Self-Assembly-Directed Organization of a Fullerene-Bisporphyrin into Supramolecular Giant Donut Structures for Excited-State Charge Stabilization.

Functional materials composed of spontaneously self-assembled electron donor and acceptor entities capable of generating long-lived charge-separated states upon photoillumination are in great demand as they are key in building the next generation of light energy harvesting devices. However, creating such well-defined architectures is challenging due to the intricate molecular design, multistep synthesis, and issues associated in demonstrating long-lived electron transfer. In this study, we have accomplished these tasks and report the synthesis of a new fullerene–bis-Zn-porphyrin e-bisadduct by tether-directed functionalization of C60 via a multistep synthetic protocol. Supramolecular oligomers were subsequently formed involving the two porphyrin-bearing arms embracing a fullerene cage of the vicinal molecule as confirmed by MALDI-TOF spectrometry and variable temperature NMR. In addition, the initially formed worm-like oligomers are shown to evolve to generate donut-like aggregates by AFM monitoring that was also supported by theoretical calculations. The final supramolecular donuts revealed an inner cavity size estimated as 23 nm, close to that observed in photosynthetic antenna systems. Upon systematic spectral, computational, and electrochemical studies, an energy level diagram was established to visualize the thermodynamic feasibility of electron transfer in these donor–acceptor constructs. Subsequently, transient pump–probe spectral studies covering the wide femtosecond-to-millisecond time scale were performed to confirm the formation of long-lived charge-separated states. The lifetime of the final charge-separated state was about 40 μs, thus highlighting the significance of the current approach of building giant self-organized donor–acceptor assemblies for light energy harvesting applications.

Self-aggregation of Synthetic 20-O-Substituted Bacteriochlorophyll-d Analogs

Zinc 20-acyloxy-bacteriochlorophyll-d analogs were prepared from naturally occurring chlorophyll(Chl)-a. The semi-synthetic Chl-a derivatives self-aggregated in an aqueous Triton X-100 micelle solution to give red-shifted and broadened visible absorption bands, similarly to native chlorosome, one of the photosynthetic light-harvesting antenna systems. The present compounds are the first examples of a 20-O-substituted chlorosomal pigment model.

Redox-state dependent blinking of single photosystem I trimers at around liquid-nitrogen temperature

Efficient light harvesting in a photosynthetic antenna system is disturbed by a ragged and fluctuating energy landscape of the antenna pigments in response to the conformation dynamics of the protein. This situation is especially pronounced in Photosystem I (PSI) containing red shifted chlorophylls (red Chls) with the excitation energy much lower than the primary donor. The present study was conducted to clarify light-harvesting dynamics of PSI isolated from Synechocystis sp. PCC6803 by using single-molecule spectroscopy at liquid‑nitrogen temperatures. Fluorescence emission at around 720 nm from the red Chls in single PSI trimers was monitored at 80–100 K. Intermittent variations in the emission intensities, so-called blinking, were frequently observed. Its time scale lay in several tens of seconds. The blinking amplitude depended on the redox state of the phylloquinone (A1). Electrochromic shifts of Chls induced by the negative charge on A1 were calculated based on the X-ray crystallographic structure. A Chl molecule, Chl-A839 (numbering according to PDB 5OY0), bound near A1 was found to have a large electrochromic shift. This Chl has strong exciton coupling with neighboring Chl (A838) whose site energy was predicted to be determined by interaction with an arginine residue (ArgF84) [Adolphs et al., 2010]. A possible scenario of the blinking was proposed. Conformational fluctuations of ArgF84 seesaw the excitation-energy of Chl-A838, which perturbs the branching ratio of excitation-energy between the red Chl and the cationic form of P700 as a quencher. The electrochromic shift of Chl-A839 enhances the effect of the conformation dynamics of ArgF84.

Quantitative modeling of energy dissipation in Arabidopsis thaliana

In photosynthesis, solar energy is absorbed and converted into chemical energy. Chlorophyll embedded in proteins absorb light and transfer excitation energy to reaction centers where charge separation occurs. However, the solar flux incident on photosynthetic organisms is highly variable, requiring complex feedback systems to regulate the excitation pressure on reaction centers and prevent excess absorbed energy from causing damage. During periods of transient high light, excess absorbed energy is dissipated as heat. This is routinely observed as the quenching of chlorophyll fluorescence, and often broadly referred to as non-photochemical quenching (NPQ). Understanding the mechanisms through which photosynthetic systems dissipate excess energy and regulate excitation pressure in response to variable light conditions requires extensive quantitative modeling of the photosynthetic system and energy dissipation to interpret experimental observations. This review discusses efforts to model energy dissipation, or quenching, in Arabidopsis thaliana and their connections to models of regulatory systems that control quenching. We begin with a review of theory used to describe energy transfer and experimental data obtained to construct energy transfer models of the photosynthetic antenna system that underlie the interpretation of chlorophyll fluorescence quenching. Second, experimental evidence leading to proposed molecular mechanisms of quenching and the implications for modeling are discussed. The initial incorporation of depictions of proposed mechanisms into quantitative energy transfer models is reviewed. Finally, the necessity of connecting energy transfer models that include molecular models of quenching mechanisms with regulatory models is discussed.

Characterization of a newly isolated freshwater Eustigmatophyte alga capable of utilizing far-red light as its sole light source.

Oxygenic phototrophs typically utilize visible light (400-700nm) to drive photosynthesis. However, a large fraction of the energy in sunlight is contained in the far-red region, which encompasses light beyond 700nm. In nature, certain niche environments contain high levels of this far-red light due to filtering by other phototrophs, and in these environments, organisms with photosynthetic antenna systems adapted to absorbing far-red light are able to thrive. We used selective far-red light conditions to isolate such organisms in environmental samples. One cultured organism, the Eustigmatophyte alga Forest Park Isolate 5 (FP5), is able to absorb far-red light using a chlorophyll (Chl) a-containing antenna complex, and is able to grow under solely far-red light. Here we characterize the antenna system from this organism, which is able to shift the absorption of Chl a to >705nm.

Dephasing Times in the Phycobiliprotein Antenna Complexes PE545 and PE555

Since long-lived quantum coherence have not only been observed in the Fenna-Matthews-Olson (FMO) complex of green sulfur bacteria but also in the phycoerythrin 545 (PE545) photosynthetic antenna system, the interest in these systems has been increased significantly. Moreover, similar results have been found for the PE555 complex in which the two alpha-beta monomers are rotated compared to the PE545 structure leading to a water filled channel. By now, all three complexes have been investigated by a sequential combination of molecular dynamics (MD) simulations, calculations of vertical excitation energies along the trajectory and quantum dynamics for the exciton transfer [1-3]. A comparison of the obtained spectral densities and dynamics will be provided. An interesting property for the systems is the dephasing time which can give an estimate for how long quantum coherences might survive in the respective system. In a first step, the relationship between dephasing time and energy gap fluctuations of the individual pigments has been determined [4]. For the above mentioned complex but also additional exciton and charge transfer systems it can be confirmed that that an inverse proportionality exists between dephasing time and average gap energy fluctuation. Interestingly, an interestingly very similar behavior has been found for all these systems. In a subsequent step, the relationship between dephasing time and excitonic energy gap fluctuations for entire complexes including the respective inter-molecular couplings has been studied.[1] C. In situ high-resolution structure of the baseplate antenna complex in Chlorobaculum tepidum.

Photosynthetic antenna systems enable organisms harvesting light and transfer the energy to the photosynthetic reaction centre, where the conversion to chemical energy takes place. One of the most complex antenna systems, the chlorosome, found in the photosynthetic green sulfur bacterium Chlorobaculum (Cba.) tepidum contains a baseplate, which is a scaffolding super-structure, formed by the protein CsmA and bacteriochlorophyll a. Here we present the first high-resolution structure of the CsmA baseplate using intact fully functional, light-harvesting organelles from Cba. tepidum, following a hybrid approach combining five complementary methods: solid-state NMR spectroscopy, cryo-electron microscopy, isotropic and anisotropic circular dichroism and linear dichroism. The structure calculation was facilitated through development of new software, GASyCS for efficient geometry optimization of highly symmetric oligomeric structures. We show that the baseplate is composed of rods of repeated dimers of the strongly amphipathic CsmA with pigments sandwiched within the dimer at the hydrophobic side of the helix.

Structure of Light-Harvesting Aggregates in Individual Chlorosomes.

Among all photosynthetic organisms, green bacteria have evolved one of the most efficient light-harvesting antenna, the chlorosome, that contains hundreds of thousands of bacteriochlorophyll molecules, allowing these bacteria to grow photosynthetically by absorbing only a few photons per bacteriochlorophyll molecule per day. In contrast to other photosynthetic light-harvesting antenna systems, for which a protein scaffold imposes the proper positioning of the chromophores with respect to each other, in chlorosomes, this is accomplished solely by self-assembly. This has aroused enormous interest in the structure-function relations of these assemblies, as they can serve as blueprints for artificial light harvesting systems. In spite of these efforts, conclusive structural information is not available yet, reflecting the sample heterogeneity inherent to the natural system. Here we combine mutagenesis, polarization-resolved single-particle fluorescence-excitation spectroscopy, cryo-electron microscopy, and theoretical modeling to study the chlorosomes of the green sulfur bacterium Chlorobaculum tepidum. We demonstrate that only the combination of these techniques yields unambiguous information on the structure of the bacteriochlorophyll aggregates within the chlorosomes. Moreover, we provide a quantitative estimate of the curvature variation of these aggregates that explains ongoing debates concerning the chlorosome structure.

A combination of classical molecular dynamics simulations, quantum chemistry and quantum dynamical calculations is employed to determine the excitation transfer dynamics in PE545. One key property of the light-harvesting system concerning the excitation transfer and dephasing phenomena is the spectral density. This quantity is determined from time series of the vertical excitation energies of the aggregate. In the present study we focus on the quantum dynamical simulations using the earlier QM/MM calculations as input. Employing an ensemble-averaged classical path-based wave packet dynamics, the excitation transfer dynamics between the different bilins in the PE545 complex is determined and analyzed. Furthermore, the nature of the environmental fluctuations determining the transfer dynamics is discussed.

Different Types of Vibrations Interacting with Electronic Excitations in Phycoerythrin 545 and Fenna-Matthews-Olson Antenna Systems.

The interest in the phycoerythrin 545 (PE545) photosynthetic antenna system of marine algae and the Fenna-Matthews-Olson (FMO) complex of green sulfur bacteria has drastically increased since long-lived quantum coherences were reported for these complexes. For the PE545 complex, this phenomenon is clearly visible even at ambient temperatures, while for the FMO system it is more prominent at lower temperatures. The key to elucidate the role of the environment in these long-lived quantum effects is the spectral density. Here, we employ molecular dynamics simulations combined with quantum chemistry calculations to study the coupling between the biological environment and the vertical excitation energies of the bilin pigment molecules in PE545 and compare them to prior calculations on the FMO complex. It is found that the overall strength of the resulting spectral densities for the PE545 system is similar to the experiment-based counterpart but also to those in the FMO complex. Molecular analysis, however, reveals that the origin for the spectral densities in the low frequency range, which is most important for excitonic transitions, is entirely different. In the case of FMO, this part of the spectral density is due to environmental fluctuations, while, in case of PE545, it is essentially only due to internal modes of the bilin molecules. This finding sheds new light on possible explanations of the long-lived quantum coherences and that the reasons might actually be different in dissimilar systems.

The site of regulation of light capture in Symbiodinium: Does the peridinin–chlorophyll a–protein detach to regulate light capture?

Dinoflagellates from the genus Symbiodinium form symbiotic associations with cnidarians including corals and anemones. The photosynthetic apparatuses of these dinoflagellates possess a unique photosynthetic antenna system incorporating the peridinin–chlorophyll a–protein (PCP). It has been proposed that the appearance of a PCP-specific 77K fluorescence emission band around 672–675nm indicates that high light treatment results in PCP dissociation from intrinsic membrane antenna complexes, blocking excitation transfer to the intrinsic membrane-bound antenna complexes, chlorophyll a–chlorophyll c2–peridinin–protein-complex (acpPC) and associated photosystems (Reynolds et al., 2008 Proc Natl Acad Sci USA 105:13674–13678).We have tested this model using time-resolved fluorescence decay kinetics in conjunction with global fitting to compare the time-evolution of the PCP spectral bands before and after high light exposure. Our results show that no long-lived PCP fluorescence emission components appear either before or after high light treatment, indicating that the efficiency of excitation transfer from PCP to membrane antenna systems remains efficient and rapid even after exposure to high light. The apparent increased relative emission at around 675nm was, instead, caused by strong preferential exciton quenching of the membrane antenna complexes associated with acpPC and reaction centers. This strong non-photochemical quenching (NPQ) is consistent with the activation of xanthophyll-associated quenching mechanisms and the generally-observed avoidance in nature of long-lived photoexcited states that can lead to oxidative damage. The acpPC component appears to be the most strongly quenched under high light exposure suggesting that it houses the photoprotective exciton quencher.

Strategy of ring-shaped aggregates in excitation energy transfer for removing disorder-induced shielding

Peripheral light harvesting complex (LH2), which is found in photosynthetic antenna systems of purple photosynthetic bacteria, has important functions in the photosynthetic process, such as harvesting sunlight and transferring its energy to the photosynthetic reaction center. The key component in excitation energy transfer (EET) between LH2s is B850, which is a characteristic ring-shaped aggregate of pigments usually formed by 18 or 16 bacteriochlorophylls in LH2. We theoretically study the strategy of the ring-shaped aggregate structure, which maximizes EET efficiency, by using the standard Frenkel exciton model and the self-consistent calculation method for the Markovian quantum master equation and Maxwell equation. As a result, we have revealed a simple but ingenious strategy of the ring-shaped aggregate structure. The combination of three key properties of the ring unit system maximizes the EET efficiency, namely the large dipole moment of aggregates causes the basic improvement of EET efficiency, and the isotropic nature and the large occupying area are critically effective to remove the disorder-induced shielding that inhibits EET in the presence of the randomness of orientation and alignment of carriers of excitation energy.

Memory-Assisted Exciton Diffusion in the Chlorosome Light-Harvesting Antenna of Green Sulfur Bacteria.

Chlorosomes are likely the largest and most efficient natural light-harvesting photosynthetic antenna systems. They are composed of large numbers of bacteriochlorophylls organized into supramolecular aggregates. We explore the microscopic origin of the fast excitation energy transfer in the chlorosome using the recently resolved structure and atomistic-detail simulations. Despite the dynamical disorder effects on the electronic transitions of the bacteriochlorophylls, our simulations show that the exciton delocalizes over the entire aggregate in about 200 fs. The memory effects associated to the dynamical disorder assist the exciton diffusion through the aggregates and enhance the diffusion coefficients as a factor of 2 as compared to the model without memory. Furthermore, exciton diffusion in the chlorosome is found to be highly anisotropic with the preferential transfer toward the baseplate, which is the next functional element in the photosynthetic system.

Computational methodologies and physical insights into electronic energy transfer in photosynthetic light-harvesting complexes

We examine computational techniques and methodologies currently in use to explore electronic excitation energy transfer in the context of light-harvesting complexes in photosynthetic antenna systems, and comment on some new insights into the underlying physics. Advantages and pitfalls of these methodologies are discussed, as are some physical insights into the photosynthetic dynamics. By combining results from molecular modelling of the complexes (structural description) with an effective non-equilibrium statistical description (time evolution), we identify some general features, regardless of the particular distribution in the protein scaffold, that are central to light-harvesting dynamics and, that could ultimately be related to the high efficiency of the overall process. Based on these general common features, some possible new directions in the field are discussed.

Two-Dimensional Spectroscopy Can Distinguish between Decoherence and Dephasing of Zero-Quantum Coherences

Recent experiments on a variety of photosynthetic antenna systems have revealed that coherences among electronic states persist longer than previously anticipated. In an ensemble measurement, the observed dephasing of a coherent state can occur because of either disorder across the ensemble or decoherence from interactions with the bath. Distinguishing how much such disorder affects the experimentally observed dephasing rate is paramount for understanding the role that quantum coherence may play in energy transfer through these complexes. Here, we show that two-dimensional electronic spectra can distinguish between the limiting cases of homogeneous dephasing (decoherence) and inhomogeneous dephasing by examining how the quantum beat frequency changes within a cross peak. For the antenna complex LH2 isolated from Rhodobacter sphaeroides , we find that dephasing of the coherence between the B850 and B800 rings arises predominantly from inhomogeneity. In contrast, within the Fenna-Matthews-Olson (FMO) complex from Chlorobium tepidum , dephasing of the coherence between the first two excitons appears quite homogeneous. Thus, the observed dephasing rate sets an upper bound on decoherence for the LH2 complex while establishing both an upper and lower bound for the FMO complex.

Selective Assembly of Photosynthetic Antenna Proteins into a Domain-Structured Lipid Bilayer for the Construction of Artificial Photosynthetic Antenna Systems: Structural Analysis of the Assembly Using Surface Plasmon Resonance and Atomic Force Microscopy

Two types of photosynthetic membrane proteins, the peripheral antenna complex (LH2) and the core antenna/reaction center complex (LH1-RC), play an essential role in the primary process of purple bacterial photosynthesis, that is, capturing light energy, transferring it to the RC where it is used in subsequent charge separation. Establishment of experimental platforms is required to understand the function of the supramolecular assembly of LH2 and LH1-RC molecules into arrays. In this study, we assembled LH2 and LH1-RC arrays into domain-structured planar lipid bilayers placed on a coverglass using stepwise combinations of vesicle-to-planar membrane formation and vesicle fusion methods. First, it was shown that assembly of LH2 and LH1-RC in planar lipid bilayers, through vesicle-to-planar membrane formation, could be confirmed by absorption spectroscopy and high resolution atomic force microscopy (AFM). Second, formation of a planar membrane incorporating LH2 molecules made by the vesicle fusion method was corroborated by AFM together with quantitative analysis by surface plasmon resonance (SPR). By combining planar membrane formation and vesicle fusion, in a stepwise manner, LH2 and LH1-RC were successfully organized in the domain-structured planar lipid membrane. This methodology for construction of LH2/LH1-RC assemblies will be a useful experimental platform with which to investigate energy transfer from LH2 to LH1-RC where the relative arrangement of these two complexes can be controlled.

The function of geometrical structure of B850 ring in bacterial photosynthetic antenna systems

AbstractA series of simulations are made to investigate the function of characteristic ring‐shaped structure of B850 ring in peripheral light harvesting complex (LH2) in typical purple photosynthetic bacteria. The results suggest that the geometrical structure of B850 ring positively functions to realize the high efficient excitation energy transfer (EET) in the photosynthetic antenna systems. It also shows the importance of geometrical structure of units in EET not only in photosynthetic systems but also any other systems containing randomness in distribution of units.
