Effect of cadmium ions on photosystem II oxygen evolution

The paper presents the results of the study of the effect of cadmium (CdSO4) ions on photosystem II (PSII) photochemical activity. The presence of Cd2+ ions in the medium was found to significantly inhibit the oxygen evolution in the thylakoid membranes, indicating a clear correlation between the concentration of Cd2+ ions and the inhibition of PSII photochemical activity. Additionally, the study identified that this inhibition is strongly dependent on the pH level of the medium. The inhibition of PSII photochemical activity occurred quickly at low cadmium concentrations (1.0-30 mM). The study also observed that the inhibition of O2 evolution by Cd2+ ions was time-dependent, with a rapid 20-40% inhibition of oxygen evolution occurring when adding Cd2+. The photochemical activity of PSII was inhibited slowly during long-term incubation. The findings imply that PSII contains at least one high-affinity cadmium binding site. Effect of antigibberellins on morphogenesis, photosynthetic apparatus, productivity and their residual content in tomato fruits

The application of growth and development regulators on crops in order to optimize their production process is one of the leading tasks of modern plant physiology. Retardants – gibberellin inhibitors are widely used for this purpose. We investigated the effect of foliar treatment with EW-250, ССС-750 and 2-СEPA on morphogenesis, leaf apparatus, content of photosynthetic pigments, indices of chlorophyll fluorescence induction, CO2 gas exchange, and residual amounts of drugs in the fruits of tomato (Lycopersicon esculentum Mill.) Bobcat hybrid. The field experiment was laid on plots with an area of 33 m2. The treatment of the plants was carried out at the budding stage. Morphometric indices were determined at the stages of flowering and fruit formation. The chlorophylls content was determined in the raw material by the spectrophotometric method. Indices of photosystem II (PSII) photochemical activity were determined according to the parameters of chlorophyll fluorescence induction after a half-hour exposure of plants in the dark using a portable single-beam fluorimeter "Floratest". The determination of the residual content of retardants in the fruits was carried out on a Shimadzu GC gas chromatograph with a mass spectrometric detector – GCMS- QP2020 EI. All gibberellin inhibitors reduced linear plant size. The number of leaves on the plants decreased under 2-CEPA treatment, and increased after the application of EW-250. Treatment with 2-CEPA decreased, EW-250 significantly increased, and CCC-750 practically did not change the leaves’ fresh and dry weight. Leaf area and leaf index decreased under 2-CEPA treatment, but practically did not change when EW-250 and ССС-750 were applied. All antigibberellin drugs increased the leaf specific leaf weight and thickened the leaf lamina due to the growth of chlorenchyma cells. At the same time, growth inhibitors increased the volume of columnar parenchyma cells and practically did not change the size of spongy parenchyma cells. Retardants increased the chlorophylls content in leaves, while the ethylene producer 2-CEPA did not change this index. The plants’ chlorophyll index after treatment with drugs increased significantly. The whole plant dry weight increased under EW-250 treatment, decreased after 2-CEPA application, and did not change under CCC-750. It was established that the photosynthetic rate increased under the EW-250 treatment, both in the flowering stage and in the stage of fruit formation, while when using 2-CEPA and CCC-750, it occurred only at the stage of fruit formation. The most significant positive changes of PSII photochemical activity indices were observed under the use of EW-250. Under the action of the drug, the maximum and actual quantum efficiency of PSII increased, the linear electron transport accelerated, and the fraction of reaction centers that did not transfer electrons from the primary acceptor QA to QB decreased, at the same time the chlorophyll fluorescence decay coefficient significantly increased, which indicates an increase in the CO2 assimilation intensity. Retardants increased the proportion of the fruit in the whole plant dry weight. All growth regulators increased net photosynthetic efficiency. A significant increase in fruit yield occurred under EW-250 treatment. When using CCC-750, the index tended to increase, while under the influence of 2-CEPA the yield decreased. The residual amounts of EW-250 and CCC-750 in the fruits did not exceed the maximum permissible concentrations.

Priming with the green leaf volatile (Z)-3-hexeny-1-yl acetate enhances drought resistance in wheat seedlings

Various studies showed that green leaf volatiles play vital roles in biotic and abiotic stresses. However, their role in drought resistance has never been studied. In order to investigate the possible role of (Z)-3-hexeny-1-yl acetate (Z-3-HAC, a type of green leaf volatile), various physiology experiments were performed to understand the physiological mechanisms of Z-3-HAC in alleviating wheat drought stress. Drought-resistant wheat seedlings were primed with 100 μM Z-3-HAC at the four-leaf stage before drought treatment. Morphological analysis showed that the primed seedlings grew better and showed higher biomass accumulation in both shoots and roots under drought stress. Additionally, exogenous Z-3-HAC significantly increased the total root length, total root surface area, and total root volume of the seedlings under drought stress. In primed seedlings, physiological measurements showed that the relative water content (RWC), net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), maximal photochemical efficiency of photosystem II (PSII) (Fv/Fm), photochemical activity of PSII (Fv′/Fm′), total chlorophyll content, activities of antioxidant enzymes, and osmolyte accumulation were higher than the control under drought conditions. Furthermore, relative electrolyte conductivity (REC), intercellular CO2 concentration (Ci), reactive oxygen species (ROS) accumulation, and malondialdehyde (MDA) content were significantly lower than the non-primed seedlings. Lastly, principal component analysis (PCA) indicated that Z-3-HAC protected wheat seedlings against damage from drought stress mainly through antioxidant and osmoregulatory systems. This study opens a new field of Z-3-HAC application, and provides a new opportunity to improve wheat drought resistance.

Alleviatory effect of rare earth micro-fertilizer on photosystem II (PSII) photoinhibition in Pseudostellaria heterophylla leaves at photosynthetic midday depression

In order to explore the mitigatory mechanism of rare earths on the midday depression of photosynthesis, this study used fast-phase chlorophyll fluorescence technology combined with an antioxidant enzyme system to analyze the effects of lanthanum and cerium on the photosystem II (PSII) function of Pseudostellaria heterophylla leaves at noon. The results show that the maximum photochemical efficiency (Fv/Fm) and the photosynthetic performance index (PIABS) of P. heterophylla leaves during the photosynthetic noon-break are only 0.73 and 0.86, respectively. The leaves of P. heterophylla show obvious photoinhibition. The application of two rare earth fertilizers promotes the light energy absorption, capture, conversion, and electron transfer efficiencies of PSII in the P. heterophylla leaves and alleviates the photoinhibition caused by excess excitation energy. Additionally, the effect of lanthanum is better than that of cerium. In the fertilization groups, P. heterophylla leaves show higher PSII photochemical activity. Fertilization protects the oxygen-evolving complex (OEC) activity of P. heterophylla, thus reducing oxidative damage to the chloroplasts and preventing the dissociation of the thylakoids to maintain the normal physiological functioning of PSII, thereby reducing the degree of photoinhibition. Additionally, fertilization increases the electron transfer ability from QA to QB on the PSII electron receptor side of P. heterophylla. Correlation analysis shows that the fluorescence parameters VL, VK, and VJ are significantly negatively correlated with antioxidant enzyme activity and are significantly positively correlated with malondialdehyde content, relative electric conductivity and reactive oxygen species (ROS), which further indicates that rare earths can enhance the ROS scavenging system in plants and reduce the degree of membrane lipid peroxidation and plasma membrane damage, thereby maintaining the structure and function of the OEC on the donor side of PSII, increasing the electron receptor pool, and contributing to the defense against photoinhibition.

The high red/far-red ratio supports the acclimation of fern Platycerium bifurcatum to high light.

High light causes disturbances in photosynthetic phosphorylation or damage to the photosystem II (PSII) structure or even assimilation tissues. The value of the red/ far-red ratio (R/FR) provides the plant with information on the environmental light conditions, regulating, among others, photosynthetic activity and pigment composition of the plant. The response of the photosynthetic apparatus of the sporotrophophylls and nest leaves of Platycerium bifurcatum, grown for 6 months at the low or high R/FR ratio, were studied. Later, the plants were transferred to high light (1,200 µmol quantum • m-2 • s-1). Changes in PSII photochemical activity were determined based on non-destructive methods of chlorophyll a fluorescence kinetics analysis. The measurement of radiation reflectance from the leaves allowed to determine the content of selected pigments related to the photosynthesis process and to assess changes in the Photochemical Reflectance Index. The calculation of reflectance difference and sensitivity analysis was used to identify so-called "stress-sensitive wavelengths". Plant growth at high R/FR ratio prepares photosynthetic apparatus of ferns to high light and enables more efficient conversion of absorbed photons. The increase in the amount of photoprotective compounds allows the protection against photoinhibition in the sporotrophophyll leaves that play key roles in plant nutrition and reproduction.

Photoprotective strategies in the motile cryptophyte alga Rhodomonas salina—role of non-photochemical quenching, ions, photoinhibition, and cell motility

We explored photoprotective strategies in a cryptophyte alga Rhodomonas salina. This cryptophytic alga represents phototrophs where chlorophyll a/c antennas in thylakoids are combined with additional light-harvesting system formed by phycobiliproteins in the chloroplast lumen. The fastest response to excessive irradiation is induction of non-photochemical quenching (NPQ). The maximal NPQ appears already after 20s of excessive irradiation. This initial phase of NPQ is sensitive to Ca2+ channel inhibitor (diltiazem) and disappears, also, in the presence of non-actin, an ionophore for monovalent cations. The prolonged exposure to high light of R. salina cells causes photoinhibition of photosystem II (PSII) that can be further enhanced when Ca2+ fluxes are inhibited by diltiazem. The light-induced reduction in PSII photochemical activity is smaller when compared with immotile diatom Phaeodactylum tricornutum. We explain this as a result of their different photoprotective strategies. Besides the protective role of NPQ, the motile R. salina also minimizes high light exposure by increased cell velocity by almost 25% percent (25% from 82 to 104μm/s). We suggest that motility of algal cells might have a photoprotective role at high light because algal cell rotation around longitudinal axes changes continual irradiation to periodically fluctuating light.

Impact of UV-B radiation on the photosystem II activity, pro-/antioxidant balance and expression of light-activated genes in Arabidopsis thaliana hy4 mutants grown under light of different spectral composition

The effect of UV-B irradiation on the photosystem II (PSII) activity, the content of photosynthetic and UV-absorbing pigments (UAPs), activity of antioxidant enzymes such as catalase (CAT) and peroxidase (POD), as well as H2O2 content in 25-day-old wild type (WT) and the cryptochrome 1 (Cry1) mutant hy4 of Arabidopsis thaliana Col-0 plants was studied. In addition, expression of photoreceptor genes Cry1, Cry2 and UVR8, photomorphogenetic gene COP1 and transcription factors genes HY5, HYH, the gene of chlorophyll-binding protein of the PSII CAB1 as well as the flavonoid biosynthesis genes CHS, PAL and thylakoid ascorbate peroxidase gene tAPX was examined. It has been shown that UV-B leads to a decrease in the photochemical activity of PSII (FV/FM) and the PSII performance index (PIABS) of WT plants grown on white (WL) and red (RL) light and also hy4 mutants grown on WL, RL and blue light (BL). In plants grown on BL and WL, the decrease in the PSII photochemical activity was significantly greater in hy4 compared to WT. The PSII of WT plants grown in BL was resistant to UV-B. The UAPs content of hy4 grown on BL and WL was lower than that in WT. The POD and CAT activities of WT grown in BL were significantly higher than in the mutant. In WT and hy4 plants grown in RL, a noticeable difference in these enzymes activity was not found. In both types of plants grown in BL and RL, the expression of photomorphogenetic genes HYH, HY5 markedly increased after UV-B treatment but the expression of the UV-B photoreceptor gene UVR8 was reduced in hy4 grown in BL and RL. It is assumed that reduced resistance of PSII in hy4 plants grown in BL and WL can be associated with low UAPs content as well as lowered POD and CAT activities. In addition, we suggest the lowered expression of UVR8 and COP1 genes caused by Cry1 deficiency leads to a shift of balance of oxidants and antioxidants towards oxidants.

Light acclimation of shade-tolerant and sun-resistant Tradescantia species: photochemical activity of PSII and its sensitivity to heat treatment.

In this work, we have compared photosynthetic characteristics of photosystem II (PSII) in Tradescantia leaves of two contrasting ecotypes grown under the low light (LL) and high light (HL) regimes during their entire growth period. Plants of the same genus, T. fluminensis (shade-tolerant) and T. sillamontana (sun-resistant), were cultivated at 50-125µmol photons m-2s-1 (LL) or at 875-1000µmol photons m-2s-1 (HL). Analyses of intrinsic PSII efficiency was based on measurements of fast chlorophyll (Chl) a fluorescence kinetics (the OJIP test). The fluorescence parameters Fv/Fm (variable fluorescence) and F0 (the initial level of fluorescence) in dark-adapted leaves were used to quantify the photochemical properties of PSII. Plants of different ecotypes showed different sustainability with respect to changes in the environmental light intensity and temperature treatment. The sun-resistant species T. sillamontana revealed the tolerance to variations in irradiation intensity, demonstrating constancy of maximum quantum efficiency of PSII upon variations of the growth light. In contrast to T. sillamontana, facultative shade species T. fluminensis demonstrated variability of PSII photochemical activity, depending on the growth light intensity. The susceptibility of T. fluminensis to solar stress was documented by a decrease in Fv/Fm and a rise of F0 during the long-term exposition of T. fluminensis to HL, indicating the loss of photochemical activity of PSII. The short-term (10min) heat treatment of leaf cuttings caused inactivation of PSII. The temperature-dependent heating effects were different in T. fluminensis and T. sillamontana. Sun-resistant plants T. sillamontana acclimated to LL and HL displayed the same plots of Fv/Fm versus the treatment temperature (t), demonstrating a decrease in Fv/Fm at t ≥ 45°C. The leaves of shadow-tolerant species T. fluminensis grown under the LL and HL conditions revealed different sensitivities to heat treatment. Plants grown under the solar stress conditions (HL) demonstrated a gradual decline of Fv/Fm at lower heating temperatures (t ≥ 25°C), indicating the "fragility" of their PSII as compared to T. fluminensis grown at LL. Different responses of sun and shadow species of Tradescantia to growth light and heat treatment are discussed in the context of their biochemical and ecophysiological properties.

Rate-limiting steps in the dark-to-light transition of Photosystem II - revealed by chlorophyll-a fluorescence induction

Photosystem II (PSII) catalyses the photoinduced oxygen evolution and, by producing reducing equivalents drives, in concert with PSI, the conversion of carbon dioxide to sugars. Our knowledge about the architecture of the reaction centre (RC) complex and the mechanisms of charge separation and stabilisation is well advanced. However, our understanding of the processes associated with the functioning of RC is incomplete: the photochemical activity of PSII is routinely monitored by chlorophyll-a fluorescence induction but the presently available data are not free of controversy. In this work, we examined the nature of gradual fluorescence rise of PSII elicited by trains of single-turnover saturating flashes (STSFs) in the presence of a PSII inhibitor, permitting only one stable charge separation. We show that a substantial part of the fluorescence rise originates from light-induced processes that occur after the stabilisation of charge separation, induced by the first STSF; the temperature-dependent relaxation characteristics suggest the involvement of conformational changes in the additional rise. In experiments using double flashes with variable waiting times (∆τ) between them, we found that no rise could be induced with zero or short ∆τ, the value of which depended on the temperature - revealing a previously unknown rate-limiting step in PSII.

The effect of light quality on the pro-/antioxidant balance, activity of photosystem II, and expression of light-dependent genes in Eutrema salsugineum callus cells.

The antioxidant balance, photochemical activity of photosystem II (PSII), and photosynthetic pigment content, as well as the expression of genes involved in the light signalling of callus lines of Eutrema salsugineum plants (earlier Thellungiella salsuginea) under different spectral light compositions were studied. Growth of callus in red light (RL, maximum 660nm), in contrast to blue light (BL, maximum 450nm), resulted in a lower H2O2 content and thiobarbituric acid reactive substances (TBARS). The BL increased the activities of key antioxidant enzymes in comparison with the white light (WL) and RL and demonstrated the minimum level of PSII photochemical activity. The activities of catalase (CAT) and peroxidase (POD) had the highest values in BL, which, along with the increased H2O2 and TBARS content, indicate a higher level of oxidative stress in the cells. The expression levels of the main chloroplast protein genes of PSII (PSBA and PSBD), the NADPH-dependent oxidase gene of the plasma membrane (RbohD), the protochlorophyllide oxidoreductase genes (POR B, C) involved in the biosynthesis of chlorophyll, and the key photoreceptor signalling genes (CIB1, CRY2, PhyB, PhyA, and PIF3) were determined. Possible mechanisms of light quality effects on the physiological parameters of callus cells are discussed.

Photosystem II activity of wild type Synechocystis PCC 6803 and its mutants with different plastoquinone pool redox states

To assess the role of redox state of photosystem II (PSII) acceptor side electron carriers in PSII photochemical activity, we studied sub-millisecond fluorescence kinetics of the wild type Synechocystis PCC 6803 and its mutants with natural variability in the redox state of the plastoquinone (PQ) pool. In cyanobacteria, dark adaptation tends to reduce PQ pool and induce a shift of the cyanobacterial photosynthetic apparatus to State 2, whereas illumination oxidizes PQ pool, leading to State 1 (Mullineaux, C. W., and Holzwarth, A. R. (1990) FEBS Lett., 260, 245-248). We show here that dark-adapted Ox(-) mutant with naturally reduced PQ is characterized by slower QA(-) reoxidation and O2 evolution rates, as well as lower quantum yield of PSII primary photochemical reactions (Fv/Fm) as compared to the wild type and SDH(-) mutant, in which the PQ pool remains oxidized in the dark. These results indicate a large portion of photochemically inactive PSII reaction centers in the Ox(-) mutant after dark adaptation. While light adaptation increases Fv/Fm in all tested strains, indicating PSII activation, by far the greatest increase in Fv/Fm and O2 evolution rates is observed in the Ox(-) mutant. Continuous illumination of Ox(-) mutant cells with low-intensity blue light, that accelerates QA(-) reoxidation, also increases Fv/Fm and PSII functional absorption cross-section (590 nm); this effect is almost absent in the wild type and SDH(-) mutant. We believe that these changes are caused by the reorganization of the photosynthetic apparatus during transition from State 2 to State 1. We propose that two processes affect the PSII activity during changes of light conditions: 1) reversible inactivation of PSII, which is associated with the reduction of electron carriers on the PSII acceptor side in the dark, and 2) PSII activation under low light related to the increase in functional absorption cross-section at 590 nm.

Preferential damaging effects of limited magnesium bioavailability on photosystem I in Sulla carnosa plants.

Magnesium deficiency preferentially inhibits photosystem I rather than photosystem II in Sulla carnosa plants. The effects of magnesium (Mg(2+)) deficiency on growth, photosynthetic performance, pigment and polypeptide composition of chloroplast membranes were studied in the halophyte Sulla carnosa (Desf.), an annual legume endemic to Tunisia and Algeria. The results demonstrate a gradual decrease in biomass production with decreasing Mg(2+) availability in the growth medium. The increase of Mg(2+) deficiency was also associated with a decline of the net CO2 assimilation (Pn) in fully expanded leaves, a decrease in the amount of photosynthetic pigments, and an increase in the lipid peroxidation in plants exposed to decreased Mg(2+) concentrations. Interestingly, while CO2 assimilation already was affected at Mg(2+) concentrations below 1.5 mM, the photochemical efficiency of photosystem II (PSII) declined only in the absence of Mg(2+). In contrast, plants of S. carnosa grown in Mg(2+)-deficient conditions exhibited a significant decrease in photosystem I (PSI) photochemistry in vivo at much higher Mg(2+) levels compared to PSII photochemical activity. The inhibitory effect of Mg(2+) deficiency on PSI photochemistry strongly correlated with significantly lower relative abundance of PSI-related chlorophyll-protein complexes and lower amounts of PSI-associated polypeptides, PsaA, PsaB, and Lhca proteins within the same range of Mg(2+) concentrations. These observations were associated with a higher intersystem electron pool size, restricted linear electron transport and a lower rate of reduction of P700(+) in the dark indicating restricted capacity for PSI cyclic electron transfer in plants exposed to Mg(2+)-deficient conditions compared to controls. These results clearly indicate that PSI, rather than PSII is preferentially targeted and damaged under Mg(2+)-deficiency conditions.

Photoprotective mechanisms in photosystem II of Ephedra monosperma during development of frost tolerance

Dissipation of light energy absorbed by photosystem II (PSII) in assimilating shoots of an ever� green shrub Ephedra monosperma was investigated during its transition from the vegetative to frosttolerant state under natural conditions of Central Yakutia. The dynamics of modulated chlorophyll fluorescence and carotenoid content was analyzed during seasonal decrease in ambient temperature. The seasonal cooling was accompanied by a stepwise decrease in photochemical activity of PSII (Fv/Fm = (Fm - F0)/Fm). The decrease in Fv/Fm occurred from the beginning of September to the end of October, when the temperature was lowered from 10 to -8°C. During winter period the residual activity of PSII was retained at about 30% of the summer values. The seasonal decrease in temperature was accompanied by a significant stimulation of pHindepen� dent dissipative processes in reaction centers and antenna of PSII. The increase in energy losses was paral� leled by a proportional increase in zeaxanthin content on the background of decreasing content of violaxan� thin and β�carotene as possible zeaxanthin precursors. At the same time, inhibition of lightinduced non� photochemical quenching in the PSII antenna was observed. The results suggest that principal photoprotec� tive mechanisms during seasonal lowering of temperature are: (1) inactivation of PSII and dissipation of exci� tation energy in PSII reaction centers and (2) zeaxanthinmediated ener gy dissipation in the antenna com� plexes. The first mechanism seems to prevail at early stages of seasonal cooling, whereas both mechanisms are recruited from the onset of sustained freezing temperatures.

Glycine betaine application in grain filling wheat plants alleviates heat and high light-induced photoinhibition by enhancing the psbA transcription and stomatal conductance

Wheat (Triticum aestivum) plants often suffer from heat stress combined with high irradiance during the grain filling stage. Glycine betaine (GB) is a compatible solute that accumulates rapidly to enhance stress tolerance in wheat under abiotic stress. In this study, the effect of foliar application of GB on photosystem II (PSII) photochemistry in wheat leaves under the heat and high light stresses were investigated. The results showed that GB-supplemented wheat plants maintained higher chlorophyll content, photochemical activity of PSII and net photosynthetic rate (P n) than stressed only plants. Moreover, GB-treated plants could retard the decrease of psbA gene transcription and accelerate the endogenous accumulation of GB in leaves. The results suggested that the increasing stress tolerance by GB accumulation was associated with an improvement in D1 protein synthesis, which accelerated the repair of PSII following stress-enhanced photoinhibition. Moreover, the accumulation of abscisic acid under the heat and high light stresses was retarded by foliar application of GB, and the decrease of stomatal conductance was inhibited as well. The results suggested that GB accumulation in vivo was involved in the regulation of stomatal conductance. Further work is required to elucidate the mechanism of GB-induced stomatal movement and PSII photoprotection.

Foliar-applied salicylic acid alleviates heat and high light stress induced photoinhibition in wheat (Triticum aestivum) during the grain filling stage by modulating the psbA gene transcription and antioxidant defense

In this study, we investigated the effects of foliar applied salicylic acid (SA) on protecting wheat (Triticum aestivum) from heat and high light stress during the grain filling stage. Specifically, the Photosystem II (PSII) photochemistry of wheat flag leaves under the treatment of SA was studied as indicators for stress response. Our results indicated that under stress, SA-supplemented wheat plants maintained higher chlorophyll content, photochemical activity of PSII, and net photosynthetic rate in comparison to non-SA treated plants, and, in addition, the SA-supplemented plants recovered more rapidly from photoinhibition when the stress was removed. SA-treated plants inhibited the decrease of the psbA gene transcription that is caused by stress and then recovered to the original control level after the stress was removed. In addition, foliar supplementation of SA could maintain or elevate the activities of antioxidative enzymes, including superoxide dismutase, ascorbate peroxidase, and catalase, which are known to provide protection against oxidative stress for wheat crops. Taken together, our results suggest that foliar application of SA can protect the PSII complex from photo-damage through enhanced transcription of the psbA gene (encoding D1 protein), as well as through mitigating photo-oxidation enabled by a high level of anti-oxidative enzyme activities, which allows for faster functional recovery of PSII from heat and high light stress.

Chilling tolerance of Cicer arietinum lines evaluated by photosystem II and antioxidant activities

Two chickpea (Cicer arietinum L.) lines (AKN 87 and AKN 290) that have different chilling susceptibilities were exposed to 2 chilling temperatures (4 and 2 °C), either cold-acclimated (10 °C) or nonacclimated (25 °C), in order to understand and compare physiological and biochemical changes at the vegetative stage. Chilling temperatures resulted in reduced growth parameters, particularly in cold-acclimated lines, whereas nonacclimated plants exhibited the lowest water contents. Cold acclimation treatment led to protective changes of increased flavonoid, proline, and antioxidant enzyme activities, mostly superoxide dismutase (SOD) and ascorbate peroxidase (APX), in the lines. However, the 10 °C treatment did not significantly influence photosystem II (PSII) activity in chickpea plants. In chilling treatments, cold-acclimated plants exhibited better tolerance; of the 2 lines, AKN 87 had the higher PSII photochemical activity. The chlorophyll contents of lines decreased, while the anthocyanin and flavonoid contents of lines increased at decreasing temperatures. MDA and proline accumulation increased with the severity of the chilling stress. In conclusion, when the plants were exposed to cold acclimation, chilling induced the enhancement of antioxidant enzyme activities, particularly SOD and APX. The results demonstrated that cold acclimation reduced the deteriorative effects of chilling and provided better tolerance, specifically in AKN 87. The line AKN 87 has greater potential for cultivation as a chilling-tolerant cultivar.

Effect of the anthocyanic epidermal layer on Photosystem II and I energy dissipation processes in Tradescantia pallida (Rose) Hunt

The effect of anthocyanic cells of the epidermal layer was investigated on photosynthetic activity of the higher plant Tradescantia pallida. To determine the possible indirect role of anthocyanin in photosynthesis, analysis was done on intact leaves and leaves where anthocyanic epidermal layer was removed. Energy dissipation processes related to Photosystem II (PSII) and Photosystem I (PSI) activity was done using simultaneously Chlorophyll a (Chl a) fluorescence and P700 transmittance signals change. In anthocyanic epidermal-less leaves, PSII photochemical activity was more decreased in dependence to increasing light irradiance exposure. We found that photoinhibition of PSII decreased PSI activity by reducing the electron flow toward PSI, especially under high light intensities. Under those conditions, it resulted in the accumulation of oxidized PSI reaction centers, which was stronger in leaves where the anthocyanic epidermal layer was removed. In conclusion, our results showed that the anthocyanic epidermal layer had a photoprotective effect only on the PSII and not on the PSI of T. pallida leaves, supporting the role of anthocyanin pigments in the regulation of photosynthesis for excess absorbed light irradiance.

The analysis of PS II photochemical activity using single and multi-turnover excitations

Paper describes chlorophyll a fluorescence measurements in algal cells, and intact plant leaves and isolated chloroplasts. It focuses on amplitude and 10 μs-resolved kinetics of variable fluorescence responses upon excitation with fluorescence-saturating pulses (SP) and with 25 μs saturating single turnover flashes (STF) which are exposed before, during and after a 100 s actinic illumination (AL) of low and high intensity. In addition to the amply documented suppression of the maximal variable fluorescence from F m to F m ′ , the relative proportion of the distinguished O–J-, J–I- and I–P-phases of an SP-induced response is shown to be distinctly different in dark- and light-adapted leaves. The O–J-phase in the 0.01–1 ms time range is much less sensitive to light adaptation than the other phases in the 1–200 ms range. In algae and chloroplasts, the amplitude F m STF of the STF-induced response is hardly affected by a shift from the dark- to the light-activated steady state. The results support the hypothesis that the maximal variable fluorescence F m induced by a multiple-turnover, fluorescence-saturating pulse (SP), is associated with the release of photochemical and photoelectrochemical quenching. It is argued that the OJIPMT- or Kautsky induction curve of variable chlorophyll fluorescence in the 0–100 s time range is the reflection of the release of photochemical quenching supplemented with a temporary Photosystem I (PSI)-dependent photoelectric stimulation and transient release of photoelectrochemical quenching of radiative energy loss in the Photosystem II (PSII) antennas, rather than solely of a decrease in PSII photochemical activity as is usually concluded.

The effect of zearalenone on PSII photochemical activity and growth in wheat and soybean under salt (NaCl) stress

The effects of mycotoxin zearalenone (ZEN) on the photochemical activity of photosystem II (PSII) in wheat and soybean leaf discs incubated in ZEN solutions as well as the after-effects of pre-sowing soaking of seeds in solutions containing ZEN on the photochemical activity of PSII and on the seedlings growth under salt stress (NaCl solutions were investigated). The incubation of wheat leaf discs in ZEN solutions strongly inhibited the energy flux per cross section (CS) for absorption (ABS/CS), trapping (TRo/CS) and electron transport (ETo/CS), while the effects of ZEN action on soybean discs were opposite and the values of those parameters significantly increased with the increase in ZEN concentration. Incubation of seeds in a ZEN solution resulted in an increase in photochemical efficiency of PSII in soybean seedlings, but did not induce any response of PSII in those of wheat at medium illuminations. Only at the stronger illumination for both species did ZEN induce an increase in efficiency of excitation energy capture by open PSII reaction centers, photochemical quenching of chlorophyll a fluorescence and quantum yield of PSII electron transport. Pre-sowing soaking of seeds in a ZEN solution decreased the photoinhibitory injuries of PSII in wheat and soybean due to safe scattering of the excess excitation energy through an increase in energy-dependent quenching (qE) and state transition quenching (qT). ZEN when added to NaCl solutions during the period of germination contributed to reduction in the growth inhibition of wheat seedlings. The incubation of wheat leaf discs in ZEN solutions strongly inhibited CS, ABS/CS, TRo/CS and ETo/CS. Possible effects of ZEN on some physiological processes in plants have been discussed especially in the context with photochemical activity of PSII and a salt stress.

Analysis of fast chlorophyll fluorescence rise (O-K-J-I-P) curves in green fruits indicates electron flow limitations at the donor side of PSII and the acceptor sides of both photosystems

Limited evidence up to now indicates low linear photosynthetic electron flow and CO(2) assimilation rates in non-foliar chloroplasts. In this investigation, we used chlorophyll fluorescence techniques to locate possible limiting steps in photosystem function in exposed, non-stressed green fruits (both pericarps and seeds) of three species, while corresponding leaves served as controls. Compared with leaves, fruit photosynthesis was characterized by less photon trapping and less quantum yields of electron flow, while the non-photochemical quenching was higher and potentially linked to enhanced carotenoid/chlorophyll ratios. Analysis of fast chlorophyll fluorescence rise curves revealed possible limitations both in the donor (oxygen evolving complex) and the acceptor (Q(A)(-)--> intermediate carriers) sides of photosystem II (PSII) indicating innately low PSII photochemical activity. On the other hand, PSI was characterized by faster reduction of its final electron acceptors and their small pool sizes. We argue that the fast reductive saturation of final PSI electron acceptors may divert electrons back to intermediate carriers facilitating a cyclic flow around PSI, while the partial inactivation of linear flow precludes strong reduction of plastoquinone. As such, the photosynthetic attributes of fruit chloroplasts may act to replenish the ATP lost because of hypoxia usually encountered in sink organs with high diffusive resistance to gas exchange.
