The effect of smoking on amylase enzyme activity in saliva

Smoking has become a common habit in Indonesia. If done regularly, smoking is known to cause problems in the oral cavity. When smoking, cigarette smoke will enter the oral cavity and affect the activity of the salivary amylase enzyme. The journal search used in this review was conducted on several databases, such as PubMed, ScienceDirect, and Google with a range of years between 2013 and 2023. A total of 14 studies from various global populations were included in this review. Thirteen studies showed an association between smoking and alpha-amylase enzyme activity, while one study did not show an association between smoking and amylase enzyme. Of the 13 studies that showed an association, ten studies showed a negative association, while the remaining three studies showed a positive association. The effect of cigarettes on amylase activity in saliva may arise due to several factors, including the toxic effects of cigarettes on the salivary glands, mechanical and chemical stimulation of cigarettes on the mucosa, smoking period, oxidative stress index, and salivary pH.

Association of NOX5 Expression with Sperm Activity and Motility in Pathospermic Infertile Men

Background: The newest NOX isoform, NOX5, has been found in mammalian spermatozoa. Many physiological and pathological situations in spermatozoa are mediated by reactive oxygen species (ROS). NOX5 is the main source of ROS in spermatozoa. Our purpose was to investigate the changes in NOX5 expression and the effect of NOX5 expression on sperm motility, chromatin integrity, and oxidative status in oligoasthenozoospermic compared to normozoospermic men. Methods: Semen samples were collected from 30 normozoospermic (NS) and 30 oligoasthenozoospermic (OAS) men. NOX5 protein expression in sperm samples was evaluated by immunohistochemistry and western blot. Oxidative stress status was evaluated by total antioxidant capacity (TAC), total oxidant capacity (TOC), and oxidative stress index (OSI) parameters. Chromatin integrity in spermatozoa was evaluated by toluidine blue staining. Results: NOX5 expression levels were significantly higher in OAS group than in NS group (p<0.001). In addition, chromatin integrity was significantly higher in the OAS group in comparison to NS group (p<0.001). TAC levels were higher in the NS group, but OSI and TOC levels were significantly higher in OAS group (p<0.001). It was found that NOX5 protein expression was positively correlated with oxidative stress and chromatin integrity and negatively correlated with motility (p<0.01). Conclusion: These results suggest that overexpression of NOX5 may be the source of excessive ROS production and oxidative stress injuries in oligoasthenozoospermic men. Considering that NOX5 expression is positively correlated with oxidative stress and chromatin integrity but negatively correlated with motility, it can be considered a biomarker to be used in assisted reproductive procedures.

Effect of different light conditions on the growth response of sugar beet to NaCl

ABSTRACT It is known that sugar beet, derived from a halophytic wild ancestor, is highly salt-tolerant and that moderate concentrations of NaCl enhance its growth. However, the mechanisms of this improved growth performance by NaCl are not fully understood. In this study, we investigated the effects of NaCl on sugar beet growth under different light conditions and examined the relationship between light-induced oxidative stress and the beneficial effects of NaCl. Sugar beet seedlings in pots filled with fertilized soil were grown with different light intensities (low: 150–250 μmol m−2s−1 or high: 600–1,000 μmol m−2s−1) and NaCl treatments (irrigation with water with or without 50 mM NaCl) for 10 days. The enhanced sugar beet growth by NaCl application was only observed in plants with high light intensity treatments. Moreover, high light intensity treatment increased the shoot concentration of malondialdehyde, an indicator of oxidative stress, while NaCl application decreased it. While there was a positive correlation between Na concentration and the compatible solute glycine betaine in the shoots and roots under both light intensities, a significant negative correlation between the concentrations of glycine betaine and malondialdehyde was found only in the shoots under high light intensity. Because it is known that glycine betaine activates antioxidant enzyme activities and reduces oxidative stress responses, glycine betaine synthesized in response to NaCl accumulation may have reduced the oxidative stress caused by high light intensity in the shoots, which may have caused the enhanced growth in sugar beet. Furthermore, the growth promotion by NaCl accumulation was accompanied by a significant decrease in NO3–N concentration, which suggested that NaCl application affects nitrogen metabolism in sugar beet.

A comparison of cognitive decline in aged mice and mice treated with aftin-4.

Dementia, especially Alzheimer's disease, presents a major clinical challenge, and researchers are still searching for an optimal animal model. To address this gap, we compared male and ovariectomized female C57BL/6 mice treated with 30mg/kg aftin-4, which induces neurodegeneration, with naturally aged (15-16 months old) mice not treated with aftin-4. We performed a series of behavioral tests; measured postmortem plasma β-amyloid levels (Aβ1-40 and Aβ1-42) and the levels of the oxidative stress indicators glutathione peroxidase (GPx), superoxide dismutase (SOD) and malondialdehyde (MDA); and evaluated astrocytic reactivity in the brain using glial fibrillary acid protein (GFAP) levels. Our results revealed no behavioral changes in the aged or aftin-4-treated mice compared with the control mice. Aftin-4 mice presented lower brain MDA levels and no detectable changes in plasma Aβ levels. In general, female mice had higher GPx and SOD levels and lower Aβ1-42 levels than male mice did. In contrast, aged and aftin-4-treated male mice presented elevated levels of GFAP, indicating astrocyte damage. Our results could not confirm that either aftin-4-treated or aged mice are reliable models for dementia. However, the observed molecular changes suggest that male animals may be more susceptible to oxidative stress and brain damage than females are. This study demonstrates the complexity of modeling dementia in animals and the importance of future studies in this area.

Berberine Mitigates Sepsis-Associated Acute Kidney Injury in Aged Rats by Preserving Mitochondrial Integrity and Inhibiting TLR4/NF-κB and NLRP3 Inflammasome Activations

Sepsis-associated acute kidney injury (SA-AKI) presents a severe challenge in the elderly due to increasing incidence, high mortality, and the lack of specific effective treatments. Exploring novel and secure preventive and/or therapeutic approaches is critical and urgent. Berberine (BBR), an isoquinoline alkaloid with anti-inflammatory, antioxidant, and immunomodulatory properties, has shown beneficial effects in various kidney diseases. This study examined whether BBR could protect against SA-AKI in aged rats. Sepsis was induced in 26-month-old male Wistar rats by cecal ligation and puncture (CLP), either with or without BBR pretreatment. CLP induction led to SA-AKI, as indicated by elevated serum levels of malondialdehyde, tumor necrosis factor-alpha, urea nitrogen, creatinine, and neutrophil gelatinase-associated lipocalin (NGAL), along with histopathological features of kidney damage. Key indicators of kidney oxidative stress, mitochondrial dysfunction, apoptosis, and activations of the Toll-like receptor 4/nuclear factor-kappa B (TLR4/NF-κB) signaling, including the nucleotide-binding domain, leucine-rich-containing family, and pyrin domain-containing-3 (NLRP3) inflammasome pathway, were also elevated following CLP induction. BBR pretreatment substantially mitigated these adverse effects, suggesting that it protects against SA-AKI in aged rats by reducing oxidative stress, preserving mitochondrial integrity, and inhibiting key inflammatory pathways. These findings highlight the potential of BBR as a therapeutic agent for managing SA-AKI in elderly populations.

Changing dynamics in daily rhythms of oxidative stress indicators in SCN and extra-SCN brain regions with aging in male Wistar rats.

The suprachiasmatic nucleus (SCN) in the hypothalamus regulates circadian timing system (CTS) by co-ordinating peripheral tissue clocks and extra-SCN oscillators in the brain. Aging disrupts the CTS, impairing physiological functions and reducing antioxidant defences, which contribute to neurodegeneration. The brain is vulnerable to oxidative damage due to its high metabolic activity, oxygen consumption, and levels of iron and lipids. Antioxidant enzymes, such as catalase (CAT), glutathione S-transferase (GST), superoxide dismutase (SOD), and lipid peroxidation (LPO), help against oxidative damage. In this study, we examined the temporal patterns of these antioxidant stress indicators in the SCN and extra-SCN brain regions (frontal cortex, cerebellum, and hippocampus) at various time points in male Wistar rats 3, 12, and 24months. The rhythmicity of GST and LPO levels persisted across brain regions with aging, while CAT rhythmicity was lost in the SCN and hippocampus of older rats. SOD rhythmicity persisted in cortex, cerebellum, and hippocampus but was lost in the SCN. The daily rhythm parameters of CAT were affected most significantly, followed by SOD, GST, and LPO. Our findings demonstrate that aging leads to desynchronization of oxidative stress indicators potentially contributing to neurodegeneration and circadian dysfunction with varying effects across different brain tissues.

Oligomeric proanthocyanidin ameliorates sepsis-associated renal tubular injury: involvement of oxidative stress, inflammation, PI3K/AKT and NFκB signaling pathways.

Sepsis is a potentially fatal infectious disease that easily causes shock and numerous organ failures. The kidney is one of the most susceptible to injury. Early intervention and renal protection significantly minimize patient mortality. Oligomeric proanthocyanidin (OPC), a naturally occurring plant compound, has a high potential for renal protection. This study was aimed at exploring the potential renoprotective role of OPC in sepsis-related renal tubular injury. C57/B6 mice were intraperitoneally injected with lipopolysaccharide (LPS) to simulate sepsis-related acute kidney injury in vivo. Renal function and pathology were assessed. RNA sequencing examined OPC mechanisms against LPS-induced renal injury. Oxidative stress indicators and inflammatory cytokines in blood serum and renal tissues were evaluated. In vitro, MTT assays assess cell viability. Apoptosis cells were detected using Hoechst 33342 and propidium iodide staining. Western blot assessed PI3K/AKT and NFκB signaling pathway proteins. OPC reduced LPS-induced renal tubular injury, improved renal functions and pathological changes, restored glutathione content, superoxide dismutase activity, and catalase activity, inhibited malondialdehyde overproduction, and suppressed LPS-induced overproduction of pro-inflammatory cytokines and the decline of anti-inflammatory cytokines. OPC attenuated LPS-induced cell morphological injury, reduced cell viability loss, and recovered the changes in proteins involved in PI3K/AKT and NFκB signaling pathways in MTEC cells. OPC protects against LPSinduced renal tubular injury by counteracting oxidative stress, inhibiting inflammatory responses, activating the PI3K/AKT signaling pathway, and inhibiting the NFκB signaling pathway. It may provide a viable solution to lessen renal injury in patients with sepsis.

Capsaicin Protects Against Nigrostriatal Neurodegeneration Induced by Rotenone

Capsaicin, the principal pungent ingredient of hot pepper exerts neuroprotective effects. In this study, the effect of capsaicin on rotenone-induced Parkinson’s disease in mice was investigated. Mice were given subcutaneous rotenone injections (1.5 mg/kg, every other day) and at the same time treated with the vehicle, L-dopa (25 mg/kg) or capsaicin at doses of 0.5 or 1.0 mg/kg orally once a day for two weeks. Biochemical indices of oxidative stress, malondialdehyde, reduced glutathione and nitric oxide were determined in brain tissue and histopathological study of the brain was done. Behavioral tests included stair, wire hanging and wood walking tests. Results showed that rotenone treatment led to significant increases in brain malondialdehyde and nitric oxide contents parallel with marked depletion of reduced glutathione. Rotenone induced degeneration of pigmented neurons in substantia nigra and of cerebral cortex and hippocampus neurons. Rotenone impaired neuromuscular strength, motor balance and coordination. Treatment with capsaicin significantly ameliorated the neuronal degeneration caused by rotenone and improved motor function. Capsaicin alleviated the increase in lipid peroxidation (malondialdehyde) and nitric oxide and prevented the depletion of reduced glutathione in brain of rotenone-treated animals. These data indicate that capsaicin protects against rotenone-induced neuronal damage and this involves decreased level of oxidative stress. Capsaicin therefore might prevent cell death in the brain of Parkinson’s disease patients.

Effects of oxidative stress on the changes of viability of Paeonia lactiflora seeds with different water content before and after cryopreservation.

Cryopreservation in liquid nitrogen (LN) is an efficient long - term seed preservation strategy and water content is one of the main factors affecting seed viability after cryopreservation. In this study, Paeonia lactiflora seeds with varying water content were used as materials to determine changes in the antioxidant system before and after cryopreservation. When the water content of P. lactiflora seeds was 15.96% - 10.12%, the viability of P. lactiflora seeds decreased significantly compared with that of the seeds without cryopreservation, but at the water content of 8.14% - 7.56% there were no significant differences. However, when the water content of P. lactiflora seeds decreased to 6.01% - 5.19% the viability was slightly increased compared to the seeds without cryopreservation. The content of superoxide anion (O2-), hydroxyl radical (·OH), hydrogen peroxide (H2O2), malondialdehyde (MDA) and protein carbonyl group (PCO) of seeds with high water content (15.96% - 10.12%) were significantly increased after cryopreservation. However, superoxide anion (O2-), hydroxyl radical (·OH) and hydrogen peroxide (H2O2) did not change significantly in seeds with low water content (6.01% - 5.19%) after cryopreservation, while oxidative stress indexes MDA and PCO decreased. These three substances were significantly negatively correlated with seed viability. In terms of antioxidant substances, the contents of catalase (CAT), ascorbic acid (AsA) and glutathione (GSH) decreased and the activities of glutathione reductase (GR) and dehydroascorbate reductase (DHAR) increased during the cryopreserved process of seeds with varying water content. These changes were significantly correlated with ROS content and the changes of MDA and PCO, among which AsA content, GSH content and CAT activity were positively correlated with seed viability. The changes of GR and DHAR activity were negatively correlated with seed viability. In summary, when the water content of the seeds ranged from 8.14% - 5.19%, ROS content did not increase significantly after cryopreservation compared with that of before preservation. The changes of MDA and PCO contents before and after cryopreservation, it was inferred that no obvious oxidative damage occurred in seeds, so the viability of seeds did not decrease compared with that of before cryopreservation. Therefore, the optimum water content of P. lactiflora seeds for cryopreservation is 8.14% - 5.19%.

Selenium mitigates vanadium toxicity through enhanced nutrition, photosynthesis, and antioxidant defense in rice (Oryza sativa L.) seedlings.

In the current industrial scenario, vanadium (V) as a metal is of great importance but poses a major threat to the ecosystem. In the present study, the effect of a toxic concentration of V, i.e., 10 µM in the soil on growth, photosynthetic pigments, gas exchange characteristics, oxidative stress biomarkers, antioxidants machinery (enzymatic and non-enzymatic antioxidants), ions uptake, proline metabolism, and V uptake in different parts of the plant was investigated with and without the exogenous application of selenium (Se) i.e., 5 µM in V-stressed rice (Oryza sativa L.). Our results depicted that V addition to the soil significantly (P < 0.05) decreased plant growth and biomass, gas exchange attributes, and minerals uptake by O. sativa as compared to the plants grown without the addition of V. However, V toxicity boosted the production of reactive oxygen species (ROS) by increasing the contents of malondialdehyde (MDA), which is the indication of oxidative stress in O. sativa and was also manifested by hydrogen peroxide (H2O2) contents to the membrane-bounded organelles. Although activities of various antioxidative enzymes like superoxidase dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX) and their gene expression Fe-SOD, POD, CAT, and APX and also non-enzymatic antioxidants like phenolic, flavonoid, and ascorbic acid, anthocyanin contents and also the proline metabolism i.e., proline, pyrroline5-carboxylate, pyrroline-5-carboxylate reductase, and pyrroline-5-carboxylate dehydrogenase were increased due to V stress. Although results also illustrated that the application of Se also decreased V toxicity in O. sativa seedlings by increasing antioxidant capacity and, thus, improved the plant growth and biomass, photosynthetic pigments, gas exchange characteristics, and decreased oxidative stress in the O. sativa seedlings, compared to those plants which were not artificially supplied by Se. Research findings, therefore, suggested that the Se application can ameliorate V toxicity in O. sativa seedlings and result in improved plant growth and composition under metal stress as depicted by balanced exudation of nutrient effluxes. This study provides novel insights into the role of selenium in mitigating vanadium-induced oxidative stress in rice, thereby offering a promising approach to enhancing crop resilience in metal-contaminated soils and advancing sustainable agricultural practices.

The impact of cooling and Moringa supplementation on oxidative stress in serum and milk, including milk cytokines, in heat stressed lactating sows and their litters

Abstract Heat stress (HS) poses a significant challenge to the United States swine industry. Sows and their piglets are particularly vulnerable to heat stress, as the peri-parturient phase is characterized by heightened metabolism and increased oxidative stress and inflammation. The study examined the effects of using conductive electronic cooling pads (ECP) and dietary supplementation with 4% Moringa (M) leaf powder on controlling oxidative stress and inflammation caused by heat stress (HS) in sows and their piglets. Forty-eight late gestation sows were assigned to four treatment groups: HS fed corn-soybean meal (HS+CS), ECP fed corn-soybean meal (ECP+CS), HS+M, and ECP+M. Blood was collected from sows on gestation (G) day 112, and lactation (L) day 14 and L20, and from piglets (2 male and 2 female) in each litter on postnatal (PN) day 1 and PN20. Colostrum was collected within 2h of birth of first neonate, and mature milk was collected on L14. Piglet fecal samples were collected on PN14 to measure calprotectin concentration as a marker of intestinal inflammation. Biological antioxidant potential (BAP), derivatives of reactive oxygen metabolites (dROMs) and oxidative stress index (OSi) were measured in blood and milk samples using Free Radical Elective Evaluator. Milk samples pooled by day of lactation and treatment group, were analyzed using cytokine array. Levels of inflammatory cytokines in colostrum were affected by Moringa supplementation and cooling, but not mature milk. Notably, the anti-inflammatory cytokines interleukin (IL)-10 and IL-1ra were 2.14 and 1.57 Log2 higher in colostrum of HS+M compared to other groups. The oxidative stress index of colostrum was higher (P = 0.0002) than mature milk. Level of BAP in sow serum was greater in ECP+CS and HS+M (P = 0.0291) compared to other groups. Moringa had an overall effect of increasing dROMs (P = 0.0035) and levels of OSi were lowest in ECP+CS (P = 0.0296) sow serum. Treatments did not affect piglet serum oxidative index (P &amp;gt; 0.05) or calprotectin levels (P &amp;gt; 0.05). Findings support further studies to investigate the efficacy of using ECP and Moringa supplementation to mitigate inflammation and oxidative stress imposed by heat stress conditions in lactating sows.

Effect of Butaphosphan and Cyanocobalamin on the Metabolic Profile, Oxidative Stress and Prophylaxis of Ovine Pregnancy Toxemia.

The study was conducted to evaluate the effect of butaphosphan and cyanocobalamin (BC) on the metabolic profile, oxidative stress in Kashmiri merino ewes and its effect on the prophylaxis of Ovine pregnancy toxemia (PT). A total of 48 ewes were randomly divided into three groups, each consisting of 16 ewes. High dose BC (HBC): Ewes were given 0.2 mL BC/kg body weight, subcutaneously (SC) weekly from 6 weeks pre-lambing to lambing. Low dose BC (LBC): Ewes were given 0.1 mL BC/kg body weight, SC weekly from 6 weeks pre-lambing to lambing. No BC (CON): Ewes were given 0.2 mL normal saline/kg body weight, SC. Weekly from six pre-lambing to lambing. A significant effect of treatment was observed on non-esterified fatty acid (NEFA; p = 0.06), beta hydroxybutyric acid (β-HBA, p = 0.008), fructosamine (p = 0.034) and oxidative stress indices (OSI; p = 0.026) with less NEFA, β-HBA, OSI and more fructosamine levels in HBC followed by LBC. The BC did not affect glucose, cholesterol, blood urea nitrogen, creatinine, total antioxidant capacity, calcium and phosphorus however a significant difference was observed along the sampling time in all the three groups. Plasma triglyceride, aspartate aminotransferase and total oxidant status levels showed a significant treatment-time interaction. Taking 0.8 mmol/L β-HBA as the cutoff limit for subclinical PT, only HBC was able to significantly control PT at 3 weeks pre-lambing (OR; 5.57; p = 0.035), at 2 weeks pre-lambing (OR: 9.53; p = 0.007), at 1-week pre-lambing (OR: 6.60; p = 0.017) and at lambing (OR: 5.57; p = 0.035). In conclusion, there was a positive effect of BC treatment on the energy metabolism in pregnant ewes and it helped in the amelioration of oxidative stress and hence can be used in the prophylaxis of PT.

Solasodine Downregulates ABCB1 Overexpression in Multidrug Resistant Cancer Cells Via Inhibiting Nrf2/Keap1 Signaling Pathway.

Multidrug-resistant (MDR) cancer cells maintain redox homeostasis to eliminate oxidative stress-mediated cell death. This study explores the effects of solasodine on regulating P-glycoprotein (P-gp) expression through the Nrf2/Keap1 signaling pathway and oxidative stress-induced sensitization of drug-resistant cancer cells to chemotherapeutics. Initially, the oxidative stress indicators such as intracellular ROS generation, the levels of 8-hydroxy-2-deoxyguanosine (8-OHdG) and gamma-H2AX (γ-H2AX) in the KBChR-8-5 drug-resistant cells were measured. Additionally, the protein expression levels of Nuclear factor erythroid 2-related factor 2 (Nrf-2), Kelch-like ECH-associated protein 1 (Keap1), and ATP Binding Cassette Subfamily B Member 1 (ABCB1)/P-gp were measured at various concentrations of solasodine (1, 5, & 10 µM) through immunofluorescence and western blot analysis. The antioxidant activities in the KBChR-8-5 cells were assessed using established protocols. In this investigation, the treatment with solasodine and doxorubicin combination showed a notable increase in intracellular ROS generation in KBChR-8-5 cells. Furthermore, this combination treatment led to enhanced nuclear condensation, elevated levels of 8-OHdG, and increased γ-H2AX foci formation in the KBChR-8-5 cells. Solasodine treatment effectively inhibited the nuclear translocation of Nrf2 and activation of the ABCB1 gene, consequently preventing overexpression of P-gp in KBChR-8-5 cells. Additionally, the combination therapy increased the lipid peroxidation levels while simultaneously reducing the activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and the levels of glutathione (GSH). These results demonstrated that solasodine disrupts redox balance, and overcomes drug resistance by downregulating P-gp via regulating Nrf2/Keap1 signaling pathway in MDR cancer cells.

Determination of selected oxysterol levels, oxidative stress, and macrophage activation indicators in children and adolescents with familial hypercholesterolemia.

Elevated levels of cholesterol in the bloodstream, also referred to as hypercholesterolemia, pose a significant risk for the onset of cardiovascular and cerebrovascular diseases. Oxysterols, cholesterol-derived oxidized compounds that form enzymatically or non-enzymatically, contribute to the development of atherosclerosis and coronary artery disease. This study aimed to examine the critical oxysterol levels in children and adolescents with hypercholesterolemia and explore the correlation between these levels, oxidative stress, and atherosclerosis progression. The study included 20 patients with familial hypercholesterolemia (FH) and 20 healthy individuals aged between 6 and 18 years. Participants were categorized into children (6-9 years) and adolescents (10-18 years). Pediatric and adolescent patients were selected from among subjects with LDL-C ≥ 130mg/dL and diagnosed with heterozygous familial hypercholesterolemia (HeFH) based on the presence of mutations in the LDL receptor (LDL-R) gene. Patients with HeFH who were receiving regular atorvastatin therapy were included in the study. There were no notable differences in catalase and paraoxonase (PON1) activities among the groups. However, the patient group displayed substantially higher levels of malondialdehyde (MDA) (P = 0.0108) and superoxide dismutase (SOD) activity (P = 0.0103). Compared to the healthy control group, serum chitotriosidase (CHITO) activity (P = 0.037) and chitinase 3-like protein 1 (YKL-40) levels (P = 0.0027) were significantly elevated in the patient group. Furthermore, the carotid intima-media thickness (CIMT) measurements of the patient group were significantly greater than those of the healthy group (**P < 0.0001****). The patient group exhibited significantly elevated levels of 5,6-α-epoxycholesterol, Cholestane-3β,5α,6β-triol (C-triol), and 7-ketocholesterol (7-KC), whereas 27-hydroxycholesterol (27-OHC) was significantly more abundant in the healthy group. On the other hand, while 27-OHC/Total cholesterol (Total-C) levels were significantly higher in healthy individuals, the C-Triol/Total-C ratio was significantly higher in patients. No significant differences were found between the groups in terms of 7-KC/Total-C and 5,6-α-epoxycholesterol/Total-C levels. This study highlights the key roles of oxysterols, oxidative stress, and macrophage activation in the development of atherosclerosis in pediatric and adolescent patients with FH. Elevated C-Triol levels in FH patients, alongside increased CIMT, point to early vascular changes despite atorvastatin therapy. In contrast, higher 27-OHC levels in healthy controls suggest differential oxysterol regulation due to cholesterol-lowering treatments in FH patients. C-Triol and 27-OHC/Total-C ratios showed potential as biomarkers to distinguish patients with FH. These findings emphasize the need for therapies targeting oxidative stress and macrophage activation in addition to cholesterol-lowering interventions.

Niosomal gel improves dermal delivery of nimbolide: a promising approach for treatment of psoriasis.

Aim: Psoriasis is a chronic inflammatory skin disorder characterized by the excessive proliferation of keratinocytes, forming thickened skin plaques due to immune-mediated cytokine responses. Delivering drugs through this barrier to target inflamed tissues remains challenging. Nimbolide (NIM), known for its anti-inflammatory and anticancer properties, shows promise in managing psoriasis. However, its efficacy is limited by its inability to penetrate the thickened horny layer of the skin. To overcome this obstacle, we have developed Nim-loaded niosomal (Nio) formulations (NIM Nio) aimed at improving dermal delivery and achieving localized sustained release at psoriasis-affected sites.Methods: The formulation characteristics were assessed using Zeta sizer, Transmission Electron Microscopy (TEM), and High-performance liquid chromatography (HPLC). The optimized formulation was evaluated for anti-psoriatic potential compared to Nim alone by using molecular techniques such as Confocal Microscopy, Flow cytometry, enzyme-linked immunosorbent assay (ELISA), and Western blotting.Results: NIM Nio showed effective penetration into psoriatic skin, resulting in reductions in keratinocyte hyperproliferation, oxidative stress, splenomegaly, inflammatory cytokines, Psoriasis Area and Severity Index (PASI), and rete ridges compared to NIM alone.Conclusion: Our findings underscore the significant anti-proliferative, antioxidant, and anti-inflammatory properties of NIM Nio in psoriasis, demonstrating its potential as a promising therapeutic option for this challenging condition.

The Effects of Different Respiratory Viruses on the Oxidative Stress Marker Levels in an In Vitro Model: A Pilot Study

Respiratory viruses are among the most common causes of human infections. Examining pathological processes linked to respiratory viral infections is essential for diagnosis, treatment strategies, and developing novel therapeutics. Alterations in oxidative stress levels and homeostasis are significant processes associated with respiratory viral infections. The study aimed to compare selected oxidative stress markers: total oxidative status (TOS), total antioxidant capacity (TAC), and the oxidative stress index (OSI) levels and glutathione peroxidase (GPx) and glutathione reductase (GR) activities in normal (MRC5 cell line) and tumor (A549 cell line) lung cells infected with human coronaviruses (HCoV) OC43 and 229E, human adenovirus type 5 (HAdV5), or human rhinovirus A (HRV A). We observed that a respiratory viral infection more significantly affected non-enzymatic oxidative stress markers in a lung adenocarcinoma model (A549 cells), while human lung fibroblasts (MRC-5 cell line) presented changes in enzymatic and non-enzymatic oxidative stress markers. We suggest that further detailed research is required to analyze this phenomenon.

L-Tryptophan improves the sperm quality of threatened Abant trout (Salmo abanticus) against glyphosate toxicity: impact on oxidative stress and DNA damage

The current study focused on the ameliorating impact of TRP (5 mM) on Salmo abanticus spermatozoa exposed to sub‑lethal concentrations of GLY (2.5, 5, and 10 mg/L). Sperm quality parameters, oxidative stress indices, and DNA damage were analyzed in the post‑exposure sperm. Our data reveal that GLY has an adverse effect on motility rate and duration, sperm kinetics parameters, antioxidant status and DNA damage, and triggers antioxidant defense mechanisms at the highest dose (10 mg/L) and it has insufficient therapeutic effect. Overall, GLY poses potential hazards to male reproductive cells and may adversely affect subsequent generations of natural populations.

Effects of Different Packaging Types and Storage Periods on Physicochemical and Antioxidant Properties of Honeys

Preserving the nutritional value of honey without compromising its properties until consumption is crucial. However, different types of honey may respond differently to packaging and storage conditions. This study aimed to assess the effects of different packaging materials (tin cans, light-colored glass jars, and dark-colored glass jars) and storage durations (initial, 6 months, and 12 months) on the physicochemical and antioxidant properties of pine, flower, and thyme honey. Nine samples were collected to conduct study on the three different types of honey. Honey samples were packaged in these materials and analyzed at the start, after 6 months, and after 12 months. The results showed that the moisture, proline content, sugar, total oxidant status (TOS), and oxidative stress index (OSI) levels were unaffected by honey type. Over time, there was a decrease in moisture, pH, proline content, diastase number, sugar, total phenolic content (TPC), total antioxidant status (TAS), and catalase (CAT) activity, alongside an increase in the electrical conductivity, hydroxymethylfurfural (HMF), free acidity, TOS, and OSI levels. The packaging type did not influence the moisture, pH, electrical conductivity, proline content, diastase number, sugar, HMF, TPC, TAS, TOS, OSI, or CAT activity levels. The findings suggest that storing pine, flower, and thyme honey in light- or dark-colored glass jars or tin cans for 12 months does not significantly impact these properties. However, given the reduction in TPC and TAS across all honey types and packaging methods after 12 months, further studies are needed to explore ways to maintain honey quality in this regard.

CORRELATION OF OXIDATIVE STRESS MARKERS IN MULTIPLE BIOFLUIDS OF END-STAGE RENAL DISEASE PATIENTS

A study established that patients with ESRD have high oxidative stress levels that spike the progression of the disease and its complications. Knowledge about the levels of the Oxidative stress biomarkers in various biofluids including Urine, serum, and saliva can explain the oxidative condition and probable diagnostic accuracy of these markers in ESRD patients. A descriptive cross-sectional exploratory research design was used to perform a comparison of 50 ESRD patients with 50 healthy subjects. On demographic, clinical, and hematological parameters data were obtained. Blood MDA, 8-OHdG, and saliva TAC were determined as the indicators of oxidative stress. Multivariate analysis was conducted to compare the levels of these markers in the biofluids and probable neurological disorders, and the participants’ demographic and clinical data were evaluated. Demographic profiling revealed that ESRD patients’ mean BMI was 26.3 ± 3.9 kg/m², compared to controls 24.8 ± 3.4 km/m², p = 0.032; more of them being smokers’ p = 0.045. ESRD patient’s general hematologic profile showed that their mean hemoglobin level was 10.2 ± 1.5 g/dL and the mean hematocrit was 32.5 ± 4.3 % and the difference was significant when compared with the control group (p&lt;0.05). ESRD was associated with an increase in WMBC, percentage of neutrophils, and CRP level compared with the control group (p&lt;0.05). Acute phase marker study indicates that the MDA level was significantly higher in serum (5.6 ± 1.1 nmol/L) than in urine (3.2 ± 0.8 nmol/L) and saliva (2.8 ± 0.7 nmol/L, p=0.016). The levels of 8-OHdG were significantly higher in serum (10.3 ± 2.1 ng/mL) than in urine and saliva (p=0.027). Salivary TAC was the least suggesting reduced antioxidant defense (p&lt;0.05). The study revealed that ESRD patients have increased levels in the markers of oxidative stress in urine and serum samples, as well as saliva compared to healthy individuals. The studies have identified serum as the most representative biofluid for evaluating oxidative stress in ESRD and therefore assured the practical application of the results for the identification of the best candidates for early interventions.

Assessing the effects of cadmium on antioxidant enzymes and histological structures in rainbow trout liver and kidney

Cadmium contamination in aquatic environments poses severe risks to aquatic organisms, particularly fish, where cadmium accumulation in tissues can lead to compromised organ functionality and reproductive issues. The present study aimed to assess the effects of cadmium (Cd) exposure on key biomarkers of oxidative stress, DNA damage, apoptosis, and enzyme activity in the liver and kidney tissues of rainbow trout (Oncorhynchus mykiss). Specifically, the study measured 8-hydroxy-2-deoxyguanosine (8-OHdG) levels, caspase-3 activation, acetylcholinesterase (AChE) activity, and oxidative stress indicators (ONOO−, MDA, GSH, SOD, and CAT) following exposure to three Cd concentrations (1, 3, and 5 mg/L) over three time points (24, 48, and 96 h). Tissue samples were collected post-exposure, and the analysis revealed a significant decrease in MDA levels in both tissues. GSH concentrations declined with prolonged exposure, while SOD activity increased, indicating a response to oxidative stress, contrasted by a reduction in CAT activity. An initial increase in ONOO− levels was observed at 24 h, followed by a subsequent decrease at the 48 and 96 h marks. These results suggest that cadmium induces oxidative stress in the liver and kidney tissues of fish. Cadmium exposure also significantly elevated 8-OHdG levels, signaling DNA damage, and increased caspase-3 activity, indicative of apoptosis, across all doses and time points (p < 0.05). The histological examination of liver and kidney showed tissue injury. Additionally, a negative correlation between AChE activity and exposure duration was noted, with prolonged exposure resulting in substantial AChE inhibition. Given the role of AChE in behavior regulation, these findings underscore the importance of exploring time-dependent, tissue-specific changes in AChE activity to further elucidate the mechanisms underlying cadmium-induced behavioral abnormalities.
