Strategic administration of antioxidant multiminerals and vitamins to transitional buffaloes augments antioxidant and udder defense mechanisms in early lactation

The aim of the study was to examine the effects of repeated administrations of antioxidant multiminerals and vitamins in transition buffaloes on udder defense mechanism, antioxidant activity and occurrence of intramammary infection (IMI) in early lactation period. Forty clinically healthy pregnant buffaloes were enrolled 45 days before expected date of calving and randomly allocated into five different supplementation groups (n = 8): only basal ration (control), vitamin E and selenium (VES), multiminerals (MM), ascorbic acid (AA) and chromium (Cr) picolinate in basal diet. The udder defense mechanism was monitored by measuring phagocytic activity (PA), myeloperoxidase (MPO) and nitric oxide (NO) productions in milk leukocytes, antioxidant activity was evaluated by measuring total antioxidant capacity (TAC) in plasma and occurrence of IMI was assessed by milk cytology, bacterial count in milk and visible clinical signs of udder until day 28 post-calving. The results showed that the VES and MM supplementations exhibited significantly higher PA, MPO and NO productions of milk leukocytes till first week of lactation whereas, elevated mean TAC in plasma was maintained from day −7 to 1 of calving in MM supplementation group as compared to control group. Statistically, no significant difference in occurrences of subclinical or clinical IMI was noted across the groups until four weeks of lactation. Taken together, it is concluded that repeated administrations of VES and MM to transition buffaloes could be an effective strategy to maintain good udder health by augmenting milk leukocyte functions and antioxidant status and preventing incidence of IMI in early lactation.

Intramammary infections and risk factors in freshly calved heifers in Swedish dairy herds

Subclinical mastitis can be common among freshly calved heifers (FCH), but the prevalence differs between herds, possibly due to variation in risk factors. The aims of this observational study were to identify differences in occurrence of intramammary infection (IMI) in FCH between herds with documented good or poorer first-parity udder health based on cow somatic cell count (CSCC) in early lactation, and to study herd differences in animal factors important for udder health, such as udder and hock skin lesions and animal cleanliness. Three groups of herds were included: those with high proportions of FCH with low CSCC (≤75,000 cells/mL) at the first 2 milk recordings after calving (LL), herds with high proportions of FCH with high CSCC (>100,000 cells/mL) at the first and low CSCC at the second recording (HL), and herds with high proportions of FCH with high CSCC at both recordings (HH). Thirty-nine herds (13 LL, 11 HL, 15 HH) were visited 3 times during a 12-mo period for observation of cleanliness and hock lesions, and sampling of udder and teat skin using swab cloths of milk-fed calves, early-pregnant heifers, and late-pregnant heifers. In 25 (9 LL, 9 HL, 7 HH) udder quarter samples from colostrum and milk on d 3 to 4 after calving were taken by the farmers from FCH during one year. The farmers also provided information on calving (individual or group), use of restraint and oxytocin at milking, and presence of teat and udder skin lesions. Bacterial growth in swab samples and quarter samples was investigated by culturing, and a selection of isolates was genotyped using whole-genome sequencing. Cleanliness, hock and udder skin lesions other than udder-thigh dermatitis, and growth of bacteria in swab samples did not differ between herd groups. It was more common that FCH from LL herds, compared with FCH in HH and HL herds, calved in a group of animals. Use of restraint at milking was more common in LL herds than in HH herds, whereas presence of udder-thigh dermatitis was lowest in LL herds. Specific infection was found in 14% of 5,593 quarter samples from 722 FCH. The most common IMI was Staphylococcus chromogenes. Growth of Staphylococcus simulans was more common in HH than in LL and HL herds. In colostrum samples, Staphylococcus haemolyticus was more common in HL and HH than in LL herds. The proportion of quarters with the same specific infection at both samplings was higher in HH than in LL herds and tended to be higher in HH than in HL herds. The proportion of quarters with Staph. chromogenes IMI at both samplings tended to differ between herd groups and was highest in HH herds. Whole-genome sequencing found the same sequence type of Staph. chromogenes and Staphylococcus aureus in both samples in almost all quarters with the same infection at both samplings. The differences in IMI between herd groups were in line with the higher somatic cell count in HH herds. The reasons for the predominance of Staph. chromogenes IMI in FCH need further studies.

Udder and teat morphometry and its relationship with occurrence of intramammary infections in dairy cattle

Bovine mastitis, an inflammatory disease of the mammary gland, is often caused by bacterial infection and is a major health problem on dairy farms. A cross-sectional study of four herds was conducted to assess udder and teat-related risk factors for the development of intramammary infections in 243 lactating cows. Udder and teat morphometric parameters were determined through visual appraisal method. Intramammary infection was assessed using California mastitis test (CMT), Somatic cell count (SCC) and bacteriology of milk samples. The frequencies of udder conformations were 65.8%, 18.9%, 7.4% and 7.8% for desirable, pendulous, round and goaty-shapes, respectively. Teat conformation revealed that 76.0%, 5.3%, 6.6%, 4.5% and 7.5% for desirable, short, funnel, bottle and conical-shapes, respectively. There was a significant (p<0.01) effect of udder shape and teat-end shape on SCC level and intramammary infections. The mean SCC and mastitis infection levels for undesirable udder was significantly (p<0.05) higher as compared to the desirable shaped udder. Similarly, significant (p<0.05) higher level SCC was found in undesired teats compared to desirable teats. The overall proportion of quarter mastitis infection in all four farms revealed 1.23% for clinical and 36.52% for subclinical mastitis. Herd-wise subclinical mastitis prevalence showed 42.8%, 35.07%, 33.09% and 2.94% for farm A, C, B and C, respectively. The most frequently isolated bacterial species were Staphylococcus aureus (56.34%) followed by Escherichia coli (22.55%), Streptococcus agalactiae (9.86%) and Bacillus spp. (8.45%). In conclusion, undesirable udder and teats conformations were more susceptible to intramammary infection and these traits must be considered accordingly while selecting dairy animals for future milk production. Observed high occurrence of subclinical Staphylococcal mastitis needs improvements in milking hygiene, mammary gland health, regular screening of animals, and use of biosecurity as an intervention tactic to limit the spread of pathogens at dairy farms.

Dynamics of Milk Parameters of Quarter Samples before and after the Dry Period on Czech Farms

This study aimed to monitor milk parameters on three different dairy farms in the Czech Republic to describe their readiness for implementing selective dry cow therapy. Fat, protein, casein, lactose, solids-not-fat content, total solids content, freezing point, titratable acidity, and somatic cell count of quarter milk samples collected from tested Holstein cows were evaluated. Associations between the tested parameters, as well as the effects of parity, farm, day of calving, and time of evaluation at dry-off and after calving, were assessed. Values of the leading milk components dynamically changed between dry-off and after calving, but only protein content was significantly affected. The most important parameter of our research, the somatic cell count of quarter milk samples, was also not affected by the time of evaluation. Even though a slight increase in the mean of somatic cell count is expected before the dry period and after calving, at dry-off, we observed 30%, 42%, and 24% of quarters with somatic cell counts above 200,000 cells per mL, while after calving, we observed 27%, 16%, and 18% of quarters with somatic cell counts above 200,000 cells per mL on Farm 1, Farm 2, and Farm 3, respectively. High somatic cell counts (>200,000 cells per mL) indicate bacterial infection, as confirmed by the significant negative correlation between this parameter and lactose content. In addition, a deficient milk fat-to-protein ratio was observed on two farms, which may indicate metabolic disorders, as well as the occurrence of intramammary infections. Despite the above, we concluded that according to the thresholds of somatic cell counts for selective dry cow therapy taken from foreign studies, a large part of the udder quarters could be dried off without the administration of antibiotics. However, it is necessary to set up more effective mechanisms for mastitis prevention.

Identification of bacterial pathogens in dry bovine udder and their antimicrobial susceptibility

Mastitis is a multi-etiological production disease of dairy cows worldwide. The likelihood of developing clinical mastitis in subsequent calving is enhanced by intramammary infections with pathogens during the late dry and post-calving period. There is an increase in the occurrence of intramammary infections during early and late dry period. Thirty apparently healthy, pregnant cross-bred dairy cows were screened for the presence of contagious mastitis pathogens in the secretions taken after 48 h of cessation of milking. A total of 118 bacterial isolates were obtained from the secretions collected from 108 quarters, and identified using morphological and cultural characteristics. Among them, 50.8 per cent were coagulase negative Staphylococci (CNS), 16 per cent were coagulase positive Staphylococci (CPS) and 27.1 per cent were Streptococci isolates. Micrococci and E. coli constituted 2.5 per cent each and Klebsiella spp. formed 0.8 per cent of the total isolates obtained. The CNS isolates identified included 22 S. chromogens, 14 S. xylosus, 12 S. saprophyticus, six S. hominis and S. epidermidis each. Among the CPS isolated 11.8 per cent were S. aureus and 4.2 per cent were non-aureus coagulase positive Staphylococcus which included three S. hyicus and two S. pseudointermedius. Molecular confirmation of S. aureus, CNS isolates, St. agalactiae were done by targeting nuc, cns and 16SrRNA gene respectively. Identification of bacterial pathogens that are present during dry period can help in formulating an effective dry cow therapy. Antibiogram studies of the isolates obtained were done using in vitro antibiotic sensitivity testing method.

Short communication: Diversity of species and transmission of antimicrobial resistance among Staphylococcus spp. isolated from goat milk

The increasing production of goat milk and its derivatives is affected by the occurrence of intramammary infections, which are highly associated with the presence of Staphylococcus species, including some with zoonotic potential. Staphylococci in general can exchange mobile genetic elements, a process that may be facilitated by the isolate's capacity of forming biofilms. In this study we identified, to the species level, Staphylococcus isolated from goat milk samples by MALDI-TOF and confirmed the identification by sequencing housekeeping genes (rrs and tuf). Eight species were identified, more than half being either Staphylococcus epidermidis or Staphylococcus lugdunensis. The isolates were shown by pulsed-field gel electrophoresis to be genetically diverse between the studied herds. Resistance to ampicillin and penicillin was widespread, and 2 Staph. epidermidis isolates contained the methicillin-resistance gene mecA. Most of the isolates that were resistant to at least 1 of the 13 antimicrobials tested harbored plasmids, one of which was demonstrated to be conjugative, being transferred from a Staph. epidermidis to a Staphylococcus aureus strain. Biofilm formation was observed in almost every isolate, which may contribute to their capacity of exchanging antimicrobial resistance genes in addition to acting as a physical barrier to the access of drugs. Our results showed that antimicrobial resistance among goat staphylococci may be emerging in a process facilitated by the exchange of mobile genetic elements between the bacteria and the establishment of biofilms, which calls for careful monitoring and more effective control therapies.

Efficacy of an internal teat seal associated with a dry cow intramammary antibiotic for prevention of intramammary infections in dairy cows during the dry and early lactation periods

ABSTRACT: The present study aimed to evaluate the use of an internal dry period teat seal containing bismuth subnitrate (Teatseal®, Zoetis®, Florham Park, Nova Jersey, USA) associated with a long-acting cloxacilin preparation (Orbenin® Extra dry cow, Zoetis®, Florham Park, Nova Jersey, USA), in preventing new infections during the dry-off and early postpartum period. A total of 150 Holstein cows (average production of 9,000 kg of milk per lactation), with four functional udder quarters without clinical mastitis was included in the study. All animals were dried-off 60 days before the expected calving date. Two teats positioned diagonal-contralaterally received only dry cow antibiotic, control group C (n=300) and the other two teats, treatment group T (n=300) received dry cow antibiotic and infusion with an internal teat seal. Data from SCC variable were transformed by log base-10 transformation. Duncan’s test was used accepting 5% as the level of statistical significance. The occurrence of intramammary infection (IMI) and chronicity rate, and frequency of microorganisms isolated at drying and immediately postpartum in teats of group C and group T were evaluated using a non-parametric Chi-square Test, accepting 10% as the statistical significance level. There was a decrease in the occurrence of new infections in the early postpartum in cows which the sealant was used (C=19.6%, T=11.4%). In the postpartum period, Gram-negative bacteria were isolated from 16 teats in C and seven in T. The greatest reduction was observed for Escherichia coli (8 vs 1) in group T. There was no effect using the internal sealant on the frequency of isolation of environmental Streptococus. The use of sealant reduced the prevalence of subclinical mastitis cows between drying-off and the early postpartum period (C=51% versus T=42%) and resulted in a lower somatic cell count (SCC) in the treatment group when compared with the control group (T=1,073x103, C=1,793x103). The use of the internal teat seal combined with dry cow antibiotic is effective in the prevention of IMI during the dry period and early lactation and results in the reduction of SCC in immediate postpartum period. The treatment is effective in reducing infection between dry-off and the immediate postpartum caused by major and minor pathogens. However, no effect on infections caused by contagious pathogens was observed.

Cow characteristics and their association with udder health after different dry period lengths.

Shortening or omitting the dry period (DP) in dairy cows is of interest because of potential beneficial effects on energy balance and metabolic health. Reported effects of a short or omitted dry period on udder health are ambiguous. This study aimed to evaluate the effect of no DP (0d), a short DP (30d), or a conventional DP (60 d) on the occurrence of intramammary infections (IMI) during the precalving period and on somatic cell counts (SCC), elevations of SCC (SCC≥200,000 cells/mL), and clinical mastitis in the subsequent lactation. The study also aimed to analyze which prepartum cow characteristics are associated with udder health after different DP lengths. Holstein-Friesian dairy cows (n=167) were randomly assigned to a DP length (0, 30, or 60 d). Cows with a 0-d DP had a greater occurrence of chronic IMI and a lower occurrence of cured IMI during the precalving period than cows with a 30-d or 60-d DP. Postpartum average SCC for lactation was greater in cows with a 0-d DP than in cows with a 30-d or 60-d DP. The number of cows with at least 1 elevation of SCC, the number of elevations of SCC per affected cow, the number of cows treated for clinical mastitis, and the number of cases of mastitis per affected cow did not differ among DP lengths. Cow characteristics related to postpartum average SCC for lactation were DP length, parity, and the following interactions: DP length with prepartum elevation of SCC, DP length with fat- and protein-corrected milk (FPCM) reduction between 150 and 67d prepartum, DP length with parity and with average SCC for lactation, and last FPCM before the conventional drying-off day with average SCC for lactation. Cows with prepartum parity 1 had a lower occurrence of at least 1 elevation of SCC in subsequent lactation compared with cows with parity >2. Last SCC before the conventional drying-off day was positively associated with occurrence of clinical mastitis in the subsequent lactation. In this study, DP length was not a risk factor for either elevation of SCC or occurrence of clinical mastitis in the subsequent lactation. The identified cow characteristics could be used in a decision support model to optimize DP length for individual cows.

Evaluation of somatic cell count thresholds to detect subclinical mastitis in Gyr cows

The objectives of this study were (1) to determine the sensitivity (Se) and specificity (Sp) of somatic cell count (SCC) thresholds to identify subclinical mastitis in Gyr cows caused by major and minor pathogens; (2) to study the effects of month of sampling, rear or front mammary quarters, herd, intramammary infection (IMI), and bacterial species on SCC at quarter level; and (3) to describe the prevalence of IMI in Gyr cows in commercial dairy herds. In total, 221 lactating Gyr cows from 3 commercial dairy farms were selected. Milk samples were collected from individual quarters once a month for 1 yr from all lactating cows for SCC and bacteriological analysis. Mammary quarters were considered the experimental units and the SCC results were log10-transformed. Four SCC thresholds (100, 200, 300 and 400 × 103 cells/mL) were used to determine Se and Sp to identify infected mammary quarters. The overall prevalence of IMI in quarter milk samples of Gyr cows was 49.8%, and the prevalence of minor pathogens was higher (31.9%) than that of major pathogens (17.8%). Quarter samples with microbial isolation presented higher SCC compared with negative samples. Sensitivity and Sp of selected SCC thresholds varied according to the group of pathogen (major and minor) involved in the IMI definition. Sensitivity increased and Sp decreased when mammary quarters with only major pathogens isolation were considered positive. The use of a single SCC analysis to classify quarters as uninfected or infected in Gyr cows may not be a useful test for this breed because Se and Sp of SCC at the studied thresholds were low. The occurrence of IMI and the bacterial species are the main factors responsible for SCC variation in mammary quarters of Gyr cows. Milk samples with major pathogens isolation elicited higher SCC than those with minor pathogens.

Stochastic bio-economic model of bovine intramammary infection

Although the dynamics of transmission play an important role in the occurrence of intramammary infection (IMI), they have not been considered in previous models used to estimate the cost of IMI. The bio-economic model described includes within-herd dynamics of pathogen-specific IMI. The model simulated Staphylococcus aureus, Streptococcus uberis, Streptococcus dysgalactiae, and Escherichia coli IMI stochastically and estimated the cost of these IMI in a herd of 100 dairy cows in a situation where a quota is applied to milk production. A Reed–Frost model was used for S. aureus, S. uberis, and S. dysgalactiae IMI and a Greenwood model for E. coli IMI. Economic analysis was conducted per pathogen for clinical and subclinical IMI. The parameters used in the model were based on the literature and were deemed credible and valid. Median annual incidence of clinical and subclinical IMI for all pathogens varied considerably. This variation was greatest for S. aureus IMI . The annual incidence of IMI in a herd of 100 dairy cows caused by S. aureus varied between 0 and 88 cases, with a median of 5 cases and the 5th and 95th percentiles of 0 to 36 for clinical IMI, and a median of 7 cases with the 5th and 95th percentiles of 0 to 52 for subclinical IMI. In consequence, the average total annual net costs also varied widely for S. aureus IMI. Clinical IMI costs were € 1375, with the 5th and 95th percentiles of 0 to 4716 and subclinical IMI costs were € 1219, with the 5th and 95th percentiles of 0 to 4030. The average annual net cost due to the 4 simulated pathogens combined was € 4896 and varied from € 915 to € 11,287 in a herd of 100 dairy cows. The bio-economic model developed for this study will be utilized as a tool to investigate the economic impact of management of pathogen-specific IMI.

Dry Cow Therapy: Effects of Method of Drug Administration on Occurrence of Intramammary Infection

Two methods of administering a dry cow infusion product were evaluated: 1) full insertion of infusion cannula into the teat cistern, and 2) partial insertion of infusion cannula into the teat canal. Eighty-six Jersey cows were treated at dry off; two quarters of each cow were infused using method 1, and two quarters were infused using method 2. The new quarter infection rate for method 1 with major intramammary pathogens was 9.9% (17 infections/172 quarters); that for method 2 was 4.1% (7 infections/171 quarters). Treatment efficacies against major pathogens for methods 1 and 2, were 57.9% (11 cures/19 quarters) and 85.7% (12 cures/14 quarters), respectively.
