Cyanobacterial Extracellular Polymeric Substances and their role in Biodeterioration of Temples and Monuments

Cyanobacteria are the ancient gram-negative prokaryotic organisms having a wide range of forms ranging from unicellular to filamentous forms. They are oxygenic photosynthetic microorganisms having cosmopolitan distribution in almost all aquatic, terrestrial habitats and even in extreme ecosystems (e.g., hot and cold deserts, hydrothermal vents, hypersaline environments and rocks) due to their tremendous adaptability to varying environmental conditions. Extracellular Polymeric Substances (EPS) are the extracellular mucilaginous substances produced by members of cyanobacteria. Production of EPS is widely known in cyanobacteria and protect the cyanobacterial cells from various stresses such as desiccation and UV-radiation. This review discusses about cyanobacterial EPS and its role in biodeterioration of monuments and temples.

Cyanobacteria-based double-mediated photo-microbial electrochemical cells are promising future energy sources for electricity generation and hydrogen production

Photoelectrons generated from water oxidation in cyanobacteria are transferred to the anode by double mediators and used for photocurrent generation and H2 production. Photocurrent and hydrogen production were successfully achieved from DM-PMECs.

A New Remote Sensing-Based System for the Monitoring and Analysis of Growth and Gas Exchange Rates of Photosynthetic Microorganisms Under Simulated Non-Terrestrial Conditions.

Oxygenic photosynthetic microorganisms are a focal point of research in the context of human space exploration. As part of the bioregenerative life-support systems, they could have a key role in the production of breathable O2, edible biomasses and in the regeneration of CO2 rich-atmospheres and wastewaters produced by astronauts. The test of the organism’s response to simulated physico-chemical parameters of planetary bodies could also provide important information about their habitability potential. It is believed that the success of future planetary and space missions will require innovative technologies, developed on the base of preliminary experiments in custom-made laboratory facilities. In this context, simulation chambers will play a pivotal role by allowing the growth of the microorganisms under controlled conditions and the evaluation in real-time of their biomass productivity and impact on atmosphere composition. We here present a system capable of addressing these requirements with high replicability and low costs. The setup is composed by three main parts: 1) a Star Light Simulator, able to generate different light intensities and spectra, including those of non-solar stars; 2) an Atmosphere Simulator Chamber where cultures of photosynthetic microorganisms can be exposed to different gas compositions; 3) a reflectivity detection system to measure from remote the Normalized Difference Vegetation Indexes (NDVI). Such a setup allows us to monitor photosynthetic microorganism’s growth and gas exchange performances under selected conditions of light quality and intensity, temperature, pressure, and atmospheres simulating non-terrestrial environments. All parameters are detected by remote sensing techniques, thus without interfering with the experiments and altering the environmental conditions set. We validated the setup by growing cyanobacteria liquid cultures under different light intensities of solar illumination, collecting data on their growth rate, photosynthetic activity, and gas exchange capacity. We utilized the reflectivity detection system to measure the reflection spectra of the growing cultures, obtaining their relative NDVI that was shown to correlate with optical density, chlorophyll content, and dry weight, demonstrating the potential application of this index as a proxy of growth.

Hybrid Ni@ZnO@ZnS-Microalgae for Circular Economy: A Smart Route to the Efficient Integration of Solar Photocatalytic Water Decontamination and Bioethanol Production.

Water remediation and development of carbon‐neutral fuels are a priority for the evermore industrialized society. The answer to these challenges should be simple, sustainable, and inexpensive. Thus, biomimetic‐inspired circular and holistic processes combing water remediation and biofuel production can be an appealing concept to deal with these global issues. A simple circular approach using helical Spirulina platensis microalgae as biotemplates to synthesize Ni@ZnO@ZnS photocatalysts for efficient solar water decontamination and bioethanol production during the recycling process is presented. Under solar irradiation, the Ni@ZnO@ZnS‐Spirulina photocatalyst exhibits enhanced activity (mineralization efficiency >99%) with minimal photocorrosion and excellent reusability. At the end of its effective lifetime for water remediation, the microalgae skeleton (mainly glycogen and glucose) of the photocatalyst is recycled to directly produce bioethanol by simultaneous saccharification and fermentation process. An outstanding ethanol yield of 0.4 L kg−1, which is similar to the highest yield obtained from oxygenic photosynthetic microorganisms, is obtained. Thus, the entire process allows effective solar photocatalytic water remediation and bioethanol production at room temperature using simple and easily scalable procedures that simultaneously fixes carbon dioxide, thereby constituting a zero‐carbon‐emission circular process.

The Development of Biophotovoltaic Systems for Power Generation and Biological Analysis.

Biophotovoltaic systems (BPVs) resemble microbial fuel cells, but utilise oxygenic photosynthetic microorganisms associated with an anode to generate an extracellular electrical current, which is stimulated by illumination. Study and exploitation of BPVs have come a long way over the last few decades, having benefited from several generations of electrode development and improvements in wiring schemes. Power densities of up to 0.5 W m−2 and the powering of small electrical devices such as a digital clock have been reported. Improvements in standardisation have meant that this biophotoelectrochemical phenomenon can be further exploited to address biological questions relating to the organisms. Here, we aim to provide both biologists and electrochemists with a review of the progress of BPV development with a focus on biological materials, electrode design and interfacial wiring considerations, and propose steps for driving the field forward.

Extending the ecological distribution of Desmonostoc genus: proposal of Desmonostoc salinum sp. nov., a novel Cyanobacteria from a saline-alkaline lake.

Cyanobacteria is an ancient phylum of oxygenic photosynthetic microorganisms found in almost all environments of Earth. In recent years, the taxonomic placement of some cyanobacterial strains, including those belonging to the genus Nostocsensu lato, have been reevaluated by means of a polyphasic approach. Thus, 16S rRNA gene phylogeny and 16S-23S internal transcribed spacer (ITS) secondary structures coupled with morphological, ecological and physiological data are considered powerful tools for a better taxonomic and systematics resolution, leading to the description of novel genera and species. Additionally, underexplored and harsh environments, such as saline-alkaline lakes, have received special attention given they can be a source of novel cyanobacterial taxa. Here, a filamentous heterocytous strain, Nostocaceae CCM-UFV059, isolated from Laguna Amarga, Chile, was characterized applying the polyphasic approach; its fatty acid profile and physiological responses to salt (NaCl) were also determined. Morphologically, this strain was related to morphotypes of the Nostocsensu lato group, being phylogenetically placed into the typical cluster of the genus Desmonostoc. CCM-UFV059 showed identity of the 16S rRNA gene as well as 16S-23S secondary structures that did not match those from known described species of the genus Desmonostoc, as well as distinct ecological and physiological traits. Taken together, these data allowed the description of the first strain of a member of the genus Desmonostoc from a saline-alkaline lake, named Desmonostoc salinum sp. nov., under the provisions of the International Code of Nomenclature for algae, fungi and plants. This finding extends the ecological coverage of the genus Desmonostoc, contributing to a better understanding of cyanobacterial diversity and systematics.

Stereochemical assignment of the unique electron acceptor 5'-hydroxyphylloquinone, a polar analog of vitamin K1 in photosystem I.

A unique electron-accepting analog of vitamin K1 found in photosystem I in several species of oxygenic photosynthetic microorganisms was confirmed to be 5'-hydroxyphylloquinone (1) through stereo-uncontrolled synthesis. Furthermore, the stereochemistry of 1 obtained from Synechococcus sp. PCC 7942 was assigned to be 5'S using proline-catalyzed stereocontrolled reactions.

Biofouling of marbles by oxygenic photosynthetic microorganisms.

Phototrophic microorganisms disfigure the surfaces of different types of stone. Stone structure is damaged by the activity of photoautotrophic and other microorganisms. However, to date few, investigations have been undertaken into the relationship between microorganisms and the properties of different types of marble. In this study, biological activity of photoautotrophic microorganisms on three types of marble (Yatagan White, Giallo Anticato and Afyon White) was investigated under laboratory conditions over a short period of time. The three types of marble supported the growth of phototrophic microbial communities on their outer and inner layers, turning their original colour from white to a yellowish green colour. The porosity of the marble types facilitated filamentous microbial growth in the presence of water. Scanning electron microscope analysis revealed the accumulation of aggregates such as small spherical, fibrillar, calcified globular bodies on the inner surfaces of the marbles. This suggests that the microscopic characteristics of particular marble types may stimulate the growth of certain types of microorganisms.

Glycogen production for biofuels by the euryhaline cyanobacteria Synechococcus sp. strain PCC 7002 from an oceanic environment

BackgroundOxygenic photosynthetic microorganisms such as cyanobacteria and microalgae have attracted attention as an alternative carbon source for the next generation of biofuels. Glycogen abundantly accumulated in cyanobacteria is a promising feedstock which can be converted to ethanol through saccharification and fermentation processes. In addition, the utilization of marine cyanobacteria as a glycogen producer can eliminate the need for a freshwater supply. Synechococcus sp. strain PCC 7002 is a fast-growing marine coastal euryhaline cyanobacteria, however, the glycogen yield has not yet been determined. In the present study, the effects of light intensity, CO2 concentration, and salinity on the cell growth and glycogen content were investigated in order to maximize glycogen production in Synechococcus sp. strain PCC 7002.ResultsThe optimal culture conditions for glycogen production in Synechococcus sp. strain PCC 7002 were investigated. The maximum glycogen production of 3.5 g L−1 for 7 days (a glycogen productivity of 0.5 g L−1 d−1) was obtained under a high light intensity, a high CO2 level, and a nitrogen-depleted condition in brackish water. The glycogen production performance in Synechococcus sp. strain PCC 7002 was the best ever reported in the α-polyglucan (glycogen or starch) production of cyanobacteria and microalgae. In addition, the robustness of glycogen production in Synechococcus sp. strain PCC 7002 to salinity was evaluated in seawater and freshwater. The peak of glycogen production of Synechococcus sp. strain PCC 7002 in seawater and freshwater were 3.0 and 1.8 g L−1 in 7 days, respectively. Glycogen production in Synechococcus sp. strain PCC 7002 maintained the same level in seawater and half of the level in freshwater compared with the optimal result obtained in brackish water.ConclusionsWe conclude that Synechococcus sp. strain PCC 7002 has high glycogen production activity and glycogen can be provided from coastal water accompanied by a fluctuation of salinity. This work supports Synechococcus sp. strain PCC 7002 as a promising carbohydrate source for biofuel production.

Ammonium tolerance in the cyanobacterium Synechocystis sp. strain PCC 6803 and the role of the psbA multigene family

Ammonium is one of the major nutrients for plants, and a ubiquitous intermediate in plant metabolism, but it is also known to be toxic to many organisms, in particular to plants and oxygenic photosynthetic microorganisms. Although previous studies revealed a link between ammonium toxicity and photodamage in cyanobacteria under in vivo conditions, ammonium-induced photodamage of photosystem II (PSII) has not yet been investigated with isolated thylakoid membranes. We show here that ammonium directly accelerated photodamage of PSII in Synechocystis sp. strain PCC6803, rather than affecting the repair of photodamaged PSII. Using isolated thylakoid membranes, it could be demonstrated that ammonium-induced photodamage of PSII primarily occurred at the oxygen evolution complex, which has a known binding site for ammonium. Wild-type Synechocystis PCC6803 cells can tolerate relatively high concentrations of ammonium because of efficient PSII repair. Ammonium tolerance requires all three psbA genes since mutants of any of the three single psbA genes are more sensitive to ammonium than wild-type cells. Even the poorly expressed psbA1 gene, whose expression was studied in some detail, plays a detectable role in ammonium tolerance.

Direct conversion of Spirulina to ethanol without pretreatment or enzymatic hydrolysis processes

Oxygenic photosynthetic microorganisms such as cyanobacteria and microalgae have attracted attention as feedstocks for next-generation biofuels. To date, however, there have been no reports on efficient bioethanol production from cyanobacterial glycogen by yeast fermentation. Additionally, multiple pretreatment and enzymatic hydrolysis steps of polysaccharides are required for conventional ethanol production from agricultural crops and microalgae. Here, we investigate direct ethanol production from Arthrospira (Spirulina) platensis, a fast-growing halophilic cyanobacterium that accumulates large amounts of glycogen, using lysozyme and a recombinant amylase-expressing yeast strain to eliminate the need for biomass pretreatment and amylase hydrolysis. In the direct conversion process from A. platensis to ethanol, 6.5 g L−1 (ethanol productivity of 1.08 g per L per day) of ethanol was produced. The total ethanol yield based on glycogen consumption was 86% of theoretical yield, which to our knowledge, is the highest yield of bioethanol from an oxygenic photosynthetic microorganism. The present findings indicate that A. platensis is a remarkable carbohydrate feedstock in the form of glycogen, which is a promising material for the production of bioethanol and various other commercially valuable chemicals.

Genome-Scale Modeling of Light-Driven Reductant Partitioning and Carbon Fluxes in Diazotrophic Unicellular Cyanobacterium Cyanothece sp. ATCC 51142

Genome-scale metabolic models have proven useful for answering fundamental questions about metabolic capabilities of a variety of microorganisms, as well as informing their metabolic engineering. However, only a few models are available for oxygenic photosynthetic microorganisms, particularly in cyanobacteria in which photosynthetic and respiratory electron transport chains (ETC) share components. We addressed the complexity of cyanobacterial ETC by developing a genome-scale model for the diazotrophic cyanobacterium, Cyanothece sp. ATCC 51142. The resulting metabolic reconstruction, iCce806, consists of 806 genes associated with 667 metabolic reactions and includes a detailed representation of the ETC and a biomass equation based on experimental measurements. Both computational and experimental approaches were used to investigate light-driven metabolism in Cyanothece sp. ATCC 51142, with a particular focus on reductant production and partitioning within the ETC. The simulation results suggest that growth and metabolic flux distributions are substantially impacted by the relative amounts of light going into the individual photosystems. When growth is limited by the flux through photosystem I, terminal respiratory oxidases are predicted to be an important mechanism for removing excess reductant. Similarly, under photosystem II flux limitation, excess electron carriers must be removed via cyclic electron transport. Furthermore, in silico calculations were in good quantitative agreement with the measured growth rates whereas predictions of reaction usage were qualitatively consistent with protein and mRNA expression data, which we used to further improve the resolution of intracellular flux values.

Metabolic engineering of algae for fourth generation biofuels production

The ecological footprint and economic performance of the current suite of biofuel production methods make them insufficient to displace fossil fuels and reduce their impact on the inventory of Green House Gas (GHG) in the global atmosphere. Algae metabolic engineering forms the basis for 4th generation biofuel production which can meet this need. The first generation biofuels are known to be made from agricultural products such as corn or sugarcane. The second generation biofuels use all forms of (lingo)cellulosic biomass. The third and fourth generation of biofuel production involves “algae-to-biofuels” technology: the former is basically processing of algae biomass for biofuel production, while the latter is about metabolic engineering of algae for producing biofuels from oxygenic photosynthetic microorganisms. Our review focuses on the research achievement of metabolic engineering of algae for biofuel production. It is concluded that 4th generation biofuel production has introduced the “cell factory” concept in this field, and shifted the research paradigm. There still exists several technical bottlenecks in algae biofuel research and development, which can only be solved by the use of post-genome tools on these photosynthetic organisms.

Hydrogen photoproduction by use of photosynthetic organisms and biomimetic systems

Hydrogen can be important clean fuel for future. Among different technologies for hydrogen production, oxygenic natural and artificial photosyntheses using direct photochemistry in synthetic complexes have a great potential to produce hydrogen, since both use clean and cheap sources: water and solar energy. Artificial photosynthesis is one way to produce hydrogen from water using sunlight by employing biomimetic complexes. However, splitting of water into protons and oxygen is energetically demanding and chemically difficult. In oxygenic photosynthetic microorganisms such as algae and cyanobacteria, water is split into electrons and protons, which during primary photosynthetic process are redirected by photosynthetic electron transport chain, and ferredoxin, to the hydrogen-producing enzymes hydrogenase or nitrogenase. By these enzymes, e- and H+ recombine and form gaseous hydrogen. Biohydrogen activity of hydrogenase can be very high but it is extremely sensitive to photosynthetic O2. In contrast, nitrogenase is insensitive to O2, but has lower activity. At the moment, the efficiency of biohydrogen production is low. However, theoretical expectations suggest that the rates of photon conversion efficiency for H2 bioproduction can be high enough (>10%). Our review examines the main pathways of H2 photoproduction by using of photosynthetic organisms and biomimetic photosynthetic systems.

Ionizing radiation impacts photochemical quantum yield and oxygen evolution activity of Photosystem II in photosynthetic microorganisms

Purpose: Long-term space exploration requires biological life support systems capable of coping with the deleterious space environment. The use of oxygenic photosynthetic microorganisms represents an intriguing topic in this context, mainly from the point of view of food and O2 production. The aim of the present study was to assess the effects of space ionizing radiation exposure on the photosynthetic activity of various microorganisms.Materials and methods: Ground-based irradiation experiments were performed using fast neutrons and gamma rays on microorganisms maintained at various light conditions. A stratospheric balloon and a European Space Agency (ESA) flight facility were used to deliver organisms to space at the altitude of 38 and 300 km, respectively. During the balloon flight, the fluorescence activity of the organisms was real-time monitored by means of a special biosensor.Results: The quantum yield of Photosystem II (PSII), measured directly in flight, varied among the microorganisms depending on the light conditions. Darkness and irradiation of cells at 120 and 180 μmol m−2 s−1 enhanced the radiation-induced inhibition of photosynthetic activity, while exposure to weaker light irradiance of 20 and 70 μmol m−2 s−1 protected the cells against damage. Cell permanence in space reduced the photosynthetic growth while the oxygen evolution capacity of the cells after the flight was enhanced.Conclusions: A potential role of PSII in capturing and utilizing ionizing radiation energy is postulated.

Use of Superoxide as an Electron Shuttle for Iron Acquisition by the Marine Cyanobacterium Lyngbya majuscula

Reduction of iron from the ferric state to the ferrous state is one strategy employed by microorganisms in nearneutral environments to increase its biological availability. In recent years, the existence of mobile reducing agents produced bymicroorganismsto promote iron reduction, known as electron shuttles, has been demonstrated. Production of electron shuttles has been shown for several organisms, employing a variety of mostly organic molecules as the electron carrier. Here we show that the coastal cyanobacterium Lyngbya majuscula produces iron-reducing superoxide radicals (02*-) and that this facilitates increased iron uptake. We suggest that superoxide is a useful electron shuttle because it reacts rapidly and almost indiscriminately with Fe(lll)-organic complexes and its precursor, dissolved oxygen, is ubiquitous in the photic zone. We further suggest that, for these reasons, the generation of superoxide by marine oxygenic photosynthetic microorganisms and its use in facilitating iron uptake may be a reasonably widespread process.

Comentary impacts into ocean: thermochemical equilibrium calculations of high-temperature O 2 generation on the early earth

The early Earth's atmosphere apparently differed from the present atmosphere mainly by its lack of free O 2 , a condition that is believed to have been indispensable for the origin of early anaerobic life forms. One of thecentral problems in Earth science is to explain the apparent transition from a primitive atmosphere (free of O 2 ) to the present atmosphere which holds 21 % of the gas. Theoretical models suggest that the initial form of O 2 in Earth's atmosphere may have been H 2 O, which was converted into atmospheric O 2 mainly through photosynthesis. I have investigated an alternative (abiotic) method for converting H 2 O to O 2 : a high temperature shock generated during the cometary impact into ocean (or on land). Calculations show that 1 % of the present atmospheric level (PAL) of O 2 could have resulted from an icy 29.2 km comet impacting the early (pre-oxygenic) Earth with a velocity between 18 and 37 km s - 1 . It is suggested that an abrupt supply (10 - 2 % of PAL) of free O 2 by the comet would be sufficient to make way for the appearance of oxygenic photosynthetic microorganisms.

Cyanobacteria associated with hot spring travertines

Cyanobacteria are the major oxygenic photosynthetic microorganisms of hot spring travertines. This review describes the known cyanobacterium flora of travertine-depositing waters > 37 °C. The communities develop either upon (epilithon) or within (endolithon) the travertine surface, where they may influence the travertine fabric by providing nucleation sites for calcium carbonate. Mat photosynthesis locally increases the amount of travertine deposited, but the importance of photosynthesis in travertine deposition is rarely significant because the main loss of CO2is by atmospheric evasion of the hot, CO2-rich waters. The Oscillatoriales are the most important group of cyanobacteria in terms of their biomass, but the taxonomy of the order is in a state of chaos. Molecular methods will ultimately disclose the true affinities of the organisms but, at present, the use of form-taxa based upon morphological characters provides a practical alternative. Identification of fossilized cyanobacteria is problematic since few of the key characters survive as fossils. The known cyanobacterium flora is tabulated and an analytical key provided to identify the form-taxa of hot spring travertines.

Massive light-dependent cycling of inorganic carbon between oxygenic photosynthetic microorganisms and their surroundings.

Membrane inlet mass spectrometry indicated massive light-dependent cycling of inorganic carbon between the medium and the cells of various phytoplankton species representing the main groups of aquatic primary producers. These included diatoms, symbiotic and free living dinoflagellates, a coccolithophorid, a green alga and filamentous and single cell cyanobacteria. These organisms could maintain an ambient CO(2) concentration substantially above or below that expected at chemical equilibrium with HCO(3) (-). The coccolithophorid Emiliania huxleyi shifted from net CO(2) uptake to net CO(2) efflux with rising light intensity. Differing responses of CO(2) uptake and CO(2) fixation to changing light intensity supported the notion that these two processes are not compulsorily linked. Simultaneous measurements of CO(2) and O(2) exchange and of the fluorescence parameters in Synechococcus sp. strain PCC 7942, showed that CO(2) uptake can serve as a sensitive probe of the energy status of the photosynthetic reaction centers. However, during transitions in light intensity, changes in CO(2) uptake did not accord with those expected from fluorescence change. Quantification of the net fluxes of CO(2), HCO(3) (-) and of photosynthesis at steady-state revealed that substantial HCO(3) (-) efflux accompanied CO(2) uptake and fixation in the case of 'CO(2) users'. On the other hand, 'HCO(3) (-) users' were characterized by a rate of net CO(2) uptake below that of CO(2) fixation. The results support the notion that entities associated with the CCM function not only in raising the CO(2) concentration at the site of Rubisco; they may also serve as a means of diminishing photodynamic damage by dissipating excess light energy.
