Curcuma and Scutellaria baicalensis extracts reduced thermoregulatory behavior in chickens subjected to moderate heat stress

Using phytogenic additives represents a new trend in the poultry feed industry because of purported beneficial impacts. There is limited information about the effects on chicken behavior, but based on new studies, some plant-based active compounds influence animal cognitive function. Therefore, the primary objective of this experiment was to investigate the impact of a feed additive consisting of a mixture of Curcuma and Scutellaria baicalensis on behaviors associated with exposure to heat stress in broiler chickens. Chickens before 21 days of age were kept in 12 littered floor boxes according to guidelines for Cobb 500 hybrid. The Cobb 500 chickens of both sexes were fed identical diets until they reached 21 days of age. From this point onward, the experimental group received in the diet a 0.1% supplement containing Curcuma and Scutellaria baicalensis extracts. The control group received a feed mixture without phytogenic additives. Both groups had six replicates, and each group comprised 75 chickens. The pens of experimental and control group were randomly selected. The experimental period spanned from 28 to 35 days of age, during which the chickens were subjected to mild constant heat stress (28°C). Ethological observations were conducted at 31, 32, and 33 days of age, divided into three observation periods at the beginning, middle, and end of the daylight period (18h) using the visual scan sampling method. After the experimental period at 35 days of age, chickens from both groups exhibited nearly identical live body weights and daily feed intake. However, the experimental group demonstrated significantly higher daily weight gain (P < 0.05). Thermoregulatory behaviors such as panting and wing spreading were suppressed (P < 0.05) in the experimental group, while there was a simultaneous increase in the number of chickens that were feeding (P < 0.05). Similarly, age significantly influenced behavioral patterns (P < 0.05), whereas time of the day only affected the number of chickens engaged in feeding (P < 0.05). In conclusion, the feed mixture enriched with Curcuma and Scutellaria baicalensis extracts exhibited performance-enhancing and behavior-changing effects, providing significant insights into the potential of this feed additive in improving performance and reducing thermoregulatory behavior manifestations within a flock under heat-stress conditions, thereby informing the poultry feed industry of a promising new development.

S100A1ct: A Synthetic Peptide Derived From S100A1 Protein Improves Cardiac Performance and Survival in Preclinical Heart Failure Models.

The EF-hand Ca2+ sensor protein S100A1 has been identified as a molecular regulator and enhancer of cardiac performance. The ability of S100A1 to recognize and modulate the activity of targets such as SERCA2a (sarcoplasmic reticulum Ca2+ ATPase) and RyR2 (ryanodine receptor 2) in cardiomyocytes has mostly been ascribed to its hydrophobic C-terminal α-helix (residues 75-94). We hypothesized that a synthetic peptide consisting of residues 75 through 94 of S100A1 and an N-terminal solubilization tag (S100A1ct) could mimic the performance-enhancing effects of S100A1 and may be suitable as a peptide therapeutic to improve the function of diseased hearts. We applied an integrative translational research pipeline ranging from in silico computational molecular modeling and in vitro biochemical molecular assays as well as isolated rodent and human cardiomyocyte performance assessments to in vivo safety and efficacy studies in small and large animal cardiac disease models. We characterize S100A1ct as a cell-penetrating peptide with positive inotropic and antiarrhythmic properties in normal and failing myocardium in vitro and in vivo. This activity translates into improved contractile performance and survival in preclinical heart failure models with reduced ejection fraction after S100A1ct systemic administration. S100A1ct exerts a fast and sustained dose-dependent enhancement of cardiomyocyte Ca2+ cycling and prevents β-adrenergic receptor-triggered Ca2+ imbalances by targeting SERCA2a and RyR2 activity. In line with the S100A1ct-mediated enhancement of SERCA2a activity, modeling suggests an interaction of the peptide with the transmembrane segments of the sarcoplasmic Ca2+ pump. Incorporation of a cardiomyocyte-targeting peptide tag into S100A1ct (cor-S100A1ct) further enhanced its biological and therapeutic potency in vitro and in vivo. S100A1ct is a promising lead for the development of novel peptide-based therapeutics against heart failure with reduced ejection fraction.

Anti-fatigue effect of an enzymatically derived deer velvet extract through muscle damage recovery and improvement of antioxidant levels

Ethnopharmacological relevanceDeer velvet (DV) has been extensively used in traditional Oriental medicine to treat various diseases. Its pharmacological spectrum encompasses tonicity, longitudinal bone growth of adolescence, blood retention, hemopoiesis facilitation, enhancement of organ function, physical function improvement, and augmentation of physical vitality. Among its myriad effects, DV notably exhibits anti-fatigue properties; however, its specific mode of action is yet to be fully elucidated. Aim of the studyThis study was undertaken to investigate the anti-fatigue and exercise performance-enhancing effects of YC-1101(HENKIV®), an enzymatically derived DV extract, in C2C12 cell lines and forced swimming mouse models. Materials and methodsThe effect of YC-1101 on increasing cell growth and lowering lactate dehydrogenase (LDH) activity was assessed in C2C12. The antioxidative effects of YC-1101 and its mechanistic underpinnings were also evaluated in C2C12 cells. Moreover, mice were subjected to an exhaustive swimming test subsequent to 3 weeks of YC-1101 extract administration. Fatigue-associated biochemical parameters, such as LDH activity and lactate, superoxide dismutase, glutathione peroxidase, and malondialdehyde levels, were measured in serum and muscle tissues. ResultsYC-1101 significantly promoted myoblast growth and reduced LDH activity, indicative of a cell-proliferative effect. Notably, free radical scavenging assays and analysis of reactive oxygen species production and antioxidant-related mRNA expression corroborated the significant involvement of YC-1101 in antioxidation, an important mechanism in anti-fatigue processes. Furthermore, animal experiments demonstrated prolonged swimming endurance and inhibition of muscle LDH accumulation in forced swimming mouse models. Serum biochemical analysis further revealed significant modulation of the expression of anti-fatigue-related biomarkers. Various bioactive low-molecular-weight DV peptides were enriched in YC-1101 compared to YHC-BE-2038 (a DV extract without enzymatic digestion). The anti-fatigue effect of YC-1101 was significantly stronger than that of YHC-BE-2038. ConclusionsThese findings suggest the potential of YC-1101 as a nutraceutical adjunct in ameliorating fatigue, concurrently facilitating muscle damage recovery, and exerting antioxidant effects via the nuclear factor E2-related factor 2-Kelch-like ECH-associated protein-1 pathway. It can be assumed that the complex action of low-molecular-weight DV peptides produced during the enzymatic degradation process was effective, and the further research is needed.

Examination of the anabolic activity and mechanisms of action of the combination of Diosgenin and Ecdysterone in C2C12 myotubes

Plant steroids such as ecdysterone (ECDY) or diosgenin (DIO) have been associated with anabolic and performance-enhancing effects for years. However, the molecular mechanisms have not yet been extensively studied in skeletal muscle cells. Consequently, the anabolic activity and associated molecular mechanisms of ECDY and DIO alone and in combination were investigated in C2C12 myotubes. Dose-dependent effects of both compounds on myotube diameter, mRNA expression of IGF-1 and PI3KR1 as well as expression of myosin heavy chain (MHC) proteins were analyzed in differentiated C2C12 cells. In addition, the binding affinities to androgen and estrogen receptors were analyzed. Treatment with ECDY and DIO significantly induced hypertrophy of C2C12 myotubes. Partially additive effects were observed. This is supported by the mRNA expression of IGF-1 and PI3KR1 as well as in the expression of MHC. However, no clear statement can be made regarding which combination has the strongest additive effects. Besides the results suggest that, in contrast to ECDY, DIO has antiandrogenic effects and bind on AR. Consequently, it indicate that two different mechanisms of action are activated in ECDY and DIO combinations. However, this must be confirmed in further cell cultures studies and human interventions concerning anti-doping regulations.

Long-Term Excretion of Roxadustat in Urine.

Roxadustat (FG-4592), an orally active hypoxia-inducible factor prolyl hydroxylase stabilizer, has been shown to enhance erythropoiesis by increasing endogenous erythropoietin. It is indicated for the treatment of anemia and chronic kidney disease and is approved for clinical use in several countries, including the European Union, Japan and others. Due to its reasonably anticipated performance-enhancing effect in athletes, roxadustat is prohibited for use in sports at all times. A few cases of adverse analytical findings in routine doping controls have been reported worldwide, some of which were claimed to be the result of contaminated dietary supplements. The present study offers new data demonstrating the long-term excretion pattern of roxadustat. Even after a single-dose administration, roxadustat can remain detectable in urine for 8 months, albeit at very low concentrations (<10 pg/mL). Following three times a week treatment with 70 to 100 mg of roxadustat, the drug was still detectable in the urine of anemic patients for between 9 and 18 months after treatment was discontinued. Lastly, an athlete who admitted use of roxadustat for almost a year (50 mg 3 to 5 times a week) has now tested positive multiple times over the course of 15 months (the first test being 12 months after the drug was discontinued), with an estimated concentration of roxadustat between 3 and 8 pg/mL. Altogether, these findings indicate the unusually prolonged terminal excretion kinetics of roxadustat, a property that testing authorities should consider in their results management process.

Substance Use and Mental Health among Canadian Social Workers.

This article reports the findings of an online survey designed to collect information about substance use (licit, illicit, or pharmaceutical) and mental health (depression or anxiety) among social workers. Among the 489 participants, Patient Health Questionnaire (PHQ-9) and Generalized Anxiety Disorder (GAD-7) screenings indicated symptoms of depression and anxiety at a higher prevalence than those of the general Canadian population. There were relatively few correlations between mental health scores and substance use. PHQ-9 total score significantly predicted past-year antidepressant use and past-year sleeping medication use. GAD-7 total score significantly predicted past-year benzodiazepine use and past-year melatonin use. Effects of substances (e.g., cannabis, alcohol, benzodiazepines, cocaine, ecstasy) were predominantly beneficial or nonproblematic (e.g., enjoyment/pleasure; socializing enhanced; concentration/focus improved). Subjective experiences of social workers should be sought to understand potential relationships between mental health scores and enhancement effects of substance use. Substances are being used, at least in part, for their performance-enhancing effects to meet the expectations of day-to-day life. Interventions can shift toward root causes, with institutions held more accountable for supporting social workers and promoting "workplace care."

Total Quality Management Lean Practices and Firm Performance: Integrated Approach Using MBNQA Criteria in the Thai Automotive Industry

Abstract In an increasingly competitive global automotive landscape, the strategic integration of Total Quality Management (TQM) and Lean Practices (LP) has emerged as a crucial determinant of operational excellence and sustained organizational performance. Grounded in empirical evidence from the Thai automotive sector, this study explores the intricate synergies between TQM with Malcom Baldrige National Quality Award (MBNQA) dimensions, lean implementation, and firm performance. This study employs the quantitative research method, where the measurement scales for each construct were adopted from the literature and reviewed by academics. Data were collected from 854 manufacturing firms in Thailand through a mail survey method. This study applied a two-step structural equation modeling (SEM) process. The findings reveal a robust direct effect of TQM (leadership, strategy planning, customer and workforce focus) on firm performance, underscoring its pivotal role. Notably, TQM also exhibited a substantial indirect effect on business performance, mediated sequentially through lean practices including continuous flow, 5S, and total preventive maintenance. Lean practices similarly demonstrated significant direct and indirect effects on firm performance. Collectively, the combined effects of MBNQA and lean practices on firm performance was also observed, substantiating their complementary contributions. These findings hold significant implications, both from an academic and a practical standpoint. From a theoretical perspective, this research enriches the discourse by providing empirical insights into the mediating roles of lean practices, elucidating the paths through which TQM's performance-enhancing effects can be transmitted. Concurrently, the magnitude of the impact these practices have on firm performance underscores their practical relevance, and insights derived from this study could play a key role in guiding automotive firms and suppliers to align critical components to drive performance excellence.

Effect of Mindful Sport Performance Enhancement in College Athletes for Reducing Sports-Caused Anxiety and Improving Self-Awareness: A Critically Appraised Topic

Mindful Sport Performance Enhancement (MSPE) training is a relatively new concept, which focuses on helping athletes manage a variety of stressors experienced throughout a season, including performing well academically, staying fit, having a productive season in their sport, and maintaining a healthy social life. A need for a critical appraisal is needed to assess the effectiveness of the intervention. Two cohort studies and one randomized control trial were included in the study and assessed using STROBE and PEDro Scale. Key results show, all 3 studies identified participants experiencing benefits after MSPE with aspects of awareness, acceptance, and emotion regulation. Furthermore, student-athletes who attended either all the sessions or more sessions after the 6-week course showed greater satisfaction with mental and physical health. Overall, there is level “B” evidence to support effectiveness of MSPE for college athletes in reducing sport anxiety and improving their overall well-being.

ESG ratings and corporate success: analyzing the environmental governance impact on Chinese companies’ performance

This study explores the impact of ESG ratings on corporate performance, focusing on achieving sustainable development and corporate sustainability through innovation within the context of high-quality global economic growth. In recent years, ESG ratings have garnered significant attention in the financial sector, influencing corporate strategy and performance management. While some argue that ESG activities might detract from profitability, others highlight that firms with strong ESG performance can access low-cost capital, thereby enhancing overall performance. Using a sample of China’s A-share listed companies from 2009 to 2021, this research examines the influence and mechanisms of ESG ratings on corporate performance. The findings indicate a significant positive relationship between ESG ratings and corporate performance, which remains robust after rigorous testing. Mediation analysis reveals that ESG ratings improve corporate performance by alleviating financing constraints and enhancing corporate reputation. Furthermore, the performance-enhancing effects of ESG ratings are more pronounced in firms with robust internal controls and private enterprises. This research provides empirical evidence to support stronger ESG investment and the refinement of the ESG rating system.

Refractory pruritus in cholestatic drug-induced liver injury secondary to androgenic anabolic steroids: Plasmapheresis to the rescue

Abstract: Anabolic androgenic steroids (AASs) have long been misused by professional athletes. Easy online and over-the-counter availability of AASs makes it an alluring option even among amateurs for their desired cosmetic and performance-enhancing effects. AASs have been associated with distinct forms of liver injury including transient transaminitis, peliosis hepatis, and hepatic tumors. An acute cholestatic syndrome with bland histology is another characteristic form of liver injury rarely reported with AASs. Plasmapheresis may provide a long-lasting relief of symptoms when conventional pharmacotherapeutic measures fail. Herein, we report a challenging case of a young male with refractory pruritus secondary to AASs, managed effectively with plasmapheresis.

Expanding the boundaries of digital orientation research: Scale development and validation

Firms keep struggling to realize the benefits of digital transformation processes. Research, therefore, proposes implementing a firm-wide organizational configuration with a digital orientation (DO) fostering digital innovation and transformation initiatives. However, exploration of DO remains somewhat constrained as existing measurement approaches are limited to secondary data. In this study, we develop a new scale to measure DO, specifically in survey-based research. Leveraging survey data from 1,488 top executives of German companies and drawing on the resource-based view, we empirically validate that our scale captures the performance-enhancing effects of DO.

Antidoping testing and testicle cancer diagnosis: An unexpected helper

Human chorionic gonadotrophin (hCG) is prohibited in male athletes due to its potential performance-enhancing effects. Some athletes with elevated hCG levels during routine doping tests have been diagnosed with testicular cancer, highlighting the inadvertent role of anti-doping screening in cancer detection. This study explores the incidental detection of testicular cancer through routine anti-doping tests among elite athletes, aiming to raise awareness about the potential secondary benefits of these screenings. We analyze cases from the past decade to demonstrate how early detection has facilitated timely medical interventions, allowing athletes to heal and return to competitive sports. Through these instances, athletes become unintentional advocates, contributing to public health awareness about this topic.

Kratom-From natural remedy to addictive drug and back

Kratom/ketum is apsychoactive herbal preparation that has been used for along time as aremedy and performance-enhancing substance in Southeast Asia. The advancement of globalization is making kratom increasingly more available in the western world, where it is becoming increasingly more used. The current research on kratom and its ingredients is presented. An overview of the use and effects of kratom is exemplary given on the basis of reports. The instrumentalization of the drug and its consequences up to the development of addiction are discussed. Consumption is accompanied by several instrumentalizeable effects so that kratom is used as atherapeutic substance in the self-management of pain, anxiety and depression as well as other substance addictions. Another benefit comes from the performance-enhancing effects on physical work and in a social context. Consumption is usually well controlled, rarely escalates and has few and mostly mild aversive side effects. The danger arises from consumption particularly when there is an escalation of the dose and from mixed consumption with other psychoactive substances. The main alkaloid mitragynine and the more potent 7‑hydroxy-mitragynine are considered mainly responsible for the effect. Both have acomplex pharmacology that involves partial µ‑opioid receptor agonism. Epidemiological, clinical and neurochemical studies have shown that kratom only has alimited addictive drug profile, which might suggest amedical use as aremedy or substitute in addiction treatment.

Label-Free and Ultra-Sensitive Detection of Dexamethasone Using a FRET Aptasensor Utilizing Cationic Conjugated Polymers.

Dexamethasone (Dex) is a widely used glucocorticoid in medical practice, with applications ranging from allergies and inflammation to cerebral edema and shock. Despite its therapeutic benefits, Dex is classified as a prohibited substance for athletes due to its potential performance-enhancing effects. Consequently, there is a critical need for a convenient and rapid detection platform to enable prompt and accurate testing of this drug. In this study, we propose a label-free Förster Resonance Energy Transfer (FRET) aptasensor platform for Dex detection utilizing conjugated polymers (CPs), cationic conjugated polymers (CCPs), and gene finder probes (GFs). The system operates by exploiting the electrostatic interactions between positively charged CCPs and negatively charged DNA, facilitating sensitive and specific Dex detection. The label-free FRET aptasensor platform demonstrated robust performance in detecting Dex, exhibiting high selectivity and sensitivity. The system effectively distinguished Dex from interfering molecules and achieved stable detection across a range of concentrations in a commonly used sports drink matrix. Overall, the label-free FRET Dex detection system offers a simple, cost-effective, and highly sensitive approach for detecting Dex in diverse sample matrices. Its simplicity and effectiveness make it a promising tool for anti-doping efforts and other applications requiring rapid and accurate Dex detection.

Interfacial B-O bonding modulated S-scheme B-doped N-deficient C3N4/O-doped-C3N5 for efficient photocatalytic overall water splitting

Photocatalytic overall pure water splitting is a promising method for generating green hydrogen energy under mild conditions. However, this process is often hindered by sluggish electron-hole separation and transport. To address this, a step-scheme (S-scheme) B-doped N-deficient C3N4/O-doped C3N5 (BN-C3N4/O-C3N5) heterojunction with interfacial B-O bonds has been constructed. Utilizing Pt and Co(OH)2 as co-catalysts, BN-C3N4/O-C3N5 S-scheme heterojunction demonstrates significantly enhanced photocatalytic activity for overall pure water splitting under visible light, achieving H2 and O2 evolution rates of 40.12 and 19.62 µmol/h, respectively. Systematic characterizations and experiments revealed the performance-enhancing effects of the enhanced built-in electric field and the interfacial B-O bonding. Firstly, the strengthened built-in electric field provides sufficient force for rapid interfacial electron transport. Secondly, by reducing the transport energy barrier and transfer distance, the interfacial B-O bonds facilitate rapid recombination of electrons and holes with relatively low redox potential via the S-scheme charge-transfer route, leaving the high-potential electrons and holes available for H+ reduction and OH− oxidation reactions. Overall, the photocatalytic efficiency of BN-C3N4/O-C3N5 S-scheme heterojunction was significantly improved, making it a promising approach for green hydrogen production through overall pure water splitting.

Simulation study of heat transfer enhancement for all-solid-state room temperature magnetic refrigeration system

To address issues relating to large thermal resistance and low heat transfer efficiency between and inside magnetocaloric materials (MCM) in all-solid-state room-temperature magnetic refrigeration technology, this study adopted the method of inserting high thermal conductivity materials (HTCM) into MCM and coupling Peltier elements between MCMs to enhance the heat transfer inside MCM and between microcells, thereby improving the refrigeration performance of the system. In doing so, a two-dimensional simulation model was developed using COMSOL Multiphysics software. The cooling performance enhancement effect of the system resulting from heat transfer improvement within MCM and between MCMs was investigated separately. Furthermore, the impacts of various system operating parameters (such as the number of micro-cell divisions, initial operating temperature, heat transfer time, magnetic field strength, Peltier input voltage), different HTCMs and insertion volume ratios on the system’s cooling performance were explored. The simulation results reveal that, in comparison with the original magnetic refrigeration system, the optimized heat transfer time of the system following enhanced heat transfer within the MCM is reduced from 180s to 8s, accompanied by a 115% increase in the maximum temperature span. Furthermore, coupling the Peltier elements elevates the maximum temperature span by 274%.

Hepatic Peliosis and Hemorrhage after Anabolic Steroid Abuse

AbstractAnabolic steroid abuse has become prevalent in modern society, with many athletes and bodybuilders using these substances for their performance-enhancing effects. A significant concern related to anabolic steroid abuse is the potential development of androgen-associated liver tumors such as hepatocellular carcinoma and adenomas. Anabolic steroid abuse has also been associated with hepatic vascular abnormalities like hepatic peliosis, a rare condition characterized by blood-filled cavities within the liver parenchyma. Hepatic peliosis can present with nonspecific symptoms or remain asymptomatic, making it difficult to diagnose without appropriate imaging studies. In this case report, the authors describe the imaging findings and management of two patients with intrahepatic bleeding associated with anabolic steroid consumption.

Doping control of estra-4,9-diene-3,17-dione in horses.

Estra-4,9-diene-3,17-dione (dienedione) is an anabolic-androgenic steroid (AAS) available on the market as a dietary supplement for bodybuilding. It is prohibited in both human and equine sports due to its potential performance-enhancing effect. With the rare presence of the 4,9-diene configuration in endogenous steroids, dienedione has been considered as a synthetic AAS. Nevertheless, the reoccurring detection of dienedione in entire male horse urine samples led to the investigation of its possible endogenous nature in horses, and its endogenous nature in entire male horses has been recently confirmed and reported by the authors' laboratory. While dienedione is not detected in castrated horses (geldings), it is essential to study its elimination and identify its metabolites for its effective control. To study the elimination and biotransformation of dienedione, administration experiments were performed by giving three castrated horses (geldings) each single oral dose of 1500 mg of dienedione powder for seven consecutive days. The postulated in vivo metabolites included 17-hydroxyestra-4,9-dien-3-one (M1a and M1b), hydroxylated dienedione (M2a, M2b, M3a, M3b, M4, M5) and hydroxylated M1 (M6a, M6b, M7a, M7b, M8a and M8b), formed from hydroxylation and reduction of dienedione. To control the misuse of dienedione in geldings, M3a and M3b are the potential targets that gave the longest detection time, which could be detected for up to 2-5 days in urine and 0.4-4 days in plasma.
