Kinetically Inert Macrocyclic Europium(III) Receptors for Phosphate

The significant role that phosphate plays in environmental water pollution and biomedical conditions such as hyperphosphatemia highlights the need to develop robust receptors that can sequester the anion effectively and selectively from complex aqueous media. Toward that goal, four macrocyclic tris-bidentate 1,2-hydroxypyridonate (HOPO) europium(III) complexes containing either a cyclen, cyclam, TACN, or TACD ligand cap were synthesized and evaluated as phosphate receptors. The solubility of EuIII-TACD-HOPO in water was insufficient for luminescent studies. Whereas EuIII-cyclen-HOPO is eight coordinate with two inner-sphere water molecules, both EuIII-cyclam-HOPO and EuIII-TACN-HOPO are nine coordinate with three inner-sphere water molecules, suggesting that the two coordination states are very close in energy. As observed previously with linear analogues of tripodal HOPO complexes, there is no relationship between the number of inner-sphere water molecules and the affinity of the complex for phosphate. Whereas all three complexes do bind phosphate, EuIII-cyclen-HOPO has the highest affinity for phosphate with the anion displacing both of its inner-sphere water molecules. On the other hand, only one or two of the three inner-sphere water molecules of EuIII-TACN-HOPO and EuIII-cyclam-HOPO are displaced by phosphate, respectively. All three complexes are highly selective for phosphate over other anions, including arsenate. All three complexes are highly stable. EuIII-cyclen-HOPO and, to a lesser extent, EuIII-TACN-HOPO are more kinetically inert than the linear EuIII-Ser-HOPO. EuIII-cyclam-HOPO, on the other hand, is not. This study highlights the significant effect that minor changes in the ligand cap can have on both the ligand exchange rate and affinity for phosphate of tripodal 1,2-dihydroxypyridinonate complexes.

A seven-coordinate Mn(II) complex with a pyridine-based 15-membered macrocyclic ligand containing one acetate pendant arm: structure, stability and relaxation properties.

A new 15-membered pyridine-based macrocyclic ligand containing one acetate pendant arm (N-carboxymethyl-3,12,18-triaza-6,9-dioxabicyclo[12.3.1]octadeca-1(18),14,16-triene, L1) was synthesized and its Mn(II) complex MnL1 was investigated in the context of MRI contrast agent development. The X-ray molecular structure of MnL1 confirmed a coordination number of seven with an axially compressed pentagonal bipyramidal geometry and one coordination site available for an inner-sphere water molecule. The protonation constants of L1 and the stability constants of Mn(II), Zn(II), Cu(II) and Ca(II) complexes were determined by potentiometry, and revealed higher thermodynamic stabilities in comparison with complexes of 15-pyN3O2, the parent macrocycle without an acetate pendant arm. The MnL1 complex is fully formed at physiological pH 7.4, but it shows fast dissociation kinetics, as followed by relaxometry in the presence of an excess of Zn(II). The short dissociation half-life estimated for physiological pH (ca. 3 minutes) is related to fast spontaneous dissociation of the non-protonated complex. At lower pH values, the proton-assisted dissociation pathway becomes important, while the Zn(II) concentration has no effect on the dissociation rate. 17O NMR and 1H NMRD data indicated the presence of one inner-sphere water molecule with a rather slow exchange (k298ex = 4.5 × 106 s-1) and provided information about other microscopic parameters governing relaxation. The relaxivity (r1 = 2.45 mM-1 s-1 at 20 MHz, 25 °C) corresponds to typical values for monohydrated Mn(II) chelates. Overall, the acetate pendant arm in L1 has a beneficial effect with respect to 15-pyN3O2 in increasing the thermodynamic stability and kinetic inertness of its Mn(II) complex, but leads to a reduced number of inner-sphere water molecules and thus lower relaxivity.

The Ligand Cap Affects the Coordination Number but Not Necessarily the Affinity for Anions of Tris-Bidentate Europium Complexes.

To evaluate the effect of ligand geometry on the coordination number, number of inner-sphere water molecules, and affinity for anions of the corresponding lanthanide complex, six tris-bidentate 1,2-hydroxypyridonate (HOPO) europium(III) complexes with different cap sizes were synthesized and characterized. Wider or more flexible ligand caps, such as in EuIII-TREN-Gly-HOPO and EuIII-3,3-Gly-HOPO, enable the formation of nine-coordinate europium(III) complexes bearing three inner-sphere water molecules. In contrast, smaller or more rigid caps, such as in EuIII-TREN-HOPO, EuIII-2,2-Li-HOPO, EuIII-3,3-Li-HOPO, and EuIII-2,2-Gly-HOPO, favor eight-coordinate europium(III) complexes that have only two inner-sphere water molecules. Notably, there is no correlation between the number of inner-sphere water molecules and the affinity of the Eu(III) complexes for phosphate. Some q = 2 (EuIII-TREN-HOPO, EuIII-3,3-Li-HOPO, and EuIII-2,2-Gly-HOPO) and some q = 3 (EuIII-TREN-Gly-HOPO) complexes have no affinity for anions, whereas one q = 2 complex (EuIII-2,2-Li-HOPO) and one q = 3 complex (EuIII-3,3-Gly-HOPO) have a high affinity for phosphate. For the latter two systems, each inner-sphere water molecule is replaced with a phosphate anion, resulting in the formation of EuLPi2 and EuLPi3 adducts, respectively.

Comparing Strategies in the Design of Responsive Contrast Agents for Magnetic Resonance Imaging: A Case Study with Copper and Zinc

Magnetic resonance imaging (MRI) has emerged over the years as one of the preferred modalities for medical diagnostic and biomedical research. It has the advantage over other imaging modalities such as positron emission tomography and X-ray of affording high resolution three-dimensional images of the body without using harmful radiation. The use of contrast agents has further expanded this technique by increasing the contrast between regions where they accumulate and background tissues. As MRI most often measures the relaxation rate of water throughout the body, contrast agents function by modulating the intensity of the water signal either via improved relaxation or via saturation transfer to selected exchangeable proton. Among the growing class of MRI contrast agents, a subset of them called "smart" contrast agents function as responsive probes. Their ability to increase or decrease their signal intensity is modulated by the presence of an analyte. These probes offer the unique ability to image the distribution of an analyte in vivo, thereby opening new possibilities for diagnostics and for elucidating the role of specific analytes in various pathologies or biological processes. A number of different strategies can be exploited to design responsive MRI contrast agents. The majority of contrast agents are based on GdIII complexes. These complexes can be rendered responsive in either of two ways: either by modulating the number of inner-sphere water molecules, q, or via modulating the rotational correlation time, τR, of the contrast agent upon substrate binding. The longitudinal relaxivity increases with the number of inner-sphere water molecules. GdIII complexes can be rendered responsive if they contain a recognition moiety that can bind to both the open coordination site of GdIII and to the analyte. When the recognition moiety leaves the lanthanide ion to bind to the analyte, q increases and therefore so does the relaxivity. The dependence of relaxivity on rotational correlation time is more complex and more pronounced at lower magnetic fields. In general, slower tumbling macromolecules have longer rotational correlation times and higher relaxivities. Analyte-triggered formation of macromolecules thus also increases relaxivity. Such macromolecules can either be analyte-templated supramolecular assemblies, or analyte-enhanced protein-contrast agent complexes. Chemical Exchange Saturation Transfer (CEST) agents are a newer class of contrast agents that offer the possibility of multifrequency and thus ratiometric imaging, which in turn enables quantitative mapping of the concentration of an analyte in vivo under conditions where the concentration of the contrast agent is not known. Such agents can be rendered responsive if the analyte changes the number of exchangeable proton(s), its exchange rate, or its chemical shift. All of these approaches have been successfully employed for detecting and imaging both copper and zinc, including in vivo. Magnetic Iron Oxide Nanoparticles (MIONs) are powerful MRI transverse relaxation agents. They can also be rendered responsive to an analyte if the latter can control the aggregation of the nanoparticles. For metal ions, this can be achieved via chemical functionalities that only react to form conjugates in the presence of the metal ion analyte.

Drawing on biology to inspire molecular design: a redox-responsive MRI probe based on Gd(iii)-nicotinamide.

A novel, reversible redox-active MRI probe, GdNR1, has been developed for the study of redox changes associated with diseased states. This system exhibits switching in relaxivity upon reduction and oxidation of the appended nicotinimidium. Relaxivity studies and cyclic voltammetry confirmed the impressive reversibility of this system, at a biologically-relevant reduction potential. A 2.5-fold increase in relaxivity was observed upon reduction of the complex, which corresponds to a change in the number of inner-sphere water molecules, as confirmed by luminescence lifetimes of the Eu(iii) analogue and NMRD studies. This is the first example of a redox-responsive MRI probe utilising the biologically-inspired nicotinimidium redox switch. In the future this strategy could enable the non-invasive identification of hypoxic tissue and related cardiovascular disease.

Evaluation of Water Exchange Kinetics on [Ln(AAZTAPh-NO2)(H2O)q](x) Complexes Using Proton Nuclear Magnetic Resonance.

Water exchange kinetics on [Ln(AAZTAPh-NO2)(H2O)q](-) (Ln = Gd(3+), Dy(3+), or Tm(3+)) were determined by (1)H nuclear magnetic resonance (NMR) measurements. The number of inner-sphere water molecules was found to change from two to one when going from Dy(3+) to Tm(3+). The calculated water exchange rate constants obtained by variable-temperature proton transverse relaxation rates are 3.9 × 10(6), 0.46 × 10(6), and 0.014 × 10(6) s(-1) at 298 K for Gd(3+), Dy(3+), and Tm(3+), respectively. Variable-pressure measurements were used to assess the water exchange mechanism. The results indicate an associative and dissociative interchange mechanism for Gd(3+) and Dy(3+) complexes with ΔV(⧧) values of -1.4 and 1.9 cm(3) mol(-1), respectively. An associative activation mode (Ia or A mechanism) was obtained for the Tm(3+) complex (ΔV(⧧) = -5.6 cm(3) mol(-1)). Moreover, [Dy(AAZTAPh-NO2)(H2O)2](-) with a very high transverse relaxivity value was found as a potential candidate for negative contrast agents for high-field imaging applications.

Luminescent europium and terbium complexes of dipyridoquinoxaline and dipyridophenazine ligands as photosensitizing antennae: structures and biological perspectives.

The europium(III) and terbium(III) complexes, namely [Eu(dpq)(DMF)2(NO3)3] (1), [Eu(dppz)2(NO3)3] (2), [Tb(dpq)(DMF)2Cl3] (3), and [Tb(dppz)(DMF)2Cl3] (4), where dipyrido[3,2-d:2',3'-f]quinoxaline (dpq in 1 and 3), dipyrido[3,2-a:2',3'-c]phenazine (dppz in 2 and 4) and N,N'-dimethylformamide (DMF) have been isolated, characterized from their physicochemical data, luminescence studies and their interaction with DNA, serum albumin protein and photo-induced DNA cleavage activity are studied. The X-ray crystal structures of complexes 1-4 show discrete mononuclear Ln(3+)-based structures. The Eu(3+) in [Eu(dpq)(DMF)2(NO3)3] (1) and [Eu(dppz)2(NO3)3] (2) as [Eu(dppz)2(NO3)3]·dppz (2a) adopts a ten-coordinated bicapped dodecahedron structure with a bidentate N,N-donor dpq ligand, two DMF and three NO3(-) anions in 1 and two bidentate N,N-donor dppz ligands and three NO3(-) anions in 2. Complexes 3 and 4 show a seven-coordinated mono-capped octahedron structure where Tb(3+) contains bidentate dpq/dppz ligands, two DMF and three Cl(-) anions. The complexes are highly luminescent in nature indicating efficient photo-excited energy transfer from the dpq/dppz antenna to Ln(3+) to generate long-lived emissive excited states for characteristic f → f transitions. The time-resolved luminescence spectra of complexes 1-4 show typical narrow emission bands attributed to the (5)D0 → (7)F(J) and (5)D4 → (7)F(J) f-f transitions of Eu(3+) and Tb(3+) ions respectively. The number of inner-sphere water molecules (q) was determined from luminescence lifetime measurements in H2O and D2O confirming ligand-exchange reactions with water in solution. The complexes display significant binding propensity to the CT-DNA giving binding constant values in the range of 1.0 × 10(4)-6.1 × 10(4) M(-1) in the order 2, 4 (dppz) > 1, 3 (dpq). DNA binding data suggest DNA groove binding with the partial intercalation nature of the complexes. All the complexes also show binding propensity (K(BSA) ∼ 10(5) M(-1)) to the bovine serum albumin (BSA) protein. The intensity of the time-gated luminescence spectral bands enhances significantly with the increasing DNA concentration in aqueous buffer medium due to displacement of bound water upon interaction with DNA, thus reducing non-radiative quenching through the O-H oscillator. Complexes 1-4 efficiently cleave supercoiled (SC) ds-DNA to its nicked circular (NC) form on exposure to UV-A light of 365 nm via formation of singlet oxygen ((1)O2) and hydroxyl radicals (HO˙) as the reactive oxygen species at micromolar concentrations under physiological conditions.

Comprehensive Evaluation of the Physicochemical Properties of LnIII Complexes of Aminoethyl‐DO3A as pH‐Responsive T1‐MRI Contrast Agents

N-Substituted aminoethyl groups were attached to 1,4,7,10-tetraazacyclododecane-1,4,7-triacetic acid (DO3A) with the aim to design pH-responsive Ln(III) complexes based on the pH-dependent on/off ligation of the amine nitrogen to the metal ion. The following ligands were synthesized: AE-DO3A (aminoethyl-DO3A), MAE-DO3A (N-methylaminoethyl-DO3A), DMAE-DO3A (N,N-dimethylaminoethyl-DO3A) and MEM-AE-DO3A (N-methoxyethyl-N-methylaminoethyl-DO3A). The physicochemical properties of the Ln(III) complexes were investigated for the evaluation of their potential applicability as magnetic resonance imaging (MRI) contrast agents. In particular, a (1) H and (17) O NMR relaxometric study was carried out for these Gd(III) complexes at two different pH values: at basic pH (pendant amino group coordinated to the metal centre) and at acidic pH (protonated amine, not interacting with the metal ion). Eu(III) complexes allow one to estimate the number of inner-sphere water molecules through luminescence lifetime measurements and obtain some structural information through variable-temperature (VT) high-resolution (1) H NMR studies. Equilibria between differently hydrated species were found for most of the complexes at both acidic and basic pH. The thermodynamic stability of Ca(II) , Zn(II) , Cu(II) and Ln(III) complexes and kinetics of formation and dissociation reactions of Ln(III) complexes of AE-DO3A and DMAE-DO3A were investigated showing stabilities comparable to currently approved Gd(III) -based CAs. In detail, higher total basicity (Σlog Ki (H) ) and higher stability constants of Ln(III) complexes were found for AE-DO3A with respect to DMAE-DO3A (i.e., log KGd-AE-DO3A =22.40 and log KGd-DMAE-DO3A =20.56). The transmetallation reactions of Gd(III) complexes are very slow (Gd-AE-DO3A: t1/2 =2.7×10(4) h; Gd-DMAE-DO3A: 1.1×10(5) h at pH 7.4 and 298 K) and occur through proton-assisted dissociation.

Dependence of the Photophysical Properties on the Number of 2,2′-Bipyridine Units in a Series of Luminescent Europium and Terbium Cryptates

The luminescence properties of a series of lanthanoid cryptates with an increasing number of 2,2'-bipyridine units have been investigated for the lanthanoids Eu and Tb in aqueous solution. The trends in important parameters that influence the photophysics in these complexes have been determined. With increasing bipyridine content, an increase is observed for the intersystem crossing efficiencies and the number of inner-sphere water molecules. In contrast, a decrease is found in the same direction for overall quantum yields, triplet energies, and sensitization efficiencies.

Activatable T 1 and T 2 Magnetic Resonance Imaging Contrast Agents

Magnetic resonance imaging (MRI) has become one of the most important diagnosis tools available in medicine. Typically MRI is not capable of sensing biochemical activities. However, recently emerged activatable MRI contrast agents (CAs), whose relaxivity is variable in response to a specific parameter change in the surrounding physiological microenvironment, potentially allow for MRI to indicate biological processes. Among the various factors influencing the relaxivity of a CA, the number of inner-sphere water molecules (q) directly coordinated to the metal center, the residence time of the coordinated water molecule (τ m), and the rotational correlation time representing the molecular tumbling time of a complex (τ R) contribute strongly to the relaxivity of an activatable CA. Tuning the ligand structure and properties has been the subject of intensive research for activatable MR CA designs. This review summarizes a variety of activatable MRI CAs sensitive to common variables in microenvironment in vivo, i.e., pH, luminescence, metal ions, redox, and enzymes, etc., with emphasis on the influence of ligand design on parameters q, τ m, and τ R.

A gadolinium(III) complex with 8-amidequinoline based ligand as copper(II) ion responsive contrast agent

A new Cu(2+)-responsive MRI contrast agent (Gd-QDOTAMA) with a quinoline-based ligand was synthesized and characterized. Relaxivity studies on Gd-QDOTAMA showed that the relaxivity increased from 4.27 mM(-1) s(-1) to 7.29 mM(-1) s(-1) in response to equimolar amounts of copper(II) ion, corresponding to ca. 71% relaxivity enhancement. Distinct changes in relaxivity were undetected upon addition of physiologically relevant alkali metal cations (K(+) or Na(+)), alkaline earth metal cations (Mg(2+) or Ca(2+)), or d-block metal cations (Zn(2+), Cu(+), Fe(2+), Fe(3+)), indicating a high selectivity for Cu(2+) over other biologically relevant metal ions. Moreover, the influence of common biological anions at physiological levels on the Cu(2+)-responsive contrast agent was also studied. Luminescence studies on the Eu counterpart Eu-QDOTAMA suggest that the enhancement in relaxivity for Gd-QDOTAMA in response to Cu(2+) is most likely due to the increased number of inner-sphere water molecules around Gd(3+) upon Cu(2+) binding to the 8-amidequinoline moiety. In vitro T(1)-weighted phantom images of Gd-QDOTAMA confirmed that signal intensity was markedly increased by the addition of equimolar amounts of Cu(2+).

Gd−Hydroxypyridinone (HOPO)-Based High-Relaxivity Magnetic Resonance Imaging (MRI) Contrast Agents

Magnetic resonance imaging (MRI) is a particularly effective tool in medicine because of its high depth penetration (from 1 mm to 1 m) and ability to resolve different soft tissues. The MRI signal is generated by the relaxation of in vivo water molecule protons. MRI images can be improved by administering paramagnetic agents, which increase the relaxation rates of nearby water protons, thereby enhancing the MRI signal. The lanthanide cation Gd(3+) is generally used because of its favorable electronic properties; high toxicity, however, necessitates strongly coordinating ligands to keep Gd(3+) completely bound while in the patient. In this Account, we give a coordination chemistry overview of contrast agents (CAs) based on Gd-hydroxypyridinone (HOPO), which show improved MRI contrast and high thermodynamic stabilities. Tris-bidentate HOPO-based ligands developed in our laboratory were designed to complement the coordination preferences of Gd(3+), especially its oxophilicity. The HOPO ligands provide a hexadentate coordination environment for Gd(3+), in which all of the donor atoms are oxygen. Because Gd(3+) favors eight or nine coordination, this design provides two to three open sites for inner-sphere water molecules. These water molecules rapidly exchange with bulk solution, hence affecting the relaxation rates of bulk water molecules. The parameters affecting the efficiency of these contrast agents have been tuned to improve contrast while still maintaining a high thermodynamic stability for Gd(3+) binding. The Gd-HOPO-based contrast agents surpass current commercially available agents because of a higher number of inner-sphere water molecules, rapid exchange of inner-sphere water molecules via an associative mechanism, and a long electronic relaxation time. The contrast enhancement provided by these agents is at least twice that of commercial contrast agents, which are based on polyaminocarboxylate ligands. Advances in MRI technology have made significant contributions to the improvement of clinical diagnostics by allowing visualization of underlying pathology. However, understanding the mechanism of a disease at the molecular level requires improved imaging sensitivity. The ultimate goal is to visually distinguish between different disease targets or markers, such as enzymes, hormones, proteins, or small molecules, at biologically relevant concentrations (from micro- to nanomolar). Although MRI techniques can provide images of the organs and tissues in which these biomarkers are regulated, the high sensitivity required to visualize the biological targets within the tissues is currently lacking; contrast enhancements of 50-fold beyond current agents are required to achieve this goal. According to the theory of paramagnetic relaxation, the contrast enhancement can be further improved by slowing the tumbling rate of the MRI agent. Theoretically, this enhancement would be greater for contrast agents with an optimal rate of water exchange. The Gd-HOPO-based contrast agents have optimal water-exchange rates, whereas the commercial agents have slower non-optimal water-exchange rates; thus, the Gd-HOPO agents are ideal for attachment to macromolecules, which will slow down the tumbling rate and increase contrast. This strategy has been recently tested with the Gd-HOPO agents via covalent attachment to virus capsids, affording contrast enhancements 10-fold beyond commercial agents.

Principles of responsive lanthanide-based luminescent probes for cellular imaging

The advent of chemical tools for cellular imaging--from organic dyes to green fluorescent proteins--has revolutionized the fields of molecular biology and biochemistry. Lanthanide-based probes are a new player in this area, as the last decade has seen the emergence of the first responsive luminescent lanthanide probes specifically intended for imaging cellular processes. The potential of these probes is still undervalued by the scientific community. Indeed, this class of probes offers several advantages over organic dyes and fluorescent proteins. Their very long luminescence lifetimes enable quantitative spatial determination of the intracellular concentration of an analyte through time-gating measurements. Their emission bands are very narrow and do not overlap, enabling the simultaneous use of multiple lanthanide probes to quantitatively detect several analytes without cross-interference. Herein we describe the principles behind the development of this class of probes. Sensors for a desired analyte can be designed by rationally manipulating the parameters that influence the luminescence of lanthanide complexes. We will discuss sensors based on varying the number of inner-sphere water molecules, the distance separating the antenna from the lanthanide ion, the energies of excited states of the antenna, and PeT switches.

Mechanistic studies of a calcium-dependent MRI contrast agent.

Intracellular Ca(2+) plays an important role in signal transduction, and we are developing new MRI techniques to study its regulation in living animals. We have reported on an MRI contrast agent (DOPTA-Gd) where the relaxivity of the complex is controlled by the presence or absence of the divalent ion Ca(2+). By structurally modulating inner-sphere access of water to a chelated Gd(3+) ion, we observe a substantial and reversible change in T(1) upon the addition of Ca(2+) and not other divalent ions. Luminescence lifetime and NMRD measurements of the complex have been acquired, and several parameters contribute to the Ca(2+) dependent relaxivity change of DOPTA-Gd. The number of inner-sphere water molecules is more than doubled after the Ca(2+) concentration is increased. This finding strongly supports the proposed conformational change of DOPTA-Gd when Ca(2+) is bound. Relaxometric measurements confirm these results and provide an indication that second-sphere water molecules are probably responsible for paramagnetic relaxation enhancement in the absence of Ca(2+). After Ca(2+) is bound to DOPTA-Gd, the molecule undergoes a substantial conformational change that opens up the hydrophilic face of the tetraazacyclododecane macrocycle. This change dramatically increases the accessibility of chelated Gd(3+) ion to bulk solvent. The design of this class of calcium-activated MR contrast agent was based primarily on the assumption that the number of coordinated inner-sphere water molecules would be the dominating factor in observed relaxivity measurements. This result has been confirmed; however, careful mechanistic studies reveal that additional factors are involved in this process.

The protonation constants of four poly(aminocarboxylates), N′-pyridylmethyl (TTDA-PY), N′-2-hydroxypropyl (TTDA-HP), N′-2-hydroxy-1-phenylethyl (TTDA-H1P) and N′-2-hydroxy-2-phenylethyl (TTDA-H2P) derivatives of TTDA (3,6,10-tri(carboxymethyl)-3,6,10-triazadodecanedioic acid), and the stability constants of their complexes formed with Ca2+, Zn2+, Cu2+, La3+, Ce3+, Nd3+, Sm3+, Gd3+, Dy3+, Ho3+, Yb3+ and Lu3+ were determined by potentiometric methods at 25.0 ± 0.1 °C and 0.10 mol dm−3 ionic strength in Me4NNO3. The stability of the Gd(III) complexes follows the order TTDA-PY > TTDA-H2P ≊ TTDA-HP ≊ TTDA-H1P. The thermodynamic resistance of the gadolinium(III) complexes, at the plasma concentration used in clinical applications, towards demetallation in the presence of important components of blood plasma, such as Ca(II), Zn(II) and Cu(II), has been evaluated. The observed water proton relaxivity values of [Gd(TTDA-PY)]−, [Gd(TTDA-HP)]−, [Gd(TTDA-H1P)]− and [Gd(TTDA-H2P)]− became constant with respect to pH changes over the range of 4–10, 6–10, 6–10 and 6–10, respectively. 17O NMR shifts showed that the [Dy(TTDA-PY)]−, [Dy(TTDA-HP)]−, [Dy(TTDA-H1P)]− and [Dy(TTDA-H2P)]− complexes at pH 6.30 and 4.30 had 0.9 and 0.9; 2.2 and 3.4; 2.0 and 3.5; 1.8 and 3.0 inner-sphere water molecules, respectively. Water proton spin–lattice relaxation rates for gadolinium(III) complexes were also consistent with the number of inner-sphere water molecules. The EPR transverse electronic relaxation rate and 17O NMR transverse relaxation time were thoroughly investigated and the results obtained were compared with that previously reported for the other gadolinium(III) complex, [Gd(DTPA)(H2O)]2−. Short exchange lifetime values were obtained for the [Gd(TTDA)(H2O)]2−, [Gd(TTDA-PY)(H2O)]−, [Gd(TTDA-HP)(H2O)2]−, [Gd(TTDA-H1P)(H2O)2]− and [Gd(TTDA-H2P)(H2O)2]− complexes. Their water-exchange rates are about 24–45 times faster than that for the [Gd(DTPA)(H2O)]2− complex, which suggested that the longer backbone of the multidentate ligand may be pulled tightly into the first coordination sphere, resulting in high steric constraints at the water binding site.

A tripod gadolinium (III) poly(amino carboxylate) relevant to magnetic resonance imaging: structural and dynamical 17O NMR and 1H NMRD studies

We present the results of multiple temperature 1H NMRD and variable temperature and pressure, multiple field 17O NMR studies on the gadolinium(III) complex of the tripod ligand H 6ttaha N-tris(2-aminoethyl)amine- N′-hexaacetate in aqueous solution. Of the ten donor atoms, seven are coordinated to the center metal, leaving one of the tripod ‘legs’ free. The coordination number is completed to nine by two water molecules ( q = 2), as confirmed by the value of the scalar coupling constant A/ h = (− 3.5 ± 0.1) × 10 6 rad s −1. The higher number of inner-sphere water molecules causes a higher 1H relaxivity as compared to commercial gadolinium(III) based MRI contrast agents. The water exchange rate was determined as K 298=(8.6±0.6) × 10 6s −1. The enthalpy and volume of activation are ΔH = (40.4±1.4) kJ mol −1 and ΔV = (2.9±1.0) cm 3mol −1, respectively, the latter indicating a dissociatively activated interchanged I d mechanism. The water exchange is unexpectedly fast for an exchange reaction that is not limiting dissociative D; this lability is interpreted as being due to the higher number of inner-sphere water molecules.

Extent of hydration of octadentate lanthanide complexes incorporating phosphinate donors: solution relaxometry and luminescence studies

The behaviour of luminescent (Eu, Tb) and highly paramagnetic (Gd) complexes of 1,4,7,10-tetraazacyclododecane containing one carboxamide and three phosphinate substituents has been studied in solution. Analyses of variable-temperature nuclear magnetic relaxation dispersion profiles indicate that there is no water molecule directly co-ordinated to gadolinium(III) ions. It has been found that the observed relaxation enhancement of solvent protons is determined, in addition to the contribution from water molecules diffusing in close proximity to the paramagnetic complex, by a relatively distant water molecule in the ‘second co-ordination sphere’. This is possible because the amide carbonyl oxygen can participate in hydrogen bonding (as a hydrogen-bond acceptor) to a local water molecule, which brings the water molecule close to the metal ion. The luminescence spectra of the complexes of Eu and Tb in water and D2O are also consistent with such a hydration scheme and there is a good correlation between the non-integral q value (number of inner-sphere water molecules) determined by this method and the distance between the metal ion and the water proton estimated by relaxometric methods. It is proposed that the hydration states q= 0 and 1 may be considered to represent boundary conditions and a given complex in solution may possess intermediate values.

Determination of the number of inner-sphere water molecules in lanthanide(III) polyaminocarboxylate complexes

The hydration numbers of various dysprosium(III) polyaminocarboxylates have been determined from the DyIII-induced 17O NMR water shifts. The results are in good agreement with those obtained by other techniques. The data indicate that formation of polynuclear species occurs in the iminodiacetate and nitrilotriacetate systems.

PMR investigation of the state of water of hydration in ethylenediaminetetraacetates of rare-earth elements

1. Analysis of the temperature dependence of band intensities in the PMR spectra of the hydrates MLnA·nH2O showed that the protons of inner-sphere water in the complex salts are manifested in the PMR spectra only in the form of a broad line, whereas those of outer-sphere water are manifested as both broad and narrow lines. 2. The number of inner-sphere water molecules in solid hydrates of rare-earth ethylenediaminetetraacetates decreases from La to Lu and is equal to three for derivatives of light lanthanides and not more than one for Yb derivatives.
