Island plants with newly discovered reproductive traits have higher capacity for uniparental reproduction, supporting Baker’s law

Uniparental reproduction is advantageous when lack of mates limits outcrossing opportunities in plants. Baker’s law predicts an enrichment of uniparental reproduction in habitats colonized via long-distance dispersal, such as volcanic islands. To test it, we analyzed reproductive traits at multiple hierarchical levels and compared seed-set after selfing and crossing experiments in both island and mainland populations of Limonium lobatum, a widespread species that Baker assumed to be self-incompatible because it had been described as pollen-stigma dimorphic, i.e., characterized by floral morphs differing in pollen-surface morphology and stigma-papillae shape that are typically self-incompatible. We discovered new types and combinations of pollen and stigma traits hitherto unknown in the literature on pollen-stigma dimorphism and a lack of correspondence between such combinations and pollen compatibility. Contrary to previous reports, we conclude that Limonium lobatum comprises both self-compatible and self-incompatible plants characterized by both known and previously undescribed combinations of reproductive traits. Most importantly, plants with novel combinations are overrepresented on islands, selfed seed-set is higher in islands than the mainland, and insular plants with novel pollen-stigma trait-combinations disproportionally contribute to uniparental reproduction on islands. Our results thus support Baker’s law, connecting research on reproductive and island biology.

QUANTIFYING BILATERAL INFECTION PATTERNS IN THE TREMATODE ALLOGLOSSIDIUM RENALE.

Within-host distributions of parasites can have relevance to parasite competition, parasite mating, transmission, and host health. We examined the within-host distribution of the adult trematode Alloglossidium renale infecting the paired antennal glands of grass shrimp. There are 4 possible parasite distributions for infections of paired organs: random, uniform, biased aggregation to 1 particular organ (e.g., left vs. right), or inconsistently biased (aggregated, but does not favor 1 side). Previous work has shown that morphological asymmetries in hosts can lead to biased infections of paired organs. Apparent symmetry between the antennal glands of grass shrimp leads to the prediction that there would be no bias for 1 particular organ. However, an alternative prediction stems from the fact that A. renale is hermaphroditic: aggregation between glands would increase outcrossing opportunities and thus, avoid inbreeding via self-mating. Existing methods to test for an overall pattern did not apply to the A. renale system because of low-intensity infections as well as many 0 values for abundance per unit of the antennal gland. Hence, we used Monte Carlo simulations to determine if the observed overall patterns differed from those expected by randomly allocating parasites into groups of 2. We found that in 3 of 4 data sets, A. renale infections did not deviate from random distributions. The fourth data set had a more uniform pattern than expected by chance. As there was no aggregation between glands and the proportion of worms in single gland infections did not differ from that expected by chance alone, we found no evidence of inbreeding avoidance as might be manifested via a within-host distribution. Given the large proportion of worms in single infections, we predict as a major evolutionary outcome that populations of A. renale will be largely inbred.

The impact of indoor residual spraying on Plasmodium falciparum microsatellite variation in an area of high seasonal malaria transmission in Ghana, West Africa.

Here, we report the first population genetic study to examine the impact of indoor residual spraying (IRS) on Plasmodium falciparum in humans. This study was conducted in an area of high seasonal malaria transmission in Bongo District, Ghana. IRS was implemented during the dry season (November–May) in three consecutive years between 2013 and 2015 to reduce transmission and attempt to bottleneck the parasite population in humans towards lower diversity with greater linkage disequilibrium. The study was done against a background of widespread use of long‐lasting insecticidal nets, typical for contemporary malaria control in West Africa. Microsatellite genotyping with 10 loci was used to construct 392 P. falciparum multilocus infection haplotypes collected from two age‐stratified cross‐sectional surveys at the end of the wet seasons pre‐ and post‐IRS. Three‐rounds of IRS, under operational conditions, led to a >90% reduction in transmission intensity and a 35.7% reduction in the P. falciparum prevalence (p < .001). Despite these declines, population genetic analysis of the infection haplotypes revealed no dramatic changes with only a slight, but significant increase in genetic diversity (H e: pre‐IRS = 0.79 vs. post‐IRS = 0.81, p = .048). Reduced relatedness of the parasite population (p < .001) was observed post‐IRS, probably due to decreased opportunities for outcrossing. Spatiotemporal genetic differentiation between the pre‐ and post‐IRS surveys (D = 0.0329 [95% CI: 0.0209 – 0.0473], p = .034) was identified. These data provide a genetic explanation for the resilience of P. falciparum to short‐term IRS programmes in high‐transmission settings in sub‐Saharan Africa.

The opportunity for outcrossing varies across the geographic range of the primarily selfing Clarkia xantiana ssp. parviflora.

The timing of self-fertilization has potentially important consequences for the trajectory of mating system evolution, the opportunity for outcrossing, and the maintenance of genetic variation in populations. For primarily selfing taxa, it remains poorly understood as to how floral variation influences the opportunity for outcrossing and whether those attributes vary among populations across geographic ranges. We examined variation in floral traits (herkogamy, protandry, flower size, stigma stage at anthesis, timing of stigma receptivity) in seven populations of Clarkia xantiana ssp. parviflora, a primarily selfing taxon, spanning from the western to eastern margins of its distribution. We also performed experimental emasculations and pollinations (followed by stigma severing) to quantify the extent of opportunities for outcrossing across flower development. There was marked among-population variation in all floral traits, particularly between far eastern and western populations. Emasculation experiments showed that the eastern populations had minimal autonomous selfing, but western populations had high rates of selfing within 24 h after anthesis. Population variation in autofertility was significantly predicted by floral trait variation, especially protandry and petal size. Greater protandry both extended the time over which outcrossing could potentially occur and reduced the probability of autonomous selfing, suggesting that there may be a tradeoff that results in fitness loss when pollinator visits are not common. The east-west pattern of differentiation in some floral traits parallels that of postglacial range expansion, suggesting that selection on the mating system may have been strong in the process of range expansion.

Evidence for inter-population variation in waiting times in a self-fertilizing flatworm

ABSTRACT Self-fertilization (selfing) is thought to evolve as a way of guaranteeing reproduction when mate availability is low. Nevertheless, because it often leads to inbreeding depression (ID), many hermaphrodites postpone reproduction via selfing when mates are scarce to wait for potential outcrossing opportunities. Owing to varying demographic histories and mate availabilities causing differing optimal selfing propensities, species exhibiting such delayed selfing may show inter-population variation in waiting time (WT) to commence selfing. To test this, we quantified WT across two natural populations of the hermaphroditic flatworm Macrostomum hystrix. In one of the populations we additionally estimated ID and sex allocation (SA), which are expected to covary with selfing propensity. We found no evidence for a WT in either population, but nevertheless observed both ID in the selfed progeny and plasticity in SA in the population we measured these traits. Our results contrast starkly to the pattern previously observed in another natural M. hystrix population, which exhibited a substantial WT and a corresponding reduction in selfed offspring fitness. Our study establishes the presence of natural diversity in selfing propensity in this species and highlights the importance of characterizing such among-population variation to further understand mating system evolution.

Variable opportunities for outcrossing result in hotspots of novel genetic variation in a pathogen metapopulation.

Many pathogens possess the capacity for sex through outcrossing, despite being able to reproduce also asexually and/or via selfing. Given that sex is assumed to come at a cost, these mixed reproductive strategies typical of pathogens have remained puzzling. While the ecological and evolutionary benefits of outcrossing are theoretically well-supported, support for such benefits in pathogen populations are still scarce. Here, we analyze the epidemiology and genetic structure of natural populations of an obligate fungal pathogen, Podosphaera plantaginis. We find that the opportunities for outcrossing vary spatially. Populations supporting high levels of coinfection -a prerequisite of sex - result in hotspots of novel genetic diversity. Pathogen populations supporting coinfection also have a higher probability of surviving winter. Jointly our results show that outcrossing has direct epidemiological consequences as well as a major impact on pathogen population genetic diversity, thereby providing evidence of ecological and evolutionary benefits of outcrossing in pathogens.

Pollination biology and breeding system in five nocturnal species of Oenothera (Onagraceae): reproductive assurance and opportunities for outcrossing

The capacity to produce seed, both by selfing and outcrossing, or mixed mating strategies, is considered a mechanism for overcoming unpredictable pollinator availability. In the present study, we investigate breeding system, insect visitations and the role of insect visitors in the pollination of five species of Oenothera subsect. Oenothera. Field experiments showed that autonomous selfing occurs at bud stage, prior to the opening of the flower. Control flowers showed similar seed set to hand-pollinated flowers, whereas emasculated flowers and those subject to open pollination set fewer seed. These results are consistent with the hypothesis that the investigated Oenothera exhibit a great capacity for autonomous selfing and that selfing is selected in order to provide reproductive assurance. Although flowers were visited mostly by nocturnal lepidopterans, these insects did not precipitate pollination and are thus considered nectar thieves. Conversely, analysis of pollen loads and behavior during foraging by diurnal insect visitors revealed that honeybees and bumblebees are the probable pollinators. We conclude that production of flowers capable of autonomous selfing at bud stage, followed by anthesis and opportunities for outcrossing, probably improves the invasive potential of these Oenothera in Europe, together with a rapid increase in their populations, even when pollinators are scarce.

The best of both worlds? A review of delayed selfing in flowering plants.

In a seminal body of theory, Lloyd showed that the fitness consequences of selfing will depend on its timing in anthesis. Selfing that occurs after opportunities for outcrossing or pollen dispersal can provide reproductive assurance when pollinators are limited and is expected to incur little cost, even when inbreeding depression is high. As a result, delayed selfing is often interpreted as a "best-of-both-worlds" mating system that combines the advantages of selfing and outcrossing. We surveyed 65 empirical studies of delayed selfing, recording floral mechanisms and examining information on inbreeding depression, autofertility, and other parameters to test the support for delayed selfing as a best-of-both-worlds strategy. Phylogenetic distribution of the diverse floral mechanisms suggests that some basic floral structures may predispose plant taxa to evolve delayed selfing. Delayed selfing appears to serve as a best-of-both-worlds strategy in some but not all species. While the capacity for autonomous selfing is often high, it is lower, in some cases, than in related species with earlier modes of selfing. In other delayed-selfers, low inbreeding depression and reduced investment in corollas and pollen suggest limited benefits from outcrossing. Despite a growing literature on the subject, experimental evidence for delayed selfing is limited and major gaps in knowledge remain, particularly with respect to the stability of delayed selfing and the conditions that may favor transitions between delayed and earlier selfing. Finally, we suggest a potential role of delayed selfing in facilitating transitions from self-incompatibility to selfing.

Does stigma curvature promote delayed selfing? An experimental investigation in Triodanis perfoliata (Campanulaceae).

Self-fertilisation that is delayed until after opportunities for outcrossing have ceased has been argued to provide both the reproductive assurance benefits of selfing and the genetic advantages of outcrossing. In the Campanulaceae, presentation of pollen on stylar hairs and progressive stigma curvature have been hypothesised to facilitate delayed selfing, but experimental tests are lacking. Stigma curvature is common in Campanula, a genus largely characterised by self-incompatibility, and therefore is unlikely to have initially evolved to promote self-fertilisation. In derived self-compatible species, however, stigma curvature might serve the secondary function of delayed selfing. We investigated delayed selfing in Triodanis perfoliata, a self-compatible relative of Campanula. Using floral manipulation experiments and pollen tube observations, we quantified the extent and timing of self-pollination. Further, we hypothesised that, if stigma curvature provides the benefit of delayed selfing in Triodanis, selection should have favoured retention of self-pollen through the loss of a stylar hair retraction mechanism. Results of a stigma removal experiment indicated that autonomous selfing produces partial seed set, but only some selfing was delayed. Pollen tube observations and a flower senescence assay also supported the finding of partial delayed selfing. Scanning electron microscopy revealed that pollen-collecting hairs retract during anthesis, which may limit the extent of delayed selfing. Delayed selfing appeared to be only partially effective in T.perfoliata. The stylar hair retraction in this species would seem to contradict selection for selfing. We suggest that caution and rigour are needed in interpreting floral traits as adaptive mechanisms for delayed selfing.

Unfertilized ovary pushes wheat flower open for cross-pollination.

Bread wheat is strongly autogamous; however, an opportunity for outcrossing occurs when self-pollination fails and florets open. The first phase of floret opening at anthesis is short and induced by lodicule turgidity. Some wheat florets re-open post-anthesis for several days, known as the 'second opening', for which the underlying mechanisms are largely unknown. We performed detailed physiological, anatomical, and histological investigations to understand the biological basis of the flower opening process. Wheat florets were observed open when the ovary was unfertilized. Unfertilized ovaries significantly increased in radial size post-anthesis, pushing the lemma and palea apart to open the florets. The absence of fertile pollen was not directly linked to this, but anther filament elongation coincided with initiation of ovary swelling. The pericarp of unfertilized ovaries did not undergo degeneration as normally seen in developing grains, instead pericarp cells remained intact and enlarged, leading to increased ovary radial size. This is a novel role for the ovary pericarp in wheat flower opening, and the knowledge is useful for facilitating cross-pollination in hybrid breeding. Ovary swelling may represent a survival mechanism in autogamous cereals such as wheat and barley, ensuring seed set in the absence of self-fertilization and increasing genetic diversity through cross-pollination.

Evolutionary consequence of a change in life cycle complexity: A link between precocious development and evolution toward female-biased sex allocation in a hermaphroditic parasite.

The evolutionary consequences of changes in the complex life cycles of parasites are not limited to the traits that directly affect transmission. For instance, mating systems that are altered due to precocious sexual maturation in what is typically regarded as an intermediate host may impact opportunities for outcrossing. In turn, reproductive traits may evolve to optimize sex allocation. Here, we test the hypothesis that sex allocation evolved toward a more female-biased function in populations of the hermaphroditic digenean trematode Alloglossidium progeneticum that can precociously reproduce in their second hosts. In these precocious populations, parasites are forced to self-fertilize as they remain encysted in their second hosts. In contrast, parasites in obligate three-host populations have more opportunities to outcross in their third host. We found strong support that in populations with precocious development, allocation to male resources was greatly reduced. We also identified a potential phenotypically plastic response in a body size sex allocation relationship that may be driven by the competition for mates. These results emphasize how changes in life cycle patterns that alter mating systems can impact the evolution of reproductive traits in parasites.

Influences of clonality on plant sexual reproduction

Flowering plants possess an unrivaled diversity of mechanisms for achieving sexual and asexual reproduction, often simultaneously. The commonest type of asexual reproduction is clonal growth (vegetative propagation) in which parental genotypes (genets) produce vegetative modules (ramets) that are capable of independent growth, reproduction, and often dispersal. Clonal growth leads to an expansion in the size of genets and increased fitness because large floral displays increase fertility and opportunities for outcrossing. Moreover, the clonal dispersal of vegetative propagules can assist “mate finding,” particularly in aquatic plants. However, there are ecological circumstances in which functional antagonism between sexual and asexual reproductive modes can negatively affect the fitness of clonal plants. Populations of heterostylous and dioecious species have a small number of mating groups (two or three), which should occur at equal frequency in equilibrium populations. Extensive clonal growth and vegetative dispersal can disrupt the functioning of these sexual polymorphisms, resulting in biased morph ratios and populations with a single mating group, with consequences for fertility and mating. In populations in which clonal propagation predominates, mutations reducing fertility may lead to sexual dysfunction and even the loss of sex. Recent evidence suggests that somatic mutations can play a significant role in influencing fitness in clonal plants and may also help explain the occurrence of genetic diversity in sterile clonal populations. Highly polymorphic genetic markers offer outstanding opportunities for gaining novel insights into functional interactions between sexual and clonal reproduction in flowering plants.

Unisexual versus bisexual mating in Cryptococcus neoformans: Consequences and biological impacts

Cryptococcus neoformans is an opportunistic human fungal pathogen and can undergo both bisexual and unisexual mating. Despite the fact that one mating type is dispensable for unisexual mating, the two sexual cycles share surprisingly similar features. Both mating cycles are affected by similar environmental factors and regulated by the same pheromone response pathway. Recombination takes place during unisexual reproduction in a fashion similar to bisexual reproduction and can both admix pre-existing genetic diversity and also generate diversity de novo just like bisexual reproduction. These common features may allow the unisexual life cycle to provide phenotypic and genotypic plasticity for the natural Cryptococcus population, which is predominantly α mating type, and to avoid Muller’s ratchet. The morphological transition from yeast to hyphal growth during both bisexual and unisexual mating may provide increased opportunities for outcrossing and the ability to forage for nutrients at a distance. The unisexual life cycle is a key evolutionary factor for Cryptococcus as a highly successful global fungal pathogen.

Genetic and ecological outcomes of Inga vera subsp. affinis (Leguminosae) tree plantations in a fragmented tropical landscape.

Planting of native trees for habitat restoration is a widespread practice, but the consequences for the retention and transmission of genetic diversity in planted and natural populations are unclear. Using Inga vera subsp. affinis as a model species, we genotyped five natural and five planted populations in the Atlantic forest of northeastern Brazil at polymorphic microsatellite loci. We studied the breeding system and population structure to test how much genetic diversity is retained in planted relative to natural populations. We then genotyped seedlings from these populations to test whether genetic diversity in planted populations is restored by outcrossing to natural populations of I. vera. The breeding system of natural I. vera populations was confirmed to be highly outcrossing (t = 0.92; FIS = −0.061, P = 0.04), with populations showing weak population substructure (FST = 0.028). Genetic diversity in planted populations was 50% less than that of natural populations (planted: AR = 14.9, HO = 0.865 and natural: AR = 30.8, HO = 0.655). However, seedlings from planted populations showed a 30% higher allelic richness relative to their parents (seedlings AR = 10.5, parents AR = 7.6). Understanding the processes and interactions that shape this system are necessary to provide ecologically sensible goals and successfully restore hyper-fragmented habitats. Future restoration plans for I. vera must consider the genetic diversity of planted populations and the potential for gene flow between natural populations in the landscape, in order to preserve ecological interactions (i.e. pollination), and promote opportunities for outcrossing.

Costs and benefits of the mixed-mating system ofNarcissus serotinus(Amaryllidaceae) in the conservation of small fragmented populations

Small fragmented populations often exhibit reduced plant–pollinator interactions and scarce outcrossing opportunities. In this context, mixed-mating systems can be advantageous since selfing can provide reproductive assurance, but they may also carry relevant costs such as those involved in inbreeding depression. This study examines the advantages and costs of selfing in Narcissus serotinus L., a mixed-mating geophyte that currently occurs in several fragmented populations in Portugal, that resulted from the construction of the largest European dam. Observation of pollinators revealed that cross-pollination is less frequent in small than in large populations. Manual self-pollinations significantly increased fruit and seed set in small-size populations, also suggesting limited insect pollination. The existence of selfing may provide reproductive assurance in small-size populations of N. serotinus where outcrossing pollination is reduced. Although floral biology and experimental pollinations showed that N. serotinus is capable of autonomous selfing, four of the six fitness traits studied showed significant inbreeding depression in all populations. The high levels of inbreeding depression found in N. serotinus suggest that the initial reproductive assurance advantage of selfing may be counterbalanced by lower survival of the resulting individuals and a decrease in the evolutionary potential of the populations.

Diplostigmaty in plants: a novel mechanism that provides reproductive assurance

Differentiation of female sexual organs in flowering plants is rare and contrasts with the wide range of male reproductive strategies. An unusual example involves diplostigmaty, the possession of spatially and temporally distinct stigmas in Sebaea (Gentianaceae). Here, the single pistil within a flower has an apical stigma, as occurs in most flowering plants, but also a secondary stigma that occurs midway down the style, which is physically discrete and receptive several days after the apical stigma. We examined the function of diplostigmaty in Sebaea aurea, an insect-pollinated species of the Western Cape of South Africa. Floral manipulations and measurements of fertility and mating patterns provided evidence that basal stigmas function to enable autonomous delayed self-pollination, without limiting opportunities for outcrossing and thus avoiding the costs of seed discounting. We suggest that delayed selfing serves as a mechanism of reproductive assurance in populations with low plant density. The possession of dimorphic stigma function provides a novel example of a flexible mixed-mating strategy in plants that is responsive to changing demographic conditions.

Protogyny and delayed autonomous self‐pollination in the desert herb Zygophyllum macropterum (Zygophyllaceae)

AbstractProtogyny and delayed selfing are two mechanisms in facilitating outcrossing and increasing female and male function in self‐compatible plant species. However, both mechanisms are rarely reported within a species. Here, we investigated pistil and stamen development, breeding system, and pollination biology of the desert herb Zygophyllum macropterum C. A. Mey. by use of field observations and manipulation experiments. Our primary aim was to assess whether protogyny and delayed autonomous self‐pollination occurred in this species. Zygophyllum macropterum is a spring flowering species with a floral longevity of approximately 3 days, including the female stage and the bisexual stage. Movement herkogamy occurred during both stages. The stigma was receptive when it was exposed from the corolla. The time of first anther dehiscence was approximately 12 h after stigma exposure. These results indicated that Z. macropterum is protogynous. Different pollination treatments showed that this species is self‐compatible and can self‐pollinate autonomously. Time of first dehiscence of anther overtopping the stigma was approximately 16 h after stigma exposure, suggesting that autonomous self‐pollination can occur in middle‐late anthesis, and thus can be defined as delayed autonomous self‐pollination. Anthophora plumipes Pallas. was the only pollinator of Z. macropterum. The unfavorable spring desert environment and the small corolla mouth diameter at the female stage would limit pollinator activity and thus lead to failure of outcrossing and occurrence of delayed self‐pollination. Therefore, both protogyny and delayed selfing play an important role in enhancing the opportunity for outcrossing and reproductive assurance of Z. macropterum in the spring desert environment.

Herkogamy and Its Effects on Mating Patterns in Arabidopsis thaliana

The evolution of mating systems, which exhibit an extraordinary diversity in flowering plants, is of central interest in plant biology. Herkogamy, the spatial separation of sexual organs within flowers, is a widespread floral mechanism that is thought to be an adaptive trait reducing self-pollination in hermaphroditic plants. In contrast with previous studies of herkogamy that focused on plants with relatively large floral displays, we here characterized herkogamy in Arabidopsis thaliana, a model plant with a strong selfing syndrome. Developmental features, reproductive consequences, and genetic architecture of herkogamy were exploited using naturally variable A. thaliana accessions, under both greenhouse and natural conditions. Our results demonstrate that the degree of herkogamy can strongly influence the mating patterns of A. thaliana: approach herkogamy can effectively promote outcrossing, no herkogamy is also capable of enhancing the opportunity for outcrossing, and reverse herkogamy facilitates efficient self-pollination. In addition, we found that the expression of herkogamy in A. thaliana was environment-dependent and regulated by multiple quantitative trait loci. This study reveals how minor modifications in floral morphology may cause dramatic changes in plant mating patterns, provides new insights into the function of herkogamy, and suggests the way for dissecting the genetic basis of this important character in a model plant.

Experimental manipulation of flowers to determine the functional modes and fitness consequences of self‐fertilization: unexpected outcome reveals key assumptions

Summary Reproductive assurance is often invoked to explain the repeated transition from outcrossing to selfing among plants. The most common experimental test of this hypothesis uses floral emasculation (anther removal) to manipulate the capacity of flowers for autogamous self‐pollination. The coastal dune endemic Camissoniopsis cheiranthifolia exhibits striking variation in floral morphology and the mating system across its geographic range in California, and reproductive assurance is likely to have played a role in the transition from large flowers and predominant outcrossing to small flowers and predominant selfing. To compare variation in the timing of self‐pollination and the extent to which it provides reproductive assurance in large‐ vs. small‐flowered populations, we emasculated flowers at different times during anthesis and measured the effects on both seed production and the proportion of seeds self‐fertilized vs. outcrossed estimated using marker genes. We also manipulated large‐ vs. small‐flowered phenotypes within two morphologically variable populations. As predicted from floral development, small‐flowered plants self‐pollinated before flowers open, whereas large‐flowered phenotypes did not. Comparing flowers that were emasculated before closing suggested that a seemingly well‐developed mechanism for delayed selfing (anthers contacting the stigma when flowers close) does not increase seed production. Using emasculation to manipulate selfing assumes that removing anthers from flowers does not compromise outcrossing. However, while not affecting the capacity of flowers to produce seed, emasculation shortened floral longevity, thereby reducing opportunities for outcrossing. Genetic estimation of the mating system revealed a negative effect of emasculation on the proportion of seed outcrossed, even in small‐flowered populations, further suggesting that flowers lacking anthers are unattractive to pollinators. These side effects of emasculation prevent a clear interpretation of the reproductive assurance value of selfing. Given that much of what we know about reproductive assurance in natural populations comes from emasculation experiments, our results suggest that the assumptions of this approach, which are rarely verified, require much more serious consideration.

Occurrence, costs and heritability of delayed selfing in a free‐living flatworm

Evolutionary theory predicts that in the absence of outcrossing opportunities, simultaneously hermaphroditic organisms should eventually switch to self-fertilization as a form of reproductive assurance. Here, we report the existence of facultative self-fertilization in the free-living flatworm Macrostomum hystrix, a species in which outcrossing occurs via hypodermic insemination of sperm into the parenchyma of the mating partner. First, we show that isolated individuals significantly delay the onset of reproduction compared with individuals with outcrossing opportunities ('delayed selfing') as predicted by theory. Second, consistent with the idea of M. hystrix being a preferential outcrosser under natural conditions, we report likely costs of selfing manifested via reduced hatchling production and offspring survival. Third, we demonstrate that selfing propensity has a genetic basis in this species, with a heritability estimated at 0.43±0.11. Variation in selfing propensity could arise due to differing costs of inbreeding among families; despite marked inter-family variation in apparent costs of inbreeding, we found no evidence for such a link. Alternatively, selfing propensity might differ across families because of heritable variation in reproductive traits that determine the likelihood of selfing. We speculate that adaptations to hypodermic insemination under outcrossing, most notably a highly modified copulatory stylet (male copulatory organ) and reduced sperm complexity, could also facilitate facultative selfing in this species.
