Behavioural studies on crude Ethanol leaf extract of Cadaba farinosa Forssk. in mice

Cadaba farinosa Forssk. (Capparaceae) is found all over the world, mostly in tropical and subtropical areas. It is used to treat pain, dysentery, rheumatism, cough, and fever, as well as an antidote and in the treatment of neurological disorders. In mice, the median lethal dose (LD50) of Crude Ethanol leaf Extract (CEE) of Cadaba farinosa was found to be 2154.1 mg/kg body weight. The presence of alkaloids, anthraquinones, carbohydrates, cardiac glycosides, flavonoids, glycosides, saponins, and tannins was discovered during a preliminary phytochemical analysis of the dark gummy mass crude extract. The CEE was then put through pharmacological testing on mice. In mice, diazepam-induced sleep, beam walking, and hole-board assays were used to investigate behavioral activities. CEE of Cadaba farinosa had no effect on the onset of sleep in mice when given at the tested doses of diazepam. In contrast, it significantly increased sleep duration at all doses tested (75 mg/kg p 0.001, 150 mg/kg p≤0.05, and 300 mg/kg p≤0.01). The CEE significantly (p≤0.01) increased the number of foot slips in the Beam walking assay at a dose of 150 mg/kg. Similarly, diazepam at 0.25 mg/kg increased the number of foot slips significantly (p≤0.001). The extract at a dose of 300 mg/kg and diazepam increased the time required to complete the task significantly (p≤0.05). At 150 mg/kg (p 0.01) and 300 mg/kg (p≤0.05), the hole-board experiment significantly reduced the number of head dips. In contrast, the standard drug (diazepam) had no discernible effect on the number of head dips. These findings suggest that CEE and Diazepam at the highest dose significantly affected the behaviour of mice in the beam walking assay. Thus, the extract showed no activity on the onset of sleep but significantly prolong duration of sleep at the tested doses. The CEE significantly produced a decrease in the exploratory behavioural pattern, indicative of the fact that CEE possesses sedative property.

Automated telemetry reveals post-reintroduction exploratory behavior and movement patterns of an endangered corvid, ʻAlalā (Corvus hawaiiensis) in Hawaiʻi, USA

Continuous movement monitoring is a powerful tool for evaluating reintroduction techniques and assessing how well reintroduced animals are adjusting to the wild. However, to date, continuous monitoring has only occurred for large-bodied species capable of carrying heavy tracking devices. In this study we used an automated VHF radio telemetry array to investigate the exploratory behavior and movement patterns of critically endangered ʻAlalā (Corvus hawaiiensis), reintroduced to the Island of Hawaiʻi in 2017. The 11 juvenile ʻAlalā we tracked exhibited high site fidelity and initial survival. Over time the birds showed decreased time spent at the supplemental feeders, and transitioned to more focused use of the landscape, suggesting increased foraging on wild food items. Birds with seemingly less spatial neophobia at release also made larger post-release exploratory movements. This study provides the first evidence that 1) supplemental feeding can support site fidelity for reintroduced ʻAlalā without restricting a transition to independent foraging, and 2) that pre-release personality metrics may be useful predictors for predicting post-release movements of ʻAlalā. Our work is the first to demonstrate the utility and power of automated telemetry for monitoring the reintroduction of small species.

The effect of playback of 22-kHz and 50-kHz ultrasonic vocalizations on rat behaviors assessed with a modified open-field test

Juvenile and adult rats emit two affectively different types of ultrasonic vocalizations (USVs), namely aversive 22-kHz and appetitive 50-kHz USVs. Aversive 22-kHz USVs are considered to be alarm calls that communicate negative affective states to conspecific receivers. Although the alarming effects of playback of 22-kHz USVs were reported recently, behavioral data showing those effects are still not abundant. Appetitive 50-kHz USVs are considered to communicate positive affective states to conspecific receivers, to pace and coordinate social behavior. In line with this, playback of 50-kHz USVs has been found to initiate behavioral activation and induce approach behavior in receiver rats. However, most of these playback studies have used male 50-kHz USVs; thus, it seems to remain unclear whether female 50-kHz USVs exert a similar social attractant effect on male rats. To investigate these issues, we performed modified open-field tests, during which USVs were continuously presented for 15 min to male receivers. In these tests, if negative affective changes are evoked in subject rats, the time spent in the open arena decreases, while the time spent on defensive behaviors increases. In contrast, when positive affective changes are evoked, the opposite phenomenon is observed. Playback of male aversive 22-kHz USVs induced anxiety-related defensive responses in receivers. However, playback of female appetitive frequency-modulated (FM) 50-kHz USVs increased opposite, appetitive pattern of exploratory behavior with increased exploration. The results indicate that playback of male aversive 22-kHz and female appetitive 50-kHz USVs might induce behavioral responses probably associated with negative and positive affective states in male rats, respectively, suggesting the validity of rat USVs as an animal model of vocal communication of emotion.

Deficits in the vesicular acetylcholine transporter alter lifespan and behavior in adult Drosophila melanogaster

The neurotransmitter acetylcholine (ACh) is involved in critical organismal functions that include locomotion and cognition. Importantly, alterations in the cholinergic system are a key underlying factor in cognitive defects associated with aging. One essential component of cholinergic synaptic transmission is the vesicular ACh transporter (VAChT), which regulates the packaging of ACh into synaptic vesicles for extracellular release. Mutations that cause a reduction in either protein level or activity lead to diminished locomotion ability whereas complete loss of function of VAChT is lethal. While much is known about the function of VAChT, the direct role of altered ACh release and its association with either an impairment or an enhancement of cognitive function are still not fully understood. We hypothesize that point mutations in Vacht cause age-related deficits in cholinergic-mediated behaviors such as locomotion, and learning and memory. Using Drosophila melanogaster as a model system, we have studied several mutations within Vacht and observed their effect on survivability and locomotive behavior. Here we report for the first time a weak hypomorphic Vacht allele that shows a differential effect on ACh-linked behaviors. We also demonstrate that partially rescued Vacht point mutations cause an allele-dependent deficit in lifespan and defects in locomotion ability. Moreover, using a thorough data analytics strategy to identify exploratory behavioral patterns, we introduce new paradigms for measuring locomotion-related activities that could not be revealed or detected by a simple measure of the average speed alone. Together, our data indicate a role for VAChT in the maintenance of longevity and locomotion abilities in Drosophila and we provide additional measurements of locomotion that can be useful in determining subtle changes in Vacht function on locomotion-related behaviors.

Ceftriaxone Attenuated Anxiety-Like Behavior and Enhanced Brain Glutamate Transport in Zebrafish Subjected to Alcohol Withdrawal.

Chronic and/or excessive consumption of alcohol followed by reduced consumption or abstention can result in Alcohol Withdrawal Syndrome. A number of behavioral changes and neurological damage result from ethanol (EtOH) withdrawal. Ceftriaxone (Cef) modulates the activity of excitatory amino acid transporters by increasing their gene expression. Zebrafish are commonly used to study alcohol exposure. The aim of this study was to evaluate the influence of Cef (100 µM) on behavior patterns, glutamate transport activity, and oxidative stress in zebrafish brains subjected to EtOH (0.3% v/v) withdrawal. The exploratory tests using Novel tank showed that EtOH withdrawal promoted a decrease in the time spent and number of entries of in the bottom displaying an anxiety-like behavior. In contrast, treatment with Cef resulted in recovery of exploratory behavioral patterns. Ceftriaxone treatment resulted in increased glutamate uptake in zebrafish subjected to EtOH withdrawal. Furthermore, EtOH withdrawal increased reactive species, as determined using thiobarbituric acid and dichlorodihydrofluorescein assays. Treatment with Cef reversed these effects. Ceftriaxone promoted a significant reduction in brain sulfhydryl content in zebrafish subjected to EtOH withdrawal. Therefore, Cef treatment in conjunction with EtOH withdrawal induced anxiolytic-like effects due to possible neuromodulation of glutamatergic transporters, potentially through mitigation of oxidative stress.

Read-the-game: System for skill-based visual exploratory activity assessment with a full body virtual reality soccer simulation

We present a novel virtual reality (VR) system to measure soccer players’ read-the-game ability. Read-the-game is a term that encompasses a conglomerate of visual exploratory behavioral patterns and cognitive elements required to make accurate in-game decisions. Our technological approach in the Sports Science domain focuses on the visuomotor component of targeted skill development in a VR simulation because VR is a powerful perception–action coupling training solution for visuomotor coordination due to its high sense of immersion and its psychological byproduct presence. Additionally, we analyze two critical aspects: psychological (i.e., sense of presence) and the human–computer interaction (HCI) domain (i.e., suitable input device for full-body immersion). To measure head movements related to visual explorations, the system tracks the user’s head excursions. Specifically, the engaged visual exploratory activity (VEA) during a VR simulation is measured frame-by-frame at runtime to study the behavior of players making passing decisions while experiencing pressure from rivals during in-game situations recreated with computer graphics (CG). Additionally, the sense of presence elicited by our system is measured via the Igroup Presence Questionnaire applied to beginner and amateur soccer players (n = 24). Regarding the HCI aspect, a comparison of input options reveals that a high presence can be achieved when using full body interactions that integrate head and body motions via a combination of an HMD and kinetic body tracking. During our system verification, a difference in the VEA performance is observed between beginner and amateur players. Moreover, we demonstrate the capacity of the system to measure the VEA while evoking immersive soccer in-match experiences with a portable VR setup.

A Dietary Ketone Ester Normalizes Abnormal Behavior in a Mouse Model of Alzheimer's Disease.

Because of a decreased sensitivity toward insulin, a key regulator of pyruvate dehydrogenase (PDH), Alzheimer’s patients have lower brain glucose utilization with reductions in Tricarboxylic Acid (TCA) cycle metabolites such as citrate, a precursor to n-acetyl-aspartate. In the 3xTgAd mouse model of Alzheimer’s disease (AD), aging mice also demonstrate low brain glucose metabolism. Ketone metabolism can overcome PDH inhibition and restore TCA cycle metabolites, thereby enhancing amino acid biosynthesis. A ketone ester of d-β-hydroxybutyrate was incorporated into a diet (Ket) and fed to 3xTgAd mice. A control group was fed a calorically matched diet (Cho). At 15 months of age, the exploratory and avoidance-related behavior patterns of the mice were evaluated. At 16.5 months of age, the animals were euthanized, and their hippocampi were analyzed for citrate, α-ketoglutarate, and amino acids. In the hippocampi of the Ket-fed mice, there were higher concentrations of citrate and α-ketoglutarate as well as higher concentrations of glutamate, aspartate and n-acetyl-aspartate compared with controls. There were positive associations between (1) concentrations of aspartate and n-acetyl-aspartate (n = 14, R = 0.9327), and (2) α-ketoglutarate and glutamate (n = 14, R = 0.8521) in animals maintained on either diet. Hippocampal n-acetyl-aspartate predicted the outcome of several exploratory and avoidance-related behaviors. Ketosis restored citrate and α-ketoglutarate in the hippocampi of aging mice. Higher concentrations of n-acetyl-aspartate corresponded with greater exploratory activity and reduced avoidance-related behavior.

Anxiolytic and mnemotropic effects of herbal extracts in an experiment on laboratory animals

We research memory, exploratory behavior and anxiety in adult rats in an experiment. These parameters were assessed through the use of behavioral assays, namely, the Suok test and passive avoidance learning. These parameters were studied after treatment with extracts of Ginkgo Biloba and salvia. The aqueous extract of salvia and Ginkgo-Biloba contributed to the activation of patterns of exploratory behavior and a decrease of anxiety in animals in the Suok-test. The Ginkgo-Biloba extract stimulates cognitive functions in rats, which is reflected in an improvement in the elaboration of the conditioned reflex and the preservation of the memorable trace after training in the passive avoidance learning.

Neural Coding of Whisker-Mediated Touch in Primary Somatosensory Cortex Is Altered Following Early Blindness.

Sensory systems do not develop and function independently of one another, yet they are typically studied in isolation. Effects of multisensory interactions on the developing neocortex can be revealed by altering the ratios of incoming sensory inputs associated with different modalities. We investigated neural responses in primary somatosensory cortex (S1) of short-tailed opossums (Monodelphis domestica; either sex) after the elimination of visual input through bilateral enucleation very early in development. To assess the influence of tactile experience after vision loss, we also examined naturally occurring patterns of exploratory behavior. In early blind (EB) animals, overall levels of tactile experience were similar to those of sighted controls (SC); locomotor activity was unimpaired and accompanied by whisking. Using extracellular single-unit recording techniques under anesthesia, we found that EB animals exhibited a reduction in the magnitude of neural responses to whisker stimuli in S1, coupled with spatial sharpening of receptive fields, in comparison to SC animals. These alterations manifested as two different effects on sensory processing in S1 of EB animals: the ability of neurons to detect single whisker stimulation was decreased, whereas their ability to discriminate between stimulation of neighboring whiskers was enhanced. The increased selectivity of S1 neurons in EB animals was reflected in improved population decoding performance for whisker stimulus position, particularly along the rostrocaudal axis of the snout, which aligns with the primary axis of natural whisker motion. These findings suggest that a functionally distinct form of somatosensory plasticity occurs when vision is lost early in development.SIGNIFICANCE STATEMENT After sensory loss, compensatory behavior mediated through the spared senses could be generated entirely through the recruitment of brain areas associated with the deprived sense. Alternatively, functional compensation in spared modalities may be achieved through a combination of plasticity in brain areas corresponding to both spared and deprived sensory modalities. Although activation of neurons in cortex associated with a deprived sense has been described frequently, it is unclear whether this is the only substrate available for compensation or if plasticity within cortical fields corresponding to spared modalities, particularly primary sensory cortices, may also contribute. Here, we demonstrate empirically that early loss of vision alters coding of sensory inputs in primary somatosensory cortex in a manner that supports enhanced tactile discrimination.

The Sensitivity of the Crayfish Reward System to Mammalian Drugs of Abuse.

The idea that addiction occurs when the brain is not able to differentiate whether specific reward circuits were triggered by adaptive natural rewards or falsely activated by addictive drugs exist in several models of drug addiction. The suitability of crayfish (Orconectes rusticus) for drug addiction research arises from developmental variation of growth, life span, reproduction, behavior and some quantitative traits, especially among isogenic mates reared in the same environment. This broad spectrum of traits makes it easier to analyze the effect of mammalian drugs of abuse in shaping behavioral phenotype. Moreover, the broad behavioral repertoire allows the investigation of self-reinforcing circuitries involving appetitive and exploratory motor behavior, while the step-wise alteration of the phenotype by metamorphosis allows accurate longitudinal analysis of different behavioral states. This paper reviews a series of recent experimental findings that evidence the suitability of crayfish as an invertebrate model system for the study of drug addiction. Results from these studies reveal that unconditioned exposure to mammalian drugs of abuse produces a variety of stereotyped behaviors. Moreover, if presented in the context of novelty, drugs directly stimulate exploration and appetitive motor patterns along with molecular processes for drug conditioned reward. Findings from these studies indicate the existence of drug sensitive circuitry in crayfish that facilitates exploratory behavior and appetitive motor patterns via increased incentive salience of environmental stimuli or by increasing exploratory motor patterns. This work demonstrates the potential of crayfish as a model system for research into the neural mechanisms of addiction, by contributing an evolutionary, comparative context to our understanding of natural reward as an important life-sustaining process.

Assessing Mongolian gerbil emotional behavior: effects of two shock intensities and response-independent shocks during an extended inhibitory-avoidance task.

Despite step-down inhibitory avoidance procedures that have been widely implemented in rats and mice to study learning and emotion phenomena, performance of other species in these tasks has received less attention. The case of the Mongolian gerbil is of relevance considering the discrepancies in the parameters of the step-down protocols implemented, especially the wide range of foot-shock intensities (i.e., 0.4–4.0 mA), and the lack of information on long-term performance, extinction effects, and behavioral patterning during these tasks. Experiment 1 aimed to (a) characterize gerbils’ acquisition, extinction, and steady-state performance during a multisession (i.e., extended) step-down protocol adapted for implementation in a commercially-available behavioral package (Video Fear Conditioning System—MED Associates Fairfax, VT, USA), and (b) compare gerbils’ performance in this task with two shock intensities – 0.5 vs. 1.0 mA—considered in the low-to-mid range. Results indicated that the 1.0 mA protocol produced more reliable and clear evidence of avoidance learning, extinction, and reacquisition in terms of increments in freezing and on-platform time as well as suppression of platform descent. Experiment 2 aimed to (a) assess whether an alternate protocol consisting of a random delivery of foot shocks could replicate the effects of Experiment 1 and (b) characterize gerbils’ exploratory behavior during the step-down task (jumping, digging, rearing, and probing). Random shocks did not reproduce the effects observed with the first protocol. The data also indicated that a change from random to response-dependent shocks affects (a) the length of each visit to the platform, but not the frequency of platform descends or freezing time, and (b) the patterns of exploratory behavior, namely, suppression of digging and rearing, as well as increments in probing and jumping. Overall, the study demonstrated the feasibility of the extended step-down protocol for studying steady performance, extinction, and reacquisition of avoidance behavior in gerbils, which could be easily implemented in a commercially available system. The observation that 1.0 mA shocks produced a clear and consistent avoidance behavior suggests that implementation of higher intensities is unnecessary for reproducing aversive-conditioning effects in this species. The observed patterning of freezing, platform descents, and exploratory responses produced by the change from random to periodic shocks may relate to the active defensive system of the gerbil. Of special interest is the probing behavior, which could be interpreted as risk assessment and has not been reported in other rodent species exposed to step-down and similar tasks.

Acute stress disrupts temporal patterns of behavioral and biochemical parameters of rats

This study aimed to answer these questions: (i) Can the temporal patterns of locomotor and exploratory activities and anxiety-like behavior be disrupted by a single session of stress? (ii) Can the temporal patterns of corticosterone, melatonin, and glucose be disrupted by a single session of stress and how long does the effect of stress upon their circadian rhythm? Adult male Wistar rats were exposed to restrain stress at four Zeitgeber Times and subdivided according to evaluation times of behavioral (open field test and elevated plus-maze test) and biochemistry (corticosterone, melatonin, and glucose levels in serum) parameters. There were temporal patterns within a course of 24 h in anxiety-like behaviors and glucose levels. Circadian rhythm to corticosterone and melatonin levels was confirmed and a single session of stress can disrupt these temporal patterns. We showed an immediate disruptive effect of stress in biomarkers levels and a delayed appearance of behavioral and locomotion pattern disruption.

The defensive behavioral patterns of captive white-lipped and collared peccary (Mammalia, Tayassuidae): an approach for conservation of the species

Defensive behavioral patterns in response to human-induced rapid environmental change can affect animals’ fitness and may play a role in species conservation status. To test this hypothesis, we compared the risk assessment and defensive behavioral responses of captive white-lipped peccary (WLP; Tayassu pecari) and collared peccary (CP; Pecari tajacu), which retain different conservation status; WLP are considered vulnerable and CP of least concern. We used an adapted paradigm of the mouse defense test battery (MDTB) comprising four consecutive tests. Two of these tests simulated a novel environment, while the other two stimulated the expression of defensive behavioral patterns. Besides differences in risk assessment and defensive threat/attack behavioral patterns between species, we compared flight initiation distance, flight speed, and plasma glucocorticoid concentrations. When facing a novel environment and risk challenges from humans’ predator-like cues, the white-lipped peccary showed more exploratory and defensive threat/attack behavioral patterns, shorter flight initiation distances, and lower flight speeds, whereas the collared peccaries showed more cautious and retreat patterns, longer flight initiation distances, and higher flight speeds. There were also correlations between physiological and behavioral parameters. We confirmed our hypothesis that the collared peccary’s cautiousness may help to prevent a decrease in its population, while the white-lipped peccary’s exploratory and confrontational behavioral patterns in overhunted areas, together with other simultaneous factors as forest fragmentation, might contribute to placing this species in the vulnerable category.

Visualising and examining sequential actions as behavioural paths that can be interpreted as markers of complex behaviours

Visualisation of pathways taken by students through problem solving environments provides the potential to identify patterns of exploratory behaviour that may relate to different levels of proficiency or different cognitive approaches. This is particularly the case when problems are situated in complex online environments and are programmed to generate large scale data for analysis.Using process data from an online collaborative problem solving task, we visualised behavioural paths of 607 pairs of students as directed graphs. The empirical paths were then examined using exploratory network analysis based on four main aspects of exploration (prominence, branches, clusters, and shortest paths).The primary purpose of such analysis is to detect sequences that are potentially relevant for establishing particular paths as meaningful markers of complex behaviours. This visualisation approach enabled us to capture all actual (as opposed to possible) pathways from the data. Although there may be an optimal pathway through a complex task according to criteria such as efficiency or correct solution, the added human factors bring a dynamic to the activity that provides a richer environment for understanding students’ collaborative processes, both cognitive and social. The method adopted in this study provides a prototype for exploration of other complex skillsets.

Amphetamine increases activity but not exploration in humans and mice.

Cross-species quantification of physiological behavior enables a better understanding of the biological systems underlying neuropsychiatric diseases such as bipolar disorder (BD). Cardinal symptoms of manic BD include increased motor activity and goal-directed behavior, thought to be related to increased catecholamine activity, potentially selective to dopamine homeostatic dysregulation. The objective of this study was to test whether acute administration of amphetamine, a norepinephrine/dopamine transporter inhibitor and dopamine releaser, would replicate the profile of activity and exploration observed in both humans with manic BD and mouse models of mania. Healthy volunteers with no psychiatric history were randomized to a one-time dose of placebo (n = 25), 10mg d-amphetamine (n = 18), or 20mg amphetamine (n = 23). Eighty mice were administered one of four doses of d-amphetamine or vehicle. Humans and mice were tested in the behavioral pattern monitor (BPM), which quantifies motor activity, exploratory behavior, and spatial patterns of behavior. In humans, the 20-mg dose of amphetamine increased motor activity as measured by acceleration without marked effects on exploration or spatial patterns of activity. In mice, amphetamine increased activity, decreased specific exploration, and caused straighter, one-dimensional movements in a dose-dependent manner. Consistent with mice, amphetamine increased motoric activity in humans without increasing exploration. Given that BD patients exhibit heightened exploration, these data further emphasize the limitation of amphetamine-induced hyperactivity as a suitable model for BD. Further, these studies highlight the utility of cross-species physiological paradigms in validating biological mechanisms of psychiatric diseases.

Modeling strategic use of human computer interfaces with novel hidden Markov models.

Immersive software tools are virtual environments designed to give their users an augmented view of real-world data and ways of manipulating that data. As virtual environments, every action users make while interacting with these tools can be carefully logged, as can the state of the software and the information it presents to the user, giving these actions context. This data provides a high-resolution lens through which dynamic cognitive and behavioral processes can be viewed. In this report, we describe new methods for the analysis and interpretation of such data, utilizing a novel implementation of the Beta Process Hidden Markov Model (BP-HMM) for analysis of software activity logs. We further report the results of a preliminary study designed to establish the validity of our modeling approach. A group of 20 participants were asked to play a simple computer game, instrumented to log every interaction with the interface. Participants had no previous experience with the game's functionality or rules, so the activity logs collected during their naïve interactions capture patterns of exploratory behavior and skill acquisition as they attempted to learn the rules of the game. Pre- and post-task questionnaires probed for self-reported styles of problem solving, as well as task engagement, difficulty, and workload. We jointly modeled the activity log sequences collected from all participants using the BP-HMM approach, identifying a global library of activity patterns representative of the collective behavior of all the participants. Analyses show systematic relationships between both pre- and post-task questionnaires, self-reported approaches to analytic problem solving, and metrics extracted from the BP-HMM decomposition. Overall, we find that this novel approach to decomposing unstructured behavioral data within software environments provides a sensible means for understanding how users learn to integrate software functionality for strategic task pursuit.

Lévy flights and self-similar exploratory behaviour of termite workers: beyond model fitting.

Animal movements have been related to optimal foraging strategies where self-similar trajectories are central. Most of the experimental studies done so far have focused mainly on fitting statistical models to data in order to test for movement patterns described by power-laws. Here we show by analyzing over half a million movement displacements that isolated termite workers actually exhibit a range of very interesting dynamical properties –including Lévy flights– in their exploratory behaviour. Going beyond the current trend of statistical model fitting alone, our study analyses anomalous diffusion and structure functions to estimate values of the scaling exponents describing displacement statistics. We evince the fractal nature of the movement patterns and show how the scaling exponents describing termite space exploration intriguingly comply with mathematical relations found in the physics of transport phenomena. By doing this, we rescue a rich variety of physical and biological phenomenology that can be potentially important and meaningful for the study of complex animal behavior and, in particular, for the study of how patterns of exploratory behaviour of individual social insects may impact not only their feeding demands but also nestmate encounter patterns and, hence, their dynamics at the social scale.

Violated expectancies: Cause and function of exploration, fear, and aggression

To be able to reproduce, animals need to survive and interact with an ever changing environment. Therefore, they create a cognitive representation of that environment, from which they derive expectancies regarding current and future events. These expected events are compared continuously with information gathered through exploration, to guide behaviour and update the existing representation. When a moderate discrepancy between perceived and expected events is detected, exploration is employed to update the internal representation so as to alter the expectancy and make it match the perceived event. When the discrepancy is relatively large, exploration is inhibited, and animals will try to alter the perceived event utilizing aggression or fear. The largest discrepancies are associated with a tendency to flee. When an exploratory, fear, or aggressive behaviour pattern proofs to be the optimal solution for a particular discrepancy, the response will become conditioned to events that previously preceded the occurrence of that discrepancy. When primary needs are relatively low, animals will actively look for or create moderately violated expectancies in order to learn about objects, behaviour patterns, and the environment. In those situations, exploratory tendencies will summate with ongoing behaviour and, when all primary needs are satiated, may even be performed exclusively. Exploratory behavior and withdrawal signs in Crayfish: Chronic central morphine injections and termination effects

Functional and evolutionary conservation of neural circuits of reward seeking >is a symbol of survival. It is found in most animals from insects to humans. Exploration is a component of a wide range of drug-elicited behaviors that reflects an appetitive motivational state when animals seek natural rewards such as food, water, and shelter for survival. Not only does the characterization of exploratory behaviors indicate the specific components of appetitive motor patterns, it also reveals how exploratory behavioral patterns are implemented via increased incentive salience of environmental stimuli. The current work demonstrates that novel stimuli appear to directly augment exploration in crayfish, while injections of morphine directly into the brain of crayfish enhanced robust arousal resulting in increased locomotion and exploration of the environment. Elimination of morphine suppressed exploratory motor patterns. Crayfish displayed atypical behavioral changes evident of withdrawal-like states when saline is injected into the brain. With proven evidence of rewarding to the exposure to mammalian drugs of abuse, modularly organized and experimentally accessible nervous system makes crayfish exceptionally suitable for characterizing the central workings of addiction at its key behavioral and neuroanatomic locations.

  The purpose of this study is to investigate the central nervous system (CNS) depressant and analgesic activities of the ethanol extract of Scutia myrtina (EESM) (Family: Rhamnaceae) in Swiss albino mice. To evaluate the CNS depressant activity by using the methods such as general behavior, exploratory behavior, muscle relaxant activity and phenobarbitone sodium-induced sleeping time were studied. Analgesic effect of EESM was evaluated in acetic acid induced writhing and hotplate tests. The results revealed that the EESM at the dose of 200 and 300 mg/kg caused a significant reduction in the spontaneous activity (general behavioral profile), remarkable decrease in exploratory behavioral pattern (Y-maze and head dip test), a reduction in muscle relaxant activity (rotarod and traction tests), and also significantly potentiated phenobarbitone sodium-induced sleeping time. The EESM also produced significant analgesic activity in both models at the dose of 200 and 300 mg/kg. Further, the EESM potentiated the morphine and aspirin induced analgesic in mice. The results suggest that EESM exhibit CNS depressant and analgesic activity in tested animal models. Key words: Scutia myrtina, central nervous system (CNS) activity, analgesic, mice, ethanol extract.
