Endophytic Penicillium oxalicum AUMC 14898 from Opuntia ficus-indica: A Novel Source of Tannic Acid Inhibiting Virulence and Quorum Sensing of Extensively Drug-Resistant Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a harmful pathogen that causes a variety of acute and chronic infections through quorum sensing (QS) mechanisms. The increasing resistance of this bacterium to numerous antibiotics has created a demand for new medications that specifically target QS. Endophytes can be the source of compounds with antibacterial properties. This research is the first to examine tannic acid (TA) produced by endophytic fungus as a potential biotherapeutic agent. A novel endophytic fungal isolate identified as Penicillium oxalicum was derived from the cladodes of Opuntia ficus-indica (L.). The species identification for this isolate was confirmed through sequencing of the internal transcribed spacer region. The metabolites from the culture of this isolate were extracted using ethyl acetate, then separated and characterized using chromatographic methods. This led to the acquisition of TA, a compound that shows strong anti-QS and excellent antibacterial effects against extensively drug-resistant P. aeruginosa strains. Furthermore, it was shown that treating P. aeruginosa with the obtained TA reduced the secretion of virulence factors controlled by QS in a dose-dependent manner, indicating that TA inhibited the QS characteristics of P. aeruginosa. Simultaneously, TA significantly inhibited the expression of genes associated with QS, including rhlR/I, lasR/I, and pqsR. In addition, in silico virtual molecular docking showed that TA could efficiently bind to QS receptor proteins. Our results showed that P. oxalicum could be a new source of TA for the treatment of infections caused by extensively drug-resistant P. aeruginosa.

Optimization, purification and characterization of laccase from a new endophytic Trichoderma harzianum AUMC14897 isolated from Opuntia ficus-indica and its applications in dye decolorization and wastewater treatment.

Hazardous synthetic dye wastes have become a growing threat to the environment and public health. Fungal enzymes are eco-friendly, compatible and cost-effective approachfor diversity of applications. Therefore, this study aimed to screen, optimize fermentation conditions, and characterize laccase from fungal endophyte with elucidating its ability to decolorize several wastewater dyes. A new fungal endophyte capable of laccase-producing was firstly isolated from cladodes of Opuntia ficus-indica and identified as T. harzianum AUMC14897 using ITS-rRNA sequencing analysis. Furthermore, the response surface methodology (RSM) was utilized to optimize several fermentation parameters that increase laccase production. The isolated laccase was purified to 13.79-fold. GFC, SDS-PAGE revealed laccase molecular weight at 72kDa and zymogram analysis elucidated a single band without any isozymes. The peak activity of the pure laccase was detected at 50°C, pH 4.5, with thermal stability up to 50°C and half life span for 4h even after 24h retained 30% of its activity. The Km and Vmax values were 0.1mM, 22.22µmol/min and activation energy (Ea) equal to 5.71kcal/mol. Furthermore, the purified laccase effectively decolorized various synthetic and real wastewater dyes. Subsequently, the new endophytic strain produces high laccase activity that possesses a unique characteristic, it could be an appealing candidate for both environmental and industrial applications.

Nutritional composition, functional and chemical characterization of Moroccan Opuntia ficus-indica cladode powder

ABSTRACT Opuntia ficus-indica, also known as the prickly pear, is a cactus belonging to the Cactaceae family. For centuries, this remarkable plant has been valued for its medicinal, environmental, and culinary uses. The primary objective of this research is to examine and contrast the chemical composition, functional attributes, and nutritional significance of Opuntia ficus-indica cladodes sourced from three distinct regions in Morocco: Oulad Boubker, Imzouren, and Skoura. According to the results, moisture content differs significantly between the three sites, from 88.21 ± 0.20% to 92.11 ± 0.10%, with an acid pH value. While the ash content remained consistent across all three samples. Carbohydrate content ranging from 55.40 ± 0.08% to 55.90 ± 0.60%, fat content from 2.30 ± 0.21 to 2.62 ± 0.09, protein from 7.54 ± 0.03% to 9.07 ± 0.16%, crude fibers from 20.52 ± 1.87% to 21.00 ± 0.20%, and soluble fibers from 15.08 ± 0.16% to 16.33 ± 1.25%. Additionally, total chlorophyll content varied between 102.64 ± 8.93 and 144.80 ± 3.08 mg.100 g−1 DM. The functional properties showed considerable differences between the three samples: the water holding capacity ranged from 109.58 ± 0.11 to 174.95 ± 0.06 g water.100 g−1 DM, the oil holding capacity from 129.27 ± 0.39 to 146.51 ± 0.39 g oil.100 g−1 DM and the solubility index from 25.75 ± 0.60 to 33.18 ± 0.15 g solids.100 g−1 DM. Mineral analysis revealed significant differences based on location. The concentrations of potassium exhibited a considerable elevation compared to those of magnesium, calcium, sodium, iron, and zinc. Moreover, the existence of diverse organic functional entities, including alcohols, ketones, and acids, was authenticated by Fourier-transform infrared spectroscopy. A differential distribution of the chemical composition of cladodes according to region was confirmed by nutritional, mineral and functional characterization and functional groups of cladodes.

Studies on the Comparative Response of Fibers Obtained from the Pastazzo of Citrus bergamia and Cladodes of Opuntia ficus-indica on In Vitro Model of Neuroinflammation.

Adhering to a healthy diet has a protective effect on human health, including a decrease in inflammatory diseases due to consuming fiber. The purpose of this manuscript was to obtain and compare two extracts based on fiber (BF and IF-C), derived from two plants particularly present in the Mediterranean region: bergamot (Citrus bergamia) and prickly pear (Opuntia ficus-indica). The parts used by these plants have been the "pastazzo" for the bergamot and the cladodes for the prickly pear. In addition to in vitro evaluations, the antioxidant activity was also measured on human neurons under inflammatory conditions. Furthermore, the extracts of interest were examined for their effects on the cell cycle and the regulation of pro-apoptotic proteins, caspase 9 and 3, induced by LPS. The results indicated that both extracts had a protective effect against LPS-induced damage, with BF consistently exhibiting superior functionality compared to IF-C. Moreover, the extracts can reduce inflammation, which is a common process of disease. By exploring this avenue, studying the consumption of dietary fiber could enhance our understanding of its positive effects, but additional experiments are needed to confirm this.

Arbuscular mycorrhizal fungi increased biomass, nutritional value, and cochineal resistance of Opuntia ficus-indica plants

BackgroundOpuntia ficus-indica (L.) Miller is dominantly growing on degraded soils in arid and semi-arid areas. The plants might establish a strong association with arbuscular mycorrhizal fungi (AMF) to adapt to nutrient, drought, and herbivore insect stress. The objective of this study was to investigate the effects of AMF inoculations and variable soil water levels (SWA) on the biomass, nutrient concentration, nutritional composition, and nutrient digestibility of the spiny and spineless O. ficus-indica by inducing resistance to cochineal stress. One mother Opuntia ficus-indica cladode was planted in a single pot in each field with 24 kg mixed soil. AMF inoculums were cultured in sorghum plants in a greenhouse and were inoculated in the planted cladodes. The planted cladodes were arranged using a completely randomized design (CRD) with three factors: AMF (present and absent); O. ficus-indica type (spiny and spineless) and four water treatments with 0–25% of plant available soil water (SWA), 25–50% of SWA, 50–75% of SWA, and 75–100% of SWA.ResultsDrought stress reduced the below and above-ground biomass, cladode nutrient content, nutritional composition, and in vitro dry matter digestibility (IVDMD) and in vitro organic matter digestibility (IVOMD). AMF colonization significantly increased biomass production with significant changes in the macro and micro-nutrient concentrations of O. ficus-indica. AMF inoculation significantly increased the IVDMD and IVOMD of both O. ficus-indica types by improving the biomass, organic matter (OM), crude protein (CP), and reduced fiber and ash contents. AMF-inoculated cladodes improved the nutrient concentrations of the cladodes. AMF caused an increase in biomass production, increased tolerance to cochineal stress, and improved nutrient concentration, nutritional composition, and nutrient digestibility performance of O. ficus-indica plants.ConclusionsAMF improved the performance of the O. ficus-indica plant to resist drought and cochineal stress and increased the biomass, nutrient concentration, nutritional composition, and nutrient digestibility. The potential of O. ficus-indica to adapt to cochineal stress is controlled by the macro and micro-nutrient concentration brought by the AMF association.

In vitro and in silico investigations of the pro-apoptotic activity of Opuntia ficus-indica cladode extracts against K562 cells

Abstract Objectives Cladodes of Opuntia ficus-indica (OFI) are thought to be an excellent source of bioactive compounds, making them an aspirant for the manufacture of health-promoting compounds. This study aims at exploring the pro-apoptotic effects of spiny and thornless OFI cladode extracts on the human immortalized myelogenous leukemia cell line (K562). Methods The ethanol extraction method was used for preparing cladode extract. Cytotoxicity was evaluated by MTT assays. Membrane permeability/damage was detected by annexin V-binding assays, and mitochondrial damage/alteration was detected by mitochondrial membrane potential measurements. The protein expression quantities of Bax and Bcl-2 were assessed by Western blotting. The pro-apoptotic potentials of the main spiny and thornless OFI extract constituents were investigated structurally and mechanistically using protein–ligand docking and interaction profiling. Results Spiny OFI extract displayed a stronger cytotoxic effect than thornless OFI extract on K562 cells. In silico findings agreed with the pro-apoptotic action observed in vitro. Conclusions Finally, our findings imply that OFI extracts cause apoptosis in K562 cells in order to have anti-cancer effects.

Opuntia ficus-indica (L.) Mill. belongs to the Cactaceae family and the genus Opuntia; it is a succulent plant that adapts to extreme climatic conditions. The aerial part of the plant consists of the cladodes, morphological changes of branches that appear green, are covered with thorns, and are essential to reduce excessive perspiration of water. The composition of cladodes is very varied, and the main constituents are water, fibers, polysaccharides, proteins, fatty acids, vitamins, sterols, minerals, and polyphenols. Polyphenols are responsible for many beneficial activities for human health, such as antioxidant, anti-inflammatory, anticancer, and nutritional properties. The purpose of this manuscript was to compare the properties of cladodes belonging to the same plant but with different stages of maturity. Relative extracts were tested both in vitro and on a cell line and antioxidant and anti-apoptotic properties were found. The antioxidant activity was tested by the Oxygen Radical Absorbance Capacity (ORAC) test, the 1,1-diphenyl-2-picrylhydrazil (DPPH) test, and the measurement of cellular accumulation of reactive oxygen species (ROS). Anti-apoptotic activity was evaluated by the annexin/PI assay and measurement of caspases 9 and 3 expression. The results obtained showed that the extracts considered possess antioxidant and anti-apoptotic properties. However, the different stages of maturity of cladodes are essential for the performance of both functions. In addition, important variations were made in the dissolution of the extracts that brought greater safety in their use. In conclusion, this manuscript provides further information on cladodes of Opuntia ficus-indica, which can be used as adjuvants in many human pathologies.

Antioxidant Activity and Seasonal Variations in the Composition of Insoluble Fiber from the Cladodes of Opuntia ficus-indica (L.) Miller: Development of New Extraction Procedures to Improve Fiber Yield.

Opuntia ficus-indica (L.) Miller is a plant belonging to the Cactaceae family adapted to live in environments characterized by long periods of drought and arid or desert climates. This plant is characterized by an aerial part composed of structures transformed by branches, called "cladodes", which are essential to reduce excessive perspiration of water and appear covered with thorns. The composition of the cladodes includes water, polysaccharides, fiber, proteins, vitamins, fatty acids, sterols, polyphenols, and minerals. The main purposes of this scientific work are (a) to compare the insoluble fiber (IF) extracted from the cladodes of O. ficus-indica belonging to the same plant but collected in different seasonal periods (winter and summer) and develop new extraction protocols that are able to improve the yield obtained and (b) evaluate the antioxidant potential of the fiber and study possible variations as a result of the extraction protocol chosen. The first objective was achieved (1) by measuring the amount of IF extracted from cladodes harvested in winter and summer (CW and CS, respectively) and (2) by modifying three variables involved in the fiber extraction protocol. To achieve the second objective, the following experiments were carried out: (1) measurement of the antioxidant potential of IF in CW and CS; (2) measurement of cellular reactive oxygen species; (3) measurement of the activity of some antioxidant enzymes; and (4) comparison of the polyphenol content in CW and CS. In conclusion, the results obtained showed that the IF extraction process can be improved, achieving a uniform yield regardless of seasonality; the antioxidant effect may vary depending on the extraction protocol.

Development of Eco-Friendly Biocomposite Films Based on Opuntia ficus-indica Cladodes Powder Blended with Gum Arabic and Xanthan Envisaging Food Packaging Applications.

Currently, food packaging is facing a critical transition period and a major challenge: it must preserve the food products' quality and, at the same time, it must meet the current requirements of the circular economy and the fundamental principles of packaging materials eco-design. Our research presents the development of eco-friendly packaging films based on Opuntia ficus-indica cladodes (OFIC) as renewable resources. OFIC powder (OFICP)-agar, OFICP-agar-gum arabic (GA), and OFICP-agar-xanthan (XG) blend films were eco-friendlily prepared by a solution casting method. The films' properties were investigated by scanning electron microscope (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (X-RD), and differential scanning calorimeter (DSC). Water solubility and moisture content were also determined. Morphology, thickness, molecular interactions, miscibility, crystallinity, and thermal properties, were affected by adjusting the gums (GA and XG) content and glycerol in the blend films. Moisture content increased with increasing glycerol and XG content, and when 1.5 g of GA was added. Water solubility decreased when glycerol was added at 50% and increased with increasing GA and XG content. FTIR and XRD confirmed strong intermolecular interactions between the different blend film compounds, which were reflected in the shifting, appearance, and disappearance of FTIR bands and XRD peaks, indicating excellent miscibility. DSC results revealed a glass transition temperature (Tg) below room temperature for all prepared blend films, indicating that they are flexible and soft at room temperature. The results corroborated that the addition of glycerol at 30% and the GA to the OFICP increased the stability of the film, making it ideal for different food packaging applications.

Opuntia ficus indica cladode as fermentation feedstock for lactic acid production by Lactobacillus acidophilus LA 5

Opuntia ficus-indica cladodes are by-products which contain high amounts of fibres, bioactive and functional compounds. Given their high annual productivity per hectare, cladodes represent a cheap and suitable substrate, usable for fermentation processes. We investigated their potential as a substrate for the growth and production of lactic acid from Lactobacillus acidophilus LA-5. A separate hydrolysis and fermentation was performed. The concentration of reducing sugars obtained after the dilute acid and enzymatic hydrolysis was 28.45 g/L. The lactobacillus count ranged from 6.03 to 8.1 log CFU/mL, whereas lactic acid yield and productivity were 0.63 g/g and 0.73 g/L h, respectively. The maximum lactic acid concentration was found to be 17.5 g/L. This study reports the possibility of using the O. ficus indica cladode for lactic acid production by LA-5 aiming to reduce costs for sustainable industrial production.

Optimization of ultrasounds assisted extraction of polysaccharides from cladodes of Opuntia ficus-indica using response surface methodology

In this work, polysaccharides are extracted from cladodes of Opuntia ficus-indica using Ultrasounds-Assisted Extraction (UAE). UAE operating conditions are optimized by a Face Centered Central Composite Response Surface Design (FCCRD) with four variables at three levels. The optimal operating conditions are obtained using Response Surface Methodology (RSM) and are the following: Solid-Liquid ratio SL = 1:10 w/v, pH = 2.5, time t = 20 min, and temperature T = 65 °C. The predicted extraction yield (12.07 ± 1.7 % dw) is in line with the experimental one (11.32 ± 0.25 % dw). The resulting extract is composed mainly of galacturonic acid (25.55 ± 0.30 %), the primary constituent of pectic material, followed by arabinose (14.34 ± 0.01 %) and galactose (13.5 ± 0.22 %). The Esterification Degree (ED) is found 42.84 ± 0.48 %, indicating the extract is Low-Methylated (LM). The FT-IR spectrum demonstrates that the extract has several peaks typical of polysaccharides. Total Phenolic Content (TPC) is equal to 41.33 ± 3.53 μgGAE/g (dw) and the anti-radical ability against the DPPH radical scavenging activity achieves 95.56 % at a concentration equal to 2 mg/mL, demonstrating that the extract is a polysaccharide functionalized with polyphenols. These results can encourage the use of the extract as a new ingredient in nutraceutical cosmetic applications.

Cladodes of Opuntia ficus-indica (L.) as a source of bioactive compounds in dairy products

Recently, the interest in improving livestock product nutraceutical profiles through sustainable feeding systems has increased. In this context, the overall quality and nutraceutical profiles were examined in dairy products obtained from 16 lactating Cinisara cows given an integrated feed in dry season with Opuntia ficus-indica cladodes. Two homogeneous groups of cows (milk yield: 6.3 ± 1.5 kg; body weight: 213 ± 55 kg) were fed with 2 different diets (CON: pasture and wheat bran; OFI: pasture, wheat bran, and cladodes), according to a 2 × 2 Latin square design. The bulk milk produced during the study was used to make Caciotta cheese and was analyzed at 0, 15, and 30 storage days. Milk and cheese samples were analyzed for chemical, physical, and microbiological traits. The nutraceutical and sensorial profiles, as well as the antioxidant capacity, were also determined in the final products. For milk, the urea content in individual samples was reduced in OFI but not in CON. In the cheese, integration of cladodes did not influence the starter cultures development with 2 strains of Streptococcus thermophilus, but it caused a higher content of polyphenols and a consequent greater antioxidant capacity and a change in the fatty acids profile. In particular, the caprylic, capric, lauric, myristic, and palmitic fatty acids were higher, as were the petroselinic, vaccenic, rumenic, and α-linolenic fatty acids. In contrast, the oleic and the γ-linolenic fatty acids were lower for OFI. The cheese from the OFI group showed better overall acceptability, and a higher yellow color, odor intensity, and butter flavor. The multivariate analysis well distinguished the cheeses belonging to the 2 groups. Further investigations should be conducted to formulate well-balanced diets that include cladodes for Cinisara lactating cows, but also to determine the content of other important bioactive compounds in fresh and treated cladodes, as well as their effects on animal welfare and production.

Precipitation solvents effect on the extraction of mucilaginous polysaccharides from Opuntia ficus-indica (Cactaceae): Structural, functional and rheological properties

This paper discusses mucilage extracted from Opuntia ficus-indica (Cactaceae) cladodes, also known as Barbary fig. Mucilaginous polysaccharides were extracted in hot water and purified with two distinct organic solvents, ethanol and isopropanol. The mucilaginous extract yield precipitated in isopropanol "MUIS″ (i.e. 23.2%) is higher than that of ethanol "MUET″ (i.e. 19%). The biochemical composition of mucilaginous extracts shows that the change in precipitation solvents has no significant effect. In addition, the microstructure of mucilaginous extracts exhibits amorphous and porous structures, which favor their capacity to retain moisture. The mucilaginous extracts analysis by ion chromatography reveals that after acid hydrolysis, MUIS has a higher total of sugars (46.9%) than MUET (37.6%). Mucilaginous extracts vary in their main content of monomers such as arabinose, galactose, glucose, and xylose as well as in the uronic acid fraction. The contact angle measurements were also established in order to investigate mucilaginous extracts’ hydrophilic or hydrophobic surfaces. Three tested liquids were used namely: distilled water, diiodomethane, and ethylene glycol. The X-ray diffraction analysis displayed that the precipitation solvent does not affect the amorphous structure. The TGA analyses of extracts under an oxidative atmosphere reveal that the oxygen flow accelerates the kinetics of degradation. Under nitrogen flow, mucilaginous extracts have intrinsic thermal resistance in specific temperature ranges. The DSC results show that the polysaccharide purification technique influences the glass temperature (Tg), which is around 53 °C and 61 °C for MUET and MUIS, respectively. In an aqueous solution, the mucilaginous extracts are highly heterogeneous, and the precipitation solvents and temperature affect their particle size. Mucilaginous extracts were concentration-dependent and reduced the surface tension of water to 43.5 mN/m and 30 mN/m for MUET and MUIS at 7 w/v%, respectively. The rheological analyses show that a non-Newtonian shear-thinning behavior was observed. The Ostwald-de Waele model successfully predicted the viscosity-shear rate relationship. The consistency coefficient (k) of MUIS at 7 w/v% (0.117 Pa.s) is higher than MUET at 7 w/v% (0.081 Pa.s). The critical concentrations (C*) of MUET and MUIS are 1.75% and 3.45 w/v%, respectively.

Egyptian Opuntia ficus-indica (OFI) Residues: Recovery and Characterization of Fresh Mucilage from Cladodes

The utilization of biopolymers gained recent attention worldwide due to their effective role in producing eco-friendly and economical products. Mucilage from Opuntia ficus-indica (OFI, and other succulent plants) has the capacity to absorb huge amounts of water, forming a viscous colloid with interesting rheological properties. It also had the potential ability for use as food additives or food technological products. We, therefore, aimed at extracting and characterizing pure liquid mucilage from the cladodes of OFI (Egyptian variety) using multidimensional approaches including HPLC-RID, FTIR, XRD, NMR, DSC, and TGA assays. The chemical composition, total polyphenols, total flavonoids, total flavonols, and antioxidant capacity by DPPH and ABTS assays were also measured. HPLC-RID analysis showed that the mucilage’s sugars are composed of glucose, xylose, rhamnose, galacturonic acid, arabinose, galactose, and fucose with total sugars of 0.375 mg/mL, which were subsequently confirmed by FTIR and NMR results. The specific signals of these sugars were also observed in 13C and 1H NMR spectra and their chemical fingerprint was obtained by FTIR. XRD patterns showed that mucilage has high calcium content, and the glass transition temperature was observed at 85.9 °C. Meanwhile, total phenolic content, flavonoids, and flavonols were about 7.96 mg GAE/g FW, 3.61 mg QE/g FW, and 1.47 mg QE/g FW, respectively. The antioxidant capacity of mucilage was around 26.15 and 22.5 µmol TE/g FW for DPPH and ABTS methods, respectively. It can be concluded that OFI cladode mucilage showed promising properties that would improve and open new opportunities and trends in the food, pharmaceutical, cosmetic, and other industries. The use of pure liquid mucilage could also be economically profitable due to its low cost, availability, and effectiveness for many edible applications.

Single and Binary Removals of Pb(II) and Cd(II) with Chemically Modified Opuntia ficus indica Cladodes.

In this study, cladodes of Opuntia ficus indica (OFIC), chemically modified with NaOH (OFICM), have been prepared, characterized, and tested as an effective biomass to remove Pb(II) and/or Cd(II) from aqueous media. At an optimum pH of 4.5, the adsorption capacity, qe, of treated OFICM was almost four times higher than that of untreated OFIC. The maximum adsorption capacities (qmax) in the single removal of Pb(II) and Cd(II) were 116.8 and 64.7 mg g-1, respectively. These values were 12.1% and 70.6% higher than those for the corresponding qmax in binary removal, which indicates the strong inhibitive effect of Pb(II) on the co-cation Cd(II) in a binary system. Structural and morphological characterization have been carried out by FTIR, SEM/EDX, and point of zero charge (pHPZC) measurements. The SEM/EDX results confirmed that the metals are adsorbed on the surface. The presence of C-O, C=O, and COO- functional groups were identified by FTIR on both OFIC and OFICM surfaces. On the other hand, we found that the adsorption processes followed the pseudo-second-order kinetics for both single and binary systems, with a fast biosorption rate of Pb(II) and Cd(II). The equilibrium data (adsorption isotherms) were better described by Langmuir and modified-Langmuir models for single and binary systems, respectively. A good regeneration of OFICM was obtained with an eluent of 0.1 M HNO3. Therefore, OFICM can be efficiently reused to remove Pb or Cd, up to three times.

Potential of Opuntia Ficus-Indica Cladodes In M’sila (North ALGERIA) as feed for ruminants: chemical composition and in vitro assessment

ABSTRACT The main objective of the present study was to investigate a strategy to bioconverte spineless cactus Opuntia Ficus-Indica (O. ficus indica) Cladodes, Medicago sativa L. (M. sativa) and barley straw (H. vulgare) based on their chemical composition, in vitro digestibility and fermentation parameters. The CP ranged from 51.41 to 156.09 g/kg DM, which was especially high within M. sativa and low in H. vulgare. O. ficus indica had the highest values in free condensed tannins. The most digestible plant sample was that from M. sativa and O.ficus indica. The maximum in vitro gas production was recorded with M. sativa and H. vulgare. The same trend was observed with the total gas production (144 h). Based on the results, it can be concluded that O. ficus indica could replace common plant species, especially in the arid and semi-arid areas in which the livestock production section frequently suffers low efficiency and big losses.

Towards local circular economy through Opuntia Ficus Indica cladodes conversion into renewable biofuels and biochars: Product distribution and kinetic modelling

Opuntia Ficus Indica (OFI) cladodes slow pyrolysis was conducted in a fixed-bed reactor at various temperature (500–600–700 °C) and residence time (30–60–120 min) with a constant heating rate (10 °C min−1). The highest biochar yield (40.08%) was obtained at 500°C and 30 min whereas more liquid (35.03%) and gaseous (31.08%) products were produced at 700 °C and 120 min. The produced biochar is an alkaline carbon rich with minerals and nutrient (N, P, K, Ca, Mg) that could be applied in agriculture to balance acidic soil. The obtained bio-oil and syngas could be used as green biofuels. The kinetic study using Vyazovkin and FWO indicates an activation energy (Eα) values of 36.05–224.41 and 47.87–242.36 kJ mol−1 respectively, with a good prediction (R2 ≥ 0.9). This integrated approach of local OFI biomass conversion into biofuels and bio-based materials is an opportunity for the implementation of a local circular economy in Tunisia.

Growth Performance, Carcass Characteristics and Economic Viability of Nguni Cattle Fed Diets Containing Graded Levels of Opuntia ficus-indica

Given the severe droughts caused by global warming, smallholder beef cattle farmers are faced with serious forage and feed scarcity. This becomes worse for resource-poor farmers who cannot afford commercial feeds. It is therefore crucial to assess the use of low-cost alternative feed resources to supplement free-range beef cattle and ensure sustainable livestock production in ways that stimulate free-range beef farmers’ participation in mainstream beef market. In an attempt to improve free-range beef cattle herds and explore the economic viability of utilizing Opuntia ficus-indica (spineless cactus) cladodes as a supplementary feed, we investigated the impact of cactus diets on animal growth performance and carcass characteristics of Nguni cattle heifers. Four dietary treatments were randomly assigned to thirty-two heifers aged 24 months, weighing, on average, 172.20 ± 27.10 kg, with each dietary treatment replicated to eight individually penned heifers for 90 days. The dietary treatments were control diet (pasture-based energy + protein sources), 10% cactus diet, 20% cactus diet and commercial diet (crop-based energy and commercial protein source). The energy concentration of the control diet was 9.35 MJ/Kg DM and the cactus was included on dry matter basis during formulation of compound diets. Thus, cactus was administered in a dry rather than wet form. The animals were confined in feeding pens 24/7 without access to pasture, with feed and water provided ad libitum. The heifers fed commercial and control diets attained significantly (p < 0.05) higher dry matter intake, average daily gains, fat thickness, carcass conformation scores and lower feed conversion ratio than those fed cactus diets. However, the final body weight gains and carcass weights, rib-eye muscle area and meat pH45min and 24h were comparable (p > 0.05) between heifers fed cactus diets and those fed commercial and control diets. The 10 and 20% cactus diets had greater gross margins (p < 0.05) of $17.47 and $18.62, respectively, than the other diets, due largely to reduced total variable costs. The comparability of carcass traits of heifers fed cactus diets and those fed non-cactus diets as well as higher economic returns from cactus inclusion warrants the use of cactus diets, particularly during drought when commercial feed prices rise.
