Arachidonic Acid and Nitroarachidonic: Effects on NADPH Oxidase Activity.

Arachidonic acid (AA) is a polyunsaturated fatty acid that participates in the inflammatory response mainly through bioactive-lipids formation in macrophages and also in the phagocytic NADPH oxidase 2 (NOX2) activation. NOX2 is the enzyme responsible for a huge superoxide formation in macrophages, essential to eliminate pathogens inside the phagosome. The oxidase is an enzymatic complex comprised of a membrane-bound flavocytochrome b 558 (gp91phox/p22phox), three cytosolic subunits (p47phox, p40phox and p67phox) and a Rac-GTPase. The enzyme becomes active when macrophages are exposed to appropriate stimuli that trigger the phosphorylation of cytosolic subunits and its migration to plasmatic membrane to form the active complex. It is proposed that AA stimulates NOX2 activity through AA interaction with different components of the NADPH oxidase complex. In inflammatory conditions, there is an increase in reactive oxygen and nitrogen species that results in the production of nitrated derivatives of AA, such as nitroarachidonic acid (NO2-AA). NO2-AA is capable to inhibit NOX2 activity by interfering with p47phox migration to the membrane without affecting phosphorylation of cytosolic proteins. Also, NO2-AA is capable to interact with protein disulfide isomerase (PDI), which is involved on NOX2 active complex formation. It has been demonstrated that NO2-AA forms a covalent adduct with PDI that could prevent the interaction with NOX2 and it would explain the inhibitory effects of the fatty acid upon NOX2. Together, current data indicate that AA is an important activator of NOX2 formed in the early events of the inflammatory response, leading to a massive production of oxidants that may, in turn, promote NO2-AA formation and shutting down the oxidative burst. Hence, AA and its derivatives could have antagonistic roles on NOX2 activity regulation.

A new ibuprofen derivative inhibits platelet aggregation and ROS mediated platelet apoptosis.

Thrombocytopenia is a serious issue connected with the pathogenesis of several human diseases including chronic inflammation, arthritis, Alzheimer's disease, cardiovascular diseases (CVDs) and other oxidative stress-associated pathologies. The indiscriminate use of antibiotics and other biological drugs are reported to result in thrombocytopenia, which is often neglected during the treatment regime. In addition, augmented oxidative stress induced by drugs and pathological conditions has also been shown to induce thrombocytopenia, which seems to be the most obvious consequence of elevated rate of platelet apoptosis. Thus, blocking oxidative stress-induced platelet apoptosis would be of prime importance in order to negotiate thrombocytopenia and associated human pathologies. The current study presents the synthesis and platelet protective nature of novel ibuprofen derivatives. The potent anti-oxidant ibuprofen derivative 4f was selected for the study and the platelet protective efficacy and platelet aggregation inhibitory property has been demonstrated. The compound 4f dose dependently mitigates the oxidative stress-induced platelet apoptosis in both platelet rich plasma and washed platelets. The platelet protective nature of compound 4f was determined by assessing various apoptotic markers such as ROS generation, cytosolic Ca2+ levels, PS externalization, cytochrome C translocation, Caspase activation, mitochondrial membrane depolarization, cytotoxicity, LDH leakage and tyrosine phosphorylation of cytosolic proteins. Furthermore, compound 4f dose dependently ameliorated agonist induced platelet aggregation. Therefore, compound 4f can be estimated as a potential candidate in the treatment regime of pathological disorders associated with platelet activation and apoptosis. In addition, compound 4f can be used as an auxiliary therapeutic agent in pathologies associated with thrombocytopenia.

Multiplex Analysis of Cardiac Hypertrophic Signaling: Reduced in vivo Phosphorylation of Glycogen Synthase Kinase-3β and Proline-Rich Akt Substrate (PRAS40)

The serine/threonine kinase Akt (PKB) is activated in response to growth factors, cytokines and other growth promoting stimuli and is involved in the regulation of a number of cellular processes including metabolism, cell growth, proliferation and survival. To study intracellular metabolic effects of left ventricular hypertrophy (LVH) induced by chronic aortic banding in a recently established swine model, protein arrays can be used to analyze changes in cytosolic protein phosphorylation levels such as cell signal transduction and protein kinase pathways. Miniaturized sandwich immunoassays allow the simultaneous analysis of several parameters in a single experiment. Bead-based protein array systems or suspension microarrays are well-established multiplex sandwich immunoassay formats. Cytosolic proteins were extracted from pig hearts. Using the bead-based Luminex system, multiplexed sandwich immunoassays have been developed to analyze the phosphorylated proteins IR[pYpY1162/1163], IGF-1R[pYpY1135 /1136], IRS-1[pS312], Akt[pS473], PRAS40[pT246], p70S6K[pTpS421/424], and GSK-3β[pS9], which were measured simultaneously in extracted myocardial tissue homogenates with the Akt Pathway Phospho 7-Plex Panel using a Luminex 100 IS system. Using this assay, tissue homogenates derived from myocardial tissue samples from 28 individual porcine in vivo model experiments in normal hearts (n=17) and LVH hearts (n=11) were analyzed for phosphorylated protein concentrations. Results obtained allowed grouping of myocardial tissue samples into a LVH and a normal group. Two candidate molecules: PRAS40 (normal vs. LVH two-tailed probability P= 0.0022) and GSK-3β (P=0.003), had lower concentrations in LVH pigs than in normal animals, suggesting loss of GSK-3β inhibitory activity and contributing to speculation of altered insulin sensitivity associated with LVH. Impaired Generation of Reactive Oxygen Species in Leprechaunism Through Downregulation of Nox4

Leprechaunism features a clinical constellation characterized by extreme insulin resistance, growth retardation, and several distinct developmental abnormalities. One puzzling observation about leprechaunism is that mutations in the insulin receptor gene frequently associated with this syndrome cannot account for the aberrant responses of cultured cells to other growth factors. Here we report that the generation of reactive oxygen species (ROS) is impaired in cells from leprechaunism patients, thus shedding new light on this issue. Stimulation of patients' skin fibroblast cells with platelet-derived growth factor (PDGF) resulted in a lower-level tyrosine phosphorylation of cytosolic proteins compared with that seen in normal cells. In addition, consistent with the hypothesis that ROS mediate the level of tyrosine phosphorylation of cytosolic proteins through inactivation of protein tyrosine phosphatases (PTPases), patient fibroblast cells showed a significantly higher phosphatase activity than normal cells. We further showed that the lower-level tyrosine phosphorylation in response to growth factors results from the downregulation of an NADPH oxidase, Nox4, which in turn results in the reduction of ROS generation. Ectopic expression of Nox4 in the patient fibroblast cells consistently restored PDGF-induced ROS production and regulation of PTPase activities. Taken together, these data provide insight into the mechanisms through which growth retardation is associated with leprechaunism syndrome.

Nitric oxide and cGMP activate the Ras-MAP kinase pathway-stimulating protein tyrosine phosphorylation in rabbit aortic endothelial cells.

The free radical nitric oxide is a very effective signal transducer, stimulating the enzyme guanylyl cyclase, the oncoprotein p21Ras, and protein tyrosine phosphorylation. In the present study using rabbit aortic endothelial cells (RAEC), it is demonstrated that the nitric-oxide-generating substances sodium nitroprusside and S-nitroso-N-acetylpenicillamine, and a stable analog of cyclic GMP, 8BrcGMP stimulate p21Ras activity. Tyrosine phosphorylation of cytosolic proteins was stimulated and intracellular production of cGMP was increased, indicating that the NO/cGMP-stimulated tyrosine phosphorylation-dependent signaling pathway is most likely associated with the activation of p21Ras. NO and cGMP-dependent activation of p21Ras result in binding of the oncoprotein to the Ras-binding domain of Raf-1 kinase. Incubation of RAEC with FPT II, a potent and selective inhibitor of p21Ras, prevented NO-dependent tyrosine phosphorylation. ODQ, a potent inhibitor of the soluble form of guanylyl cyclase, inhibited the signal as well. Conversely, the use of KT5823, a cGMP-dependent protein kinase (PKG) blocker, showed no effect on protein tyrosine phosphorylation. To further establish a role for p21Ras on the NO-stimulated tyrosine phosphorylation-signaling pathway, RAEC were constitutively transfected with a dominant negative mutant of p21Ras, N17Ras. NO and cGMP-stimulated tyrosine phosphorylation were prevented in N17Ras-expressing RAEC exposed to NO donors and 8BrcGMP. The above findings indicate that NO and cGMP stimulation of protein tyrosine phosphorylation requires the participation of fully functional p21Ras. ERK1/2 MAP kinases and their subsequent targets, the transcription factors, lie downstream to Ras, Raf-1 kinase, and MEK. Treatment of both RAEC and mock-transfected RAEC with NO resulted in phosphorylation and activation of ERK1/2. On the other hand, NO did not stimulate phosphorylation of ERK1/2 in N17Ras-expressing RAEC. In addition, PD98059, a MEK inhibitor, prevented overall tyrosine phosphorylation and phosphorylation of ERK1/2. Upstream to Ras ERK1/2 MAP kinases target the EGF receptor. Incubation of RAEC or mock-transfected RAEC with NO donors resulted in activation of the EGF receptor autophosphorylation. PD98059 effectively blocked this activation. EGF receptor autophosphorylation was insensitive to NO stimulation in N17Ras-expressing RAEC. It is concluded that NO and cGMP stimulate a signaling pathway involving p21Ras-Raf-1 kinase-MEK-ERK1/2. Activation of this signaling pathway is connected to NO-stimulated overall tyrosine phosphorylation that also involves the transactivation of the EGF receptor mediated by ERK1/2.

Expression of ZAP-70 is associated with increased B-cell receptor signaling in chronic lymphocytic leukemia

We examined isolated leukemia B cells of patients with chronic lymphocytic leukemia (CLL) for expression of zeta-associated protein 70 (ZAP-70). CLL B cells that have nonmutated immunoglobulin variable region genes (V genes) expressed levels of ZAP-70 protein that were comparable to those expressed by normal blood T cells. In contrast, CLL B cells that had mutated immunoglobulin variable V genes, or that had low-level expression of CD38, generally did not express detectable amounts of ZAP-70 protein. Leukemia cells from identical twins with CLL were found discordant for expression of ZAP-70, suggesting that B-cell expression of ZAP-70 is not genetically predetermined. Ligation of the B-cell receptor (BCR) complex on CLL cells that expressed ZAP-70 induced significantly greater tyrosine phosphorylation of cytosolic proteins, including p72(Syk), than did similar stimulation of CLL cells that did not express ZAP-70. Also, exceptional cases of CLL cells that expressed mutated immunoglobulin V genes and ZAP-70 also experienced higher levels tyrosine phosphorylation of such cytosolic proteins following BCR ligation. Following BCR ligation, ZAP-70 underwent tyrosine phosphorylation and became associated with surface immunoglobulin and CD79b, arguing for the involvement of ZAP-70 in BCR signaling. These data indicate that expression of ZAP-70 is associated with enhanced signal transduction via the BCR complex, which may contribute to the more aggressive clinical course associated with CLL cells that express nonmutated immunoglobulin receptors.

Minor participation of cAMP on the protein kinase phosphorylation of mitochondrial and cytosolic fractions from Ascaris suum: a comparative study with porcine heart muscle

In contrast to porcine heart muscle in which cAMP effectively activated the phosphorylation of cytosolic proteins, cAMP exerted a minor effect on the phosphorylation of proteins from the soluble fraction of Ascaris suum muscle. Similarly, cAMP did not enhance the kinase activity in the mitochondrial membranes from porcine heart and A. suum, although major differences in protein phosphorylation were observed between both fractions. However, cAMP-dependent protein kinases (PKA) were evidenced in the parasitic soluble mitochondrial fraction, since the phosphorylation of histone IIA and kemptide was augmented in this fraction, in the presence of cAMP. An increase in the phosphorylation of exogenously added A. suum phosphofructokinase was also obtained when cAMP was added to the parasite soluble mitochondrial fraction. The phosphorylation of phosphofructokinase by this fraction was inhibited when kemptide and cAMP were included in the reaction mixture, suggesting substrate competition for the same PKA. Although PKI (6–22), a reported inhibitor of the catalytic subunit of mammalian cAMP-dependent PKAs, did not affect the endogenous phosphorylation of proteins in the various A. suum fractions, an inhibition on the phosphorylation of exogenously added kemptide and phosphofructokinase was observed when PKI (6–22) was incubated with the parasite mitochondrial soluble fraction.

Free radical mediated photoreceptor damage in uveitis.

Uveitis is a major cause of blindness, with the visual loss that occurs being due primarily to retinal tissue damage. The tissue damage is mediated mainly by phagocytic inflammatory cells, such as macrophages, by the release of various proteolytic enzymes, arachidonic acid metabolites, cytokines and free radicals. The latter are found to be potent cytotoxic agents that readily cause tissue damage by peroxidation of lipid cell membranes. Recent studies of experimental uveitis indicate that other potent oxidants are generated in uveitis by macrophages. One of these is ONOO-, which is formed from *NO and O(-)2. The macrophages generate *NO preferentially in the outer retina following iNOS expression. In these phagocytes, outer retinal proteins, especially arrestin, are found to be potent iNOS inducers. Current studies of RPE show that these cells protect the retina from ONOO- mediated damage in uveitis by releasing a novel protein called retinal pigment epithelial protective protein. This protein is found to suppress O(-)2 and *NO generation by the phagocytes, in both in vitro and in vivo uveitis models. The protective protein expression is restricted to RPE, its suppressive effect is a result of the inhibition of the phosphorylation of cytosolic proteins, p47-phox, required for the assembly of NADPH and activation of NFkappaB, which are required for generation of 0(-)2 and expression of iNOS respectively. Direct interaction between p47phox and protein kinase C: evidence for targeting of protein kinase C by p47phox in neutrophils

p47(phox) is an essential component of the NADPH oxidase, and phosphorylation of p47(phox) is associated with activation of the enzyme. Here we have used p47(phox) affinity chromatography to extract a p47(phox) kinase from neutrophil cytosol. The kinase activity was purified by gel filtration and Mini Q chromatography and shown to be indistinguishable from the catalytic fragments of protein kinase C (PKC)-beta(I), -beta(II) and -delta. The C-terminus of p47(phox) represented the site of interaction with PKC. Co-immunoprecipitation experiments revealed that the interaction between PKC isotypes and p47(phox) takes place in intact cells. However PKC-beta and -delta showed different time courses of co-immunoprecipitation, suggesting that the interactions may serve different functions for the various PKC isotypes. Using cells lacking p47(phox), we investigated the functional relevance of the interaction between PKC and p47(phox). Subcellular fractionation revealed an abnormal recruitment of PKC-beta(I) and -beta(II), but not PKC-delta, to particulate fractions in p47(phox)-deficient cells. Phosphorylation of cytosolic proteins was generally increased in stimulated p47(phox)-deficient neutrophils as compared with normal neutrophils. Furthermore, the cytoskeletal protein coronin was not phosphorylated upon stimulation of p47(phox)-deficient neutrophils. These findings were confirmed in an in vitro-reconstituted system using rat brain cytosol in which addition of p47(phox) affected phosphorylation by PKC/PKM (PKM is the catalytic fragment of PKC). These results indicate that p47(phox) can act as a regulator of PKC in neutrophils.

Activation of the L Voltage-sensitive Calcium Channel by Mitogen-activated Protein (MAP) Kinase following Exposure of Neuronal Cells to β-Amyloid: MAP KINASE MEDIATES β-AMYLOID-INDUCED NEURODEGENERATION

Neuronal degeneration in Alzheimer's disease (AD) has been variously attributed to increases in cytosolic calcium, reactive oxygen species, and phosphorylated forms of the microtubule-associated protein tau. beta-Amyloid (betaA), which accumulates extracellularly in AD brain, induces calcium influx in culture via the L voltage-sensitive calcium channel. Since this channel is normally activated by protein kinase A-mediated phosphorylation, we examined kinase activities recruited following betaA treatment of cortical neurons and SH-SY-5Y neuroblastoma. betaA increased channel phosphorylation; this increase was unaffected by the protein kinase A inhibitor H89 but was reduced by the mitogen-activated protein (MAP) kinase inhibitor PD98059. Pharmacological and antisense oligonucleotide-mediated reduction of MAP kinase activity also reduced betaA-induced accumulation of calcium, reactive oxygen species, phospho-tau immunoreactivity, and apoptosis. These findings indicate that MAP kinase mediates multiple aspects of betaA-induced neurotoxicity and indicates that calcium influx initiates neurodegeneration in AD. betaA increased MAP kinase-mediated phosphorylation of membrane-associated proteins and reduced phosphorylation of cytosolic proteins without increasing overall MAP kinase activity. Increasing MAP kinase activity with epidermal growth factor did not increase channel phosphorylation. These findings indicate that redirection, rather than increased activation, of MAP kinase activity mediates betaA-induced neurotoxicity.

Gender differences in the mechanism of dioxin toxicity in rodents and in nonhuman primates

This study examined the differences in mechanisms of toxicity when adipose cells from males and females were exposed to 2,3,7,8-tetrachlorodibenzo- p-dioxin (TCDD). Glucose uptake by adipose tissue in vitro was decreased significantly in male guinea pigs within 1 d of intraperitoneal injection of TCDD, but there was no significant effect in females, even at 28 d after treatment. A similar difference between male and female guinea pigs was detected in the effect of TCDD on lipoprotein lipase (LPL) activity, except that a significant decrease in LPL activity was observed 28 d after treatment. Experiments with adipose tissue expiants from untreated guinea pigs and macaques revealed similar gender differences in the effect of TCDD in vitro on glucose uptake and LPL activity. Both time-course studies and dose-response studies with TCDD in vitro confirmed the greater sensitivity of male tissues to TCDD toxicity. TCDD induced lipid peroxidation in the adipose tissues of male guinea pigs, while it had no effect in females. 3H-TCDD binding affinity studies in adipose expiant tissues showed that tissues from male guinea pigs and monkeys had a higher binding capacity for TCDD than female tissues. TCDD induced a significant reduction in nuclear protein phosphorylation and an increase in cytosolic protein phosphorylation in adipose tissue from male guinea pigs; the effects in female tissues were opposite: nuclear protein phosphorylation increased and cytosolic protein phosphorylation decreased. In a cell-free system in the absence of the nucleus, adipose tissues from male guinea pigs and monkeys responded to TCDD with a rapid stimulation of tyrosine kinase activity but female tissues from both species had a significantly lower and slower response. TCDD induced the DNA binding of AP-1 in adipose tissues of male guinea pigs, but in female tissues TCDD reduced the DNA binding of AP-1. In summary, the results of this study demonstrate gender differences in the response of nonreproductive cells to TCDD. Some of these differences involve different mechanisms of toxicity in both the cytoplasmic and nuclear compartments of the cell.

Tyrosine phosphorylation in neutrophils from synovial fluid of patients with rheumatoid arthritis.

The priming of neutrophils is associated with an increase in the level of tyrosine phosphorylation of cytosolic proteins, specifically of proteins with mol. wts of 42 and 74 kDa. We show here, using dot blots and Western blotting, that neutrophils isolated from rheumatoid synovial fluid have increased tyrosine phosphorylation of these target proteins. The level of tyrosine phosphorylation within neutrophils in the synovial fluid was increased when compared with neutrophils from the blood of the same patients, normal blood or neutrophils from the synovial fluid of patients without rheumatoid arthritis. Neutrophils from the rheumatoid synovial fluid were also more active and were unable to be further primed by exogenous primers. These data suggest that this elevation of tyrosine phosphorylation was the result of the action of local priming agents within the rheumatoid synovial fluid.

Inhibition by phospholipids, lysophospholipids and gangliosides of melittin-induced phosphorylation in bovine mammary gland

Melittin, a toxic peptide from bee venom known to inhibit protein kinase C (PKC) activity, induces, like sphingosine, phosphorylation of proteins, such as the 19 kDa and 27 kDa, in bovine mammary gland. This phosphorylation is inhibited by the addition of phosphatidylserine (PS). The present study was conducted to examine whether phospholipids (other than PS), lysophospholipids and gangliosides can inhibit the phosphorylation of cytosolic proteins from bovine mammary gland. The lipid effects were also explored in the sphingosine-induced phosphorylation for comparison. Like PS, phosphatidic acid, phosphatidylglycerol and phosphatidylinositol inhibited the melittin-induced phosphorylation. Phosphatidylcholine was less potent and phosphatidylethanolamine was inactive in inhibiting the phosphorylation. Similar results were obtained when sphingosine was used instead of melittin. Of the lysophospholipids tested, lysophosphatidic acid was most effective in inhibiting the phosphorylation induced by melittin. Lysophosphatidylcholine was inactive in reversing the melittin-induced phosphorylation, but it selectively suppressed the 19 kDa phosphorylation induced by sphingosine. Gangliosides inhibited both melittin- and sphingosine-induced phosphorylation and their effectiveness was: GTlb > GDla = GD3 > GM3. Of the phosphoproteins, the 19 kDa was most sensitive to ganglioside inhibition. These results suggest that melittin-induced phosphorylation is selectively inhibited by specific lipids and also that the enzyme(s) responsible for the phosphorylation is not identical to the sphingosine-activated protein kinases.

Membrane and cytosolic protein phosphorylation patterns in the early stages of DEN-induced hepatocarcinogenesis in rats fed a high or low protein diet.

The membrane and cytosolic protein phosphorylation patterns in the early stages of diethylnitrosamine-induced rat liver carcinogenesis, promoted by 2-acetylaminofluorene in the diet plus partial hepatectomy (DEN-AAF-PH), were analyzed by two-dimensional gel electrophoresis in animals fed a low protein (5% casein) diet, or the original high protein (24% casein) diet, in order to modulate the development of GST-P-positive preneoplastic lesions. Compared with untreated controls, membrane and cytosolic protein phosphorylation patterns changed only slightly in low protein-fed rats 7 days post-hepatectomy, with no appearance of enzyme-altered hyperplastic foci in the liver sections. By contrast, high protein-fed animals demonstrated GST-P-positive preneoplastic lesions 7 days post-hepatectomy and several acidic and more basic high M(r) phosphorylated membrane (between 97 and 116 kDa) as well as cytosolic (between 97 and 200 kDa) proteins could be detected. In the presence of enzyme-altered hepatocytes in the liver sections, low protein-fed rats demonstrated at 60 days post-hepatectomy cytosolic protein phosphorylation patterns remarkably similar to those shown by 24% casein-fed animals at 7 days post-hepatectomy, suggesting close correlation between protein phosphorylation patterns and development of preneoplastic lesions during the early stages of DEN-AAF-PH liver carcinogenesis. This may arise by a constitutive activation of one or more signal transduction pathways, possibly involving protein kinase C, during liver tumour promotion.

APO-1(CD95)-mediated apoptosis in Jurkat cells does not involve src kinases or CD45

Tyrosine phosphorylation has been reported to be an early event required for APO-1/Fas(CD95) signalling in lymphocytes [Eischen, C.M., Dick, C.J. and Leibson, P.J. (1994) J. Immunol. 153, 1947–1954]. We have compared two mutant Jurkat cells, one largely deficient in expression of CD45 (J45.01) and a second one deficient in expression of p56 lck (JCaM1.6) with wild type Jurkat cells for their ability to undergo APO-1-induced apoptosis. No significant difference was observed among the three cell lines. In the mutant Jurkat cells APO-1 triggering did not result in increased tyrosine phosphorylation of cytosolic proteins. Furthermore, herbimycin A did not inhibit but rather augmented apoptosis at concentrations which effectively degraded the src related kinases lck and fyn. The data suggest that APO-1-mediated signalling is independent from src kinases and CD45.

Activation of Protein Kinases and the Inactivation of Protein Phosphatase 2A in Tumour Necrosis Factor and Interleukin-1 Signal-Transduction Pathways

We report the identification of 16 of the 30 cellular proteins which are rapidly phosphorylated in tumour-necrosis-factor-(TNF)-treated or interleukin-1-(IL-1)-treated primary human fibroblasts. Phosphorylation assays of proteins found in the cytosolic extract of human fibroblasts by in vitro assays indicate that at least 12 of these proteins are likely to be substrates for mitogen-activated protein kinase(s) (MAP kinase), mitogen-activated protein-kinase-activated protein kinase 2 (MAPKAP kinase 2), a pp60c-src-like tyrosine kinase as well as for a putative dual nucleotide protein kinase (DNK) in TNF-treated or IL-1-treated cells. Comparison of the phosphorylation of cytosolic proteins in vitro by exogenously added protein kinases with that observed in cells treated with TNF or IL-1 enabled the identification of cellular substrates of TNF-activated and IL-1-activated cellular protein kinases. Comparison of protein kinase activities of cytosolic extracts derived from TNF-treated or IL-1-treated and control fibroblasts also show the activation of MAP kinase, MAPKAP kinase 2, a putative DNK and a pp60src-like tyrosine kinase 3-19 fold. The data suggest TNF or IL-1 signal transduction may involve the phosphorylation of protein phosphatase type 2A by a pp60src-like tyrosine kinase, followed by the activation of MAP kinase, MAPKAP kinase 2 and the putative DNK. However, the activation of MAP kinase and MAPKAP kinase 2 may be independent of the earlier activation of pp60src-like tyrosine kinase and the inactivation of protein phosphatase type 2A.

1,25-Dihydroxyvitamin D3 Effects in Rat Kidney: Regulation of Protein Phosphorylation

This study tested the hypothesis that 1,25-dihydroxyvitamin D3 treatment alters the pattern of protein phosphorylation in rat kidney. Treatment with 1,25-dihydroxyvitamin D3 (100 ng/day s.c. × 7d) in vitamin D-deficient rats markedly decreased phosphorylation of a particulate protein (91 ± 0.6 kDa, n = 9) and moderately increased phosphorylation of a cytosolic protein (108 ± 0.8 kDa, n = 9) in the kidney. The decreased phosphorylation of the 91-kDa particulate protein showed a graded dose response (0-200 ng/day), as did the more moderate increase in phosphorylation of the 108-kDa cytosolic protein. In conclusion, this study has provided evidence that 1,25-dihydroxyvitamin D3 plays an important role in the regulation of protein phosphorylation in rat kidney and adds these biochemical events to the growing list of 1,25-dihydroxyvitamin D3 effects in this poorly understood target tissue.

An elevation of cytosolic protein phosphorylation modulates trimeric G-protein regulation of secretory vesicle formation from the trans-Golgi network.

The role of protein phosphorylation in the formation of secretory vesicles from the trans-Golgi network (TGN) and in the regulation of this process by TGN-associated trimeric G-proteins was investigated, using a previously established and a novel cell-free system derived from the neuroendocrine cell line PC12. In the absence of exogenous activators of trimeric G-proteins, okadaic acid, an inhibitor of protein serine/threonine phosphatase types 1, 2A, and PPX, had no significant effect on secretory vesicle formation as reconstituted in a postnuclear supernatant. However, okadaic acid antagonized the inhibition of secretory vesicle formation which occurred upon activation of trimeric G-proteins by either aluminum fluoride or guanosine 5'-3-O-(thio)-triphosphate (GTP gamma S). Microcystin-LR, a protein phosphatase inhibitor structurally distinct from okadaic acid, also antagonized the trimeric G-protein-mediated inhibition of secretory vesicle formation but, in contrast to okadaic acid, alone was sufficient to stimulate this process. The antagonistic effect of the phosphatase inhibitors was abolished by a broad spectrum protein kinase inhibitor, staurosporine, which alone, however, did not affect vesicle formation. The effect of okadaic acid was promoted by activators of protein kinase C (phorbol myristate acetate) and protein kinase A (cyclic AMP). To investigate the subcellular localization of the phosphoprotein that is involved in the antagonistic effect of protein phosphatase inhibitors, a novel cell-free system was established which reconstitutes the formation of secretory vesicles from TGN membranes supplemented with cytosol. Using this cell-free system, the relevant phosphoprotein was found to reside in the cytosol. In conclusion, our results suggest that serine/threonine protein phosphorylation is not required for secretory vesicle formation from the TGN but modulates, via a cytosolic phosphoprotein, the regulation of this process by TGN-associated trimeric G-proteins.

Patterns of protein synthesis and phosphorylation during anoxia in the land snail Otala lactea

Protein labeling patterns with [32P]phosphorous and [35S]methionine plus [35S]cysteine were examined in normoxic and anoxic tissues of Otala lactea. Analysis of protein phosphorylation showed differences between normoxic and anoxic individuals in both the extent of labeling and the pattern of protein phosphorylation. These changes were confined to the hepatopancreas where anoxia caused overall protein phosphorylation to decrease by a factor of 3. Subcellular fractionation of the hepatopancreas showed that this overall decrease in protein phosphorylation was due to a lower level of protein phosphorylation in individual fractions. Thus, the level of protein phosphorylation during anoxia was 45% (plasma membrane fraction), 21% (mitochondrial fraction), 24% (microsomal fraction), and 22% (cytosolic fraction) of that measured in normoxic tissues. Isoelectric focusing and sodium dodecyl sulfate polyacrylamide gel electrophoresis showed several changes in protein phosphorylation patterns. For example, the relative phosphorylation of cytosolic proteins of molecular mass 65, 49, 30, 25, 21, and 18 kDa was increased in normoxic individuals. Cytosol from anoxic snails showed a relative increase in the phosphorylation of proteins of molecular mass 68, 52, 44, 37, 18, and 16 kDa. No differences in the protein labeling patterns of the mitochondrial and microsomal fractions were apparent. In contrast, analysis of proteins labeled with [35S]methionine plus [35S]cysteine showed no differences when anoxic and normoxic snail tissues were compared.

Modulation of cytosolic protein phosphorylation by sphingosylphosphorylcholine

Sphingosylphosphorylcholine (SPC), a putative product of sphingomyelin N-deacylation, has been shown to exert potent mitogenic activity (Desai, N.N. and Spiegel, S. (1991) Biochem. Biophys. Res. Commun. 181, 361–366). In order to explore potential mechanisms of action of SPC, the effects of SPC on endogenous protein phosphorylation was examined in vitro. In cytosol derived from Jurkat T cells, SPC was found to exert dual effects on protein phosphorylation. SPC (10–100 μM) induced the phosphorylation of a number of protein substrates with molecular weights of 32, 35, and 87 kDa. Higher concentrations of SPC (50–200 μM) inhibited the phosphorylation of proteins with estimated molecular weights of 22, 56, and 60 kDa by inhibiting the activity of endogenous protein kinases. Stimulation of protein kinases by SPC required distinct structural features (amino base and the hydrophobic character) from those required for inhibition of protein kinases (the choline phosphate headgroup as well as the hydrophobic character). The SPC-dependent protein kinases were distinct from protein kinase C, cyclic-nucleotide-dependent protein kinases, and calcium-dependent protein kinases, but may be related to casein kinase II. These studies suggest that SPC may act, at least in part, by modulating the activity of endogenous protein kinases.
