Morphological and functional spleen development in crossbreed rabbits

The accelerated growth of muscle mass in productive broiler breeds is often associated with delayed organ development of integral body systems, particularly the immune structures. The spleen is the largest secondary immune organ in the mammalian body and is responsible for initiating immune responses to blood-borne antigens. It can only perform this function successfully if all of its tissue components are fully morphologically and functionally mature. The spleen was studied in meat production rabbits (early maturing crossbreed Hyplus) at 1, 10, 20, 30, 60 and 90 days of age. Morphological studies included anatomical dissection, clarification of topography, determination of mass parameters, preparation of smears and histological sections. Spleen histological sections were stained with haematoxylin and eosin, according to Van Gieson, and impregnated with silver nitrate, followed by microscopy. Qualitative and quantitative indicators of spleen cellular and tissue component development were determined using ImageJ software. It was found that in crossbred rabbits the topography of the spleen corresponds to general anatomical principles of localisation, has a fixed place and is an anatomically formed organ. In day-old animals, the histological differentiation of the spleen is limited to the connective tissue stroma and the parenchyma. The organ's parenchyma is formed by reticular tissue, with hematopoietic and lymphoid cells among the cells, without differentiation into white and red pulp. In 10-day-old rabbits, the white pulp is represented by the periarterial lymphatic sheath (PALS) and marginal zones. Single primary lymphoid nodules without germinal centres are seen in the spleen from 20 days of age. By 30 days of age, the white splenic pulp has all the major structural and functional zones, including formed lymphoid nodules with germinal centres and mantle zones. During the second and third months of life, the spleen gradually increases the relative area of all white pulp functional zones, reaching a maximum at 90 days of age. In productive rabbits, the cellular composition of the splenic white pulp is represented by lymphocytes (small, medium, large), reticular cells, macrophages and plasma cells. During postnatal ontogenesis, the number of small lymphocytes increases in all white pulp functional zones, reaching maximum values in 90-day-old animals. On the contrary, the relative number of medium and large lymphocytes decreases. The number of reticulocytes in the periarterial lymphatic sheath and lymphoid nodules zones does not change significantly, and in the marginal zone it decreases substantially by the end of the study. The results of determining the timing of morphological and functional maturation of immunocompetent structures in the spleen of meat rabbits are of great biomedical and economic importance. They will serve as a control for comparing changes in rabbit spleen lymphoid tissues during the development of pathological processes, as well as under the influence of external factors.

Antibody preparation and age-dependent distribution of TLR8 in Bactrian camel spleens

BackgroundToll-like receptor 8 (TLR8) can recognize specific pathogen-associated molecular patterns and exert multiple immunological functions through activation of signaling cascades. However, the precise distribution and age-related alterations of TLR8 in the spleens of Bactrian camels have not yet been investigated. This study aimed to prepare a rabbit anti-Bactrian camel TLR8 polyclonal antibody and elucidate the distribution of TLR8 in the spleens of Bactrian camels at different age groups. The methodology involved the construction of the pET-28a-TLR8 recombinant plasmid, followed by the expression of TLR8 recombinant protein via prokaryotic expression. Subsequently, rabbits were immunized with the purified protein to prepare the TLR8 polyclonal antibody. Finally, twelve Alashan Bactrian camels were categorized into four groups: young (1–2 years), pubertal (3–5 years), middle-aged (6–16 years) and old (17–20 years). These camels received intravenous sodium pentobarbital (20 mg/kg) anesthesia and were exsanguinated to collect spleen samples. Immunohistochemical techniques were employed to observe and analyze the distribution patterns and age-related changes of TLR8 in the spleen.ResultsThe results showed that the TLR8 recombinant protein was expressed in the form of inclusion body with a molecular weight of 52 kDa, and the optimal induction condition involved 0.3 mmol/L IPTG induction for 8 h. The prepared antibody yielded a titer of 1:32 000, and the antibody demonstrated specific binding to TLR8 recombinant protein. TLR8 positive cells exhibited a consistent distribution pattern in the spleen across different age groups of Bactrian camels, primarily scattered within the periarterial lymphatic sheath of the white pulp, marginal zone, and red pulp. The predominant cell type expressing TLR8 was macrophages, with expression also observed in neutrophils and dendritic cells. Statistical analysis revealed that there were significant differences in the distribution density of TLR8 positive cells among different spleen regions at the same age, with the red pulp, marginal zone, and white pulp showing a descending order (P<0.05). Age-related changes indicated that the distribution density in the marginal zone and red pulp exhibited a similar trend of initially increasing and subsequently decreasing from young to old camels. As camels age, there was a significant decrease in the distribution density across all spleen regions (P<0.05).ConclusionsThe results confirmed that this study successfully prepared a rabbit anti-Bactrian camel TLR8 polyclonal antibody with good specificity. TLR8 positive cells were predominantly located in the red pulp and marginal zone of the spleen, signifying their pivotal role in the innate immune response of the spleen. Aging was found to significantly reduce the density of TLR8 positive cells, while leaving their scattered distribution characteristics unaffected. These findings provide valuable support for further investigations into the immunomorphology and immunosenescence of the spleen in Bactrian camels.

Comparative Histomorphological Study of Spleen in Goat, Sheep, Cow and Buffalo

Immunity in some animals, such as goats, stands for unique properties by which they withstand unpleasant environments. Spleen influences animals' immunity through its hematopoietic and immunological roles. Because of that, we emphasized the average morphological and histological properties of the Goat, Sheep, Cow and Buffalo spleen. We randomly collected three spleens for every species from healthy slaughtered Goats, Sheep, Cows and Buffalo. All the animals were adult and weighed about 71, 80, 400 and 500 grams, respectively. Histological cutting was followed according to the paraffin method; then, the samples were ready for microscopic exam. The structure results revealed that the spleen of animals is supported externally by a thick fibromuscular connective tissue capsule, which vascularizes by subcapsular sinus. Stromal trabeculae emerge from the capsule, entering the splenic parenchyma. The parenchyma of animals' spleen differentiates into white pulp, which organizes into the periarterial lymphatic sheath along with lymph nodules, red pulp ropes in splenic sinuses among splenic cords, and marginal zone in which macrophage occupies excellent importance. This study concluded that the amount of smooth muscle in the spleen septa in goats was more than in the rest of the experimental animals. Keywords: Histomorphology; Spleen; Goat; Sheep; Cow; Buffalo

Morphological, histological, and histochemical study of the adult golden hamster (Mesocricetus auratus) spleen.

The golden hamster is a choice model for investigating many visceral and splenic infections and neoplastic and retrospective lesions. To study hamsters' spleen's morphological, histological, and histochemical structure. Samples were collected from eight healthy adult golden hamsters and then fixed with 10% buffered formalin. Later, samples were processed, sectioned, and stained with Hematoxylin and Eosin as well as Masson's Trichrome stain. Other slides were further stained with Periodic Acid Schiff and Alcian blue 2.5 stain (PAS) for histochemical evolution; the gross measurement was performed for the splenic length, width, and thickness, while the histological measures included the splenic capsular and trabecula thickness, diameter of white pulp follicles, splenic sinusoids and central arteries and proportion of white and red pulps. The macroscopic findings revealed that the spleen was red-brown lanciform on the left side of the dorsolateral abdominal wall. The morphological measurements for splenic length, width, and thickness were 26.6 ± 7.67, 4.17 ± 1.65, and 1.70 ± 0.01 mm, respectively. The histological observations showed that the splenic capsule was composed of two layers (serosal and subserosal). The inner layer sends trabeculae dividing the splenic parenchyma irregularly, and the splenic parenchyma comprises the white and red pulp. The white pulp follicles included the mantle, marginal zones, and the PALS (periarterial lymphatic sheath), while the red pulp constituted splenic cords and sinuses. The histomorphological findings showed that white pulp follicles and the central artery mean diameter were 252.62 ± 8.07 µm and 54.45 ± 0.36 µm respectively, the proportion of white to a red pulp was 0.49 ± 0.01, the splenic capsule, trabecula and the wall of splenic arteries showed an intense positive activity to PAS stain and negative or weak in other splenic structures. The similarities and differences in the spleen between the laboratory animals and hamsters were apparent in this article, so understanding the morphological and histological structure of the spleen presents significant assistance with species identification to select the appropriate experimental animal model in future medical research.

Sympathetic innervation of human and porcine spleens: implications for between species variation in function

BackgroundThe vagus nerve affects innate immune responses by activating spleen-projecting sympathetic neurons, which modulate leukocyte function. Recent basic and clinical research investigating vagus nerve stimulation to engage the cholinergic anti-inflammatory pathway (CAP) has shown promising therapeutic results for a variety of inflammatory diseases. Abundant sympathetic innervation occurs in rodent spleens, and use of these species has dominated mechanistic research investigating the CAP. However, previous neuroanatomical studies of human spleen found a more restricted pattern of innervation compared to rodents. Therefore, our primary goal was to establish the full extent of sympathetic innervation of human spleens using donor tissue with the shortest procurement to fixation time. Parallel studies of porcine spleen, a large animal model, were performed as a positive control and for comparison.MethodsHuman and porcine spleen tissue were fixed immediately after harvest and prepared for immunohistochemistry. Human heart and porcine spleen were stained in conjunction as positive controls. Several immunohistochemical protocols were compared for best results. Tissue was stained for tyrosine hydroxylase (TH), a noradrenergic marker, using VIP purple chromogen. Consecutive tissue slices were stained for neuropeptide Y (NPY), which often co-localizes with TH, or double-labelled for TH and CD3, a T cell marker. High-magnification images and full scans of the tissue were obtained and analyzed for qualitative differences between species.ResultsTH had dominant perivascular localization in human spleen, with negligible innervation of parenchyma, but such nerves were abundant throughout ventricular myocardium. In marked contrast, noradrenergic innervation was abundant in all regions of porcine spleen, with red pulp having more nerves than white pulp. NPY stain results were consistent with this pattern. In human spleen, noradrenergic nerves only ran close to T cells at the boundary of the periarterial lymphatic sheath and arteries. In porcine spleen, noradrenergic nerves were closely associated with T cells in both white and red pulp as well as other leukocytes in red pulp.ConclusionSympathetic innervation of the spleen varies between species in both distribution and abundance, with humans and pigs being at opposite extremes. This has important implications for sympathetic regulation of neuroimmune interactions in the spleen of different species and focused targeting of the CAP in humans.

Epidermal growth factor receptors and their ligands are expressed in the spleen of the Japanese Quail (Coturnix coturnix japonica) during the post-hatch period

ABSTRACT 1. The epidermal growth factor (EGF) family plays an important role in the development, differentiation, migration and apoptosis of cells, as well as in wound healing, which are all essential to the viability of multicellular organisms. The avian spleen is a principal organ of systemic immunity and its importance in disease resistance is presumably accentuated by the scarcity of avian lymph nodes. 2. The aim of this study was to determine whether EGF receptors (ErbB1-4) and their ligands (EGF, AREG and NRG) are expressed in the structural components of the quail spleen during the post-hatch period. At each selected age, from 1 d to 7, 14, 21 and 60 d, 10 quails were euthanised under ether anaesthesia and their spleens were fixed in a 10% formaldehyde-alcohol solution. Following routine histological processing, the streptavidin-biotin-peroxidase method was used for immunohistochemical examination. 3. Strong cytoplasmic immunoreactions for ErbB2, ErbB4 and NRG were observed in the ellipsoid associated cells (EAC) of the quail spleen throughout the post-hatch period. This immunoreactivity in the EAC increased after the 7th d post-hatch. ErbB1 and ErbB3 immunoreactions were relatively similar and weak in all components of the spleen during the post-hatch period. Some immune cells of the peri-arterial lymphatic sheath (PALS) and peri-ellipsoidal lymphatic sheath (PELS) showed positive immunoreactivity for the ErbB receptors and their ligands. In the vascular smooth muscle cells, immunoreactivity for ErbB2 was stronger than that for the other ErbB receptors and their ligands. 4. The data showed that ErbB receptors and their ligands (EGF, AREG and NRG) are expressed by different structural components of the quail spleen during the post-hatch period.

Porcine reproductive and respiratory syndrome virus reinfection causes the distribution of porcine interleukin-4 in close proximity to B lymphocytes within lymphoid follicles and a reduction in B and T lymphocytes

Interleukin 4 (IL-4) plays a major role in T-lymphocyte development and is thought to be a central regulator as a cofactor in resting B-lymphocyte proliferation. Primary infection with porcine reproductive and respiratory syndrome virus (PRRSV) induces minimal IL-4 production, whereas an IL-4 response occurs in the peripheral blood of piglets reinfected by PRRSV. The locations and interaction partners for the massive volume of IL-4 triggered by PRRSV reinfection remain unclear. This study aimed to investigate the characteristics of IL-4 secretion and location changes in peripheral immune organs induced by PRRSV infection and reinfection. Our results show that PRRSV reinfection induced higher levels of IL-4 mRNA and protein expression in the peripheral immune organs (e.g., lymph node and spleen) and peripheral blood compared with PRRSV primary infection. Importantly, we found that, following PRRSV reinfection, an obvious large-scale migration of IL-4 occurred in the lymph nodes. During PRRSV primary infection, IL-4 was mainly concentrated around the lymphoid follicles and paracortical regions of the lymph node and also located in the marginal area and periarterial lymphatic sheath region of the spleen. During PRRSV reinfection, the now abundant IL-4 gathered into the lymphoid follicles of the lymph node and spleen. Notably, IL-4 changed its location state from scattered and sparse during primary infection to clinging to B lymphocytes in the lymphoid follicles during reinfection. During reinfection, IL-4 was often co-localized with T and B lymphocytes; furthermore, the percentages of several T lymphocyte subsets, N protein-specific antibody levels, and viral load in the peripheral blood or lymph tissues underwent remarkable variation. Another important finding of this study was that the numbers of B lymphocytes and T lymphocytes in the lymphoid nodes were significantly reduced after PRRSV infection or reinfection, presumably due to PRRSV-induced acute bone marrow failure and autophagy in thymic epithelial cells. This study revealed the characteristics of IL-4 migration and distribution in the peripheral lymph organs induced by PRRSV reinfection and provides valuable clues for further exploration of the interactions between IL-4, B lymphocytes, and T lymphocytes during PRRSV infection and reinfection.

Histological features of the spleen and pancreas of the Egyptian Nile monitor lizard (Varanus niloticus)

The present study investigated the histological features of the spleen and pancreas of the Egyptian Nile monitor lizard (Varanus niloticus). After gross observations the representative samples were collected from three adult male monitor lizards and were processed for histological examination. Grossly, the spleen was brownish in color and embedded in the mesentery near the stomach and pancreas. The pancreas was yellowish in color and consisted of a ventrally placed principal lobe which was pyramidal and extended in the mesentery, whereas the dorsally located splenic lobe was small and was attached to the spleen. a single duct was emerged from the pancreas. Histologically, the spleen was nodular and was encapsulated by the connective tissue capsule covered by mesothelium. it was composed of white and red pulps. Many ellipsoids, melano-macrophage centers, periarterial and periellipsoidal lymphatic sheaths (PALS and PELS; respectively) were observed. The pancreas demonstrated a thicker capsule covered by mesothelium and the exocrine portion was formed of tubule-acinar cells. Main pancreatic duct passed across the length of the pancreas and was lined by stratified cuboidal epithelium. The Islets were found in the ventral lobe where it was in various sizes. This is the first report about the histological structure of the spleen and pancreas of the Egyptian Nile monitor lizard, where the spleen showed a unique structure while the pancreas was similar to what was described in other varanus species. The information in this study will be a baseline knowledge for future studies regarding endocrine or immune organs in this species.

Scanning Electron Microscopic Study of the White Pulp of Spleen in Adult Goats

The white pulp of the spleen in adult goats was thoroughly screened under the scanning electron microscope (VEGA3 TESCAN). The study revealed the presence of lymphoid and non-lymphoid cells. The T lymphocytes and B lymphocytes could morphologically be differentiated under the magnification of 8000 to 20000 times by Scanning electron microscopy. The changes in number of cells in relation to the age was also recorded. The peri arterial lymphatic sheath and its structural connections with Reticular cells and other non-lymphoid cells were clearly demonstrated. The types of lymphoid cells and their arrangement around the central arteries upto the marginal zones were observed in detail. A conclusion about the type of circulation in the spleen of goats was arrived after three dimentional observation of the tissue under scanning electron microscope.

Tissue distribution and developmental changes of PTEN in the immune organs of chicken and effect of IBDV infection on it

Phosphatase and tensin homolog (PTEN), a tumor suppressor gene, functions in antiviral innate immunity and regulates the development and function of T cells and B cells. However, limited information about PTEN is available in poultry. In the present study, quantitative real-time polymerase chain reaction and immunohistochemistry staining were used to study the tissue distribution and developmental changes of PTEN in the main immune organs of chicken. The effects of infectious bursal disease virus (IBDV) infection on PTEN mRNA expression in the bursa of Fabricius (BF) of chickens were also investigated. The results are as follows. 1) The order of PTEN mRNA expression levels at the 18th d of hatching (E18) was: muscle and immune organs (spleen and thymus) > visceral organs (heart, lung, kidney, and liver) > hypothalamus and digestive tracts (duodenum, jejunum, ileum, cecum, proventriculus, BF [originates from cloaca], and cecum tonsil [locates at the lamina propria of cecum]). However, at the 15th d of raising (D15), the PTEN mRNA expression in the heart was the highest among all the tissues, followed by those in the liver, proventriculus, and kidney. The PTEN mRNA expression levels in the rest tissues were very low and were only 1.20 to 19.47% as much as that in the heart (P < 0.05). 2) The changes in the expression of PTEN mRNA in the BF, spleen, and thymus from E15 to D15 had no obvious regularity. PTEN-immunopositive (PTEN-ip) cells in the BF were distributed in epithelium mucosa, bursal follicles and interfollicles before hatching, but only in bursal follicles after hatching. PTEN-ip cells in the spleen were expressed in the periarterial lymphatic sheath from E18 to D15. Most of PTEN-ip cells distributed in the thymic medulla and only a few distributed in the thymic cortex during the whole experiment. 3) Chicken with IBDV infection had a remarkable decrease in PTEN mRNA expression from 1 d postinfection (dpi) to 7 dpi. Although PTEN mRNA level was reversed at 7 dpi, it was still significantly lower than that at 0 dpi (P < 0.05). These findings suggest that the PTEN of chicken might play important roles in the development of embryos and T/B lymphocytes, and the downregulation of PTEN in chickens infected with IBDV might be a mechanism of IBDV evasion from host immunity. Strategies designed to restore PTEN expression may be a therapy for preventing chickens from IBDV infection.

Effect of Selenium on Quantitative Structural Changes in Dexamethasone-Induced Immunodeficiency Rat Models.

Background: One of the major indices of immunodeficiency is lymphoid organ atrophy. Some trace elements are candidates for the treatment of this defect. These conditions may induce structural changes in the sub-components of lymphoid organs. Therefore, this study evaluated the effect of selenium on volumetric changes in dexamethasone (DEX)-induced lymphoid organ atrophy in an animal model.Methods:This study was conducted at Histomorphometry and Stereology Research Centre, Shiraz University of Medical Sciences, Shiraz, Iran, in September 2016 to September 2017. Thirty-two male rats were divided into four groups: Group I; control (normal saline, 0.5 mL/kg, intraperitoneally), Group II; DEX (0.4 mg/kg; intraperitoneally), Group III; selenium plus DEX (similar to Group II and Group IV), and Group IV; selenium (0.1 mg/kg; orally). At the end of the experiment, the rats’ thymus, spleen, and lymph nodes were removed, processed, and stained by hematoxylin and eosin (H&E). The volume and volume density of theses organs were estimated by stereology. The results were analyzed using the Mann–Whitney U-test and the Kruskal–Wallis test.Results: The volume of the thymus as well as its cortex and medulla; the volume of the spleen as well as the volume density of its white pulp, periarterial lymphatic sheath zone, and follicles; and the volume of the lymph nodes as well as their inner (P=0.001) and outer (P=0.007) cortices showed a significant reduction in the DEX-treated animals in comparison with the controls. In the DEX plus selenium-treated animals, maximum effects were observed on the increment in the thymic cortex (P=0.001), the outer cortex of the lymph nodes (P=0.012), and the splenic follicles (P=0.018) in comparison with the DEX group. There was no significant difference between the animals receiving selenium treatment and the controls in terms of lymphoid organs. Conclusion: Selenium may improve lymphoid organ structures in an immunodeficiency rat model but has no effect on normal lymphoid tissues.

Microscopical Study of White and Red Pulp of Spleen of Young Porcine (Sus Scrofa) Spleen

The spleen of six young porcain aging (2 month) each spleen was examined macroscopically and was studied using light and transmission electron microscopy. The spleen is an important lymphoid organ of immunological defence. The type of pig spleen is sinusal type spleen It has a role in many hematological functions as storage, filtration and maturation of reticulocytes. The splenic parenchyma of young pig consisted of white and red pulp the whit pulp formed of few sporadic lymphocytes and seldomly found lymphatic nodules, occasionally single ovoid mass of lymphoid nodule the peri-arterial lymphatic sheath was few in pig spleen Ellipsoids (pericapillary macrophage sheath) were not clearly defined in the marginal zone which is not well demarcated. The red pulp occupied the majority of pig spleen it consisted of splenic cords and presence of few and small sized blood sinusoids lined by flat endothelial cells containing different blod cells as neutrophils and blood platlets. In spite of that the histology of the spleen was describe in different animals species, to the best of our knowledge there is no available literature concerned with the histology of the Egyptian pig spleen so we aimed in our study to illustrate the main difference of splenic parechyma.

Morphological characterization of postembryonic development of blood–spleen barrier in duck

The spleen is the largest peripheral lymphoid organ and an important site of immune response, in which the blood–spleen barrier (BSB) plays a significant role to resist various pathogens. The BSB structure of duck spleen is different from that of chicken and mammals. However, no information about the development of BSB after the postembryonic age has been reported in ducks. The current study observed the spleen of 1, 7, 14, 21, 35, and 60-day-old ducks by light and electron microscopy to analyze the cellular structural development. The results showed that the spleen index was continuously increased from 1 to 14-day-old ducks. During their early age, the spleen of ducks showed no definite zone of white and red pulp, but the area of the white pulp was large compared to that of the red pulp. The diameter of the ellipsoid was constantly increased in up to 35-day-old duck spleen, while the periellipsoidal lymphatic sheath (PELS) and periarterial lymphatic sheath continuously developed after 1 D. The reticular fibers developed with age; their branching reached the ellipsoidal wall to show a developed framework in the BSB of 14-day-old ducks. After 7 D, the endothelial cells of the sheathed capillary showed a typical cuboidal shape; between these cells, the gaps increased as age advanced, while the thickness of the basement membrane and collagen fibers increased in 35-day-old ducks. The mechanical filtration function of BSB by intravenous injection showed a 1-layer ring of carbon particles restricted in the white pulp in 1-day-old duck spleen; however, in 14 to 60 D, these particles were restricted in the ellipsoid and PELS, forming 2-layer rings of carbon particles. Collectively, the cellular features of the duck BSB developed up to 35 D of postembryonic age to perform their immune function.

Architecture and Cellular Composition of the Spleen in the Japanese Quail (Coturnix japonica).

The spleen is considered a key player in birds' immunity. The stroma and the parenchyma of the spleen of the adult quail were demonstrated histologically, histochemically, and ultrastructurally. A thin capsule and the absence of trabeculae were the most characteristics of spleen stroma. The demarcation between white pulp and red pulp was not observed in the quail. White pulp formed from the periarterial lymphatic sheath and the periellipsoidal lymphatic sheath, both of which were surrounded by arteriole and ellipsoid, respectively. Ellipsoids appeared more numerous and were characterized by cuboidal lining of the epithelium and supporting cells. Red pulp consisted of sinuses and cords. White pulp and red pulp of the quail spleen contained various cells, such as red blood cells, macrophages, heterophils with characteristic granules, lymphocytes of different sizes, dendritic cells, plasma cells, and telocytes. In addition, closed circulation and open circulation established the blood flow on the spleen.

Aging-associated immunosenescence via alterations in splenic immune cell populations in rat

AimImmunosenescence is the decline of the host immune system due to aging, resulting in various complications. The splenic lymphoid nodule is the pivotal compartment involved in immunosenescence. In this study, we investigated the important changes in the splenic immune cell populations of aged rats (18–24 months) in comparison with young rats (3–5 months). Materials and methodsWe, also, studied the effects of aging on the activities of total superoxide dismutase (T-SOD) and malondialdehyde (MDA) levels in spleen of both groups, besides the changes of the splenic architecture. Furthermore, immunohistochemical staining was performed to detect the aging effects in T cells, B cells, macrophages, granulocytes, mast cells, proliferating cells, apoptotic cells, and cells positive for interleukin-1β (IL-1β), interleukin-6 (IL-6), and Toll-like receptor 4 (TLR4). Key findingsThe aged rats had significantly lower spleen/body weight ratios and smaller splenic nodules, indicating a decline in general immunity in them. With aging, T-SOD activities were decreased, while MDA levels were increased, exhibiting that oxidative stress increases in spleens. In addition, the aged group also had significantly fewer T and B cells, macrophages, granulocytes, IL-6 and TLR4 immuno-positive cells, and proliferating cells in the periarterial lymphatic sheaths, marginal zone, and lymphoid follicles compared with the young group. On the other hand, the number of mast cells and apoptotic cells was significantly increased with age. Therefore, we can conclude that cellular immunity and humoral immunity were crumpled with age.

A New Role for the Spleen: Aggravation of the Systemic Inflammatory Response in Rats with Severe Acute Pancreatitis

Little is known about the role of the spleen in mediating systemic inflammatory responses in severe acute pancreatitis (SAP). We investigated the role played by the spleen in rats after SAP induction. Splenectomy was performed at designated time points after SAP induction. Pancreatic tissue and serum samples were collected and subjected to histologic, immunohistochemical, and immunologic analyses. After SAP induction, the splenic immune response was enhanced during SAP progression, as shown by the increased diameter of the splenic periarterial lymphatic sheath and the thickness of the splenic marginal zone. Rats with splenectomy developed acute pancreatitis more slowly than rats without splenectomy. In addition, pancreatic tissues of rats with splenectomy contained lower levels of serum amylase, tumor necrosis factor-α, and IL-6 and exhibited less acinar cell death, leukocyte infiltration, and interstitial edema than those of rats without splenectomy. Compared with splenectomy alone, cotreatment with splenectomy and the administration of splenic cells originating from a rat with SAP 12 hours after induction increased systemic inflammation in SAP rats. Splenic factors exacerbated SAP-associated liver and lung injury and accentuated intestinal mucosal barrier dysfunction. Splenectomy altered the serum cytokine profile in rats with SAP. In a rat model of SAP, the spleen exacerbated the systematic inflammatory responses and injury to multiple organs, indicating a new role for the spleen in SAP.

The dynamic distribution of duck Tembusu virus in the spleen of infected shelducks

BackgroundDuck Tembusu virus (DTMUV) is a novel member of Flavivirus. The isolated and purified DTMUV strain XZ-2012 was used as a strain model, to intramuscularly inject the six-month egg-laying shelducks with the infective dose of 104TCID50. The dynamic distribution of the virus in spleen at different time post-infection (pi) was studied using RT-PCR, RT-qPCR, ELISA, immunofluorescence and transmission electron microscopy (TEM).ResultThe results showed that the virus occurred in the spleen after 2 hpi and lasted up to 18 dpi. The registered viral load increased from 2 hpi to 3 dpi, and then it diminished from 6 dpi to 18 dpi with a slight rise at 12 dpi. From 2 hpi to 6 dpi the DTMUV particles were mostly distributed in the periellipsoidal lymphatic sheath (PELS) of spleen white pulp, few being found in the sheathed capillary. From 9 dpi to 18 dpi, the DTMUV particles were migrating into periarterial lymphatic sheaths (PALS) around the central artery through the red pulp. Under TEM, the virus particles could be observed mostly in lymphocytes and macrophages.ConclusionIt was suggested that DTMUV invaded lymphocytes and macrophages of the spleen at 2 hpi and replicated significantly from 1 dpi to 3 dpi, being eliminated from 9 dpi to 18 dpi. This is the first study on the dynamic distribution of DTMUV from invasion to elimination in duck spleen conducted by molecular and morphological methods. It could provide theoretical basis for the occurrence, development and detoxification of the virus in the organs of the immune system.

Comparative Histoarchitectural Study of Splenic Components in Sheep and Goat

Animal husbandry practices like sheep and goat rearing offer many advantages for beginners and also experienced farmers. They are well suited and productive for the prevailing agro-climatic conditions in India. In mammals, health status is governed by the organ of lymphatic system. Though spleen is the secondary lymphatic organ, it is the largest among the lymphoid organs which plays a crucial role in immune function. For the present study, splenic tissue from adult sheep and goat (6 Nos. each) were collected from Corporation slaughter house, Chennai. Gross, microscopic and immunohistochemical observations were done. Micrometric observations of various structures were also recorded. The spleen of sheep was triangular, whereas, in goat it was roughly quadrangular in outline. In both the species, parenchyma was covered by a thick capsule, predominantly made up of collagen, elastic and smooth muscle fibres. Thickness of capsule in sheep was 150±10.33μ and in goat it was 141±11.5μ. Trabeculae originated from inner side of the capsule and extended in to parenchyma in both the species. Parenchyma was made up of white pulp and red pulp and the proportion of these were almost equal. The white pulp was composed of splenic nodule and peri-arterial lymphatic sheath. Red pulp was compost of irregular splenic cords separated by splenic sinusoids. Immunohistochemical localization of T-lymphocytes revealed the distribution of T-lymphocytes around the PALS and also scattered in the red pulp.

Age-related histomorphological studies on the spleen of Chinese yellow quail

This study was aimed to investigate the morphology, growth, histological structure and development of the spleen of the Chinese yellow quail at different ages(from 0 to 38 weeks). The yellow Quail spleen was spherical, brownish red, located at the right dorsal of the junction of the muscle stomach and glandular stomach. The absolute quality, length and width of the spleen gradually increased with age, reaching a maximum for each of these measurements at the age of 6 weeks and then remainied unchanged between 10 and 26 weeks of age. Finally, these measurements of the spleen were no significant difference at 30 weeks of age compared to at 34 to 38 weeks (P&gt;0.05). Although, these measurements were found to gradually decrease during 30-38 weeks of age, the decrease was not obvious. The basic structure of the spleen included red pulp, white pulp and the edge area. In addition, the average area and diameter of the splenic nodules, the white pulp ellipsoid and the periarterial lymphatic sheath gradually increased, reaching maximum sizes at the age of 26 weeks. It was seen that a sustained developmental period occurred at 0-6 weeks of age and a maturation period occurred at 10-38 weeks during the development of quail spleen.

Molecular identification and gene expression profiles of the T cell receptors and co-receptors in developing red-tailed phascogale (Phascogale calura) pouch young

Expressed coding sequences were identified for the T cell receptors TCRα, TCRβ, TCRγ, TCRδ, TCRμ, and co-receptors CD3ε, CD4, and CD8α in the red-tailed phascogale (Phascogale calura). Expression of these genes was examined in the developing thymus and spleen of pouch young and juvenile individuals to detect any changes in expressionrelative to developmental stages. Transcripts of CD3ε, CD4, TCRδ and TCRμ were first detected in the thorax at 1 day postpartum (dpp), and transcripts of all target genes were detected in the thymus from 3 dpp, and the spleen from 17 dpp. The relative expression of all target genes changed significantly over the course of pouch life and was associated with histological milestones of both the thymus and the spleen. Peak gene expression was observed early in pouch life in the thymus, with a decrease in expression associated with timing of thymic involution, while expression peaked later in the spleen and was associated with the initial differentiation of the red and white pulp and a population of T cells forming the periarterial lymphatic sheath (PALS). Active expression of these target genes within the thymus and spleen indicates that the young marsupials are actively developing immunocompetence during pouch life.
