Phenotypic plasticity as the main driver of alien plant trait variation in urban versus rural microclimate for the model species Veronica persica.

Urban environments are warmer than the rural surroundings, impacting plant phenotypic traits. When plants are present over areas with contrasted conditions such as along urbanization gradients, their phenotypes may differ, and these differencesdepend on different processes, including phenotypic plasticity, maternal environmental effects and genetic differentiation (local adaptation and/or genetic drift). Successful establishment of alien species along environmental gradients has been linked to high phenotypic plasticity and rapid evolutionary responses, which are easier to track for species with a known residence time. The mechanisms explaining trait variation in plants in urban versus rural microclimatic conditions have received little attention. Using the alien Veronica persica as model species, we measured leaf traits in urban and rural populations and performed a reciprocal common-garden experiment to study how germination, leaf, growth, and flowering traits varied in response to experimental microclimate (rural or urban) and population origin environment (rural or urban). Veronica persica displayed phenotypic plasticity in all measured traits, with reduced germination, development, and flowering under urban microclimate which suggests more stressful growing conditions in the urban than in the rural microclimate. No significant effect of the rural or urban origin environment was detected, providing no evidence for local adaptation to urban or rural environments. Additionally, we found limited signs of maternal environmental effects. We noted the importance of the mother plant and the population identities suggesting genetically based differences. Our results indicate that urban environments are more hostile than rural ones, and that V. persica does not show any adaptation to urban environments despite genetic differences between populations.

Regulation of muscle pyruvate dehydrogenase activity and exercise fuel use in high-altitude deer mice.

Adult, lab-reared, highland deer mice acclimate to hypoxia by increasing reliance on carbohydrates to fuel exercise. Yet neither the underlying mechanisms for this shift in fuel use pattern, nor the impact of lifetime hypoxia exposure experienced in the high alpine, are fully understood. Thus, we assessed exercise fuel use in wild highland deer mice running in their native environment. We examined a key step in muscle carbohydrate oxidation, pyruvate dehydrogenase (PDH) and its regulation with exercise at altitude in wild highlanders and using first generation (G1) lab born and raised highlanders acclimated to normoxia or hypoxia. PDH activity was also determined in the gastrocnemius of G1 highlanders using an in situ muscle preparation. We found wild highlanders had a high reliance on carbohydrates while running in their native environment, consistent with data from hypoxia acclimated G1 highlanders. PDH activity in the gastrocnemius was similar post exercise between G1 and wild highlanders. However, when the gastrocnemius was stimulated at a light work rate in situ, PDH activity was higher in hypoxia acclimated G1 highlanders, and associated with a lower intramuscular lactate. These findings were supported by lower PDH kinase 2 protein expression in hypoxia acclimated G-1 mice. Our findings indicate that adult phenotypic plasticity in response to low oxygen is sufficient to increase exercise carbohydrate reliance in highland deer mice. Additionally, variation in PDH regulation with hypoxia acclimation contributes to shifts in whole-animal fuel use patterns and are likely to improved exercise performance via elevated energy yield per mole O2.

Manipulation of photosensory and circadian signaling restricts phenotypic plasticity in response to changing environmental conditions in Arabidopsis

Plants exploit phenotypic plasticity to adapt their growth and development to prevailing environmental conditions. Interpretation of light and temperature signals is aided by the circadian system, which provides a temporal context. Phenotypic plasticity provides a selective and competitive advantage in nature but is obstructive during large-scale, intensive agricultural practices since economically important traits (including vegetative growth and flowering time) can vary widely depending on local environmental conditions. This prevents accurate prediction of harvesting times and produces a variable crop. In this study, we sought to restrict phenotypic plasticity and circadian regulation by manipulating signaling systems that govern plants’ responses to environmental signals. Mathematical modeling of plant growth and development predicted reduced plant responses to changing environments when circadian and light signaling pathways were manipulated. We tested this prediction by utilizing a constitutively active allele of the plant photoreceptor phytochrome B, along with disruption of the circadian system via mutation of EARLY FLOWERING3. We found that these manipulations produced plants that are less responsive to light and temperature cues and thus fail to anticipate dawn. These engineered plants have uniform vegetative growth and flowering time, demonstrating how phenotypic plasticity can be limited while maintaining plant productivity. This has significant implications for future agriculture in both open fields and controlled environments.

Altering developmental oxygen exposure influences thermoregulation and flight performance of Manduca sexta.

Endothermic, flying insects are capable of some of the highest recorded metabolic rates. This high aerobic demand is made possible by the insect's tracheal system, which supplies the flight muscles with oxygen. Many studies focus on metabolic responses to acute changes in oxygen to test the limits of the insect flight metabolic system, with some flying insects exhibiting oxygen limitation in flight metabolism. These acute studies do not account for possible changes induced by developmental phenotypic plasticity in response to chronic changes in oxygen levels. The endothermic moth Manduca sexta is a model organism that is easy to raise and exhibits a high thorax temperature during flight (∼40°C). In this study, we examined the effects of developmental oxygen exposure during the larval, pupal and adult stages on the adult moth's aerobic performance. We measured flight critical oxygen partial pressure (Pcrit-), thorax temperature and thermoregulating metabolic rate to understand the extent of developmental plasticity as well as effects of developmental oxygen levels on endothermic capacity. We found that developing in hypoxia (10% oxygen) decreased thermoregulating thorax temperature when compared with moths raised in normoxia or hyperoxia (30% oxygen), when moths were warming up in atmospheres with 21-30% oxygen. In addition, moths raised in hypoxia had lower critical oxygen levels when flying. These results suggest that chronic developmental exposure to hypoxia affects the adult metabolic phenotype and potentially has implications for thermoregulatory and flight behavior.

Testing the evolutionary potential of an alpine plant: Phenotypic plasticity in response to growth temperature far outweighs parental environmental effects and other genetic causes of variation

Abstract Phenotypic plasticity and rapid evolution are fundamental processes by which organisms can maintain their function and fitness in the face of environmental changes. Here we quantified the plasticity and evolutionary potential of an alpine herb Wahlenbergia ceracea. Utilising its mixed-mating system, we generated outcrossed and self-pollinated families that were grown in either cool or warm environments, and that had parents that had also been grown in either cool or warm environments. We then analysed the contribution of environmental and genetic factors to variation in a range of phenotypic traits including phenology, leaf mass per area, photosynthetic function, thermal tolerance, and reproductive fitness. The strongest effect was that of current growth temperature, indicating strong phenotypic plasticity. All traits except thermal tolerance were plastic, whereby warm-grown plants flowered earlier, grew larger, produced more reproductive stems compared to cool-grown plants. Flowering onset and biomass were heritable and under selection, with early flowering and larger plants having higher relative fitness. There was little evidence for transgenerational plasticity, maternal effects, or genotype-by-environment interactions. Inbreeding delayed flowering and reduced reproductive fitness and biomass. Overall, we found that W. ceracea has the capacity to respond rapidly to climate warming via plasticity, and the potential for evolutionary change.

Current evidence of climate-driven colour change in insects and its impact on sexual signals.

The colours of insects function in intraspecific communication such as sexual signalling, interspecific communication such as protection from predators, and in physiological processes, such as thermoregulation. The expression of melanin-based colours is temperature-dependent and thus likely to be impacted by a changing climate. However, it is unclear how climate change drives changes in body and wing colour may impact insect physiology and their interactions with conspecifics (e.g. mates) or heterospecific (e.g. predators or prey). The aim of this review is to synthesise the current knowledge of the consequences of climate-driven colour change on insects. Here, we discuss the environmental factors that affect insect colours, and then we outline the adaptive mechanisms in terms of phenotypic plasticity and microevolutionary response. Throughout we discuss the impact of climate-related colour change on insect physiology, and interactions with con-and-heterospecifics.

Environmental versus phylogenetic controls on leaf nitrogen and phosphorous concentrations in vascular plants

Global patterns of leaf nitrogen (N) and phosphorus (P) stoichiometry have been interpreted as reflecting phenotypic plasticity in response to the environment, or as an overriding effect of the distribution of species growing in their biogeochemical niches. Here, we balance these contrasting views. We compile a global dataset of 36,413 paired observations of leaf N and P concentrations, taxonomy and 45 environmental covariates, covering 7,549 sites and 3,700 species, to investigate how species identity and environmental variables control variations in mass-based leaf N and P concentrations, and the N:P ratio. We find within-species variation contributes around half of the total variation, with 29%, 31%, and 22% of leaf N, P, and N:P variation, respectively, explained by environmental variables. Within-species plasticity along environmental gradients varies across species and is highest for leaf N:P and lowest for leaf N. We identified effects of environmental variables on within-species variation using random forest models, whereas effects were largely missed by widely used linear mixed-effect models. Our analysis demonstrates a substantial influence of the environment in driving plastic responses of leaf N, P, and N:P within species, which challenges reports of a fixed biogeochemical niche and the overriding importance of species distributions in shaping global patterns of leaf N and P.

Plasticity in resource allocation of the invasive Phytolacca americana: Balancing growth, reproduction, and defense along urban–rural gradients

In response to varying environments along urban and rural gradients, invasive plants may strategically allocate resources to enhance their invasiveness. However, how invasive plants balance their resources for growth, reproduction, and defense as responses to biotic and abiotic factors across these gradients remain unclear. We conducted field surveys on the growth, reproduction, and herbivory of the invasive species Phytolacca americana across diverse urban and rural habitats. Leaf samples were collected to analyze the nutritional content, primary and secondary metabolites. We found that plant growth rates, specific leaf area, leaf nitrogen content, and concentrations of flavonoids and saponins were higher in urban habitats, while reproduction, herbivory, and carbon-to‑nitrogen ratios were lower than those in rural habitats. We also found a trade-off between growth rate and herbivory, as well as trade-offs among defense traits associated with herbivory (e.g., leaf mass per area, the inverse of leaf nitrogen content, and carbon‑nitrogen ratio) and the production of metabolites associated with abiotic stress tolerance (e.g., soluble sugars, flavonoids, and saponins). As earlier studies showed low levels of genetic diversity within and between populations, our findings suggest that the urban–rural gradient patterns of resource allocation are primarily phenotypic plasticity in response to herbivory in rural areas and abiotic factors in urban areas. Our study sheds light on the mechanisms by which urbanization affects plant invasions and offers insights for the implementation of their management strategies.

Proteomic and physiological signatures of altitude adaptation in a Myrsine coriacea population under common garden conditions

Plants exhibit phenotypic plasticity in response to environmental variations, which can lead to stable genetic and physiological adaptations if exposure to specific conditions is prolonged. Myrsine coriacea demonstrates this through its ability to thrive in diverse environments. The objective of the article is to investigate potential differences in protein accumulation and physiological responses of M. coriacea by cultivating plants from seeds collected from four populations at different altitudes in a common garden experiment. Additionally, we aim to evaluate whether these differences exhibit genetic fixation. Through integrated physiological and proteomic analyses, we identified 170 differentially accumulated proteins and observed significant physiological differences among the populations. The high-altitude population (POP1) exhibited a unique proteomic profile with significant down-regulation of proteins involved in carbon fixation and energy metabolism, suggesting a potential reduction in photosynthetic efficiency. Physiological analyses showed lower leaf nitrogen content, net CO2 assimilation rate, specific leaf area, and relative growth rate in stem height for POP1, alongside higher leaf carbon isotopic composition (δ13C) and leaf carbon (C) content. These findings provide insight into the complex interplay between proteomic and physiological adaptations in M. coriacea and underscore the importance of local adaptations. SignificanceWe investigate the adaptive responses of M. coriacea, a shrub with a broad phenotypic range, by cultivating plants from seeds collected at four different altitudes in a common garden experiment. These findings provide insight into the complex interplay between proteomic and physiological adaptations in M. coriacea and underscore the importance of local adaptations in the face of climate change. This study contributes to advancing our understanding of the influence of altitude-specific selection pressures on the molecular biology and physiology of plants in natural populations. Our findings provide valuable insights that enhance our ability to predict and comprehend how plants respond to climate change.

Consistent geographical gradient of water use efficiency evidences local adaptations to drought across the complete latitudinal distribution of Quercus suber

RationaleIncreased aridity has led to drought-induced mortality or loss of health for many tree species. Of particular interest is to explore the response of the Mediterranean tree species cork oak (Quercus suber) to this declining phenomenon due to its severity and its large implications for the local economy and the provision of highly relevant ecosystem services. ApproachTo assess geographical variations in the response to water stress, we analyzed under controlled conditions the effects of four watering levels and the resistance to a terminal drought on seedlings collected from nine populations covering the complete latitudinal distribution of the species. We explored the response of a number of physiological traits and markers of oxidative stress potentially related with drought-resistance. FindingsWe found a highly plastic phenotypic response of most variables to water availability and a large influence of seedlings provenance in the drought-resistance strategies. Although the oxidative stress enzymes ruled out differential water stress throughout the distribution range, we found that seedlings from the southern limit are less vulnerable to drought than other populations. Southern seedlings adapted to xeric conditions displayed a larger sensitivity of stomata to changes in soil humidity and a higher water use efficiency. These physiological local adaptations coupled with larger acorn size in the southern populations, resulted in larger aboveground biomass and higher drought resistance at the southern distribution edge. ConclusionsOur data suggest that this evergreen tree species relies on its physiological plasticity to develop adaptative features that allows it to overcome water shortage and that southern populations hold genetic diversity that could improve the specie´s adaptation in the forecasted drought context with relevant implications for conservation programs.

Effects of long-term voluntary wheel running and selective breeding for wheel running on femoral nutrient canals.

The nutrient artery provides ~50%-70% of the total blood volume to long bones in mammals. Studying the functional characteristics of this artery invivo can be difficult and expensive, so most researchers have measured the nutrient foramen, an opening on the outer surface of the bone that served as the entry point for the nutrient artery during development and bone ossification. Others have measured the nutrient canal (i.e., the passage which the nutrient artery once occupied), given that the external dimensions of the foramen do not necessarily remain uniform from the periosteal surface to the medullary cavity. The nutrient canal, as an indicator of blood flow to long bones, has been proposed to provide a link to studying organismal activity (e.g., locomotor behavior) from skeletal morphology. However, although external loading from movement and activity causes skeletal remodeling, it is unclear whether it affects the size or configuration of nutrient canals. To investigate whether nutrient canals can exhibit phenotypic plasticity in response to physical activity, we studied a mouse model in which four replicate high runner (HR) lines have been selectively bred for high voluntary wheel-running behavior. The selection criterion is the average number of wheel revolutions on days 5 and 6 of a 6-day period of wheel access as young adults (~6-8 weeks old). An additional four lines are bred without selection to serve as controls (C). For this study, 100 female mice (half HR, half C) from generation 57 were split into an active group housed with wheels and a sedentary group housed without wheels for 12 weeks starting at ~24 days of age. Femurs were collected, soft tissues were removed, and femora were micro-computed tomography scanned at a resolution of 12 μm. We then imported these scans into AMIRA and created 3D models of femoral nutrient canals. We tested for evolved differences in various nutrient canal traits between HR and C mice, plastic changes resulting from chronic exercise, and the selection history-by-exercise interaction. We found few differences between the nutrient canals of HR versus C mice, or between the active and sedentary groups. We did find an interaction between selection history and voluntary exercise for the total number of nutrient canals per femur, in which wheel access increased the number of canals in C mice but decreased it in HR mice. Our results do not match those from an earlier study, conducted at generation 11, which was prior to the HR lines reaching selection limits for wheel running. The previous study found that mice from the HR lines had significantly larger total canal cross-sectional areas compared to those from C lines. However, this discrepancy is consistent with studies of other skeletal traits, which have found differences between HR and C mice to be somewhat inconsistent across generations, including the loss of some apparent adaptations with continued selective breeding after reaching a selection limit for wheel-running behavior.

Climate is changing, are European bats too? A multispecies analysis of trends in body size

AbstractAnimal size, a trait sensitive to spatial and temporal variables, is a key element in ecological and evolutionary dynamics. In the context of climate change, there is evidence that some bat species are increasing their body size via phenotypic responses to higher temperatures at maternity roosts. To test the generality of this response, we conducted a >20‐year study examining body size changes in 15 bat species in Italy, analysing data from 4393 individual bats captured since 1995. In addition to examining the temporal effect, we considered the potential influence of sexual dimorphism and, where relevant, included latitude and altitude as potential drivers of body size change. Contrary to initial predictions of a widespread increase in size, our findings challenge this assumption, revealing a nuanced interplay of factors contributing to the complexity of bat body size dynamics. Specifically, only three species (Myotis daubentonii, Nyctalus leisleri, and Pipistrellus pygmaeus) out of the 15 exhibited a discernible increase in body size over the studied period, prompting a reassessment of bats as reliable indicators of climate change based on alterations in body size. Our investigation into influencing factors highlighted the significance of temperature‐related variables, with latitude and altitude emerging as crucial drivers. In some cases, this mirrored patterns consistent with Bergmann's rule, revealing larger bats recorded at progressively higher latitudes (Plecotus auritus, Myotis mystacinus, and Miniopterus schreibersii) or altitudes (Pipistrellus kuhlii). We also observed a clear sexual dimorphism effect in most species, with females consistently larger than males. The observed increase in size over time in three species suggests the occurrence of phenotypic plasticity, raising questions about potential long‐term selective pressures on larger individuals. The unresolved question of whether temperature‐related changes in body size reflect microevolutionary processes or phenotypic plastic responses adds further complexity to our understanding of body size patterns in bats over time and space.

The incredible shrinking puffin: Decreasing size and increasing proportional bill size of Atlantic puffins nesting at Machias Seal Island.

Climate change imposes physiological constraints on organisms particularly through changing thermoregulatory requirements. Bergmann's and Allen's rules suggest that body size and the size of thermoregulatory structures differ between warm and cold locations, where body size decreases with temperature and thermoregulatory structures increase. However, phenotypic plastic responses to malnutrition during development can result in the same patterns while lacking fitness benefits. The Gulf of Maine (GOM), located at the southern end of the Labrador current, is warming faster than most of the world's oceans, and many of the marine species that occupy these waters exist at the southern edge of their distributions including Atlantic puffins (Fratercula arctica; hereafter "puffin"). Monitoring of puffins in the GOM, at Machias Seal Island (MSI), has continued annually since 1995. We asked whether changes in adult puffin body size and the proportional size of bill to body have changed with observed rapid ocean warming. We found that the size of fledgling puffins is negatively related to sea surface temperature anomalies (warm conditions = small fledgers), adult puffin size is related to fledgling size (small fledgers = small adults), and adult puffins have decreased in size in recent years in response to malnutrition during development. We found an increase in the proportional size of bill to wing chord, likely in response to some mix of malnutrition during development and increasing air temperatures. Although studies have assessed clinal variation in seabird morphology with temperature, this is the first study addressing changes in seabird morphology in relation to ocean warming. Our results suggest that puffins nesting in the GOM have morphological plasticity that may help them acclimate to ocean warming.

Effect of Simulated Grazing on Morphological Plasticity and Resource Allocation of Aeluropus lagopoides

Aeluropus lagopoides, a dominant palatable species in various sabkha and coastal regions of Saudi Arabia, can withstand harsh saline environments through phenotypic plasticity. When subjected to grazing, how A. lagopoides adapt phenotypically is currently unknown. There is a breakage in the chain of study on the spatial and temporal expansion strategy of A. lagopoides plants when subjected to different grazing stresses in different saline soil habitats. A grazing experiment was conducted to investigate the phenotypic plasticity and resource allocation pattern response of A. lagopoides in different saline soils. Individual A. lagopoides rhizomes from five saline regions were grown and exposed to varied grazing treatments in the form of clipping, viz; light, moderate, and heavy grazing, as compared to a grazing exclusion control. Our results showed that heavy grazing/clipping significantly decreased the shoot system and above-ground biomass in high-saline region plants in the early season. Plant length, root length, root and shoot biomass, the number of stolons, average stolon length, leaf area, and SLA of A. lagopiodes responded significantly to grazing intensities. A. lagopoides from the Qareenah, Qaseem, and Jizan regions were more tolerant to light grazing than A. lagopoides from the Salwa and Jouf regions. Light grazing showed significantly good re-growth, especially during the late season. Light grazing decreased the synthesis of chlorophyll content. Also, A. lagopiodes reduced the risk caused by reactive oxygen species via the increased accumulation of proline content. Overall, plants adapted to different morphological and physiological strategies to tolerate different levels of grazing intensities by adapting their morphological attributes. Though heavy grazing damages the plant, light and moderate grazing can be allowed to maintain the productivity and economic benefits of sabka habitats where soil conditions are moderately saline.

Transcriptome analysis revealed that short-term stress in Blattella germanica to β-cypermethrin can reshape the phenotype of resistance adaptation

Previous studies on insect resistance have primarily focused on resistance monitoring and the molecular mechanisms involved, while overlooking the process of phenotype formation induced by insecticide stress. In this study, we compared the expression profiles of a beta-cypermethrin (β-CYP) resistant strain (R) and a susceptible strain (S) of Blattella germanica after β-CYP induction using transcriptome sequencing. In the short-term stress experiment, we identified a total of 792 and 622 differentially expressed genes (DEGs) in the S and R strains. Additionally, 893 DEGs were identified in the long-term adaptation experiment. To validate the RNA-Seq data, we performed qRT-PCR on eleven selected DEGs, and the results were consistent with the transcriptome sequencing data. These DEGs exhibited down-regulation in the short-term stress group and up-regulation in the long-term adaptation group. Among the validated DEGs, CUO8 and Cyp4g19 were identified and subjected to knockdown using RNA interference. Subsequent insecticide bioassays revealed that the mortality rate of cockroaches treated with β-CYP increased by 69.3% and 66.7% after silencing the CUO8 and Cyp4g19 genes (P<0.05). Furthermore, the silencing of CUO8 resulted in a significant thinning of the cuticle by 59.3% and 53.4% (P<0.05), as observed through transmission electron microscopy and eosin staining, in the S and R strains, respectively. Overall, our findings demonstrate that the phenotypic plasticity in response to short-term stress can reshape the adaptive mechanisms of genetic variation during prolonged exposure to insecticides. And the identified resistance-related genes, CUO8 and Cyp4g19, could serve as potential targets for controlling these pest populations.

Identification of a morphogene required for tapered filament termini in filamentous cyanobacteria.

Although the photosynthetic cyanobacteria are monophyletic, they exhibit substantial morphological diversity across species, and even within an individual species due to phenotypic plasticity in response to life cycles and environmental signals. This is particularly prominent among the multicellular filamentous cyanobacteria. One example of this is the appearance of tapering at the filament termini. However, the morphogenes controlling this phenotype and the adaptive function of this morphology are not well defined. Here, using the model filamentous cyanobacterium Nostoc punctiforme ATCC29133 (PCC73102), we identify tftA, a morphogene required for the development of tapered filament termini. The tftA gene is specifically expressed in developing hormogonia, motile trichomes where the tapered filament morphology is observed, and encodes a protein containing putative amidase_3 and glucosaminidase domains, implying a function in peptidoglycan hydrolysis. Deletion of tftA abolished filament tapering inidcating that TftA plays a role in remodelling the cell wall to produce tapered filaments. Genomic conservation of tftA specifically in filamentous cyanobacteria indicates this is likely to be a conserved mechanism among these organisms. Finally, motility assays indicate that filaments with tapered termini migrate more efficiently through dense substratum, providing a plausible biological role for this morphology.

Sharks and their relatives: can their past help predict their future?

Elasmobranchs (i.e., sharks, skates, and rays) have survived five mass extinction events and changed relatively little throughout their ~450-million-year evolutionary history. Therefore, elasmobranchs may provide critical evolutionary perspectives on how species and populations can elicit phenotypic plasticity and adaptation responses to climate change. Unfortunately, despite their roles as critical apex- and meso-predators, most elasmobranch species are considered to be highly vulnerable to the impacts of fisheries exploitation and climate change, which is compounded by their K-selected life history strategies. Furthermore, the future of elasmobranchs is uncertain at best in the face of anthropogenic climate change because there have only been a handful of studies that have directly investigated the effects of climate change related stressors. Phenotypic plasticity in response to climate change, specifically ocean warming, may be a species’ best chance of resilience given the expedited rate of environmental change. However, despite extensive research on plasticity within and across generations in teleost fishes, there remains a knowledge gap for elasmobranch species, owing to their extended life spans and delayed sexual maturity. Here, we present four case studies on different elasmobranch species to lend perspectives on the capacity for phenotypic plasticity within the context of ocean warming. Furthermore, we discuss potential research avenues and modern technologies that may enable future investigations to empirically explore the capacity for phenotypic plasticity in elasmobranchs.

Endocrine-disrupting chemicals and epigenetic reprogramming in developmental origin of uterine fibroids.

Endocrine-disrupting chemicals (EDCs) are a class of exogenous substances that mimic the effects of hormones in the body, inducing hormonal dysregulation and contributing to various disorders. Epigenome regulation has emerged as an important mechanism for maintaining organ function in health and disease. Dissecting epigenomic and resultant gene expression changes provides unprecedented insight into the chromatin state, which underlines disease development and shapes risk and phenotypic plasticity in response to the environment and internal cues. The cutting-edge, high throughput technologies provide new routes to understanding the etiology of disease and new footholds on the promising path to better treatment and disease prevention. We have recently revealed that myometrial stem cells (MMSCs), the cell origin of UFs, are the target of developmental EDC exposure. The EDC-induced epigenetic changes in MMSCs identified by multi-omics approaches include DNA methylation and histone modification modulated by DNA methyltransferases and MLL1, which characterized the molecular mechanism underlying EDC-related risk in hormone-dependent UFs. Future studies are needed to determine the link between real-life exposures to EDCs and their impact on the development of human diseases and transgenerational epigenetic inheritance, which can help explore strategies that may prevent adverse outcomes linked to EDC exposure.

Climatic Stress Test of Scots Pine Provenances in Northeastern Europe Reveals High Phenotypic Plasticity and Quasi-Linear Response to Warming

Research Highlights: Scots pine (Pinus sylvestris L.) phenotypic plasticity will buffer and even benefit from temperature increases in Northeast Europe this century, except for the southern peripheries of the range. Objectives: The “stress test” aimed to assess the inherent potential of existing populations to withstand projected changes in their lifetimes at their original location. Materials and Methods: This study applied an alternative analytic approach to calculate response and transfer equations from historic height growth data from provenance tests in the former USSR and Hungary. Results: Contrary to earlier analyses, the populations displayed quasi-linear responses to mimicked warming without clear ecological optima, forecasting a general growth acceleration north of Lat. 53° N. Climate-triggered mortality is predicted for the near future in the southern peripheries. Locally adapted populations at the distribution confines of the northern and southern limits deserve special attention. Conclusions: The observed adaptability to warming moderates the necessity of genetic management interventions such as assisted migration. The support of natural processes of adaptation and acclimation will be sufficient in boreal and central Northeast Europe this century. Evacuating heat and drought-tolerant populations should be envisaged in the endangered zone to conserve valuable genetic resources.

Phenotypic plasticity of growth ring traits in Pinus hartwegii at the ends of its elevational gradient.

Phenotypic plasticity (PP) could be an important short-term mechanism to modify physiological and morphological traits in response to climate change and global warming, particularly for high-mountain tree species. The objective was to evaluate PP response of growth ring traits to temperature and precipitation in Pinus hartwegii Lindl. populations located at the ends of its elevational gradient on two volcanic mountains in central Mexico (La Malinche and Nevado de Toluca). Increment cores collected from 274 P. hartwegii trees were used to estimate their PP through reaction norms (RN), which relate the ring width and density traits with climate variables (temperature and precipitation). We estimated the trees' sensitivity (significant RN) to climatic variables, as well as the relative proportion of RN with positive and negative slope. We also estimated the relationship between the PP of ring width and density traits using correlation and Principal Component (PC) analyses. Over 70% of all trees showed significant RN to growing season and winter temperatures for at least one growth ring trait, with a similar proportion of significant RN at both ends of the gradient on both mountains. Ring width traits had mostly negative RN, while ring density traits tended to have positive RN. Frequency of negative RN decreased from lower to higher elevation for most traits. Average PP was higher at the lower end of the gradient, especially on LM, both for ring width and ring density traits, although high intrapopulation variation in PP was found on both mountains. Results indicate that P. hartwegii presents spatially differentiated plastic responses in width and density components of radial growth. PP was particularly strong at the lower elevation, which has higher temperature and water stress conditions, putting these populations at risk from the continuing global warming driven by climate change.
