Pd-Catalyzed Coupling of Bromo-N-(β-glucopyranpsyl)quinolin-2-ones with Amides: Synthesis of N-glucosyl-6BrCaQ Conjugates with Potent Anticancer Activity.

A series of N-glycosyl- 6BrCaQ conjugates was synthesized through a Pd-catalyzed cross-coupling reaction between brominated N-glycosyl quinolin-2-one derivatives and various nitrogen nucleophiles. Antiproliferative assays revealed that this new series of analogues represents a promising class of antitumor compounds as illustrated by the high biological activity observed for several derivatives towards different cancer cell lines compared to the non-glycosylated congeners.

Synthesis of Heterocycles by a C–C Cross-Coupling/Alkyne-Carbonyl-Metathesis Strategy

AbstractThe present article presents a personal account on the synthesis of heterocycles by the combination of regioselective Pd-catalyzed cross-coupling reactions of polyhalogenated heterocycles, i.e., Suzuki–Miyaura and Sonogashira reactions, with alkyne-carbonyl-metathesis (ACM) reactions. The products, which show interesting optical, electronic or medicinal properties, are not readily available by other methods.

How do we get across? Impact of bridging aromatic units on the optical, electronic, and photovoltaic properties of donor-acceptor polymers

A series of donor - π - acceptor (D- π -A) type polymers (P1, P2, and P3) containing benzodithiophene (BDT) as donor unit and benzothiadiazole (BT) as acceptor unit were designed and synthesized by incorporating diverse electron-rich π bridges as thiophene (T), selenophene (Se) and thieno[3,2-b] thiophene (TT) via Pd-catalyzed Stille cross-coupling reaction. The effect of the various π-bridges on the optical and electrochemical properties of P1, P2, and P3 was examined, and for P1 and P2, the effect of π-bridges on photovoltaic properties was examined. Density functional theory (DFT) calculations were performed on tetramer model structures to understand the effect of π-bridges on the structural, electronic, and optical properties of the resulting polymers. Insertion of different π-bridges results in polymers with up-shifted highest occupied molecular orbital (HOMO). Thiophene π-bridged polymer P1:PC71BM showed the best photovoltaic performance with a power conversion efficiency (PCE) of 3.48 %, a short-circuit density (JSC) of 9.19 mA/cm2, an open-circuit voltage (VOC) of 0.75 V, and a fill factor (FF) of 50.30 %. The highest PCE obtained from a P2:PC71BM-based photovoltaic device was 2.42 %.

Synthesis and Properties of 4- and 10-Benzoyl-1-azapyrenes.

A series of 4- and 10-benzoyl-1-azapyrenes were prepared by a combination of Pd-catalyzed cross-coupling reactions and Brønsted-acid-mediated alkyne-carbonyl-metathesis (ACM). The photophysical and electrochemical properties of the products were studied and compared to theoretical results.

Adventures in CH-Arylation Chemistry

AbstractThe present article provides a personalized account on CH-arylation reactions employed for the synthesis of heterocycles. The presence of a nitro group allowed for direct and regioselective CH-arylations of pyrazoles, imidazoles, indoles and a variety of purine analogues. Direct CH-arylations without the presence of an activating nitro-group were employed for inter- and intramolecular reactions of purine derivatives, which allowed for the synthesis of a great variety of polycyclic systems. Domino C–N coupling / hydroamination / CH-activation reactions of diarylacetylenes with anilines allowed for the synthesis of polycondensated N-heterocycles. Products include indolo- and azaindolo[1,2-f]phenanthridines, quinolino[3′,4′:4,5]pyrrolo[1,2-f]phenanthridines, pyrimido[5′,4′:4,5]pyrrolo[1,2-f]phenanthridines, and benzothieno[2′,3′:4,5]pyrrolo[1,2-f]phenanthridines. The reaction of N-heterocycles, such as indoles, with 1,1-difluoroalkenes resulted in a twofold addition-elimination reaction to give 1,1-diaminoalkenes, which were transformed by CH-arylation into various polycondensated heterocycles, such as indoloisoquinolines, thienoindolizines, oxepines and helicenes. Pyridofluoranthenes, diindenopyrene and azadiindenopyrenes were prepared by a combination of Pd-catalyzed cross-coupling reactions with acid-mediated cycloisomerizations and Pd-catalyzed intramolecular CH-arylations. Bis(carbazoles), benzodithiazoles, benzodithiophenes and 2,5-diarylpyrroles were prepared by inter- and intramolecular CH-arylation reactions.

Pd-Catalyzed Suzuki-Miyaura Cross-Coupling Reaction in Glycerol Using KOH as Base and Glycerol-Triethanolamine Deep Eutectic Solvent under Inorganic Base‑Free Conditions

Glycerol and glycerol-triethanolamine deep eutectic solvent are both environmentally benign, cost-effective, and practical solvents for the Pd-catalyzed Suzuki-Miyaura cross-coupling of aryl halides with arylboronic acids. Utilizing KOH as base, the reaction in glycerol proceeded smoothly with low catalyst loadings (up to 0.5 mol% of PdCl2(PPh3)2) providing excellent yields (up to 99%) of the cross-coupling products, which can be readily extracted with hexane. However, the recyclability of the glycerol medium containing the catalyst is limited to a few cycles. Furthermore, it was explored the use of chlorodiphenylphosphine as a pre-ligand. Although glycerol phosphinite was not generated in situ, PPh2Cl proved to be an excellent pre-ligand, yielding the coupling product with a 98% yield. Compared to glycerol, glycerol-triethanolamine deep eutectic solvent (TEOA:G DES) proves to be a superior solvent for the cross-coupling reaction. In addition, the presence of amine group in the solvent allowed to obtain an inorganic base-free cross-coupling protocol. Aryl bromides and aryl boronic acids containing both electron releasing and attracting functional groups can be coupled without requiring the addition of any further base, affording the biphenyl products in moderate to good yields. The cross-coupling products can be easily isolated by extraction with hexane. Moreover, we observed the formation of triethanolamine phenylboronate during the reaction. The triethanolamine boronate was subsequently synthesized through the condensation of phenylboronic acid with triethanolamine and fully characterized. 11B nuclear magnetic resonance (NMR) spectroscopy confirmed the coordination of the nitrogen to the boron atom of the triethanolamine boronate.

Synthesis of Functionalized Hexahydrocarbazoles by Beckmann Elimination and Nucleophile-Intercepted Beckmann Fragmentation.

The Beckmann elimination and nucleophile-intercepted Beckmann fragmentation (NuBFr) of oximes starting from regioisomeric indolinyl bicyclic ketones lead to products that are subjected to further synthetic manipulations and ultimately result in the stereospecific formation of densely functionalized hexahydrocarbazoles. The Pd-catalyzed Suzuki-Miyaura cross-coupling reaction of a key alkenyl bromide intermediate with various boronic acids gives arylated products.

Elongation of N6-benzyladenosine scaffold via Pd-catalyzed C–C bond formation leads to derivatives with antiflaviviral activity

Decoration of nucleoside analogues with lipophilic groups often leads to compounds with improved antiviral activity. For example, N6-benzyladenosine derivatives containing elongated lipophilic substituents in the benzyl core efficiently inhibit reproduction of tick-borne encephalitis virus (TBEV), while N6-benzyladenosine itself potently inhibits reproduction of human enterovirus A71 (EV-A71). We have extended a series of N6-benzyladenosine analogues using effective synthetic methods of CC bond formation based on Pd-catalyzed cross-coupling reactions (Sonogashira and Suzuki) in order to study the influence of bulky lipophilic substituents in the N6 position of adenosine on the antiviral activity against flaviviruses, such as TBEV, yellow fever virus (YFV) and West Nile virus (WNV), as well as a panel of enteroviruses including EV-A71, Echovirus 30 (E30), and poliovirus type 2 (PV2). Reproduction of tested flaviviruses appeared to be inhibited by the micromolar concentrations of the compounds, while cytotoxicity in most cases was beyond the detection limit. Time-of-addition studies demonstrated that the hit compounds inhibited the stage of viral RNA synthesis, but not the stages of the viral entry or protein translation. As a result, several new promising antiflaviviral leads have been identified. On the other hand, none of the synthesized compounds inhibited enterovirus reproduction, indicating a possibility of involvement of flavivirus-specific pathways in their mechanism of action.

Catalytic stannane–fluorine activation triggered by Pd → Sn–F interaction

The chemistry of σ-acceptor (Z-type) ligands has been attracting significant interest for some time, and the geometric and electronic structures featuring Z-type ligands have provided a profound understanding of their distinctive chemical bonds, chemical properties, and reactivities. We herein report that the coordination of Lewis acidic fluorostannane as Z-type ligands enhances their nucleophilicities, opening new opportunities for the development of Pd-catalyzed cross-coupling reaction of fluorostannanes as coupling reagents. Thanks to phosphine chelation, fluorostannane serves as a Lewis acid (Z-type ligand) toward Pd atom and withdraws electron density from the Pd center via the σ*(Sn–F) orbital. Thus, the strong and polar Sn–F bond is efficiently weakened via σ-donation from the Pd center. Accordingly, the F atom in fluorostannane was readily abstracted by even moderate Lewis acidic boranes, leading to the conversion of Z-type stannane to X-stannyl complexes. This key conversion enabled the catalytic arylation of fluorostannane for the first time.

Induction of Chirality on Nanographenes.

Chirality induction is an important topic in studies of nanographenes (NGs). We report chirality enhancements of NGs through postsynthetic chemical modifications of NGs with pyrene and m-terphenyl groups. These substituents were installed into N-(p-bromophenylethyl)imides on the edges of the NGs with Pd-catalyzed cross-coupling reactions. Circular dichroism (CD) spectra demonstrated that these bulky substituents improved the induced CD signal of the NGs compared to those previously reported and suggested that they induced the opposite chirality. Density functional theory calculations indicated possible edge structures for the NGs and indicated that π/π and CH/π interactions among the neighboring substituents influenced the orientations of the imides. These imides distorted the edges, and the distorted edges eventually generated the chiral environments of the NGs. The interactions among the substituents are, therefore, likely to allow detection of the CD signals in the visible region and induction of the opposite chirality.

Important Role of NH-Carbazole in Aryl Amination Reactions Catalyzed by 2-Aminobiphenyl Palladacycles.

2-Aminobiphenyl palladacycles are among the most successful precatalysts for Pd-catalyzed cross-coupling reactions, including aryl amination. However, the role of NH-carbazole, a byproduct of precatalyst activation, remains poorly understood. Herein, the mechanism of the aryl amination reactions catalyzed by a cationic 2-aminobiphenyl palladacycle supported by a terphenyl phosphine ligand, PCyp2ArXyl2 (Cyp = cyclopentyl; ArXyl2 = 2,6-bis(2,6-dimethylphenyl)phenyl), P1, has been thoroughly investigated. Combining computational and experimental studies, we found that the Pd(II) oxidative addition intermediate reacts with NH-carbazole in the presence of the base (NaO t Bu) to yield a stable aryl carbazolyl Pd(II) complex. This species functions as the catalyst resting state, providing the amount of monoligated LPd(0) species required for catalysis and minimizing Pd decomposition. In the case of a reaction with aniline, an equilibrium between the carbazolyl complex and the on-cycle anilido analogue is established, which allows for a fast reaction at room temperature. In contrast, heating is required in a reaction with alkylamines, whose deprotonation involves coordination to the Pd center. A microkinetic model was built combining computational and experimental data to validate the mechanistic proposals. In conclusion, our study shows that despite the rate reduction observed in some reactions by the formation of the aryl carbazolyl Pd(II) complex, this species reduces catalyst decomposition and could be considered an alternative precatalyst in cross-coupling reactions.

Palladium-Catalyzed Synthesis, Acetylcholinesterase Inhibition, and Neuroprotective Activities of N-Aryl Galantamine Analogues.

A series of new N-aryl galantamine analogues (5a-5x) were designed and synthesized by modification of galantamine, using Pd-catalyzed Buchwald-Hartwig cross-coupling reaction in good to excellent yields. The cholinesterase inhibitory and neuroprotective activities of N-aryl derivatives of galantamine were evaluated. Among the synthesized compounds, the 4-methoxylpyridine-galantamine derivative (5q) (IC50 = 0.19 μM) exhibited excellent acetylcholinesterase inhibition activity, as well as significant neuroprotective effect against H2O2-induced injury in SH-SY5Y cells. Molecular docking, staining, and Western blotting analyses were performed to demonstrate the mechanism of action of 5q. Derivative 5q would be a promising multifunctional lead compound for the treatment of Alzheimer's disease.

Negishi or Suzuki–Miyaura Pd-Catalyzed Cross-Coupling Reaction: Which Reaction Mechanism is Ahead for the Formation of Well-Known Anticancer Drug Combretastatin A-4 Analogue?

In recent years, scientists have become increasingly interested in finding high-efficacy, low-toxicity anti-tumor compounds. Combretastatin A-4 (CA-4) is supposed to be one of the materials that has excellent anti-tumor properties. This study intends to develop a more acceptable pathway and cross-coupling mechanism to resolve the contest between Negishi and Suzuki–Miyaura cross-coupling to form a combretastatin analogue A-4 by using the CAM-B3LYP-D3 theory level with DEF2-SVP basis set in the presence of [Formula: see text],[Formula: see text]-dimethylformamide as a solvent. First, due to the experimental data for the formation of 4-methyl-[Formula: see text]-methoxybiphenyl in the Suzuki–Miyaura and Negishi reactions, two designated reactions were used to determine the 4-methyl-[Formula: see text]-methoxybiphenyl formation cycle. The mechanism for the progression of the regioselective compound 2-methoxy-5-(3-(3,4,5-trimethoxy phenyl) furan-2-yl) phenol via Suzuki–Miyaura and Negishi reactions can theoretically be reconciled with a more appropriate cross-coupling and pathway.

Synthesis and Properties of Diphenylbenzo[j]naphtho[2,1,8-def][2,7]phenanthrolines.

A series of hitherto unknown 5,14-diphenylbenzo[j]naphtho[2,1,8-def][2,7]phenanthrolines, containing a 5-azatetracene and a 2-azapyrene subunit, were prepared by combination of Pd-catalyzed cross-coupling reactions with a one-pot Povarov/cycloisomerization reaction. In the final key step four new bonds are formed in one step. The synthetic approach allows for a high degree of diversification of the heterocyclic core structure. The optical and electrochemical properties were studied experimentally and by DFT/TD-DFT and NICS calculations. Due to the presence of the 2-azapyrene subunit, the typical electronic nature and characteristics of the 5-azatetracene moiety are lost and the compounds are electronically and optically more related to 2-azapyrenes.

Bromine-Radical-Mediated Bromoallylation of C–C Unsaturated Bonds: A Facile Access to 1,4-, 1,5-, 1,6-, and 1,7-Dienes and Related Compounds

AbstractThe radical bromoallylation of alkynes, allenes, and vinylidene cyclopropanes proceeds efficiently in the presence of a radical initiator to give 2-bromo-substituted 1,4-, 1,5-, and 1,6-diene derivatives, respectively. Three-component reactions comprised of allenes, electron-deficient alkenes, and allyl bromides give 1,7-dienes in good yields. The bromoallylation of an arylalkene can override β-scission of the bromine radical from β-bromoalkyl radicals to give 5-bromoalkenes, whilst the bromoallylation of vinylcyclopropanes is accompanied by 5-exo ring closure to give 1-(bromomethyl)-2-vinylcyclopentane derivatives in good yields. Accessing spiropiperidines from dihydropyridones through tandem triflation-allylation and ring-closing metathesis (RCM).

A novel approach to build 2-spiropiperidine moieties starting from dihydropyridones was developed. The triflic anhydride-promoted conjugate addition of allyltributylstannane onto dihydropyridones allowed for the formation of gem bis-alkenyl intermediates that were converted to the corresponding spirocarbocycles with excellent yields via ring closing metathesis. The vinyl triflate group generated on these 2-spiro-dihydropyridine intermediates could be successfully used as a chemical expansion vector for further transformations namely Pd-catalyzed cross-coupling reactions.

Application of hindered ether solvents for palladium catalyzed Suzuki-Miyaura, Sonogashira and cascade Sonogashira cross-coupling reactions.

Cross-coupling and cascade reactions typically rely on unsustainable and toxic volatile organic solvents. 2,2,5,5-Tetramethyloxolane (TMO) and 2,5-diethyl-2,5-dimethyloxolane (DEDMO) are both inherently non-peroxide forming ethers, and have been used in this work as effective, more sustainable, and potentially bio-based alternative solvents for Suzuki-Miyaura and Sonogashira reactions. Suzuki-Miyaura reactions demonstrated good yields for a range of substrates, 71-89% in TMO and 63-92% in DEDMO. In addition, a Sonogashira reaction exhibited the excellent yields of 85-99% performed in TMO, which was significantly higher than traditional volatile organic solvents, THF or toluene, and higher than those reported for another non-peroxide forming ether, namely eucalyptol. Cascade Sonogashira reactions utilizing a simple annulation methodology were particularly effective in TMO. Furthermore, a green metric assessment confirmed that the methodology employing TMO was more sustainable and greener than the traditional solvents THF and toluene, thereby demonstrating the promise of TMO as an alternative solvent for Pd-catalyzed cross-coupling reactions.

A general approach to stereospecific Pd-catalyzed cross-coupling reactions of benzylic stereocenters.

We have developed a general process for the formation of enantioenriched benzylic stereocenters via stereospecific Pd-catalyzed cross-coupling reactions of enantioenriched benzylic tricyclohexyltin nucleophiles. This process proceeds with excellent stereospecificity for a remarkably broad scope of electrophilic coupling partners including aryl and heteroaryl halides and triflates, acid chlorides, thioesters, chloroformates, and carbamoyl chlorides. Thus, enantioenriched 1,1-diarylalkanes as well as formal products of asymmetric enolate arylation are readily accessed using this approach. We additionally provide the first demonstration of a Sn-selective cross-coupling reaction using a vicinal alkylborylstannane nucleophile. In these reactions, the presence of cyclohexyl spectator ligands on tin is essential to ensure selective transfer of the secondary benzylic unit from tin to palladium.

Functionalized Chromans from ortho-Quinone Methides and Arylallenes.

ortho-Quinone methides (o-QMs) underwent formal [4 + 2]-cycloaddition reactions with arylallenes regioselectively at the styrenyl olefin to furnish the corresponding 3-methylene-2-arylchromans in moderate to good yields (up to 88%). When R ≠ H, the reactions also proceeded with moderate stereoselectivity (up to 5:1) which was governed by the nature of the R group. The 3-methylene-2-arylchromans could serve as common intermediates for further functionalization including epoxidation, oxidative cleavage/Baeyer-Villiger oxidation, Riley oxidation, acid-catalyzed rearrangement, and Pd-catalyzed cross-coupling reactions to furnish the corresponding derivatives in moderate to good yields.
