Characterization of the Effect of a Novel Production Technique for 'Not from Concentrate' Pear and Apple Juices on the Composition of Phenolic Compounds.

The consumption of 'not-from-concentrate' (NFC) fruit juices can be a convenient and enjoyable way to incorporate the nutritional benefits and flavors of fruits into one's diet. This study will focus on the effect of production of juices from apple and pear fruits, by using centrifugal decanter and tangential filtration, on the profile of polyphenols as a valuable source of bioactive compounds. Likewise, by-products from the juice industry were characterized in order to understand the high-value-added potential based on their composition of polyphenols. Briefly, apple and apple juice showed great contents of chlorogenic acid (0.990 ± 0.021 mg/g of DW), the dihydrochalcone phloridzin (1.041 ± 0.062 mg/g of DW), procyanidins (0.733 ± 0.121 mg/g of DW) and quercetin derivatives (1.501 ± 0.192 mg/g of DW). Likewise, the most abundant compounds in pear and pear juices were chlorogenic acid (0.917 ± 0.021 mg/g of DW), caffeoylquinic acid (0.180 ± 0.029 mg/g of DW), procyanidins (0.255 ± 0.016 mg/g of DW) and quercetin derivatives (0.181 ± 0.004 mg/g of DW). Both temperature and tangential speed affect the amount of phenolic compounds in fruit juices, highlighting the need to control the technological process to obtain a more nutritious/healthier beverage. Overall, NFC juices arise as a better option when compared with concentrated juices. Furthermore, the higher yield of phenolic compounds found in fruit pomace clearly open new ways for upcycling this fruit by-product as a high-value-added ingredient.

Fabrication of montmorillonite/ionic liquid composite coated solid-phase microextraction fibers for determination of phenolic compounds in fruit juices by gas chromatography and liquid chromatography

Novel montmorillonite composites including ionic liquid were prepared and utilized as coating materials for solid phase microextraction fibers. Ionic liquids containing amino terminated imidazolium cations with methyl and benzyl groups and hydrophobic anions (bis(trifluoromethylsulfonyl)imide) and hydrophilic anions (bromide and tetrafluoroborate) were intercalated to KSF-montmorillonite using stainless steel wire as support by using the layer-by-layer technique. After optimization of experimental conditions, solid phase microextraction (SPME) coupled to gas chromatography/mass spectrometry (SPME-GC/MS) after derivatization by silylation and high performance liquid chromatography/diode array detector (SPME-HPLC/DAD) were developed for the determination of 16 phenolic compounds. The developed SPME-GC/MS method had wider linearity and lower limit of detection than the developed SPME-HPLC/DAD method with similar repeatability (for 5 runs, less than 4.2 %) for a single fiber. The percentage of the variance between two different fibers was less than 6.0 % in both methods. The developed SPME methods were applied successfully to fresh fruit juices and the relative recoveries and the repeatabilities from spiked fruit juice samples were satisfactorily achieved. The results obtained with both chromatographic methods were in good agreement with each other.

Color, physicochemical parameters and antioxidant potential of whole grape juices subject to different UV-C radiation doses

ABSTRACT Knowing that moderate stress such as UV radiation can activate defense mechanisms in plants, the use of UV-C radiation appears as hypothesis of a promising technique that would help to stimulate and enhance beneficial compounds for health, through a clean and healthy technology. In this study, the possible induction of secondary metabolism, the increase in the content of phytochemical compounds and physicochemical changes through the use of UV-C radiation were evaluated on whole grape juices produced with Vitis labrusca grapes, cultivar Isabel Precoce. Grapes were harvested, sanitized, exposed to UV-C radiation at doses of 0, 2, 4 and 6 KJ m-2, and then the juices were prepared and packed into amber glass bottles at room temperature. Analyses were performed at 0, 30, 60, 90 and 120 days of storage. Based on results obtained and conditions in which the experiment was performed, UV-C treatment in grapes caused abiotic stress in the fruits, affecting color, titratable acidity, soluble solids/titratable acidity ratio, vitamin C and percentage of protection against oxidation. Application of UV-C did not change levels of phenolic compounds in fruit juices or the percentage of scavenging free radicals, pH and soluble solids.

A general analytical strategy for the characterization of phenolic compounds in fruit juices by high-performance liquid chromatography with diode array detection coupled to electrospray ionization and triple quadrupole mass spectrometry

In the present study, a methodology based on liquid chromatography with diode array detection (HPLC/DAD) coupled to an electrospray ionization (ESI) interface and a triple quadrupole mass spectrometer for the simultaneous identification of phenolic compounds in fruit juices has been developed. 72 available phenolic compound standards from diverse families present in fruits have been studied in order to analyze their fragmentation pattern. As a result, a general strategy for the characterization of unknown phenolic compounds in fruit juices was designed: (i) taking into account its UV–visible spectrum and elution order, assign the unknown polyphenol to a polyphenol class, (ii) identify the quasi-molecular ion using positive and negative MS spectra, being supported by adducts generated with solvent or sodium and molecular complexes, (iii) determinate the pattern of glycosylation in positive mode using ESI(+)-CID MS/MS product ion scan experiments, selecting the quasi-molecular ion as precursor ion, and finally, (iv) study the identity of the aglycone through ESI(+)-CID MS/MS product ion spectra from the protonated aglycone, [Y 0] +. This strategy was successfully employed for the characterization of known and unknown phenolic compounds in juices from 17 different fruits.

INFLUENCE OF CONVENTIONAL CLARIFICATION AND ULTRAFILTRATION ON THE PHENOLIC COMPOSITION OF GOLDEN DELICIOUS APPLE JUICE

Abstract The effects of conventional clarification and ultrafiltration techniques on the phenolic composition of apple juice was studied. Since the health benefit of fruits and fruit juices have been ascribed, in part, to the phenolic antioxidants, it is very important to know what the effects of processing steps are on the level of phenolic compounds in fruit juices. The total losses of phenolic compounds were determined to be 28% and 7% during conventional clarification using gelatin and bentonite, and ultrafiltration, respectively. A total of 14 phenolic compounds were identified in apple juice prepared from Golden Delicious apples. The phenolic compounds were found to be based mainly on hydroxycinnamic acids (∼ 50% of total phenolics) and chlorogenic acid was found to be the major phenolic compound with its concentration as high as 465.45 mg 1−1 in raw apple juice.

Separation and determination of organic acids and phenolic compounds in fruit juices and drinks by high-performance liquid chromatography.

A high-performance liquid chromatographic (HPLC) separation method with photo-diode array detection has been developed for the simultaneous determination of organic acids and phenolic compounds in juices and drinks. The chromatographic analysis of organic acids and phenolic compounds was carried out after their elution with sulphuric acid solution (pH 2.5) and methanol from C18 stationary phase. The mobile phase employed was sulphuric acid solution working at a flow-rate of 0.35 ml min(-1) for the whole run, while methanol was linearly increased to 0.45 ml min(-1) from 15 to 75 min followed by a 5-min isocratic elution. Ten organic acid acids were eluted in 30 min and 21 phenolic compounds, which include phenolic acids and flavonoids, were eluted in the following 50 min. Target compounds were detected at 215 nm. The repeatability (n=3) and between day precision of peak area (n=3) were all within 5.0% RSD. The within-day repeatability (n=3) and between-day precision (n=10) of retention times were within 0.3 and 1.6% relative standard deviation (RSD), respectively. The accuracy of the method was confirmed with an average recovery ranging between 85 and 106%. The method was successfully used to measure a variety of organic acids and phenolic compounds in juices and beverages. This method could also be used to evaluate the authenticity, spoilage or micronutrient contents of juices.
