Necrosulfonamide promotes hair growth and ameliorates DHT-induced hair growth inhibition

BackgroundAlopecia affects patients' appearance and psychology. Mixed-lineage kinase domain-like pseudokinase (MLKL)-mediated necrosis plays a role in various skin diseases, but its effect on hair growth is unclear. ObjectiveTo investigate the effects of MLKL on hair growth and its regulatory mechanisms and to determine the potential clinical value of Necrosulfonamide (NSA, a MLKL-targeting inhibitor) in promoting hair growth and counteracting dihydrotestosterone (DHT) inhibition of hair growth. MethodsThe expression level of MLKL was detected in the scalp of androgenetic alopecia (AGA) patients and the skin tissues of mice. Knock down MLKL expression or use NSA to observe hair growth in vivo and in vitro. ResultsIn AGA patients, MLKL expression is elevated in the alopecia areas. In mice, MLKL is significantly expressed in the outer root sheath (ORS) cells of hair follicles, peaking during the catagen phase. Knockdown expression of MLKL in mice skin promoted hair growth. NSA enhanced hair growth and prevented hair follicle regression via the Wnt signaling. Reduced MLKL boosts ORS cell proliferation without directly impacting DPCs' growth. Interestingly, NSA boosts DPCs' proliferation and induction when co-cultured with ORS cells. Besides, NSA alleviated the inhibition of DHT on hair growth in vivo and vitro. ConclusionNSA inhibited the activation of MLKL in ORS cells, promoted the activation of Wnt signal in DPC cells, and improved the inhibition of hair growth by DHT, illuminating a new alopecia mechanism and aiding anti-alopecia drug development.

Transcriptome Analysis Reveals an Inhibitory Effect of Dihydrotestosterone-Treated 2D- and 3D-Cultured Dermal Papilla Cells on Hair Follicle Growth.

Dermal papillae are a target of androgen action in patients with androgenic alopecia, where androgen acts on the epidermis of hair follicles in a paracrine manner. To mimic the complexity of the dermal papilla microenvironment, a better culture model of human dermal papilla cells (DPCs) is needed. Therefore, we evaluated the inhibitory effect of dihydrotestosterone (DHT)-treated two-dimensional (2D)- and 3D-cultured DPCs on hair follicle growth. 2D- and 3D-cultured DPC proliferation was inhibited after co-culturing with outer root sheath (ORS) cells under DHT treatment. Moreover, gene expression levels of β-catenin and neural cell adhesion molecules were significantly decreased and those of cleaved caspase-3 significantly increased in 2D- and 3D-cultured DPCs with increasing DHT concentrations. ORS cell proliferation also significantly increased after co-culturing in the control-3D model compared with the control-2D model. Ki67 downregulation and cleaved caspase-3 upregulation in DHT-treated 2D and 3D groups significantly inhibited ORS cell proliferation. Sequencing showed an increase in the expression of genes related to extracellular matrix synthesis in the 3D model group. Additionally, the top 10 hub genes were identified, and the expression of nine chemokine-related genes in DHT-treated DPCs was found to be significantly increased. We also identified the interactions between transcription factor (TF) genes and microRNAs (miRNAs) with hub genes and the TF–miRNA coregulatory network. Overall, the findings indicate that 3D-cultured DPCs are more representative of in vivo conditions than 2D-cultured DPCs and contribute to our understanding of the molecular mechanisms underlying androgen-induced alopecia.

Hair growth promotion by Necrostatin-1s

Necrostatins (Necs) have been developed as a receptor-interacting protein kinase 1 (RIPK1) inhibitor, thus inhibiting necroptosis. In this current study, we have investigated the possible involvement of necroptosis in the hair cycle regulation and further examined its underlying molecular mechanisms. Diverse RIPK1/3 inhibitors and siRNA were tested in the human outer-root sheath (ORS) cells and animal models. The expression and hair cycle-dependent expression of RIPK 1, respectively, were investigated in the hair follicles (HF) of human, pig, and the mouse. Resulting from the experiment, Nec-1s was most effective in the hair growth promotion among several inhibitors. Nec-1s induced the ORS cell proliferation and migration, and increased the HF length in mouse and pig organ cultures. In addition, it accelerated the telogen-to-anagen transition and elongated the anagen period in the mouse model. Both apoptosis and necroptosis were detected in hair cycle. RIPK1 and RIPK3 were highly expressed in ORS cells during the hair regression period. Nec-1s upregulated the mRNA expression of Wnt3a and Wnt5b, and the activity of β-catenin. Collectively, Nec-1s promotes hair growth through inhibiting necroptosis and activating the Wnt/β-catenin pathway. Necroptosis is involved in hair cycle regression, and Nec-1s is a promising target for hair-loss treatment.

Nerve growth factor regulates the proliferation of cashmere goat outer root sheath cells through the activation of cAMP-binding protein

The quality and yield of cashmere are closely related to the growth and development of the secondary hair follicles of cashmere goats. Outer root sheath cell (ORSC) proliferation is considered particularly important for the growth and development of hair follicles; however, the factors affecting this are unclear. We previously showed that the expression of nerve growth factor (NGF) in secondary follicle ORSCs was significantly higher in the anagen phase than in the catagen phase, suggesting that NGF promotes ORSC proliferation. To investigate whether ORSCs secrete NGF, and to investigate its role in ORSCs from goat secondary hair follicles, we used enzyme linked immunosorbent assay (ELISA) to show that NGF levels in cultured ORSC medium decreased significantly with the passage of time (P < 0.05). Immunofluorescence analysis detected the transcription factor cAMP-binding protein (CREB) in ORSCs, with a stronger signal intensity observed in the nucleus than in the cytoplasm. An alkaline phosphatase reporter system assay showed that the exogenous addition of 20 ng/mL or 100 ng/mL recombinant NGF protein significantly enhanced the activity of CREB in ORSCs, while treatment with K252α, which inhibits tropomyosin receptor kinase A (TrkA), revealed that NGF enhanced intracellular CREB activity in an NGF/TrkA-dependent manner. Real-time quantitative PCR investigated the expression of proliferating cell nuclear antigen (PCNA) mRNA after treatment with NGF or the CREB inhibitor ICG-001. The NGF/TrkA/CREB system was found to play an important role in maintaining and promoting the expression of PCNA, which will provide a theoretical basis for marker-assisted breeding of cashmere goats.

Morroniside regulates hair growth and cycle transition via activation of the Wnt/\u03b2-catenin signaling pathway

Hair loss is characterized by a shortened hair anagen phase and hair follicles (HF) miniaturization. Morroniside is the most abundant iridoid glycoside extracted from Cornus officinalis and has various bioactivities in different cell functions and tissue regeneration. In this study, we investigated the effects and the underlying mechanism of morroniside on hair growth and regulation of HF cycle transition. Morroniside treatment significantly enhanced outer root sheath cell (ORSC) proliferation and migration in vitro. Additionally, morroniside upregulated Wnt10b, β-catenin and lef1. The enhanced ORSC proliferation and migration due to morroniside treatment were partly rescued by a Wnt/β-catenin signaling inhibitor, DKK1. Furthermore, in a hair-induced mouse model, morroniside injection accelerated the onset of anagen and delayed HF catagen, as shown by histological examination. Immunohistochemical analyses revealed that Wnt/β-catenin signaling pathway expression was upregulated in the HFs. These findings suggest that morroniside regulates HF growth and development partly through the Wnt/β-catenin signaling pathway and may be a potential treatment for hair loss.

Expression of nerve growth factor in skin tissues and its effect on the proliferation of outer root sheath cells in cashmere goats

Nerve growth factor (NGF) is widely expressed in the nervous system and best understood for its neuronal function. The present study was designed to describe the characteristics of hair follicles and the expression pattern of NGF and its receptor, tyrosine kinase receptor (TrkA), in the skin of Liaoning cashmere goats during hair follicle cycling. Results of hematoxylin and eosin staining revealed that there was a high ratio of secondary to primary follicles, with more active secondary follicles during anagen phase than other phases. The results of quantitative real-time polymerase chain reaction and western blot showed that both NGF and TrkA mRNA and protein levels were persistently detected in the skin tissue during hair follicle cycling. Furthermore, NGF and TrkA mRNA and protein levels during anagen were significantly higher than that during catagen. Using immunohistochemistry, the especially high immunoreactivities of TrkA in the outer root sheath (ORS) and NGF in the internal root sheath (IRS) and ORS were observed during anagen of hair follicles. Meanwhile, NGF and TrkA immunoreactivities were also observed in the epidermis of cashmere goats at similar expression levels as hair follicles. ORS cells were isolated from secondary follicle, and NGF treatment experiments were also studied. Result show that 20ng/mL or 100ng/mL NGF could significantly promote the proliferation of ORS cells in vitro, but the promotion could be blocked by the addition of 20ng/mL K252a, the inhibitor of TrkA. The present study indicates that NGF might be involved in the reconstruction and growth of hair follicles through TrkA tyrosine kinase dependent pathways.

Sox9 Increases the Proliferation and Colony-forming Activity of Outer Root Sheath Cells CulturedIn Vitro

β-catenin plays a pivotal role in hair follicle development and hair growth cycle. The aim of this study was to identify β-catenin-regulated genes in cultured human hair outer root sheath (ORS) cells. Primary cultured ORS cells were transduced with recombinant adenovirus expressing N-terminal truncated β-catenin (constitutive active form), and β-catenin-regulated genes were identified. Overexpression of the constitutively active form of β-catenin led to induction of Sox9 expression at both mRNA and protein levels. To investigate the potential role of Sox9, we made the recombinant adenovirus expressing green fluorescent protein-tagged Sox9, and then transduced into cultured ORS cells. Interestingly, Sox9 induced the expression of keratin 15, increased the proliferation of ORS cells in vitro, and enhanced colony-forming activity. Our results suggest that Sox9 is a β-catenin-regulated gene in ORS cells, and has potential importance in the regulation of hair follicle homeostasis.

Improved isolation of outer root sheath cells from human hair follicles and their proliferation behavior under serum-free condition

Hair follicle is a small but very complex and dynamic miniorgan of the human body. It is easy to isolate and culture mesenchymal cells but not epithelial cells of hair follicle. It is necessary for intact and healthy outer root sheath (ORS) cells to be isolated and cultured. In this study we developed an appropriate isolation method to yield 6.4±0.75×104 cells/hair follicle, which is about 9-fold comparing to our previous data. This yield was achieved by modifications such as different kinds of enzyme uses, fragmentation, and mechanical stimuli. Especially we detected that the different kinds of isolation enzyme could affect proliferation of ORS cells during primary culture. In addition, bovine pituitary extract (BPE) was needed for ORS cells to proliferate and to form colonies under serum-free, feeder layer-free culture condition, but type I collagen as a substratum did not have any positive effect. Moreover, ORS cells under BPE-added condition contained stem/progenitor cells expressing β1-integrin. CK19, and CD34. These results can provide useful cell culture information, not only in the study of hair biology but also in the field of tissue engineering and cell therapy for the treatment of alopecia.

Role of Androgen in Mesenchymal Epithelial Interactions in Human Hair Follicle

Human hair follicles, which are distributed in various and specific sites of the body, appear to have an inherited susceptibility for androgen-dependent growth. Beard, axillary, and frontal scalp dermal papilla cells (DPC) were recently shown to possess the characteristics of androgen target cells. These DPC show strong expression of androgen receptors, and the expression of type II 5alpha reductase is restricted to beard and frontal scalp DPC. These findings suggest that DPC mediate the signals of androgen to follicular epithelial cells in a paracrine fashion. We developed an in vitro co-culture system using DPC and keratinocytes (KC) to characterize the mode of androgen action in human hair follicles. Androgen significantly stimulated the proliferation of KC co-cultured with beard DPC, indicating that beard DPC produce androgen-dependent diffusible growth factors. Insulin-like growth factor-I was identified as one of the androgen-dependent paracrine growth factors produced by beard DPC. We also identified the inhibitory role of androgen on the growth of KC co-cultured with DPC from androgenetic alopecia (AGA) when the DPC were transfected with an expression vector encoding the androgen receptor. This growth suppression of KC was mediated by transforming growth factor-beta1 (TGF-beta1) derived from DPC of AGA, suggesting that TGF-beta1 is a paracrine mediator for AGA.

Fibroblast Growth Factor 5 Inhibits Hair Growth by Blocking Dermal Papilla Cell Activation

Fibroblast growth factor (FGF) 5 inhibits hair growth and induces catagen in mouse hair follicles, in vivo. Given that FGF-5 receptor (FGFR1) is expressed in dermal papilla cells (DPCs), which are known to stimulate outer root sheath cell (ORSC) proliferation, we hypothesized that FGF-5 attenuates DPC-mediated ORSC proliferation. In the present study, DPCs and ORSCs were isolated from rat vibrissae, after which the effects of FGF-5 on proliferation of ORSCs cultured in DPC-conditioned medium were assessed. We first confirmed that FGFR1 was expressed in cultured DPCs and detected FGFR2-4 as well. ORSC proliferation was increased approximately twofold when the cells were cultured in DPC-conditioned medium, and the effect was unaltered by FGF-5. In addition, FGF-5 did not directly inhibit ORSC proliferation; indeed, it actually promoted proliferation of both DPCs and ORSCs. When DPCs were first activated by exposure to FGF-1 and FGF-2, which are expressed in hair follicles during anagen, ORSC proliferation observed in the resultant conditioned medium was substantially greater than in medium conditioned by unstimulated DPCs. The FGF-1-induced enhancement was reversed by FGF-5, diminishing ORSC proliferation to control levels. By contrast, the enhancement of DPC-mediated ORSC proliferation by FGF-2 was not suppressed by FGF-5. Proliferation of ORSCs did not depend on DPC proliferation, nor did FGF-1 directly promote ORSC proliferation. Dermal papillae thus appear to require activation before they will efficiently stimulate hair growth, and FGF-5 appears to inhibit hair growth and induce catagen by blocking that activation.

Inhibition of hair growth by testosterone in the presence of dermal papilla cells from the frontal bald scalp of the postpubertal stumptailed macaque.

Hair-follicle regression in the bald scalps of stumptailed macaques develops after puberty, which corresponds to an elevation of serum testosterone and dihydrotestosterone. Using the cultured cells from the pre- and postpubertal macaques, we examined the role of dermal papilla cells in testosterone-induced inhibition of outer root sheath cell proliferation. Testosterone showed no effects on proliferation of either dermal papilla cells or outer root sheath cells cultured alone. Testosterone-induced inhibition of outer root sheath cell proliferation occurred only in coculture with dermal papilla cells derived from the bald scalps of adult macaques but not with dermal papilla cells from the hairy occipital scalps of adult macaques or the prebald frontal scalps of juvenile macaques. Furthermore, RU 58841, an androgen receptor blocker, antagonized this testosterone-elicited inhibition. Together our data indicate that the inhibitory effect of testosterone on proliferation of epithelial cells is age dependent, and androgen may play an essential role in hair growth either by inducing repressor(s) from dermal papilla cells, which may then inhibit the growth of epithelial cells of the hair follicle, or by inducing growth factor(s) from dermal papilla cells, which, in turn, may trigger the induction of some repressors in epithelial cells, thereby inhibiting the epithelial cell growth. Our animal studies also showed that RU 58841 has a dramatic effect on hair regrowth in the bald frontal scalp of the stumptailed macaque, which may further support our in vitro culture studies showing that antiandrogens can antagonize testosterone-elicited hair growth. In summary, our studies may provide a model for further isolation of androgen-regulated repressor(s)/growth factors, which may help control hair growth and baldness.

Mechanism of action of androgen in hair follicles

In order to investigate the mode of action of testosterone (T) on human hair follicles we studied the metabolism of T and localization of androgen receptors in outer root sheath cells (ORSC) and dermal papilla cells (DPC) from different body sites. T was principally metabolized to androstenedione (Δ 4) even in beard ORSC as well as epidermal keratinocytes (EK), and the ratio of apparent 5α-reductase (5α-R) to 17β-hydroxysteroid dehydrogenase (17β-HSD) did not differ between these two kinds of cells. The 5α-R activity in beard DPC was 3 times as high as that in occipital scalp and axillary DPC. The 5α-R of beard DPC exhibited a narrow optimum pH of 5.5, which is characteristic of type 2 enzyme present in androgen target cells. In contrast, 5α-R of DPC from axillary and occipital scalp hair showed a broad optimum pH range between 6.5–9.0 corresponding to type 1 5α-R. Androgen receptors were detected in the DPC of beard and axillary hair follicles, but not in those of occipital scalp hair follicles using immunohistochemical staining with polyclonal anti-androgen receptor antibody. Epithelial cells of the hair bulb were not stained by the antibody. Androgen receptors were also detected in the nuclei of cultured beard and axillary DPC, but the DPC from occipital scalp hair follicles showed little staining with the antibody. We also examined the effects of T on the DNA synthesis and proliferation of cultured ORSC and DPC. T did not have a proliferative effect on either type of cell when cultured alone. In contrast, T significantly stimulated the proliferation of ORSC which were cocultured with beard or axillary DPC without cell contact. No such effect was observed when dermal papilla cells from occipital scalp hair follicles were used in the cultures. Since this proliferative effect of T was antagonized by cyproterone acetate (CA), it was apparently mediated by the androgen receptor pathway. These results suggest that the dermal papilla cells from the beard and axillary hair follicles produce androgen-dependent diffusible growth factors which stimulate follicular epithelial cells.

Organotypic cocultures as models to study cell-cell and cell-matrix interactions of human hair follicle cells.

In the hair follicle complex interactions of specialized epithelial and mesenchymal cells as well as extracellular components are crucial for regulation of proliferation and differentiation. In order to mimic this situation in vitro, techniques to cultivate follicular cells such as outer root sheath (ORS) cells, hair matrix cells (HMC) and hair papilla cells (HPC) were developed. Human dermal fibroblasts (HDF) and HPC markedly enhanced proliferation of ORS cells when cocultured spatially separated in a two-chamber system, which was more pronounced with postmitotic than with mitotic mesenchymal cells. In organotypic cocultures of ORS cells on HDF- or HPC-populated collagen gels lifted at the air-medium interface, stratified epithelia developed largely reminiscent of the epidermis. The morphology was well structured and differentiation markers were expressed (e.g. suprabasal keratins, filaggrin, involucrin, basement membrane components). Both proliferation and differentiation were dependent on the presence of HDF or HPC. When grown embedded in extracellular matrix (Matrigel) without HDF, ORS cells formed large spheroids with inward directed differentiation. Also herein HDF or HPC sustained both cell growth and balanced, epidermistype differentiation. While in organotypic cocultures with HDF, HMC also organized into stratified epithelia, epidermis-type stratification was prevented by HPC. Similarly, in the presence of HDF, HMC in Matrigel formed keratinizing spheroids, whereas this was largely suppressed in the presence of HPC. However, hair-type differentiation was not observed, suggesting a crucial role of other yet unknown components or of the surrounding matrix.

Effects of 1 alpha,25-dihydroxy-vitamin D3 and calcipotriol on organotypic cultures of outer root sheath cells: a potential model to evaluate antipsoriatic drugs.

In the human hair follicle, outer root sheath (ORS) cells constitutively express the hyperproliferation-associated keratins 6, 16 and 17 instead of keratins 1 and 10 found in interfollicular epidermis. In organotypic cultures. ORS cells form a stratified epithelium which in many respects resembles psoriatic skin: it has a hyperplastic tissue architecture and a poorly developed granular layer, and expresses hyperproliferation-associated keratins. Therefore, we studied the effects of the antipsoriatic compounds 1 alpha,25-dihydroxy-vitamin D3 (1 alpha,25-(OH)2-D3) and its synthetic derivative calcipotriol on cultured ORS cells. In monolayer cultures, 10(-6) M 1 alpha,25-(OH)2-D3 or calcipotriol completely blocked ORS cell proliferation. This inhibitory effect was substantially reduced at 10(-8) M. Incubation of organotypic ORS cultures with both vitamin D analogues resulted in a marked thinning of the living cell compartment concomitant with a thickening of the horny layer. A reduced expression of differentiation markers such as keratins 10, 16 and 17, involucrin and filaggrin paralleled the thinning of the stratum Malpighi. As determined by quantification of BrdU-positive cells, ORS cell proliferation was apparently not affected by the vitamin D analogues, indicating that these compounds mainly operate by accelerating the differentiation pathway within the suprabasal living cell compartment. No alteration in the expression of the alpha 6- and beta 1-integrin chains was found.

The effect of 1,25-dihydroxyvitamin D 3 on the growth and differentiation of cultured human outer root sheath cells from normal subjects and patients with vitamin D-dependent rickets type II with alopecia

We examined the effect of 1,25-dihydroxyvitamin D 3 (1,25(OH) 2D 3) on the growth and differentiation of cultured human hair outer root sheath cells (ORSC) from normal subjects and patients with vitamin D-dependent rickets type II (DDR-II) with alopecia. 1,25(OH) 2D 3 dose-dependently suppressed the plating efficiency, clonal growth, and DNA synthesis of normal ORSC. It enhanced the cornified envelope formation and caused morphological changes in the cells. All results indicated the existence of specific receptors for 1,25(OH) 2D 3 in the ORSC, and suggest that 1,25(OH) 2D 3 is a potent inhibitor of proliferation of ORSC as well as a stimulator of terminal differentiation. However, the cells from DDR-II patients with alopecia did not respond to 1,25(OH) 2D 3, suggesting a lack of the specific receptors in the cells. The differences in the cellular response to the hormone between the normal ORSC and those from the patients were apparent and easily distinguishable, therefore this experiment may be a rapid and simple diagnostic test for DDR-II patients with alopecia. Large number of hairs were difficult to obtain from patients with alopecia, and we developed a new culture method to accomplish these studies from a few plucked hair follicles. Our system may be useful in the culture of ORSC from limited number of follicles, and could be utilized to analyse the cellular characteristics of ORSC in patients with hair diseases.
