Sensitive Analysis of Recombinant Human Erythropoietin Glycopeptides by On-Line Phenylboronic Acid Solid-Phase Extraction Capillary Electrophoresis Mass Spectrometry.

In this study, several chromatographic sorbents: porous graphitic carbon (PGC), aminopropyl hydrophilic interaction (aminopropyl-HILIC), and phenylboronic acid (PBA) were assessed for the analysis of glycopeptides by on-line solid-phase extraction capillary electrophoresis mass spectrometry (SPE-CE-MS). As the PBA sorbent provided the most promising results, a PBA-SPE-CE-MS method was developed for the selective and sensitive preconcentration of glycopeptides from enzymatic digests of glycoproteins. Recombinant human erythropoietin (rhEPO) was selected as the model glycoprotein and subjected to enzymatic digestion with several proteases. The tryptic O126 and N83 glycopeptides from rhEPO were targeted to optimize the methodology. Under the optimized conditions, intraday precision, linearity, limits of detection (LODs), and microcartridge lifetime were evaluated, obtaining improved results compared to that from a previously reported TiO2-SPE-CE-MS method, especially for LODs of N-glycopeptides (up to 500 times lower than by CE-MS and up to 200 times lower than by TiO2-SPE-CE-MS). Moreover, rhEPO Glu-C digests were also analyzed by PBA-SPE-CE-MS to better characterize N24 and N38 glycopeptides. Finally, the established method was used to analyze two rhEPO products (EPOCIM and NeuroEPO plus), demonstrating its applicability in biopharmaceutical analysis. The sensitivity of the proposed PBA-SPE-CE-MS method improves the existing CE-MS methodologies for glycopeptide analysis and shows a great potential in glycoprotein analysis to deeply characterize protein glycosites even at low concentrations of the protein digest.

A hybrid nano-MOF/polymer material for trace analysis of fluoroquinolones in complex matrices at microscale by on-line solid-phase extraction capillary electrophoresis

A hybrid material (nano-metal organic framework@organic polymer, named as nano-MOF@polymer) was applied for the first time as sorbent for on-line solid-phase extraction capillary electrophoresis with ultraviolet detection (SPE-CE-UV). The resulting material was prepared building layer-by-layer a HKUST-1 (Hong Kong University of Science and Technology-1) nano-MOF onto the polymer surface, which allowed controlling the thickness and maximizing the active surface area. The sorbent was widely characterized at micro- and nano-scale to validate the synthesis and to establish the material properties. Then, fritless microcartridges (2 mm) were assembled by packing only a few micrograms of sorbent particles and investigated for preconcentration of fluoroquinolones (FQs) in several real samples (river water, human urine and whole cow milk). Under the optimized conditions, the sample (ca. 60 μL) was loaded in separation background electrolyte (BGE, 50 mM phosphate (pH 7)), and retained analytes were eluted using a small volume of 2% v/v formic acid in methanol (ca. 50 nL). The SPE-CE-UV method was validated in terms of linearity, limit of detection (LOD), limit of quantification (LOQ), repeatability, reproducibility and reusability. The developed method showed a LOD decreasing until 1 ng L−1 when larger volumes of sample were loaded (ca. 180 μL), which was 500,000 times lower than by CE-UV. This undescribed sensitivity enhancement would arise from the homogenous and populated MOF nano-domains and the appropriate permeability of the hybrid material, which would promote high extraction efficiency and loading capacity. Furthermore, the sorbent showed appropriate selectivity regardless the analyzed complex environmental, biological or food matrix samples, achieving excellent detectability and recoveries (>90%).

Evaluation of on-line solid-phase extraction capillary electrophoresis–mass spectrometry with a nanoliter valve for the analysis of peptide biomarkers

On-line solid-phase extraction capillary electrophoresis-mass spectrometry (SPE-CE-MS) is a powerful technique for high throughput sample clean-up and analyte preconcentration, separation, detection, and characterization. The most typical design due to its simplicity and low cost is unidirectional SPE-CE-MS. However, in this configuration, the sample volumes introduced by pressure depend on the dimensions of the separation capillary and some matrix components could be irreversibly adsorbed in its inner walls. Furthermore, in many cases, the requirements of on-line preconcentration are incompatible with the background electrolyte necessary for an efficient separation and sensitive MS detection. Here, we present SPE-CE-MS with a nanoliter valve (nvSPE-CE-MS) to overcome these drawbacks while keeping the design simple. The nvSPE-CE-MS system is operated with a single CE instrument and two capillaries for independent and orthogonal SPE preconcentration and CE separation, which are interfaced through an external and electrically isolated valve with a 20 nL sample loop. The instrumental setup is proved for the analysis of opioid and amyloid beta peptide biomarkers in standards and plasma samples. NvSPE-CE-MS allowed decreasing the limits of detection (LODs) 200 times with regard to CE-MS. Compared to unidirectional SPE-CE-MS, peak efficiencies were better and repeatabilities similar, but total analysis times longer and LODs for standards slightly higher due to the heart-cut operation and the limited volume of the valve loop. This small difference on the LODs for standards was compensated for plasma samples by the improved tolerance of nvSPE-CE-MS to complex sample matrices. In view of these results, the presented setup can be regarded as a promising versatile alternative to avoid complicated matrix samples entering the separation capillary in SPE-CE-MS.

Metabolic profiling for the identification of Huntington biomarkers by on-line solid-phase extraction capillary electrophoresis mass spectrometry combined with advanced data analysis tools.

In this work, an untargeted metabolomic approach based on sensitive analysis by on-line solid-phase extraction capillary electrophoresis mass spectrometry (SPE-CE-MS) in combination with multivariate data analysis is proposed as an efficient method for the identification of biomarkers of Huntington's disease (HD) progression in plasma. For this purpose, plasma samples from wild-type (wt) and HD (R6/1) mice of different ages (8, 12, and 30 weeks), were analyzed by C18 -SPE-CE-MS in order to obtain the characteristic electrophoretic profiles of low molecular mass compounds. Then, multivariate curve resolution alternating least squares (MCR-ALS) was applied to the multiple full scan MS datasets. This strategy permitted the resolution of a large number of metabolites being characterized by their electrophoretic peaks and their corresponding mass spectra. A total number of 29 compounds were relevant to discriminate between wt and HD plasma samples, as well as to follow-up the HD progression. The intracellular signaling was found to be the most affected metabolic pathway in HD mice after 12 weeks of birth, when mice already showed motor coordination deficiencies and cognitive decline. This fact agreed with the atrophy and dysfunction of specific neurons, loss of several types of receptors, and changed expression of neurotransmitters.

On-Line Solid-Phase Extraction Capillary Electrophoresis Mass Spectrometry for Preconcentration and Clean-Up of Peptides and Proteins.

One of the major drawbacks of capillary electrophoresis (CE) and other microscale separation techniques, for the analysis of low abundant peptides and proteins in complex samples, are the poor concentration limits of detection. Several strategies have been developed to improve CE sensitivity. Here, we describe an on-line solid-phase extraction capillary electrophoresis mass spectrometry method with a commercial C18 sorbent for clean-up and preconcentration of neuropeptides from highly diluted biological samples.

Developments in coupled solid-phase extraction-capillary electrophoresis 2013-2015.

An overview of the design and application of coupled solid-phase extraction-capillary electrophoresis (SPE-CE) systems reported in the literature between July 2013 and June 2015 is provided in this paper. The present article is a continuation of our previous review papers on this topic which covered the time period 2000-2013 (Electrophoresis 2008, 29, 108-128; Electrophoresis 2010, 31, 44-54; Electrophoresis 2012, 33, 243-250; Electrophoresis 2014, 35, 128-137). The use of in-line and on-line SPE-CE approaches is treated and outlined in this review. Recent advancements, such as, for example, the use of aptamers as affinity material for in-line SPE-CE, the use of a bead string design for in-line fritless SPE-CE, and new interfacing techniques for the on-line coupling of SPE to CE, are outlined. Selected examples demonstrate the applicability of the coupled SPE-CE systems for biomedical, pharmaceutical, environmental, and food studies. A complete overview of the recent SPE-CE studies is given in table format, providing information on sample type, SPE sorbent, coupling mode, detection mode, and LOD. Finally, some general conclusions and perspectives are provided.

A high performance system to study the influence of temperature in on-line solid-phase extraction capillary electrophoresis

A novel high performance system to control the temperature of the microcartridge in on-line solid phase extraction capillary electrophoresis (SPE–CE) is introduced. The mini-device consists in a thermostatic bath that fits inside of the cassette of any commercial CE instrument, while its temperature is controlled from an external circuit of liquid connecting three different water baths. The circuits are controlled from a switchboard connected to an array of electrovalves that allow to rapidly alternate the water circulation through the mini-thermostatic-bath between temperatures from 5 to 90°C. The combination of the mini-device and the forced-air thermostatization system of the commercial CE instrument allows to optimize independently the temperature of the sample loading, the clean-up, the analyte elution and the electrophoretic separation steps.The system is used to study the effect of temperature on the C18-SPE–CE analysis of the opioid peptides, Dynorphin A (Dyn A), Endomorphin1 (END) and Met-enkephalin (MET), in both standard solutions and in spiked plasma samples. Extraction recoveries demonstrated to depend, with a non-monotonous trend, on the microcartridge temperature during the sample loading and became maximum at 60°C. Results prove the potential of temperature control to further enhance sensitivity in SPE–CE when analytes are thermally stable.

Preparation and evaluation of open tubular C18-silica monolithic microcartridges for preconcentration of peptides by on-line solid phase extraction capillary electrophoresis

In this study, C18-silica monoliths were synthesized as a porous layer in open tubular capillary columns, to be cut later into microcartridges for the analysis of neuropeptides by on-line solid-phase extraction capillary electrophoresis with UV and MS detection (SPE-CE-UV and SPE-CE-MS). First, several types of C18-silica monolithic (MtC18) microcartridges were used to analyse standard solutions of five neuropeptides (i.e. dynorphin A (1–7), substance P (7–11), endomorphin 1, methionine enkephalin and [Ala]-methionine enkephalin). The MtC18 sorbents were especially selective against endomorphin 1 and substance P (7–11)). The best results in terms of sensitivity and inter-microcartridge reproducibility were achieved with the microcartridges obtained from a 10-cm open tubular capillary column with a thin monolithic coating with large through-pores (1–5μm). Run-to-run repeatability, microcartridge durability, linearity ranges and LODs were studied by MtC18-SPE-CE-MS. As expected due to their greater selectivity, the best LOD enhancement was obtained for End1 and SP (7–11) (50 times with regard to CE-MS). Finally, the suitability of the methodology for analysing biological fluids was tested with plasma samples spiked with End1 and SP (7–11). Results obtained were promising because both neuropeptides could be detected at 0.05μgmL−1, which was almost the same concentration level as for the standard solutions (0.01μgmL−1).

The present paper is an update of two previous review papers covering the years 2000-2009 (Electrophoresis 2008, 29, 108-128; Electrophoresis 2010, 31, 44-54). Both in-line and on-line SPE-CE approaches are treated and outlined. Attention is paid to emerging technological developments, such as the use of carbon nanotubes and magnetic particles for on-line extraction of sample components prior to CE analysis. Selected examples illustrate the applicability of SPE-CE in biomedical, pharmaceutical, environmental and food analysis. A full overview of recent SPE-CE studies is given in table format, providing information about sample type, SPE sorbent, coupling mode, detection mode and limit of detection. Finally, some general conclusions and future perspectives are given.

Transient isotachophoresis in on‐line solid phase extraction capillary electrophoresis time‐of‐flight‐mass spectrometry for peptide analysis in human plasma

In this study, we evaluated the combination of transient isotachophoresis with on-line solid-phase extraction capillary electrophoresis time-of-flight mass spectrometry (SPE-tITP-CE-TOF-MS) to improve sensitivity of peptide analysis, using several opioid peptides as model compounds. First, standard solutions were analyzed in order to establish the tITP-CE methodology using UV and TOF-MS detection. The volume and composition of the leading and terminating electrolytes (i.e. LE and TE) for an efficient tITP were investigated to obtain optimum detection sensitivity and electrophoretic separation. In the best cases, LODs in tITP-CE-TOF-MS were tenfold better than those obtained in CE-TOF-MS (i.e. 5 versus 50 ng/mL). Afterwards, the tITP-CE-TOF-MS methodology was adapted to perform SPE-tITP-CE-TOF-MS. Repeatability, linearity and LODs were investigated and compared to the values obtained by SPE-CE-TOF-MS. Furthermore, human plasma samples fortified with the opioid peptides were analyzed in order to show the potential of SPE-tITP-CE-TOF-MS for peptide analysis in biological fluids. The LODs attained in standard solutions and plasma samples for some of the studied peptides (i.e. 0.01 and 0.1 ng/mL, respectively) were tenfold better than those obtained in SPE-CE-TOF-MS, proving the enhanced sensitivity that could be achieved when both on-line preconcentration approaches were combined together.

Lowering the concentration limits of detection by on-line solid-phase extraction–capillary electrophoresis–electrospray mass spectrometry

The use of solid-phase extraction coupled on-line to capillary electrophoresis using electrospray mass spectrometry detection (SPE-CE-ESI-MS) is described for the analysis of peptides in dilute solutions. A SPE microcartridge or analyte concentrator containing C 18 derivatized silica particles as the extraction sorbent was easily constructed near the inlet of the separation capillary using commercially available materials. The reversed-phase sorbent selectively retained the target peptides, enabling large volumes of the sample to be introduced (>100 μL). The captured analytes were eluted in a small volume of an appropriate solution (20–50 nL). This resulted in sample clean-up and concentration enhancement, with minimum sample handling. As the SPE-CE conditions were compatible with on-line ESI-MS detection, the potential for identifying and characterizing the preconcentrated analytes by SPE-CE-ESI-MS using a sheath-flow CE-ESI-MS interface is also shown. Using separation electrolytes containing N-[carbamoylmethyl]-2-aminoethanesulfonic acid (ACES) at pH 7.4, an elution plug of 80:20 (v/v) (25 mM of formic acid in MeCN):H 2O and a sheath liquid of 20 mM of acetic acid in 50:50 (v/v) methanol:H 2O the concentration limits of detection for the analyzed peptides in the positive ion mode were lowered to nanogram per milliliter levels. The systematic optimization of the operational parameters involved in the development of the SPE-CE method is described in detail, in order to promote robust and quantitative SPE-CE-ESI-MS analysis and facilitate the widespread use of the technique.

Determination of pesticides in waters by automatic on-line solid-phase extraction–capillary electrophoresis

The separation of seven pesticides by micellar electrokinetic capillary chromatography in spiked water samples is described, allowing the analysis of pesticides mixtures down to a concentration of 50 μg l −1 in less than 13 min. Calibration, pre-concentration, elution and injection into the sample vial was carried out automatically by a continuous flow system (CFS) coupled to a capillary electrophoresis system via a programmable arm. The whole system was electronically coupled by a micro-processor and completely controlled by a computer. A C 18 solid-phase mini-column was used for the pre-concentration, allowing a 12-fold enrichment (as an average value) of the pesticides from fortified water samples. Under the optimal extraction conditions, recoveries between 90 and 114% for most of the pesticides were obtained.

Characterization of a solid-phase extraction device for discontinuous on-line preconcentration in capillary electrophoresis-based peptide mapping

Peptide mapping by capillary electrophoresis (CE) with UV detection is problematic for the characterization of proteins that can only be obtained at low micromolar concentrations. Dilution of peptide fragments during digestion of the protein can further reduce the detection sensitivity in peptide mapping to the point where analysis at sub-micromolar concentrations is not possible. A remedy to this problem is preconcentration (sample enrichment) of the proteolytic digest by solid-phase extraction (SPE). To minimize non-specific adsorptive losses during sample handling, on-line SPE–CE is preferred. However, packed-inlet SPE–CE is not always feasible due to either instrument or sample limitations. We describe here a simple method of preconcentration by discontinuous on-line SPE–CE, specifically applied to peptide mapping in low-pH separation buffer after protein digestion in a solid-phase enzyme microreactor. The SPE–CE system does not require application of a low pressure during electrophoretic separation to overcome reversed electroosmotic flow because the preconcentrator device is disconnected from the separation capillary before the electric field is applied. Up to a 500-fold preconcentration factor can be achieved with this device, which can be reused for many samples. Parameters such as the volume of desorption solution, the adsorption/desorption (chromatographic) process, reproducibility of packing the SPE preconcentrator and effects of sample concentration on the peptide map are investigated.

Capillary Electrophoresis of Insulin-like Growth Factors: Enhanced Ultraviolet Detection Using Dynamically Coated Capillaries and On-Line Solid-Phase Extraction

Insulin-like growth factor-I and -II (IGF-I and IGF-II) are difficult to separate and measure as a result of their homology, both structurally and immunologically. A number of binding proteins (BPs) which interact with the IGFs with high affinity complicate the ability to measure the IGFs accurately and reproducibly. Current methodology for measuring IGF is immuno-based and involves dissociation from the IGFs and removal of the binding proteins through sample acidification and removal by solid-phase adsorption. However, the net result is an assay that is time-consuming and, at best, semiquantitative. In an attempt to improve the reproducibility and accuracy of IGF-I and -II measurement, electrophoretic systems employing dynamically coated and bare silica capillaries were evaluated. Separations in bare silica capillaries in the presence or absence of the cationic additive, decamethonium bromide were ineffective for resolving IGF-I and IGF-II. However, when the capillary was coated dynamically with polybrene, IGF-I and -II could be resolved in a BSA sample matrix using a low pH buffer. Despite the fact that the IGFs could be resolved in the presence of an IGF-I analog used as an internal standard, polybrene recoating was required after as few as 12 runs and poor coating-to-coating reproducibility was observed. Use of polydiallyldimethylammonium chloride (PDMAC) as a dynamic cationic coating and a low pH buffer containing 0.5% PDMAC was found to be much more effective, providing reproducible separation of IGF-I and -II. It was found that PDMAC need not be included in the separation buffer to obtain reproducible analyses regarding IGF separation. Subsequently, functionality remained intact for as many as 35–40 consecutive analyses before recoating was required. Without the need for PDMAC in the buffer, on-line solid-phase extraction–capillary electrophoresis could be accomplished for detection of IGF-I and -II at concentrations as low 195 ng/ml.

On-line solid-phase extraction–capillary electrophoresis for enhanced detection sensitivity and selectivity: application to the analysis of metallothionein isoforms in sheep fetal liver

A simple and rapid capillary electrophoresis (CE) method for analysis of metallothionein (MT) isoforms is described. A modified two-step solvent extraction procedure was used in combination with CE and an on-line solid-phase pre-concentration device for sensitive and reproducible detection of MT isoforms in sheep fetal liver. Preparation of twenty samples was practicable within a working day with subsequent automated overnight analysis by solid-phase extraction (SPE)–CE. A commercially available divinylbenzene-based reversed-phase resin was found to be most suitable for the SPE device because it is resistant to extremes of pH and can be adequately regenerated between analyses. Each SPE device was readily constructed from commonly available materials and was used for the reproducible separation of over 100 biological samples before replacement. Precision of analysis within or between sample batches was <10% and usually <5% while detection limits were at least 28 ng/ml for standards and 272 ng/ml for biological samples. This would indicate a detection limit of about 0.5 μg/g wet weight of tissue. Recovery of MT from tissue cytosols by solvent extraction was measured using radiolabeled MT and was found to be just over 50% increasing to almost 70% by addition of NaCl to the homogenisation buffer. The combined solvent extraction and SPE–CE methodology was applied to the analysis of MT in sheep fetal liver and the results compared favorably with those obtained by high-resolution chromatography. MT-1 levels were 2 to 4-times higher than those of MT-2 and both isoforms decreased from day 89 to day 136 of gestation. These results were compared with MT levels in fetal liver from sheep embryos that had been perturbed by temporary transfer to an advanced uterine environment. Hepatic MT levels at day 136 of gestation were 3 to 8-times higher than in normal fetal liver and significant differences were observed with both isoforms.
