola tricolor

The objective of this study was to assess the effect of metalliferous conditions on the functioning of photosynthetic electron transport in waste heap populations of a pseudometallophyte, Viola tricolor L. Measurements of chlorophyll a fluorescence and the absorbance changes at 830 nm enabled a non-invasive assessment of photosynthetic apparatus performance. This was complemented by the evaluation of the chlorophyll content. Low temperature chlorophyll fluorescence emission spectra were also recorded. Based on the OJIP test performed in situ, we demonstrated a disturbed condition of photosystem II (PSII) in three metalliferous populations in comparison with a non-metallicolous one. The combined effects of elevated concentrations of zinc, cadmium and lead in soil resulted in the decline of some parameters describing the efficiency and electron flow through PSII. The differences between waste heap populations seemed to be partly correlated with the concentration of heavy metals in the soil. The characteristic of electron transport at photosystem I (PSI) in the light-adapted state revealed increased values of PSI donor-side limitation (YND) and a declined PSI quantum efficiency (YI). It was also demonstrated that the waste heap conditions negatively affect the total chlorophyll content in leaves and led to an increased ratio of fluorescence emission at 77 K (F730/F685). The obtained data indicate that, regardless of the high adaptation of metallicolous populations, photosynthetic electron transport is hampered in V. tricolor plants at metal polluted sites.

Morphophysiological responses of Billbergia zebrina Lindl. (Bromeliaceae) in function of types and concentrations of carbohydrates during conventional in vitro culture

Abstract When propagated in vitro, explants receive all the nutrients needed for their growth, including carbohydrates, from the culture medium. However, it is not well understood how the type and concentration of carbohydrates can affect the functioning of the photosynthetic apparatus (particularly photosystem II) of these plants. The aim was to assess the morphophysiological responses of Billbergia zebrina plants in function of sources and concentrations of carbohydrates during in vitro culture. Side shoots of plants previously established in vitro were individualized and transferred to a culture medium containing fructose, glucose or sucrose in four concentrations (0, 15, 30 or 45 g L−1). After growth for 55 days, the chlorophyll a fluorescence transient, leaf anatomy and growth were analyzed. The concentration and type of carbohydrate employed during in vitro culture did not decrease the photosynthetic apparatus performance. However, concentrations above 30 g L−1 led to anatomical modifications, revealing some degree of stress suffered by the plants. When grown in concentrations of 15 and 30 g L−1, irrespective of the carbohydrate used, the plants presented greater stomatal density. The supplementation of the culture medium with monosaccharides caused alterations in the development of the xylem vessels, such as increased number and diameter, allowing adjustment to the microenvironmental conditions. The in vitro conditions influenced the photosynthetic and anatomical responses of plants. The concentration interval from 15 to 30 g L−1 sucrose had a better effect by not causing large changes in the performance of the photosynthetic apparatus and anatomy of plants.

Efficiency of the photosynthetic apparatus of Myriophyllum spicatum L. under anthropopression – a case study from Upper Silesian

The current type of global economy and environmental management requires a change in its approach to the use of natural resources in order to meet human needs. A system approach and an analysis of the importance of anthropogenic ecosystems appears to be an effective tool for sustainable development of urban and post-industrial areas. One example of anthropogenic ecosystems are mining water reservoirs, formed in the basins of subsidence, affecting the increase of biodiversity in the area poor in the surface water reservoirs. This study presents analysis of the impact of water quality in anthropogenic reservoirs on the Myriophyllum spicatum L. photosynthetic apparatus efficiency. Five reservoirs with different water quality were analysed. The photosynthetic apparatus performance of macrophytes was measured using the chlorophyll fluorescence signal through a Pocket PEA fluorimeter. The results showed that electron transport decreased with deteriorating water quality of reservoirs. This leads to a weakening of the population of Myriophyllum spicatum, which, as a further consequence, may lead to an acceleration of eutrophication of reservoirs.

Effectiveness of 2-Hydroxybenzoic Acid in Physiologic Response of Basket Willow (Salix viminalis L.) on Stress Induced by Cadmium

The aim of the study was an evaluation of the effect of exogenous application of 2-hydroxybenzoic acid on the performance of photosynthetic apparatus of two basket willow (Salix viminalis L.) varieties growing in elevated concentrations of cadmium salts and an evaluation of the suitability of fluorescence method to characterize the physiological state of the examined willow genotypes under stress conditions caused by an increased content of cadmium ions. The influence of cadmium ions at a concentration of 50 mg/dm3 and salicylic acid at a concentration of 138 mg/dm3 on the physiological reaction of basket willow of Bjor and Tora varieties cultivated in hydroponics was investigated. The applied dose of salicylic acid effectively eliminated the influence of cadmium, activating the synthesis of chlorophyll a and b and carotenoids as well as increasing the intensity of the determined fluorescence processes and indicators. The use of the fluorescent method to characterize the physiological state of the examined willow genotypes under the conditions of stress caused by an increased content of cadmium ions allows a better understanding of the immune system’s response to these metal ions.

Exploration of Chlorophyll a Fluorescence and Plant Gas Exchange Parameters as Indicators of Drought Tolerance in Perennial Ryegrass.

Perennial ryegrass (Lolium perenne L.) belongs to the common cultivated grass species in Central and Western Europe. Despite being considered to be susceptible to drought, it is frequently used for forming the turf in urban green areas. In such areas, the water deficit in soil is recognized as one of the most important environmental factors, which can limit plant growth. The basic aim of this work was to explore the mechanisms standing behind the changes in the photosynthetic apparatus performance of two perennial ryegrass turf varieties grown under drought stress using comprehensive in vivo chlorophyll fluorescence signal analyses and plant gas exchange measurements. Drought was applied after eight weeks of sowing by controlling the humidity of the roots ground medium at the levels of 30, 50, and 70% of the field water capacity. Measurements were carried out at four times: 0, 120, and 240 h after drought application and after recovery (refilling water to 70%). We found that the difference between the two tested varieties’ response resulted from a particular re-reduction of P700+ (reaction certer of PSI) that was caused by slower electron donation from P680. The difference in the rate of electron flow from Photosystem II (PSII) to PSI was also detected. The application of the combined tools (plants’ photosynthetic efficiency analysis and plant gas exchange measurements) allowed exploring and explaining the specific variety response to drought stress.

JIP-test in assessing sensitivity to nitrogen deficiency in two cultivars of Actinidia arguta (Siebold et Zucc.) Planch. ex Miq.

Optimal nitrogen availability is necessary to keep high photosynthetic efficiency and in consequence, good physiological condition and growth of plants. In order to examine the effect of N nutrition on photosynthetic apparatus performance we used the chlorophyll a fluorescence (ChF) technique in two differing Actinidia cultivars and we aimed to select the most reliable ChF parameters showing N deficiency. The 4-years-old specimens growing on an experimental plantation were treated with three differing N doses and ChF data were collected during two years. The results showed that PSII performance indices, performance index on the absorption basis and total performance index, are useful to detect the overall decrease in photosynthetic apparatus vitality. Efficiency of electron movement beyond QA into the electron transport chain was more susceptible to different N nutritions than maximum quantum efficiency, while a pool of QA reducing reaction centres per PSII antenna chlorophyll, RC/ABS, and a number of QA reducing reaction centres per cross section of a sample, RC/CS0, were the best indicators of N deficiency.

Extended temperature optimum of photosynthetic reaction centers in Rhodobacterales

Temperature is one of the most important physical factors affecting microbial and biochemical processes. We investigated the performance of photosynthetic apparatus of marine photoheterotrophic bacterium Dinoroseobacter shibae under various temperatures. The primary photochemistry and electron transport was measured using variable infra-red fluorometry in the cells grown between 8-35°C. It was found that the photosynthetic electron transport had a broad temperature optimum between 25-50°C. Moreover, the primary charge separation stayed functional even after rising temperature up to 55°C. The same phenomenon was observed also in other phototrophic Rhodobacterales. The psychrotolerant bacterium Roseisalinus antarcticus reached its maximum electron transport rate at 48°C, 30°C above its growth temperature. We propose that the extended temperature stability may be crucial to maintain photosynthetic function under situation when photosynthetic membranes heat up above their ambient temperature due to the heat dissipation of the excess light energy.
