Learning from Projection to Reconstruction: A Deep Learning Reconstruction Framework for Sparse-View Phase Contrast Computed Tomography via Dual-Domain Enhancement

Phase contrast computed tomography (PCCT) provides an effective non-destructive testing tool for weak absorption objects. Limited by the phase stepping principle and radiation dose requirement, sparse-view sampling is usually performed in PCCT, introducing severe artifacts in reconstruction. In this paper, we report a dual-domain (i.e., the projection sinogram domain and image domain) enhancement framework based on deep learning (DL) for PCCT with sparse-view projections. It consists of two convolutional neural networks (CNN) in dual domains and the phase contrast Radon inversion layer (PCRIL) to connect them. PCRIL can achieve PCCT reconstruction, and it allows the gradients to backpropagate from the image domain to the projection sinogram domain while training. Therefore, parameters of CNNs in dual domains are updated simultaneously. It could overcome the limitations that the enhancement in the image domain causes blurred images and the enhancement in the projection sinogram domain introduces unpredictable artifacts. Considering the grating-based PCCT as an example, the proposed framework is validated and demonstrated with experiments of the simulated datasets and experimental datasets. This work can generate high-quality PCCT images with given incomplete projections and has the potential to push the applications of PCCT techniques in the field of composite imaging and biomedical imaging.

High-Resolution X-ray Phase-Contrast Imaging and Sensory and Rheometer Tests in Cooked Edamame.

Although several reports exist on the use of X-ray analysis in vegetables and fruits to examine internal disorders, cavities, and porosity, information on X-ray analysis of qualities, such as texture, is lacking as well as information on X-ray analysis of legumes. Therefore, this study aimed to perform X-ray analysis with sensory and rheometer tests in cooked vegetable soybean (edamame). Edamame is popular worldwide due to its deliciousness and nutritional value. Vascular structures and cracks around them were clearly visualized using X-ray phase-contrast computed tomography (CT) imaging. In addition, we observed the fine structure of the seed coat, which could be important for seed development, germination, and processing. The density in the edamame beans declined as the boiling time increased, promoting a reduction in hardness described in sensory and rheometer tests. The reduction in density proceeded from the gap between cotyledons, the opposite side of the hypocotyl, and the crack. Collectively, the findings show that the high-resolution X-ray phase-contrast CT imaging conducted in a nondestructive manner may help in effectively evaluating the quality of vegetables and in observing the internal structures related to plant development.

Prototype system of noninterferometric phase-contrast computed tomography utilizing medical imaging components

Grating-based x-ray phase-contrast imaging has been demonstrated to provide more information and higher-contrast images for low-Z soft tissues, compared with conventional absorption-based imaging. However, the existing Talbot–Lau phase-contrast devices are operated in either a two- or three-dimensional mode at low energy with a small field of view and long exposure time. This is because of coherence limitations, difficulties in fabricating high aspect ratio gratings, and the slow readout speed of the detector. For preclinical or even clinical applications, a variable x-ray energy, a large field of view, and fast phase-contrast computed tomography (CT) devices are desirable. The noninterferometric grating-based phase-contrast imaging method is a good candidate, as it relaxes requirements on gratings, including grating period and aspect ratio. Based on the noninterferometric imaging principle, we constructed a prototype phase-contrast CT system, at the National Synchrotron Radiation Laboratory of the University of Science and Technology of China, with medical imaging components. This prototype system enables a large field of view and fast phase-contrast CT imaging under medical imaging energies. In this paper, the prototype system and preliminary experimental results are reported, and possible optimization for forthcoming work is also discussed.

Quantifying the hydroxyapatite orientation near the ossification front in a piglet femoral condyle using X-ray diffraction tensor tomography

While a detailed knowledge of the hierarchical structure and morphology of the extracellular matrix is considered crucial for understanding the physiological and mechanical properties of bone and cartilage, the orientation of collagen fibres and carbonated hydroxyapatite (HA) crystallites remains a debated topic. Conventional microscopy techniques for orientational imaging require destructive sample sectioning, which both precludes further studies of the intact sample and potentially changes the microstructure. In this work, we use X-ray diffraction tensor tomography to image non-destructively in 3D the HA orientation in a medial femoral condyle of a piglet. By exploiting the anisotropic HA diffraction signal, 3D maps showing systematic local variations of the HA crystallite orientation in the growing subchondral bone and in the adjacent mineralized growth cartilage are obtained. Orientation maps of HA crystallites over a large field of view (~ 3 × 3 × 3 mm3) close to the ossification (bone-growth) front are compared with high-resolution X-ray propagation phase-contrast computed tomography images. The HA crystallites are found to predominantly orient with their crystallite c-axis directed towards the ossification front. Distinct patterns of HA preferred orientation are found in the vicinity of cartilage canals protruding from the subchondral bone. The demonstrated ability of retrieving 3D orientation maps of bone-cartilage structures is expected to give a better understanding of the physiological properties of bones, including their propensity for bone-cartilage diseases.

Method of reconstructing region of interest for differential phase contrast computed tomography imaging

X-ray differential phase contrast computed tomography imaging based on grating interferometer system can reconstruct not only the linear attenuation coefficient, but also the phase shift coefficient and the linear scattering coefficient of the object. In practical application, it is very difficult to make a large area grating, so the sample is often larger than the grating. When the sample is scanned with a grating smaller than the sample, the part of the sample beyond the field of view of the grating will cause the differential phase projection information to be truncated. In this paper, a method of reconstructing the region of interest for differential phase contrast computed tomography is proposed. The method is based on the approximate linear relation between the phase shift coefficient of the object and the linear attenuation coefficient (i.e. the decrement in the real part of the refractive index and the imaginary part of the refractive index), the phase shift coefficient of the region of interest is approximately reconstructed by the polynomial of Lambda function of the phase shift coefficient and Lambda inverse function of linear attenuation coefficient. In this paper, according to the Fresnel diffraction theory and differential phase grating phase step-by-step method of imaging a simulation experiment is performed. In the experiment, conducted is the approximate reconstruction by using the first order polynomial and quadratic polynomial of Lambda function of the phase shift coefficient and Lambda inverse function of linear attenuation coefficient. The sample size is five times of grating imaging field, and the results show that this method can approximately reconstruct the region of interest for the sample image. We also carry out the actual data experiment. The actual data are obtained by the Talbot grating interferometer system of Shanghai synchrotron radiation BL13W1 station, and the standard model and biological sample are imaged. The method of reconstructing the region of interest is proposed in this paper. This method can be applied to the multi-material samples with a similar relationship between the decrement in the real part of the refractive index and the decrement in the imaginary part of the refractive index, and also to single-material samples. The comparison between the numerical simulations and the actual experimental results verifies the effectiveness of the proposed method.

Unsupervised/Supervised Hybrid Deep Learning Framework for Low Dose Phase Contrast Imaging

X-ray phase contrast computed tomography (PCCT) has better imaging quality than conventional attenuation X-ray CT and has demonstrated promising application prospects in medical diagnosis. However, reducing the radiation dose during PCCT imaging still remains a major challenge. Recently, deep learning (DL) techniques have been applied to low dose CT and obtain significant progress. Most of them require massive paired images to train the network in a supervised manner, which may hamper their practical applications because the ground-truth images are hard to be obtained in most cases. To address this issue, we report a hybrid deep learning framework for low dose PCCT which capsules unsupervised and supervised learning manners. It combines the advantages of convolutional neural network (CNN) and total variation (TV) and is suitable for both unlabelled datasets and labelled datasets. This framework has been validated and demonstrated with experimental data. It will be helpful to push the practical application of low dose PCCT.

CT Radiomics for Distinction of Human Epidermal Growth Factor Receptor 2 Negative Gastric Cancer

The purpose of this study was to investigate the role of computed tomography (CT) radiomics for the prediction of the human epidermal growth factor 2 (HER2) status in patients with gastric cancer. One hundred and thirty two consecutive patients with advanced gastric cancer undergoing radical gastrectomy were retrospectively reviewed. All patients received preoperative contrast CT examination, and immunohistochemistry results of their HER2 status were available. All the subjects were randomly divided into a training cohort (n = 90) and a test cohort (n = 42). Arterial phase (AP) and portal phase (PP) contrast CT images were retrieved for tumor segmentation and feature extraction. Receiver operating characteristics (ROC) curves and area under the curve (AUC) were used to evaluate the performance of the radiomics classifiers. Among the 132 patients, a total of 99 patients were HER2 negative, and the remaining 33 patients were border line or positive. The AP radiomics model could distinguish HER2-negative cases with an AUC of 0.756 (95% confidence interval [CI]: 0.656-0.840) in the training cohort, which was confirmed in the test cohort with AUC of 0.830 (95% CI: 0.678-0.930). The PP radiomics model showed AUCs of 0.715 (95% CI: 0.612-0.804) and 0.718 (95% CI: 0.554-0.849) in the training and test cohort for distinction of negative HER2 cases, respectively. Radiomics models based on standard-of-care CT images hold promise for distinguishing HER2-negative gastric cancer.

Quantitative characterization of ex vivo breast tissue via x-ray phase-contrast tomography.

Grating-based X-ray phase-contrast imaging (GPCI) has received growing interests in recent years due to its high capability of visualizing soft tissue. Breast imaging is one of the most promising candidates for the first clinical application of this imaging modality. In this work, quantitative breast tissue characterization based on GPCI computed tomography (CT) is investigated with a laboratory X-ray tube through a comparison between attenuation-based CT images and phase-contrast CT images. The Hounsfield units (HU) scale was introduced to phase-contrast images due to its wide application in clinical medicine. In this work, instead of water, plastic cylinders composed of polyethylene terephthalate (PET) was treated as the calibration material. An alternative test-retest reliability (TRR) was presented to evaluate the repeatability of GPCI. Comparison between attenuation-based CT imaging and GPCI CT imaging was operated with the use of statistical analysis methods like histograms and receiver operating characteristic (ROC) curves. The determined mean TRR related to cylinders is slightly larger in phase-contrast imaging (0.93) than that in attenuation-based imaging (0.89). With respect to distinguishing breast tissues, the AUC (area under curve) values of ROC curves of phase-contrast images are higher than that of attenuation-based images. An ex vivo study of GPCI shows that it is a stable imaging modality for visualizing the breast tissue with good repeatability, and that it could be of potential for the diagnosis of breast cancer as well.

Three-dimensional visualization of fibrous tissues in cirrhotic rats via X-ray phase-contrast computed tomography with iodine staining.

To accurately characterize cirrhosis, knowledge of the 3D fibrous structures is essential. Histology is the gold standard in cirrhosis screening, but it mainly provides structural information in 2D planes and destroys the 3D samples in the process. The aim of this study was to evaluate the potential of X-ray phase-contrast computed tomography (PCCT) with iodine staining for the 3D nondestructive visualization of internal structural details in entire cirrhotic livers with histopathologic correlation. In this study, cirrhotic livers induced by carbon tetrachloride (CCl4) in rats were imaged via PCCT and then histopathologically processed. Characteristics of the cirrhosis, i.e. abnormal nodules surrounded by annular fibrosis, were established and a 3D reconstruction of these structures was also performed via PCCT. Fibrosis area, septal width and nodular size were measured and the correlation for these quantitative measurements between PCCT and histopathologic findings was analyzed. The results showed that fibrous bands, small nodules and angio-architecture in cirrhosis were clearly presented in the PCCT images, with histopathologic findings as standard reference. In comparison with histopathology, PCCT was associated with a very close value for fibrosis area, septal width and nodular size. The quantitative measurements showed a strong correlation between PCCT and histopathology. Additionally, the 3D structures of fibrous bands and microvasculature were presented simultaneously. PCCT provides excellent results in the assessment of cirrhosis characteristics and 3D presentation of these feature structures compared with histopathology. Thus, the technique may serve as an adjunct nondestructive 3D modality for cirrhosis characterization.

Large-area single-photon-counting CdTe detector for synchrotron radiation computed tomography: adedicated pre-processing procedure.

Large-area CdTe single-photon-counting detectors are becoming more and more attractive in view of low-dose imaging applications due to their high efficiency, low intrinsic noise and absence of a scintillating screen which affects spatial resolution. At present, however, since the dimensions of a single sensor are small (typically a few cm2), multi-module architectures are needed to obtain a large field of view. This requires coping with inter-module gaps and with close-to-edge pixels, which generally show a non-optimal behavior. Moreover, high-Z detectors often show gain variations in time due to charge trapping: this effect is detrimental especially in computed tomography (CT) applications where a single tomographic image requires hundreds of projections continuously acquired in several seconds. This work has been carried out at the SYRMEP beamline of the Elettra synchrotron radiation facility (Trieste, Italy), in the framework of the SYRMA-3D project, which aims to perform the world's first breast-CT clinical study with synchrotron radiation. An ad hoc data pre-processing procedure has been developed for the PIXIRAD-8 CdTe single-photon-counting detector, comprising an array of eight 30.7 mm × 24.8 mm modules tiling a 246 mm × 25 mm sensitive area, which covers the full synchrotron radiation beam. The procedure consists of five building blocks, namely dynamic flat-fielding, gap seaming, dynamic ring removal, projection despeckling and around-gap equalization. Each block is discussed and compared, when existing, with conventional approaches. The effectiveness of the pre-processing is demonstrated for phase-contrast CT images of a human breast specimen. The dynamic nature of the proposed procedure, which provides corrections dependent upon the projection index, allows the effective removal of time-dependent artifacts, preserving the main image features including phase effects.

MÖNCH detector enables fast and low-dose free-propagation phase-contrast computed tomography of in situ mouse lungs.

Due to the complexity of the underlying pathomechanism, in vivo mouse lung-disease models continue to be of great importance in preclinical respiratory research. Longitudinal studies following the cause of a disease or evaluating treatment efficacy are of particular interest but challenging due to the small size of the mouse lung and the fast breathing rate. Synchrotron-based in-line phase-contrast computed tomography imaging has been successfully applied in lung research in various applications, but mostly at dose levels that forbid longitudinal in vivo studies. Here, the novel charge-integrating hybrid detector MÖNCH is presented, which enables imaging of mouse lungs at a pixel size of 25 µm, in less than 10 s and with an entrance dose of about 70 mGy, which therefore will allow longitudinal lung disease studies to be performed in mouse models.

Three-dimensional visualization and pathologic characteristics of cartilage and subchondral bone changes in the lumbar facet joint of an ovariectomized mouse model

Background ContextLow back pain (LBP) is more prevalent among postmenopausal women than men. Ovariectomy (OVX) is an established animal model that mimics the estrogen deficiency of postmenopausal women. Little is known about the three-dimensional (3D) morphologic properties of cartilage and subchondral bone changes in the lumbar facet joint (LFJ) of an OVX mouse model. PurposeThe purpose of this study was to characterize the 3D morphologic change of cartilage and subchondral bone in the LFJ of an OVX mouse model. Study DesignThree-dimensional visualization and a histologic study on degenerative changes in cartilage and subchondral bone in the LFJ of an OVX mouse model were conducted. Materials and MethodsOvariectomy is performed to mimic postmenopausal changes in adult female mice. We present an imaging tool for 3D visualization of the pathologic characteristics of cartilage and subchondral bone changes LFJ degradation using propagation-based phase-contrast computed tomography (PPCT). The samples were further dissected, fixed, and stained for histologic examination. ResultsPropagation-based phase-contrast computed tomography imaging provides a 3D visualization of altered cartilage with a simultaneous high detail of the subchondral bone abnormalities in an OVX LFJ model. A quantitative analysis demonstrated that the cartilage volume, the surface area, and thickness were decreased in the OVX group compared with the control group (p<.05). Meanwhile, these decreases were accompanied by an obvious destruction of the subchondral bone surface and a loss of trabecular bone in the OVX group (p<.05). The delineation of the 3D pathologic changes in the PPCT imaging was confirmed by a histopathologic method with Safranin-O staining. Tartrate-resistant acid phosphatase staining revealed an increased number of osteoclasts in the subchondral bone of the OVX mice compared with that of the control group. ConclusionsThese results demonstrated that a mouse model of OVX-induced LFJ osteoarthritis (OA)-like changes was successfully established and showed a good resemblance to the human OA pathology. Propagation-based phase-contrast computed tomography has great potential to becomea powerful 3D imaging method to comprehensively characterize LFJ OA and to effectively monitor therapeutics. Moreover, degenerative LFJ possesses a severe morphologic change in the subchondral bone, may be the source of postmenopausal LBP, and has the potential to be a novel therapeutic target for LBP treatment.

High-resolution 3D visualization of ductular proliferation of bile duct ligation-induced liver fibrosis in rats using x-ray phase contrast computed tomography

X-ray phase-contrast computed tomography (PCCT) can provide excellent image contrast for soft tissues with small density differences, and it is particularly appropriate for three-dimensional (3D) visualization of accurate microstructures inside biological samples. In this study, the morphological structures of proliferative bile ductules (BDs) were visualized without contrast agents via PCCT with liver fibrosis samples induced by bile duct ligation (BDL) in rats. Adult male Sprague-Dawley rats were randomly divided into three groups: sham operation group, 2-week and 6-week post-BDL groups. All livers were removed after euthanasia for a subsequent imaging. The verification of the ductular structures captured by PCCT was achieved by a careful head-to-head comparison with their corresponding histological images. Our experimental results demonstrated that PCCT images corresponded very well to the proliferative BDs shown by histological staining using cytokeratin 19 (CK19). Furthermore, the 3D density of proliferative BDs increased with the progression of liver fibrosis. In addition, PCCT accurately revealed the architecture of proliferative BDs in a 3D fashion, including the ductular ramification, the elongation and tortuosity of the branches, and the corrugations of the luminal duct surface. Thus, the high-resolution PCCT technique can improve our understanding of the characteristics of ductular proliferation from a new 3D perspective.

Phase-contrast CT: Qualitative and Quantitative Evaluation of Capillarized Sinusoids and Trabecular Structure in Human Hepatocellular Carcinoma Tissues

Capillarization of sinusoids and change of trabecular thickness are the main histologic features in hepatocellular carcinoma (HCC). Of particular interest are the three-dimensional (3D) visualization and quantitative evaluation of such alterations in the HCC progression. X-ray phase-contrast computed tomography (PCCT) is an emerging imaging method that provides excellent image contrast for soft tissues. This study aimed to explore the potential of in-line PCCT in microstructure imaging of capillarized sinusoids and trabecular structure in human HCC tissues and to quantitatively evaluate the alterations of those fine structures during the development of HCC. This project was designed as an ex vivo experimental study. The study was approved by the institutional review board, and informed consent was obtained from the patients. Eight human resected HCC tissue samples were imaged using in-line PCCT. After histologic processing, PCCT images and histopathologic data were matched. Fine structures in HCC tissues were revealed. Quantitative analyses of capillarized sinusoids (ie, percentage of sinusoidal area [PSA], sinusoidal volume) and trabecular structure (ie, trabecular thickness, surface-area-to-volume ratio [SA/V]) in low-grade (well or moderately differentiated) and high-grade (poorly differentiated) HCC groups were performed. Using PCCT, the alterations of capillarized sinusoids and trabecular structure were clearly observed in 3D geometry, which was confirmed by the corresponding histologic sections. The 3D qualitative analyses of sinusoids in the high-grade HCC group were significantly different (P < 0.05) in PSA (7.8 ± 2.5%) and sinusoidal volume (2.9 ± 0.6 × 107 µm3) from those in the low-grade HCC group (PSA, 12.9 ± 2.2%; sinusoidal volume, 2.4 ± 0.3 × 107 µm3). Moreover, the 3D quantitative evaluation of the trabecular structure in the high-grade HCC group showed a significant change (P < 0.05) in the trabecular thickness (87.8 ± 15.6 µm) and SA/V (2.2 ± 1.3 × 103 µm-1) compared to the low-grade HCC group (trabecular thickness, 75.9 ± 7.1 µm; SA/V, 7.5 ± 1.3 × 103 µm-1). This study provides insights into the 3D alterations of microstructures such as capillarized sinusoids and the trabecular structure at a micrometer level, which might allow for an improved understanding of the development of HCC.

Three Dimensional Visualization of Human Cardiac Conduction Tissue in Whole Heart Specimens by High-Resolution Phase-Contrast CT Imaging Using Synchrotron Radiation.

The feasibility of synchrotron radiation-based phase-contrast computed tomography (PCCT) for visualization of the atrioventricular (AV) conduction axis in human whole heart specimens was tested using four postmortem structurally normal newborn hearts obtained at autopsy. A PCCT imaging system at the beamline BL20B2 in a SPring-8 synchrotron radiation facility was used. The PCCT imaging of the conduction system was performed with "virtual" slicing of the three-dimensional reconstructed images. For histological verification, specimens were cut into planes similar to the PCCT images, then cut into 5-μm serial sections and stained with Masson's trichrome. In PCCT images of all four of the whole hearts of newborns, the AV conduction axis was distinguished as a low-density structure, which was serially traceable from the compact node to the penetrating bundle within the central fibrous body, and to the branching bundle into the left and right bundle branches. This was verified by histological serial sectioning. This is the first demonstration that visualization of the AV conduction axis within human whole heart specimens is feasible with PCCT.

Statistical iterative reconstruction algorithm for X-ray phase-contrast CT.

Grating-based phase-contrast computed tomography (PCCT) is a promising imaging tool on the horizon for pre-clinical and clinical applications. Until now PCCT has been plagued by strong artifacts when dense materials like bones are present. In this paper, we present a new statistical iterative reconstruction algorithm which overcomes this limitation. It makes use of the fact that an X-ray interferometer provides a conventional absorption as well as a dark-field signal in addition to the phase-contrast signal. The method is based on a statistical iterative reconstruction algorithm utilizing maximum-a-posteriori principles and integrating the statistical properties of the raw data as well as information of dense objects gained from the absorption signal. Reconstruction of a pre-clinical mouse scan illustrates that artifacts caused by bones are significantly reduced and image quality is improved when employing our approach. Especially small structures, which are usually lost because of streaks, are recovered in our results. In comparison with the current state-of-the-art algorithms our approach provides significantly improved image quality with respect to quantitative and qualitative results. In summary, we expect that our new statistical iterative reconstruction method to increase the general usability of PCCT imaging for medical diagnosis apart from applications focused solely on soft tissue visualization.

X-ray phase contrast tomography by tracking near field speckle.

X-ray imaging techniques that capture variations in the x-ray phase can yield higher contrast images with lower x-ray dose than is possible with conventional absorption radiography. However, the extraction of phase information is often more difficult than the extraction of absorption information and requires a more sophisticated experimental arrangement. We here report a method for three-dimensional (3D) X-ray phase contrast computed tomography (CT) which gives quantitative volumetric information on the real part of the refractive index. The method is based on the recently developed X-ray speckle tracking technique in which the displacement of near field speckle is tracked using a digital image correlation algorithm. In addition to differential phase contrast projection images, the method allows the dark-field images to be simultaneously extracted. After reconstruction, compared to conventional absorption CT images, the 3D phase CT images show greatly enhanced contrast. This new imaging method has advantages compared to other X-ray imaging methods in simplicity of experimental arrangement, speed of measurement and relative insensitivity to beam movements. These features make the technique an attractive candidate for material imaging such as in-vivo imaging of biological systems containing soft tissue.

X-ray phase-contrast computed tomography visualizes the microstructure and degradation profile of implanted biodegradable scaffolds after spinal cord injury

Tissue engineering strategies for spinal cord repair are a primary focus of translational medicine after spinal cord injury (SCI). Many tissue engineering strategies employ three-dimensional scaffolds, which are made of biodegradable materials and have microstructure incorporated with viable cells and bioactive molecules to promote new tissue generation and functional recovery after SCI. It is therefore important to develop an imaging system that visualizes both the microstructure of three-dimensional scaffolds and their degradation process after SCI. Here, X-ray phase-contrast computed tomography imaging based on the Talbot grating interferometer is described and it is shown how it can visualize the polyglycolic acid scaffold, including its microfibres, after implantation into the injured spinal cord. Furthermore, X-ray phase-contrast computed tomography images revealed that degradation occurred from the end to the centre of the braided scaffold in the 28 days after implantation into the injured spinal cord. The present report provides the first demonstration of an imaging technique that visualizes both the microstructure and degradation of biodegradable scaffolds in SCI research. X-ray phase-contrast imaging based on the Talbot grating interferometer is a versatile technique that can be used for a broad range of preclinical applications in tissue engineering strategies.

A sparse-projection computed tomography reconstruction method for in vivo application of in-line phase-contrast imaging

BackgroundIn recent years, X-ray phase-contrast imaging techniques have been extensively studied to visualize weakly absorbing objects. One of the most popular methods for phase-contrast imaging is in-line phase-contrast imaging (ILPCI). Combined with computed tomography (CT), phase-contrast CT can produce 3D volumetric images of samples. To date, the most common reconstruction method for phase-contrast X-ray CT imaging has been filtered back projection (FBP). However, because of the impact of respiration, lung slices cannot be reconstructed in vivo for a mouse using this method. Methods for reducing the radiation dose and the sampling time must also be considered.MethodsThis paper proposes a novel method of in vivo mouse lung in-line phase-contrast imaging that has two primary improvements compared with recent methods: 1) using a compressed sensing (CS) theory-based CT reconstruction method for the in vivo in-line phase-contrast imaging application and 2) using the breathing phase extraction method to address the lung and rib cage movement caused by a live mouse’s breathing.ResultsExperiments were performed to test the breathing phase extraction method as applied to the lung and rib cage movement of a live mouse. Results with a live mouse specimen demonstrate that our method can reconstruct images of in vivo mouse lung.ConclusionsThe results demonstrate that our method could deal with vivo mouse’s breathing and movements, meanwhile, using less sampling data than FBP while maintaining the same high quality.

Evaluation of the potential of phase-contrast computed tomography for improved visualization of cancerous human liver tissue

PurposePhase-contrast X-ray computed tomography (PCCT) is currently investigated and developed as a potentially very interesting extension of conventional CT, and can offer several advantages for specific indications in diagnostic imaging. Current absorption-based computed tomography (CT) without the application of contrast material is limited in the detection of minor density differences in soft-tissue. The purpose of this study is to test whether PCCT can improve soft tissue contrast in healthy and tumorous human liver specimens. Materials and MethodsTwo specimens of human liver (one healthy and one metastasized liver sample) were imaged with brilliant X-ray beam at the synchrotron radiation source ESRF in Grenoble, France. For correlation the same specimens were imaged with a magnetic resonance imaging system at 1.5 T. The histopathology confirmed our findings in the corresponding sections of the specimens. ResultsIn the phase-contrast CT images we observed a significantly enhanced soft-tissue contrast when compared to simultaneously recorded standard absorption CT measurements. Further, we found that the pathological and morphological information in the PCCT reconstructions show significant improvement when compared to those performed on MRI. Based on matching of prominent features, a good correlation between PCCT and the histological section is demonstrated; especially the tumor capsule and the surrounding vascular structures are visible in PCCT. In addition, our study revealed the ability of PCCT to visualize the blood vessels structure in the tumorous liver without the need of any contrast agents. ConclusionGrating-based PCCT significantly improves the soft-tissue contrast in ex-vivo liver specimens and holds the potential to overcome the need of contrast materials for visualization of the tumor vascularization.
