Combined FCS and PCH Analysis to Quantify Protein Dimerization in Living Cells.

Protein dimerization plays a crucial role in the regulation of numerous biological processes. However, detecting protein dimers in a cellular environment is still a challenge. Here we present a methodology to measure the extent of dimerization of GFP-tagged proteins in living cells, using a combination of fluorescence correlation spectroscopy (FCS) and photon counting histogram (PCH) analysis of single-color fluorescence fluctuation data. We named this analysis method brightness and diffusion global analysis (BDGA) and adapted it for biological purposes. Using cell lysates containing different ratios of GFP and tandem-dimer GFP (diGFP), we show that the average brightness per particle is proportional to the fraction of dimer present. We further adapted this methodology for its application in living cells, and we were able to distinguish GFP, diGFP, as well as ligand-induced dimerization of FKBP12 (FK506 binding protein 12)-GFP. While other analysis methods have only sporadically been used to study dimerization in living cells and may be prone to errors, this paper provides a robust approach for the investigation of any cytosolic protein using single-color fluorescence fluctuation spectroscopy.

Fluorescence Correlation Spectroscopy and Photon Counting Histograms in Finite, Bounded Domains

Analysis of fluctuations arising as fluorescent particles pass through a focused laser beam has enabled quantitative characterization of a broad range of molecular kinetic processes. Two key mathematical frameworks that have enabled these quantifications are fluorescence correlation spectroscopy (FCS) and photon counting histogram (PCH) analysis. Although these frameworks are effective and accurate when the focused laser beam is well approximated by an infinite Gaussian beam with a waist that is small compared to the size of the region over which the fluorescent particles can diffuse, they cannot be applied to situations in which this region is bounded at the nanoscale. We therefore derived general forms of the FCS and PCH frameworks for bounded systems. The finite-domain form of FCS differs from the classical form in its boundary and initial conditions and requires development of a new Fourier space solution for fitting data. Our finite-domain FCS predicts simulated data accurately and reduces to a previous model for the special case when the system is much larger than the Gaussian beam and can be considered to be infinite. We also derived the PCH form for the bounded systems. Our approach enables estimation of the concentration of diffusing fluorophores within a finite domain for the first time, to our knowledge. The method opens the possibility of quantification of kinetics in several systems for which this has never been possible.

Scanning fluorescence correlation spectroscopy comes full circle.

In this article, we review the application of fluorescence correlation spectroscopy (FCS) methods to studies on live cells. We begin with a brief overview of the theory underlying FCS, highlighting the type of information obtainable. We then focus on circular scanning FCS. Specifically, we discuss instrumentation and data analysis and offer some considerations regarding sample preparation. Two examples from the literature are discussed in detail. First, we show how this method, coupled with the photon counting histogram analysis, can provide information on yeast ribosomal structures in live cells. The combination of scanning FCS with dual channel detection in the study of lipid domains in live cells is also illustrated.

Changes in Cholesterol Level Alter Integrin Sequestration in Raft-Mimicking Lipid Mixtures

The influence of cholesterol (CHOL) level on integrin sequestration in raft-mimicking lipid mixtures forming coexisting liquid-ordered (lo) and liquid-disordered (ld) lipid domains is investigated using complementary, single-molecule-sensitive, confocal detection methods. Systematic analysis of membrane protein distribution in such a model membrane environment demonstrates that variation of CHOL level has a profound influence on lo-ld sequestration of integrins, thereby exhibiting overall ld preference in the absence of ligands and lo affinity upon vitronectin addition. Accompanying photon-counting histogram analysis of integrins in the different model membrane mixtures shows that the observed changes of integrin sequestration in response to variations of membrane CHOL level are not associated with altering integrin oligomerization states. Instead, our experiments suggest that the strong CHOL dependence of integrin sequestration can be attributed to CHOL-mediated changes of lipid packing and bilayer thickness in coexisting lo and ld domains, highlighting the significance of a biophysical mechanism of CHOL-mediated regulation of integrin sequestration. We envision that this model membrane study may help clarify the influence of CHOL in integrin functionality in plasma membranes, thus providing further insight into the role of lipid heterogeneities in membrane protein distribution and function in a cellular membrane environment.

Fission yeast Myo2: Molecular organization and diffusion in the cytoplasm.

Myosin-II is required for the assembly and constriction of cytokinetic contractile rings in fungi and animals. We used electron microscopy, fluorescence recovery after photobleaching (FRAP), and fluorescence correlation spectroscopy (FCS) to characterize the physical properties of Myo2 from fission yeast Schizosaccharomyces pombe. By electron microscopy, Myo2 has two heads and a coiled-coiled tail like myosin-II from other species. The first 65 nm of the tail is a stiff rod, followed by a flexible, less-ordered region up to 30 nm long. Myo2 sediments as a 7 S molecule in high salt, but aggregates rather than forming minifilaments at lower salt concentrations; this is unaffected by heavy chain phosphorylation. We used FRAP and FCS to observe the dynamics of Myo2 in live S. pombe cells and in cell extracts at different salt concentrations; both show that Myo2 with an N-terminal mEGFP tag has a diffusion coefficient of ∼ 3 µm2 s-1 in the cytoplasm of live cells during interphase and mitosis. Photon counting histogram analysis of the FCS data confirmed that Myo2 diffuses as doubled-headed molecules in the cytoplasm. FCS measurements on diluted cell extracts showed that mEGFP-Myo2 has a diffusion coefficient of ∼ 30 µm2 s-1 in 50 to 400 mM KCl concentrations.

Distinct Effects of Multivalent Macroion and Simple Ion on the Structure and Local Electric Environment of a Weak Polyelectrolyte in Aqueous Solution.

Adding ionic species can critically affect the structure of weak polyelectrolyte (PE) chains, whose charge density in aqueous solution can be greatly regulated by bathing solution conditions such as pH and added ions. Distinct from simple ions that can be treated as point charges, multivalent macroions of finite size, including many charged nanoparticles and biopolymers, could show strong electrostatic coupling with PEs and effectively modify the conformation and assembly of PEs in aqueous solution. In this work, we have compared the effects of hydrophilic multivalent macroion of finite size and simple divalent ion on the conformational transition of a model weak polybase, poly(2-vinylpyridine) (P2VP), in dilute aqueous solution. By using fluorescence correlation spectroscopy combined with photon counting histogram analysis, we have examined the swollen-to-collapsed conformational transition and local electric potential of a P2VP chain in ionic aqueous solution at a single-molecule level. Adding inorganic polytungstate ([W12]) macroion bearing eight negative charges per [W12] of ∼0.8 nm in diameter at increased concentration from 10-9 to 10-5 mM can lead to a shift of the critical conformational transition pH, pHcr, of P2VP to lower pH values, in an opposite trend to the previously reported effect of adding simple monovalent anion. Conversely, adding simple divalent sulfate anion can lead to a nonmonotonic change of pHcr when increasing its concentration from 0.03 to 15 mM. Additionally, at pH < pHcr where P2VP is highly protonated and adopts a swollen conformation, a monotonic decrease of P2VP size is observed with increased sulfate ionic concentration, exhibiting the typical ionic screening effect. In contrast, the size of the P2VP chain shows little change with increasing [W12] concentration before the precipitation of P2VP from water. To investigate the distinct effects of multivalent ion and macroion on the conformational transition of P2VP in aqueous solution, we have also measured the local proton concentration in the vicinity to a P2VP chain by an attached pH-sensitive fluorescence probe. In both cases, we have observed the monotonic reduction of the local electric potential of a swollen P2VP chain with increased ionic concentration, despite the increased protonation degree of P2VP. The results suggest that counterion condensation of multivalent ion and macroion can modify the effective net charge density of P2VP chains in dilute aqueous solution. However, possibly due to its high multivalency and finite size, multivalent [W12] macroion is much more effective in modifying the local electric environment and structure of P2VP chains at 3-7 orders of magnitude lower concentrations than simple sulfate counterion.

Probing Cholesterol-Dependence of Integrin-Urokinase Receptor Complex Formation using Confocal Dual-Color Fluorescence Intensity Analysis

The modern view of a plasma membrane is that of a complex supramolecular assembly of notable lipid/protein heterogeneity, in which the local lipid environment may have a profound impact on membrane protein distribution and function. Yet, the underlying mechanisms of lipid-mediated regulation of membrane protein functionality remain elusive, in part due to the limitations of existing methodologies and the often small size and transient nature of lipid/protein heterogeneities in cellular membranes. Exemplary, we still have rather limited knowledge about the significance of lipid composition in hetero-protein complex formation in such membrane systems. To address this fascinating, but poorly understood topic, here we introduce a single molecule-sensitive confocal experimental strategy, comprised of dual-color fluorescence peak analysis and photon counting histogram (PCH) method, which not only allows the detection of hetero-protein complexes in well-defined lipid environments, but also provides accurate insight into the composition of such complexes in the membrane. By applying the described methodology, here we demonstrate the formation of complexes of αvβ3 integrins and GPI-anchored urokinase plasminogen activator receptors (uPAR) in cholesterol-containing model lipid mixtures. PCH analysis of fluorescence intensity signals of complexes confirms furthermore that αvβ3-uPAR complexes are comprised of one integrin and two uPAR proteins. Remarkably, formation of comparable hetero-protein complexes is largely suppressed in a cholesterol-free lipid composition, highlighting the importance of cholesterol in αvβ3-uPAR complex formation. Our results are intriguing in light of the already established significance of cholesterol in integrin function in cellular membranes.

The use of fluorescence correlation spectroscopy to characterize the molecular mobility of fluorescently labelled G protein-coupled receptors.

The membranes of living cells have been shown to be highly organized into distinct microdomains, which has spatial and temporal consequences for the interaction of membrane bound receptors and their signalling partners as complexes. Fluorescence correlation spectroscopy (FCS) is a technique with single cell sensitivity that sheds light on the molecular dynamics of fluorescently labelled receptors, ligands or signalling complexes within small plasma membrane regions of living cells. This review provides an overview of the use of FCS to probe the real time quantification of the diffusion and concentration of G protein-coupled receptors (GPCRs), primarily to gain insights into ligand–receptor interactions and the molecular composition of signalling complexes. In addition we document the use of photon counting histogram (PCH) analysis to investigate how changes in molecular brightness (ε) can be a sensitive indicator of changes in molecular mass of fluorescently labelled moieties.

Probing the Inhomogeneous Charge Distribution on Annealed Polyelectrolyte Star Polymers in Dilute Aqueous Solutions.

The conformational structure of a polyelectrolyte chain in dilute aqueous solution is strongly coupled with its surrounding electrostatic environment. With the introduction of branched topology, the distribution of counterions in the vicinity of a polyelectrolyte chain becomes highly inhomogeneous, giving rise to complex structures of branched polyelectrolytes in dilute aqueous solution. To directly probe the local electrostatic conditions near a branched polyelectrolyte in aqueous solutions, star-shaped poly(2-vinylpyridine) (P2VP) polymers with precise labeling of one single fluorophore at different locations, for example, the star center or the terminal group of one arm, were synthesized using reversible addition-fragmentation chain transfer (RAFT) polymerization of vinyl-terminated P2VP macromonomers. Using fluorescence correlation spectroscopy (FCS) combined with photon counting histogram (PCH) analysis, the conformational structures and local electric potential of P2VP star polyelectrolytes were investigated in dilute aqueous solutions of varied pH at a single molecule level. Despite the same hydrodynamic radius of P2VP stars, pH-sensitive fluorophores labeled at different locations sensitively differentiated the higher electric potential at the star center from the lower electric potential at the periphery in dilute aqueous solutions.

Fluorescence cumulants analysis with non-ideal observation profiles

One of the challenges of fluorescence fluctuation fpectroscopy (FFS) is an adequate approximation of a brightness profile. The key feature of fluorescence intensity distribution analysis (FIDA) is a polynomial approximation of a brightness profile. A broad range of brightness profile shapes can be well described by this approximation. A different approach consisting of the introduction of additional fitting parameters, defined as a relative difference between integrals of the actual brightness profile and its Gaussian approximation, is used in photon counting histogram (PCH) analysis. It is sufficient to introduce only one additional fitting parameter (first-order correction) to get an adequate fit to the experimental data in many practical applications.In the current study, we apply these approaches to the theory of time integrated fluorescence cumulants analysis. We demonstrate that developed corrections improve results of FFS analysis applied to simulated and experimental data. The use of different brightness profile approximations and normalizations in PCH and FIDA leads to different estimates of brightness and the number of molecules, even though they represent the same physical quantities. Based on the developed theory, we derive equations that relate brightness and the number of molecules in PCH and FIDA.

Plasma membrane dynamics and tetrameric organisation of ABCG2 transporters in mammalian cells revealed by single particle imaging techniques

ABCG2 is one of three human ATP binding cassette (ABC) transporters involved in the export from cells of a chemically and structurally diverse range of compounds. This multidrug efflux capability, together with a broad tissue distribution in the body, means that ABCG2 exerts a range of effects on normal physiology such as kidney urate transport, as well as contributing towards the pharmacokinetic profiles of many exogenous drugs. The primary sequence of ABCG2 contains only half the number of domains required for a functioning ABC transporter and so it must oligomerise in order to function, yet its oligomeric state in intact cell membranes remains uncharacterized. We have analysed ABCG2 in living cell membranes using a combination of fluorescence correlation spectroscopy, photon counting histogram analysis, and stepwise photobleaching to demonstrate a predominantly tetrameric structure for ABCG2 in the presence or absence of transport substrates. These results provide the essential basis for exploring pharmacological manipulation of oligomeric state as a strategy to modulate ABCG2 activity in future selective therapeutics.

Analysis of AKAP7γ Dimerization.

A-kinase anchoring proteins (AKAPs) constitute a family of scaffolding proteins that contribute to spatiotemporal regulation of PKA-mediated phosphorylation events. In particular, AKAP7 is a family of alternatively spliced proteins that participates in cardiac calcium dynamics. Here, we demonstrate via pull-down from transfected cells and by direct protein-protein association that AKAP7γ self-associates. Self-association appears to be an isoform specific phenomenon, as AKAP7α did not associate with itself or with AKAP7γ. However, AKAP7γ did associate with AKAP7δ, suggesting the long isoforms of the AKAP can form heterodimers. Surface plasmon resonance found that the AKAP7γ self-association occurs via two high affinity binding sites with K D values in the low nanomolar range. Mapping of the binding sites by peptide array reveals that AKAP7γ interacts with itself through multiple regions. Photon counting histogram analysis (PCH) of AKAP7γ-EGFP expressed in HEK-293 cells confirmed that AKAP7γ-EGFP self-associates in a cellular context. Lastly, computational modeling of PKA dynamics within AKAP7γ complexes suggests that oligomerization may augment phosphorylation of scaffolded PKA substrates. In conclusion, our study reveals that AKAP7γ forms both homo- and heterodimers with the long isoforms of the AKAP and that this phenomenon could be an important step in mediating effective substrate phosphorylation in cellular microdomains.

Exploring the Dynamics of Cell Processes through Simulations of Fluorescence Microscopy Experiments

Fluorescence correlation spectroscopy (FCS) methods are powerful tools for unveiling the dynamical organization of cells. For simple cases, such as molecules passively moving in a homogeneous media, FCS analysis yields analytical functions that can be fitted to the experimental data to recover the phenomenological rate parameters. Unfortunately, many dynamical processes in cells do not follow these simple models, and in many instances it is not possible to obtain an analytical function through a theoretical analysis of a more complex model. In such cases, experimental analysis can be combined with Monte Carlo simulations to aid in interpretation of the data. In response to this need, we developed a method called FERNET (Fluorescence Emission Recipes and Numerical routines Toolkit) based on Monte Carlo simulations and the MCell-Blender platform, which was designed to treat the reaction-diffusion problem under realistic scenarios. This method enables us to set complex geometries of the simulation space, distribute molecules among different compartments, and define interspecies reactions with selected kinetic constants, diffusion coefficients, and species brightness. We apply this method to simulate single- and multiple-point FCS, photon-counting histogram analysis, raster image correlation spectroscopy, and two-color fluorescence cross-correlation spectroscopy. We believe that this new program could be very useful for predicting and understanding the output of fluorescence microscopy experiments.

Single Molecule Analyses of Fluorescent Protein Tagged FSHR Reveals Homodimerization and Specific Heterodimerization with LHR

Gonadotropin hormones (Follicle stimulating hormone/FSH, Luteinizing Hormone/LH) play essential roles in ovarian follicle maturation. FSH induces granulosa cell proliferation and differentiation including expression of LH receptor (LHR). Since FSH receptor (FSHR) and LHR are concurrently expressed in granulosa cells, we attempted to monitor trafficking of FSHR and possible heterodimerization with LHR in HEK293 cells. We constructed an FSHR‐fluorescent protein (FSHR‐FP) but found that FSHR‐FP did not traffic to the plasma membrane. The FSHR C‐tail was swapped with the C‐tail of LHR (LHRcT) to create a chimera to which was added a fluorescent protein (FP) [GFP, YFP, or mCherry]. The FSHR‐LHRcT‐FP bound FSH and transduced FSH binding to produce cAMP. Quantitative FRET analysis of FSHR‐LHRcT‐FP showed an average FRET E% of 12.9± 5.7. Fluorescence Correlation Spectroscopy coupled with Photon Counting Histogram analyses demonstrated that the FSHR‐LHRcT‐FP exists as a freely diffusing homodimer on the cell surface. FSHR‐LHRcT‐FP co‐expressed with LHR‐FP showed an average FRET E% of 14.4±7.5, demonstrating the formation of FSHR and LHR heterodimers/hetero‐oligomers. These data provide a structural imprimatur to understand the consequences of FSHR/LHR heteromerization in granulosa cells as a possible additional level of FSHR signal control and modulation during the process of differentiation. Single-molecule analyses of fully functional fluorescent protein-tagged follitropin receptor reveal homodimerization and specific heterodimerization with lutropin receptor.

We have previously shown that the carboxyl terminus (cT) of human follicle-stimulating hormone (FSH, follitropin) receptor (FSHR) is clipped before insertion into the plasma membrane. Surprisingly, several different constructs of FSHR fluorescent fusion proteins (FSHR-FPs) failed to traffic to the plasma membrane. Subsequently, we discovered that substituting the extreme cT of luteinizing hormone (LH) receptor (LHR) to create an FSHR-LHRcT chimera has no effect on FSHR functionality. Therefore, we used this approach to create an FSHR-LHRcT-FP fusion. We found this chimeric FSHR-LHRcT-FP was expressed in HEK293 cells at levels similar to reported values for FSHR in human granulosa cells, bound FSH with high affinity, and transduced FSH binding to produce cAMP. Quantitative fluorescence resonance energy transfer (FRET) analysis of FSHR-LHRcT-YFP/FSHR-LHRcT-mCherry pairs revealed an average FRET efficiency of 12.9 ± 5.7. Advanced methods in single-molecule analyses were applied in order to ascertain the oligomerization state of the FSHR-LHRcT. Fluorescence correlation spectroscopy coupled with photon-counting histogram analyses demonstrated that the FSHR-LHRcT-FP fusion protein exists as a freely diffusing homodimer in the plasma membrane. A central question is whether LHR could oligomerize with FSHR, because both receptors are coexpressed in differentiated granulosa cells. Indeed, FRET analysis revealed an average FRET efficiency of 14.4 ± 7.5 when the FSHR-LHR cT-mCherry was coexpressed with LHR-YFP. In contrast, coexpression of a 5-HT2cVSV-YFP with FSHR-LHR cT-mCherry showed only 5.6 ± 3.2 average FRET efficiency, a value indistinguishable from the detection limit using intensity-based FRET methods. These data demonstrate that coexpression of FSHR and LHR can lead to heterodimerization, and we hypothesize that it is possible for this to occur during granulosa cell differentiation.

Lipopolymer Crowding in Polymer-Tethered Lipid Bilayers Alters Lipid Mixing Behavior and Protein Sequestration in the Presence of Raft-Mimicking Lipid Mixtures

It is now well established that the cytoplasm and plasma membrane of cells are characterized by high concentrations of proteins. Consequently, macromolecular crowding and confinement effects are believed to play important roles in the organization, dynamics, and function of proteins in cellular environments. However, the functional significance of molecular crowding remains unclear and conclusions are still controversial, due to the complexity of cellular systems. To address this experimental challenge, here we introduce a model membrane system on the basis of a polymer-tethered lipid bilayer, in which lipopolymers act as crowding agents. Our experiments show that changes in lipopolymer concentration have a profound effect on the lipid mixing behavior of raft-mimicking lipid mixtures forming micron-size liquid-ordered (lo) and liquid-disordered (ld) phase separations. Complementary fluorescence correlation spectrometry (FCS), confocal fluorescence intensity analysis, and photon counting histogram (PCH) analysis experiments demonstrate that lipopolymer-mediated molecular crowding also influences the lateral diffusion of lipids and membrane proteins, as well as membrane protein sequestration in raft-mimicking lipid mixtures. Interestingly, our experiments confirm that elevated lipopolymer concentrations cause the formation of buckling structures, which can be seen as a stress relaxation phenomenon. Notably, the extent of membrane buckling is substantially different in lo and ld lipid regions.

Measuring protein dynamics in live cells: protocols and practical considerations for fluorescence fluctuation microscopy.

Quantitative analysis of protein complex stoichiometries and mobilities are critical for elucidating the mechanisms that regulate cellular pathways. Fluorescence fluctuation spectroscopy (FFS) techniques can measure protein dynamics, such as diffusion coefficients and formation of complexes, with extraordinary precision and sensitivity. Complete calibration and characterization of the microscope instrument is necessary in order to avoid artifacts during data acquisition and to capitalize on the full capabilities of FFS techniques. We provide an overview of the theory behind FFS techniques, discuss calibration procedures, provide protocols, and give practical considerations for performing FFS experiments. One important parameter recovered from FFS measurements is the relative molecular brightness that can correlate with oligomerization. Three methods for measuring molecular brightness (fluorescence correlation spectroscopy, photon-counting histogram, and number and brightness analysis) recover similar values when measuring samples under ideal conditions in vitro. However, examples are given illustrating that these different methods used for calculating molecular brightness of fluorescent molecules in cells are not always equivalent. Methods relying on spot measurements are more prone to bleaching and movement artifacts that can lead to underestimation of brightness values. We advocate for the use of multiple FFS techniques to study molecular brightnesses to overcome and compliment limitations of individual techniques.

Defining the Oligomerization State of γ-Synuclein in Solution and in Cells

γ-Synuclein is expressed at high levels in neuronal cells and in multiple invasive cancers. Like its family member α-synuclein, γ-synuclein is thought to be natively unfolded but does not readily form fibrils. The function of γ-synuclein is unknown, but we have found that it interacts strongly with the enzyme phospholipase Cβ (PLCβ), altering its interaction with G proteins. As a first step in determining its role, we have characterized its oligomerization using fluorescence homotransfer, photon-counting histogram analysis, and native gel electrophoresis. We found that when its expressed in Escherichia coli and purified, γ-synuclein appears monomeric on chromatographs under denaturing conditions, but under native conditions, it appears as oligomers of varying sizes. We followed the monomer-to-tetramer association by labeling the protein with fluorescein and following the concentration-dependent loss in fluorescence anisotropy resulting from fluorescence homotransfer. We also performed photon-counting histogram analysis at increasing concentrations of fluorescein-labeled γ-synuclein and found concentration-dependent oligomerization. Addition of PLCβ2, a strong γ-synuclein binding partner whose cellular expression is correlated with γ-synuclein, results in disruption of γ-synuclein oligomers. Similarly, its binding to lipid membranes promotes the monomer form. When we exogenously express γ-synuclein or microinject purified protein into cells, the protein appears monomeric. Our studies show that even though purified γ-synuclein form oligomers, when binding partners are present, as in cells, it dissociates to a monomer to bind these partners, which in turn may modify protein function and integrity.

Spatial Organization of Oncogenic Ras on Supported Membranes

Ras is a membrane protein which acts as a molecular switch in the activation of many signal transduction pathways involved in cellular processes such as proliferation and apoptosis. Ras has been a subject of great interest due to the causal role found between its hyperactivity and cancer, and studies have shown that approximately 30% of all human cancers have oncogenic Ras mutations. Although Ras has been studied for more than thirty years, the spatial organization of oncogenic Ras on membranes is still not well characterized. It has been suggested that Ras may organize into clusters in cell membranes but the molecular scale structure of these organizations remain unknown. Molecular dimerization of Ras has also been suggested and we quantitatively characterize this in an array of Ras mutants in supported lipid bilayer membranes using fluorescence correlation spectroscopy (FCS) and photon counting histogram analysis (PCH). The implications of Ras clustering on downstream signaling will be discussed.

Fluorescence Correlation Spectroscopy Analysis of Serotonin, Adrenergic, Muscarinic, and Dopamine Receptor Dimerization: The Oligomer Number Puzzle

The issue of G protein-coupled receptor (GPCR) oligomer status has not been resolved. Although many studies have provided evidence in favor of receptor-receptor interactions, there is no consensus as to the exact oligomer size of class A GPCRs. Previous studies have reported monomers, dimers, tetramers, and higher-order oligomers. In the present study, this issue was examined using fluorescence correlation spectroscopy (FCS) with photon counting histogram (PCH) analysis, a sensitive method for monitoring diffusion and oligomer size of plasma membrane proteins. Six different class A GPCRs were selected from the serotonin (5-HT2A), adrenergic (α1b-AR and β2-AR), muscarinic (M1 and M2), and dopamine (D1) receptor families. Each GPCR was C-terminally labeled with green fluorescent protein (GFP) or yellow fluorescent protein (YFP) and expressed in human embryonic kidney 293 cells. FCS provided plasma membrane diffusion coefficients on the order of 7.5 × 10(-9) cm(2)/s. PCH molecular brightness analysis was used to determine the GPCR oligomer size. Known monomeric (CD-86) and dimeric (CD-28) receptors with GFP and YFP tags were used as controls to determine the molecular brightness of monomers and dimers. PCH analysis of fluorescence-tagged GPCRs revealed molecular brightness values that were twice the monomeric controls and similar to the dimeric controls. Reduced χ(2) analyses of the PCH data best fit a model for a homogeneous population of homodimers, without tetramers or higher-order oligomers. The homodimer configuration was unaltered by agonist treatment and was stable over a 10-fold range of receptor expression level. The results of this study demonstrate that biogenic amine receptors freely diffusing within the plasma membrane are predominantly homodimers.
