Degradation of petroleum hydrocarbon contaminants by Rhodococcus erythropolis KB1 synergistic with alfalfa (Medicago sativa L.).

Petroleum hydrocarbons are a stubborn pollutant that is difficult to degrade globally, and plant-microbial degradation is the main way to solve this type of pollutant. In this study, the physiological and ecological responses of alfalfa to petroleum hydrocarbons in different concentrations of petroleum hydrocarbon-contaminated soil with KB1 (Rhodococcus erythropolis) were analyzed and determined by laboratory potting techniques. The growth of alfalfa (CK) and alfalfa with KB1 (JZ) in different concentrations of petroleum hydrocarbons contaminated soil was compared and analyzed. The results of the CK group showed that petroleum hydrocarbons could significantly affect the activity of alfalfa antioxidant enzyme system, inhibit the development of alfalfa roots and the normal growth of plants, especially in the high-concentration group. KB1 strain had the ability to produce IAA, form biofilm, fix nitrogen, produce betaine and ACC deaminase, and the addition of KB1 could improve the growth traits of alfalfa in the soil contaminated with different concentrations of petroleum hydrocarbons, the content of soluble sugars in roots, and the stress resistance and antioxidant enzyme activities of alfalfa. In addition, the degradation kinetics of the strain showed that the degradation rate of petroleum could reach 75.2% after soaking with KB1. Furthermore, KB1 can efficiently degrade petroleum hydrocarbons in advance and significantly alleviate the damage of high concentration of petroleum hydrocarbons to plant roots. The results showed that KB1 strains and alfalfa plants could effectively enhance the degradation of petroleum hydrocarbons, which provided new ideas for improving bioremediation strategies.

Remediation of petroleum hydrocarbon contaminated soils by nZVI coupled with electrokinetic activation of persulfate

This paper explores the use of nanoscale zero-valent iron (nZVI) coupled with electrically activated persulfate (nZVI-EK/PS) for remediating petroleum hydrocarbon-contaminated soil. The effects of persulfate (PS) dosage, nZVI dosage, and soil organic matter content on the remediation of petroleum hydrocarbons were investigated. The study concludes that persulfate (PS) and nanoscale zero-valent iron (nZVI) are the primary factors influencing the remediation of petroleum hydrocarbons in the nZVI-EK/PS system. The optimal reaction conditions of PS = 170 mmol and nZVI = 17 mmol increased the efficiency of petroleum hydrocarbon treatment to 40.45%, with the highest removal efficiency of petroleum hydrocarbons observed at position S2, reaching 48.5%. With the addition of organic matter at 5%, the efficiency of petroleum hydrocarbon treatment reached 50.08%, representing a 9.68% enhancement in the nZVI-EK/PS system. Furthermore, potential mechanisms for the degradation of petroleum hydrocarbons by the nZVI-EK/PS system were identified using gas chromatography-mass spectrometry (GC-MS). The degradation order of different components was found to be esters < alkenes < alkanes < phenols.

Investigating the influence of sulphur amendment and temperature on microbial activity in bioremediation of diesel-contaminated soil

This study investigated the effectiveness of incorporating sulphur (S) with nitrogen (N) and phosphorus (P) for enhancing microbial activity in diesel-contaminated soil during ex-situ bioremediation. While N and P amendments are commonly used to stimulate indigenous microorganisms, the potential benefits of adding S have received less attention. The study found that historically contaminated soil with a moderate concentration of total petroleum hydrocarbons (TPH; 1270 mg/kg) did not have nutrient limitation, and incubation temperature was found to be more critical for enhancing microbial activity. However, soil spiked with an additional 5000 mg/kg of diesel showed increased activity following NP and NPS amendment. Interestingly, NPS amendment at 10 °C resulted in higher microbial activity than at 20 °C, indicating the potential for a tailored nutrient amendment approach to optimize bioremediation in cold conditions. Overall, this study suggests that incorporating S with N and P can enhance microbial activity in diesel-contaminated soil during ex-situ bioremediation. Furthermore, the study highlights the importance of considering incubation temperature in designing a nutrient amendment approach for bioremediation, especially in cold conditions. These findings can guide the design and implementation of future effective bioremediation strategies for petroleum hydrocarbon-contaminated soil.

Evaluation of lipopeptides biosurfactant from Raoultella planticola for bioremediation in n-hexadecane-contaminated soil

Biosurfactants combined with bioaugmentation for the remediation of petroleum hydrocarbon-contaminated soils proved to be one of the most effective ways. However, research on lipopeptides combined with bioaugmentation is rare, and the biological mechanisms are undefined. Therefore, the effect of Raoultella planticola and the lipopeptides it produces to restore contaminated soils was investigated using n-hexadecane as a model of petroleum hydrocarbons. After 60 d, n-hexadecane removal rate increased from 31.3% in contaminated soil to 39.9% with R. planticola alone and 59.0% with R. planticola together with 1.0 g intracellular lipopeptide (INLS) and 7.5 g sodium gluconate (SG)/kg dry soil. Meanwhile, the nitrogen and phosphorus components of the soil increased by 13.9% and 8% in the experimental group, respectively. Besides, DHA activity was 6.7 times that of the control group at 28 d, indicating a considerable increase in DHA activity. Microbial community analysis revealed a considerable increase in functional genera of Sphingomonas, Nocardia, Pseudoalteromonas, Povalibacter, Apiotrichum, Pseudallecheria, and Ascodesmis. The amount of fungus in the experimental group was more than 30 times that of the control group after 60 d. Microbial co-occurrence networks showed improved interactions of Firmicutes and Ascomycota, encouraging synergistic cooperation among microbes and thereby accelerating n-hexadecane breakdown. At once, the metabolic pathways ko00071 and ko00980 for n-hexadecane degradation were greatly elevated, indicating that functional genes such as alkane 1-monooxygenase and NADPH-cytochrome P450 reductase were activated. These results implied that combining hydrocarbon-degrading microbial with INLS was a promising bioremediation technique to remove soil petroleum hydrocarbons.

Effect of the Application of Ochrobactrum sp.-Immobilised Biochar on the Remediation of Diesel-Contaminated Soil.

The immobilisation of bacteria on biochar has shown potential for enhanced remediation of petroleum hydrocarbon-contaminated soil. However, there is a lack of knowledge regarding the effect of bacterial immobilisation on biosolids-derived biochar for the remediation of diesel-contaminated soil. This current study aimed to assess the impact of the immobilisation of an autochthonous hydrocarbonoclastic bacteria, Ochrobacterium sp. (BIB) on biosolids-derived biochar for the remediation of diesel-contaminated soil. Additionally, the effect of fertiliser application on the efficacy of the BIB treatment was investigated. Biochar (BC) application alone led to significantly higher hydrocarbon removal than the control treatment at all sampling times (4887-11,589 mg/kg higher). When Ochrobacterium sp. was immobilised on biochar (BIB), the hydrocarbon removal was greater than BC by 5533 mg/kg and 1607 mg/kg at weeks 10 and 22, respectively. However, when BIB was co-applied with fertiliser (BIBF), hydrocarbon removal was lower than BIB alone by 6987-11,767 mg/kg. Quantitative PCR (q-PCR) analysis revealed that the gene related to Ochrobacterium sp. was higher in BIB than in the BC treatment, which likely contributed to higher hydrocarbon removal in the BIB treatment. The results of the q-PCR analysis for the presence of alkB genes and FTIR analysis suggest that the degradation of alkane contributed to hydrocarbon removal. The findings of this study demonstrate that bacterial immobilisation on biosolids-derived biochar is a promising technique for the remediation of diesel-contaminated soil. Future studies should focus on optimising the immobilisation process for enhanced hydrocarbon removal.

Research Progress on in-situ Remediation of Typical Heavy Metals in Petroleum Hydrocarbon-contaminated Soil Enrichment by Plants

Petroleum hydrocarbon is one of the dangerous substances in the process of petroleum development, refining, processing, transportation, and production. In the related activities of the petroleum industry, the output is large, and improper treatment will cause pollution to the surrounding environment. It is an urgent problem to conduct harmless and resource treatment of petroleum hydrocarbon polluted soil. Plant enrichment, as an environmentally friendly and pollution-free technical means, has the advantages of low cost and small change to the soil environment and effectively solves the problems of excessive heavy metals in petroleum hydrocarbons through plant enrichment. In this paper, the development process of plant enrichment, remediation methods, and plant enrichment of typical heavy metals (Cd, Hg, Zn) in petroleum hydrocarbon-polluted soil were systematically introduced. Through investigation, the mechanism and influencing factors of plant enrichment of heavy metals in the presence of petroleum hydrocarbons were summarized and analyzed, and the possible development direction of plant enrichment technology in the future was prospected.

Performance evaluation of electrokinetic bioremediation for weathered petroleum hydrocarbon-contaminated soil

This study investigates the impact of Electrokinetic Remediation (EKR) time on total petroleum hydrocarbons (TPH) removal from soil, focusing on electroosmotic phenomena guided by Helmholtz-Smoluchowski theory. Soil samples were exposed to a constant 2 V/cm voltage gradient for 8, 16, and 24 hours, utilizing a 0.05 M Na2SO4 solution as an electrolyte to enhance soil conductivity. Biostimulation was combined with EKR, and TPH levels and microbial colonies (TPC) were monitored over a 4-week period. Maintaining optimal conditions (25-40°C, pH 6-8, 30-60% water content) was crucial for effective biodegradation. Results showed that a 24-hour EKR duration was most effective, achieving an 87.9% removal rate. The 16-hour duration closely followed at 85.7%, while the 8-hour duration exhibited lower effectiveness at 62.5%. TPC counts increased in the fourth week for the 16-hour and 24-hour durations but decreased for the 8-hour duration. Post-EKR, a gradual decline in TPH levels indicated electroosmotic flow’s (EOF) positive impact on TPH desorption and biodegradation. Electroosmosis influence was evident in varying TPH concentrations among segments after EKR, with higher levels near the cathode in the 16-hour and 24-hour durations. Lower TPH near the cathode in the 8-hour variation suggested electroosmosis-induced desorption, while reduced concentrations in regions A (near anode) and B (middle) resulted from biodegradation and microbial mobility due to electrophoresis. Confirmation of electroosmosis across all durations was supported by observed water content and EOF volume. The 24-hour duration exhibited the highest EOF proportion in the cathode chamber at 22.2%, followed by 16 hours at 16.1%, and 8 hours at 8.6%.

Developing a Robust Bioventing Model

Bioventing is a widely recognized technique for the remediation of petroleum hydrocarbon-contaminated soil. In this study, the objective was to identify an optimal mathematical model that balances accuracy and ease of implementation. A comprehensive review of various models developed for bioventing was conducted wherein the advantages and disadvantages of each model were evaluated and compared regarding the different numerical methods used to solve relevant bioventing equations. After investigating the various assumptions and methods from the literature, an improved foundational bioventing model was developed that characterizes gas flow in unsaturated zones where water and non-aqueous phase liquid (NAPL) are present and immobile, accounting for interphase mass transfer and biodegradation, incorporating soil properties through a rate constant correlation. The proposed model was solved using the finite volume method in OpenFOAM, an independent dimensional open-source coding toolbox. The preliminary simulation results of a simple case indicate good agreement with the exact analytical solution of the same equations. This improved bioventing model has the potential to enhance predictions of the remediation process and support the development of efficient remediation strategies for petroleum hydrocarbon-contaminated soil.

Response of the soil microbial community to petroleum hydrocarbon stress shows a threshold effect: research on aged realistic contaminated fields.

Microbes play key roles in maintaining soil ecological functions. Petroleum hydrocarbon contamination is expected to affect microbial ecological characteristics and the ecological services they provide. In this study, the multifunctionalities of contaminated and uncontaminated soils in an aged petroleum hydrocarbon-contaminated field and their correlation with soil microbial characteristics were analyzed to explore the effect of petroleum hydrocarbons on soil microbes. Soil physicochemical parameters were determined to calculate soil multifunctionalities. In addition, 16S high-throughput sequencing technology and bioinformation analysis were used to explore microbial characteristics. The results indicated that high concentrations of petroleum hydrocarbons (565-3,613 mg•kg-1, high contamination) reduced soil multifunctionality, while low concentrations of petroleum hydrocarbons (13-408 mg•kg-1, light contamination) might increase soil multifunctionality. In addition, light petroleum hydrocarbon contamination increased the richness and evenness of microbial community (p < 0.01), enhanced the microbial interactions and widened the niche breadth of keystone genus, while high petroleum hydrocarbon contamination reduced the richness of the microbial community (p < 0.05), simplified the microbial co-occurrence network, and increased the niche overlap of keystone genus. Our study demonstrates that light petroleum hydrocarbon contamination has a certain improvement effect on soil multifunctionalities and microbial characteristics. While high contamination shows an inhibitory effect on soil multifunctionalities and microbial characteristics, which has significance for the protection and management of petroleum hydrocarbon-contaminated soil.

Effect of the bacterial community assembly process on the microbial remediation of petroleum hydrocarbon-contaminated soil.

The accumulation of petroleum hydrocarbons (PHs) in the soil can reduce soil porosity, hinder plant growth, and have a serious negative impact on soil ecology. Previously, we developed PH-degrading bacteria and discovered that the interaction between microorganisms may be more important in the degradation of PHs than the ability of exogenous-degrading bacteria. Nevertheless, the role of microbial ecological processes in the remediation process is frequently overlooked. This study established six different surfactant-enhanced microbial remediation treatments on PH-contaminated soil using a pot experiment. After 30 days, the PHs removal rate was calculated; the bacterial community assembly process was also determined using the R language program, and the assembly process and the PHs removal rate were correlated. The rhamnolipid-enhanced Bacillus methylotrophicus remediation achieved the highest PHs removal rate, and the bacterial community assembly process was impacted by deterministic factors, whereas the bacterial community assembly process in other treatments with low removal rates was affected by stochastic factors. When compared to the stochastic assembly process and the PHs removal rate, the deterministic assembly process and the PHs removal rate were found to have a significant positive correlation, indicating that the deterministic assembly process of bacterial communities may mediate the efficient removal of PHs. Therefore, this study recommends that when using microorganisms to remediate contaminated soil, care should be taken to avoid strong soil disturbance because directional regulation of bacterial ecological functions can also contribute to efficient removal of pollutants.

Numerical Simulation of Heat Transfer Characteristics of Petroleum Hydrocarbon-Contaminated Soil During Ex-Situ Indirect Thermal Desorption

ABSTRACT Indirect thermal desorption process of petroleum hydrocarbon contaminated soil is very complex and difficult to be analyzed by engineering full-scale numerical simulation considering multi-phase flow and interphase heat as well as mass transfer. Therefore, it is urgent to develop a simple and rapid calculation method to guide technology development and engineering applications. In this paper, the method of equivalent specific heat approximation was proposed to simplify the mass transfer process on the material side to the heat transfer process under variable specific heat conditions, and the numerical simulation of engineering scale indirect thermal desorption equipment was studied by coupling the diffusion flame combustion model, weighted-sum-of-gray-gases model, and discrete ordinates model. The heating process of soil in thermal desorption chamber was simulated, and the influence of soil residence time and designed feed rate on process parameters was obtained. The research results showed that soil residence time should be extended appropriately under the condition of less than 1.75 hours and designed feed rate be limited within 4 t/h to ensure reasonable soil discharge temperature that was directly related to the removal efficiency of petroleum hydrocarbons in contaminated soil. Meanwhile, the modification scheme of power distribution of combustion system was further studied to improve the heating uniformity of tubes.

Remediation of petroleum contaminated soil by persulfate oxidation coupled with microbial degradation

This study was conducted on soil polluted with high concentrations of crude oil (12,835 ± 572.76 mg/kg) with different dosages of persulfate (PS) coupled with hydrocarbon-degrading mixed bacteria (including Enterobacteriaceae, Stenotrophomonas, Pseudomonas, Acinetobacter, and Achromobacter) obtained from long-term oil-contaminated soil. The interaction among total petroleum hydrocarbons (TPH), soil parameters, and microbial communities during a 187-d combined remediation process was evaluated. The result showed that the TPH removal of 1% PS oxidation combined with bioremediation was 80.05%; this was 4.88% and 20.94% higher than that of bioremediation and 1% PS oxidation combined with natural attenuation. This is a result of 1%PS stimulated enzyme secretion and dissolved organic carbon content, especially fulvic acid, improving TPH mobility and bioavailability in soil. Furthermore, introducing mixed bacteria after oxidation further increased enzyme activities and TPH-degrading ability, thus promoting carbon substrate conversion and biodegradation rate. Conversely, the excessive oxidation of 3% and 5% PS had a negative impact on the soil environment and microbial diversity resulting in lower TPH degradation (36%-57%). High-throughput sequencing revealed that the abundance and diversity of soil bacteria decreased in all remediation groups compared to those in the pristine soil. Redundancy analysis showed that the main factors affecting 1% PS oxidation combined with bioremediation during subsequent microbial remediation were pH, cation exchange capacity, and lipase activity, which may indirectly influence TPH biodegradation by increasing the abundance of genus Immundisolibacteraceae, Comamonadaceae, and Acinetobacter. This study provides data on in situ chemical oxidation integrated with the bioremediation of petroleum hydrocarbon-contaminated soils.

Helichrysum microphyllum subsp. tyrrhenicum, Its Root-Associated Microorganisms, and Wood Chips Represent an Integrated Green Technology for the Remediation of Petroleum Hydrocarbon-Contaminated Soils

Phytoremediation and the use of suitable amendments are well-known technologies for the mitigation of petroleum hydrocarbon (PHC) contaminations in terrestrial ecosystems. Our study is aimed at combining these two approaches to maximize their favorable effects. To this purpose, Helichrysum microphyllum subsp. tyrrhenicum, a Mediterranean shrub growing on sandy and semiarid soils, was selected. The weathered PHC-polluted matrix (3.3 ± 0.8 g kg−1 dry weight) from a disused industrial site was employed as the cultivation substrate with (WCAM) or without (UNAM) the addition and mixing of wood chips. Under the greenhouse conditions, the species showed a survival rate higher than 90% in the UNAM while the amendment administration restored the totality of the plant survival. At the end of the greenhouse test (nine months), the treatment with the wood chips significantly increased the moisture, dehydrogenase activity and abundance of the microbial populations of the PHC degraders in the substrate. Cogently, the residual amount of PHCs was significantly lower in the UNAM (3–92% of the initial quantity) than in the WCAM (3–14% of the initial quantity). Moreover, the crown diameter was significantly higher in the WCAM plants. Overall, the results establish the combined technology as a novel approach for landscaping and the bioremediation of sites chronically injured by PHC-weathered contaminations.

Microbial Removal of Petroleum Hydrocarbons from Contaminated Soil under Arsenic Stress.

The contamination of soils with petroleum and its derivatives is a longstanding, widespread, and worsening environmental issue. However, efforts to remediate petroleum hydrocarbon-polluted soils often neglect or overlook the interference of heavy metals that often co-contaminate these soils and occur in petroleum itself. Here, we identified Acinetobacter baumannii strain JYZ-03 according to its Gram staining, oxidase reaction, biochemical tests, and FAME and 16S rDNA gene sequence analyses and determined that it has the ability to degrade petroleum hydrocarbons. It was isolated from soil contaminated by both heavy metals and petroleum hydrocarbons. Strain JYZ-03 utilized diesel oil, long-chain n-alkanes, branched alkanes, and polycyclic aromatic hydrocarbons (PAHs) as its sole carbon sources. It degraded 93.29% of the diesel oil burden in 7 days. It also had a high tolerance to heavy metal stress caused by arsenic (As). Its petroleum hydrocarbon degradation efficiency remained constant over the 0-300 mg/L As(V) range. Its optimal growth conditions were pH 7.0 and 25-30 °C, respectively, and its growth was not inhibited even by 3.0% (w/v) NaCl. Strain JYZ-03 effectively bioremediates petroleum hydrocarbon-contaminated soil in the presence of As stress. Therefore, strain JYZ-03 may be of high value in petroleum- and heavy-metal-contaminated site bioremediation.

Effect of fresh and aged biochar on electrogenic hydrocarbon degradation in soil microbial electrochemical remediation

The remediation of petroleum hydrocarbon-contaminated soil is a global problem. A bioelectrochemical system (BES) is an environmentally friendly technique. In BESs, microbes on the anode play central roles in electricity generation and pollutant degradation. Here, we utilized fresh and aged biochar (marked by FB and AB) as amendments to assess the spatial-temporal hydrocarbon removal, bacterial community shift and functional gene changes in the anode and cathode vicinity. After 78 days, an accumulated charge of 750 C for FB was gained, which was 12% and 98% higher than that of AB and the no-biochar control (CK), respectively. Moreover, the removal of total petroleum hydrocarbons was enhanced in the order of CK (17–26%) < AB (22–28%) < FB (24–29%) in the cathodic area, while in the anodic region, the removal was enhanced in the order CK (22–31%) < FB (24–30%) < AB (28–30%) on days 43–78. Additionally, the top six phyla were Proteobacteria, Desulfobacterota, Firmicutes, Chloroflexi, Actinobacteriota and Bacteroidota, accounting for > 92% of the total bacterial community, with a declining tendency over time. The cathode vicinity favoured Proteobacteria growth, whereas the anode region favoured Desulfobacterota and Firmicutes. Functional genes emphasized petroleum hydrocarbons as the carbon source and relationships with nitrogen, sulfur and iron compounds in soil. This study validated the aptitudes and application effects of fresh and aged biochar in BESs.

Green Remediation Technology for Total Petroleum Hydrocarbon-Contaminated Soil

In order to improve the bioremediation efficiency of petroleum-contaminated soil, five test groups were selected in this study, including native bacteria, Acinetobacter venetianus, Vetiveria zizanioides L., and Vetiveria zizanioides L. combined with Acinetobacter venetianus and biochar to compare the degradation efficiency of petroleum-contaminated soil. The results of the study showed that after a six-month bioremediation, there was no significant difference between the native bacteria and the A.V. bacteria group in the removal efficiency of TPH, and the proportion of degradable TPH accounts for about 50~70%. The removal efficiency of TPH could be increased by 18.1–29% by increasing the phytoremediation of Vetiveria zizanioides L. The cultivation of Vetiveria zizanioides L. could not only stabilize the soil’s pH and conductivity but could also increase the soil’s bacterial abundance. It was suggested that bioremediation could be carried out through the combination of native bacteria and the planting of Vetiveria zizanioides L. Although the addition of biochar to the soil was able to improve the remediation effect of Vetiveria zizanioides L., it would increase the soil conductivity and reduce the abundance of soil bacteria. Therefore, it was recommended to reduce the conductivity of biochar before adding it, which may improve processing efficiency.

Coupling biostimulation and phytoremediation for the restoration of petroleum hydrocarbon-contaminated soil

Total petroleum hydrocarbons (TPH) continue to be among the most common pollutants in soil worldwide. Bioremediation and phytoremediation have become sustainable ways of dealing with TPH contamination and biostimulation-assisted phytoremediation is considered as a potential approach for the treatment of pollutants. In this study, the response surface was used to optimize the single-factor biological stimulation experiment of moisture content, leavening agent content and compound fertilizer content and got the best experimental plan of biological stimulation. It was found that TPH degradation rate was 28.6% by biostimulation after 70 days. Further, from 20 kinds of plant seeds, 5 kinds of suitable or growth and high germination rate were selected for petroleum hydrocarbon degradation experiment. In the phytoremediation, peanut was selected as the best plant species by measuring the TPH degradation rate, bacteria count, growth of test plants, germination rate and amount of catalase in the soil and it could achieved 31.1% degradation rate of petroleum hydrocarbons after 70 days. Finally, the artificial biostimulation and phytoremediation combined degradation experiment of petroleum hydrocarbons-contaminated soil was designed and it achieved 38.9% TPH degradation rate after 70 days.
