Toxicological effects of meloxicam on physiological and antioxidant status of common carp (Cyprinus carpio).

Fish in aquatic environments are end consumers of the food chain and are widely used for the evolution effects of environmental pollution and their interactions in aquatic ecosystem. In the present study, common carp (Cyprinus carpio) fingerlings were selected to assess the potential risk and aquatic toxicity of meloxicam as a non-steroidal anti-inflammatory and a commonly used pharmaceutical drug. In order to evaluate meloxicam toxicological effect on haematological, antioxidant status, enzymological and histological parameters, based on its LC50 24h acute toxicity (10.05mgL-1 ), fish fingerlings were exposed to four doses of meloxicam including; 0 (control), 0.1 (low), 1 (medium) and 2mgL-1 (high) under static bioassay method for 28 days. The results showed that sublethal doses of meloxicam significantly decreased alanine aminotransferase, alkaline phosphatase, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) levels in comparison with the control group after 28 days (p<0.05). However, red blood cell, haematocrit, haemoglobin and malondialdehyde values in fish exposed to meloxicam significantly increased alongside its concentration (p<0.05) more than the control group after 28 days. SOD, CAT and GPX mRNA expression levels in gill, liver, kidney and brain organ of fish under meloxicam treatment were significantly down-regulated compared to the control group (p<0.05). Histopathological assessment showed the increased vacuolation in hepatocytes in liver of fish exposure to medium and high doses of meloxicam. In conclusion, meloxicam induces oxidative stress in common carp which results a disruption of physiological and health status of this species based on our current findings.

Short-term exposure to dexamethasone at environmentally relevant concentrations impairs embryonic development in Cyprinus carpio: Bioconcentration and alteration of oxidative stress-related gene expression patterns

In recent years and as a result of the Covid-19 pandemic, the consumption of dexamethasone (DXE) has increased. This favors that this corticosteroid is highly released in aquatic environments, generating deleterious effects in aquatic organisms. The information on the toxic effects of DXE in the environment is still limited. Thus, the objective of this work was to determine whether DXE at short-term exposure can cause alterations to embryonic development and alteration of oxidative stress-related gene expression patterns in Cyprinus carpio. For this purpose, common carp embryos (2 hpf) were exposed to realistic concentrations of DXE until 96 hpf. Alterations to embryonic development were evaluated at 12, 24, 48, 72 and 96 hpf. In addition, oxidative stress in carp embryos at 72 and 96 hpf was evaluated by cellular oxidation biomarkers (lipoperoxidation level, hydroperoxide and carbonyl protein content) and antioxidant enzymes activities (superoxide dismutase and catalase). Oxidative stress-related gene expression (sod, cat and gpx1) was also evaluated. Our results showed that DXE concentrations above 35 ng/L are capable of producing alterations to embryonic development in 50 % of the embryo population. Furthermore, DXE was able to induce alterations such as scoliosis, hypopigmentation, craniofacial malformations, pericardial edema and growth retardation, leading to the death of half of the population at 50 ng/L of DXE. Concerning oxidative stress, the results demonstrated that DXE induce oxidative damage on the embryos of C. carpio. In conclusion, DXE is capable of altering embryonic development and generating oxidative stress in common carp C. carpio.

Toxic effects of polyethylene microplastics on transcriptional changes, biochemical response, and oxidative stress in common carp (Cyprinus carpio)

Aquatic ecosystems have become a place for accumulating microplastics (MPs). MPs can directly or indirectly damage organisms. Although studies of the toxicity of MPs, there are insufficient literature reports on the effects of MPs on freshwater aquatic life. Therefore, this study aimed to evaluate the effect of MPs toxicity on Cyprinus carpio. In this study, biochemical parameters, oxidative biomarkers, and gene expression were assayed in fish exposed to 0, 175, 350, 700, and 1400 μg L−1 of MPs for 30 days. MPs were detected in the liver and intestine of fish using FTIR-analysis. Mt1, Ces2, and P450 mRNA expression were enhanced in the hepatocytes of fish exposed to MPs, while Mt2 gene expression was significantly decreased. After exposure to MPs, MDA and carbonyl protein levels were higher than those of the reference group. The antioxidant capacity and glycogen contents in the hepatocytes significantly declined. MPs significantly inhibited glutathione reductase (GR), glucose 6-phosphate dehydrogenase (G6PDH), and catalase (CAT) activities. However, superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities increased. MPs decreased the total protein, globulin levels, and butyrylcholinesterase (BChE) activity in blood. In contrast, aspartate aminotransferase (AST), alanine aminotransferase (ALT), gamma-glutamyl transferase (GGT), lactate dehydrogenase (LDH), alkaline phosphatase (ALP), and creatine phosphokinase (CPK) activities increased in treated-fish with MPs. Glucose, creatinine, cholesterol and triglyceride concentrations in fish exposed to MPs were significantly higher than that of the reference group. Consequently, MPs exposure could disrupt biochemical homeostasis, oxidative stress and alter the expression of genes involved in detoxification.

Effect of selenium-rich Bacillus subtilis against mercury-induced intestinal damage repair and oxidative stress in common carp

Mercury (Hg) poisoning in humans and fish represents a significant global problem. Hg is one of the most dangerous threats to the aquatic ecosystem due to its high toxicity. Mercury has a high oxidative stress-inducing potential, and can compounds exert toxic effects by interacting with many important enzymes involved in the regulation of antioxidants. Selenium (Se) supplementation can reactivate the mercury-inhibited enzymes viability. The probiotic Bacillus subtilis is widely used in aquaculture, and it has a certain adsorption effect on heavy metals. The interactions between Hg and Se have been rigorously investigated, particularly due to the observed protective effects of Se against Hg toxicity. The objective of this study was to evaluate whether Se-rich B. subtilis ameliorated Hg-induced toxicity in C. carpio var. specularis. Fish were exposed to waterborne Hg (0.03 mg/L) and fed a diet supplemented with 105 cfu/g Se-rich B. subtilis for 30 days. Fish were sampled, antioxidant activity, and Intestinal damage repair were assessed. Our results indicated that Se-rich B. subtilis protected the Intestinal from Hg-induced morphological changes. Hg treatment significantly decreased the activity levels of SOD, CAT and GSH-PX while increasing the activity levels of MDA, GST, and GSH. Hg treatment also upregulated the mRNA expression of Nrf2, CAT, GSH-PX and HO-1, and reduced expression of keap1. Se-rich B. subtilis had a significant protective effect against Hg-induced oxidative stress.

The ameliorative role of geranium (Pelargonium graveolens) essential oil against hepato-renal toxicity, immunosuppression, and oxidative stress of profenofos in common carp, Cyprinus carpio (L.)

The present study investigated the possible effect of profenofos (PFF) on hepato-renal functions, non specific-immunity, and oxidants/antioxidants status in common carp, Cyprinus carpio (L.), and the supportive role of geranium (Pelargonium graveolens) essential oil (GEO). A static 96-h acute toxicity test was performed to determine the 96-h LC50 of PFF in common carp fingerlings and it was found to be 47.46 μg L−1 calculated by Finney probit analysis. One hundred and twenty fish were randomly allocated into four groups in triplicate. The first served as control and second groups were fed on basal diets that were enriched with 0.0 and 400 mg kg−1 of diet GEO, respectively without PFF exposure. The third and fourth groups were fed on the same diets and in addition, were exposed to 1/10 of the 96 h-LC50 of PFF (4.74 μg L−1), respectively for 60 days. The results revealed that the PFF exposure induced marked increases in the levels of serum alanine and aspartate aminotransferases (ALT and AST), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), creatinine, urea, cholesterol, triglycerides (TG), and malondialdehyde (MDA). Meanwhile, the serum total protein (TP), globulins, glucose, lysozyme activity, complement3 (C3), nitric oxide (NO), and hepatic antioxidant enzyme activities as well as the reduced glutathione (GSH) content were markedly declined in the PFF-exposed fish compared to the control group. Additionally, vacuolation, pyknosis, and degeneration of hepatocytes and epithelial cells of renal tubules with mononuclear cell infiltration were observed. A strong caspase-3 and weak BCL-2 immunohisto-reactivity in the liver and kidney tissues of PFF-exposed fish were noted respective to the control. Interestingly, GEO supplementation improved these variables with nearly restoring to control values with improving the histological architecture of liver and kidney tissues. The apoptotic condition of the examined tissues was markedly restricted in the combination group than PFF-exposed group. These results concluded that GEO has ameliorative influence against toxic impacts of PFF on hepato-renal functions, immunological parameters, and oxidative stress in common carp, and therefore, maybe a valuable candidate as a dietary supplement in aquaculture.

Limited oxidative stress in common carp (Cyprinus carpio, L., 1758) exposed to a sublethal tertiary (Cu, Cd and Zn) metal mixture

Analyzing effects of metal mixtures is important to obtain a realistic understanding of the impact of mixed stress in natural ecosystems. The impact of a one-week exposure to a sublethal metal mixture containing copper (4.8 μg/L), cadmium (2.9 μg/L) and zinc (206.8 μg/L) was evaluated in the common carp (Cyprinus carpio). To explore whether this exposure induced oxidative stress or whether defense mechanisms were sufficiently fitting to prevent oxidative stress, indicators of apoptosis (expression of caspase 9 [CASP] gene) and of oxidative stress (malondialdehyde [MDA] level and xanthine oxidase [XO] activity) were measured in liver and gills, as well as activities and gene expression of enzymes involved in antioxidant defense (superoxide dismutase [SOD], catalase [CAT], glutathione peroxidase [GPx], glutathione reductase [GR] and glutathione-S-transferase [GST]). The total antioxidative capacity (T-AOC) was also quantified. No proof of oxidative stress was found in either tissue but there was indication of apoptosis in the liver. CAT, GPx, GR and GST total activities were reduced after 7 days, suggesting a potential decrease of glutathione levels and risk of increased free radicals if the exposure would have lasted longer. There were no major changes in the total activities of antioxidant enzymes in the gills, but the relative expression of the genes coding for CAT and GR were triggered, suggesting a response at the transcription level. These results indicate that C. carpio is well equipped to handle these levels of metal pollution, at least during short term exposure.

Acesulfame potassium: Its ecotoxicity measured through oxidative stress biomarkers in common carp (Cyprinus carpio)

Acesulfame potassium (ACS) is a widely-used sweetener worldwide. Its presence has been demonstrated in diverse bodies of water. However, the deleterious effects this causes in aquatic organisms has not yet been identified, which generates controversy concerning the risks that ACS represents after its disposal into the bodies of water. Thus, the objective of this work was to evaluate if the exposure of ACS in environmentally-relevant concentrations was capable of producing oxidative stress in blood, liver, gill, brain and muscle of common carp (Cyprinus carpio). With this finality, the carp were exposed to two environmentally-relevant concentrations (0.05 and 149 μg L−1) at different exposure times (12, 24, 48, 72 and 96 h), having controls in the same conditions for each exposure time. Posteriorly, the following biomarkers of damage were evaluated: hydroperoxide content (HPC), level of lipoperoxidation (LPX) and protein carbonyl content (PCC), as well as the activity of antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT). The results showed that ACS produces significant increase in damage biomarkers evaluated in all organs, mainly in gill, brain and muscle, as well as significant changes in the activity of antioxidant enzymes in the same organs. Thus, it is concluded that ACS is capable of producing oxidative stress in common carp (Cyprinus carpio).

Effects of dimethoate alone and in combination with Bacilar fertilizer on oxidative stress in common carp, Cyprinus carpio

Insecticides and fertilizers are the most common agrochemicals that enter aquatic ecosystems from farmlands via different ways and consequently affect fish. In this study, effects of dimethoate, and Bacilar, as a bio-fertilizer, on oxidative stress and biochemical parameters were measured in common carp. For this purpose, fish were exposed to 16 and 32 μg L−1 dimethoate, 0.1 and 0.2 ml L−1 Bacilar and the mixtures of both compounds for 14 days. The results showed that Bacilar increased aspartate aminotransferase (AST) and LDH in gills, AST in liver and the activity of catalase (CAT) (0.2 ml L−1) in kidney, but decreased the activity of lactate dehydrogenase (LDH) and glucose 6-phsphate dehydrogenase (G6PDH) in liver. Dimethoate, alone or in combination with Bacilar, significantly increased LDH, alkaline phosphatase (ALP), AST and CAT activities in gills. Fish exposure to dimethoate, alone or in combination with Bacilar, resulted in a significant decrease in LDH, G6PDH activities, total antioxidant (TA) and glycogen levels in liver and TA in kidney. A significant increase was observed in malondialdehyde (MDA), ALP, AST, alanine aminotransferase (ALT) and CAT activity in liver and kidney of fish exposed to dimethoate alone or in combination with Bacilar. The results showed that cytotoxicity of Bacilar and dimethoate combination depended on their concentrations. Higher concentrations of Bacilar induced significant changes in some biochemical parameters. Dimethoate or/and Bacilar, in sub-lethal concentrations, induced oxidative damages in fish. Finally, these data support the hypothesis that changes in biochemical parameters were induced by exposure to dimethoate and/or Bacilar.

Environmentally relevant concentrations of glibenclamide induce oxidative stress in common carp (Cyprinus carpio)

The hypoglycemic pharmaceutical glibenclamide (GLB) is widely used around the world. This medication is released into the environment by municipal, hospital and industrial wastewater discharges. Although there are reports of its environmental occurrence in the scientific literature, toxicity studies on aquatic species of commercial interest such as the common carp Cyprinus carpio are scarce. The present study aimed to evaluate the oxidative stress induced on C. carpio by environmentally relevant concentrations of GLB. Biomarkers of oxidative damage such as hydroperoxide content, lipid peroxidation and protein carbonyl content were evaluated as well as the activity of the antioxidant enzymes superoxide dismutase and catalase. The concentration of GLB was determined in water as well as in gill, liver, muscle, brain and blood of carp at 12, 24, 48, 72 and 96 h. The findings obtained in the study prove that GLB induces increases in biomarkers of oxidative damage and antioxidant enzyme activity in the teleost C. carpio, that this response is not concentration dependent and that the organs evaluated bioconcentrate this hypoglycemic agent. These findings permit us to conclude that the presence of GLB in water bodies represents a risk for aquatic species.

Effect of cadmium on oxidative stress and immune function of common carp (Cyprinus carpio L.) by transcriptome analysis

Cadmium (Cd) is an increasingly important environmental pollutant which causes irreversible toxicity to fish. To understand how Cd impacts the immune response and oxidative stress in common carp, we performed transcriptomic profiles for head kidney, the immune organ of common carp which were underwent Cd exposure. Totally there are 42,489,124 and 48,562,526 high quality clean reads obtained from the Cd exposure groups, and 44,677,578 and 44,106,696 clean reads from the control groups. Among them, 308 genes were differently expressed, including 101 upregulated and 207 down-regulated genes. The identified genes were enriched using databases of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Antioxidant systems and immune function genes and pathways were identified and validated by quantitative real-time RT-PCR. Our results showed that Cd exposure leads to oxidative stress and immunosuppression in head kidney of common carp. These results provide new insights for unveiling the biological effects of Cd in common carp.

Chlorpyrifos exposure in common carp (Cyprinus carpio L.) leads to oxidative stress and immune responses

Chlorpyrifos (CPF) is an environmental pollutant with increasing importance due to its high toxicity to fish and aquatic animals. To understand how CPF impacts immune response and oxidative stress in common carp (Cyprinus carpio L.), we investigated the transcriptomic profiles of the head kidneys from common carp exposed to CPF for 15days. A total of 52, 297, 928 and 52, 273, 784 high quality clean reads were obtained from CPF exposure groups, 44,677,578 and 44,106,696 high quality clean reads were obtained from corresponding control groups. Among them, 456 genes were differentially expressed, including 246 up-regulated genes and 210 down-regulated genes identified and enriched in databases of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Antioxidant systems, and immune response genes and pathways were verified by quantitative real-time RT-PCR. We found that CPF-induced ROS regulates immune response by stimulating the antigen-presenting ability of head kidney in carp. These results provide new insights for unveiling the biological effects of CPF in common carp.

Modulatory role of L-carnitine against microcystin-LR-induced immunotoxicity and oxidative stress in common carp

Microcystin-LR (MCLR), one of the most popular microcystins (MCs) found in many field water bodies around the world, poses great health risks to animals and humans. In the present study, healthy common carp (initial weight 24.8±2.3g) were randomly assigned to five groups. Group I was fed on normal diet as control. Group II was maintained on normal diet and received MCLR intraperitoneal injection (150μgkg-1 BW). Common carp in groups III, IV, and V were daily pretreated with L-carnitine (LC) at doses of 0.5, 1.0, and 2.0gkg-1 of the diet for 4weeks prior to MCLR intraperitoneal injection. The results showed that MCLR alone led to a significant downregulation in immune response, including serum complement C3, lysozyme, and bactericidal activity. However, oxidative stress response: catalase (CAT), superoxide dismutase (SOD), glutathione (GSH), glutathione peroxidase (GPx), and lipid peroxidation (LPO) levels were significantly increased. Similarly, gene expressions of inflammatory IL-1β, TNF-α, IFN I, and heat shock proteins (HSP70 and HSP90) were also upregulated after challenged with MCLR. However, LC pretreated group caused a significant elevation in immune response (C3, lysozyme, and bactericidal activity) and gene expressions of inflammatory IL-1β, TNF-α, IFN I, and heat shock proteins (HSP70 and HSP90) after MCLR stress. Antioxidant activities (CAT, SOD, GSH, GPx, and LPO) were returned to background levels at 96h after MCLR challenge. Strikingly, LC supplementation at 2.0gkg-1 has been considered the optimum for common carp since it exhibited enhancement of immune response and antioxidant activity over the level 0.5 and 1.0gkg-1, and even better than that of control level. It was concluded that LC as a functional feed additive significantly inhibited the progression of MCLR-induced immunotoxicity and oxidative stress in common carp.

Cyto-genotoxicity and oxidative stress in common carp (Cyprinus carpio) exposed to a mixture of ibuprofen and diclofenac.

Thirty million people worldwide consume each day nonsteroidal anti-inflammatory drugs (NSAIDs), a heterogeneous group of pharmaceuticals used for its analgesic, antipyretic, and anti-inflammatory properties. Recent studies report high NSAID concentrations in wastewater treatment plant effluents, in surface, ground, and drinking water, and in sediments. NSAIDs are also known to induce toxicity on aquatic organisms. However, toxicity in natural ecosystems is not usually the result of exposure to a single substance but to a mixture of toxic agents, yet only a few studies have evaluated the toxicity of mixtures. The aim of this study was to evaluate the toxicity induced by diclofenac (DCF), ibuprofen (IBP), and their mixture on a species of commercial interest, the common carp Cyprinus carpio. The median lethal concentration of IBP and DCF was determined, and oxidative stress was evaluated using the following biomarkers: lipid peroxidation and activity of the antioxidant enzymes superoxide dismutase, catalase and glutathione peroxidase. Cyto-genotoxicity was evaluated by micronucleus test, comet assay, and the specific activity of caspase-3. Results show that DCF, IBP, and a mixture of these pharmaceuticals induced free radical production, oxidative stress and cyto-genotoxicity in tissues of C. carpio. However, a greater effect was elicited by the mixture than by either pharmaceutical alone in some biomarkers evaluated, particularly in gill. © 2016 Wiley Periodicals, Inc. Effect of pollutants in Tigris river in Mosul on lipid peroxidation and antioxidant parameters in tissues of the common carp (Cyprinus carpio L.)

This research was conducted for evaluation of the effect of pollutants of Tigris on some parameters of oxidative stress in common carp. In addition to detection of Glutathione, Malondialdehyde, vitamins E and C and the results were compared with control group. The results revealed that the presence of congestion on blood vessels in kidney with vacuolar degeneration in hepatic cells with infiltration of inflammatory cells and swelling and fatty change in the epithelial cells lining the renal tubules in addition to coagulative necrosis with infiltration of inflammatory cells in the interstitial tissue of kidney. Furthermore fatty change in cardiac muscles with infiltration of inflammatory cells and congestion of blood vessels in heart were observed. The statistical analysis referred to a significant decrease (P≤0.05) in glutathione and increase in the malondialdehyde concentrations levels for the most tissues but there was no difference in E and C vitamins except that of the kidney which show a decrease in C vitamin level. It is concluded from the present study that there is a worse effect of pollutants in river of Tigris on the levels of lipid peroxidation indicators for carp fish tissues naturally exposed to those pollutants which may affects the nutritional and hygienic values of these fishes.
