Time- and temperature-dependent dynamics of prothoracicotropic hormone and ecdysone sensitivity co-regulate pupal diapause in the green-veined white butterfly Pieris napi

Diapause, a general shutdown of developmental pathways, is a vital adaptation allowing insects to adjust their life cycle to adverse environmental conditions such as winter. Diapause in the pupal stage is regulated by the major developmental hormones prothoracicotropic hormone (PTTH) and ecdysone. Termination of pupal diapause in the butterfly Pieris napi depends on low temperatures; therefore, we study the temperature-dependence of PTTH secretion and ecdysone sensitivity dynamics throughout diapause, with a focus on diapause termination. While PTTH is present throughout diapause in the cell bodies of two pairs of neurosecretory cells in the brain, it is absent in the axons, and the PTTH concentration in the haemolymph is significantly lower during diapause than during post diapause development, indicating that the PTTH signaling is reduced during diapause. The sensitivity of pupae to ecdysone injections is dependent on diapause stage. While pupae are sensitive to ecdysone during early diapause initiation, they gradually lose this sensitivity and become insensitive to non-lethal concentrations of ecdysone about 30 days into diapause. At low temperatures, reflecting natural overwintering conditions, diapause termination propensity after ecdysone injection is precocious compared to controls. In stark contrast, at high temperatures reflecting late summer and early autumn conditions, sensitivity to ecdysone does not return. Thus, here we show that PTTH secretion is reduced during diapause, and additionally, that the low ecdysone sensitivity of early diapause maintenance is lost during termination in a temperature dependent manner. The link between ecdysone sensitivity and low-temperature dependence reveals a putative mechanism of how diapause termination operates in insects that is in line with adaptive expectations for diapause.

Egfr Signaling Is a Major Regulator of Ecdysone Biosynthesis in the Drosophila Prothoracic Gland.

Understanding the mechanisms that determine final body size of animals is a central question in biology. In animals with determinate growth, such as mammals or insects, the size at which the immature organism transforms into the adult defines the final body size, as adult individuals do not grow [1]. In Drosophila, the growth period ends when the immature larva undergoes the metamorphic transition to develop the mature adult [2]. This metamorphic transition is triggered by a sharp increase of the steroid ecdysone, synthetized in the prothoracic gland (PG), that occurs at the end of the third instar larvae (L3) [3-6]. It is widely accepted that ecdysone biosynthesis in Drosophila is mainly induced by the activation of tyrosine kinase (RTK) Torso by the prothoracicotropic hormone (Ptth) produced into two pairs of neurosecretory cells that project their axons onto the PG [7, 8]. However, the fact that neither Ptth nor torso-null mutant animals arrest larval development but only present a delay in the larva-pupa transition [9-11] mandates for a reconsideration of the conventional model. Here, we show that Egfr signaling, rather than Ptth/torso, is the major contributor of ecdysone biosynthesis in Drosophila. We found that Egfr signaling is activated in the PG in an autocrine mode by the EGF ligands spitz and vein, which in turn are regulated by the levels of ecdysone. This regulatory positive feedback loop ensures the production of ecdysone to trigger metamorphosis by a progressive Egfr-dependent activation of MAPK/ERK pathway, thus determining the animal final body size.

CAPA neuropeptides and their receptor form an anti-diuretic hormone signaling system in the human disease vector, Aedes aegypti

Insect CAPA neuropeptides are homologs of mammalian neuromedin U and are known to influence ion and water balance by regulating the activity of the Malpighian ‘renal’ tubules (MTs). Several diuretic hormones are known to increase primary fluid and ion secretion by insect MTs and, in adult female mosquitoes, a calcitonin-related peptide (DH31) called mosquito natriuretic peptide, increases sodium secretion to compensate for the excess salt load acquired during blood-feeding. An endogenous mosquito anti-diuretic hormone was recently described, having potent inhibitory activity against select diuretic hormones, including DH31. Herein, we functionally deorphanized, both in vitro and in vivo, a mosquito anti-diuretic hormone receptor (AedaeADHr) with expression analysis indicating highest enrichment in the MTs where it is localized within principal cells. Characterization using a heterologous in vitro system demonstrated the receptor was highly sensitive to mosquito CAPA neuropeptides while in vivo, AedaeADHr knockdown abolished CAPA-induced anti-diuretic control of DH31-stimulated MTs. CAPA neuropeptides are produced within a pair of neurosecretory cells in each of the abdominal ganglia, whose axonal projections innervate the abdominal neurohaemal organs, where these neurohormones are released into circulation. Lastly, pharmacological inhibition of nitric oxide synthase (NOS) and protein kinase G (PKG) signaling eliminated anti-diuretic activity of CAPA, highlighting the role of the second messenger cGMP and NOS/PKG in this anti-diuretic signaling pathway.

Serotonin receptor B may lock the gate of PTTH release/synthesis in the Chinese silk moth, Antheraea pernyi; a diapause initiation/maintenance mechanism?

The release of prothoracicotropic hormone, PTTH, or its blockade is the major endocrine switch regulating the developmental channel either to metamorphosis or to pupal diapause in the Chinese silk moth, Antheraea pernyi. We have cloned cDNAs encoding two types of serotonin receptors (5HTRA and B). 5HTRA-, and 5HTRB-like immunohistochemical reactivities (-ir) were colocalized with PTTH-ir in two pairs of neurosecretory cells at the dorsolateral region of the protocerebrum (DL). Therefore, the causal involvement of these receptors was suspected in PTTH release/synthesis. The level of mRNA5HTRB responded to 10 cycles of long-day activation, falling to 40% of the original level before activation, while that of 5HTRA was not affected by long-day activation. Under LD 16:8 and 12:12, the injection of dsRNA5HTRB resulted in early diapause termination, whereas that of dsRNA5HTRA did not affect the rate of diapause termination. The injection of dsRNA5HTRB induced PTTH accumulation, indicating that 5HTRB binding suppresses PTTH synthesis also. This conclusion was supported pharmacologically; the injection of luzindole, a melatonin receptor antagonist, plus 5th inhibited photoperiodic activation under LD 16:8, while that of 5,7-DHT, induced emergence in a dose dependent fashion under LD 12:12. The results suggest that 5HTRB may lock the PTTH release/synthesis, maintaining diapause. This could also work as diapause induction mechanism.

Molecular characterization and developmental expression of the gene encoding the prothoracicotropic hormone in the beet armyworm, Spodoptera exigua

Prothoracicotropic hormone (PTTH), a neuropeptide hormone stimulating the prothoracic glands to synthesize ecdysone, plays an important role in regulating postembryonic development in insects. The cDNA encoding PTTH was isolated and sequenced from the beet armyworm, Spodoptera exigua (Spe). The deduced amino acid sequence is composed of a signal peptide, a peptide (65 amino acids) of unknown function, and a mature PTTH molecule (111 amino acids). The Spe-PTTH shows similarities (45.5%-70.3%) to other known PTTHs reported in Lepidoptera species, but 7 cysteine residues and the hydrophobic regions were conserved. Whole-mount immunocytochemistry by using an antiserum against recombinant Helicoverpa armigera PTTH showed that Spe-PTTH was synthesized in two pairs of neurosecretory cells in the S. exigua brain. Northern blot analysis demonstrates the presence of a 1.2-kb transcript in the brain. The Spe-PTTH mRNA is detectable at high levels at the wandering larval stage, early pupal stage, and pharate adult stage, suggesting that the Spe-PTTH gene might be correlated with molting, metamorphosis, and reproduction.

Rhythmic Firing Activity of Median Neurosecretory Cells of the Subesophageal Ganglion and its Coordination with Periodically Occurring Abdominal Movements in the Mealworm Beetle, Tenebrio obscurus

An immunocytochemical study using an FXPRLamide antiserum revealed three clusters of neurosecretory cells along the midline of the subesophageal ganglion of the tenebrionid [corrected] beetle, Zophobas atratus [corrected] Firing activity of five pairs of neurosecretory cells in the mandibular and maxillary clusters was recorded from an axonal tract of the cells throughout the entire pupal period. The population of neurosecretory cells became active during the middle and late pupal periods, and they usually discharged clusters of action potentials at an interval of 30-90 min. The ultradian activity rhythm of the cells in Z. atratus [corrected] was related to a periodic discharge of electrical activity in developing flight muscles, as has been observed in the homologous cells in the silkmoth, Bombyx mori. Furthermore, the rhythmic activity of the neurosecretory cells in the mealworm was closely synchronized with periodically occurring rhythmic abdominal movements that caused extracardiac hemocoelic pulsations, which facilitate hemolymph circulation and exchange of respiratory gases. The results suggest that the secretory products of the neurosecretory cells may activate and/or orchestrate physiological mechanisms supporting morphogenesis during metamorphosis.

Immunological Localization of Mosquito Ovary Ecdysteroidogenic Hormone I and Fruit Fly Insulin Receptor in Adult <I>Phormia regina</I> (Diptera: Calliphoridae)

The neurohormone ovary ecdysteroidogenic hormone I (OEH I), originally identified from Aedes aegypti (L.), has an effect on mosquito ecdysteroidogenesis. Would there be a similar presence and function in other adult flies, e.g., black blow fly, Phormia regina (Meigen)? One pair of median neurosecretory cells (MNCOEH I) and one pair of lateral neurosecretory cells (LNCOEH I) were immunopositive to OEH I antiserum in the brain of sugar-fed and liver-fed female P. regina. In addition, two pairs of neurosecretory cells (SOGNCOEH I) positive to this antiserum were found in the suboesophageal ganglion only in adult female P. regina at 16, 20, 23, 28, 36, and 72 h after a liver meal. In addition to the positive pairs of MNCOEH I and LNCOEH I in females, there were four more pairs of positive neurons (MSNFNCOEH I) in the region between the MNCOEH I and the esophageal foramen in P. regina males, but not in females. The presence of OEH I in male flies needs further attention as to the function of this hormone in male dipterans. We also found substances positive to Drosophila melanogaster Meigen insulin receptor antiserum located in the corpus cardiacum and the nerves going to the corpus allatum of adult female P. regina at 6 and 30 h after the liver meal, but not to the brain.

Molecular cloning of the prothoracicotropic hormone from the tobacco hornworm, Manduca sexta

A cDNA encoding a putative precursor of prothoracicotropic hormone (PTTH) from the tobacco hornworm, Manduca sexta, was isolated and sequenced. This clone contains an open reading frame encoding a 226-amino acid prepropeptide hormone. The deduced amino acid sequence is composed of a signal sequence, a precursor domain and a mature hormone and shows similarities to the other PTTHs that have been cloned from closely related lepidopteran species, Bombyx mori, Samia cynthia ricini, Antheraea peryni, and Hyalophora cecropia. Although these cDNAs showed slightly less similarities in predicted amino acid sequences, seven cysteine residues and the hydrophobic regions within those mature peptides were conserved. In situ hybridization using a cDNA probe encoding the Manduca PTTH showed that PTTH mRNA was in two pairs of neurosecretory cells in the Manduca brain. The recombinant putative Manduca PTTH produced in E. coli was biologically active, both causing a larval molt in neck-ligated Manduca 4th instar larvae (ED 50=50 pM) and the adult molt of diapausing Manduca pupae (ED 50=79 pM), but was unable to stimulate molting of debrained Bombyx pupae.

Synchronous firing dynamics in a heterogeneous neurosecretory-cell population in an insect

Five pairs of neurosecretory cells in the subesophageal ganglion of the silkmoth Bombyx mori discharge action potentials in (near) synchrony to release a pheromone biosynthesis-activating neuropeptide (PBAN). Waveforms of compound action potentials recorded extracellularly from axonal tracts were analyzed to determine the firing activity, timing of spikes and the combination of active cells. Analyses revealed a heterogeneous cellular organization of the neurosecretory cell system. There was a gradient in the firing activity among the cells and the activity of a cell was closely related to relative timing of firing: the most active cell was usually the first to fire and participated in about 90% of all synchronous firing events, while the least active unit was mostly the last to fire and contributed to only 40% of all firing events. A cell with a higher firing activity had a higher potential to mediate propagation of synchronous firing in the cell system. Firing activities of right and left cell groups usually differed and the difference increased in case of a low temperature. Synchronous firings occurred more frequently among the same subgroup of cells rather than different subgroups. Heterogeneous cellular organization and coupling may be important for producing a graded pattern of active cell numbers, which seems to be suitable for maintaining a stable firing (secretory) activity of the cell system for a long period of time.

Phe-X-Pro-Arg-Leu-NH 2 peptide producing cells in the central nervous system of the silkworm, Bombyx mori

Members of the neuropeptide family having Phe-X-Pro-Arg-Leu-NH 2 (FXPRLamide; X=Ser, Thr, Val, or Gly) at the C-terminus serve as regulators of oviduct and visceral muscle contraction, sex pheromone production, and diapause induction. Antibody raised against Bombyx mori diapause hormone recognized a variety of FXPRLamide peptides. Using this antibody, the antigen was immunocytochemically localized in the central nervous system (CNS) of the silkworm, Bombyx mori. Immunoreactive somata were observed in all ganglia of the CNS including the brain. Twelve somata localized at the midline of the suboesophageal ganglion (SG) were most intensely stained, and their neurite projections reached the retrocerebral complex. Thus, these cells in the SG exhibited typical features of neuroendocrine neurons. Marked reduction in immunoreactivity was observed in a pair of neurosecretory cells in the labial neuromere in SG of diapause type pupae, which indicates an active release of FXPRLamide peptides from these cells. No clear connection to neurohemal sites were observed in immunoreactive cells in the brain, thoracic or abdominal ganglia, suggesting that the immunoreactive peptides in these organs are likely to serve as neurotransmitters or neuromodulators.

Electrical properties of the cerebral prothoracicotropic hormone cells in diapausing and non-diapausing pupae of the tobacco hornworm, Manduca sexta.

Prothoracicotropic hormone (PTTH) is an insect brain neuropeptide that is a primary factor regulating an insect development. Curtailment of its release is thought to be responsible for the pupal diapause of tobacco hornworm, Manduca sexta. The cell synthesizing and secreting the PTTH has been identified as a pair of neurosecretory cells in the pars lateralis on each brain hemisphere. Using intracellular recording techniques, we have demonstrated electrical properties of the PTTH cells in different physiological status, i.e., diapausing and developing pupae. In diapausing pupae, they showed threshold value increasing and input resistance decreasing with the progress of diapausing state, indicating that they were getting unexcitable. Spontaneous action potentials and excitatory postsynaptic potentials (EPSPs) were rarely observed in deeply diapausing state. Non-diapausing PTTH cells were almost silent except day-2, showing rather constant values of electrical properties. On day-2, a significant proportion of the cells had spontaneous action potentials, showing less negative membrane potential values than inactive cells. Exclusively inhibitory postsynaptic potentials (IPSPs) were observed in significant numbers of the cells during the period from day-2 to day-5. On the basis of the results obtained, we proposed a working hypothesis that electrical activities of the PTTH cell may be primarily regulated by its membrane properties which are further modulated by the synaptic mediation.

The vasopressin-like immunoreactive (VPLI) neurons of the locust, Locusta migratoria. II. Physiology

The two vasopressin-like immunoreactive (VPLI) neurons of the locust, Locusta migratoria, have cell bodies in the suboesophageal ganglion and extensive arborizations throughout the CNS. One of the two peptides responsible for AVP-like immunoreactivity is a vasopressin-related peptide with putative 'diuretic hormone' properties. These neurons also have FLRF-like immunoreactivity, probably due to the FMRF-amide-related peptide. SchistoFLRF-amide, isolated from Schistocerca gregaria. This peptide has cardioinhibitory activity and a dual potentiation/inhibition of slow motoneuron induced muscle-twitch tension. Although haemolymph AVP-like peptide titre fluctuates under various conditions, the mechanism that regulates neurohaemal release of this peptide is not understood. Very little is known of the release of SchistoFLRF-amide. We have used intracellular recording from VPLI neurons in vivo to reveal synaptic inputs that lead to changes in their level of spiking activity, and probably, release of both the AVP-like peptides and SchistoFLRF-amide. This pair of neurosecretory cells has a major, common excitatory input whose sustained rate of activity is inversely related to light intensity; VPLI spiking activity, driven by this input, is greater in the dark than in light. This input is from a pair of descending brain interneurons. Their light-sensitivity persists after ablation of compound eyes, optic lobes and ocelli, showing them to be part of an extra-ocular photoreceptor system. Attempts to record from, and individually stain, the descending neuron have been unsuccessful, although its axon location and diameter in the circumoesophageal connective have been determined. Possible locations for its cell body have been identified; one region, close to the pars intercerebralis, is known to be photosensitive in some insects. Mechanosensory stimuli also lead to brief increases in VPLI spiking activity via the descending interneuron, though this modality rapidly habituates. We detect no changes in VPLI spiking activity that consistently correlate with the osmolality of perfusion salines; such changes might have been expected from their previously proposed role in water homeostasis. Alternative roles for VPLI cells are discussed.

Histological observations on circadian changes in the neurosecretory cells of cockroach sub-oesophageal ganglia

Six pairs of neurosecretory cells in the sub-oesophageal ganglion of Periplaneta americana stain intensely with paraldehyde fuchsin; their morphology is described. These cells were subjected to a detailed histological study throughout the 24 hr. No clear evidence of a circadian cycle in the movement of neurosecretory material was detected in any of them. The nuclei of one pair of neurosecretory cells believed to be concerned in the control of the circadian rhythm of locomotion were significantly larger during the period of maximum locomotor activity than at other times, but no sharp change in their nuclear volume was detected at the onset of activity. Possible errors in the estimation of nuclear volume are discussed. The results are considered in relation to those of other workers and in relation to the possibility that circadian metabolic changes in the organism as a whole may be reflected in synchronous changes in many of its cells.
