Retinal waves align the concentric orientation map in mouse superior colliculus to the center of vision

Neurons in the mouse superior colliculus (SC) are arranged in a concentric orientation map, which is aligned to the center of vision and the optic flow experienced by the mouse. The origin of this map remains unclear. Here, we propose that spontaneous retinal waves during development provide a scaffold to establish the concentric orientation map within the SC and its alignment to the optic flow. We test this hypothesis by modeling the orientation-tuned SC neurons that receive ON/OFF retinal inputs. Our model suggests that the propagation direction bias of stage III retinal waves, together with OFF-delayed responses, shapes the spatial organization of the orientation map. The OFF delay establishes orientation-tuned neurons by segregating their ON/OFF receptive subfields, the wave-like activities form the concentric pattern, and the direction biases align the map to the center of vision. Together, retinal waves may play an instructive role in establishing functional properties of single SC neurons and their spatial organization within maps.

Investigating the Serotonergic Modulation of Orientation Tuning of Neurons in Primary Visual Cortex of Anesthetized Mice

Introduction: Sensory processing is profoundly regulated by brain neuromodulatory systems. One of the main neuromodulators is serotonin which influences higher cognitive functions, such as different aspects of perceptual processing. Accordingly, malfunction in the serotonergic system may lead to visual illusion in psychiatric disorders, such as autism and schizophrenia. This study aims to investigate the serotonergic modulation of visual responses of neurons to stimulus orientation in the primary visual cortex. Methods: Eight-week-old naive mice were anesthetized and a craniotomy was done on the region of interest in the primary visual cortex. Spontaneous and visual-evoked activities of neurons were recorded before and during the electrical stimulation of the dorsal raphe nucleus using in vivo whole-cell patch-clamp recording. The square-wave grating of 12 orientations was presented. The data were analyzed and the Wilcoxon signed-rank test was used to compare the data of two conditions that belong to the same neurons, with or without electrical stimulation. Results: The serotonergic system changed the orientation tuning of nearly 60% of recorded neurons by decreasing the mean firing rate in two independent visual response components, namely gain and baseline response. It also increased the mean firing rate in a small number of neurons (about 20%). Additionally, it left the preferred orientation and sensitivity of neurons unchanged. Conclusion: Serotonergic modulation showed a bidirectional effect. It causes predominately divisive and subtractive decreases in the visual responses of the neurons in the primary visual cortex that can modify the balance between internal and external sensory signals and result in disorders.

A two-photon imaging study on the roles of macaque V1 and V4 in texture segregation

The segregation of visual scenes on the basis of texture differences is a fundamental process in early/middle vision. Previous studies are inconsistent on whether V1 neurons could signal texture based figures from grounds (Lamme, 1995), or simply detect local feature discontinuities through surround modulation (Rossi_et_al., 2001). Here we used two-photon calcium imaging to study neuronal activities associated with texture-based figure-ground stimuli in V1, as well as in V4, in awake fixating macaques. Responses to parafoveal texture stimuli were obtained from 1237 orientation-tuned V1 neurons in two macaques, and from 1049 orientation-tuned V4 neurons in two additional macaques. The figure-ground texture stimuli consisted of a 32x32 arcdeg array of oriented lines in 4 orientations (ground), within which a 4x4 arcdeg square patch (figure) was formed by orthogonally orientated line segments. The center position of the figure patch was varied relative to the pRF of the recording site, so that the pRF fell within the figure, on the figure-ground boundary, or within the ground. Responses to a single oriented bar (to identify orientation-tuned neurons and measure tuning functions) and the uniform textures were also recorded. V1 neurons’ responses to texture stimuli were suppressed compared to neurons’ peak responses to an oriented bar. Slightly less suppression was observed when the figure-ground boundary fell on the pRF, regardless of neurons’ orientation preferences. Similar effects were not observed in V4 neurons, likely because larger V4 RFs made it difficult to align with a specific part of the texture stimuli. However, V4 neurons that preferred higher SFs (≥4 cpd) responded strongly to the figure-ground boundary oriented in the neurons’ preferred orientation. These results suggest that V1 neurons may weakly signal the figure-ground boundary by detecting the local feature discontinuities, while V4 neurons participate in texture segregation by detecting the second-order figure-ground boundary.

Effects of Chronic Alcohol Use Disorder on the Visual Tilt Illusion.

Rationale: Among the serious consequences of alcohol use disorder (AUD) is the reduced ability to process visual information. It is also generally agreed that AUD tends to occur with disturbed excitation–inhibition (EI) balance in the central nervous system. Thus, a specific visual behavioral probe could directly qualify the EI dysfunction in patients with AUD. The tilt illusion (TI) is a paradigmatic example of contextual influences on perception of central target. The phenomenon shows a characteristic dependence on the angle between the inducing surround stimulus and the central target test. For small angles, there is a repulsion effect; for larger angles, there is a smaller attraction effect. The center-surround inhibition in tilt repulsion is considered to come from spatial orientational interactions between orientation-tuned neurons in the primary visual cortex (V1), and tilt attraction is from higher-level effects of orientation processing in the visual information processing.Objectives: The present study focuses on visual spatial information processing and explores whether chronic AUD patients in abstinence period exhibited abnormal TI compared with healthy controls.Methods: The participants are 30 male volunteers (20–46 years old) divided into two groups: the study group consists of 15 clinically diagnosed AUD patients undergoing abstinence from alcohol, and the control group consists of 15 healthy volunteers. The TI consists of a center target surround with an annulus (both target and annulus are sinusoidal grating with spatial frequency = 2 cycles per degree). The visual angle between center and surround is a variable restricted to 0°, ±15°, ±30°, or ±75°. For measuring the TI, participants have to report whether the center target grating orientation tilted clockwise or counterclockwise from the internal vertical orientation by pressing corresponding keys on the computer keyboard. No feedback is provided regarding response correctness.Results: The results reveal significantly weaker tilt repulsion effect under surround orientation ±15° (p < 0.05) and higher lapse rate (attention limitation index) under all tested surround orientations (all ps < 0.05) in patients with chronic AUD compared with health controls.Conclusions: These results provide psychophysical evidence that visual perception of center-contextual stimuli is different between AUD and healthy control groups.

Plaid Detectors in Macaque V1 Revealed by Two-Photon Calcium Imaging.

Neuronal responses to one-dimensional orientations are combined to represent two-dimensional composite patterns; this plays a key role in intermediate-level vision such as texture segmentation. However, where and how the visual cortex starts to represent composite patterns, such as a plaid consisting of two superimposing gratings of different orientations, remains neurophysiologically elusive. Psychophysical and modeling evidence has suggested the existence of early neural mechanisms specialized in plaid detection [1-6], but the responses of V1 neurons to an optimally orientated grating are actually suppressed by a superimposing grating of different orientation (i.e., cross-orientation inhibition) [7, 8]. Would some other V1 neurons be plaid detectors? Here, we used two-photon calcium imaging [9] to compare the responses of V1 superficial-layer neurons to gratings and plaids in awake macaques. We found that many non-orientation-tuned neurons responded weakly to gratings but strongly to plaids, often with plaid orientation selectivity and cross-angle selectivity. In comparison, most (∼94%) orientation-tuned neurons showed more or less cross-orientation inhibition, regardless of the relative stimulus contrasts. Only a small portion (∼8%) of them showed plaid facilitation at off-peak orientations. These results suggest separate subpopulations of plaid and grating responding neurons. Because most of these plaid neurons (∼95%) were insensitive to motion direction, they were plaid pattern detectors, not plaid motion detectors.

Aging Potentiates Lateral but Not Local Inhibition of Orientation Processing in Primary Visual Cortex.

Aging-related declines in vision can decrease well-being of the elderly. Concerning early sensory changes as in the primary visual cortex, physiological and behavioral reports seem contradictory. Neurophysiological studies on orientation tuning properties suggested that neuronal changes might come from decreased cortical local inhibition. However, behavioral results either showed no clear deficits in orientation processing in older adults, or proposed stronger surround suppression. Through psychophysical experiments and computational modeling, we resolved these discrepancies by suggesting that lateral inhibition increased in older adults while neuronal orientation tuning widths, related to local inhibition, stayed globally intact across age. We confirmed this later result by re-analyzing published neurophysiological data, which showed no systematic tuning width changes, but instead displayed a higher neuronal noise with aging. These results suggest a stronger lateral inhibition and mixed effects on local inhibition during aging, revealing a more complex picture of age-related effects in the central visual system than people previously thought.

Feature-Specific Organization of Feedback Pathways in Mouse Visual Cortex

Higher and lower cortical areas in the visual hierarchy are reciprocally connected [1]. Although much is known about how feedforward pathways shape receptive field properties of visual neurons, relatively little is known about the role of feedback pathways in visual processing. Feedback pathways are thought to carry top-down signals, including information about context (e.g., figure-ground segmentation and surround suppression) [2-5], and feedback has been demonstrated to sharpen orientation tuning of neurons in the primary visual cortex (V1) [6, 7]. However, the response characteristics of feedback neurons themselves and how feedback shapes V1 neurons' tuning for other features, such as spatial frequency (SF), remain largely unknown. Here, using a retrograde virus, targeted electrophysiological recordings, and optogenetic manipulations, we show that putatively feedback neurons in layer 5 (hereafter "L5 feedback") in higher visual areas, AL (anterolateral area) and PM (posteromedial area), display distinct visual properties in awake head-fixed mice. AL L5 feedback neurons prefer significantly lower SF (mean: 0.04 cycles per degree [cpd]) compared to PM L5 feedback neurons (0.15 cpd). Importantly, silencing AL L5 feedback reduced visual responses of V1 neurons preferringlow SF (mean change in firing rate: -8.0%), whereas silencing PM L5 feedback suppressed responses of high-SF-preferring V1 neurons (-20.4%). These findings suggest that feedback connections from higher visual areas convey distinctly tuned visual inputs to V1 that serve to boost V1 neurons' responses to SF. Such like-to-like functional organization may represent animportant feature of feedback pathways in sensorysystems and in the nervous system in general.

Interlayer Repulsion of Retinal Ganglion Cell Mosaics Regulates Spatial Organization of Functional Maps in the Visual Cortex.

In higher mammals, orientation tuning of neurons is organized into a quasi-periodic pattern in the primary visual cortex. Our previous model studies suggested that the topography of cortical orientation maps may originate from moiré interference of ON and OFF retinal ganglion cell (RGC) mosaics, but did not account for how the consistent spatial period of maps could be achieved. Here we address this issue with two crucial findings on the development of RGC mosaics: first, homotypic local repulsion between RGCs can develop a long-range hexagonal periodicity. Second, heterotypic interaction restrains the alignment of ON and OFF mosaics, and generates a periodic interference pattern map with consistent spatial frequency. To validate our model, we quantitatively analyzed the RGC mosaics in cat data, and confirmed that the observed retinal mosaics showed evidence of heterotypic interactions, contrary to the previous view that ON and OFF mosaics are developed independently.SIGNIFICANCE STATEMENT Orientation map is one of the most studied functional maps in the brain, but it has remained unanswered how the consistent spatial periodicity of maps could be developed. In the current study, we address this issue with our developmental model for the retinal origin of orientation map. We showed that local repulsive interactions between retinal ganglion cells (RGCs) can develop a hexagonal periodicity in the RGC mosaics and restrict the alignment between ON and OFF mosaics, so that they generate a periodic pattern with consistent spatial frequency for both the RGC mosaics and the cortical orientation maps. Our results demonstrate that the organization of functional maps in visual cortex, including its structural consistency, may be constrained by a retinal blueprint.

Selective enhancement of orientation tuning before saccades.

Saccadic eye movements cause a rapid sweep of the visual image across the retina and bring the saccade's target into high-acuity foveal vision. Even before saccade onset, visual processing is selectively prioritized at the saccade target. To determine how this presaccadic attention shift exerts its influence on visual selection, we compare the dynamics of perceptual tuning curves before movement onset at the saccade target and in the opposite hemifield. Participants monitored a 30-Hz sequence of randomly oriented gratings for a target orientation. Combining a reverse correlation technique previously used to study orientation tuning in neurons and general additive mixed modeling, we found that perceptual reports were tuned to the target orientation. The gain of orientation tuning increased markedly within the last 100 ms before saccade onset. In addition, we observed finer orientation tuning right before saccade onset. This increase in gain and tuning occurred at the saccade target location and was not observed at the incongruent location in the opposite hemifield. The present findings suggest, therefore, that presaccadic attention exerts its influence on vision in a spatially and feature-selective manner, enhancing performance and sharpening feature tuning at the future gaze location before the eyes start moving.

Aversive learning shapes neuronal orientation tuning in human visual cortex.

The responses of sensory cortical neurons are shaped by experience. As a result perceptual biases evolve, selectively facilitating the detection and identification of sensory events that are relevant for adaptive behaviour. Here we examine the involvement of human visual cortex in the formation of learned perceptual biases. We use classical aversive conditioning to associate one out of a series of oriented gratings with a noxious sound stimulus. After as few as two grating-sound pairings, visual cortical responses to the sound-paired grating show selective amplification. Furthermore, as learning progresses, responses to the orientations with greatest similarity to the sound-paired grating are increasingly suppressed, suggesting inhibitory interactions between orientation-selective neuronal populations. Changes in cortical connectivity between occipital and fronto-temporal regions mirror the changes in visuo-cortical response amplitudes. These findings suggest that short-term behaviourally driven retuning of human visual cortical neurons involves distal top–down projections as well as local inhibitory interactions.

Probability and the changing shape of response distributions for orientation.

Spatial attention and feature-based attention are regarded as two independent mechanisms for biasing the processing of sensory stimuli. Feature attention is held to be a spatially invariant mechanism that advantages a single feature per sensory dimension. In contrast to the prediction of location independence, I found that participants were able to report the orientation of a briefly presented visual grating better for targets defined by high probability conjunctions of features and locations even when orientations and locations were individually uniform. The advantage for high-probability conjunctions was accompanied by changes in the shape of the response distributions. High-probability conjunctions had error distributions that were not normally distributed but demonstrated increased kurtosis. The increase in kurtosis could be explained as a change in the variances of the component tuning functions that comprise a population mixture. By changing the mixture distribution of orientation-tuned neurons, it is possible to change the shape of the discrimination function. This prompts the suggestion that attention may not "increase" the quality of perceptual processing in an absolute sense but rather prioritizes some stimuli over others. This results in an increased number of highly accurate responses to probable targets and, simultaneously, an increase in the number of very inaccurate responses.

Relationship between the Local Structure of Orientation Map and the Strength of Orientation Tuning of Neurons in Monkey V1: A 2-Photon Calcium Imaging Study

A majority of neurons in the monkey primary visual cortex (V1) are tuned to stimulus orientations. Preferred orientations and tuning strengths vary among V1 neurons. The preferred orientation of neurons gradually changes across the cortex with occasional failures of this organization. How V1 neurons are arranged by the strength of orientation tuning and whether neuronal arrangement for tuning strength relates to orientation preference maps remains controversial. In this study, we performed in vivo two-photon calcium imaging in macaque V1 to examine the local spatial organization of orientation tuning at the level of single cells. We recorded fluorescence signals from individual neurons loaded with a calcium-sensitive dye in layer 2 and the uppermost tier of layer 3. The strength of orientation tuning was shared by nearby neurons, and changed across the cortex. The neurons with similar tuning strength were distributed across at least the entire thickness of layer 2. The tuning strength was weaker in regions where neurons exhibited heterogeneous preferred orientations, as compared with regions where neurons shared similar orientation preferences. Nearby direction-selective neurons often shared their preferred directions, although only a few neurons were direction selective in the layers examined. Thus, the orientation tuning strength of V1 neurons is partially predictable from the local structure of orientation map. The weaker orientation tuning we found in regions with heterogeneous orientation preferences suggests that orientation-independent interactions among local populations of V1 neurons play a critical role in determining their orientation tuning.

Effects of stimulus spatial frequency, size, and luminance contrast on orientation tuning of neurons in the dorsal lateral geniculate nucleus of cat

It is generally thought that orientation selectivity first appears in the primary visual cortex (V1), whereas neurons in the lateral geniculate nucleus (LGN), an input source for V1, are thought to be insensitive to stimulus orientation. Here we show that increasing both the spatial frequency and size of the grating stimuli beyond their respective optimal values strongly enhance the orientation tuning of LGN neurons. The resulting orientation tuning was clearly contrast-invariant. Furthermore, blocking intrathalamic inhibition by iontophoretically administering γ-aminobutyric acid (GABA)A receptor antagonists, such as bicuculline and GABAzine, slightly but significantly weakened the contrast invariance. Our results suggest that orientation tuning in the LGN is caused by an elliptical classical receptive field and orientation-tuned surround suppression, and that its contrast invariance is ensured by local GABAA inhibition. This contrast-invariant orientation tuning in LGN neurons may contribute to the contrast-invariant orientation tuning seen in V1 neurons.

Changes in V1 orientation tuning when blocking astrocytic glutamate transporters: models for extra- and intrasynaptic mechanisms

Several recent studies reveal a close interplay of neurons and astrocytes in information processing [1]. Astrocytes affect neural transmission by their control of glutamate concentrations by glutamate transporters [2] in direct vicinity to the synaptic cleft as well as extrasynaptically. An in-vivo study in primary visual cortex of ferrets [3] demonstrated the impact of astrocytic glutamate transport on neuronal response by blocking glutamate uptake pharmacologically with TBOA. A severe effect on neuronal orientation tuning curves to a full-field stationary grating, tilted every second by additional 10 degrees, was observed. While glutamate transporters were blocked tuning curves were broadened (HWHM increased from 28 deg. to 39 deg.) and response at preferred orientation was enhanced. However, it is unclear if the change in response originates from a prolongation of synaptic glutamate clearance or locally increased ambient glutamate concentration. Here we investigate in a computational model of ferret V1 how the intra- and extrasynaptic mechanisms affect orientation tuning. We implemented both mechanisms in a highly recurrent single layer 2d map model based on [4]. Neurons are placed on a 50 × 50 grid and connected to their neighbors by randomly drawn connections from a Gaussian. On every grid point an excitatory neuron is placed, additionally every third grid point is occupied by an inhibitory neuron. Peak conductivities for the four types of connections are set to be in the recurrent regime, exceeding the conductivity of the additional afferent input. The afferent input specific to each grid location induces a pinwheel-domain organization of neurons in the network. For the implementation of the synaptic mechanism the glutamate concentration follows a bi-exponential description with a decay time prolonged if glutamate uptake is reduced [2]. Synaptic glutamate then activates complex kinetic NMDA and AMPA-receptors [5]. The synaptic mechanism can however have a stronger influence on excitatory-to-excitatory or excitatory-to-inhibitory connections, as the different synapse geometries also affects glutamate clearance. Extrasynaptically ambient glutamate provides a constant NMDA-receptor mediated somatic current [6]. Here different sensitivities (different NMDA-receptor densities) of excitatory and inhibitory neurons to ambient glutamate may affects tuning. For both mechanism possible parameter combinations are assessed in a grid search. We observed that a selective increase in modalities towards inhibitory neurons even leads to a sharpening of orientation tuning. While selective enhancement in the modalities towards excitatory neurons leads first to a drop in orientation tuning and very fast for further increase to pathological firing rates. For both mechanisms the closest fit in orientation tuning (HWHM) to the experimental observation with TBOA was found for a stronger effect on excitatory neurons along with a simultaneous but weaker effect on the inhibitory population. While both models can explain the current data, they, however, provide different predictions for sub-threshold properties and for neurons close to pinwheels or domain centers.

Adaptation Shifts Preferred Orientation of Tuning Curve in the Mouse Visual Cortex

In frontalized mammals it has been demonstrated that adaptation produces shift of the peak of the orientation tuning curve of neuron following frequent or lengthier presentation of a non-preferred stimulus. Depending on the duration of adaptation the shift is attractive (toward the adapter) or repulsive (away from the adapter). Mouse exhibits a salt-and-pepper cortical organization of orientation maps, hence this species may respond differently to adaptation. To examine this question, we determined the effect of twelve minutes of adaptation to one particular orientation on neuronal orientation tuning curves in V1 of anesthetized mice. Multi-unit activity of neurons in V1 was recorded in a conventional fashion. Cells were stimulated with sine-wave drifting gratings whose orientation tilted in steps. Results revealed that similarly to cats and monkeys, majority of cells shifted their optimal orientation in the direction of the adapter while a small proportion exhibited a repulsive shift. Moreover, initially untuned cells showing poor tuning curves reacted to adaptation by displaying sharp orientation selectivity. It seems that modification of the cellular property following adaptation is a general phenomenon observed in all mammals in spite of the different organization pattern of the visual cortex. This study is of pertinence to comprehend the mechanistic pathways of brain plasticity.

Spatial warping by oriented line detectors can counteract neural delays

The slow speed of neural transmission necessitates that cortical visual information from dynamic scenes will lag reality. The “perceiving the present” (PTP) hypothesis suggests that the visual system can mitigate the effect of such delays by spatially warping scenes to look as they will in ~100 ms from now (Changizi, 2001). We here show that the Hering illusion, in which straight lines appear bowed, can be induced by a background of optic flow, consistent with the PTP hypothesis. However, importantly, the bowing direction is the same whether the flow is inward or outward. This suggests that if the warping is meant to counteract latencies, it is accomplished by a simple strategy that is insensitive to motion direction, and that works only under typical (forward-moving) circumstances. We also find that the illusion strengthens with longer pulses of optic flow, demonstrating motion integration over ~80 ms. The illusion is identical whether optic flow precedes or follows the flashing of bars, exposing the spatial warping to be equally postdictive and predictive, i.e., peri-dictive. Additionally, the illusion is diminished by cues which suggest the bars are independent of the background movement. Collectively, our findings are consistent with a role for networks of visual orientation-tuned neurons (e.g., simple cells in primary visual cortex) in spatial warping. We conclude that under the common condition of forward ego-motion, spatial warping counteracts the disadvantage of neural latencies. It is not possible to prove that this is the purpose of spatial warping, but our findings at minimum place constraints on the PTP hypothesis, demonstrating that any spatial warping for the purpose of counteracting neural delays is not a precise, on-the-fly computation, but instead a heuristic achieved by a simple mechanism that succeeds under normal circumstances.

Pull-Push Neuromodulation of LTP and LTD Enables Bidirectional Experience-Induced Synaptic Scaling in Visual Cortex

Neuromodulatory input, acting on G protein-coupled receptors, is essential for the induction of experience-dependent cortical plasticity. Here we report that G-coupled receptors in layer II/III of visual cortex control the polarity of synaptic plasticity through a pull-push regulation of LTP and LTD. In slices, receptors coupled to Gs promote LTP while suppressing LTD; conversely, receptors coupled to Gq11 promote LTD and suppress LTP. In vivo, the selective stimulation of Gs- or Gq11-coupled receptors brings the cortex into LTP-only or LTD-only states, which allows the potentiation or depression of targeted synapses with visual stimulation. The pull-push regulation of LTP/LTD occurs via direct control of the synaptic plasticity machinery and it is independent of changes in NMDAR activation or neuronal excitability. We propose these simple rules governing the pull-push control of LTP/LTD form a general metaplasticity mechanism that may contribute to neuromodulation of plasticity in other cortical circuits.

Dendritic organization of sensory input to cortical neurons in vivo

In sensory cortex regions, neurons are tuned to specific stimulus features. For example, in the visual cortex, many neurons fire predominantly in response to moving objects of a preferred orientation. However, the characteristics of the synaptic input that cortical neurons receive to generate their output firing pattern remain unclear. Here we report a novel approach for the visualization and functional mapping of sensory inputs to the dendrites of cortical neurons in vivo. By combining high-speed two-photon imaging with electrophysiological recordings, we identify local subthreshold calcium signals that correspond to orientation-specific synaptic inputs. We find that even inputs that share the same orientation preference are widely distributed throughout the dendritic tree. At the same time, inputs of different orientation preference are interspersed, so that adjacent dendritic segments are tuned to distinct orientations. Thus, orientation-tuned neurons can compute their characteristic firing pattern by integrating spatially distributed synaptic inputs coding for multiple stimulus orientations.

Paint It Black (or Red, or Green): Optical and Genetic Tools Illuminate Inhibitory Contributions to Cortical Circuit Function

Visual responses in the cortex are generated by the interactions of feedforward thalamic inputs with local inhibitory and excitatory circuitry, but the exact roles of different cell types in establishing response selectivity are unclear. Two papers in this issue of Neuron (Kerlin etal. and Runyan etal.) combine two-photon imaging with guided electrical recordings to measure orientation tuning in molecularly defined groups of interneuron types.
