Peripheral blood lymphocyte subsets and their relation with early response to treatment in patients with low or intermediate risk myelodysplastic syndrome (IPSS score ≤ 1.0) and chronic aplastic anemia

The purpose of this study was retrospectively to analyze the peripheral blood lymphocyte subset distribution in patients with low or intermediate risk myelodysplastic syndromes (IPSS ≤ 1.0) and chronic aplastic anemia (CAA), and their hematological changes of peripheral blood after treatment, so as to understand differences and their relation with early treatment response. The lymphocyte subsets in peripheral blood of 67 patient with low or intermediate risk MDS (IPSS ≤ 1.0), 54 patients with CAA and 73 healthy individuals were analyzed by flow cytometry. The results showed that Th cells, Th/Ts ratio in peripheral blood of low or intermediate risk MDS were 42.94% ± 10.80% and 1.80% ± 0.99% respectively, and were significantly higher than those in control group; the CD16(+) CD56(+) cell ratio was 11.22% ± 7.97%, and was significantly lower than that in control group, the difference was statistically significant (P < 0.05); Ts cells and CD19(+) cell ratio in peripheral blood of CAA patients were 30.87% ± 9.11% and 16.98% ± 7.40% respectively, and were significantly higher than those in control group; CD16(+) CD56(+) cell ratio was 9.81% ± 7.00%, and was significantly lower than that in normal control group, and the difference was statistically significant (P < 0.05); while the Th cells and Th/Ts ratio in low or intermediate risk MDS group were significantly higher than those in CAA group, and the difference was statistically significant (P < 0.05). After treatment for 6 mouths, the HI-E and HI-N rates in CD19(+) cell normal group of low or intermediate risk MDS patients were 18.2% (4/22) and 13.6% (3/22), and were significantly lower than that in the increased group and decreased group. In Ts cell increased group HI-N rate was 15.4% (2/13), and was significantly lower than that in normal group and decreased group. In Th/Ts ratio decreased group HI-N rate was 14.3% (2/14), and was significantly lower than that in the increase group and normal group, the difference was statistically significant (P < 0.05). After treatment of CAA for 6 months, the effective rate for CD3(+) cells, Th cells, Th/Ts ratio in decreased group was 71.4% (5/7), 56.3% (9/16), 50.0% (10/20), and were significantly higher than those in increased and normal group, and the difference was statistically significant (P < 0.05). It is concluded that the peripheral blood lymphocyte subsets of low or intermediate risk MDS(IPSS score ≤ 1.0) and CAA are abnormal, and these lymphocyte subsets are related with hematologic changes after early response to treatment.

Comparative study of peripheral blood leukocytes in healthy dogs and in dogs with cancer and inflammatory diseases

In the present study, the peripheral blood lymphocyte subset distribution was compared between healthy dogs and dogs with chronic gastrointestinal disease, dental and skin conditions, and cancer. The immunophenotype of the group with chronic gastrointestinal disease and the group with dental and skin conditions showed no statistically significant differences with other groups of healthy or diseased dogs. When compared with healthy dogs, animals with cancer showed significantly lower absolute values of T cytotoxic cells (CD3+, CD8+) and lymphocytes that express major histocompatibility complex (MHC) class II (MHC-II+) in peripheral blood. The results suggest that peripheral blood immunophenotype is mainly altered in dogs with cancer but not in other diseases. Further studies are required to evaluate the clinical relevance of these findings.

Immunophenotypic evaluation of working Labrador Retrievers and German Shepherd dogs living in the same environment

Multiparametric flow cytometry was used to compare peripheral blood lymphocyte subset distribution between healthy working police Labrador Retrievers (LRs; n=12) and German Shepherd dogs (GS; n=11) living in the same environment. The CD4/CD8 ratio was significantly higher in LR than in GS because of the lower percentage of CD8+ T lymphocytes in LR. GS showed the highest relative percentage of CD3-/CD21- lymphocytes, whereas LR had the highest percentages of MHC II+ lymphocytes. Because age, sex, environmental and housing conditions, dietary patterns, and training or working routines were similar in both breeds in the study, differences in peripheral blood lymphocyte subset distribution could be attributed to the influence of breed on the immune system.

Peripheral blood lymphocyte subset distribution and function in patients with Alzheimer's disease and other dementias.

Until recently, new data on immune aspects of Alzheimer's disease (AD) have suggested that some facets of AD pathogenesis may be immune related. However, the effects of dementia itself on immune function have not been considered. To compare the distribution of peripheral blood lymphocyte subsets and their function in patients with AD and other dementias. Peripheral blood lymphocyte numbers, T cell subset distribution, proliferative responses to mitogens and suppressor cell assay were studied in a well characterised group of patients with AD, and compared to patients with other forms of dementia. Age and sex matched elderly controls were screened to exclude dementia, and young controls were medical, paramedical and laboratory staff. Analysis of variance (ANOVA) and student's test were used for statistical analysis. The CD8+ lymphocyte population was reduced in AD and in other forms of dementia, when compared with non-demented elderly and young controls. Concanavalin A induced lymphocyte transformation was reduced in all dementia groups and in elderly compared with young controls. The changes in T cell numbers and function were not specific for Alzheimer's disease, but were found also in other forms of dementia.

Study of peripheral blood lymphocyte subset distribution and IL-2 receptor (IL-2 R) p55–p75 subunit expression in patients with cancer of different sites

The aim of the study was to evaluate the subset distribution and the IL-2 R p55-p75 subunit expression on unstimulated and phytohemagglutinin (PHA)-stimulated (at 3-d) peripheral blood mononuclear cells (PBMC), of patients with solid cancers of different sites. Indeed the expression of the two subunits of IL-2 R is an essential prerequisite for the action of the IL-2 on CD8+, CD16+ lymphocytes as effectors in antitumor activity (LAK-cell). The subset distribution (CD3, CD4, CD8, CD16, DR) was assessed by cytofluorometry with specific monoclonal antibodies (MAbs); the p55 (CD25) and p75 subunit expression was evaluated by specific MAb (OKT26a and anti-p75). Ninety patients with advanced cancer (mainly non-small cell lung cancer [NSCLC], head and neck cancer, and gynecological cancer; mean age 55 yr; range 27-80) were studied. Thirty-five age- and sex-matched healthy subjects were studied as controls. Our data show that there is no significant difference in the subset distribution between cancer patients and controls. Furthermore, no difference has been found in the expression of p55 subunits on unstimulated PBMC between cancer patients and controls. No difference has been found in the expression of both p55 and p75 subunits on PHA-stimulated PBMC between cancer patients and controls. Our results can support the rationale for further clinical trials with IL-2 in solid malignancies.
