Prenatal and childhood chlordecone exposure, cognitive abilities and problem behaviors in 7-year-old children: the TIMOUN mother–child cohort in Guadeloupe

BackgroundChlordecone is a highly persistent organochlorine insecticide that was intensively used in banana fields in the French West Indies, resulting in a widespread contamination. Neurotoxicity of acute exposures in adults is well recognized, and empirical data suggests that prenatal exposure affects visual and fine motor developments during infancy and childhood, with greater susceptibility in boys.ObjectiveTo assess the associations between pre- and postnatal exposures to chlordecone and cognitive and behavioral functions in school-aged children from Guadeloupe.MethodsWe examined 576 children from the TIMOUN mother–child cohort in Guadeloupe at 7 years of age. Concentrations of chlordecone and other environmental contaminants were measured in cord- and children’s blood at age 7 years. Cognitive abilities of children were assessed with the Wechsler Intelligence Scale for Children-IV (WISC-IV), and externalizing and internalizing problem behaviors documented with the Strengths and Difficulties Questionnaire (SDQ) completed by the child’s mother. We estimated covariate-adjusted associations between cord- and 7-years chlordecone concentrations and child outcomes using structural equations modeling, and tested effect modification by sex.ResultsGeometric means of blood chlordecone concentrations were 0.13 µg/L in cord blood and 0.06 µg/L in children’s blood at age 7 years. A twofold increase in cord blood concentrations was associated with 0.05 standard deviation (SD) (95% Confidence Interval [CI]: 0.0, 0.10) higher internalizing problem scores, whereas 7-years chlordecone concentrations were associated with lower Full-Scale IQ scores (FSIQ) and greater externalized behavioral problem scores. A twofold increase in 7-year chlordecone concentrations was associated with a decrease of 0.67 point (95% CI: -1.13, -0.22) on FSIQ and an increase of 0.04 SD (95% CI: 0.0, 0.07) on externalizing problems. These associations with Cognitive abilities were driven by decreases in perceptive reasoning, working memory and verbal comprehension. Associations between 7-year exposure and perceptive reasoning, working memory, and the FSIQ were stronger in boys, whereas cord blood and child blood associations with internalizing problems were stronger in girls.ConclusionsThese results suggests that cognitive abilities and externalizing behavior problems at school age are impaired by childhood, but not in utero, exposure to chlordecone, and that prenatal exposure is related to greater internalizing behavioral problems.

Soil ingestion, a key determinant of exposure to environmental contaminants. The case study of chlordecone exposure in free-range pigs in the French West Indies.

Ingested soil may expose free-range animals to environmental pollutants. In pigs, soil ingestion is few described whereas their burrowing behaviour suggests that it could be high. Although highly productive pigs are generally reared indoor, free-range farming is increasing in view of ethical considerations for animal welfare and is a common practice for subsistence agriculture systems. The experiment lasted 8 weeks (2 for adaptation, 6 for measurements) with 24 growing pigs of Guadeloupean Creole (CR) or Large White (LW) breeds. Pigs were assigned to 3 outdoor treatments: high pasture HP (>60 days of regrowth), low pasture LP (35 days of regrowth), and sweet potato SP (sweet potato field). Titanium (soil marker) and chromium (faecal output marker) contents of faeces, vegetation and soil samples were used to estimate individual daily soil ingestions. The average, 10th and 90th percentiles were 440, 200 and 726 g of dry soil per 100 kg body weight, respectively, without significant differences between the 3 outdoor treatments or the 2 breeds but with a significant period (i.e. week of measurements) × treatment interaction (P < 0.001). In the French West Indies, animals may be exposed to chlordecone (CLD), a very persistent organochlorine insecticide. Simulations of CLD tissue contamination due to ingestion of contaminated soil were carried out and compared to the maximum residue limit. These results show that grazing management needs to be adapted to effectively limit soil ingestion by pigs and the impact of a contaminated environment on the sustainability of pig systems.

Endosulfan causes oxidative stress in the liver and brain that involves inhibition of NADH dehydrogenase and altered antioxidant enzyme status in rat

Endosulfan, a neurotoxic, highly persistent organochlorine insecticide, is known for its acute and chronic toxicity. We have shown that a single sublethal dose of endosulfan caused high induction of oxidative stress in the liver and brain by altering the antioxidant status, as shown by reduction in the antioxidant enzymes SOD, GPx, GST, GR along with increased ROS and lipid peroxidation. The cerebral region in the brain showed a higher level of oxidative stress than the cerebellum, revealing differential sensitivity of the brain regions to endosulfan. Depletion of natural antioxidants causes the imbalance of redox status in cells, and the role of mitochondrial distress causally related to the cellular oxidative stress in vivo is not well understood. We have shown that reduction in the mitochondrial NADH dehydrogenase activity in the brain is associated with the induction of ROS in endosulfan-treated rats. Although oxidative stress is induced in both the liver and brain, the oxidative damage to the brain has implications for the toxic outcome in view of the brain's lower antioxidant defenses and high oxygen consumption.

Prenatal and childhood exposure to chlordecone and adiposity of seven-year-old children in the Timoun mother\u2013child cohort study in Guadeloupe (French West Indies)

BackgroundExposure to persistent environmental organic pollutants may contribute to the development of obesity among children. Chlordecone is a persistent organochlorine insecticide with estrogenic properties that was used in the French West Indies (1973–1993) and is still present in the soil and the water and food consumed by the local population. We studied the association between prenatal and childhood exposure to chlordecone and the adiposity of prepubertal children.MethodsWithin the Timoun Mother–Child Cohort Study in Guadeloupe (French West Indies), 575 children had a medical examination at seven years of age, including adiposity measurements. A Structural Equation Modeling approach was used to create a global adiposity score from four adiposity indicators: the BMI z-score, percentage of fat mass, sum of the tricipital and subscapular skinfold thickness, and waist-to-height ratio. Chlordecone concentrations were measured in cord blood at birth and in the children’s blood at seven years of age. Models were adjusted for prenatal and postnatal covariates. Sensitivity analyses accounted for co-exposure to PCB-153 and pp’-DDE. Mediation analyses, including intermediate birth outcomes, were conducted.ResultsPrenatal chlordecone exposure tended to be associated with increased adiposity at seven years of age, particularly in boys. However, statistical significance was only reached in the third quartile of exposure and neither linear nor non-linear trends could be formally identified. Consideration of preterm birth or birth weight in mediation analyses did not modify the results, as adjustment for PCB-153 and pp’-DDE co-exposures.ConclusionGlobally, we found little evidence of an association between chlordecone exposure during the critical in utero or childhood periods of development and altered body-weight homeostasis in childhood. Nevertheless, some associations we observed at seven years of age, although non-significant, were consistent with those observed at earlier ages and would be worth investing during further follow-ups of children of the Timoun Mother–Child Cohort Study when they reach puberty.

Removal of multiple pesticide residues from water by low-pressure thin-film composite membrane

The study evaluated removal efficiency of 43 pesticides from water by thin-film composite polyamide membrane indigenously prepared by interfacial polymerization of 1,3-phenylenediamine and 1,3,5 trimesoyl chloride coated on asymmetric polysulfone support. Membrane performance was evaluated by gas and liquid chromatography mass spectroscopy determination of multiple pesticides remaining in feed and permeated water following the application of pesticides each @ 0.02, 0.05, and 0.10 mg/L in de-ionized water. The membrane was most efficient in the rejection of persistent organochlorine insecticides, viz. endosulfans (100%), dichlorodiphenyltrichloroethane (95%), and hexachlorocyclohexane (92%). Out of 43 selected pesticides, 33 were removed by > 80%. Size exclusion mass transfer played a significant role for molecules to pass through the membrane as observed for endosulfan isomers, endosulfan sulphate, and difenoconazole with molecular weight > 400. Pesticide rejection was also related to hydrophobicity (Log P). Hydrophobic pesticides with log P > 4.5 were rejected by > 80%, while monocrotophos with less hydrophobicity (log P = − 0.22) exhibited poor rejection (38%). Water flux decreased with an increase in pesticide concentration. The process of pesticide filtration was optimized at 200 psi. The results indicated the potential of the membrane to remove pesticides from water.

Dietary antioxidants impact DDT resistance in Drosophila melanogaster

Insects experience a diversity of subtoxic and/or toxic xenobiotics through exposure to pesticides and, in the case of herbivorous insects, through plant defensive compounds in their diets. Many insects are also concurrently exposed to antioxidants in their diets. The impact of dietary antioxidants on the toxicity of xenobiotics in insects is not well understood, in part due to the challenge of developing appropriate systems in which doses and exposure times (of both the antioxidants and the xenobiotics) can be controlled and outcomes can be easily measured. However, in Drosophila melanogaster, a well-established insect model system, both dietary factors and pesticide exposure can be easily controlled. Additionally, the mode of action and xenobiotic metabolism of dichlorodiphenyltrichloroethane (DDT), a highly persistent neurotoxic organochlorine insecticide that is detected widely in the environment, have been well studied in DDT-susceptible and -resistant strains. Using a glass-vial bioassay system with blue diet as the food source, seven compounds with known antioxidant effects (ascorbic acid, β-carotene, glutathione, α-lipoic acid, melatonin, minocycline, and serotonin) were orally tested for their impact on DDT toxicity across three strains of D. melanogaster: one highly susceptible to DDT (Canton-S), one mildly susceptible (91-C), and one highly resistant (91-R). Three of the antioxidants (serotonin, ascorbic acid, and β-carotene) significantly impacted the toxicity of DDT in one or more strains. Fly strain and gender, antioxidant type, and antioxidant dose all affected the relative toxicity of DDT. Our work demonstrates that dietary antioxidants can potentially alter the toxicity of a xenobiotic in an insect population.

Lithospherically sorbed cyclodiene insecticide sustainable remediation via green adsorbent derived from Arachis hypogaea shells

ABSTRACT The current investigation has attempted to investigate the adsorption pattern of a persistent organochlorine insecticide, Endosulfan via the standard batch equilibrium method. Results for the model application are expressed adsorption co-efficient Kd ranging from 0.7 to 10 µg/mL. Existence of physisorption was confirmed through an exothermic and low interaction between Endosulfan and soil samples are shown via Gibbs free energy (ΔG) with negative values. Statistical significance of the results was shown by negative association between soil pH and Kd (R 2 = −0.74 and p = 0.01), while organic matter and adsorption were related to one another in a positive manner (R 2 = 0.92). Additionally, statistical significance of the results was also expressed by ANOVA analysis results, indicating a p value of <0.05. In the sub-sequential manner, activated peanut shells powder, as a green adsorbent, has been used for Endosulfan pesticide removal from the selected soil samples. Activated peanut shells powder was exposed to the FT-IR analysis showing presence and augmentation of different groups. Endosulfan maximum removal was observed in 5 ppm concentration to be 78%, while in 7.5 ppm, it was 70%. Currently devised method can be efficiently used in a cost-effective manner for the removal of obnoxious pesticides from environmental compartments.

Analysis of Carbamates Pesticides: Immunogical Technique by Local Development of Enzyme-linked Immuno-Sorbent Assay

Carbamates insecticides are used increasingly in agriculture as a replacement for environmentally more persistent organochlorine insecticides for the control of insect pests. It was the most used pesticides in Loukkous-Morocco. Conventional methods employed to detect/analyze carbamates residues are time consuming and require sophisticated equipment only available in well-equipped laboratories. In addition, the conventional methods usually require a lot of complex pre-treatment of samples. Therefore, convenient and rapid pesticide detection system is urgently needed. In this order, we aimed to develop a rapid ELISA (enzyme-linked immuno-Sorbent Assay) for detection of some carbamates such as carbendazim [methyl 2-benzimidazole carbamate] and carbofuran [2,3-dihydro-2,2-dimethyl-7-benzofuranylmethyl carbamate]. To develop an immunoassay for carbendazim and carbofuran we have synthesized molecules with acid function (haptens with 4 and 5 carbons) that are coupled with BSA protein and injected to the rabbits, collected antibodies are used for the achievements of the immunoanalytical assay.

Prostate cancer clinical presentation, incidence, mortality and survival in Guadeloupe over the period 2008-2013 from a population-based cancer registry.

The Caribbean population of Guadeloupe has one of the highest incidence rates of prostate cancer worldwide. In 2008, a population-based cancer registry was set up for the monitoring of cancer incidence in the aftermath of the environmental pollution with chlordecone, a persistent organochlorine insecticide formerly used in banana plantations. We describe the clinical presentation, incidence, mortality and survival of prostate cancer for the period 2008-2013. The Guadeloupe cancer registry has been routinely collecting all incident cases of cancer since 2008. We compared age-specific incidence rates between different populations, and calculated incidence and mortality rates standardized to the world population. Kaplan-Meier observed survival and estimated age-standardized net survival were calculated by category for age, PSA level, and Gleason score using the Pohar-Perme method. Overall, 3,295 cases of prostate cancer were recorded. World-standardized incidence and mortality were respectively 184.1 [177.8-190.4] and 23.9 [21.9-25.7] per 100,000 person-years. At diagnosis, the mean age of patients was 68 ± 9.6years old and 22% were aged over 75. Median PSA level was 8.9 [IQR: 6.0-16.0] and 13.6% of the patients had a Gleason ≥ 8. Five-year observed and net survivals were, respectively, 79.6% [77.9-81.2] and 90.7% [88.6-92.8]. The incidence of prostate cancer in Guadeloupe is among the highest in the world, along with those of the neighboring Caribbean countries and US African-Americans. We observed no decrease in incidence rates, and a decreasing but non-significant trend in mortality rates, which nonetheless remain higher than in high-income countries. Many Genome-Wide Association Studies are conducted to identify genetic markers involved in prostate cancer risk. In the Caribbean, complementary studies on both lifestyle and behavioral factors should highlight potential common risks among populations who share both genetic and environmental characteristics.

Chronic endosulfan exposure impairs immune response rendering Clarias gariepinus susceptible to microbial infection

Endosulfan, a persistent organochlorine insecticide affects several off-target organisms including fish though the underlying mechanisms remain obscure. In the present study, we monitored the effect of chronic endosulfan exposure on headkidney (HK), an important immune organ in fish and on fish immune system thereof. Clarias gariepinus were exposed to a non-lethal concentration of endosulfan 2.884ppb (1/10th LC50) for 30 d which resulted in suppressed phagocytosis and bactericidal potential of headkidney macrophages (HKM). The same non-lethal concentration of endosulfan also interfered with T-cell proliferation and serum antibody titer in fish. Endosulfan-exposed fish were challenged with non-lethal dose of fish pathogenic bacteria Aeromonas hydrophila and the ‘exposure-challenge’ study revealed endosulfan-exposed C. gariepinus severely immunocompromised and prone to bacterial infections. Depuration for 30 d suggested that except for phagocytosis and serum agglutination titer other endosulfan-induced immune aberrations could not be restored significantly. Nonetheless, compared to exposed-challenged fish the depurated fish showed significant improvement in viability on challenge with A. hydrophila. Collectively, these findings suggest chronic endosulfan exposure has prolonged effect on fish making them prone to microbial infections.

Prenatal exposure to chlordecone, gestational weight gain, and birth weight in a Guadeloupean birth cohort

BackgroundChlordecone is a persistent organochlorine insecticide with well-defined estrogenic properties. It was intensively used in the French West Indies until 1993 to control the banana root borer. Because of the long-term contamination of soils and water, the population is currently exposed to chlordecone through food consumption. Chlordecone has been found in the blood of pregnant women and in cord blood. It has been shown to be an endocrine-disrupting chemical and exposure during pregnancy may affect fetal growth. ObjectivesThe objective of our study was to examine the association between prenatal exposure to chlordecone and fetal growth based on the TIMOUN birth cohort conducted in Guadeloupe, with a focus on the potential modification of this relationship by maternal body mass index (BMI) and gestational weight gain (GWG). MethodsChlordecone was determined in cord plasma at birth in 593 babies. Birth weight was the indicator of fetal growth. Maternal pre-pregnancy BMI and GWG were determined. Adherence to GWG recommendations of the US Institute of Medicine based on maternal pre-pregnancy BMI was assessed. Birth weight was analyzed relative to cord blood chlordecone levels using linear and non-linear regression models. ResultsOverall chlordecone in cord blood was not associated with birth weight, but we found an interaction between chlordecone exposure with GWG and adherence to GWG recommendations. After stratification by GWG, we found a significant U-shaped association between birth weight and chlordecone exposure, within the upper quartiles of GWG or excessive GWG. ConclusionChlordecone exposure may affect fetal growth, particularly when excessive GWG is present.

Hexachlorocyclohexane: persistence, toxicity and decontamination

Hexachlorocyclohexane (HCH), a persistent organochlorine insecticide, has been extensively used in the past for control of agricultural pests and vector borne diseases. The use of HCH has indeed accrued benefits, however the unusual production of the insecticidal isomer; γ-HCH (lindane) and unregulated disposal of HCH muck has created various dumpsites all over the world, leading to serious environmental concerns. HCH isomers have been ranked as possible human carcinogens and endocrine disruptors with proven teratogenic, mutagenic and genotoxic effects, hence making its decontamination mandatory. Efforts in this direction have led to the isolation of various HCH degrading bacteria from the dumpsites, reflecting their role in HCH bioremediation. This review summarizes the problem of environmental persistence of HCH isomers along with their toxicity and possible solutions for their decontamination.

Chlordecone fate and mineralisation in a tropical soil (andosol) microcosm under aerobic conditions

Chlordecone is a persistent organochlorine insecticide that, even decades after its ban, poses a threat to the environment and human health. Nevertheless, its environmental fate in soils has scarcely been investigated, and elementary data on its degradation and behaviour in soil are lacking. The mineralisation and sorption of chlordecone and the formation of possible metabolites were evaluated in a tropical agricultural andosol. Soil microcosms with two different soil horizons (S-A and S-B) were incubated for 215days with 14C-chlordecone. At five different times (1, 33, 88, 150 and 215days) the extractability of 14C-chlordecone was analysed. Mineralisation was monitored using 14CO2 traps of NaOH. The appearance of metabolites was studied using thin layer and gas chromatography techniques. At the end of the experiment, the water soluble 14C-activity was 2% of the remaining 14C-chlordecone for S-A and 8% for S-B. Only 12% of the remaining activity was non extractable and more than 80% remained extractable with organic solvents. For the first time to our knowledge, a significant mineralisation of chlordecone was measured in a microcosm under aerobic conditions (4.9% for S-A and 3.2% for S-B of the initial 14C-activity). The drastically lower emission of 14CO2 in sterilised microcosms indicated the biological origin of chlordecone mineralisation in the non-sterilised microcosms. No metabolites could be detected in the soil extracts. The mineralisation rate of chlordecone decreased by one order of magnitude throughout the incubation period. Thus, the chlordecone content in the soil remained large. This study confirms the existence of chlordecone degrading organisms in a tropical andosol. The reasons why their activity is restricted should be elucidated to allow the development of bioremediation approaches. Possible reasons are a heterogeneous distribution a chlordecone between sub-compartments with different microbial activities or a degradation of chlordecone by co-metabolic processes controlled by a limited supply of nutrients.

Chlordecone retention in the fractal structure of volcanic clay

Chlordecone (CHLD), a soil and foodstuff pollutant, as well as an environmentally persistent organochlorine insecticide, was used intensively in banana fields. The chlordecone uptake of three crops was measured for two types of polluted soils: allophanic and non-allophanic. The uptake is lower for allophanic soils even if their chlordecone content is higher than with non-allophanic soils. The fractal structure of the allophane aggregates was characterized at the nanoscale by small angle X-rays scattering, pore size distribution and transmission electron microscopy. We showed that clay microstructures should be an important physico-chemical factor governing the fate of chlordecone in the environment. Allophanic clays result in two counterintuitive findings: higher contaminant trappings yet lower contaminant availability. We propose that this specific, tortuous structure, along with its associated low accessibility, partly explains the low availability of chlordecone confined in allophanic soils.CapsuleThe fractal and tortuous microstructure of allophane clay favours the chlordecone retention in soils and disfavours the crop uptake.

Organochlorine Insecticides Lindane and Dieldrin and Their Binary Mixture Disturb Calcium Homeostasis in Dopaminergic PC12 Cells

Current hypotheses link long-term environmental exposure of humans to persistent organochlorine (OC) insecticides lindane (HCH) and dieldrin (HEOD) to the development of neurodegenerative disorders, such as Parkinson's disease. Primary adverse neurological effects of these insecticides are directed at inhibition of GABA(A) and glycine receptors, although GABA-independent effects have also been reported. In this paper we describe the effect of dieldrin and a binary mixture of dieldrin and lindane on a critical parameter of neuronal function and survival, i.e., intracellular calcium homeostasis. The intracellular calcium concentration ([Ca(2+)](i)) has been monitored using real-time single-cell fluorescence microscopy in dopaminergic PC12 cells loaded with the calcium-sensitive dye Fura-2. The results demonstrate that nanomolar concentrations of dieldrin time- and concentration-dependently inhibit depolarization-evoked influx of Ca(2+). Co-exposure of PC12 cells to a mixture of dieldrin and lindane revealed an additive inhibition of the depolarization-evoked increase in [Ca(2+)](i), whereas the lindane-induced increase in basal [Ca(2+)](i) is inhibited by dieldrin. The combined findings indicate that dieldrin and binary mixtures of organochlorines affect [Ca(2+)](i) already at concentrations below commonly accepted effect concentrations and close to human internal dose levels. Consequently, current findings illustrate the need to take mixtures of OC insecticides into account in human risk assessment.

Expression of a human cytochrome P4502E1 in Nicotiana tabacum enhances tolerance and remediation of γ-hexachlorocyclohexane

Lindane (γ-hexachlorocyclohexane), a persistent organo-chlorine insecticide widely used in developing countries, has a negative effect as a polluting agent of soil and surface waters. Plants can be used for remediation of organic pollutants and their efficiency can be enhanced by introduction of heterologous genes. Mammalian cytochrome P4502E1 (CYP2E1), an important monooxygenase is involved in the degradation of a wide range of xenobiotics including environmental pollutants/herbicides and pesticides. Here, we report the development of transgenic tobacco plants expressing human CYP2E1 and the efficacy of plants for remediation of lindane. Transgenic tobacco plants with CYP2E1 showed enhanced tolerance to lindane when grown in hydroponic medium and soil compared to control plants. Remediation of (14)C-labeled lindane from hydroponic medium was higher in transgenic plants compared to that of control plants, with the best performing line showing 25% higher removal of lindane from solution than control plants. Similar results were seen in plants grown in soil spiked with lindane. The present study has shown that transgenic plants expressing CYP2E1 gene have potential use for remediation of lindane from contaminated solutions and soil.

Pesticide exposure of pregnant women in Guadeloupe: Ability of a food frequency questionnaire to estimate blood concentration of chlordecone

Chlordecone, an environmentally persistent organochlorine insecticide used intensively in banana culture in the French West Indies until 1993, has permanently polluted soils and contaminated foodstuffs. Consumption of contaminated food is the main source of exposure nowadays. We sought to identify main contributors to blood chlordecone concentration (BCC) and to validate an exposure indicator based on food intakes. We used a food frequency questionnaire (FFQ) completed by a sample of 194 pregnant women to estimate their dietary exposure to chlordecone and compared it to blood levels. In a first approach, chlordecone daily intake was estimated as the product of daily eaten quantity of 214 foodstuffs, multiplied by their chlordecone content, and summed over all items. We then predicted individual blood chlordecone concentration with empirical weight regression models based on frequency of food consumption, and without contamination data. Among the 191 subjects who had BCC determination, 146 (76%) had detectable values and mean BCC was 0.86 ng/mL (range < LOD-13.2). Mean per capita dietary intake of chlordecone was estimated at 3.3 microg/day (range: 0.1-22.2). Blood chlordecone levels were significantly correlated with food exposure predicted from the empirical weight models (r=0.47, p<0.0001) and, to a lesser extent, with chlordecone intake estimated from food consumption and food contamination data (r=0.20, p=0.007). Main contributors to chlordecone exposure included seafood, root vegetables, and Cucurbitaceous. These results show that the Timoun FFQ provides valid estimates of chlordecone exposure. Estimates from empirical weight models correlated better with blood levels of chlordecone than did estimates from the dietary intake assessment.

Endosulfan increases seric interleukin-2 like (IL-2L) factor and immunoglobulin M (IgM) of Nile tilapia ( Oreochromis niloticus) challenged with Aeromona hydrophila

Endosulfan is a persistent organochlorine insecticide which is extremely toxic to fish. It is known to induce immunological alterations in juvenile Nile tilapia ( Oreochromis niloticus) such as increases in phagocytic activity and reactive oxygen species production of spleen macrophages. The purpose of the present study was to demonstrate the effects of acute exposure to a sublethal concentration of endosulfan (7 ppb, 96 h) on parameters of the adaptive humoral immune response of the aforementioned aquatic organism. The effect of endosulfan on the capacity of immune cells to produce interleukin-2 like (IL-2L) factor and immunoglobulin M (IgM) in response to a challenge with ½ LD50 of the infectious bacteria Aeromonas hydrophila was evaluated. Experimental results indicate that short, sublethal, endosulfan exposure triggers a succession of events beginning with non-specific activation of macrophages followed by an exacerbated synthesis of the IL-2L factor by activated B cells. This leads to significantly increased secretion of IgM and could in turn facilitate autoantibody production and the development of autoimmune pathologies.

Pesticide Exposure of Pregnant Women in Guadeloupe: Ability of a Food Frequency Questionnaire to Estimate Blood Concentration of Chlordecone

ISEE-0045 Background and Objective: Chlordecone, an environmentally persistent organochlorine insecticide used intensively in banana culture in the French West Indies until 1993, has permanently polluted soils and contaminated foodstuffs. Consumption of contaminated food is the main source of exposure nowadays. Exposure indicators are required to assess its potential health effects today. We sought to validate an exposure indicator based on food intake as an alternative to blood measurements. Methods: We used a food frequency questionnaire completed by a sample of 194 pregnant women to estimate their dietary exposure to chlordecone and compared it to blood levels. In a first approach, chlordecone daily intake was estimated as the product of frequency of consumption of 214 foodstuffs, multiplied by their chlordecone content, and summed over all items. We then predicted individual blood chlordecone levels with empirical weight linear regression models based on frequency of food consumption and without contamination data. Results: Mean blood chlordecone levels were 1.3 ng/mL (range: 0.5-13.2) among the 59% of subjects with detectable values. Mean per capita dietary intake of chlordecone was estimated at 3.3μg.day−1 (range: 0.1-22.2). Blood chlordecone levels were significantly correlated with food exposure predicted from the empirical weight models (r = 0.47, P <0.0001) and, to a lesser extent, with chlordecone intake estimated from food consumption and food contamination data (r = 0.16, P = 0.03). Conclusion: These results demonstrate that our food frequency questionnaire is able to estimate dietary chlordecone exposure. Estimates from empirical weight models correlated better with blood levels of chlordecone than did estimates from the dietary intake assessment.

Spatial distribution of lindane in topsoil of Northern France

Lindane is a persistent organochlorine insecticide and the use of this insecticide in agriculture was banned in France in 1998. In this study we investigated the concentrations of lindane in topsoil in Northern France and used robust geostatistics to map the geographical distribution of lindane. The study was based on a 16 km × 16 km grid covering an area of ca 25 000 km 2. Lindane was found in all soils, even those from non-agricultural-application areas. Very low ratios of α-/γ-HCH and δ-/γ-HCH suggested that a long time had passed since technical HCH was used in the studied area, or that emission sources of lindane were still present. A strong gradient in lindane concentration was observed, with the highest lindane concentrations in an area located in the northern region. Results suggested that some of the lindane observed in the high concentration area may have come from volatilization of old lindane applied to intensively cultivated areas, which was then transported by prevailing winds coming from the south-west and deposited in a densely inhabited depression.
