Dry tobacco leaves: an in vivo and in silico approach to the consequences of occupational exposure.

Exposure of tobacco workers handling dried tobacco leaves has been linked to an increased risk of toxicity and respiratory illness due to the presence of nicotine and other chemicals. This study aimed to evaluate the DNA damage caused by the exposure of tobacco growers during the dry leaf classification process and the relation to cellular mechanisms. A total of 86 individuals participated in the study, divided into a group exposed to dry tobacco (n = 44) and a control group (n = 42). Genotoxicity was evaluated using the alkaline comet assay and lymphocyte micronucleus (MN) assay (CBMN-Cyt), and measurement of telomere length. The levels of oxidative and nitrosative stress were evaluated through the formation of thiobarbituric acid reactive species, and nitric oxide levels, respectively. The inorganic elements were measured in the samples using particle-induced X-ray emission method. The combination of variables was demonstrated through principal component analysis and the interactions were expanded through systems biology. Comet assay, MN, death cells, thiobarbituric acid reactive species, and nitrosative stress showed a significant increase for all exposed groups in relation to the control. Telomere length showed a significant decrease for exposed women and total exposed group in relation to men and control groups, respectively. Bromine (Br) and rubidium (Rb) in the exposed group presented higher levels than control groups. Correlations between nitrate and apoptosis; Br and MN and necrosis; and Rb and telomeres; besides age and DNA damage and death cells were observed. The systems biology analysis demonstrated that tobacco elements can increase the nuclear translocation of NFKB dimers inducing HDAC2 expression, which, associated with BRCA1 protein, can potentially repress transcription of genes that promote DNA repair. Dry tobacco workers exposed to dry leaves and their different agents showed DNA damage by different mechanisms, including redox imbalance.

Impact of dry deposition flux and dust intrusion phenomena on elemental concentrations of air particulate matter

Depositions of trace elements of air particulates in the atmosphere have gained much attention in recent times due to their environmental, climatic and health impacts. In the current study, trace elements of size-segregated rural and urban particulate matter (PM2.5 and PM2.5−10) were characterized using particle-induced X-ray emission method. Effects of dry deposition flux and Saharan dust intrusion on the elemental concentrations were determined. The mean elemental concentration in the rural fine PM ranged between 0.0002 ± 0.0002 μg m−3 (for Se) and 0.6658 ± 0.5816 μg m−3 (for S) whereas in the urban fine PM the concentration ranged between 0.0007 ± 0.0008 μg m−3 (for As) and 0.5869 ± 0.3429 μg m−3 (for Ca). In the coarse PM fraction, concentration ranged between 0.0001 ± 0.0001 μg m−3 (for Se) and 0.5886 ± 0.5343 μg m−3 (for Ca) at the rural site; and between 0.0002 ± 0.0002 μg m−3 (for Se) and 1.9201 ± 1.911 μg m−3 (for Ca) at the urban site. Mainly crustal elements (Al, Ca, Fe) and ‘pollution’ source element (S) recorded high and significant dry deposition fluxes across the two sites in both PM modes. Deposition flux was relatively higher in the coarse than in the fine PM mode. The average dry deposition flux estimated for fine and coarse PM at the rural and urban sites were 33.68 ± 57.19 μg m−2 day−1 and 47.71 ± 92.19 μg m−2 day−1; 36.66 ± 61.95 μg m−2 day−1 and 107.02 ± 237.71 μg m−2 day−1, respectively. Mainly crustal (Al, Ca, Fe) and “pollution” (S) element recorded high and significant dry deposition fluxes across the two sites and in the two PM modes. The impacts of dust on climate, air quality, and the environment in general have increased the need to better understand the dust flow system. In the current study, dust intruding the study area from the Sahara was analyzed using the NMMB/BSC dust model. The model’s output indicated that, the highest daily surface dust concentration was observed in the central and south-west of Turkey which corresponded with the peak period of Saharan dust transport to the Eastern Mediterranean in the month of March.

Sub-cellular elemental imaging of human muscle tissues affected by neuromuscular diseases

Abstract Various types of neuromuscular diseases differ in symptoms, pathology, and clinical picture but one of their common elements is muscle weakness, which could lead to human motor activities impairment and in many cases to shortening of life span and even death due to respiratory failure. That is why it is very important to better understand the underlying causes of these diseases to be able to implement new methods of treatment more effectively. This paper presents the results of the elemental analysis of human muscular tissues affected by dystrophy and myopathy. For this purpose, the particle-induced X-ray emission method was used, which is perfectly suited for measuring light elements. The samples were analysed for differences in the elemental composition of Na, Mg, P, S, Cl, K, Fe, Zn, and Br. The results were presented in the form of elemental concentration maps and a thorough statistical analysis of the obtained data using the advanced statistical methods.

Investigation on major, minor and trace elements in some medicinal plants using Particle Induced X-ray Emission

Elemental concentrations namely P, S, Cl, K, Ca, Sc, Ti, Mn, Fe, Cu, Zn, Rb, Br and Sr were determined using Particle Induced X-ray Emission method in leaves and flowers of medicinal plants namely flowers of Hibiscus rosa-sinensis, leaves and flowers of Annona reticulata, leaves of holy basil and leaves of Psidium guajava. No toxic elements were detected in these samples. The method was validated by analyzing the reference materials namely IAEA RM V-10 and NIST SRM 1632a. The main objective of this study is to provide a good source of elemental concentration data for Ayurvedic medicine dosage.

Sequential changes in serum zinc concentrations in calves with experimentally induced endotoxin shock measured by the particle-induced X-ray emission method.

The aim of the present study was to measure changes in the serum concentrations of some elements in endotoxin-challenged calves using a particle-induced X-ray emission analysis and to screen for elements useful as diagnostic markers. The results obtained revealed that serum Zn concentrations were more accurate diagnostic markers for detecting endotoxin shock in calves than other elements. Serum Zn level in endotoxin-challenged calve was significantly lower from 8 to 12 hr after the endotoxin challenge than pre-challenge values. In addition, serum Zn concentrations in calves from 4 to 24 hr after endotoxin challenges were significantly lower than those of control. Our results indicate that serum Zn concentration has potential as diagnostic markers for detecting inflammation in calves with endotoxin shock.

PIXE analysis of concrete composition in a research reactor facility under decommissioning

This study conducted an evaluation of elemental composition of concrete in an old nuclear reactor facility using the particle-induced X-ray emission (PIXE) technique. Given that the physical volume of the concrete is huge, easy, and quick analysis of many samples via the PIXE method can expedite the decommissioning work. The result of this study confirmed that it is possible to use the PIXE method for detecting light and medium-heavy elements (from Na to Fe) in concrete without the complicated chemical treatment. Some of these elements contain parent nuclides of Na-24, Al-28, and Fe-55, which are dominant components with residual radioactivity for a period of up to 10 years after facility shutdown.

Pixe Analysis of Some Ghanaian Coins Using a 1.7 mv Pelletron Accelerator

The elemental composition of some Ghanaian coins spanning from the years 1967 to 2007 have been successfully analysed using a newly installed 1.7 MV Pelletron accelerator at the Ghana Atomic Energy Commission. Using Particle Induced Xray Emission (PIXE) spectrometry, this study attempts to relate the major composition of these coins to the economic state of the country at the year of minting and also to validate the PIXE method used at the Centre. This work demonstrates that the Accelerator Research Centre has the capacity to analyse mintage coins in Ghana.

Standardisation of the ion beam facility at Chandigarh cyclotron for simultaneous PIXE and PESA analysis

Abstract The proton beam facility at variable energy cyclotron (VEC) Panjab University, Chandigarh, India is being used for Particle Induced X-ray Emission (PIXE) analysis of different environmental, biological and industrial samples. The PIXE method, however, does not provide any information of low Z elements like carbon, nitrogen, oxygen and fluorine. As a result of the increased need for rapid and multi-elemental analysis of biological and environmental samples, the PIXE facility was upgraded and standardized to facilitate simultaneous measurements using PIXE and Proton Elastic Scattering Analysis (PESA). Both PIXE and PESA techniques were calibrated and standardized individually. Finally, the set up was tested by carrying out simultaneous PIXE and PESA measurements using a 2 mm diameter proton beam of 2.7 MeV on few multilayered thin samples. The results obtained show excellent agreement between PIXE and PESA measurements and confirm adequate sensitivity and precision of the experimental set up.

Total Calcium and Albumin Are Decreased in the Deeper Epidermis of Patients with Chronic Kidney Disease-Associated Pruritus

Background/Aims: In our earlier studies, we reported high concentrations of intra- and extracellular calcium ions (Ca²⁺) in the deeper epidermis of patients with chronic kidney disease (CKD) and associated pruritus. To determine the cause of this phenomenon, we measured total calcium (TCa) concentrations in the deeper epidermis and performed immunostaining of epidermal albumin, which binds to Ca²⁺. Methods: This study included 45 patients with CKD-stage 5, which was defined as severely reduced kidney function (i.e., estimated glomerular filtration rate less than 15 mL/min or on dialysis). Subjects were divided into the pruritus group, consisting of patients with mild, moderate, or severe uremic pruritus, and the non-pruritus group, consisting of patients with no or slight pruritus. The particle-induced X-ray emission method was used to measure elements including TCa. Furthermore, we have immunostained epidermal albumin using anti-albumin antibodies and compared the results in the pruritus and non-pruritus groups. Results: The TCa concentration in the spinous layer of patients with CKD with CKD-associated pruritus was lower than in patients with CKD without pruritus (median [range], 395 [235-1,063] vs. 476 [342-1,243] μg/g). The intensity of epidermal albumin expression in the spinous layer was weaker in patients with CKD with CKD-associated pruritus than in those without. Conclusion: Patients with CKD with CKD-associated pruritus demonstrated higher Ca²⁺ concentrations but lower TCa concentrations than patients without CKD-associated pruritus. This could be in part due to low concentrations of epidermal albumin, which binds to Ca²⁺, in those with CKD-associated pruritus. These results clarify the pathophysiology of CKD-associated pruritus, providing a valuable foundation for the future development of treatments for this condition.

Chemical analysis of archaeological clay potteries by PIGE and PIXE methods using proton beams from tandem accelerator for provenance study

Particle induced gamma-ray emission (PIGE) and particle induced X-ray emission (PIXE) methods using proton beams from tandem accelerator were applied for chemical analysis of sixteen archaeological clay potteries. The samples were characterized by Fourier transform infrared spectroscopy to obtain firing temperature. PIGE method was used for determination of concentrations of Si and Al and PIXE was used to determine concentrations of eight transition elements. Concentration ratios of SiO2 to Al2O3 and statistical cluster analysis using transition elemental concentrations of samples indicated two major groups. PIGE and PIXE methods were validated by analyzing reference materials from IAEA and NIST.

Evaluation of Toxicological Effects of an Aqueous Extract of Shells from the Pecan Nut Carya illinoinensis (Wangenh.) K. Koch and the Possible Association with Its Inorganic Constituents and Major Phenolic Compounds.

Background. Industrial processing of the pecan nut Carya illinoinensis K. Koch generated a large amount of shells, which have been used to prepare nutritional supplements and medicinal products; however, the safe use of shells requires assessment. This study evaluated the toxic, genotoxic, and mutagenic effects of pecan shell aqueous extract (PSAE) and the possible contribution of phenolic compounds, ellagic and gallic acids, and inorganic elements present in PSAE to induce toxicity. Results. Levels of inorganic elements like K, P, Cl, and Rb quantified using the Particle-Induced X-Ray Emission method were higher in PSAE than in pecan shells, while Mg and Mn levels were higher in shells. Mice showed neurobehavioral toxicity when given high PSAE doses (200–2,000 mg kg−1). The LD50 was 1,166.3 mg kg−1. However, PSAE (50–200 mg·kg−1) and the phenolic compounds (10–100 mg·kg−1) did not induce DNA damage or mutagenicity evaluated using the comet assay and micronucleus test. Treatment with ellagic acid (10–100 mg·kg−1) decreased triglyceride and glucose levels, while treatments with PSAE and gallic acid had no effect. Conclusion. Pecan shell toxicity might be associated with high concentrations of inorganic elements such as Mn, Al, Cu, and Fe acting on the central nervous system, besides phytochemical components, suggesting that the definition of the safe dose should take into account the consumption of micronutrients.

PIXE in the studies of stable cesium sorption from water solutions

The possibility of cesium content evaluation by the particle induced X-ray emission method from X-ray emission L-shell at sorption from water solutions has been investigated. Consideration has been given to the processes of dynamic cesium sorption by zeolites from water solutions. The technique has been developed for preparing targets analyzable in the proton beam of energy up to 2.0 MeV. The relationship has been determined between the cesium content and the intensity of X-ray emission L-shell. Copyright © 2015 John Wiley & Sons, Ltd.

Determination of the density of silicon–nitride thin films by ion-beam analytical techniques (RBS, PIXE, STIM)

This work presents the investigation of some commercially available and commonly used Si3N4 foils prepared with LPCVD technique. The density and the stoichiometry of these films were determined by Rutherford backscattering spectroscopy and profilometry, while the study of impurities was achieved with particle induced X-ray emission method. It was found that the density of the studied Si3N4 films is significantly less (~2.71 g cm−3), while the stoichiometry is close to the values of the bulk material. The results were verified by measuring the ion energy loss through the films by scanning transmission ion microscopy.

PIXE analysis of trace elements included in oral lichen planus-affected mucosa

The purpose of our research is to identify causative metals by using particle-induced X-ray emission (PIXE) method to directly analyze trace elements in the oral mucosa with oral lichen planus (OLP). Subjects were 72 patients with OLP, and the patients are 25 men and 47 women, with a mean age of 60.1 years (OLP group). The control is elemental analysis data by PIXE method of 100 cases of the oral mucosa of healthy persons (control group). Seventeen essential elements and 12 contaminating elements were detected from the oral mucosa of the OLP and control groups. As to appearance ratio of elements in oral mucosa, among the contaminating elements, levels of Al, Ga, Sb, Hg and Pb were significantly lower, whereas Au and Y were significantly higher in the OLP group than in the control group. About concentration of elements in oral mucosa, the contaminating elements in the oral mucosa of the OLP group had a lower appearance ratio than in the control but each of the contaminating elements was more abundant. Among 25 subjects in the OLP group and seven in the control group, serum, oral mucosa and saliva were collected from the same individual. Abundance in serum and saliva varied depending on the element, but in general, a tendency was seen for contaminating elements to be more abundant in saliva than in serum and for essential elements to be more abundant in serum than in saliva.

Doslidzhennya metodamy yadernoho mikroanalizu poverkhnevykh nanoshariv struktury zoloto−kremniy

The Rutherford backscattering and particle-induced X-ray emission methods are used to study the surface layers in gold–silicon structures, the parameters of which govern the operational characteristics of electron devices constructed on their basis. The measurements are performed on a high-precision microanalytical unit “Nuclear scanning probe” recently put into operation at the “Spectrum” laboratory. The thicknesses of a gold layer sputtered onto the specimen surface were about 17 and 20 nm for two different specimens. The layer non-uniformity was less than 1.6 nm and did not exceed the experimental error. A substantial amount of the fluorine impurity was revealed under the gold layer in the gold–silicon interface layer. Probably, it may be remnants of fluorine remaining after the etching of the silicon surface in a mixture of acids that included the hydrofluoric one (HF). The amount of fluorine detected in a series of measurements was found to strongly correlate with the current of alpha-particles at the target surface during the spectrum measurement. Since the local heating of the target depends on the current, it is evident that the local diffusion rate of fluorine atoms varied over the target surface.

Magnetic properties of carbon nanodisk and nanocone powders

We have investigated the magnetic properties of carbon powders which consist of nanodisks, nanocones, and a small fraction of carbon-black particles. Magnetization measurements were carried out using a superconducting quantum interference device in magnetic fields $-5<\mu_{0}H<5\:\mathrm{T}$ for temperatures in the range $2\leq T<350\:\mathrm{K}$. Measurements of the magnetization $M$ versus temperature $T$ and magnetic field $\mu_{0}H$ for these carbon samples show diamagnetism and paramagetism with an additional ferromagnetic contribution. The ferromagnetic magnetization is in agreement with the calculated magnetization from Fe impurities as determined by the particle-induced x-ray emission method ($<75\:\mu\mathrm{g/g}$). Magnetization measurements in weak magnetic fields show thermal hysteresis, and for strong fields the magnetization $M$ decreases as $M\sim aT^{-\alpha}$ with $\alpha<1$, which is slower than the Curie law ($\alpha=1$), when the temperature increases. The magnetization $M$ versus magnetic field $\mu_{0}H$ shows paramagnetic free-spin $S=\frac{1}{2}$ and $\frac{3}{2}$ behaviors for temperatures $T=2\:\mathrm{K}$ and $15\leq T\leq50\:\mathrm{K}$, respectively. A tendency for localization of electrons was found by electron spin resonance when the temperature $T$ decreases ($2<T<40\:\mathrm{K}$). The magnetic properties in these carbon cone and disk powder samples are more complex than a free-spin model predicts, which is apparently valid only for the temperature $T=2\:\mathrm{K}$.

Particle-Induced X-ray Emission Analysis of Elements in Plasma from Wild and Captive Sea Turtles (Eretmochelys imbricata, Chelonia mydas, and Caretta caretta) in Okinawa, Japan

The aim of this study was to evaluate the reliability of direct determination of trace and major element concentrations in plasma samples from wild (six hawksbill, nine green, and nine loggerhead) and captive sea turtles (25 howksbill, five green, and three loggerhead) in Okinawa, Japan. The particle induced X-ray emission method allowed detection of 23 trace and major elements (Al, As, Br, Ca, Cl, Cr, Cu, Fe, Hg, K, Mg, Mn, Mo, Ni, P, Pb, S, Se, Si, Sr, Ti, Y, and Zn). The wild sea turtles were found to have high concentrations of As and Pb in plasma compared with captive, but there were no significant changes in the Al and Hg concentrations. Loggerhead sea turtles were found to have significantly higher accumulation of As and Pb in plasma in comparison to other species. These findings may be useful when adjusting environmental and species-related factors in severely polluted marine ecosystems. Our results indicate that measuring the plasma As and Pb concentrations in wild sea turtles might be of help to assess the level of pollution in marine ecosystems, keeping in mind that loggerhead sea turtles had been shown to have higher levels of As and Pb in plasma.

Forensic analysis of tempered sheet glass by particle induced X-ray emission (PIXE)

The elemental concentrations of five trace elements in tempered sheet glass fragments were determined using particle-induced X-ray emission (PIXE) spectrometry. The trace element concentrations for calcium, iron, manganese, strontium, and titanium are compared to those obtained by inductively-coupled plasma-atomic emission spectrometry (ICP-AES) following complete digestion by hydrofluoric acid. For these five elements, the absolute concentrations obtained by both methods are shown to agree well over a wide range of concentrations. The limits of detection for trace elements are typically lower for the ICP-AES method. However, we show that the concentrations of these five elements can be accurately measured by the PIXE method. Since PIXE is an entirely non-destructive method, there exists a niche for this technique to be used as a complement to the more sensitive ICP-AES technique in the forensic analysis of sheet glass.

Trace and Major Elements Status in Bronchoalveolar Lavage Fluid in Dogs with or without Bronchopneumonia

The aim of this study was to investigate the relationships between the bronchopneumonia and mean concentrations of those trace elements in bronchoalveolar lavage fluid (BALF). Twenty-nine dogs were included this study (17 healthy dogs and 12 dogs with respiratory disease). Each BALF sample had been obtained during bronchoscope examination by use of a standardized method. The concentrations of Al, Br, Ca, Cu, Fe, K, Ni, P, Si, Sr and Zn in BALF were measured by the particle-induced X-ray emission method. We found no relationship between the bronchopneumonia and the levels of elements in the BALF, except Ca, P and Zn. The dogs with respiratory disease were found to have a large amount of Ca and Zn, and a high Ca/P and Zn/Cu ratios in BALF compared to those without respiratory disease.

Determination of the metal balance shift induced in small fresh water fish by X-ray irradiation using PIXE analysis

The balance of essential elements in organisms can be changed by environmental stresses. A small fresh water fish, the medaka, was irradiated with X-rays (total dose: 17 Gy, which is not a lethal dose for this fish). Essential elements in the liver, gall bladder, kidney, spleen, heart and brain of the fish were measured by the particle induced X-ray emission (PIXE) method and compared with those of a control fish. Various changes in the elemental balance shift were observed. The PIXE method can analyze many elements in a small sample simultaneously, and so the changes in elemental content induced by irradiation were readily determined.
