Triplet state delocalization in a conjugated porphyrin dimer probed by transient electron paramagnetic resonance techniques.

The delocalization of the photoexcited triplet state in a linear butadiyne-linked porphyrin dimer is investigated by time-resolved and pulse electron paramagnetic resonance (EPR) with laser excitation. The transient EPR spectra of the photoexcited triplet states of the porphyrin monomer and dimer are characterized by significantly different spin polarizations and an increase of the zero-field splitting parameter D from monomer to dimer. The proton and nitrogen hyperfine couplings, determined using electron nuclear double resonance (ENDOR) and X- and Q-band HYSCORE, are reduced to about half in the porphyrin dimer. These data unequivocally prove the delocalization of the triplet state over both porphyrin units, in contrast to the conclusions from previous studies on the triplet states of closely related porphyrin dimers. The results presented here demonstrate that the most accurate estimate of the extent of triplet state delocalization can be obtained from the hyperfine couplings, while interpretation of the zero-field splitting parameter D can lead to underestimation of the delocalization length, unless combined with quantum chemical calculations. Furthermore, orientation-selective ENDOR and HYSCORE results, in combination with the results of density functional theory (DFT) calculations, allowed determination of the orientations of the zero-field splitting tensors with respect to the molecular frame in both porphyrin monomer and dimer. The results provide evidence for a reorientation of the zero-field splitting tensor and a change in the sign of the zero-field splitting D value. The direction of maximum dipolar coupling shifts from the out-of-plane direction in the porphyrin monomer to the vector connecting the two porphyrin units in the dimer. This reorientation, leading to an alignment of the principal optical transition moment and the axis of maximum dipolar coupling, is also confirmed by magnetophotoselection experiments.

Simultaneous acquisition of pulse EPR orientation selective spectra

High resolution pulse EPR methods are usually applied to resolve weak magnetic electron-nuclear or electron–electron interactions that are otherwise unresolved in the EPR spectrum. Complete information regarding different magnetic interactions, namely, principal components and orientation of principal axis system with respect to the molecular frame, can be derived from orientation selective pulsed EPR measurements that are performed at different magnetic field positions within the inhomogeneously broadened EPR spectrum. These experiments are usually carried out consecutively, namely a particular field position is chosen, data are accumulated until the signal to noise ratio is satisfactory, and then the next field position is chosen and data are accumulated. Here we present a new approach for data acquisition of pulsed EPR experiments referred to as parallel acquisition. It is applicable when the spectral width is much broader than the excitation bandwidth of the applied pulse sequence and it is particularly useful for orientation selective pulse EPR experiments. In this approach several pulse EPR measurements are performed within the waiting (repetition) time between consecutive pulse sequences during which spin lattice relaxation takes place. This is achieved by rapidly changing the main magnetic field, B0, to different values within the EPR spectrum, performing the same experiment on the otherwise idle spins. This scheme represents an efficient utilization of the spectrometer and provides the same spectral information in a shorter time. This approach is demonstrated on W-band orientation selective electron-nuclear double resonance (ENDOR), electron spin echo envelope modulation (ESEEM), electron–electron double resonance (ELDOR) – detected NMR and double electron–electron resonance (DEER) measurements on frozen solutions of nitroxides. We show that a factors of 3–6 reduction in total acquisition time can be obtained, depending on the experiment applied.

Principles and applications of ENDOR spectroscopy for structure determination in solution and disordered matrices

Both electron paramagnetic resonance (EPR) and electron nuclear double resonance (ENDOR) spectroscopies are extremely powerful and versatile methods for the characterisation of paramagnetic systems in biology, chemistry, and physics. However, by comparison to EPR, ENDOR remains a less widely used technique. In this tutorial review the basic principles of continuous wave ENDOR are described. The theory of orientation selective ENDOR, for structure determination in frozen solutions and powders, is then described. A range of examples, illustrating the type of information obtained from the ENDOR spectrum, is finally presented.

The17O Hyperfine Interaction in V17O(H217O)52+and Mn(H217O)62+Determined by High Field ENDOR Aided by DFT Calculations

The 17O hyperfine interaction of the water ligands and the V=O oxygen in the vanadyl aquo complex and of the water ligands in the Mn2+ aquo complex in a frozen solution were determined by W-band (95 GHz) electron-nuclear double resonance (ENDOR). Orientation selective ENDOR spectra of the vanadyl complex exhibited two distinct signals assigned to the vanadyl oxygen and the water ligands. The assignment of the signals was done based on the orientation of the principal axis system of the hyperfine interaction and through comparison with the hyperfine interaction predicted by DFT calculations. The latter showed good agreement with the experimental values thus providing clear evidence that the vanadyl oxygen is exchangeable. The interaction of the vanadyl oxygen, especially its anisotropic part, was significantly larger than that of the water oxygens due to a relatively large negative spin density on the oxygen p orbitals. The 17O hyperfine interaction of the water ligand in the Mn2+ complex was found to be similar to that of the water ligand in the vanadyl complex and was in good agreement with earlier single-crystal data. Here, due to the large thermal polarization, it was also possible to determine the absolute sign of the hyperfine coupling by selecting different EPR transitions.

An orientation-selected ENDOR and HYSCORE study of the Ni-C active state of Desulfovibrio vulgaris Miyazaki F hydrogenase

Electron nuclear double resonance (ENDOR) and hyperfine sublevel correlation spectroscopy (HYSCORE) are applied to study the active site of catalytic [NiFe]-hydrogenase from Desulfovibrio vulgaris Miyazaki F in the reduced Ni-C state. These techniques offer a powerful tool for detecting nearby magnetic nuclei, including a metal-bound substrate hydrogen, and for mapping the spin density distribution of the unpaired electron at the active site. The observed hyperfine couplings are assigned via comparison with structural data from X-ray crystallography and knowledge of the complete g-tensor in the Ni-C state (Foerster et al. (2003) J Am Chem Soc 125:83-93). This is found to be in good agreement with density functional theory calculations. The two most strongly coupled protons (a(iso)=13.7, 11.8 MHz) are assigned to the beta-CH(2) protons of the nickel-coordinating cysteine 549, and a third proton (a(iso)=8.9 MHz) is assigned to a beta-CH(2) proton of cysteine 546. Using D(2)O exchange experiments, the presence of a hydride in the bridging position between the nickel and iron-recently been detected for a regulatory hydrogenase (Brecht et al. (2003) J Am Chem Soc 125:13075-13083)-is experimentally confirmed for the first time for catalytic hydrogenases. The hydride exhibits a small isotropic hyperfine coupling constant (a(iso)=-3.5 MHz) since it is bound to Ni in a direction perpendicular to the z-axis of the Ni (3d(z)(2)) orbital. Nitrogen signals that belong to the nitrogen N(epsilon) of His-88 have been identified. This residue forms a hydrogen bond with the spin-carrying Ni-coordinated sulfur of Cys-549. Comparison with other hydrogenases reveals that the active site is essentially the same in all proteins, including a regulatory hydrogenase.

2D TRIPLE in orientationally disordered samples—a means to resolve and determine relative orientation of hyperfine tensors

The two-dimensional (2D) TRIPLE experiment provides correlations between electron-nuclear double resonance (ENDOR) frequencies that belong to the same electron-spin manifold, M S , and therefore allows to assign ENDOR lines to their specific paramagnetic centers and M S manifolds. This, in turn, also provides the relative signs of the hyperfine couplings. So far this experiment has been applied only to single crystals, where the cross-peaks in the 2D spectrum are well resolved with regular shapes. Here we introduce the application of the 2D TRIPLE experiment to orientationally disordered systems, where it can resolve overlapping powder patterns. Moreover, analysis of the shape of the cross-peaks shows that it is highly dependent on the relative orientation of the hyperfine tensors of the two nuclei contributing to this particular peak. This is done initially through a series of simulations and then demonstrated experimentally at a high field (W-band, 95 GHz). The first example concerned the 1H hyperfine tensors of the stable radical α,γ-bisdiphenylene-β-phenylallyl (BDPA) immobilized in a polystyrene matrix. Then, the experiment was applied to a more complex system, a frozen solution of Cu(II)-bis(2,2 ′:6 ′,2″ terpyridine) complex. There, the 2D TRIPLE experiment was combined with the variable mixing time (VMT) ENDOR experiment, which determined the absolute sign of the hyperfine couplings involved, and orientation selective ENDOR experiments. Analysis of the three experiments gave the hyperfine tensors of a few coupled protons.

Structure of copper(II)-histidine based complexes in frozen aqueous solutions as determined from high-field pulsed electron nuclear double resonance.

W-band (95 GHz) pulsed EPR and electron-nuclear double resonance (ENDOR) spectroscopic techniques were used to determine the hyperfine couplings of different protons of Cu(II)-histidine complexes in frozen solutions. The results were then used to obtain the coordination mode of the tridentate histidine molecule and to serve as a reference for Cu(II)-histidine complexation in other, more complex systems. Cu(II) complexes with L-histidine and DL-histidine-alpha-d,beta-d2 were prepared in H2O and in D2O, and orientation-selective W-band 1H and 2H pulsed ENDOR spectra of these complexes were recorded at 4.5 K. These measurements lead to the unambiguous assignment of the signals of the H alpha, H beta, imidazole H epsilon, and the exchangeable amino, Ham, protons. The 14N superhyperfine splitting observed in the X-band EPR spectrum and the presence of only one type of H alpha and H beta protons in the W-band ENDOR spectra show that the complex is a symmetric bis complex. Its g parallel is along the molecular symmetry axis, perpendicular to the equatorial plane that consists of four coordinated nitrogens in histamine-like coordinations (NNNN). Simulations of orientation-selective ENDOR spectra provided the principal components of the protons' hyperfine interaction and the orientation of their principal axes with respect to g parallel. From the anisotropic part of the hyperfine interaction of H alpha and H beta and applying the point-dipole approximation, a structural model was derived. An unexpectedly large isotropic hyperfine coupling, 10.9 MHz, was found for H alpha. In contrast, H alpha of the Cu(II)-1-methyl-histidine complex where only the amino nitrogen is coordinated, showed a much smaller coupling. Thus, the hyperfine coupling of H alpha can serve as a signature for a histamine coordination where both the amino and imino nitrogens of the same molecule bind to the Cu(II), forming a six-membered chelating ring. Unlike H alpha the hyperfine coupling of H epsilon is not as sensitive to the presence of a coordinated amino nitrogen of the same histidine molecule.

W- and X-Band Pulsed Electron Nuclear Double-Resonance Study of a Sodium−Nitric Oxide Adsorption Complex in NaA Zeolites

Pulsed electron nuclear double-resonance (ENDOR) spectroscopy at W- and X-band frequencies has been employed to characterize the structure of NO adsorption sites involving sodium cations in zeolite NaA. The principal values of the sodium hyperfine and nuclear quadrupole coupling tensors as well as the orientation of their principal axes system with respect to the g tensor coordinate frame of the Na+−NO adsorption complex could be determined by orientation-selective ENDOR spectroscopy. Such orientation-selective experiments benefit especially from the high spectral resolution at W-band frequencies. Furthermore, the sodium ENDOR spectrum is drastically simplified at high frequencies where the limit of weak hyperfine couplings is fulfilled. The dipolar sodium hyperfine coupling tensor reveals a bent structure of the formed adsorption complexes and gives access to the bond distance between the NO molecule and the cations. The 23Na nuclear quadrupole data indicate that the adsorption complexes are preferentially formed with the sodium ions at the six-membered rings of the NaA zeolite structure. An analysis of the sodium and nitrogen hyperfine coupling data shows that 96% of the unpaired electron spin density in the Na+−NO adsorption complex is localized in the nitrogen and oxygen 2pπ orbitals of the NO ligand molecule.

Proton ENDOR spectroscopy of cyanide-inhibited copper-zinc superoxide dismutase in randomly oriented samples

Electron nuclear double resonance (ENDOR) spectroscopy of frozen solutions of samples of CN-inhibited superoxide dismutase (CN-SOD) was applied in order to unravel the structural changes and the changes in electronic parameters which occur in native SOD upon CN ligation. Two samples were prepared from bovine sources in H 2O and in D 2O in order detect buffer exchangeable protons in hydrogen bonds. Two further samples from human sources were employed which contained specifically deuterated histidine carbon-bound proton positions. Measurements were taken at about 10 K from the proton ENDOR response at different magnetic field positions of the electron paramagnetic resonance spectrum. The experimental results from the samples at different deuteration levels were reproduced by simulations of orientation-selected ENDOR powder-type spectra. These data were integrated into a new structure of the copper site of CN-SOD together with positions of the g tensor axes and the spin densities in the complex. Comparison is made in relation to the native SOD in both structural and electronic parameters aspects and with the structural proposal from X-ray determination of CN-SOD single crystals.

Nitrogen and proton ENDOR of cytochrome d, hemin, and metmyoglobin in frozen solutions

Orientation-selected electron nuclear double resonance (ENDOR) spectra have been obtained from iron-linked nitrogens of the d heme of the cytochrome d oxidase complex in frozen solution. Heme d is a high-spin chlorin moiety which exists in the cytochrome d complex located in the inner membrane of Escherichia coli. This complex is a terminal oxidase in the E. coli aerobic respiratory chain. Comparison of the spectra of cytochrome d with hemin and metmyoglobin strongly implies that the d heme does not contain an axial nitrogen ligand. By computer simulation of the ENDOR spectra for each magnetic field, the nitrogen hyperfine interaction matrices and quadrupole coupling tensors to the three heme groups were determined
